Aims Obesity and asthma are widely prevalent and associated disorders. Recent studies of our group revealed that Substance P (SP) is involved in pathophysiology of obese-asthma phenotype in mice through its selective NK1 receptor (NK1-R). Lymphangiogenesis is impaired in asthma and obesity, and SP activates contractile and inflammatory pathways in lymphatics. Our aim was to study whether NK1-R expression was involved in lymphangiogenesis on visceral (VAT) and subcutaneous (SAT) adipose tissues and in the lungs, in obese-allergen sensitized mice. Main methods Diet-induced obese and ovalbumin (OVA)-sensitized Balb/c mice were treated with a selective NK1-R antagonist (CJ 12,255, Pfizer Inc., USA) or placebo. Lymphatic structures (LYVE-1 +) and NK1-R expression were analyzed by immunohistochemistry. A semi-quantitative score methodology was used for NK1-R expression. Key findings Obesity and allergen-sensitization together increased the number of LYVE-1 + lymphatics in VAT and decreased it in SAT and lungs. NK1-R was mainly expressed on adipocyte membranes of VAT, blood vessel areas of SAT, and in lung epithelium. Obesity and allergen-sensitization combined increased the expression of NK1-R in VAT, SAT and lungs. NK1-R antagonist treatment reversed the effects observed in lymphangiogenesis in those tissues. Significance The obese-asthma phenotype in mice is accompanied by increased expression of NK1-R on adipose tissues and lung epithelium, reflecting that SP released during inflammation may act directly on these tissues. Blocking NK1-R affects lymphangiogenesis, implying a role of SP, with opposite physiological consequences in VAT, and in SAT and lungs. Our results provide a clue for a novel SP role in the obese-asthma phenotype.

A eliminação da prática da tradução no ensino de línguas estrangeiras, aliada ao conhecimento por vezes insuficiente da língua-fonte por parte dos alunos de tradução, faz com que estes se concentrem excessivamente no signo linguístico e que o texto de chegada produzido seja o resultado de uma operação quase mecânica de substituição lexical. Numa tentativa de evitar fórmulas prescritivas de ensino unidireccional professor/aluno e favorecendo uma perspectiva de construção de significado através da interacção com o texto, propõe-se uma actividade simples em que se passa do nível do nível da palavra e da frase ao nível do texto como um todo. Tanto pela sua própria natureza como pela sua abundância no par SP-PT, os falsos cognatos oferecem uma boa oportunidade para pôr de manifesto a necessidade de utilizar procedimentos que considerem o texto como a unidade de tradução.

This study performs a sustainability evaluation of biodiesel from microalga Chlamydomonas sp. grown in 20 % (v/v) of brewery’s wastewater, blended with pentose sugars (xylose, arabinose or ribose resulting from the hydrolysis of brewer’s spent grains (BSG). The life cycle steps considered for the study are: microalgae cultivation, biomass processing and lipids extraction at the brewery site, and its conversion to biodiesel at a dedicated external biofuel’s plant. Three sustainability indicators (LCEE, FER and GW) were considered and calculated using experimental data. Literature data was used, whenever necessary, to complement life cycle data, thus allowing a more accurate sustainability evaluation. A comparative analysis of the biodiesel life cycle steps was also conducted, with the main goal of identifying which steps need to be improved. Results show that biomass processing, especially cell harvesting, microalgae cultivation, and lipids extraction are the main process bottlenecks. It is also analysed the influence on the microalgae biodiesel sustainability of adding each pentose sugar to the cultivation media, concluding that it strongly influences the biomass and lipid productivity. In particular, the addition of xylose is preferable in terms of lipid productivity, but from a sustainability point of view, ribose is the best, though the difference from xylose is not significant. Nevertheless, culture without pentose addition presents the best sustainability results.

