The aim of this study was to verify the effect of a double distillation on the reduction of the ethyl carbamate content in sugar cane spirit. Ethyl carbamate is a potentially carcinogenic compound normally present at critical levels in sugar cane spirit, constituting a public health problem and therefore hindering the export of this beverage. The ethanol, copper and ethyl carbamate contents were evaluated, using gas chromatography/mass spectroscopy, during a double distillation of the fermented sugar cane juice. The distillate fraction from the first distillation accumulated 30% of the ethyl carbamate formed. In the second distillation, the ethyl carbamate and the copper content increased during the process as the alcohol content decreased, and only 3% of the ethyl carbamate formed was collected in the spirit. Double distillation decreased the ethyl carbamate content in the sugar cane spirit by 97%. (C) Copyright 2012 The Institute of Brewing & Distilling

Objective: The aim of this study was to screen CO2 laser (10.6 mu m) parameters to increase enamel resistance to a continuous-flow erosive challenge. Background data: A new clinical CO2 laser providing pulses of hundreds of microseconds, a range known to increase tooth acid-resistance, has been introduced in the market. Methods: Different laser parameters were tested in 12 groups (n = 20) with varying fluences from 0.1 to 0.9 J/cm(2), pulse durations from 80 to 400 mu s and repetition rates from 180 to 700 Hz. Non-lased samples (n = 30) served as controls. All samples were eroded by exposure to hydrochloric acid (pH 2.6) under continuous acid flow (60 mu L/min). Calcium and phosphate release into acid was monitored colorimetrically at 30 sec intervals up to 5 min and at 1 min intervals up to a total erosion time of 15 min. Scanning electron microscopic (SEM) analysis was performed in lased samples (n = 3). Data were statistically analysed by one-way ANOVA (p < 0.05) and Dunnett's post-hoc tests. Results: Calcium and phosphate release were significantly reduced by a maximum of 20% over time in samples irradiated with 0.4 J/cm(2) (200 mu s) at 450 Hz. Short-time reduction of calcium loss (<= 1.5 min) could be also achieved by irradiation with 0.7 J/cm(2) (300 mu s) at 200 and 300 Hz. Both parameters revealed surface modification. Conclusions: A set of CO2 laser parameters was found that could significantly reduce enamel mineral loss (20%) under in vitro erosive conditions. However, as all parameters also caused surface cracking, they are not recommended for clinical use.

Background Natural antioxidants present in common foods and beverages have drawn great attention to cancer prevention due to its health benefits, remarkable lack of toxicity and side effects. Copaifera langsdorffii, known as “copaiba”, “capaiva”, or “pau-de-óleo“, belongs to the Leguminosae family and occurs in fields and grasslands in the northern and northeastern parts of Brazil. Biological studies of Copaifera corroborate its widespread use by the population. This paper describes the effects of C. langsdorffii leaves hydroalcoholic extract on the 1,2-dimethylhydrazine (DMH)-induced DNA damage and aberrant crypt foci (ACF) in the colon of male Wistar rats. Methods The hydroalcoholic extract of C. langsdorffii was administered to rats by gavage at daily doses of 20, 40 and 80 mg/kg body weight. To evaluate DNA damage by the comet assay, animals received the C. langsdorffii extract for seven days and a single subcutaneous injection (sc) of 1,2-dimethylhydrazine (DMH) at a dose of 40 mg/kg on day 7. Animals were sacrificed 4 h after injection of DMH, to assess DNA damage. For the ACF assay, animals were acclimatized for one week (week 1) and then treated with the C. langsdorffii extract five times a week for four weeks (weeks 2 to 5). The rats received sc injections of DMH (40 mg/kg) on days 2 and 5 of weeks 2 and 3, to induce ACF. Animals were euthanized at week 5; i.e., four weeks after the first DMH treatment. Results Animals treated with different doses of the C. langsdorffii extract combined with DMH had significantly lower frequency of DNA damage as compared with the positive control (animals treated with DMH only). The percentage of reduction in the frequency of DNA damage ranged from 14.30% to 38.8%. The groups treated with 40 and 80 mg/kg C. langsdorffii extract during and after DMH treatment presented significantly lower numbers of ACF and aberrant crypts compared with the control. Conclusion The C. langsdorffii extract significantly reduced the extent of DNA damage and ACF induced by DMH, suggesting that the extract has a protective effect against colon carcinogenesis.

