Este trabalho propõe o desenvolvimento de um procedimento analítico para a determinação em linha de estanho em soluções analíticas empregando a técnica de espectrometria de absorção atômica com nebulização térmica em tubo metálico aquecido na chama (TS-FF-AAS) como alternativa ao uso da chama de óxido nitroso-acetileno em espectrometria de absorção atômica, cujo comburente apresenta um custo elevado frente ao ar comprimido. Foram avaliados parâmetros para a otimização do sistema, tais como: vazão do carregador (ar), volume de amostra injetada e concentração do ácido (HCl). Foi construída curva analítica linear [A= -0,00163 + 0,00319 [Sn], (r= 0,9998)] no intervalo de 10 a 80 mg Sn L-1. O limite de detecção, o desvio padrão relativo (n=12) e a freqüência analítica foram: 1,7 mg L-1 Sn, £ 2,7% and 120 h-1 respectivamente. O sistema TS-FF-AAS é uma alternativa para a determinação de Sn.

This work describes the development of an analytical procedure for on-line tin determination using thermospray flame furnace atomic absorption spectrometry (TS-FF-AAS). Two tubes were evaluated as atomization cells: a metallic tube (Ni-Cr, principal components composition: 73.95% Ni and 16.05% Cr) and a ceramic tube (99.8% Al2O3). The use of air as the carrier was made by employing a Rheodyne valve to inject the samples, allowing an analytical frequency of 90 h(-1) and avoiding sample dispersion. The carrier flow rate (air), sample volume injected, and acid concentration (HCl) were evaluated for the optimization of the TS-FF-AAS system. The sensitivity for 50 mL of analytical solution with TS-FF-AAS was 2 and 5 times higher (to metallic and ceramic tube, respectively) than using an acetylene-nitrous oxide flame with pneumatic aspiration (requiring a sample volume of approximately 20 times higher.

Thermospray flame furnace Atomic Absorption Spectrometry (TS-FF-AAS) was used for the total determination of Cd, Pb and Zn in fresh water and seawater samples at µg L-1 levels, and in marine sediment samples at µg g-1 levels. Using a sample loop of 50 µL and a peristaltic pump the samples were transported into the metallic tube placed over an air/acetylene flame, through a ceramic capillary (o.d. = 3.2 mm) containing two parallel internal orifices (i.d = 0.5 mm). The detection limits determined for Cd, Pb and Zn using a synthetic water matrix (2.5% m/v NaCl, 0.5% m/v MgCl2 and 0.8% m/v CaCl2) were 0.32 µg L-1; 2.6 µg L-1 and 0.21 µg L-1 respectively. The methodology by TS-FF-AAS was validated by determination of Cd, Pb and Zn in certified reference materials of water and marine sediment, and the t-test for differences between means was applied. No statistically significant differences were established in fresh water and seawater (p>0.05), whereas differences became apparent in marine sediment (p<0.03).

Jundiaí river is the main stream in the Jundiaí Hydrographic Basin and its water is not considered safe for public supply. The water problem is getting worse with the development of the region. The Pirai stream, have been one of the last potable water resource. With the purpose of investigating the sediment quality, due to its influence on the water quality, bioavailable and total Cd and Pb were determined using TS-FF-AAS. Total Cd and Pb up to 2.47 and 24.7 µg g-1 were measured, respectively. The wastewater sludge showed concentrations of 4.01 and 171 µg g-1, for the same metals.

A flow injection on-line pre-concentration system coupled to thermospray flame furnace atomic absorption spectrometry (TS-FF-AAS) for cadmium determination at sub μg L-1 levels in seawater samples was developed. The on-line system was evaluated by analysing cadmium containing in a synthetic seawater matrix (2.5% m/v NaCl, 0.5% m/v MgCl2 and 0.8% m/v CaCl2). A sample volume of 2 mL allows determining Cd with a detection limits of 30 ng L-1 (3* σblank/slope), pre-concentration factor of 34 and repeatability of 1,8% (calculated as RSD, N=8 and containing 200 ng L-1 of Cd ).

