The production of hydrogen by steam reforming of bio-oils obtained from the fast pyrolysis of biomass requires the development of efficient catalysts able to cope with the complex chemical nature of the reactant. The present work focuses on the use of noble metal-based catalysts for the steam reforming of a few model compounds and that of an actual bio-oil. The steam reforming of the model compounds was investigated in the temperature range 650-950 degrees C over Pt, Pd and Rh supported on alumina and a ceria-zirconia sample. The model compounds used were acetic acid, phenol, acetone and ethanol. The nature of the support appeared to play a significant role in the activity of these catalysts. The use of ceria-zirconia, a redox mixed oxide, lead to higher H-2 yields as compared to the case of the alumina-supported catalysts. The supported Rh and Pt catalysts were the most active for the steam reforming of these compounds, while Pd-based catalysts poorly performed. The activity of the promising Pt and Rh catalysts was also investigated for the steam reforming of a bio-oil obtained from beech wood fast pyrolysis. Temperatures close to, or higher than, 800 degrees C were required to achieve significant conversions to COx and H-2 (e.g., H-2 yields around 70%). The ceria-zirconia materials showed a higher activity than the corresponding alumina samples. A Pt/ceria-zirconia sample used for over 9 h showed essentially constant activity, while extensive carbonaceous deposits were observed on the quartz reactor walls from early time on stream. In the present case, no benefit was observed by adding a small amount of O-2 to the steam/bio-oil feed (autothermal reforming, ATR), probably partly due to the already high concentration of oxygen in the bio-oil composition. Ionising radiation plays a key role in therapy due to its ability to directly induce DNA damage, in particular DNA double-strand breaks leading to cell death. Cells have multiple repair pathways which attempt to maintain genomic stability. DNA repair proteins have become key targets for therapy, using small molecule inhibitors, in combination with radiation and or chemotherapeutic agents as a means of enhancing cell killing. Significant advances in our understanding of the response of cells to radiation exposures has come from the observation of non-targeted effects where cells respond via mechanisms other than those which are a direct consequence of energy-dependent DNA damage. Typical of these is bystander signalling where cells respond to the fact that their neighbours have been irradiated. Bystander cells show a DNA damage response which is distinct from directly irradiated cells. In bystander cells, ATM- and Rad3-related (ATR) protein kinase-dependent signalling in response to stalled replication forks is an early event in the DNA damage response. The ATM protein kinase is activated downstream of ATR in bystander cells. This offers the potential for differential approaches for the modulation of bystander and direct effects with repair inhibitors which may impact on the response of tumours and on the protection of normal tissues during radiotherapy. he influence of poly(ethylene glycol) (PEG) plasticiser content and molecular weight on the physicochemical properties of films cast from aqueous blends of poly(methyl vinyl ether-co-maleic acid) was investigated using thermal analysis, swelling studies, scanning electron microscopy (SEM) and attenuated total reflectance (ATR)-Fourier transform infrared (FTIR) spectroscopy. FTIR spectroscopy revealed a shift of the CO peak from 1708 to 1731 cm-1, indicating that an esterification reaction had occurred upon heating, thus producing crosslinked films. Higher molecular weight PEGs (10,000 and 1000 Da, respectively), having greater chain length, producing hydrogel networks with lower crosslink densities and higher average molecular weight between two consecutive crosslinks. Accordingly, such materials exhibited higher swelling rates. Hydrogels crosslinked with a low molecular weight PEG (PEG 200) showed rigid networks with high crosslink densities and, therefore, lower swelling rates. Polymer:plasticizer ratio alteration did not yield any discernable patterns, regardless of the method of analysis. The polymer–water interaction parameter (?) increased with increases in the crosslink density. SEM studies showed that porosity of the crosslinked films increased with increasing PEG MW, confirming what had been observed with swelling studies and thermal analysis, that the crosslink density must be decreased as the Mw of the crosslinker is increased. Hydrogels containing PMVE/MA/PEG 10,000 could be used for rapid delivery of drug, due to their low crosslink density. Moderately crosslinked PMVE/MA/PEG 1000 hydrogels or highly crosslinked PMVE/MA/PEG 200 systems could then be used in controlling the drug delivery rates. We are currently evaluating these systems, both alone and in combination, for use in sustained release drug delivery devices.