Este giro tiene como telón de fondo el tránsito del monopolio estatal de ambas bebidas al ámbito privado y la defensa que los pueblos de indios hicieron de su consumo, en un momento de transición social y económica hacia el Estado agroexportador cafetalero.

Polyphenols, including flavonoids and stilbenes, are an essential part of human diet and constitute one of the most abundant and ubiquitous group of plant secondary metabolites. The level of these compounds is inducible by stress or fungal attack, so attempts are being made to identify likely biotic and abiotic elicitors and to better understand the underlying mechanism. Resveratrol (3,5,4’-trihydroxystilbene), which belongs to the stilbene family, is a naturally occurring polyphenol, found in several fruits, vegetables and beverages including red wine. It is one of the most important plant polyphenols with proved benefic activity on animal health. In the last two decades, the potential protective effects of resveratrol against cardiovascular and neurodegenerative diseases, as well as the chemopreventive properties against cancer, have been largely investigated. The most important source of polyphenols and in particular resveratrol for human diet is grape (Vitis vinifera). Since stilbenes and flavonoids play a very important role in plant defence responses and enviromental interactions, and their effects on human health seem promising, the aim of the research of this Thesis was to study at different levels the activation and the regulation of their biosynthetic pathways after chitosan treatment. Moreover, the polyphenol production in grape cells and the optimisation of cultural conditions bioreactor scale-up, were also investigated. Cell suspensions were obtained from cv. Barbera (Vitis vinifera L.) petioles and were treated with a biotic elicitor, chitosan (50 μg/mL, dissolved in acetic acid) to promote phenylpropanoid metabolism. Chitosan is a D-glucosamine polymer from fungi cell wall and therefore mimes fungal pathogen attack. Liquid cultures have been monitored for 15 days, measuring cell number, cell viability, pH and grams of fresh weight. The endogenous and released amounts of 7 stilbenes (trans and cis isomers of resveratrol, piceid and resveratroloside, and piceatannol), gallic acid, 6 hydroxycinnamic acids (trans-cinnamic, p-coumaric, caffeic, ferulic, sinapic and chlorogenic acids), 5 catechines (catechin, epicatechin, epigallocatechin-gallate (EGCG), epigallocatechin and epicatechin-gallate) and other 5 flavonoids (chalcon, naringenin, kaempferol, quercetin and rutin) in cells and cultural medium, were measured by HPLC-DAD analysis and total anthocyanins were quantified by spectrophotometric analysis. Chitosan was effective in stimulating trans-resveratrol endogenous accumulation with a sharp peak at day 4 (exceeding acetic acid and water controls by 36% and 63%, respectively), while it did not influence the production of the cis-isomer. Compared to both water and acetic acid controls, chitosan decreased the release of both trans- and cis-resveratrol respect to controls. No effect was shown on the accumulation of single resveratrol mono-glucoside isomers, but considering their total amount, normalized for the relative water control, it was possible to evidence an increase in both accumulation and release of those compounds, in chitosan-treated cells, throughout the culture period and particularly during the second week. Many of the analysed flavonoids and hydroxycinnamic acids were not present or detectable in trace amounts. Catechin, epicatechin and epigallocatechin-gallate (EGCG) were detectable both inside the cells and in the culture media, but chitosan did not affect their amounts. On the contrary, total anthocyanins have been stimulated by chitosan and their level, from day 4 to 14, was about 2-fold higher than in both controls, confirming macroscopic observations that treated suspensions showed an intense brown-red color, from day 3 onwards. These elicitation