Background: Cryptic species complexes are common among anophelines. Previous phylogenetic analysis based on the complete mtDNA COI gene sequences detected paraphyly in the Neotropical malaria vector Anopheles marajoara. The ""Folmer region"" detects a single taxon using a 3% divergence threshold. Methods: To test the paraphyletic hypothesis and examine the utility of the Folmer region, genealogical trees based on a concatenated (white + 3' COI sequences) dataset and pairwise differentiation of COI fragments were examined. The population structure and demographic history were based on partial COI sequences for 294 individuals from 14 localities in Amazonian Brazil. 109 individuals from 12 localities were sequenced for the nDNA white gene, and 57 individuals from 11 localities were sequenced for the ribosomal DNA (rDNA) internal transcribed spacer 2 (ITS2). Results: Distinct A. marajoara lineages were detected by combined genealogical analysis and were also supported among COI haplotypes using a median joining network and AMOVA, with time since divergence during the Pleistocene (< 100,000 ya). COI sequences at the 3' end were more variable, demonstrating significant pairwise differentiation (3.82%) compared to the more moderate 2.92% detected by the Folmer region. Lineage 1 was present in all localities, whereas lineage 2 was restricted mainly to the west. Mismatch distributions for both lineages were bimodal, likely due to multiple colonization events and spatial expansion (similar to 798 - 81,045 ya). There appears to be gene flow within, not between lineages, and a partial barrier was detected near Rio Jari in Amapa state, separating western and eastern populations. In contrast, both nDNA data sets (white gene sequences with or without the retention of the 4th intron, and ITS2 sequences and length) detected a single A. marajoara lineage. Conclusions: Strong support for combined data with significant differentiation detected in the COI and absent in the nDNA suggest that the divergence is recent, and detectable only by the faster evolving mtDNA. A within subgenus threshold of >2% may be more appropriate among sister taxa in cryptic anopheline complexes than the standard 3%. Differences in demographic history and climatic changes may have contributed to mtDNA lineage divergence in A. marajoara.

This paper presents results of research into the use of the Bellman-Zadeh approach to decision making in a fuzzy environment for solving multicriteria power engineering problems. The application of the approach conforms to the principle of guaranteed result and provides constructive lines in computationally effective obtaining harmonious solutions on the basis of solving associated maxmin problems. The presented results are universally applicable and are already being used to solve diverse classes of power engineering problems. It is illustrated by considering problems of power and energy shortage allocation, power system operation, optimization of network configuration in distribution systems, and energetically effective voltage control in distribution systems. The present paper proposes a flexible consensus scheme for group decision making, which allows one to obtain a consistent collective opinion, from information provided by each expert in terms of multigranular fuzzy estimates. It is based on a linguistic hierarchical model with multigranular sets of linguistic terms, and the choice of the most suitable set is a prerogative of each expert. From the human viewpoint, using such model is advantageous, since it permits each expert to utilize linguistic terms that reflect more adequately the level of uncertainty intrinsic to his evaluation. From the operational viewpoint, the advantage of using such model lies in the fact that it allows one to express the linguistic information in a unique domain, without losses of information, during the discussion process. The proposed consensus scheme supposes that the moderator can interfere in the discussion process in different ways. The intervention can be a request to any expert to update his opinion or can be the adjustment of the weight of each expert`s opinion. An optimal adjustment can be achieved through the execution of an optimization procedure that searches for the weights that maximize a corresponding soft consensus index. In order to demonstrate the usefulness of the presented consensus scheme, a technique for multicriteria analysis, based on fuzzy preference relation modeling, is utilized for solving a hypothetical enterprise strategy planning problem, generated with the use of the Balanced Scorecard methodology. This paper presents results of research related to multicriteria decision making under information uncertainty. The Bell-man-Zadeh approach to decision making in a fuzzy environment is utilized for analyzing multicriteria optimization models (< X, M > models) under deterministic information. Its application conforms to the principle of guaranteed result and provides constructive lines in obtaining harmonious solutions on the basis of analyzing associated maxmin problems. This circumstance permits one to generalize the classic approach to considering the uncertainty of quantitative information (based on constructing and analyzing payoff matrices reflecting effects which can be obtained for different combinations of solution alternatives and the so-called states of nature) in monocriteria decision making to multicriteria problems. Considering that the uncertainty of information can produce considerable decision uncertainty regions, the resolving capacity of this generalization does not always permit one to obtain unique solutions. Taking this into account, a proposed general scheme of multicriteria decision making under information uncertainty also includes the construction and analysis of the so-called < X, R > models (which contain fuzzy preference relations as criteria of optimality) as a means for the subsequent contraction of the decision uncertainty regions. Hepatitis C virus (HCV) is a major cause of hepatic disease and of liver transplantation worldwide. Mannan-binding lectin (MBL), encoded by the MBL2 gene, can have an important role as an opsonin and complement activating molecule in HCV persistence and liver injury. We assessed the MBL2 polymorphism in 102 Euro-Brazilian patients with moderate and severe chronic hepatitis C, paired for gender and age with 102 HCV seronegative healthy individuals. Six common single nucleotide polymorphisms in the MBL2 gene, three in the promoter (H/L, X/Y and P/Q) and three in exon 1 (A, the wild-type, and B, C or D also known as O) were evaluated using real-time polymerase chain reaction with fluorescent hybridization probes. The concentration of MBL in plasma was measured by enzyme-linked immunosorbent assay. The frequency of the YA/YO genotype was significantly higher in the HCV patients compared with the controls (P = 0.022). On the other hand, the genotypes associated with low levels of MBL (XA/XA, XA/YO and YO/YO) were decreased significantly in the patients with severe fibrosis (stage F4), when compared with the patients with moderate fibrosis (stage F2) (P = 0.04) and to the control group (P = 0.011). Furthermore, MBL2 genotypes containing X or O mutations were found to be associated with non-responsiveness to pginterferon and ribavirin treatment (P = 0.023). MBL2 polymorphisms may therefore be associated not only with the development of chronic hepatitis C, but also with its clinical evolution and response to treatment.

