Righteousness, justice or faithfulness? The Hebrew Root ṣdq in the Psalter of the Finnish Church Bible of 1992 This study attempts to answer three questions. Firstly, what do the derivates of the root ṣdq mean in the Hebrew Psalter? Secondly, with which equivalents are these Hebrew words translated in the Psalter of the Finnish Church Bible of 1992 and why? And thirdly, how is the translation of the root ṣdq in the Psalter placed in comparison with the translations of the root ṣdq in certain ancient and modern Bible translations? The root ṣdq has a very wide semantic field in Biblical Hebrew. The basic meaning of the root ṣdq is ‘right’ or ‘to be in the right’. The traditional English equivalent of the root ṣdq is righteousness. In many European languages the equivalent of the root ṣdq has some connection with the word ‘right’, but this is not the case in Finnish. The Finnish word vanhurskaus has been present since the first Finnish Bible translation by Mikael Agricola in 1548. However, this word has nothing to do with the Finnish word for ‘right’. The word vanhurskaus has become a very specific religious and theological word in Finnish, and it can be a word that is not obvious or at all understandable even to a native Finnish speaker. In the Psalter of the earlier Finnish Church Bible of 1938 almost every derivate of the root ṣdq (132/139) was translated as vanhurskaus. In the Psalter of the Finnish Church Bible of 1992 less than half of these derivates (67/139) are translated as that. Translators have used 20 different equivalents of the Hebrew derivates of the root ṣdq. But this type of translation also has its own problems. The most disputed is the fact that in it the Bible reader finds no connections between many Bible verses that have obvious connections with each other in the Hebrew Bible. For example, in verse Ps. 118, 15 one finds a Finnish word for ‘saved’ and in verse Ps. 142, 8 one finds another Finnish word for ‘friends’, while in the Hebrew Bible the same word is used in both verses, ṣaddîqīm. My study will prove that it is very challenging to compare or fit together the semantics of these two quite different languages. Tutkimuksessa tarkastellaan yhteiskunnallista yrittäjyyttä suomalaisesta näkökulmasta ja pyritään kartoittamaan niitä tekijöitä, jotka voivat vaikuttaa sen institutionalisaatioon Suomessa. Yhteiskunnallinen yrittäjyys on viimeisen kahden vuosikymmenen aikana kansainvälisesti huomiota saanut pyrkimys yhdistää innovatiivisella tavalla taloudellisesti kestävä yritystoiminta ja sosiaalisen muutoksen aikaansaaminen. Erityisesti yhteiskunnalliset yritykset on nähty yhtenä mahdollisena vastauksena tulevaisuudessa yhä ajankohtaisempaan kysymykseen siitä, miten tärkeinä pidetyt hyvinvointipalvelut saadaan tuotettua huoltosuhteen heikentyessä ja julkisen talouden kiristyessä. Tutkimuksen tarkoituksena on selvittää avainhenkilöhaastatteluiden avulla, miten yhteiskunnallinen yrittäjyys ymmärretään Suomessa, ja mitä mahdollisuuksia tai haasteita nähdään sen vakiintumiselle. Tutkimus muodostuu kahdesta osuudesta. Ensimmäisen osan muodostaa yhteiskunnallisen yrittäjyyden käsitteen selventäminen heijastamalla haastatteluissa nousseita käsityksiä aiempaan pääosin kansainväliseen tutkimuskirjallisuuteen. Toinen osuus on teoriasidonnainen. Aineistosta pyritään kartoittamaan niitä tekijöitä, jotka haastateltavien mukaan vaikuttavat yhteiskunnallisen yrittäjyyden institutionalisaatioon Suomessa. Tutkimuksen teoreettisen viitekehyksen muodostaa (uusi) institutionalismi ja erityisesti teoria institutionalisaatiosta. Uuden alan institutionalisaatioon vaikuttavat Lawrencen ja Phillipsin mukaan sekä makrokulttuuriset instituutiot, eli yhteiskunnan erilaiset rakenteet, että institutionaalinen yrittäjyys, eli aktiivisten yksilöiden toiminta. Olemassa olevat rakenteet voivat olla uuden alan vakiintumista mahdollistavia tai rajoittavia. Tutkimuksen aineiston valinta perustuu teoreettiseen otokseen.Tutkimukseen on haastateltu avainhenkilöitä niistä sidosryhmistä, jotka ovat keskeisessä asemassa uuden alan tai ilmiön vakiintumiseen nähden. Haastattelutekniikkana on käytetty puolistrukturoitua teemahaastattelua. Haastatteluista saatua aineistoa on analysoitu laadullisen sisällönanalyysin avulla. Yhteiskunnallisesta yrittäjyydestä ei vallinnut haastateltujen kesken yhtenäistä kuvaa. Yhteiskunnallisen tehtävän ensisijaisuus nähtiin kuitenkin keskeisenä ilmiötä määrittävänä tekijänä. Haastatteluissa ilmenevät käsitykset ovat myös melko yhdenmukaisia Iso-Britannian mallin kanssa. Keskeisinä tuloksina yhteiskunnallisen yrittäjyyden institutionalisaatioon vaikuttavina tekijöinä nousivat mahdollistavina esiin vahva kolmas sektori, vakiintunut osuustoiminta, myönteinen poliittinen linjaus, vastuullisen kansalaisuuden ja individualismin vahvistuminen sekä tiivis kansainvälinen yhteistyö. Institutionalisaatiota rajoittavina institutionaalisina tekijöinä nousivat esiin vakiintuneet (erityisesti yrityspuolen) organisaatiot, hankintalaki ja sen sovellukset, perinteinen yksipuolinen näkemys yrittäjyydestä, hallintoalamaisuuskulttuuri sekä käsitteiden sekaantuminen ja leimautuminen haihatteluksi.

