O presente estudo determina e analisa a importância relativa de diversos descritores ambientais, de pastoreio e de influência humana directa na distribuição da geneta (Genetta genetta L., 1758). O estudo decorreu em Monfurado, Sítio de Importância Comunitária, no Sul de Portugal. A área de estudo é predominantemente agrícola, maioritariamente ocupada por montado. A análise do padrão de distribuição deste carnívoro através de partição da variância e de ITMC (lnformation Theoretic Model Comparison) teve como objectivo orientar as prioridades na gestão das actividades humanas, compatíveis com a presença da espécie. Os resultados mostram que a distribuição da geneta é maioritariamente influenciada pelos descritores ambientais, apresentando uma relação positiva com densidade de montado com matos, conteúdo de matéria orgânica do solo e índice de Shannon de diversidade vertical da vegetação. A sua presença parece também ser promovida por níveis intermédios de pastoreio e fora das zonas de caça do regime cinegético especial. ABSTRACT; This study determines and analyses the relative importance of several environmental, livestock and human descriptors in the distribution of the common genet (Genetta genetta L., 1758). The study was conducted in Monfurado, a Site of Communitary lmportance, in South Portugal. The study area is Mediterranean farmland and is dominated by evergreen oak tree stands, named "montado". Modeling the distribution of this carnivore species was evaluated on the basis of ITMC (lnformation Theoretic Model Comparison) and variation partitioning techniques aiming to define human management activities compatible with the species conservation. The results show that the species distribution is mainly influenced by the environmental descriptors, and is positively related with montado and shrubs density, soil organic matter and Shannon's index of vertical vegetation diversity. Genet presence is favoured by intermediate levels of grazing and outside of game estates areas.

Os ecossistemas de água doce – responsáveis por funções ambientais importantes e pelo fornecimento de bens e serviços insubstituíveis – têm vindo a ser severamente afectados por perturbações antropogénicas. A conversão de floresta em terreno agrícola afecta os sistemas aquáticos através de uma série de mecanismos: sedimentação; excesso de nutrientes; contaminação; alterações hidrológicas; e remoção de vegetação ripícola. As comunidades de macroinvertebrados de água doce – devido à sua diversidade, ubiquidade e sensibilidade às perturbações ambientais – revelam-se como particularmente adequadas para estudos de avaliação da integridade ecológica destes sistemas expostos simultaneamente a múltiplos factores de impacto. O uso sistemático de respostas biológicas para avaliação de mudanças ambientais – ou biomonitorização – pode ser levado a cabo através de diversas metodologias, que, de uma forma geral, não consideram aspectos funcionais das comunidades biológicas e têm aplicabilidade geograficamente restrita. A biomonitorização através de atributos biológicos (características que reflectem a adaptação das espécies ao seu meio ambiente) revela-se como uma ferramenta promissora na resolução dos problemas referidos, apresentando vantagens adicionais: relações causa-efeito directas; melhoria na diferenciação de impactos; e integração da variabilidade natural. O presente estudo apresenta uma revisão critica do estado-da-arte actual na área do uso de atributos biológicos em biomonitorização. Até à data de publicação, não estava disponível nenhum outro trabalho com a base conceptual do uso de atributos de macroinvertebrados enquanto descritores de comunidades e para efeitos de biomonitorização e gestão de sistemas de água doce. Descrevem-se as teorias ecológicas de suporte destas metodologias (conceitos de habitat-molde e de filtros paisagísticos) e os estudos que aplicaram estas teorias em cenários reais, tendo-se chamado a atenção para questões técnicas e possíveis soluções. As necessidades futuras nesta área englobam: o desenvolvimento de uma só ferramenta de biomonitorização de aplicação alargada; uma maior compreensão da variabilidade natural nas comunidades biológicas; diminuição dos efeitos de soluções de compromisso biológico e sindromas; realização de estudos autoecológicos adicionais; e detecção de impactos específicos em cenários de impacto complexos. Um dos objectivos deste estudo foi contribuir para a melhoria das técnicas de biomonitorização através de atributos, focalizando em comunidades de macroinvertebrados ribeirinhas em diferentes regiões biogeográficas (as bacias hidrográficas dos rios: Little e Salmon em New Brunswick, Canadá; Anllóns na Galiza, Espanha; Reventazón em Cartago, Costa Rica). Em cada região, foram estudados gradientes de uso agrícola de solo, incluindo desde bacias hidrográficas quase exclusivamente cobertas por floresta até bacias sob a influência maioritária de actividades agrícolas