An important step in breeding for nutritionally enhanced varieties is determining the effects of the post-harvest supply chain on phytochemicals and the changes in VOCs produced over time. TD- GC-TOF-MS was used and a technique for the extraction of VOCs from the headspace using portable tubes is described. Forty-two compounds were detected; 39 were identified by comparison to NIST libraries. Thirty-five compounds had not been previously reported in Eruca sativa. Seven accessions were assessed for changes in headspace VOCs over 7 days. Relative amounts of VOCs across 3 time points were significantly different - isothiocyanate-containing molecules being abundant on 'Day 0'. Each accession showed differences in proportions/types of volatiles produced on each day. PCA revealed a separation of VOC profiles according to the day of sampling. Changes in VOC profiles over time could provide a tool for assessment of shelf-life.

The ability of nanoassisted laser desorption-ionization mass spectrometry (NALDI-MS) imaging to provide selective chemical monitoring with proper spatial distribution of lipid profiles from tumor tissues after plate imprinting has been tested. NALDI-MS imaging identified and mapped several potential lipid biomarkers in a murine model of melanoma tumor (inoculation of B16/F10 cells). It also confirmed that the in vivo treatment of tumor bearing mice with synthetic supplement containing phosphoethanolamine (PHO-S) promoted an accentuated decrease in relative abundance of the tumor biomarkers. NALDI-MS imaging is a matrix-free LDI protocol based on the selective imprinting of lipids in the NALDI plate followed by the removal of the tissue. It therefore provides good quality and selective chemical images with preservation of spatial distribution and less interference from tissue material. The test case described herein illustrates the potential of chemically selective NALDI-MS imaging for biomarker discovery.

Zusammenfassung der DoktorarbeitDie MALDI-TOF-Massenspektrometrie (Matrix Assisted Laser Desorption and Ionisation–Time Of Flight) ist in der Lage, Moleküle mit einem Molekulargewicht bis zu mehreren Hunderttausend Da intakt in die Gasphase zu überführen. Dabei wird die Fragmentierung des Analyten stark eingeschränkt bzw. gänzlich vermieden. Diese Methode findet daher zunehmend Verwendung für die Charakterisierung von Biopolymeren und synthetischen Polymeren. Ziel dieser Arbeit war, die MALDI-TOF-Massenspektrometrie zur Charakterisierung von Makromolekülen einzusetzen, bei denen die konventionellen polymeranalytischen Methoden nur unzureichende Informationen oder gar falsche bzw. gar keine Ergebnisse liefern. Mittels einer methodischen Entwicklung der MALDI-TOF-Massenspektrometrie gelang es, die bisherigen Grenzen der Methode zu erweitern und neue Anwendungsbereiche der Polymeranalytik aufzuzeigen. Anhand der erzielten Ergebnisse wurden darüber hinaus neue Erklärungsansätze formuliert, die zu einem besseren Verständnis des noch immer ungeklärten MALDI-Prozesses beitragen können. Besonders vielversprechend sind zum einen die Ergebnisse der Fragmentionenanalyse synthetischer Polymere und zum anderen die Charakterisierung von unlöslichen PAHs (Polycyclic Aromatic Hydrocarbons). Die Möglichkeiten und Aussagekraft der Fragmentionenanalyse wurde an synthetischen Polymeren getestet. Mit Hilfe dieser neuen Technik konnte die komplizierte Endgruppenverteilung einer Polycarbonat-Probe sowie die Zusammensetzung eines Poly-para-phenylenethynylen-b-Polyethylenoxid-Diblock-Copolymers eindeutig bestimmen werden, während die konventionellen MALDI-Massenspektren nur über einen wesentlich geringeren Informationsgehalt verfügten. Auf dem Gebiet der Analytik von unlöslichen PAHs wurde mit der Entwicklung einer neuen MALDI-Probenvorbereitung eine Methode gefunden, die über die PAH Analytik hinaus von großem Nutzen ist. Diese erstmalig angewendete Probenvorbereitung unterscheidet sich von den üblichen MALDI-Probenpräparationen, indem sie auf die Beteiligung eines Lösungsmittels vollkommen verzichtet. Damit konnte speziell ein unlöslicher, zuvor nicht nachweisbarer PAH von ca. 2700 Da mit MALDI eindeutig charakterisiert werden.