In the study, two upper elementary school health education textbooks were investigated. The purpose of the study was to examine the health discourses and subject- and reader positions constructed in text. Theoretically, the study is based on poststructuralist thought and critical sociology of health promotion. Methodologically, it draws mainly on critical lingvistics and new rhetorics. Textbooks were understood as informative, argumentative and persuasive texts in which different lexical and grammatical methods to secure the readers´ responsiveness were utilized. Also, the relations of the text to wider genres, social situations, structures, institutions and practices were investigated. The interpersonal and ideational dimensions of the texts were analysed with the aim of finding out the kinds of identities for and relations between the speaker and the ideal reader were constructed and the kinds of representations of health and the world around were produced in the textbooks. Multiple discourses of health, and genres and styles characteristic for many kinds of contexts and situations were found. The identities of and the relationships between the speaker and the ideal reader of the text were also multiple and changing. The text echoes both biomedical health discourse emphasizing prevention of illness and holistic discourse emphasizing personal welfare, fulfillment and happiness. Furthermore, traces for example from development psychological, ecological and civilization critical discourses were perceived. Formal scientific genre was found to be mixed with informal chatting imitating close and equal relationship between participants characteristic for advertisements and other persuasive texts, and obliging and ordering expression typical for school context and other situations where the relationship between participants is unequal and distant. The ideal reader of the text can be characterized as adolescent living in the world saturated by advertising and media. He or she is interested in the life of the celebrities, and is interested rather in her or his appearance, image and short-term enjoyment than health and long-term welfare. In the textbooks, healthy way of life is attempted to create a product which appeals to the values and interests of the imaginary public, the ideal reader of the text. Marketing healthy choices tend to reproduce stereotyped ideas of happiness, good life, youth and sex. Furthermore, individualizing approach mixed with wide definition of health legitimizes easily an erraneous impression of health, beauty and success being personal achievements dependent only on attitudes and competences.

Tämän pro gradu -tutkielman tarkoituksena on ollut tavoittaa niitä vaikutuksia, joita tietojärjestelmien käytöllä on sosiaali- ja terveysalalla. Tarkastelu sijoittuu lastensuojelun sosiaalityöhön. Yhden toimintayksikön tapahtumien kuvaus ja analyysi on puheenvuoro, jolla olen pyrkinyt tavoittamaan yhteyksiä tietojärjestelmien käyttöön liittyviin yleisiin ilmiöihin. Tutkielma on tiheä kuvaus lastensuojelun sosiaalityöstä asiakastietojärjestelmän muutosvaiheessa. Tapahtumia on jäsennetty toimijuuden käsitteen kautta (Archer 2003, Heiskala, 2000). Sosiaalityöntekijän toimijuus on ollut keskiössä tutkimussuunnitelman mukaisesti. Asiakastietojärjestelmä työntyi muutosvaiheessa toimijan rooliin vaikuttaen muun muassa lastensuojelun sosiaalityön työprosesseihin. Tietojärjestelmän toimijuus vaikuttaa sosiaalityöntekijään toimijana ja työntekijän toimijan asemaan. Aineistosta nousi teoretisoinnin (Layder, 1998) kautta esille jännitteisyys näiden keskeisten toimijoiden välillä. Jännite jäsentyi kolmelle kentälle. Lainsäädännön kenttä sulkee sisäänsä organisaatiota ja sosiaalityöntekijän perustehtävää kuvaavat kentät. Kenttien sisällöissä havaitsin yhtäläisyyttä Heiskalan (2000) yhteiskunnallisia pakkoja koskevaan jaotteluun. Pakot välittyvät semioottisesti tarkastellen merkkien eli lakimerkkien, rahamerkkien ja rituaalisesti vahvistettujen symbolien välityksellä. Tutkielman aineiston pohjalta on analysoitavaksi noussut kysymys uuden julkisjohtamisen (NPM) (Harvey, 2006, Julkunen, 2006) vaikutuksista lastensuojelun sosiaalityössä. Uuden julkisjohtamisen elementit tuottavat toimintaan jännitteitä edellä kuvatuilla kolmella kentällä. Yhtä