The a-b plane dielectric function (epsilon) of c-axis YBa2Cu3O7-delta thin films with T-c > 85 K was measured at lambda = 3.392 mum in the temperature range 85-300 It, using an attenuated total reflectance (ATR) technique based on the excitation of surface plasmons, The results show that \epsilon (r)\ decreases quasi-linearly with increasing temperature, while Ei is invariant to temperature within experimental uncertainties. Typical values are epsilon (ab) = -23 + 16.5i at similar to 295 R and epsilon (ab) = -27 + 15.5i at similar to 90 K. A generalised Drude analysis yields effective scattering rates (1/tau*) that increase with temperature from similar to 1500 to similar to 1900 cm(-1). The temperature dependent rates best fit an equation of the form 1/tau* = a + bT(alpha) with alpha = 1.46 +/- 0.40. The effective plasma frequencies of w(p)* similar to 18,500 cm(-1) are almost independent of temperature. The uniquely detailed temperature dependence of the results confirm and consolidate data obtained by other groups using normal reflectance methods, but contradict our previously published ATR measurements. Cdk2 and cdk1 are individually dispensable for cell-cycle progression in cancer cell lines because they are able to compensate for one another. However, shRNA-mediated depletion of cdk1 alone or small molecule cdk1 inhibition abrogated S phase cell-cycle arrest and the phosphorylation of a subset of ATR/ATM targets after DNA damage. Loss of DNA damage-induced checkpoint control was caused by a reduction in formation of BRCA1-containing foci. Mutation of BRCA1 at S1497 and S1189/S1191 resulted in loss of cdk1-mediated phosphorylation and also compromised formation of BRCA1-containing foci. Abrogation of checkpoint control after cdk1 depletion or inhibition in non-small-cell lung cancer cells sensitized them to DNA-damaging agents. Conversely, reduced cdk1 activity caused more potent G2/M arrest in nontransformed cells and antagonized the response to subsequent DNA damage. Cdk1 inhibition may therefore selectively sensitize BRCA1-proficient cancer cells to DNA-damaging treatments by disrupting BRCA1 function.

Otto configuration attenuated total reflection (ATR) measurements of the excitation of surface plasmons in the infrared have been carried out on YBCO films deposited on MgO (100) substrates. The dielectric constants for YBCO at 3.392 mu m are determined to be -10 + 15i for c-axis material. The anisotropic nature of the cuprate is seen from films with other orientations: nearly a-axis material has constants of 4.0 + 7.0i. It is thus not metallic in its optical response along the c-axis which lies parallel to the substrate plane. Ellipsometric measurements in the visible on c-axis material point to a maximum surface plasmon energy of 1 eV.

BRCA1 is a major player in the DNA damage response. This is evident from its loss, which causes cells to become sensitive to a wide variety of DNA damaging agents. The major BRCA1 binding partner, BARD1, is also implicated in the DNA damage response, and recent reports indicate that BRCA1 and BARD1 co-operate in this pathway. In this report, we utilized small interfering RNA to deplete BRCA1 and BARD1 to demonstrate that the BRCA1-BARD1 complex is required for ATM/ATR (ataxia-telangiectasia-mutated/ATM and Rad3-related)-mediated phosphorylation of p53(Ser-15) following IR- and UV radiation-induced DNA damage. In contrast, phosphorylation of a number of other ATM/ATR targets including H2AX, Chk2, Chk1, and c-jun does not depend on the presence of BRCA1-BARD1 complexes. Moreover, prior ATM/ATR-dependent phosphorylation of BRCA1 at Ser-1423 or Ser-1524 regulates the ability of ATM/ATR to phosphorylate p53(Ser-15) efficiently. Phosphorylation of p53(Ser-15) is necessary for an IR-induced G(1)/S arrest via transcriptional induction of the cyclin-dependent kinase inhibitor p21. Consistent with these data, repressing p53(Ser-15) phosphorylation by BRCA1-BARD1 depletion compromises p21 induction and the G(1)/S checkpoint arrest in response to IR but not UV radia-tion. These findings suggest that BRCA1-BARD1 complexes act as an adaptor to mediate ATM/ATR-directed phosphorylation of p53, influencing G(1)/S cell cycle progression after DNA damage.