A nova ponte pedonal sobre o Rio Ave foi recentemente construída em Santo Tirso, no âmbito do projecto de requalificação da frente de rio da autarquia. Trata-se de uma estrutura em arco metálico, muito abatido, com 60m de corda central e 6m de flecha, e com extensão total de 84m. Tendo esta obra sido projectada pelo autor, o presente trabalho tem como objectivo descrever as sucessivas fases do estudo do seu comportamento dinâmico, com especial atenção às vibrações induzidas pela passagem de peões em corrida e ao desenvolvimento de um sistema de controlo de vibrações, composto por amortecedores de massas sintonizadas (TMD’s), que foi instalado na estrutura. Neste contexto, numa fase inicial são discutidos diversos aspectos sobre o comportamento dinâmico de pontes sob a acção pedonal, nomeadamente a caracterização experimental da referida acção, modelos de carga a aplicar em projecto e limites de conforto a respeitar no âmbito das recomendações internacionais actualmente existentes. São também apresentados sucintamente os sistemas de controlo de vibrações mais comuns em pontes pedonais, detalhando-se o funcionamento e princípios de dimensionamento dos TMD’s. Em seguida é apresentado o projecto da nova ponte pedonal sobre o Rio Ave, descrevendo-se a solução estrutural e os estudos dinâmicos levados a cabo em fase de projecto. São posteriormente apresentados e discutidos os resultados das campanhas de ensaios dinâmicos, realizados após o final da construção, que permitiram confirmar as conclusões obtidas em fase de projecto, assim como verificar a eficiência do sistema de controlo de vibrações dimensionado. O trabalho termina com a apresentação das principais conclusões e também com um conjunto de ideias que permitem perspectivar novos tópicos de interesse em termos de investigação na continuidade do trabalho realizado.

A plant growth-promoting bacterial (PGPB) strain SC2b was isolated from the rhizosphere of Sedum plumbizincicola grown in lead (Pb)/zinc (Zn) mine soils and characterized as Bacillus sp. based on (1) morphological and biochemical characteristics and (2) partial 16S ribosomal DNA sequencing analysis. Strain SC2b exhibited high levels of resistance to cadmium (Cd) (300 mg/L), Zn (730 mg/L), and Pb (1400 mg/L). This strain also showed various plant growth-promoting (PGP) features such as utilization of 1-aminocyclopropane-1-carboxylate, solubilization of phosphate, and production of indole-3-acetic acid and siderophore. The strain mobilized high concentration of heavy metals from soils and exhibited different biosorption capacity toward the tested metal ions. Strain SC2b was further assessed for PGP activity by phytagar assay with a model plant Brassica napus. Inoculation of SC2b increased the biomass and vigor index of B. napus. Considering such potential, a pot experiment was conducted to assess the effects of inoculating the metal-resistant PGPB SC2b on growth and uptake of Cd, Zn and Pb by S. plumbizincicola in metal-contaminated agricultural soils. Inoculation with SC2b elevated the shoot and root biomass and leaf chlorophyll content of S. plumbizincicola. Similarly, plants inoculated with SC2b demonstrated markedly higher Cd and Zn accumulation in the root and shoot system, indicating that SC2b enhanced Cd and Zn uptake by S. plumbizincicola through metal mobilization or plant-microbial mediated changes in chemical or biological soil properties. Data demonstrated that the PGPB Bacillus sp. SC2b might serve as a future biofertilizer and an effective metal mobilizing bioinoculant for rhizoremediation of metal polluted soils.

The phenolic profile and antioxidant activity of three endemic Laurus sp. from Portugal were analysed. Dried leaves of L. nobilis L., L. azorica (Seub.) Franco, and L. novocanariensis Rivas Mart., Lousã, Fern. Prieto, E. Días, J. C. Costa & C. Aguiar, collected in the mainland and in the Azores and Madeira archipelagos, respectively, were used to prepare different extracts (aqueous, ethanolic and hydroalcoholic). They were studied regarding their DPPH˙ scavenging activity, total phenolic and flavonoid contents, and the main phenolic compounds were identified by HPLC-DAD-ESI-MS/MS. Total flavonoid contents were 30.1, 46.3, and 36.7 mg of epicatechin equivalents per g of sample (dry weight) for L. nobilis, L. azorica and L. novocanariensis, respectively. Epicatechin was the major compound, representing ∼12.1% of total flavan-3-ols in L. nobilis, ∼25.6% in L. azorica, and ∼19.9% in L. novocanariensis. Although all samples presented a similar phenolic profile, significant differences were observed in their total contents and antioxidant activity.