toimintayksikköä koskeva aineisto antaa viitteitä siitä, että NPM toimii lastensuojelun sosiaalityössä toimintaa ohjaavana käyttöteoriana. Käyttöteorian (Argyris & Schön, 1996) käsitteellä ymmärretään yksilön tai organisaation todellista toimintaa ohjaavaa teoriaa, mikä voi olla ristiriidassa julkilausuttujen toiminnan teorioiden kanssa. Yksilö tai organisaatio ei välttämättä ole tietoinen käyttöteorian ja julkilausutun teorian välisestä kuilusta, minkä esiin saaminen edellyttäisi reflektiota. Tutkimuskohteena on ollut Helsingin kaupungin sosiaaliviraston ruotsinkielisiä lastensuojelun palveluja tuottava yksikkö. Tutkimuksen aineisto on kerätty uuden asiakastietojärjestelmän käyttöönottovaiheessa 1.11.2008–28.2.2009. Menetelmällisesti tutkimus sijoittuu toimintatutkimukselliseen kehykseen (Carr & Kemmis, 1986). Kenttävaiheen aikana yhdistin tutkijan ja toimijan roolit työskennellen sosiaalityöntekijänä tutkimukseni kohteena olleessa yksikössä. Kirjallinen aineisto koostuu järjestelmämuutosta koskevista päätöksistä, tiedotteista ja ohjeista. Järjestelmän muutosvaiheessa kokosin aineistoa osallistuvan havainnoinnin menetelmällä. Aineistossa on mukana myös yksikön työntekijöiden kanssa tuotettua materiaalia muutosvaiheen aikaisten sisältöjen haltuun ottamiseksi, esimerkiksi työn prosessikuvauksia. Aineisto on analysoitu sisällönanalyysillä. Analyysin edetessä kävin jäsennyksistäni keskustelua toimintayksikön työntekijöiden kanssa. Tapaamisten jälkeen jatkoin analyysiä ja teoretisointia käyttäen semioottista sosiologiaa metateoriana ja aktanttimallia muutosvaihetta kuvaavien kertomusten koostamisessa. Kertomusten pohjalta jatkoin toimijuuden analyysiä subjektiaseman (Törrönen, 2000) ja toimijan aseman (Alasuutari 2007) käsitteitä käyttäen.

This study identified the molecular defects underlying three lethal fetal syndromes. Lethal Congenital Contracture Syndrome 1 (LCCS1, MIM 253310) and Lethal Arthrogryposis with Anterior Horn Cell Disease (LAAHD, MIM 611890) are fetal motor neuron diseases. They affect the nerve cells that control voluntary muscle movement, and eventually result in severe atrophy of spinal cord motor neurons and fetal immobility. Both LCCS1 and LAAHD are caused by mutations in the GLE1 gene, which encodes for a multifunctional protein involved in posttranscriptional mRNA processing. LCCS2 and LCCS3, two syndromes that are clinically similar to LCCS1, are caused by defective proteins involved in the synthesis of inositol hexakisphosphate (IP6), an essential cofactor of GLE1. This suggests a common mechanism behind these fetal motor neuron diseases, and along with accumulating evidence from genetic studies of more late-onset motor neuron diseases such as Spinal muscular atrophy (SMA) and Amyotrophic lateral sclerosis (ALS), implicates mRNA processing as a common mechanism in motor neuron disease pathogenesis. We also studied gle1-/- zebrafish in order to investigate whether they would be a good model for studying the pathogenesis of LCCS1 and LAAHD. Mutant zebrafish exhibit cell death in their central nervous system at two days post fertilization, and the distribution of mRNA within the cells of mutant zebrafish differs from controls, encouraging further studies. The third lethal fetal syndrome is described in this study for the first time. Cocoon syndrome (MIM 613630) was discovered in a Finnish family with two affected individuals. Its hallmarks are the encasement of the limbs under the skin, and severe craniofacial abnormalities, including the lack of skull bones. We showed that Cocoon syndrome is caused by a mutation in the gene encoding the conserved helix-loop-helix ubiquitous kinase CHUK, also known as IκB kinase α (IKKα). The mutation results in the complete lack of CHUK protein expression. CHUK is a subunit of the IκB kinase enzyme that inhibits NF-κB transcription factors, but in addition, it has an essential, independent role in controlling keratinocyte differentiation, as well as informing morphogenetic events such as limb and skeletal patterning. CHUK also acts as a tumor suppressor, and is frequently inactivated in cancer. This study has brought significant new information about the molecular background of these three lethal fetal syndromes, as well as provided knowledge about the prerequisites of normal human development.