intensivas. Em cada gradiente de uso de solo, a caracterização da comunidade biológica (por amostragem de macroinvertebrados em troços de rápidos) foi acompanhada pela caracterização do habitat circundante (incluindo propriedades da bacia hidrográfica, análise química das águas e outras propriedades à escala local). A comunidade de macroinvertebrados foi caracterizada através de informação taxonómica, métricas estruturais, índices de diversidade, métricas de tolerância, índices bióticos e através da compilação de atributos biológicos e fisiológicos gerais, de história de vida e de resistência a perturbações. Análises estatísticas univariadas e multivariadas foram usadas para evidenciar os gradientes biológicos e físico-químicos, confirmar a sua co-variação, testar a significância da discriminação de níveis de impacto e estabelecer comparações inter-regionais. A estrutura de comunidades revelou os complexos gradientes de impacto, que por sua vez co-variaram significativamente com os gradientes de uso de solo. Os gradientes de impacto relacionaram-se sobretudo com entrada de nutrientes e sedimentação. Os gradientes biológicos definidos pelas medidas estruturais seleccionadas co-variaram com os gradientes de impacto estudados, muito embora apenas algumas variáveis estruturais tenham individualmente discriminado as categorias de uso de solo definidas a priori. Não foi detectada consistência nas respostas das medidas estruturais entre regiões biogeográficas, tendo-se confirmadado que as interpretações puramente taxonómicas de impactos são difíceis de extrapolar entre regiões. Os gradientes biológicos definidos através dos atributos seleccionados também co-variaram com os gradientes de perturbação, tendo sido possível obter uma melhor discriminação de categorias de uso de solo. Nas diferentes regiões, a discriminação de locais mais impactados foi feita com base num conjunto similar de atributos, que inclui tamanho, voltinismo, técnicas reproductivas, microhabitat, preferências de corrente e substrato, hábitos alimentares e formas de resistência. Este conjunto poderá vir a ser usado para avaliar de forma predictiva os efeitos das modificações severas de uso de solo impostas pela actividade agrícola. Quando analisadas simultaneamente através dos atributos, as comunidades das três regiões permitiram uma moderada mas significativa discriminação de níveis de impacto. Estas análises corroboram as evidências de que as mudanças nas comunidades de macroinvertebrados aquáticos em locais sob a influência de agricultura intensiva podem seguir uma trajectória convergente no espaço multidimensional, independentemente de factores geográficos. Foram fornecidas pistas para a identificação de parâmetros específicos que deverão ser tidos em conta no planeamento de novos programas de biomonitorização com comunidades de macroinvertebrados bentónicos, para aplicação numa gestão fluvial verdadeiramente ecológica, nestas e noutras regiões. Foram ainda sugeridas possíveis linhas futuras de investigação.

The main goal of this study was to analyse the development of a pedagogy for autonomy tuned to the current situation in which Higher Education finds itself, namely of implementation of the Bologna Process in Europe, and in particular at the Escola Superior Agrária of the Polytechnic School at Coimbra. For this purpose new course units were created, new descriptors written, new materials developed and new methodologies tried. A case study was undertaken to observe and analyse the changes the institution was undergoing and to assess the impact of the new methodologies, particularly regarding the development of student autonomy. The target population for this study were the first year students on the Biotechnology degree in the year 2007/2008, to whom a battery of tests were applied, namely proficiency tests, belief tests, metacognitive tests, as well as methods of ethnographic research. The study relied on the European Framework of Reference for Languages and the European Language Portfolio to establish recognised levels and competences, and employed the portfolio as its main tool for measuring student autonomy. This portfolio was simultaneously biographical and descriptive and was organised by the students in their own time. Great incentive was given to this independent work. The ethnographic component of the study was of significant importance being given to students learning histories and to the collection of students’ reflections. This component aimed in particular to allow students the opportunity to express their thoughts on the experience they underwent during 20 weeks. From the analysis of the data, a significant improvement in self-confidence and motivation for lifelong learning is observable in the development of multiple tasks. Regarding the students’ progression towards proficiency, this improvement is less clear, but the final results of educational success were generally of great relevance.

