BACKGROUND: Effects of 17beta-estradiol and progesterone on rhesus monkey oocyte maturation in vitro were evaluated by embryo development subsequent to IVF. METHODS AND RESULTS: In experiment 1, immature cumulus-oocyte complexes collected from unstimulated adult females during the non-breeding season were matured in modified medium CMRL-1066 containing various combinations of gonadotrophins (FSH + LH), estradiol and/or progesterone. Formation of morulae and blastocysts was greatest in oocytes matured in medium containing estradiol and/or progesterone, with or without gonadotrophins (morula 38-46%, blastocyst 14-20%) than in control oocytes matured without estradiol or progesterone (morula 14%, blastocyst 0%). In experiment 2, cumulus-oocyte complexes from unstimulated prepubertal female monkeys were matured in medium with gonadotrophins, estradiol or progesterone. The best development to the morula stage was obtained with oocytes matured with gonadotrophins and estradiol or gonadotrophins and progesterone (43 and 25 morulae, respectively), while control oocytes matured with gonadotrophins but without steroid hormones gave the poorest morula developmental response (12%). However, there was no difference in blastocyst development across all groups (0-3%). CONCLUSIONS: These results demonstrate that during rhesus monkey oocyte maturation in vitro: (i) estradiol or progesterone can improve oocyte developmental competence; (ii) immature oocytes from prepubertal versus adult females have differential responses to challenge with estradiol or progesterone.

The hemizona assay (HZA) in Rhesus monkeys was employed to study the correlation of zona-binding ability with sperm motility or with naturally developing oocytes at various maturational stages. Oocytes from unstimulated ovaries were retrieved within 2 hr from monkeys sacrificed for vaccine production (in reproductive season, but with their menstrual cycles not determined). Oocytes were divided into four groups based on their morphological maturation: 1) Oocytes surrounded by more than one cumulus layer (MC); 2) Oocytes retaining intact germinal vesicle nuclei (GV); 3) Oocytes with germinal vesicle breakdown showing distinct perivitelline space (PVS); and 4) Oocytes extruding the first polar body (PB1). The mean numbers of sperm bound to hemizona for PBI, PVS, GV, and MC groups were 132.9 +/- 12.0, 71.5 +/- 10.1, 36.1 +/- 4.0, and 20.1 +/- 2.9 (Mean +/- SE), respectively. The four groups showed significant differences from each other in sperm/egg binding ability (P < 0.01). The number of bound sperm significantly increased with oocyte maturation. The present study also showed that zona-binding ability was also affected by sperm motility. For sperm with 67.7% motility and sperm with 31.2% motility, the average numbers of bound sperm were 43.5 +/- 2.2 and 25.3 +/- 2.9 (Mean +/- SE), respectively. There was significantly higher binding ability for sperm with higher motility (P < 0.01). The results suggest that: 1)The rhesus monkey model can serve as a very sensitive model for studying sperm/egg interaction by HZA; 2) Sperm motility positively correlated with sperm/egg binding; and 3) Sperm/egg binding ability increases with oocyte maturation. The binding ability is highest when oocytes matured to the PB1 stage, which is also the best opportunity for fertilization. This is strong evidence for the ''zona maturation'' hypothesis. The objective of this study was to provide a simple cryopreservation method for oocytes from Yunnan Yellow Cattle and facilitate preservation efforts in this native Chinese breed, which is threatened by agricultural modernization. Cumulus-oocyte complexes (COCs) were collected from slaughterhouse ovaries and matured in vitro for 22-24 h, then selected for cryopreservation. Vitrification in open pulled straws (OPS) or in microdrops on a cooled metal surface (solid surface vitrification, SSV) was compared. The OPS vitrification solution consisted of 20% ethylene glycol (EG) and 20% DMSO. The SSV solution was a mixture of 35% EG, 5% polyvinyl-pyrrolidon (PVP) and 0.4 M trehalose. Vitrified and warmed oocytes were either fertilized in vitro or parthenogenetically activated. The rates of cleavage and development to blastocysts of fertilized oocytes following OPS versus SSV were not statistically different (38.3 and 12.5% versus 35.8 and 6.0%, respectively). The corresponding rates of parthenogenetic development to blastocysts were also not different (8.2 versus 3.5%, respectively). Development to blastocysts of non-vitrified controls following fertilization was significantly higher than that of the vitrified oocytes (22.6%, P < 0.05). These results demonstrate for the first time, that although both OPS and SSV procedures reduced embryonic development, Yunnan Yellow Cattle oocytes are capable of developing to blastocysts following cryopreservation. This study evaluated the effects of different amino acid formulations on supporting meiotic and cytoplasmic maturation of rhesus monkey (Macacca mulatta) oocytes in vitro. Five hundred and forty-six cumulus-oocyte complexes (COCs) aspirated from unstimulated adult monkey follicles (greater than or equal to 1000 mum in diameter) were cultured in either modified Connaught Medical Research Laboratories 1066 medium (mCMRL-1066) or in one of eight chemically defined media (modified basic medium 5 supplemented with 5.5 mmol glucose l(-1), 0.003 mmol pantothenic acid l(-1) and different amino acid formulations) as below: (1) modified basic medium 5 (mBM5) containing no amino acid; (2) mBM5 + 0.2 mmol glutamine l(-1); (3) mBM5 + 11 amino acids from hamster embryo culture medium 6 (HECM-6) (11 AA); (4) mBM5 + Eagle's non-essential amino acids (NEA); (5) mBM5 + NEA + 0.2 mmol glutamine l(-1); (6) mBM5 + Eagle's essential amino acids (EA) without glutamine; (7) mBM5 + EA + 0.2 mmol glutamine l(-1); (8) mBM5 + Eagle's 20 amino acids (20 AA) + 0.2 mmol glutamine l(-1); and (9) mCMRL-1066 (control). All media contained FSH, LH, oestradiol and progesterone. After maturation, mature oocytes were subjected to the same fertilization and embryo culture procedures. COCs matured in treatment 5 had greater potential to progress to metaphase II (66%; P < 0.05) than did those in treatments 1 (37.3%), 2 (48.3%)f 3 (41%), 6 (41%) and 9 (43%). Oocytes matured in treatment 8 had the best morula (53%) and blastocyst (18%) developmental responses (P<0.05). The lowest (P<0.05) morula and blastocyst developmental responses were obtained from COCs matured in treatments 1 (0%) and 6 (8%). The other media supported intermediate embryonic development (range 11-38% of morula and blastocyst). These results indicate that the choice of amino acids affects the competence of oocyte maturation and that Eagle's 20 AA with 0.2 mmol glutamine l(-1) is more efficient than the other amino acid formulations for maturation of rhesus monkey oocytes.