Pituitary adenomas are common benign neoplasms. Although most of them are sporadic, a minority occurs in familial settings. Heterozygous germline mutations in the aryl hydrocarbon receptor interacting protein (AIP) gene were found to underlie familial pituitary adenomas, a condition designated as pituitary adenoma predisposition (PAP). PAP confers incomplete penetrance of mostly growth hormone (GH) secreting adenomas in young patients, who often lack a family history of pituitary adenomas. This thesis work aimed to clarify the molecular and clinical characteristics of PAP. Applying the multiplex ligation-dependent probe amplification assay (MLPA), we found large genomic AIP deletions to account for a subset of PAP. Therefore, MLPA could be considered in PAP suspected patients with no AIP mutations found with conventional sequencing. We generated an Aip mouse model to examine pituitary tumorigenesis in vivo. The heterozygous Aip mutation conferred complete penetrance of pituitary adenomas that were mostly GH-secreting, rendering the phenotype of the Aip mouse similar to that of PAP patients. We suggest that AIP may function as a candidate gatekeeper gene in somatotrophs. To clarify molecular mechanisms of tumorigenesis, we elucidated the expression of AIP-related molecules in human and mouse pituitary tumors. The expression of aryl hydrocarbon receptor nuclear translocator (ARNT) was reduced in mouse Aip-deficient adenomas, and similar ARNT reduction was also evident in human AIP mutation positive adenomas. This suggests that in addition to participating in the hypoxia pathway, estrogen receptor signaling and xenobiotic response pathways, ARNT may play a role in AIP-related tumorigenesis. We also studied the characteristics and the response to therapy of PAP patients and found them to have an aggressive disease phenotype with young age at onset. Therefore, improvement in treatment outcomes of PAP patients would require their efficient identification and earlier diagnosis of the pituitary adenomas. The possible role of the RET proto-oncogene in tumorigenesis of familial AIP mutation negative pituitary adenomas was evaluated, but none of the found RET germline variants were considered pathogenic. Surprisingly, RET immunohistochemistry suggested possible underexpression of RET in AIP mutation positive pituitary adenomas an observation that merits further investigation.

Home Economics Classrooms as Part of Developing the Environment Housing Activities and Curriculums Defining Change --- The aim of the research project was to develop home economics classrooms to be flexible and versatile learning environments where household activities might be practiced according to the curriculum in different social networking situations. The research is based on the socio-cultural approach, where the functionality of the learning environment is studied specifically from an interactive learning viewpoint. The social framework is a natural starting point in home economics teaching because of the group work in classrooms. The social nature of learning thus becomes a significant part of the learning process. The study considers learning as experience based, holistic and context bound. The learning environment, i.e. home economics classrooms and the material tools there, plays a significant role in developing students skills to manage everyday life. --- The first research task was to analyze the historical development of household activities. The second research task was to develop and test criteria for functional home economics classrooms in planning both the learning environment and the students activities during lessons. The third research task was to evaluate how different professionals (commissioners, planners and teachers) use the criteria as a tool. The research consists of three parts. The first contains a historical analysis of how social changes have created tension between traditional household classrooms and new activities in homes. The historical analysis is based on housing research, regulations and instructions. For this purpose a new theoretical concept, the tension arch, was introduced. This helped in recognizing and solving problems in students activities and in developing innovations. The functionality criteria for home economics classrooms were developed based on this concept. These include technical (health, safety and technical factors), functional (ergonomic, ecological, aesthetic and economic factors) and behavioural (cooperation and interaction skills and communication technologies) criteria. --- The second part discusses how the criteria were used in renovating school buildings. Empirical data was collected from two separate schools where the activities during lessons were recorded both before and after classrooms were renovated. An analysis of both environments based on video recordings was conducted. The previously created criteria were made use of, and problematic points in functionality looked for particularly from a social interactive viewpoint. The results show that the criteria were used as a planning tool. The criteria facilitated layout and equipment solutions that support both curriculum and learning in home economics classrooms taking into consideration cooperation and interaction in the classroom. With the help of the criteria the home economics classrooms changed from closed and complicated space into integrated and open spaces where the flexibility and versatility of the learning environment was emphasized. The teacher became a facilitator and counselor instead a classroom controller. --- The third part analyses the discussions in planning meetings. These were recorded and an analysis was conducted of how the criteria and research results were used in the planning process of new home economics classrooms. The planning process was multivoiced, i.e. actors from different interest groups took part. All the previously created criteria (technical, functional and behavioural) emerged in the discussions and some of them were used as planning tools. Planning meetings turned into planning studios where boundaries between organizations were ignored and the physical learning environments were developed together with experts. The planning studios resulted in multivoiced planning which showed characteristics of collaborative and participating planning as well as producing common knowledge and shared expertise. --- KEY WORDS: physical learning environment, socio-cultural approach, tension arch, boundary crossing, collaborative planning.