results suggest that chitosan selectively up-regulates specific biosynthetic pathways, without modifying the general accumulation pattern of other flavonoids. Proteins have been extracted from cells at day 4 of culture (corresponding to the production peak of trans-resveratrol) and separated by bidimensional electrophoresis. The 73 proteins that showed a consistently changed amount between untreated, chitosan and acetic acid (chitosan solvent) treated cells, have been identified by mass spectrometry. Chitosan induced an increase in stilbene synthase (STS, the resveratrol biosynthetic enzyme), chalcone-flavanone isomerase (CHI, that switches the pathway from chalcones to flavones and anthocyanins), pathogenesis-related proteins 10 (PRs10, a large family of defence proteins), and a decrease in many proteins belonging to primary metabolisms. A train of six distinct spots of STS encoded by the same gene and increased by chitosan, was detected on the 2-D gels, and related to the different phosphorylation degree of STS spots. Northern blot analyses have been performed on RNA extracted from cells treated with chitosan and relative controls, using probes for STS, PAL (phenylalanine ammonia lyase, the first enzyme of the biosynthetic pathway), CHS (chalcone synthase, that shares with STS the same precursors), CHI and PR-10. The up-regulation of PAL, CHS and CHI transcript expression levels correlated with the accumulation of anthocyanins. The strong increase of different molecular weight PR-10 mRNAs, correlated with the 11 PR-10 protein spots identified in proteomic analyses. The sudden decrease in trans-resveratrol endogenous accumulation after day 4 of culture, could be simply explained by the diminished resveratrol biosynthetic activity due to the lower amount of biosynthetic enzymes. This might be indirectly demonstrated by northern blot expression analyses, that showed lower levels of phenylalanine ammonia lyase (PAL) and stilbene synthase (STS) mRNAs starting from day 4. Other possible explanations could be a resveratrol oxidation process and/or the formation of other different mono-, di-glucosides and resveratrol oligomers such as viniferins. Immunolocalisation experiments performed on grape protoplasts and the subsequent analyses by confocal microscope, showed that STS, and therefore the resveratrol synthetic site, is mostly associated to intracellular membranes close to the cytosolic side of plasma membrane and in a smaller amount is localized in the cytosol. STS seemed not to be present inside vacuole and nucleus. There were no differences in the STS intracellular localisation between the different treatments. Since it was shown that stilbenes are largely released in the culture medium and that STS is a soluble protein, a possible interaction of STS with a plasma membrane transporter responsible for the extrusion of stilbenes in the culture medium, might be hypothesized. Proteomic analyses performed on subcellular fractions identified in the microsomial fraction 5 proteins taking part in channel complexes or associated with channels, that significantly changed their amount after chitosan treatment. In soluble and membrane fractions respectively 3 and 4 STS and 6 and 3 PR-10 have been identified. Proteomic results obtained from subcellular fractions substantially confirmed previous result obtained from total cell protein extracts and added more information about protein localisation and co-localisation. The interesting results obtained on Barbera cell cultures with the aim to increase polyphenol (especially stilbenes) production, have encouraged scale up tests in 1 litre bioreactors. The first trial fermentation was performed in parallel with a normal time-course in 20 mL flasks, showing that the scale-up (bigger volume and different conditions) process influenced in a very relevant way stilbenes production. In order to optimise culture parameters such as medium sucrose amount, fermentation length and inoculum cell concentration, few