Im ersten Teil dieser Arbeit wird die Anwendung der MALDI-TOF Massenspektrometrie auf annähernd monodisperse und eng verteilte Verbindungen polymerer Gestalt beschrieben. Dazu werden mit Polyphenylendendrimeren, Polybutadien-Sternpolymeren, Propionylethyleniminen und Polyethylmethylsiloxanen weniger gängige Verbindungen ausgewählt, für deren Analyse keine Standardmethodik vorliegt. Dabei wird gezeigt, dass andere Analysemethoden (wie z.B. GPC oder NMR) der MALDI-TOF Massenspektrometrie oftmals deutlich unterlegen sind - in einigen Fällen werden deren Ergebnisse gar widerlegt. In anderen Fällen wird allerdings auch festgestellt, dass diese Methoden die Massenspektrometrie unterstützen oder ergänzen können. Im darauf folgenden Teil werden durch Mischen eng verteilter Polymerstandards breite Verteilung simuliert, um so die Gründe für die Limitierung der Anwendbarkeit der MALDI-TOF Massenspektrometrie auf breite Verteilungen zu ermitteln. Die Problematik der Messung von breiten Verteilungen wird oftmals durch eine vorherige Fraktionierung mit der GPC gelöst. Auch dieses Verfahren wird in dieser Arbeit an Hand von simulierten breiten Verteilungen untersucht und bewertet. Neben qualitativen Untersuchungen mit der MALDI-TOF Massenspektrometrie wird in dieser Arbeit abgeschätzt, inwieweit diese Methode auch zur quantitativen Analyse eingesetzt werden kann. Dazu werden die Untersuchungen von Oligo(para-phenylene) (OPP) und von Cyclodehydrierungsprodukten sehr großer Dendrimere (C385, C474) beschrieben. Dabei stellt sich heraus, das neben der direkten UV-Absorption (im Falle der OPPs) auch die zunehmende Ionisierungswahrscheinlichkeit und die sinkende Desorptionswahrscheinlichkeit mit fortschreitender Cyclodehydrierung eine Quantifizierung problematisch gestalten. In einem abschließenden Kapitel wird der Versuch beschrieben, aus polyzyklischen aromatischen Kohlenwasserstoffen (PAHs) durch den Beschuss mit den UV-Strahlen des MALDI-TOF Massenspektrometers Fullerene zu erzeugen. Der Einsatz von NaCl als sogenannte Inertmatrix führt zu vielversprechenden ersten Ergebnissen.

Species of the family Pasteurellaceae play an important role as primary or opportunistic, predominantly respiratory, pathogens in domestic and wild animals. Some of them cause severe disease with high economic losses in commercial animal husbandry. Hence, rapid and accurate differentiation of Pasteurellaceae is important and signifies a particular challenge to diagnostic laboratories. Identification and differentiation of Pasteurellaceae is mostly done using phenotypic tests or genetic identification based on sequence similarity of housekeeping genes, such as the rrs gene encoding the 16S ribosomal RNA (16S rRNA). Both approaches are time consuming, laborious, and costly, therefore often delaying the final diagnosis of disease or epidemics. Matrix-assisted laser desorption/ionization time-of-flight (MALDI-TOF) mass spectrometry represents an alternative rapid and reliable method for the differentiation of most members of the family Pasteurellaceae. It is able to differentiate within a few minutes the currently known 18 genera and most of the over 60 species and subspecies of Pasteurellaceae including many members encountered in veterinary diagnostic laboratories. A few closely related species and subspecies that cannot be discriminated by MALDI-TOF are easily identified further by complementary simple tests, such as hemolysis done simultaneously or routinely during pathogen isolation.