In mammals, the ATM (ataxia-telangiectasia-mutated) and ATR (ATM and Rad3-related) protein kinases function as critical regulators of the cellular DNA damage response. The checkpoint functions of ATR and ATM are mediated, in part, by a pair of checkpoint effector kinases termed Chk1 and Chk2. In mammalian cells, evidence has been presented that Chk1 is devoted to the ATR signaling pathway and is modified by ATR in response to replication inhibition and UV-induced damage, whereas Chk2 functions primarily through ATM in response to ionizing radiation (IR), suggesting that Chk2 and Chk1 might have evolved to channel the DNA damage signal from ATM and ATR, respectively. We demonstrate here that the ATR-Chk1 and ATM-Chk2 pathways are not parallel branches of the DNA damage response pathway but instead show a high degree of cross-talk and connectivity. ATM does in fact signal to Chk1 in response to IR. Phosphorylation of Chk1 on Ser-317 in response to IR is ATM-dependent. We also show that functional NBS1 is required for phosphorylation of Chk1, indicating that NBS1 might facilitate the access of Chk1 to ATM at the sites of DNA damage. Abrogation of Chk1 expression by RNA interference resulted in defects in IR-induced S and G(2)/M phase checkpoints; however, the overexpression of phosphorylation site mutant (S317A, S345A or S317A/S345A double mutant) Chk1 failed to interfere with these checkpoints. Surprisingly, the kinase-dead Chk1 (D130A) also failed to abrogate the S and G(2) checkpoint through any obvious dominant negative effect toward endogenous Chk1. Therefore, further studies will be required to assess the contribution made by phosphorylation events to Chk1 regulation. Overall, the data presented in the study challenge the model in which Chk1 only functions downstream from ATR and indicate that ATM does signal to Chk1. In addition, this study also demonstrates that Chk1 is essential for IR-induced inhibition of DNA synthesis and the G(2)/M checkpoint.

Transcription byRNApolymerase I (Pol-I) is the main driving force behind ribosome biogenesis, a fundamental cellular process that requires the coordinated transcription of all three nuclear polymerases. Increased Pol-I transcription and the concurrent increase in ribosome biogenesis has been linked to the high rates of proliferation in cancers. The ellipticine family contains a number of potent anticancer therapeutic agents, some having progressed to stage I and II clinical trials; however, the mechanism by which many of the compounds work remains unclear. It has long been thought that inhibition of Top2 is the main reason behind the drugs antiproliferative effects. Here we report that a number of the ellipticines, including 9-hydroxyellipticine, are potent and specific inhibitors of Pol-I transcription, with IC50 in vitro and in cells in the nanomolar range. Essentially, the drugs did not affect Pol-II and Pol-III transcription, demonstrating a high selectivity.Wehave shown that Pol-I inhibition occurs by a p53-, ATM/ATR-, and Top2-independent mechanism. We discovered that the drug influences the assembly and stability of preinitiation complexes by targeting the interaction between promoter recognition factor SL1 and the rRNA promoter. Our findings will have an impact on the design and development of novel therapeutic agents specifically targeting ribosome biogenesis.

A new experimental procedure based on attenuated total reflection infrared spectroscopy has been developed to investigate surface species under liquid phase reaction conditions. The technique has been tested by investigating the enhanced selectivity in the hydrogenation of α,β-unsaturated aldehyde citral over a 5% Pt/SiO2 catalyst toward unsaturated alcohols geraniol/nerol, which occurs when citronellal is added to the reaction. The change in selectivity is proposed to be the result of a change in the citral adsorption mode in the presence of citronellal. Short time on stream attenuated total internal reflection infrared spectroscopy has allowed identification of the adsorption modes of citral. With no citronellal, citral adsorbs through both the C═C and C═O groups; however, in the presence of citronellal, citral adsorption occurs through the C═O group only, which is proposed to be the cause of the altered reaction selectivity.