other fermentations were performed. Chitosan treatments were also performed. The modification of each parameter brought relevant variations in stilbenes and catechins levels, so that the production of a certain compound (or class of compounds) could be hypothetically promoted by modulating one or more culture parameters. For example the catechin yield could be improved by increasing sucrose content and the time of fermentation. The best results in stilbene yield were obtained in a 800 mL fermentation inoculated with 10.8 grams of cells and supplemented with chitosan. The culture was fed with MS medium added with 30 g/L sucrose, 25 μg/mL rifampicin and 50 μg/mL of chitosan, and was maintained at 24°C, stirred by marine impeller at 100 rpm and supplied of air at 0.16 L/min rate. Resveratroloside was the stilbene present in the larger amount, 3-5 times more than resveratrol. Because resveratrol glucosides are similarly active and more stable than free resveratrol, their production using a bioreactor could be a great advantage in an hypothetical industrial process. In my bioreactor tests, stilbenes were mainly released in the culture medium (60-80% of the total) and this fact could be another advantage for industrial applications, because it allows recovering the products directly from the culture medium without stopping the fermentation and/or killing the cells. In my best cultural conditions, it was possible to obtain 3.95 mg/L of stilbenes at day 4 (maximum resveratrol accumulation) and 5.13 mg/L at day 14 (maximum resveratroloside production). In conclusion, chitosan effect in inducing Vitis vinifera defense mechanisms can be related to its ability to increase the intracellular content of a large spectrum of antioxidants, and in particular of resveratrol, its derivates and anthocyanins. Its effect can be observed at transcriptional, proteomic (variation of soluble and membrane protein amounts) and metabolic (polyphenols production) level. The chitosan ability to elicit specific plant matabolisms can be useful to produce large quantities of antioxidant compounds from cell culture in bioreactor.

Hefen der Gattungen Brettanomyces/Dekkera sind in der Produktion von fermentierten Getränken, insbesondere in der Bier-, Sekt- und Weinherstellung bekannt. Sie können als Schädlingshefen insbesondere durch die Bildung von charakteristischen Sekundärmetaboliten zu einer negativen geschmacklichen Veränderung des Getränks führen. Aufgrund ihres langsamen Wachstums werden diese Hefen bei Routineanalysen mit konventionellen Kultivierungsmethoden leicht übersehen. Ein schneller und eindeutiger Nachweis von Brettanomyces/Dekkera-Hefen ist bis heute problematisch. In der vorliegenden Arbeit wurde eine Methode zur sicheren Detektion und Identifizierung aller fünf bekannten Spezies dieser Gattungen entwickelt. Die Fluoreszenz in situ Hybridisierung (FISH) mit Cy3-markierten DNA-Sonden ermöglichte einen direkten mikroskopischen Nachweis dieser Mikroorganismen in der Untersuchungsprobe. Im Hinblick auf die Generierung Art-spezifischer Sonden wurden die ribosomalen Gen-Cluster der verschiedenen Spezies hinsichtlich potentieller Zielregionen analysiert. Eine signifikante Steigerung des Sonden-vermittelten Fluoreszenz-Signals konnte durch die Anwendung eines neuen Sonden-Konzepts (Gemeinschafts-Sonden) auf hochvariable Bereichen der 26S rRNAs, unter Berücksichtigung ihrer Sekundärstrukturen, realisiert werden. Die Untersuchung der regionalen Verbreitung dieser Hefen in der Weinbauregion Rheinhessen ergab, dass bei 15 % der untersuchten Winzerbetriebe D. bruxellensis in Rotweinproben vorhanden war. Insgesamt konnten bei den Probenuntersuchungen aus 299 Weinen 44 D. bruxellensis-Stämme isoliert werden. Im Rahmen dieser Arbeit wurden darüber hinaus verschiedene Vitalitätsfärbungen hinsichtlich ihrer Anwendbarkeit auf Brettanomyces/Dekkera evaluiert und eine Differenzierung dieser Hefen durch einen physiologischen Mikrotiterplatten-Test (Biolog, USA) überprüft.