Noble gas analysis in early solar system materials, which can provide valuable information about early solar system processes and timescales, are very challenging because of extremely low noble gas concentrations (ppt). We therefore developed a new compact sized (33 cm length, 7.2cm diameter, 1.3 L internal volume) Time-of-Flight (TOF) noble gas mass spectrometer for high sensitivity. We call it as Edel Gas Time-of-flight (EGT) mass spectrometer. The instrument uses electron impact ionization coupled to an ion trap, which allows us to ionize and measure all noble gas isotopes. Using a reflectron set-up improves the mass resolution. In addition, the reflectron set-up also enables some extra focusing. The detection is via MCPs and the signals are processed either via ADC or TDC systems. The objective of this work is to understand the newly developed Time-Of-Flight (TOF) mass spectrometer for noble gas analysis in presolar grains of the meteorites. Chapter 1 briefly introduces the basic idea and importance of the instrument. The physics relevant to time-of-flight mass spectrometry technique is discussed in the Chapter 2 and Chapter 3 will present the oxidation technique of nanodiamonds of the presolar grains by using copper oxide. Chapter 4 will present the details about EGT data analysis software. Chapter 5 and Chapter 6 will explain the details about EGT design and operation. Finally, the performance results will be presented and discussed in the Chapter 7, and whole work is summarized in Chapter 8 and also outlook of the future work is given.

Il teff (Eragrostis tef) è uno pseudocereale originario dell’Etiopia che recentemente si sta diffondendo in vari Paesi. Infatti presenta numerose caratteristiche positive (buon contenuto di fibra e composti fenolici, indice glicemico ridotto, assenza di glutine) che potrebbero renderlo adatto alla formulazione di alimenti funzionali o arricchiti, destinati a categorie della popolazione con particolari esigenze (diabetici, celiaci, neonati). I dati attualmente disponibili riguardo il profilo fenolico del teff sono estremamente carenti. Pertanto, lo scopo di questa tesi è la determinazione dei composti fenolici liberi e legati in una farina di teff, mediante la combinazione della cromatografia liquida ad alte prestazioni e la spettrometria di massa con analizzatore a tempo di volo (HPLC-ESI-TOF-MS). L’estrazione dei fenoli liberi e legati dal campione di farina è stata condotta seguendo il metodo messo a punto da Verardo et al. (2011). Dall'analisi dei rispettivi base peak chromatogram sono stati individuati 15 composti fenolici nell'estratto solubile e 5 nell'estratto insolubile. I fenoli liberi riscontrati sono esclusivamente flavonoidi: 11 derivati dall'apigenina, 2 derivati dalla luteolina e 2 composti acetilati derivati dalla vitexina, identificati per la prima volta nella farina di teff. L'estratto insolubile risulta composto da acidi fenolici, rappresentati dall'acido ferulico e i suoi derivati. Non è stato possibile effettuare la determinazione dei composti fenolici totali mediante saggio al reattivo di Folin-Ciocalteu a causa dell’intorbidimento dell’estratto in fase di diluizione: questo ha confermato il limite di tale metodologia, sebbene ampiamente utilizzata, nella determinazione dei composti fenolici.

Corynebacterium species (spp.) are among the most frequently isolated pathogens associated with subclinical mastitis in dairy cows. However, simple, fast, and reliable methods for the identification of species of the genus Corynebacterium are not currently available. This study aimed to evaluate the usefulness of matrix-assisted laser desorption ionization/mass spectrometry (MALDI-TOF MS) for identifying Corynebacterium spp. isolated from the mammary glands of dairy cows. Corynebacterium spp. were isolated from milk samples via microbiological culture (n=180) and were analyzed by MALDI-TOF MS and 16S rRNA gene sequencing. Using MALDI-TOF MS methodology, 161 Corynebacterium spp. isolates (89.4%) were correctly identified at the species level, whereas 12 isolates (6.7%) were identified at the genus level. Most isolates that were identified at the species level with 16 S rRNA gene sequencing were identified as Corynebacterium bovis (n=156; 86.7%) were also identified as C. bovis with MALDI-TOF MS. Five Corynebacterium spp. isolates (2.8%) were not correctly identified at the species level with MALDI-TOF MS and 2 isolates (1.1%) were considered unidentified because despite having MALDI-TOF MS scores >2, only the genus level was correctly identified. Therefore, MALDI-TOF MS could serve as an alternative method for species-level diagnoses of bovine intramammary infections caused by Corynebacterium spp.