In previous studies, we determined that beta 1 integrins from human colon tumors have elevated levels of alpha 2-6 sialylation, a modification added by beta-galactosamide alpha-2,6-sialyltranferase I (ST6Gal-I). Intriguingly, the beta 1 integrin is thought to be a ligand for galectin-3 (gal-3), a tumor-associated lectin. The effects of gal-3 are complex; intracellular forms typically protect cells against apoptosis through carbohydrate-independent mechanisms, whereas secreted forms bind to cell surface oligosaccharides and induce apoptosis. In the current study, we tested whether alpha 2-6 sialylation of the beta 1 integrin modulates binding to extracellular gal-3. Herein we report that SW48 colonocytes lacking alpha 2-6 sialylation exhibit beta 1 integrin-dependent binding to gal-3-coated tissue culture plates; however, binding is attenuated upon forced expression of ST6Gal-I. Removal of alpha 2-6 sialic acids from ST6Gal-I expressors by neuraminidase treatment restores gal-3 binding. Additionally, using a blot overlay approach, we determined that gal-3 binds directly and preferentially to unsialylated, as compared with alpha 2-6-sialylated, beta 1 integrins. To understand the physiologic consequences of gal-3 binding, cells were treated with gal-3 and monitored for apoptosis. Galectin-3 was found to induce apoptosis in parental SW48 colonocytes ( unsialylated), whereas ST6Gal-I expressors were protected. Importantly, gal-3-induced apoptosis was inhibited by function blocking antibodies against the beta 1 subunit, suggesting that beta 1 integrins are critical transducers of gal-3-mediated effects on cell survival. Collectively, our results suggest that the coordinate up-regulation of gal-3 and ST6Gal-I, a feature that is characteristic of colon carcinoma, may confer tumor cells with a selective advantage by providing a mechanism for blockade of the pro-apoptotic effects of secreted gal-3.

The composition of the Pyrgulidae and its relationships to other member families of the caenogastropod superfamily Rissooidea are examined after a consideration of new anatomical (including gross anatomy, sperm ultrastructure), conchological (including protoconch features), ecological, biogeographical and palaeontological data and a re-evaluation of existing literature. Pyrgulidae can be distinguished from hydrobiids unequivocally only with the aid of the radula. Sperm ultrastructural features suggest a very close relationship between the Pyrgulidae, the Hydrobiidae and the Bithyniidae (in fact no family-diagnostic sperm characters can be found to separate these three taxa). Based upon neontological and fossil evidence it is likely that pyrgulids represent a Miocene offshoot from a paratethyal hydrobiid lineage. Pyrgulids may also represent the stock from which the baicaliids arose, in which case the Pyrgulidae must be considered a paraphyletic group. The huge biogeographic gap between the Caspian Sea and Lake Baikal is to some extent bridged by the finding of a Neogene pyrgulid from the Altai Mountains. An alternative scenario for the origin of baicaliids is presented.

Se nos estados federais os mecanismos de coopera????o e coordena????o intergovernamental j?? s??o relevantes, no caso brasileiro eles ganham centralidade ainda maior. A Constitui????o Federal de 1988 agregou complexidade ao desenho federativo brasileiro, reconhecendo o munic??pio como ente federado. Esse processo foi acompanhado por intensa descentraliza????o de pol??ticas p??blicas, pelo fortalecimento do poder local e por mecanismos pouco coordenados de rela????o vertical e horizontal entre os entes federativos. Ao mesmo tempo, a aus??ncia de pol??ticas de desenvolvimento regional acentuou as desigualdades locais e regionais observadas historicamente no pa??s. Diferentes experi??ncias de consorciamento foram levadas a cabo por munic??pios no pa??s e ainda hoje esse ?? um instrumento de larga utiliza????o. De forma diversa, as iniciativas de coopera????o entre estados s??o ainda incipientes. As limita????es institucionais e jur??dicas dos desenhos utilizados pelos munic??pios, no entanto, levaram ?? aprova????o da Emenda Constitucional n?? 19/98, que instituiu os cons??rcios p??blicos e a gest??o associada de servi??os p??blicos.