Das Wachstum von Milchsäurebakterien-Arten der Gattungen Lactobacillus, Pediococcus und Leuconostoc während der Weinfermentation kann durch die Bildung verschiedener Stoffwechselprodukte zu Weinfehlern führen. Um rechtzeitig Gegenmaßnahmen ergreifen zu können und einem Weinverderb vorzubeugen, bedarf es geeigneter Identifizierungsmethoden. Klassische mikrobiologische Methoden reichen oft nicht aus, um Mikroorganismen auf Art- und Stammniveau gezielt zu identifizieren. Wegen ihrer schnellen Durchführbarkeit und Zuverlässigkeit sind molekularbiologische Identifizierungsmethoden zur Kontrolle der mikrobiellen Flora während der Lebensmittelfermentierung in der heutigen Zeit unabdingbar. In der vorliegenden Forschungsarbeit wurden die 23S rRNA-Gensequenzen von neun Pediococcus-Typstämmen sequenziert, analysiert und phylogenetische Analysen durchgeführt. Zur Art-Identifizierung der Pediokokken wurden PCR-Primer generiert und ein Multiplex PCR System entwickelt, mit dem alle typischen Arten simultan in einer Reaktion nachgewiesen werden konnten. Die Ergebnisse der Multiplex PCR-Identifizierung von 62 Pediococcus-Stämmen aus Kulturensammlungen und 47 neu isolierten Stämmen aus Wein zeigten, dass einige Stämme unter falschen Artnamen hinterlegt waren, und dass P. parvulus im Weinanbaugebiet Rheinhessen weit verbreitet war. Die Fähigkeit der Pediococcus-Stämme zur Exopolysaccharid-Synthese wurde durch den Nachweis zweier Gene überprüft. Auf Basis der 23S rDNA-Sequenzen wurden rRNA-Sekundärstrukturen mit der neu entwickelten Software Structure Star generiert, die zum Auffinden von Zielbereichen für fluoreszenzmarkierte DNA-Sonden geeignet waren. Die Sequenzunterschiede zwischen den Pediococcus-Arten reichten aus, um zwei Gruppen durch Fluoreszenz in situ Hybridisierung differenzieren zu können. Die Verwendung unmarkierter Helfer-sonden verbesserte die Zugänglichkeit der Sonden an die rRNA, wodurch das Fluoreszenz-Signal verstärkt wurde. Um Milchsäurebakterien durch Denaturierende Gradienten Gel Elektrophorese differenzieren zu können, wurden Primer entwickelt, mit denen ein hochvariabler 23S rDNA-Bereich amplifiziert werden konnte. Die Nested Specifically Amplified Polymorphic DNA (nSAPD)-PCR wurde in der vorliegenden Arbeit zur Art- und Stamm-Differenzierung pro- und eukaryotischer Organismen angewandt. Es wurden vor allem weinrelevante Milchsäurebakterien der Gattungen Oenococcus, Lactobacillus, Pediococcus und Leuconostoc und Hefen der Gattungen Dekkera / Brettanomyces und Saccharomyces untersucht. Die Cluster-Analyse der Pediococcus-Typstämme führte zu einer unterschiedlichen Baum-Topologie im Vergleich zum phylogenetischen 23S rDNA-Stammbaum. Die Verwandtschaftsverhältnisse der untersuchten O. oeni-Stämme aus Starterkulturen konnten in Bezug auf eine frühere Cluster-Analyse reproduziert werden. Die Untersuchung von 40 B. bruxellensis-Stämmen aus rheinhessischen Weinproben zeigte eine Gruppierung der Stämme gemäß dem Ort der Probennahme. Beim Vergleich der Verwandtschaftsverhältnisse von Stämmen der Arten P. parvulus und B. bruxellensis, die aus denselben Weinproben isoliert wurden, konnte eine hohe Übereinstimmung der beiden Baum-Topologien beobachtet werden. Anhand der SAPD-PCR Untersuchung von Sekthefen aus Starterkulturen konnten alle Stämme der Art S. cerevisiae zugeordnet werden. Die nSAPD-PCR war darüber hinaus geeignet, um höhere Eukaryoten wie Weinreben zu differenzieren und es konnten die Verwandtschaftsverhältnisse von Mäusen und menschlichen Individuen durch Cluster-Analysen nachvollzogen werden. Mit Hilfe der Sequence Characterized Amplified Region (SCAR)-Technik wurden (n)SAPD-Marker in SCAR-Marker konvertiert. Die neu generierten SCAR-Primer konnten zur simultanen Art-Identifizierung von sieben weinschädlichen Milchsäurebakterien in einer Multiplex PCR erfolgreich eingesetzt werden. Die in dieser Arbeit entwickelten molekularbiologischen Identifizierungsmethoden können zum Beispiel in der mikrobiologischen Qualitätskontrolle Anwendung finden.

