O objetivo deste trabalho foi estudar a influência das superfícies geomórficas na variação espacial da perda de solo por erosão na região de Pereira Barreto, São Paulo (SP). Os solos foram amostrados nos pontos de cruzamento de uma malha, georreferenciados, com intervalos de 350m, na profundidade de 0,0-0,2m, totalizando 67 pontos. Foram feitas determinações da composição granulométrica e do conteúdo de matéria orgânica. Foram avaliados os fatores de erosão locais, tais como erosividade (R), erodibilidade (K), fator topográfico (LS), uso e manejo (C), práticas conservacionistas (P), potencial natural de erosão (PNE), perda de solo com e sem práticas conservacionistas (A e *A), tolerância de perda de solo (T) e risco de erosão (RE). As variáveis A, PNE e RE apresentaram forte correlação espacial com o fator topográfico (LS), indicando a forte relação do relevo sobre os fatores de erosão. As perdas de solo (A e *A) apresentaram comportamento coerente com a conceituação de superfícies geomórficas, evidenciando as relações de dependência do processo erosivo do solo aos ambientes geomórficos.

The purpose of this study was the estimation of current and potential water erosion rates in Castellon Province (Spain) using RUSLE3D (Revised Universal Soil Loss Equation-3D) model with Geographical Information System (GIS) support. RUSLE3D uses a new methodology for topographic factor estimation (LS factor) based on the impact of flow convergence allowing better assessment of sediment distribution detached by water erosion. In RUSLE3D equation, the effect that vegetation cover has on soil erosion rate is reflected by the C factor. Potential erosion indicates soil erosion rate without considering C factor in RUSLE3D equation. The results showed that 57% of estimated current erosion does not exceed 10 t/ha.year (low erosion). In the case of potential erosion rates, 5% of the area of Castellon Province does not exceed 10 t/ha.year but 55% exceed 200 t/ha.year. Based on these results, the current vegetation cover of Castellon Province is adequate but needs to be conserved to avoid an increase in the current soil erosion rates as shown by potential erosion rates.

A cultura da cana-de-açúcar é a segunda maior movimentação econômica na cadeia do agronegócio no Brasil. Gera riquezas através da fabricação de açúcar, etanol e cogeração de energia elétrica, além de outros subprodutos. Considerada fonte de energia renovável, a cana-de- açúcar a princípio tinha sua imagem associada a impactos negativos principalmente devido as queimadas realizadas nas lavouras para colheita manual. Nos últimos anos, baseado em decretos e no Protocolo Agro-ambiental, essa prática vem sendo abolida. Para se manter e até mesmo aumentar o rendimento das colhedoras nos canaviais, os gestores têm adotado práticas para reduzir os terraços agrícolas, com impacto nos sistemas de conservação de solos. Assim, este trabalho teve como objetivo identificar os impactos ambientais provocados pela mecanização agrícola decorrente do mau manejo e dimensionamento dos mecanismos de conservação do solo. Neste estudo também se realizou uma análise à Equação Universal de Perdas de Solo (EUPS), como ferramenta para o dimensionamento de terraços agrícolas. O estudo foi realizado em uma microbacia hidrográfica, denominada Ribeirão da Bocaina, localizada na UGRHI-13 (Tietê - Jacaré). Foi possível identificar a variabilidade amostral do solo para o dimensionamento conservacionista, gerando curvas de nível com Desníveis Verticais (D.V) desuniformes, contrariando a sistemática atual de terraços que respeita cotas múltiplas ou mesmo dimensionamentos empiristas, segundo o conhecimento local e o histórico recente da área. Algumas sugestões também foram feitas afim de torná-la uma ferramenta ainda mais eficiente, considerando condições particulares à cultura da cana-de-açúcar, tais como a influência da palhada, sulcos de plantio e diversos tipos de terraços como meios de controle à erosão. A metodologia foi satisfatória, no que tange a compreensão pelos meios de correlação entre as práticas conservacionistas e modelos de predição de perda de solo, trazendo luz à ciência na interpretação das ferramentas existentes e as lacunas a serem preenchidas.