Este trabalho faz uma breve revis??o sobre as diferentes maneiras de se estruturar uma iniciativa or??ament??ria de g??nero, seja ela elaborada pelo governo ou pela sociedade civil, em n??vel local ou nacional, considerando todo o or??amento ou setores selecionados e dependendo do est??gio do ciclo or??ament??rio em que ela ?? realizada. Este estudo delineia uma estrutura para a an??lise de g??nero de or??amentos, relacionando os insumos ??s atividades, aos resultados e aos impactos e, a partir da??, apresenta alguns exemplos de an??lises de or??amento a partir da perspectiva de g??nero, em diferentes momentos do ciclo or??ament??rio, buscando experi??ncias em diferentes pa??ses. Tamb??m destaca dois princ??pios chave na an??lise do or??amento com enfoque no g??nero, a saber: 1) a avalia????o do impacto do or??amento tanto sobre o indiv??duo como sobre a fam??lia; e 2) o reconhecimento da contribui????o econ??mica do trabalho dom??stico n??o remunerado, a cargo basicamente das mulheres.

Por outro lado, a incorpora????o de t??cnicas de gest??o avan??adas ?? considerada indispens??vel para o aumento da efic??cia e da efici??ncia no setor p??blico. Os principais problemas discutidos no artigo incluem: racionalidade do setor p??blico racionalidade das empresas privadas; incentivos e motiva????o dos servidores p??blicos; rigidez dos procedimentos administrativos; margem de decis??o dos gestores; sistemas de acesso a informa????o; import??ncia da gest??o de pessoal e da valoriza????o do trabalho no setor p??blico; papel dos gerentes na moderniza????o das institui????es p??blicas; cargos de carreira, cargos de confian??a; mecanismos de recrutamento e reten????o e sistemas de capacita????o dos servidores p??blicos. Os autores concluem que, devido ??s inova????es constantes em mat??ria de ger??ncia p??blica, n??o existe hoje um perfil gerencial definitivo como resposta ??s necessidades do Estado. Nos ??ltimos anos, o debate sobre a melhoria da qualidade do gasto p??blico tem ganhado espa??o crescente. As quest??es mais gerais do tema s??o relevantes, mas n??o substituem o exame espec??fico dos gastos sociais dos diversos minist??rios ??? raz??o pela qual a avalia????o das pol??ticas p??blicas ?? instrumento fundamental para melhoria do gasto. O trabalho corrente faz uma avalia????o da din??mica or??ament??ria das pol??ticas do Minist??rio do Trabalho e Emprego. Essa an??lise revela um crescente desequil??brio: enquanto o valor destinado ??s pol??ticas passivas de emprego aumentou brutalmente, os valores destinados ??s pol??ticas ativas de emprego ca??ram significativamente. Esse cen??rio ??? oposto ao que seria desej??vel ??? ?? ainda mais preocupante na medida em que as pol??ticas passivas de emprego n??o beneficiam os mais pobres. Neste contexto, o trabalho avalia a possibilidade de que parte dos recursos reservados para pol??ticas passivas seja dirigido ??s pol??ticas ativas de emprego.

No contexto da reforma do Estado brasileiro, a descentraliza????o das compet??ncias pol??ticas e administrativas na arena ambiental tem se mostrado um processo din??mico. Muitas inst??ncias locais j?? respondem pela quest??o ambiental. No entanto, isso n??o significa mais compet??ncia administrativa, sustentabilidade institucional, tampouco participa????o democr??tica. Dois casos de pol??tica ambiental s??o retratados no texto: o licenciamento industrial pelos munic??pios brasileiros e o a gest??o florestal pelo Estado do Mato Grosso. A descentraliza????o do licenciamento ambiental para o ??mbito municipal ainda parece fr??gil em munic??pios menores, em um processo que parece ser induzido pelo Estado e n??o pelo controle social local. Em rela????o ?? pol??tica florestal, o caso do Mato Grosso ?? emblem??tico. Ele mostra que a coopera????o com o poder p??blico federal e, em determinados momentos, sua coordena????o s??o de suma import??ncia para que as pol??ticas p??blicas ambientais n??o fiquem ?? merc?? de governos estaduais, que podem envies??-las em favor do ???desenvolvimentismo???, nem percam a legitimidade adiante da fragilidade financeira e institucional dos ??rg??os ambientais locais.