The present paper deals on the histological description of the hake ovary made on the basis of gonad observations of 394 females during the period April 1966 March 1967. The material was obtained from weekly sampling of commercial catches carried out at the Institute of Marine Biology (Prov. Buenos Aires, Argentina). The anatomical and histological description of the standard ovary and the adopted terminology are given. The maturation process is divided into five periods, from ovocyte formation to yolked ovocyte formation, with its histological description. Ovary changes are analyzed on detail. The following conclusions were outlined: 1. Analysis demonstrated that although some specimens were totally spawned others, at the end of the spawning period, retaining a great number of ovocytes in different maturity stages. Therefore, postspawners have been classified as follows: Postspawned II : This stages is characterized by the empty ovarian structure, with ovocytes in stage II, which will remain in the resting phase untill next spawning season. Postspawned III and IV: Their main characteristics are: tissue destruction, bloody residuals and remaining ovocytes in stage III and IV, respectively. 2. Some transformations were found in ovaries of postspawned III and IV. They are classified as follows, according to its origin and structure: Developed from follicular cell membrane – a) Glandular formations, b) Epiteloid formations - Originated from remaining ovocytes, c) Ovocyte disintegration, d) Ovocyte with follicular cell infiltration. 3. All those structures derived from postpawners III and IV have a temporary character and will be reabsorbed. Their presence delay the recuperation of the organ and its reproductive functions. Consequently, the possibility of those structures acting as control mechanisms is suggested. 4. Transformations pointed out in paragraph Nº 2 prevent the possibility of consecutive spawning originated from the remaining ovocytes (II and IV). 5. No structures originated from postspawners III and IV were found during summer season. 6. Reproductive cycle of hake has been described monthly. It was observed that maturing ovaries predominate in summer (November-December).