Myb is a key transcription factor that can regulate proliferation, differentiation, and apoptosis, predominantly in the haemopoietic system. Abnormal expression of Myb is associated with a number of cancers, both haemopoietic and non-haemopoietic. In order to better understand the role of Myb in normal and tumorigenic processes, we undertook a cDNA array screen to identify genes that are regulated by this factor. In this way, we identified the gene encoding vascular endothelial growth factor (VEGF) as being potentially regulated by the Myb oncoprotein in myeloid cells. To determine whether this was a direct effect on VEGF gene transcription, we examined the activity of the murine VEGF promoter in the presence of either wild-type (WT) or mutant forms of Myb. It was found that WT Myb was able to activate the VEGF promoter and that a minimal promoter region of 120 bp was sufficient to confer Myb responsiveness. Surprisingly, activation of the VEGF promoter was independent of DNA binding by Myb. This was shown by the use of DNA binding-defective Myb mutants and by mutagenesis of a potential Myb-binding site in the minimal promoter. Mutation of Sp1 sites within this region abolished Myb-mediated regulation of a reporter construct, suggesting that Myb DNA binding-independent activation of VEGF expression occurs via these Sp1 binding elements. Regulation of VEGF production by Myb has implications for the potential role of Myb in myeloid leukaemias and in solid tumours where VEGF may be functioning as an autocrine growth factor. SUMMARY : Eukaryotic DNA interacts with the nuclear proteins using non-covalent ionic interactions. Proteins can recognize specific nucleotide sequences based on the sterical interactions with the DNA and these specific protein-DNA interactions are the basis for many nuclear processes, e.g. gene transcription, chromosomal replication, and recombination. New technology termed ChIP-Seq has been recently developed for the analysis of protein-DNA interactions on a whole genome scale and it is based on immunoprecipitation of chromatin and high-throughput DNA sequencing procedure. ChIP-Seq is a novel technique with a great potential to replace older techniques for mapping of protein-DNA interactions. In this thesis, we bring some new insights into the ChIP-Seq data analysis. First, we point out to some common and so far unknown artifacts of the method. Sequence tag distribution in the genome does not follow uniform distribution and we have found extreme hot-spots of tag accumulation over specific loci in the human and mouse genomes. These artifactual sequence tags accumulations will create false peaks in every ChIP-Seq dataset and we propose different filtering methods to reduce the number of false positives. Next, we propose random sampling as a powerful analytical tool in the ChIP-Seq data analysis that could be used to infer biological knowledge from the massive ChIP-Seq datasets. We created unbiased random sampling algorithm and we used this methodology to reveal some of the important biological properties of Nuclear Factor I DNA binding proteins. Finally, by analyzing the ChIP-Seq data in detail, we revealed that Nuclear Factor I transcription factors mainly act as activators of transcription, and that they are associated with specific chromatin modifications that are markers of open chromatin. We speculate that NFI factors only interact with the DNA wrapped around the nucleosome. We also found multiple loci that indicate possible chromatin barrier activity of NFI proteins, which could suggest the use of NFI binding sequences as chromatin insulators in biotechnology applications. RESUME : L'ADN des eucaryotes interagit avec les protéines nucléaires par des interactions noncovalentes ioniques. Les protéines peuvent reconnaître les séquences nucléotidiques spécifiques basées sur l'interaction stérique avec l'ADN, et des interactions spécifiques contrôlent de nombreux processus nucléaire, p.ex. transcription du gène, la réplication chromosomique, et la recombinaison. Une nouvelle technologie appelée ChIP-Seq a été récemment développée pour l'analyse des interactions protéine-ADN à l'échelle du génome entier et cette approche est basée sur l'immuno-précipitation de la chromatine et sur la procédure de séquençage de l'ADN à haut débit. La nouvelle approche ChIP-Seq a donc un fort potentiel pour remplacer les anciennes techniques de cartographie des interactions protéine-ADN. Dans cette thèse, nous apportons de nouvelles perspectives dans l'analyse des données ChIP-Seq. Tout d'abord, nous avons identifié des artefacts très communs associés à cette méthode qui étaient jusqu'à présent insoupçonnés. La distribution des séquences dans le génome ne suit pas une distribution uniforme et nous avons constaté des positions extrêmes d'accumulation de séquence à des régions spécifiques, des génomes humains et de la souris. Ces accumulations des séquences artéfactuelles créera de faux pics dans toutes les données ChIP-Seq, et nous proposons différentes méthodes de filtrage pour réduire le nombre de faux positifs. Ensuite, nous proposons un nouvel échantillonnage aléatoire comme un outil puissant d'analyse des données ChIP-Seq, ce qui pourraient augmenter l'acquisition de connaissances biologiques à partir des données ChIP-Seq. Nous avons créé un algorithme d'échantillonnage aléatoire et nous avons utilisé cette méthode pour révéler certaines des propriétés biologiques importantes de protéines liant à l'ADN nommés Facteur Nucléaire I (NFI). Enfin, en analysant en détail les données de ChIP-Seq pour la famille de facteurs de transcription nommés Facteur Nucléaire I, nous avons révélé que ces protéines agissent principalement comme des activateurs de transcription, et qu'elles sont associées à des modifications de la chromatine spécifiques qui sont des marqueurs de la chromatine ouverte. Nous pensons que lés facteurs NFI interagir uniquement avec l'ADN enroulé autour du nucléosome. Nous avons également constaté plusieurs régions génomiques qui indiquent une éventuelle activité de barrière chromatinienne des protéines NFI, ce qui pourrait suggérer l'utilisation de séquences de liaison NFI comme séquences isolatrices dans des applications de la biotechnologie.