Tutkielmassa tarkastellaan osallistuvaa demokratiaa ja kansalaiskuulemista sosiaali-ja terveyspolitiikan kehyksissä. Tutkittava tapaus on sosiaali-ja terveyspalvelulta kehittämään pyrkivä Kuntien parhaat palvelukäytännöt -hanke, joka on räätälöity vauhdittamaan vuonna 2005 käynnistettyä Paras-hanketta. Paras-hankkeessa on kyse sekä kuntien ja valtion vallanjaosta että palvelujärjestelmän kehittämisestä. Kuntien parhaat palvelukäytännöt - hankkeen päämäärä on ollut kehittää sosiaali- ja terveyspalvelulta eri tuottajatoimijöiden kesken. Hankkeiden taustalla vaikuttavat niin uuden julkisjohtamisen ja julkispalvelun eli governance poliittishallinnolliset ideologiat. Tutkittava aineisto koostuu kolmesta osasta: 1) keskusteluista otakantaa-foorumilla a) Parempia sosiaali- ja terveyspalvelulta - miten ja millaisin toimenpitein? sekä b) Miten kehittää kuntien, yksityisen ja kolmannen sektorin yhteistyötä?, 2) virkamiesten laatimasta yhteenvedosta keskustelusta a) ja 3) virkamieshaastatteluista. Aineistoa tarkastellaan sosiaalisen konstruktivismin teoreettismetodologisesta viitekehyksestä. Metodeina käytetään kriittistä diskurssianalyysiä, teoreettista sisällönanalyysiä sekä kehysanalyysiä. Tutkimuksen yleisenä viitekehyksenä toimivat poliittis-ideologiset hallinnolliset muutokset sekä konfliktikeskeinen deliberatiivinen demokratiateoria (Rosanvalion 2008). Tarkennettuna viitekehyksenä toimii osallistuva ja itsepalvelupolitiikka ja epäpolitisoiva ja politisoiva keskustelu niiden kehyksissä.Tutkimus osoittaa keskustelu sosiaali- ja terveyspalveluiden kehittämisestä on etenkin virkamiesten osalta epäpolitisoivaa. Uudistuksia käsitellään teknisinä ratkaisuina, ei poliittisina valintoina. Kansalaiskeskustelusta välittyy poliittisempia sävyjä, mitkä eivät kuitenkaan välity ylhäältä alas -ohjatulla keskustelulla. Kansalaisten ja virkamiesten välinen suhde ei ole hankkeessa vuorovaikutteinen, joten keskustelun ei voida kutsua osallistuvaksi demokratiaksi, vaan kansalaiskuulemiseksi. Tutkimus osoittaa myös, että tutkittua kansalaiskuulemista voidaan tarkastella itsepalveludemokratian viitekehyksestä. Markkinaohjautuvan sosiaali- ja terveyspolitiikan kehyksissä kansalaiset näyttäytyvät asiakas-subjekteina ja demokratian sovellukset kuten otakantaa.fi -foorumi edustavat hankkeen osalta ylhäältä alas -johdettua konsultaatiodemokratiaa, jolloin foorumin rooli demokraattisen keskustelun laajentamisessa jää vähäiseksi.

Human parvovirus B19 (B19V) is known to cause anemia, hydrops fetalis, and fetal death especially during the first half of pregnancy. Women who are in occupational contact with young children are at increased risk of B19V infection. The role of the recently discovered human parvovirus, human bocavirus (HBoV), in reproduction is unknown. The aim of this research project was to establish a scientific basis for assessing the work safety of pregnant women and for issuing special maternity leave regulations during B19V epidemics in Finland. The impact of HBoV infection on the pregnant woman and her fetus was also defined. B19V DNA was found in 0.8% of the miscarriages and in 2.4% of the intrauterine fetal death (IUFD; fetal death after completed 22 gestational weeks). All control fetuses (from induced abortions) were B19V-DNA negative. The findings on hydropic B19V DNA-positive IUFDs with evidence of acute or recent maternal B19V infection are in line with those of previous Swedish studies. However, the high prevalence of B19V-related nonhydropic IUFDs noted in the Swedish studies was mostly without evidence of maternal B19V infection and was not found during the third trimester. HBoV was not associated with miscarriages or IUFDs. Almost all of the studied pregnant women were HboV-IgG positive, and thus most probably immune to HBoV. All preterm births, perinatal deaths, smallness for gestational age (SGA) and congenital anomaly were recorded among the infants of child-care employees in a nationwide register-based cohort study over a period of 14 years. Little or no differences in the results were found between the infants of the child-care employees and those of the comparison group. The annual B19V seroconversion rate was over two-fold among the child-care employees, compared to the women in the comparison group. The seropositivity of the child-care employees increased with age, and years from qualification/joining the trade union. In general, the child-care employees are not at increased risk for adverse pregnancy outcome. However, at the population level, the risk of rare events, such as adverse pregnancy outcomes attributed to infections, could not be determined. According to previous studies, seronegative women had a 5 10% excess risk of losing the fetus during the first half of their pregnancy, but thereafter the risk was very low. Therefore, an over two-fold increased risk of B19V infection among child-care employees is considerable, and should be taken into account in the assessment of the occupational safety of pregnant women, especially during the first half of their pregnancy.