The mafic-ultramafic layered intrusions in the Panxi, China contain large V-Ti-magnetite deposits. These layered intrusions are related with the Emeishan continental flood basalts in space and time. Two layered intrusions, Hongge and Xinjie have clear PGE mineralization at the base of the intrusions. Thus the detailed investigations of these two intrusions not only have a geological but also have an economic significance. This thesis aims to characterize the elemental and Sr-Nd isotopic features of diverse rock zones within the intrusion on the basis of systematic studies of the major, trace element and isotope ratios, therefore to constrain the petrogenesis, mantle source and evolution of the Hongge and Xinjie intrusions. Generally, both Hongge and Xinjie intrusions show the same Fe-Ti-rich and Si-M-poor characteristics. They are also enriched in rare-earth elements (REE) and large-ion lithophile elements (LILE) as well as in Sr-Nd isotope ratios (Hongge: initial Sr = 0.7056-0.7076, ε_(Nd)(t) and (Nd/Sm)_N-ε_(Nd)(t) plots, the Hongge intrusion has a similar elemental and isotopic features to the Emeishan low-Ti (LT) basalts, whereas the Xinjie intrusion was close to the Emeishan high-Ti (HT) basalt. Therefore, the Hongge intrusion may be co-genetic with the LT basalt, formed by the partial melting of the spinel-garnet transition mantle that had a slight enriched isotope character. In contrast, the Xinjie intrusion and the HT basalts are probably derived from the garnet-phases mantle with a primitive isotope character. The involvement of the components of mantle wedge into the source is considered to be the major reason of the REE and LILE enrichment and Nd isotope depletion in the Xinjie intrusion. In contrast with the systematic variations in TiO_2 content, Mg#, transition elements (Ni, Cu, Co), REE concentrations, and La/Yb, La/Sm ratios from the lower zone to upper zone, the different rock zones of the Hongge intrusion have no clear Sr-Nd isotope variations. This suggests that the Hongge intrusions were formed by the crystal fractionation from the same magma source. The rhythm may be formed by slow injection of the co-genetic magma during the crystal fractionation. The increase in K_2O and Al_2O_3 contents, REE abundance, and the degree of the REE fractionation in the base of the intrusion, together with the relatively low ε_(Nd)(t) value, may imply that the base of the Hongge intrusion was contaminated with the local crust rocks. Xinjie intrusion shows the clearly elemental and isotopic differences in diverse cumulus cycles. The observation of the systematic variations in TiO_2 content, Mg# value, transition elements (Ni, Cu, Co), REE concentrations, and La/Yb, La/Sm ratios in first cycle was not occurred in second cumulus cycle. In addition, the ε_(Nd)(t) value in second cumulus cycle is apparently higher than that of the first one. Thus the abruptly elemental and isotopic changes at the base of second cycle demonstrate that there is considerable new and depleted magma addition to the residue magma after the crystallization of the first cycle. These features are very similar to those of the well-known PGE-rich Bushveld and Stillwater layered intrusions. The PGE mineralization in Xinjie intrusion is much better than in Hongge intrusion. Therefore, the layered intrusion similar to the Xinjie in Panxi area posses the better prospects for the PGE deposits.

‘Work on the move’ is a design, process-driven methodology, which uses multiple locations within an outdoors setting and movement between locations, all of which function as learning places, confined to a specified time period. Between 2012 and 2015, a team of international Higher Education product design educators (all members of Carousel, a co-operation of Erasmus members in Zwolle, Edinburgh, Nantes, Rome, Kortrijk and Oslo), industry professionals and product design students developed and tested four case studies. Each case study was conducted in a different international location and was constructed with a different focus, to help define and refine a definitive working methodology. ‘Work on the move’ explores the influence of ‘place’ upon design, in terms of the impact it has on productivity and creative problem-solving, when working away from the traditional studio/office-based environment. It also explores the significance of shared place, when working directly with a client in situ, and experiencing the place-based influences upon their businesses. While identifying location as part of the design process, the study also seeks to understand the effects of time restriction and working in transit upon creativity and productivity, within the context of specific projects.

L'article présente quelques éléments de la procédure mise en place pour traiter un corpus écrit comportant 617 textes (près de 500 000 mots) relatifs aux eurorégions. Complexe et hétérogène à plusieurs titres (technique, linguistique, éditorial, générique, énonciatif), le corpus pose la difficulté majeure de l’appréhension de données multilingues (français, italien, espagnol, anglais, allemand, néerlandais). Sa manipulation a nécessité une réflexion adaptée et une démarche de modélisation que nous qualifions d’« agile » en raison de son caractère souple et itératif. La plateforme d’analyse élaborée permet de disposer de résultats utiles à l’analyse qualitative ultérieure du discours eurorégional. Elle articule un logiciel d'analyse morphosyntaxique éprouvé (TreeTagger) à des programmes (Perl) et à une base de données (SQLite) développés pour optimiser les requêtes multilingues simultanées et l’exportation automatique des résultats. Les fonctionnalités liées à la localisation contextualisée de mots- pivots, au recueil de dénominations et à la détection de segments répétés nous servent ici de guides pour exprimer les besoins de la recherche, les problèmes rencontrés et les solutions proposées. L'analyse d'observables récurrents, à savoir les notions de décision et de responsabilité, illustre le propos.