Radiotherapy is an important treatment option for many human cancers. Current research is investigating the use of molecular targeted drugs in order to improve responses to radiotherapy in various cancers. The cellular response to irradiation is driven by both direct DNA damage in the targeted cell and intercellular signalling leading to a broad range of bystander effects. This study aims to elucidate radiation-induced DNA damage response signalling in bystander cells and to identify potential molecular targets to modulate the radiation induced bystander response in a therapeutic setting. Stalled replication forks in T98G bystander cells were visualised via bromodeoxyuridine (BrdU) nuclear foci detection at sites of single stranded DNA. γH2AX co-localised with these BrdU foci. BRCA1 and FANCD2 foci formed in T98G bystander cells. Using ATR mutant F02-98 hTERT and ATM deficient GM05849 fibroblasts it could be shown that ATR but not ATM was required for the recruitment of FANCD2 to sites of replication associated DNA damage in bystander cells whereas BRCA1 bystander foci were ATM-dependent. Phospho-Chk1 foci formation was observed in T98G bystander cells. Clonogenic survival assays showed moderate radiosensitisation of directly irradiated cells by the Chk1 inhibitor UCN-01 but increased radioresistance of bystander cells. This study identifies BRCA1, FANCD2 and Chk1 as potential targets for the modulation of radiation response in bystander cells. It adds to our understanding of the key molecular events propagating out-of-field effects of radiation and provides a rationale for the development of novel molecular targeted drugs for radiotherapy optimisation.

Purpose: To assess the demographics and distribution of corneal astigmatism before cataract surgery in Chinese patients. Setting: State Key Laboratory of Ophthalmology, Zhongshan Ophthalmic Center, Sun Yat-sen University, Guangzhou, China. Design: Clinic-based cross-sectional study. Methods: From July 2009 to May 2011, preoperative bilateral partial coherence interferometry (IOLMaster) was performed in consecutive patients having cataract surgery. Patient demographics and keratometric data were recorded. Results: The mean age of the 2849 patients (4831 eyes) was 70.56 years ± 9.55 (SD); there was a predominance of women patients (64.0%). The mean axial length was 23.58 ± 1.13 mm. The mean corneal astigmatism in this cohort was 1.01 D (range 0.05 to 6.59 D). Corneal astigmatism was between 0.25 D and 1.25 D in 67.7% of eyes, 1.25 D or higher in 27.5% eyes, and less than 0.25 D in 4.8% of eyes. Astigmatism was with the rule in 25.1% of eyes, against the rule (ATR) in 58.2% of eyes, and oblique in 16.7% of eyes. The mean steep keratometry measurement was 44.76 ± 1.56 D. Against-the-rule astigmatism increased significantly with older age. Conclusions: Corneal astigmatism largely fell between 0.25 D and 1.25 D in these predominantly elderly female Chinese patients, and ATR astigmatism increased with age. Atendendo à produção de epóxidos em larga escala e à sua importância como intermediários versáteis, muita atenção tem sido dada à epoxidação de olefinas. Destaca-se a implementação do processo industrial de epoxidação de propileno em fase líquida com tBHP, usando complexos de molibdénio como catalisadores homogéneos (Halcon-ARCO). Neste trabalho foram investigados novos complexos à base de molibdénio como catalisadores (ou precursores) para epoxidação de olefinas em fase líquida. Foi objecto de estudo a identificação das espécies activas e a estabilidade dos catalisadores através da sua separação no final das reacções catalíticas, caracterização e reutilização. Escolheu-se como reacção modelo a epoxidação do ciscicloocteno com tBHP (em decano, tBHPdec), a 55 ºC. Estendeu-se o estudo dos desempenhos catalíticos a diferentes substratos, oxidantes, solventes e métodos de aquecimento. A maior actividade catalítica