El tractament de les fístules perianals de la malaltia de Crohn es basa en procediments poc invasius, amb èxit variable. L’ús de PDGF (factors de creixement derivats de les plaquetes autòlogues), és habitual en altres camps quirúrgics per millorar la curació de les ferides. No s’ha estudiat la seva aplicació en la malaltia de Crohn. Aquest és un estudi pilot combinant PDGF amb tècniques habituals. La curació va ser del 80% dels malalts i el 88’8% de trajectes fistulosos. L’ús de PDGF és una tecnologia barata, poc invasiva i senzilla, i que sembla augmentar l’èxit del tractament.

Antecedentes: Existe la percepción de que los pacientes con trastorno mental severo (TMS) presentan una mayor prevalencia de factores de riesgo cardiovascular (FRCV) y un menor control de los mismos en comparación con la población general. Objetivos: Conocer la prevalencia de los FRCV y hábitos tóxicos en pacientes con TMS y evaluar el seguimiento por la Atención Primaria (AP) en los que presentan un riesgo vascular elevado. Diseño y metodología: Se realizó un estudio descriptivo de prevalencia y de seguimiento en una muestra representativa de los pacientes con diagnóstico de TMS seguidos en el Centro de salud mental de Mataró. Se registraron los parámetros antropométricos, los FRCV, hábitos tóxicos, perfil lipídico y tratamientos farmacológicos. Se calculó el RCV mediante las tablas de REGICOR. Los pacientes con un RCV & 10% fueron derivados a AP para valorar la estrategia terapéutica. Se evaluó el seguimiento por parte de los especialistas a los 4 meses y en Atención Primaria a los 6-9 meses. Resultados: Se realizaron 112 entrevistas en el centro de Salud Mental de Mataró; 62% eran hombres y 38% mujeres, con una edad media de 43 ± 11,9 años. La obesidad abdominal fue el FRCV más prevalente (62%), le siguió el tabaquismo en un 58,9%, el sedentarismo con un 48,2% y la dislipemia 42%. El 66,8% de los pacientes eran portadores de tratamiento con antipsicóticos atípicos, lo que se asocia con un mayor RCV. Un total de 102 pacientes (91% de la muestra) presentaban uno o más FRCV, 98 de los mismos tenían un RCV & 10 y 4 presentaron un RCV & 10. Se derivaron 6 pacientes a la AP para control de los FRCV, de los que 3 nunca habían sido atendidos en AP. A los 9 meses fueron visitados el 50% de los derivados a AP, y se consiguió reducir el RCV & 10 en un 50% de los que se le hizo la recomendación de seguimiento. Conclusión: Los pacientes con TMS acuden habitualmente a las consultas de AP para el control de los FRCV. La prevalencia de FRCV en pacientes con TMS es superior a la población general. El porcentaje de pacientes con RCV elevado según el REGICOR es bajo.

Objetivo. Determinar si la cantidad de sangre y su localización es el espacio subaracnoideo, medible con la escala de Fisher en las primeras 24 horas de ocurrido el ictus hemorrágico, es un factor predictor para el desarrollo de hidrocefalia en pacientes con diagnóstico de hemorragia subaracnoidea (HSA) espontánea, vistos en el Hospital Universitario Clínica San Rafael (HUCSR) con seguimiento de 12 meses. Métodos. 251 pacientes fueron incluidos en una cohorte retrospectiva. La asociación entre la escala de Fisher y el desarrollo de hidrocefalia en pacientes con HSA espontánea fue analizada a través de un análisis bivariado y multivariado. Resultados. La edad promedio de los pacientes fue de 55,5 ± 15 años; con predominancia en el sexo femenino 65,7%. La prevalencia de hidrocefalia fue de 27,1% en la cohorte y la etiología de la HSA fue en su mayoría por ruptura de aneurismas de arterias cerebrales; 78,5%. La sobrevida a 12 meses fue de 65,7%. Tanto el grado 4 en la escala de Fisher como Hunt-Hess III se asocian con el desarrollo de hidrocefalia: ORA; 2.93 IC 95%: 1.51-5.65, P <0.001, ORA 2.83 IC 95%: 1.31-6.17 P=0.008 respectivamente. Conclusión. La presencia de sangrado Intraventricular o intraparenquimatoso ( Fisher 4) en las primeras 24 horas, asociado a un deterioro neurológico al ingreso Hunt-Hess III están asociados con el desarrollo de hidrocefalia en los 251 pacientes evaluados con diagnóstico de HSA espontánea; hallazgos consistentes con lo reportado en la literatura mundial.