Pro gradu -tutkielmani aihe on nörtin stereotypia ja kuinka se on mahdollisesti muuttunut teknologiahypen ja yhteiskunnan muutoksen myötä. Tarkastelen eri aikakausien elokuvien tarjoamaa kuvaa nörteistä ja sitä miten tämä kuva on muuttunut. Lähtökohtana muutokselle on 1980-luvun stereotyyppinen kuva nörtistä teknisesti lahjakkaana, mutta epäsuosittuna ja sosiaalisesti lähes avuttomana mieshenkilönä. Hypoteesina tutkielmassani on muutos. Niin kutsutun nörttiyden yleistyessä ja muun muassa vuosituhannen vaihteen it-kuplan ja teknologiahypen myötä kuva nörtistä on muuttunut. Tutkielman tavoitteena on selventää mitä nörttiys voi olla nykyään ja mitä se on ollut ennen. Alustavat tutkimuskysymykseni, joita lähden tässä työssäni kehittelemään ja syventämään, ovat: – Kuinka nörtin stereotypia on muuttunut ajan myötä, 1980-luvulta nykyaikaan? – Mitkä asiat vaikuttavat nörtin stereotypiaan? Mistä nörttiys muodostuu? – Onko nörttiys oma alakulttuurinsa? Nörtin tarinaa lähden aluksi seuraamaan yhdysvaltalaisten elokuvien kautta. Elokuvissa henkilöhahmot usein esitetään tietynlaisina ihmistyyppeinä. Lähden etsimään nörttityyppiä sieltä, mistä sen voi nähdä syntyneen, yhdysvaltalaisesta nuorisokulttuurista ja sitä kuvaavista elokuvista. Etsin elokuvien tarjoamasta kuvasta nörtteihin liittyviä yleistyksiä ja lainalaisuuksia. Tämä gradu rakentuu erilaisten prismojen varaan, joiden keskiössä on nörtti. Lainaan prisman käsitettä luonnontieteistä, joissa prismalla tarkoitetaan optista laitetta, jolla voidaan taittaa, heijastaa tai hajottaa valoa valospektrin eri väreihin. Esimerkiksi auringon valkoinen valo taittuu prisman läpi kulkiessaan sateenkaaren kaikkiin väreihin. Spektri on se värien kirjo, johon esimerkiksi juuri auringon valo jakautuu. Nämä luonnontieteestä lainatut termit saattavat aluksi kuulostaa hieman kaukaa haetuilta ja käytänkin niitä tässä gradussa metaforana asioiden useammista näkökulmista ja tarkastelusuunnista. Tarkastelussa tässä tutkielmassa ovat sukupuolijärjestelmien prisma, toisessa päässään ”miehisyys” ja toisessa ”naiseus”. Toisella prismalla löytyvät etnisyyteen liittyvät asiat. Spektrit jakautuvat toisena ääripäänään ”rodullistettu herruus” ja toisena päätynä ”rodullistettu alisteisuus”. Yksi lähestymiskulma on uuden teknologian herättämät pelot sekä nörtti sivuroolissa, kritiikittömänä, urkinnan apulaisena. Viimeinen piirtämäni prisma vaikuttaa koko nörtti-imagon taustalla. Se liittyy yhteiskunnan muutokseen ja työnjakoon yhteiskunnassa. Kuten kaikki liittyy kaikkeen, myös tämä prisma liittyy jokaiseen aikaisemmin mainittuun näkemykseen

Bacteria growing in paper machines can cause several problems. Biofilms detaching from paper machine surfaces may lead to holes and spots in the end product or even break the paper web leading to expensive delays in production. Heat stable endospores will remain viable through the drying section of paper machine, increasing the microbial contamination of paper and board. Of the bacterial species regularly found in the end products, Bacillus cereus is the only one classified as a pathogen. Certain B. cereus strains produce cereulide, the toxin that causes vomiting disease in food poisonings connected to B. cereus. The first aim of this thesis was to identify harmful bacterial species colonizing paper machines and to assess the role of bacteria in the formation of end product defects. We developed quantitative PCR methods for detecting Meiothermus spp. and Pseudoxanthomonas taiwanensis. Using these methods I showed that Meiothermus spp. and Psx. taiwanensis are major biofoulers in paper machines. I was the first to be able to show the connection between end product defects and biofilms in the wet-end of paper machines. I isolated 48 strains of primary-biofilm forming bacteria from paper machines. Based on one of them, strain K4.1T, I described a novel bacterial genus Deinobacterium with Deinobacterium chartae as the type species. I measured the transfer of Bacillus cereus spores from packaging paper into food. To do this, we constructed a green fluorescent protein (GFP) labelled derivative of Bacillus thuringiensis and prepared paper containing spores of this strain. Chocolate and rice were the recipient foods when transfer of the labelled spores from the packaging paper to food was examined. I showed that only minority of the Bacillus cereus spores transferred into food from packaging paper and that this amount is very low compared to the amount of B. cereus naturally occurring in foods. Thus the microbiological risk caused by packaging papers is very low. Until now, the biological function of cereulide for the producer cell has remained unknown. I showed that B. cereus can use cereulide to take up K+ from environment where K+ is scarce: cereulide binds K+ ions outside the cell with high affinity and transports these ions across cell membrane into the cytoplasm. Externally added cereulide increased the growth rate of cereulide producing strains in medium where potassium was growth limiting. In addition, cereulide producing strains outcompeted cereulide non-producing B. cereus in potassium deficient environment, but not when the potassium concentration was high. I also showed that cereulide enhances biofilm formation of B. cereus.

