Bronsted acid-base ionic liquids (GILs) based on guanidine and acetic acid are efficient reaction media for palladium-catalyzed Heck reactions. They offer the advantages of high activity and reusability. GIL2 plays multiple roles in the reaction: it could act as solvent, as a strong base to facilitate beta-hydride elimination, and as a ligand to stabilize activated Pd species.

Nickel appears the most promising for the replacement of palladium among the inexpensive transition metals for that it is active for Heck reaction and about 500 times cheaper than palladium. In this article, we describe our recent results in the area of nickel-catalyzed Heck vinylations. Main focus is on the influence of ligand concentration in different organic solvents and the effects of the addition of water. The conversion is high in NMP and increases with increasing of PPh3/Ni ratio; in ethanol and toluene the conversion presents a maximum value on Ni(PPh3)(2). The effect of the addition of water is sensitive to the concentration of PPh3 and the nature of solvent, it was well explained with the formation and transfer of the active species in the different solvent systems.

A highly efficient Pd(OAc)(2)/guanidine aqueous system for the room temperature Suzuki cross-coupling reaction has been developed. The new water-soluble and air-stable catalyst Pd(OAc)(2)(.)(1f)(2) from Pd(OAc)(2) and 1,1,3,3-tetramethyl-2-n-butylguanidine (1f) was synthesized and characterized by X-ray crystallography. In the presence of Pd(OAc)(2)(.)(1f)(2), coupling of arylboronic acids with a wide range of aryl halides, including aryl iodides, aryl bromides, even activated aryl chlorides, was carried out smoothly in aqueous solvent to afford the cross-coupling products in good to excellent yields and high turnover numbers (TONs) (TONs up to 850 000 for the reaction of 1-iodo-4-nitrobenzene and phenylboronic acid). Furthermore, this mild protocol could tolerate a broad range of functional groups.

Palladium substituted in cerium dioxide in the form of a solid solution, Ce-0.98 Pd-0.02 O-1.98 is a new heterogeneous catalyst which exhibits high activity and 100% trans-selectivity for the Heck reactions of aryl bromides including heteroaryls with olefins. The catalytic reactions work without any ligand. Nano-crystalline Ce-0.98 Pd-0.02 O-1.98 is prepared by solution combustion method and Pd is in +2 state. The catalyst can be separated, recovered and reused without significant loss in activity.

A highly regioselective alkenylation of indole at the C2-position has been achieved using the Ru(II) catalyst by employing a directing group strategy. This strategy offers rare selectivity for the alkenylation N-benzoylindole at the C-2 position in the presence of the more active C3- and C7-position of indole and the ortho-positions of the benzoyl protecting group. A simple deprotection of the benzoyl group has also been exemplified, and the resulting product serves as a useful synthon for natural product syntheses.

The Heck reaction, performed in room temperature ionic liquids, has been studied by in situ XAFS, which indicates that palladium clusters of 0.8-1.6 nm diameter are the main species present during reaction.

This thesis describes work towards the total synthesis of a 7-aza analogue of the Amaryllidaceae alkaloid narciclasine, a potent anticancer compound which suffers from a poor solubility profile. A key strategy in the formation of the C-ring is the biotransformation of bromobenzene by E.coli JM109. The densely substituted heterocyclic A-ring is obtained by sequential directed ortho-metalation and the fragment union accomplished with an amide coupling and subsequent intramolecular Heck reaction.

An efficient, flexible, and stereoselective convergent route for constructing the trans-10-hydroxy1,1-dimethyloctahydrodibenzo[a,d]cyclohepten-7-ones (5a-c) was achieved via intramolecular Heck reaction. This strategy has been successfully implemented for the syntheses of (+/-)-komaroviquinone (3) through (+/-)-coulterone dimethyl ether (5c) and (+/-)-faveline methyl ether (1a).

(formula presented) The air, water, and highly thermally stable sulfur-containing palladacycles, mainly derived from the ortho-palladation of benzylic thioethers, are exceptional catalyst precursors for the Heck reaction. The reaction can be performed with aryl iodides, bromides, and chlorides, with acrylic esters and styrene, leading to turnover numbers up to 1 850 000.

A novel method for the synthesis of pyrrolidine C-nucleosides has been developed. The key step of the synthesis is the palladium(0)-mediated coupling of a disubstituted N-protected 2-pyrroline and 5-iodouracil. C-Nucleoside 14 and its N-methyl derivative 15 can easily be converted to the corresponding phosphoramidite building blocks for DNA synthesis

A study towards the synthesis of a new fulvestrant analogue with improved bioavailability was carried out. In this work a twelve-step synthetic route starting from β-estradiol was optimized and a palladium (Pd)-catalyzed endo-selective Heck reaction for the functionalization of an advanced intermediate was investigated.