Introducción: una de las causas de pobre ganancia visual luego de un tratamiento exitoso de desprendimiento de retina, sin complicaciones, es el daño de los fotoreceptores, reflejada en una disrupción de la capa de la zona elipsoide y membrana limitante externa (MLE). En otras patologías se ha demostrado que la hiperautofluorescencia foveal se correlaciona con la integridad de la zona elipsoide y MLE y una mejor recuperación visual. Objetivos: evaluar la asociación entre la hiperautofluorescencia foveal, la integridad de la capa de la zona elipsoide y recuperación visual luego de desprendimiento de retina regmatógeno (DRR) exitosamente tratado. Evaluar la concordancia inter-evaluador de estos exámenes. Metodología: estudio de corte transversal de autofluorescencia foveal y tomografía óptica coherente macular de dominio espectral en 65 pacientes con DRR evaluados por 3 evaluadores independientes. La concordancia inter-evaluador se estudio mediante Kappa de Cohen y la asociación entre las diferentes variables mediante la prueba chi cuadrado y pruebas Z para comparación de proporciones. Resultados: La concordancia de la autofluorescencia fue razonable y la de la tomografía óptica coherente macular buena a muy buena. Sujetos que presentaron hiperautofluorescencia foveal asociada a integridad de la capa de la zona elipsoide tuvieron 20% más de posibilidad de recuperar agudeza visual final mejor a 20/50 que los que no cumplieron éstas características. Conclusión: Existe una asociación clínicamente importante entre la hiperautofluorescencia foveal, la integridad de la capa de zona elipsoide y la mejor agudeza visual final, sin embargo ésta no fue estadísticamente significativa (p=0.39)

This work examined how the conceptus modulates endometrial tissue remodeling and vascular development prior to implantation in mares. A macroscopic uterine examination was completed at day 21 of pregnancy. In situ morphology revealed that the endometrium involved in encroachment is restricted to the dorsal endometrium immediately overlying the yolk sac. The amount of stromal area occupied by blood vessels and the number of endometrial glands were increased during early pregnancy. Endometrial histomorphometry as well as the endometrial mRNA abundance and immunolocalization of VEGF, VEGFR1, VEGFR2, and Ki-67 was completed at days 14 and 21 of pregnancy, at day 10 of the estrous cycle, and during estrus. No obvious differences in VEGF and VEGFR1 protein localization were detected between pregnant and cycling mares but differential staining pattern for VEGFR2 and Ki-67 was observed. VEGFR2 localized to luminal and glandular epithelium of pregnant mares, while luminal epithelium was negative in cycling mares. Ki-67 staining was weak during the luteal phase but exhibited prominent luminal epithelium staining during estrus. In pregnant mares, all endometrial layers were Ki-67 positive. Quantitative RT-PCR revealed a greater abundance of VEGF mRNA during pregnancy. VEGFR2 transcript abundance was greatest in pregnant mares on day 21. This study supports the concept that the conceptus plays an active role in directing vasculogenesis within the uterus and thereby establishing hemotrophic nutrition that supports pregnancy after implantation. The present investigation observed the sealing ability of low shrinkage composite resins in large and deep cavities, placed and photocured in one increment. Large, deep cavities (5.0 mm diameter and 2.5 mm deep) surrounded by enamel were prepared in bovine teeth, which were then divided into five groups. Groups 1, 2, 3 and 4: acid conditioning + Adper Single Bond (3M/ESPE, St Paul, MN, USA) and restoration with Aelite LS Posterior (BISCO Inc. Schaumburg, IL, USA) (G1); Filtek Z-350 (3M/ESPE,St Paul, MN, USA) (G2); Filtek Z-350 Flow (3M/ESPE, St Paul, MN, USA) (G3); Premisa (KERR Corporation, Orange, CA, USA) (G4). Group 5: Silorane Adhesive system (3M/ESPE, St Paul, MN, USA) + restoration with Filtek Low Shrinkage Posterior P90 (3M/ESPE, St Paul, MN, USA). After polymerization, the teeth were immersed in 0.5% basic fuchsine solution and immediately washed. Using the Imagetool Software, the extent of dye along the margins was calculated as a percentage of total perimeter. The restorations were then transversally sectioned and the depth of dye penetration was calculated in mm, using the same software. Kruskal-Wallis analysis for all groups showed no statistical differences for extent (p = 0.54) or depth (p = 0.8364) of dye penetration. According to this methodology, the so-called low shrinkage composite resins had the same sealing ability compared to regular and flowable nanocomposite materials.