Im Rahmen dieser Arbeit wurde die Bindung von Koffein und verwandten Oxopurinen in C¬3-symmetrischen Rezeptoren auf der Basis von Triphenylenketalen untersucht. Dabei stand vor allem die Evaluierung für eine spätere Anwendung im Vordergrund. Für die Anwendung als Chemosensor wurden mehrere optische Verfahren getestet. Die Verwendung von UV/Vis-Spektroskopie gelingt nur unter Einsatz eines elektronenarmen Konkurrenzgastes, welcher durch das stärker bindende Koffein unter Entfärbung verdrängt wird. Obwohl dieser Effekt sogar mit bloßem Auge zu erkennen ist und somit eine einfache Untersuchung ermöglichen würde, machen die besondere Reaktivität des Konkurrenzgastes und dessen geringe Affinität zum Rezeptor eine weitere Anwendung als Chemosensor für Koffein unwahrscheinlich. Den entscheidenden Durchbruch lieferte der Wechsel auf Fluoreszenzspektroskopie. Die Bindung von Gästen lässt sich mit dieser Methode direkt beobachten und für quantitative Studien nutzen. Die Signalzunahme bei Zugabe von Koffein liegt bei maximal 30%. Durch Verwendung eines vom Koffein abgeleiteten Konkurrenzgastes können weitere Verbesserungen erzielt werden. So konnte eine maximale Signaldynamik von fast 400% erzielt werden. Durch die Entwicklung eines geeigneten Probenvorbereitungsprotokolls war es möglich, mit dem fluoreszenzbasierten System einen Nachweis von Koffein an kommerziell verfügbaren Getränkeproben durchzuführen. Die Ergebnisse waren in guter Übereinstimmung mit HPLC-Kontrollexperimenten. Die Eignung von Rezeptoren auf Triphenylenketalbasis für die enantiofaciale Differenzierung an Heteroaromaten wurde durch Untersuchung verschiedener Wirt-Gast-Komplexe mittels CD-Spektroskopie und Tieftemperatur-NMR systematisch demonstriert. Rezeptoren mit Menthyl-Substituenten liefern laut NMR die stärkste Seitendifferenzierung. Anhand des CD wird ein vollständiges und schlüssiges Bild über den Zusammenhang zwischen dem Raumbedarf am Gast, der Ausrichtung der chiralen Gruppen am Wirt und dem erhaltenen CD hergestellt. Durch umfangreiche molekulardynamische Simulationen und nachfolgende semiempirische Berechnungen wurden Referenzspektren berechnet, welche die Zuordnung der Stereochemie anhand des CD eindeutig belegen. Die Ergebnisse sind zudem in guter Übereinstimmung mit den Ergebnissen aus röntgenkristallographischen Untersuchungen. (Diese Methode ließ sich erfolgreich auf die helicale Faltung von Alkanen in Kapseln von Rebek, jr. umsetzen.) Obwohl die Energieunterschiede zwischen den diastereomeren Komplexen klein sind, konnte anhand der CD-Spektroskopie somit erstmalig die enantiofaciale Differenzierung an einem heterocyclischen System bei Raumtemperatur beobachtet werden. Die beste enantiofaciale Differenzierung erzielen die Menthyl-abgeleiteten Rezeptoren. Diese sind hinsichtlich einer möglichen Anwendung als chirales „Auxiliar“ ungeeignet, da sie mit den sperrigen Cyclohexylgruppen auch den Raum oberhalb des gebundenen Gastes blockieren. Daher wird für die weitere Entwicklung auf die praktische Einführung chiraler Information in Form des Isocyanats verzichten werden müssen. Stattdessen zielen aktuelle Bemühungen auf den Aufbau chiraler Rückgrate, welche den Raum in der unteren Peripherie des Gastes beeinflussen.

