EXTRACT (SEE PDF FOR FULL ABSTRACT): General circulation models (GCMs) are probably the most sophisticated theoretical tools we have to simulate possible climatic effects of increasing carbon dioxide and other greenhouse gases. ... As will be illustrated here using a variety of examples, although the models do simulate "reality" very well on the "grand" scale (e.g., global, hemispheric, zonal), substantial differences are more apparent as the scale is reduced to areas particularly relevant to regional planners. It is particularly important that workers more clearly recognize the potential dangers in relying too heavily on simple summary statistics such as averages estimated over large regional scales. Many shortcomings are apparent in the model simulations of the present climate, indicating that further model improvements are needed to achieve reliable regional and seasonal projections of the future climatic conditions.

Twenty five different species belonging to 16 genera of freshwater fishes were analysed for protein, fat, moisture, ash, carbohydrate, phosphorus, calcium and total iron content in their muscle. Calorific value for protein, fat and carbohydrate fractions and total calories for each species were also calculated.

一种新型的2,4-二取代氨基-6-取代-[1,3,5]三嗪或1,3-嘧啶衍生物及其制备方法、药物组合物和其药理用途,其结构通式如式(I)所示,其中R1、R2、R3、R4、R5、R6、A、B、X、Y和Z的定义如说明书中所述。该类化合物与HIV-1整合酶具有很高结合活性,并且在底物竞争测试中能够有效的抑制整合酶对底物的结合。因此该类化合物是较强的HIV-1整合酶抑制剂,有望开发成为新的抗HIV病毒药物。

To search for compounds with superior anti-human immunodeficiency virus type 1 (HIV-1) activity, ten 5,5'-(p-phenylenebisazo)-8-hydroxyquinoline sulfonates (4a-j) were synthesized and preliminarily evaluated as HIV-1 inhibitors in vitro for the first time. Some compounds demonstrated anti-HIV-1 activity, especially 5,5'-(p-phenylenebisazo)-8-hydroxyquinoline p-ethylbenzenesulfonate (4g) and 5,5'-(p-phenylenebisazo)-8-hydroxyquinoline p-chlorobenzenesulfonate (41) showed the more potent anti-HIV-1 activity with 50% effective concentration (EC50) values of 2.59 and 4.01 mu g/ml, and therapeutic index (TI) values of 31.77 and 24.51, respectively.

Chronic exposure to opiates impairs hippocampal long-term potentiation (LTP) and spatial memory, but the underlying mechanisms remain to be elucidated. Given the well known effects of adenosine, an important neuromodulator, on hippocampal neuronal excitability and synaptic plasticity, we investigated the potential effect of changes in adenosine concentrations on chronic morphine treatment-induced impairment of hippocampal CA1 LTP and spatial memory. We found that chronic treatment in mice with either increasing doses (20-100 mg/kg) of morphine for 7 d or equal daily dose (20 mg/kg) of morphine for 12 d led to a significant increase of hippocampal extracellular adenosine concentrations. Importantly, we found that accumulated adenosine contributed to the inhibition of the hippocampal CA1 LTP and impairment of spatial memory retrieval measured in the Morris water maze. Adenosine A(1) receptor antagonist 8-cyclopentyl-1,3-dipropylxanthine significantly reversed chronic morphine-induced impairment of hippocampal CA1 LTP and spatial memory. Likewise, adenosine deaminase, which converts adenosine into the inactive metabolite inosine, restored impaired hippocampal CA1 LTP. We further found that adenosine accumulation was attributable to the alteration of adenosine uptake but not adenosine metabolisms. Bidirectional nucleoside transporters (ENT2) appeared to play a key role in the reduction of adenosine uptake. Changes in PKC-alpha/beta activity were correlated with the attenuation of the ENT2 function in the short-term (2 h) but not in the long-term (7 d) period after the termination of morphine treatment. This study reveals a potential mechanism by which chronic exposure to morphine leads to impairment of both hippocampal LTP and spatial memory.

根据鲫鱼类PKR蛋白激酶基因(CaPKR-like)的全长cDNA序列,克隆CaPKR-likeZα结构域cDNA(Zα1、Zα2和Zα1Zα2),原核表达成功获得3种融合蛋白PZα1、PZα2和PZα1Zα2。凝胶阻滞实验结果显示:PZα1、PZα2、PZα1和PZα2混合表达蛋白不能与聚肌胞苷酸(PolyI∶C)结合,而表达完整的Zα结构域的PZα1Zα2与PolyI∶C有明显的结合现象。另外,3种表达多肽PZα1、PZα2和PZα1Zα2在体外分别都能聚合形成二聚体。与PZα1相比,PZα2和PZα

We fabricate a saturable absorber mirror by coating a graphenefilm on an output coupler mirror. This is then used to obtain Q-switched mode-locking from a diode-pumped linear cavity channel waveguide laser inscribed in Ytterbium-doped Bismuthate Glass. The laser produces 1.06 ps pulses at ∼1039 nm, with a 1.5 GHz repetition rate, 48% slope efficiency and 202 mW average output power. This performance is due to the combination of the graphene saturable absorber and the high quality optical waveguides in the laser glass. © 2013 Optical Society of America.

TNF receptor associated factor 1 (TRAF1) plays an important role in regulating the TNF signaling and protecting cells from apoptosis. In the present study, a TRAF1 gene has been cloned from grass carp (Ctenopharyngodon idella) by reverse transcription (RT)-PCR and rapid amplification of cDNA ends (RACE). The full-length cDNA is 2235 bp, including a 250 bp 5' UTR (untranslated region), a 1659 bp open reading frame, and a 326 bp 3'UTR. The polyadenylation signal (AATAAA, AATAA) and one mRNA instability motif (AUUUA) were found followed by a poly (A) tail in the 3'UTR. No signal peptide or transmembrane region has been found in the putative amino acids of grass carp TRAF1 (gcTRAF1). The putative amino acids of gcTRAF1 share 72% identity with the homologue in zebrafish. It is characterized by a zinc finger at the N-terminus and a TRAF domain (contains one TRAF-C and one TRAF-N) at the C-terminus. The identity of the TRAF domain among all the TRAF1 homologues in vertebrates varies from 52% to 58%, while the identities of TRAF-C were almost the same as 70%. The recombinant gcTRAF1 has been constructed successfully and expressed in Escherichia coli by using pET-32a expression vector. The polyclonal antibody for rabbit has been successfully obtained. The expression of gcTRAF1 in different organs was examined by real-time quantitative PCR and Western blotting, respectively. It was widely distributed in heart, head kidney, thymus, brain, gill, liver, spleen, and trunk kidney. This is the first report of TRAF1 homologue molecule found in fish. (c) 2007 Elsevier B.V. Several studies have suggested that Otu domain had de-ubiquitinating activity and Tudor domain was important for the formation of germ cells. Here, we reported a novel zebrafish ovary-specific gene containing Otu and Tudor domain, z-otu, which was expressed at stages I-III oocytes and embryonic stages from zygotes to early blastula during embryonic cells maintained their totipotency. Therefore, z-otu might link the ubiquitin signaling pathway to early oogenesis and maintaining the totipotency of embryonic cell. (c) 2005 Elsevier B.V. All rights reserved.