Vitis vinifera L., the most widely cultivated fruit crop in the world, was the starting point for the development of this PhD thesis. This subject was exploited following on two actual trends: i) the development of rapid, simple, and high sensitive methodologies with minimal sample handling; and ii) the valuation of natural products as a source of compounds with potential health benefits. The target group of compounds under study were the volatile terpenoids (mono and sesquiterpenoids) and C13 norisoprenoids, since they may present biological impact, either from the sensorial point of view, as regards to the wine aroma, or by the beneficial properties for the human health. Two novel methodologies for quantification of C13 norisoprenoids in wines were developed. The first methodology, a rapid method, was based on the headspace solid-phase microextraction combined with gas chromatography-quadrupole mass spectrometry operating at selected ion monitoring mode (HS-SPME/GC-qMS-SIM), using GC conditions that allowed obtaining a C13 norisoprenoid volatile signature. It does not require any pre-treatment of the sample, and the C13 norisoprenoid composition of the wine was evaluated based on the chromatographic profile and specific m/z fragments, without complete chromatographic separation of its components. The second methodology, used as reference method, was based on the HS-SPME/GC-qMS-SIM, allowing the GC conditions for an adequate chromatographic resolution of wine components. For quantification purposes, external calibration curves were constructed with β-ionone, with regression coefficient (r2) of 0.9968 (RSD 12.51 %) and 0.9940 (RSD of 1.08 %) for the rapid method and for the reference method, respectively. Low detection limits (1.57 and 1.10 μg L-1) were observed. These methodologies were applied to seventeen white and red table wines. Two vitispirane isomers (158-1529 L-1) and 1,1,6-trimethyl-1,2-dihydronaphthalene (TDN) (6.42-39.45 μg L-1) were quantified. The data obtained for vitispirane isomers and TDN using the two methods were highly correlated (r2 of 0.9756 and 0.9630, respectively). A rapid methodology for the establishment of the varietal volatile profile of Vitis vinifera L. cv. 'Fernão-Pires' (FP) white wines by headspace solid-phase microextraction combined with comprehensive two-dimensional gas chromatography with time-of-flight mass spectrometry (HS-SPME/GCxGC-TOFMS) was developed. Monovarietal wines from different harvests, Appellations, and producers were analysed. The study was focused on the volatiles that seem to be significant to the varietal character, such as mono and sesquiterpenic compounds, and C13 norisoprenoids. Two-dimensional chromatographic spaces containing the varietal compounds using the m/z fragments 93, 121, 161, 175 and 204 were established as follows: 1tR = 255-575 s, 2tR = 0,424-1,840 s, for monoterpenoids, 1tR = 555-685 s, 2tR = 0,528-0,856 s, for C13 norisoprenoids, and 1tR = 695-950 s, 2tR = 0,520-0,960 s, for sesquiterpenic compounds. For the three chemical groups under study, from a total of 170 compounds, 45 were determined in all wines, allowing defining the "varietal volatile profile" of FP wine. Among these compounds, 15 were detected for the first time in FP wines. This study proposes a HS-SPME/GCxGC-TOFMS based methodology combined with classification-reference sample to be used for rapid assessment of varietal volatile profile of wines. This approach is very useful to eliminate the majority of the non-terpenic and non-C13 norisoprenic compounds, allowing the definition of a two-dimensional chromatographic space containing these compounds, simplifying the data compared to the original data, and reducing the time of analysis. The presence of sesquiterpenic compounds in Vitis vinifera L. related products, to which are assigned several biological properties, prompted us to investigate the antioxidant, antiproliferative and hepatoprotective activities of some sesquiterpenic compounds. Firstly, the antiradical capacity of trans,trans-farnesol, cis-nerolidol, α-humulene and guaiazulene was evaluated using chemical (DPPH• and hydroxyl radicals) and biological (Caco-2 cells) models. Guaiazulene (IC50= 0.73 mM) was the sesquiterpene with higher scavenger capacity against DPPH•, while trans,trans-farnesol (IC50= 1.81 mM) and cis-nerolidol (IC50= 1.48 mM) were more active towards hydroxyl radicals. All compounds, with the exception of α-humulene, at non-cytotoxic levels (≤ 1 mM), were able to protect Caco-2 cells from oxidative stress induced by tert-butyl hydroperoxide. The activity of the compounds under study was also evaluated as antiproliferative agents. Guaiazulene and cis-nerolidol were able to more effectively arrest the cell cycle in the S-phase than trans,trans-farnesol and α-humulene, being the last almost inactive. The relative hepatoprotection effect of fifteen sesquiterpenic compounds, presenting different chemical structures and