Symptomatic hypertrophic breasts cause a health burden with physical and psychosocial morbidity. The value of reduction mammaplasty in the treatment of symptomatic breast hypertrophy has been consistently reported by patients and has been well recognised by plastic surgeons for a long time. However, the scientific evidence of the effects of reduction mammaplasty has been weak or lacking. During the design of this study most of the previous studies were retrospective and the few prospective studies had methodological limitations. Therefore, an obvious need for prospective randomised studies was present. Nevertheless, practical and ethical considerations seemed to make this study design impossible, because the waiting time for the operation was several years. The legislation and subsequent introduction of the uniform criteria for access to non-emergency treatment in Finland removed these obstacles, as all patients received their treatment within a reasonable time. As a result, a randomised controlled trial with a six-month follow-up time was designed and conducted. In addition, a follow-up study with two to five years follow-up was also carried out later. The effects of reduction mammaplasty on the patients breast-related symptoms, psychological symptoms, pain and quality of life was assessed. In addition, factors affecting the outcome were investigated. This study was carried out in the Hospital District of Helsinki and Uusimaa, Finland. Eighty-two out of the approximately 300 patients on the waiting list in 2004 agreed to participate in the study. Patients were randomised either to be operated (40 patients) on or to be followed up (42 patients). The follow-up time for both groups was six months. The patients were operated on by plastic surgeons or trainees at the Department of Plastic Surgery at Helsinki University Central Hospital or at the Department of Surgery at Hyvinkää Hospital. The patients completed five questionnaires: the SF-36 and the 15D quality of life questionnaires, the Finnish Breast-Associated Symptoms questionnaire (FBAS), a mood questionnaire (Raitasalo s modification of the short form of the Beck Depression Inventory, RBDI), and a pain questionnaire (The Finnish Pain Questionnaire, FPQ). Sixty-two out of the original 82 patients agreed to participate in the prospective follow-up study. In this study, patients completed the 15D quality of life questionnaire, the Finnish Breast-Associated Symptoms questionnaire, and the RBDI mood questionnaire. After six months follow-up, patients who had undergone reduction mammaplasty had a significantly better quality of life, fewer breast-associated symptoms and less pain, and they were less depressed or anxious when compared to patients who had not undergone surgery. The change in quality of life was more than two times the minimal clinically important difference. The patients preoperative quality of life was significantly inferior when compared to the age-standardised general population. This health burden was removed with reduction mammaplasty. The health loss related to symptomatic breast hypertrophy was comparable to that of patients with major joint arthrosis. In terms of change in quality of life, the intervention effect of reduction mammaplasty was comparable to that of hip joint replacement and more pronounced than that of knee joint replacement surgery. The outcome of reduction mammaplasty was affected more by preoperative psychosocial factors than by changes in breast dimensions. The effects of reduction mammaplasty remained stable at two to five years follow-up. In terms of quality of life, symptomatic breast hypertrophy causes a considerable health loss comparable to that of major joint arthrosis. Patients who undergo surgery have fewer breast-associated symptoms and less pain, and they are less depressed or anxious and have an improved quality of life. The intervention effect is comparable to that of major joint replacement surgery, and it remains stable at two to five years follow-up. The outcome of reduction mammaplasty is affected by preoperative psychosocial factors.