Shellac is the purified product of the natural polymer Lac. Shellac types, from different origins and with different ages, all purified by the solvent extraction process were compared in this study. Their physicochemical properties acid value, glass transition temperatures, color numbers and molecular sizes were determined. Metoprolol tartrate pellets were coated by air suspension coating with these different grades of shellac. Two coating levels 20% w/w and 25% w/w were applied and then subjected to in vitro dissolution testing. Enteric resistance was achieved for all tested brands for the two coating levels. At pH 6.8, 7.2 and 7.4, significant variations were obvious between the brands. rnMoreover the molecular size of shellac has a pronounced effect in that shellac types with larger molecular size show a higher and faster release than others, while the one with the smaller molecular size show the opposite effect on the release of metoprolol.rnIn this study commercially available ready for use aqueous shellac solutions (SSB AQUAGOLD), which are based on shellac SSB 57 (Dewaxed Orange Shellac, Bysakhi-Ber type refined in a solvent extraction process), with different manufacturing dates were used. rnTo improve the enteric coating properties of films from aqueous shellac solutions, different aqueous polymeric solutions of hydroxypropyl methylcellulose (HPMC), hydroxypropyl cellulose (HPC), carboyxmethyl cellulose (CMC), gum arabic and polysaccharides (Pullulan®) were used. These water soluble polymers will act as pore formers to enhance drug release from pellets coated with the combination of shellac and these polymers. The influence of these polymers on the gloss of the shellac films, mechanical properties of the films and drug release from metoprolol tartrate pellets were studied.rnThe potential of ethanol to alter the rate of drug release from shellac coated pellets was assessed by using a modified in vitro dose dumping in alcohol (DDA) method and the test concluded that shellac coated dosage forms can be co-administered with alcohol beverages containing ≤ 5% with no effect of alcohol on the shellac coat.rnPellets coated with shellac sodium salts, showed higher release rates than pellets coated with shellac as ammonium salt forms. rn

A global metabolic profiling methodology based on gas chromatography coupled to time-of-flight mass spectrometry (GC-TOFMS) for human plasma was applied to a human exercise study focused on the effects of beverages containing glucose, galactose, or fructose taken after exercise and throughout a recovery period of 6 h and 45 min. One group of 10 well trained male cyclists performed 3 experimental sessions on separate days (randomized, single center). After performing a standardized depletion protocol on a bicycle, subjects consumed one of three different beverages: maltodextrin (MD)+glucose (2:1 ratio), MD+galactose (2:1), and MD+fructose (2:1), consumed at an average of 1.25 g of carbohydrate (CHO) ingested per minute. Blood was taken straight after exercise and every 45 min within the recovery phase. With the resulting blood plasma, insulin, free fatty acid (FFA) profile, glucose, and GC-TOFMS global metabolic profiling measurements were performed. The resulting profiling data was able to match the results obtained from the other clinical measurements with the addition of being able to follow many different metabolites throughout the recovery period. The data quality was assessed, with all the labelled internal standards yielding values of <15% CV for all samples (n=335), apart from the labelled sucrose which gave a value of 15.19%. Differences between recovery treatments including the appearance of galactonic acid from the galactose based beverage were also highlighted.