Palladium plus magnesium nitrates with and without Ir, Ru and W were evaluated for the simultaneous determination of As, Cu and Pb in cachaca by graphite furnace atomic absorption spectrometry. For 20 mu L sample, 5 mu L Pd(NO(3))(2) and 3 mu L Mg(NO(3))(2) dispensed together onto the Ir-coated platform of the THGA, analytical curves in the 0-30.0 mu g L(-1) As, 0-1.50 mg L(-1) Cu and 0-60.0 mu g L(-1) Pb were built up and typical linear correlation coefficients were always better than 0.999. The limit of detection was 1.30 mu g L(-1) As, 140 mu g L(-1) Cu and 0.90 mu g L(-1) Pb. As, Cu and Pb contents in 10 cachaca samples agreed with those obtained by ICP-MS. Recoveries of spiked samples varied from 96% to 106% (As), 97% to 112% (Cu) and 92% to 108% (Pb). The relative standard deviation (n = 12) was typically 2.7%, 3.3% and 1.9%. (C) 2008 Elsevier Ltd. Mobile Lipids detected using H-1-NMR in stimulated lymphocytes were correlated with cell cycle phase, expression of the interleukin-2 receptor alpha and proliferation to assess the activation status of the lymphocytes. Mobile lipid levels, IL-2R alpha expression and proliferation increased after treatment with PMA and ionomycin. PMA or ionomycin stimulation alone induced increased IL-2R alpha expressiom but not proliferation, PMA- but not ionomycin-stimulation generated mobile lipid, Treatment with anti-CD3 antibody did not increase IL-2R alpha expression or proliferation but did generate increased amounts of mobile lipid, The cell cycle status of thymocytes treated with anti-CD3, PMA or ionomycin alone indicated an. accumulation of the cells in the G(1) phase of the cell cycle, The generation of mobile lipid was abrogated in anti-CD3 antibody-stimulated thymic lymphocytes but not in splenic lymphocytes, using a phosphatidylcholine-specific phospholipase C (PC-PLC) inhibitor which blocked cells in the G(1)/S phase of the cell cycle, This suggests that the H-1-NMR-detectable mobile Lipid may be generated in anti-CD3 antibody-stimulated thymic lymphocytes by the action of PC-PLC activity via the catabolism of PC, in the absence of classical signs of activation. O mercúrio é um elemento que ainda necessita constante monitoramento devido sua capacidade tóxica em concentrações em níveis de traço com possibilidade de contaminação e exposição por uma variedade de formas e compostos presentes no ambiente. Esse estudo possuiu como objetivo a obtenção de um procedimento simplificado e eficiente para a determinação de teores de mercúrio em tecidos de rato Wistar a partir da otimização de um método de determinação, utilizando um sistema de geração de vapor frio acoplado a espectrometria de absorção atômica com forno de grafite (CV-GF AAS). Houve a comparação entre métodos de digestão, comparação de métodos de amalgamação, otimização, verificação do método a partir da obtenção de figuras de mérito e análise de amostras de fígado e rins de ratos tratados com exposição crônica ao mercúrio. Os resultados comprovaram que o método de digestão em bloco digestor, a utilização de amalgamação por recobrimento de tubo em ouro em conjunto com rede de ouro e adição pré-análise à solução de simeticona e isopropanol apresentou os melhores resultados, com teores de recuperação médios entre 92 e 114 %, viabilizando a utilização do procedimento proposto.

E10-5A3 is a dhfr-ts- Leishmania major double knockout auxotrophic shown previously to induce substantial protection against virulent L. major infection in both genetically susceptible and resistant mice. We investigated the capacity of dhfr-ts- to protect against heterologous infection by L. amazonensis. The degree of protection was evaluated by immunization of BALB/c or C57BL/6 mice with E10-5A3, followed by L. amazonensis challenge. Whether immunized by subcutaneous (SC) or intravenous (IV) inoculation, susceptible and resistant mice displayed a partial degree of protection against challenge with virulent L. amazonensis. SC-immunized BALB/c mice developed lesions 40 to 65% smaller than non immunized mice, while IV immunization led to protection ranging from 40 to 75% in four out of six experiments compared to non immunized animals. The resistant C57BL/6 mice displayed comparable degrees of protection, 57% by SC and 49% by IV immunization. Results are encouraging as it has been previously difficult to obtain protection by SC vaccination against Leishmania, the preferred route for human immunization.