The nature of a burial is always ritualistic. This is often forgotten when dealing with Finnish inhumation burials containing animal bones. Only the animal bones found close to the deceased have traditionally been thought to have a ritualistic purpose. The animal bones found in the filling of the grave, which is still part of the burial, has on the other hand, often been neglected in the previous research. In this Master s thesis I will discuss the function and interpretation of animal bones in graves. The base of this study is six sites, all of different nature, from Finland. Luistari in Eura is from the western coast and is dated to Late Iron Age (and possibly Medieval period), the Medieval hamlet of Finno is situated in Espoo which is situated on the southern coast. Two town burials, Turku and Porvoo, are also included in this study. The graves from Turku are dated to Late Medieval period and Early Renaissance, whereas the cemetery in Porvoo is from the 18th century. Visulahti in Mikkeli is from the Late Iron Age and represents Eastern Finnish burial tradition, the same as Suotniemi from Käkisalmi parish, which is nowadays part of Russia. While parts of the animal bones had already been analysed before, the author also analysed animal bones for the purpose of the present Master´s thesis. The bones were compared to the burial contexts, when possible. Based on the comparisons I have made interpretations which might explain the existence of animal bones in the graves. The interpretations are among others sacrifice, commemoration meals and animal burials. The site could also have been a settlement site prior to the graves, thus the bones in the graves would belong to the settlement phase. When comparing the date of the studied sites, the town burials are later and the animal bones are probably related to previous or contemporary use of the sites as graveyards. On top of this there does not seem to be much difference in burial tradition between Eastern and Western Finland, although at least from the hamlet burials of Finno there are aspects that could be linked to Eastern burials. In making the interpretations I have taken into consideration the aspects of belief during different time periods when they could be accounted as relevant. Also the problems with bone preservation were relevant and challenging for the study. Often only the hardest substance of the skeleton, namely teeth, has been preserved. For this reason the quality of the archaeological documentation was a key issue in this study. In producing quality interpretations of the animal bones in graves, the bones, contexts and their relationship to the surrounding site should be documented with care.

Lung cancer accounts for more cancer-related deaths than any other cancer. In Finland, five-year survival ranges from 8% to 13%. The main risk factor for lung cancer is long-term cigarette smoking, but its carcinogenesis requires several other factors. The aim of the present study was to 1) evaluate post-operative quality of life, 2) compare clinical outcomes between minimally invasive and conventional open surgery, 3) evaluate the role of oxidative stress in the carcinogenesis of non-small lung cancer (NSCLC), and 4) to identify and characterise targeted agents for therapeutic and diagnostic use in surgery. For study I, pneumonectomy patients replied to 15D quality of life and baseline dyspnea questionnaires. Study III involved a prospective quality of life assessment using the 15D questionnaire after lobectomy or bi-lobectomy. Study IV was a retrospective comparison of clinical outcomes between 212 patients treated with open thoracotomy and 116 patients who underwent a minimally invasive technique. Study II measured parameters of oxidative metabolism (myeloperoxidase activity, glutathione content and NADPH oxidase activity) and DNA adducts. Study V employed the phage display method and identified a core motif for homing peptides. This method served in cell-binding, cell-localisation, and biodistribution studies. Following both pneumonectomy and lobectomy, NSCLC patients showed significantly decreased long-term quality of life. No significant correlation was noted between post-operative quality of life and pre-operative pulmonary function tests. Women suffered more from increased dyspnea after pneumonectomy which was absent after lobectomy or bi-lobectomy. Patients treated with video-assisted thoracoscopy showed significantly decreased morbidity and shorter periods of hospitalization than did open surgery patients. This improvement was achieved even though the VATS patients were older and suffered more comorbid conditions and poorer pulmonary function. No significant differences in survival were noted between these two groups. An increase in NADPH oxidase activity was noted in tumour samples of both adenocarcinoma and squamous cell carcinoma. This increase was independent from myeloperoxidase activity. Elevated glutathione content was noted in tumour tissue, especially in adenocarcinoma. After panning the clinical tumour samples with the phage display method, an amino acid sequence of ARRPKLD, the Thx, was chosen for further analysis. This method proved selective of tumour tissue in both in vitro and in vivo cell-binding assay, and biodistribution showed tumour accumulation. Because of the significantly reduced quality of life following pneumonectomy, other operative strategies should be implemented as an alternative (e.g. sleeve-lobectomy). To treat this disease, implementation of a minimally invasive surgical technique is safe, and the results showed decreased morbidity and a shorter period of hospitalisation than with thoracotomy. This technique may facilitate operative treatment of elderly patients with comorbid conditions who might otherwise be considered inoperable. Simultaneous exposure to oxidative stress and altered redox states indicates the important role of oxidative stress in the pathogenesis and malignant transformation of NSCLC. The studies showed with great specificity and with favourable biodistribution that Thx peptide is specific to NSCLC tumours. Thx thus shows promise in imaging, targeted therapy, and monitoring of treatment response.