foi observada para os complexos [MoO2Cl2L2] (L=ligando dialquilamida), mais estáveis e fáceis de manusear que [MoO2Cl2] e complexos análogos com L {THF, MeCN} (Cap. 2). A partir destes complexos podem-se formar in situ espécies activas intermediárias do tipo [(MoO2ClL2)2(μ-O)]. O complexo [MoO2(Lzol)], Lzol= ligando oxazolina quiral (Cap. 3), é um catalisador estável e versátil, activo para a epoxidação de diversas olefinas (selectividades elevadas para epóxidos, mas enantioselectividades baixas), desidrogenação oxidativa de álcoois e sulfoxidação de sulfuretos. O catalisador foi também reciclado eficientemente, usando um líquido iónico (LI). O complexo iónico [MoO2Cl{HC(3,5-Me2pz)3}]BF4 (Cap.4) converteu-se nos complexos activos [{MoO2(HC(3,5-Me2pz)3)}2(μ-O)](BF4)2, [Mo2O3(O2)2(μ-O){HC(3,5-Me2pz)3}] e [MoO3{HC(3,5-Me2pz)3}]; quando dissolvido num LI, o catalisador foi reciclado com sucesso. A presença de água e o meio oxidante influenciaram a formação destas espécies. Os complexos [CpMo(CO)3Me] (Cap.5) e [CpMo(CO)2(η3- C3H5)] (Cap.6) originaram espécies activas similares (baseado nos testes catalíticos e nos espectros FT-IR ATR dos sólidos recuperados). Para [Cp'Mo(CO)2(η3-C3H5)], a influência do Cp' na actividade catalítica sugeriu a formação de espécies activas com este ligando. A partir dos complexos [Mo(CO)4L] formaram-se in situ catalisadores estáveis, que podem ser heterogéneos: para L=2-[3(5)-pirazolil]piridina formou-se [Mo4O12L4]; para L=[3- (2-piridil)-1-pirazolil]acetato de etilo formou-se [Mo8O24L4] (Cap.7). O uso de microondas (MO) como método de aquecimento em vez de um banho de óleo (BO) resultou no aumento da velocidade da reacção catalítica, devido ao aquecimento mais rápido da mistura reaccional (Caps. 5 e 7). A utilização da solução aquosa de tBHP em vez de tBHPdec era preferível, porque excluía o decano do sistema reaccional e mantinham-se elevados os rendimentos em epóxido (Caps. 2 e 6); optimizou-se o desempenho catalítico removendo a água das misturas reaccionais (Caps. 4 e 7). O melhor resultado para a epoxidação de limoneno foi observado para [CpMoCO3Me]: 88% de rendimento em epóxido (2 h, 55 ºC, método de aquecimento MO).

Os lenhosulfonatos representam um sub-produto formado durante o cozimento ao sulfito ácido, sendo queimados para a regeneração da base e recuperação de energia. No entanto, os lenhosulfonatos são também considerados uma importante matéria-prima para a produção de vários produtos de valor acrescentado. Os objectivos principais deste trabalho foram contribuir para uma melhor compreensão sobre a caracterização química e estrutural dos lenhosulfonatos do Eucalyptus globulus, assim como, para complementar a informação disponível sobre a síntese e a caracterização estrutural e térmica de materiais poliméricos obtidos a partir de compostos modelo dos produtos de oxidação dos lenhosulfonatos. O licor de cozimento ao sulfito foi analisado em termos do teor de cinzas, extractáveis, compostos voláteis, açúcares e lenhosulfonatos. O teor de cinzas e açúcares no licor de cozimento é muito elevado, tendo sido necessário purificar o mesmo (2,8-13,8 % e 3,2-9,1 %, respectivamente). A análise dos açúcares mostrou uma quantidade considerável de pentoses, sendo o açúcar predominante a xilose. Os lenhosulfonatos foram purificados, isolados e caracterizados por química molhada (titulação potenciométrica e oxidação com permanganato), análise elementar, espectroscopia de ultravioleta/visível (UV/Vis), espectroscopia de infravermelho de transformada de Fourier (FTIR), espectroscopia de ressonância magnética nuclear de protão (RMN de 1H) e carbono (RMN de 13C), espectrometria de massa de ionização por electrospray (ESI-MS), cromatografia de permeação em gel (GPC), termogavimetria (TGA) e calorimetria diferencial de varrimento (DSC). Os lenhosulfonatos são constituídos principalmente por unidades S, são parcialmente sulfonados e possuem