The reconstruction of the external ear to correct congenital deformities or repair following trauma remains a significant challenge in reconstructive surgery. Previously, we have developed a novel approach to create scaffold-free, tissue engineering elastic cartilage constructs directly from a small population of donor cells. Although the developed constructs appeared to adopt the structural appearance of native auricular cartilage, the constructs displayed limited expression and poor localization of elastin. In the present study, the effect of growth factor supplementation (insulin, IGF-1, or TGF-β1) was investigated to stimulate elastogenesis as well as to improve overall tissue formation. Using rabbit auricular chondrocytes, bioreactor-cultivated constructs supplemented with either insulin or IGF-1 displayed increased deposition of cartilaginous ECM, improved mechanical properties, and thicknesses comparable to native auricular cartilage after 4 weeks of growth. Similarly, growth factor supplementation resulted in increased expression and improved localization of elastin, primarily restricted within the cartilaginous region of the tissue construct. Additional studies were conducted to determine whether scaffold-free engineered auricular cartilage constructs could be developed in the 3D shape of the external ear. Isolated auricular chondrocytes were grown in rapid-prototyped tissue culture molds with additional insulin or IGF-1 supplementation during bioreactor cultivation. Using this approach, the developed tissue constructs were flexible and had a 3D shape in very good agreement to the culture mold (average error <400 µm). While scaffold-free, engineered auricular cartilage constructs can be created with both the appropriate tissue structure and 3D shape of the external ear, future studies will be aimed assessing potential changes in construct shape and properties after subcutaneous implantation.

This clinical study has investigated the antigenic activity of bacterial contents from exudates of acute apical abscesses (AAAs) and their paired root canal contents regarding the stimulation capacity by levels of interleukin (IL)-1 beta and tumor necrosis factor alpha (TNF-α) throughout the root canal treatment against macrophage cells. Paired samples of infected root canals and exudates of AAAs were collected from 10 subjects. Endodontic contents were sampled before (root canal sample [RCS] 1) and after chemomechanical preparation (RCS2) and after 30 days of intracanal medication with calcium hydroxide + chlorhexidine gel (Ca[OH]2 + CHX gel) (RCS3). Polymerase chain reaction (16S rDNA) was used for detection of the target bacteria, whereas limulus amebocyte lysate was used to measure endotoxin levels. Raw 264.7 macrophages were stimulated with AAA exudates from endodontic contents sampled in different moments of root canal treatment. Enzyme-linked immunosorbent assays were used to measure the levels of TNF-α and IL-1 beta. Parvimonas micra, Porphyromonas endodontalis, Dialister pneumosintes, and Prevotella nigrescens were the most frequently detected species. Higher levels of endotoxins were found in samples from periapical exudates at RCS1 (P < .005). In fact, samples collected from periapical exudates showed a higher stimulation capacity at RCS1 (P < .05). A positive correlation was found between endotoxins from exudates with IL-1 beta (r = 0.97) and TNF-α (r = 0.88) production (P < .01). The significant reduction of endotoxins and bacterial species achieved by chemomechanical procedures (RCS2) resulted in a lower capacity of root canal contents to stimulate the cells compared with that at RCS1 (P < .05). The use of Ca(OH)2 + CHX gel as an intracanal medication (RCS3) improved the removal of endotoxins and bacteria from infected root canals (P < .05) whose contents induced a lower stimulation capacity against macrophages cells at RCS1, RCS2, and RCS3 (P < .05). AAA exudates showed higher levels of endotoxins and showed a greater capacity of macrophage stimulation than the paired root canal samples. Moreover, the use of intracanal medication improved the removal of bacteria and endotoxins from infected root canals, which may have resulted in the reduction of the inflammatory potential of the root canal content.