commonly found in plants and plant-derived foods and beverages, was assessed. Endogenous lipid peroxidation and induced lipid peroxidation with tert-butyl hydroperoxide were evaluated in liver homogenates from Wistar rats. With the exception of α-humulene, all the sesquiterpenic compounds under study (1 mM) were effective in reducing the malonaldehyde levels in both endogenous and induced lipid peroxidation up to 35% and 70%, respectively. The developed 3D-QSAR models, relating the hepatoprotection activity with molecular properties, showed good fit (R2LOO > 0.819) with good prediction power (Q2 > 0.950 and SDEP < 2%) for both models. A network of effects associated with structural and chemical features of sesquiterpenic compounds such as shape, branching, symmetry, and presence of electronegative fragments, can modulate the hepatoprotective activity observed for these compounds. In conclusion, this study allowed the development of rapid and in-depth methods for the assessment of varietal volatile compounds that might have a positive impact on sensorial and health attributes related to Vitis vinifera L. These approaches can be extended to the analysis of other related food matrices, including grapes and musts, among others. In addition, the results of in vitro assays open a perspective for the promising use of the sesquiterpenic compounds, with similar chemical structures such as those studied in the present work, as antioxidants, hepatoprotective and antiproliferative agents, which meets the current challenges related to diseases of modern civilization.

Estuaries are poles of attraction for human settlement which is a source of pressures to surface water bodies. The implementation of the European Water Framework Directive (WDF, 2000/60/EC) has increased the investigation in order to develop methodologies to assess the Ecological Quality Status (EQS) of aquatic ecosystems. Transitional systems are naturally stressed and characterized by highly dynamic physical, chemical and hydro-morphologic conditions and by species with a higher level of tolerance to change, being more difficult to develop suitable quality indicators for these systems. The general purpose of this study is to test the ability of synthesis descriptors, including primary (S, taxa richness) and derived biological variable (H’, Shannon-Wiener diversity), biotic indices (AMBI and M-AMBI), body size properties (abundance distribution by body size classes, length, weight and length-weight relationships) and non-taxonomic indices (ISS), as well as functional indicators related to the decomposition rates of various experimental substrates, a macrophyte (Phragmites australis) and an alga (Fucus vesiculosus), to evaluate the environmental quality in transitional systems. This study was carried out in one of the most pristine channels of the Ria the Aveiro, Mira Channel, along a full salinity gradient and in a metals and metalloid sediment contamination area, the Estarreja Channel, and two reference channels (Canelas and Salreu). In this study were used different sampling techniques, the leaf-bag technique and a hand-held corer. In Mira Channel, the alga and the macrophyte presented an opposite trend in the decomposition rate along the salinity gradient, with the decomposition rates of the alga always higher than those of the macrophyte. The decomposition rates of the macrophyte and the alga were higher in the mid estuary and in higher salinity areas, respectively, corresponding to the preferencial distribution areas of each species. The macrobenthic fauna associated with the decaying and an artificial substrate (control) showed equally well the benthic succession from the marine to the freshwater areas and, despite the strong differences in the decay rates, no significant differences were found between the benthic communities associated with the alga and the macrophyte. The body size properties of the macrobenthic fauna associated with the P. australis leaf-bag (1mm and 5mm) and corer samples were studied along the full salinity gradient. The dominant species of the sub-set of measured specimens were not the same of the original macrobenthic fauna sampled but, despite that, the sub-set of measured specimens was also able to show the benthic succession from the marine to the freshwater areas. The body size abundance distribution of the benthic macroinvertebrates according to the ISS size classes did not show a particular trend in any sampler along the salinity gradient. Significant differences were found in the length, weight and length-weight relationships of Annelids, , Molluscs and even some species along the salinity gradient. No significant differences were found in the AMBI, M-AMBI and ISS values along the salinity gradient for all the samplers. The EQS of the corer samples obtained using the M-AMBI was lower than that of the leaf-bags. The EQS obtained with the ISS was higher than that obtained with the M-AMBI in the leaf-bags