In abstinence maintenance programs, for reissuing the driving licence and in workplace monitoring programs abstinence from ethanol and its proof are demanded. Various monitoring programs that mainly use ethyl glucuronide (EtG) as alcohol consumption marker have been established. To abstain from ethanol, but not from the taste of alcoholic beverages, in particular non-alcoholic beer has become more and more popular. In Germany, these "alcohol-free" beverages may still have an ethanol content of up to 0.5vol.% without the duty of declaration. Due to severe negative consequences resulting from positive EtG tests, a drinking experiment with 2.5L of non-alcoholic beer per person was performed to address the question of measurable concentrations of the direct metabolites EtG and EtS (ethyl sulphate) in urine and blood. Both alcohol consumption markers - determined by LC-MS/MS - were found in high concentrations: maximum concentrations in urine found in three volunteers were EtG 0.30-0.87mg/L and EtS 0.04-0.07mg/L, i.e., above the often applied cut-off value for the proof of abstinence of 0.1mg EtG/L. In the urine samples of one further volunteer, EtG and EtS concentrations cumulated over-night and reached up to 14.1mg/L EtG and 16.1mg/L EtS in the next morning's urine. Ethanol concentrations in blood and urine samples were negative (determined by HS-GC-FID and by an ADH-based method).

This study aims to show that sensitive detection of ethyl glucuronide in dried blood spotted onto various surfaces after a period of 24h is feasible. At present, there is insufficient information how tightly ethyl glucuronide (EtG) binds to various materials and how easily it can be eluted. 4ml aliquots of blood samples obtained from seven volunteers after consumption of alcoholic beverages were applied to six different surfaces. After drying and a 24h-storage at 20±2°C the samples were re-dissolved in water, and EtG was subsequently analyzed by a LC-MS Paul-type ion trap. A comparison was made between dried and corresponding fluid samples. EtG was detectable in all subjects' samples following consumption of alcohol. EtG was also detectable after a storage time of four weeks at 4°C in whole blood that had been preserved with EDTA. EtG was detectable in all samples dried on different surfaces and its concentration remained relatively constant irrespective of the particular condition of the material. Detection of EtG in blood spots from the scene may indicate recent alcohol consumption in cases where collection of blood remained undone or could not be performed.

OBJECTIVE: Craving for alcohol is probably involved in acquisition and maintenance of alcohol dependence to a substantial degree. However, the brain substrates and mechanisms that underlie alcohol craving await more detailed elucidation. METHOD: Positron emission tomography was used to map regional cerebral blood flow (CBF) in 21 detoxified patients with alcohol dependence during exposure to alcoholic and non-alcoholic beverages. RESULTS: During the alcohol condition compared with the control condition, significantly increased CBF was found in the ventral putamen. Additionally, activated areas included insula, dorsolateral prefrontal cortex and cerebellum. Cerebral blood flow increase in these regions was related to self-reports of craving assessed in the alcoholic patients. CONCLUSIONS: In this investigation, cue-induced alcohol craving was associated with activation of brain regions particularly involved in brain reward mechanisms, memory and attentional processes. These results are consistent with studies on craving for other addictive substances and may offer strategies for more elaborate studies on the neurobiology of addiction.

BACKGROUND: The term 'isolated extraordinary daytime urinary frequency' designates an abnormally increased diurnal frequency of painless urination in a completely toilet-trained child with normal urinalysis. METHODS: We report the history of 26 children (16 boys and 10 girls; aged between 4.1 and 10 years) who were referred to us between 2002 and 2006 and subsequently diagnosed with this condition. RESULTS: Possible psychosocial problems, or recent emotional stressors, were disclosed in the majority of the children: recent (36 months or less) asylum seekers (n = 9), school-related problems (n = 4), parental divorce (n = 2) or death of the mother (n = 1). Possible dietary causes were observed in 9 patients: oxalate-rich beverages (n = 5) and liberal ingestion of 'acidic' juices (n = 4). A diet low in oxalates was recommended when children were consuming large quantities of oxalate-rich beverages; and a diet low in acidic juice was recommended in those liberally ingesting acidic juices. Reassurance and observation were the approach in the remaining cases. The median duration of the symptoms was 5 months. A longer (p < 0.05) duration was noted in children of asylum seekers. CONCLUSIONS: This functional condition is easily identifiable, but often under- or misdiagnosed. Confounding the condition might result in redundant investigation.