um peso molecular relativamente baixo (Mw = 1250-2400 Da). A ruptura das ligações β-O-4 e α-O-4 da lenhina do Eucalyptus globulus após cozimento ao sulfito ácido originam olígomeros de baixo peso molecular cuja estrutura foi elucidada por RMN 1D/2D e ESI-MS. A degradação térmica dos lenhosulfonatos apresentou dois máximos de degradação a 188-190ºC e a 315-380ºC. As curvas de DSC mostraram um pico endotérmico para temperaturas inferiores a 130ºC e um pico exotérmico a 300-500ºC. Os lenhosulfonatos foram despolimerizados na presença de oxigénio molecular em meio alcalino. Os produtos de oxidação principais foram o aldeído siríngico, a vanilina, o ácido vanílico e o ácido siríngico. A adição do catalisador (sal de cobre) promoveu a oxidação dos lenhosulfonatos aumentando o rendimento dos aldeídos aromáticos (< 50%). A presença de açúcares nos lenhosulfonatos teve um efeito negativo no rendimento dos produtos de oxidação principais. Alguns compostos modelo dos produtos de oxidação dos lenhosulfonatos foram polimerizados por poliadição (catiónica e radicalar) e policondensação. Os monómeros e os polímeros foram caracterizados por espectroscopia de infravermelho de transformada de Fourier e reflectância total atenuada (FTIR-ATR), RMN em solução e no estado sólido, UV/Vis no estado sólido, GPC, difracção de raios-X (XRD), TGA e DSC. Os compostos modelo estudados foram os estirenos metoxi-substituídos (p-metoxiestireno e 3,4-dimetoxiestireno) e os ácidos hidroxi aromáticos metoxi-substituídos (ácido vanílico e ácido siríngico). O 3,4-dimetoxiestireno foi ainda copolimerizado com o éter isobutil vinílico e os seus copolímeros foram desmetilados, assim como, o poli(p-metoxiestireno) e o poli(3,4-dimetoxiestireno). A polimerização catiónica do p-metoxiestireno e 3,4-dimetoxiestireno é mais rápida e mais completa do que a polimerização radicalar produzindo polímeros com pesos moleculares elevados. O poli(p-metoxiestireno) (Mw = 235000 Da) possui um peso molecular maior do que o poli(3,4-dimetoxiestireno) (Mw = 18800 Da). A estabilidade térmica e a temperatura de transição vítrea diminuiram com a presença do segundo grupo metoxilo. A desmetilação dos homopolímeros foi bem sucedida, tendo sido corroborada por FTIR-ATR e RMN. A policondensação do ácido siríngico foi dificultada pela presença do segundo grupo metoxilo, tendo sido necessário adicionar uma maior quantidade do agente de condensação devido a factores estéricos. O poli(ácido vanílico) e poli(ácido siríngico) são insolúveis na maior parte dos solventes orgânicos, sendo parcialmente solúveis em clorofórmio, ácido triflúoracético, 1,1,2,2- tetracloroetano, dimetilsulfóxido, tetrahidrofurano, N,N’-dimetilformamida e 1,1,1,3,3,3-hexaflúor-2-propanol. A estabilidade térmica diminuiu com a presença do segundo grupo metoxilo e os dois polímeros não exibiram temperatura de transição vítrea. O poli(ácido vanílico) e poli(ácido siríngico) apresentaram uma estrutura muito cristalina (grau de cristalinidade 70% e 50%, respectivamente). O segundo grupo metoxilo aumentou o valor da absorvância, mas a forma do espectro de UV/Vis foi similar. A polimerização catiónica do éter isobutil vinílico resultou na produção de um polímero muito viscoso com peso molecular elevado (Mw = 20400 Da). A degradação térmica do polímero ocorreu em várias gamas de temperatura e foi completa (0% de resíduo a 800ºC). A copolimerização catiónica do 3,4-dimetoxiestireno com o éter isobutil vinílico foi realizada com proporções diferentes 80:20, 50:50 e 20:80. Os copolímeros apresentaram uma viscosidade elevada e um peso molecular baixo (Mw = 2000-4000 Da) que aumentou com a quantidade de éter isobutil vinílico. A degradação térmica dos copolímeros ocorreu também em várias gamas de temperatura, sendo a sua degradação completa (0,9-1,5% de resíduo a 800ºC). A desmetilação dos copolímeros não foi bem sucedida, tendo sido confirmada por FTIR-ATR e RMN.