but not in the corer samples. The ecological effects of contaminated sediments associated with the industrial chemical effluents discharged in the Estarreja Channel were studied a decade after ceasing the emissions, using the Sediment Quality Triad approach and two reference channels. The results showed that despite the emissions ceased in 2004, the sediment remains polluted with high levels of metals and metalloid, available to bioaccumulation and with severe consequences at the community level. The sediment contamination problem was also studied using the leaf-bag technique with a macrophyte, an alga and a control substrate. The results showed that the decay rates, the associated macrofauna and the application of the AMBI, M-AMBI and ISS indices to the mesh-bag samples were not able to identify the sediment contamination. Contrarily to the AMBI, the M-AMBI and the ISS showed significant differences between the contaminated and the reference channels for the corer samples. Although such statistical significance, the interest of using these complex biotic indices could be questioned, when much simple ones, like the S and H’ allow to reach the same conclusions.

The purpose of this research is to develop and validate a measurement scale to assess golf destinations’ brand personality and therefore to perceive the destination personality of the Algarve as a golf destination. Based on literature review on human personality, brand personality, destination brand image and marketing scales validation procedures, an initial 36 unrepeated items were the base for a survey instrument. Those items were generated from the literature, from the results of individual interviews with experts in tourism and golf in the Algarve and from promotional texts in golf- related websites. After content validation, the items were allocated into categories of attributes by a panel of expert judges. A survey was then applied to a convenient sample of 600 golf players in the Algarve, and 545 (valid) questionnaires were analysed to refine the scale. Golf players assessed the components of the relational brand personality (functional, symbolic and experiential) as well as the Algarve as a golf destination. A taxonomy of brand personality was developed and tested in the Algarve as it is recognized as one of the world best golf destination. The developed taxonomy of brand personality was assessed in two ways: 1) through the overall perception of the Algarve as a golf destination and 2) through the perception of specific attributes of the destination grouped into three main categories (functional, symbolic and experiential). Therefore, two multi-dimensional brand personality models were estimated by using structural equation modelling. Findings of this study indicate that golf players ascribe personality characteristics to destinations. The brand personality of the Algarve is translated into three main dimensions enjoyableness, distinctiveness and friendliness when tourists/golf players reveal their overall perception of the destination. The brand personality of golf destination Algarve is reflected in the dimensions reliability, hospitality, uniqueness and attractiveness when tourists assess the components of the relational brand personality. Refined scales consisting of 10 and 12 items were finally derived meeting both reliability and validity requirements. This study does not replicate Aaker’s (1997) personality dimensions and very little parallelism can be drawn with Aaker’s (1997) brand personality scale since only three items from her scale were validated in both models: friendly and cheerful, (sincerity), reliable (competence). The same is verified concerning the ‘Big-five’. The human personality traits (HPT) validated to describe golf destinations personality are only four helpful, pleasant (agreeableness), relaxed (emotional stability), and innovative (intellect or openness). As far as destination image descriptors (DID) are concerned, the items appealing, relaxed and safe were validated, while traits suggested by the interviews and website promotional texts such as calm, natural, spectacular, unique, welcoming, and the best (destination-specific traits) appear to be appropriate to describe the personality of a golf destination. The results suggest that the overall perception of the Algarve´s brand personality is described by the dimensions enjoyableness, distinctiveness and friendliness. Moreover, the relational perspective revealed that the functional attributes of the destination are described by the dimension reliablility, while the symbolic attributes are described by the dimensions hospitablility and uniqueness and finally its experiential attributes are described by the dimension attractiveness. These results show that a golf destination´s brand personality should not just be based on good golf practices. Theoretical and practical implications are discussed in the context of destination brand personality.
