Four new piperidine alkaloids, leptophyllin A (4), 3-acetylleptophyllin A (5), leptophyllin B (8), and (-)-spectaline (1), and 3 known piperidine alkaloids, (-)-spectalinine (2), canavaline (3), and iso-6-canavaline (7), were isolated by a bioassay-guided fractionation of a leaf extract of Cassia leptophylla. The alkaloids 1 - 3 were active in a mechanism-based DNA-modifying yeast assay and 2 was moderately active in Vero monkey and Chinese hamster ovary cell cytotoxicity assays.

The antinociceptive activity of (-)-spectaline (1), a piperidine alkaloid isolated from Cassia leptophylla Vog. (Leguminosae), was investigated. We have also studied the acute oral toxicity of 1 in mice and it did not show any signals of toxicity in doses lower than 400 mumol/kg. The antinociceptive effect of 1 was evaluated on chemical (acetic acid, formalin and capsaicin) and thermal (hot plate and tail flick) pain models in mice, using classical standard drugs. Dipyrone ID50 = 14.68 mumol/kg (4.8 mg/kg), in-domethacin ID50 = 0.78 mumol/kg (0.28 mg/kg) and (-)-spectaline ID50 = 48.49 mumol/kg (15.75 mg/kg), all produced a significant inhibition of acetic acid-induced abdominal writhing in mice. (-)-Spectaline was inactive in the hyperalgesic model of formalin and did not show any central analgesic activity (hot plate and tail flick models). In the capsaicin-induced neurogenic pain model, (-)-spectaline presented an important inhibitory effect with an ID50 = 20.81 mug/paw and dipyrone ID50 = 19.89 mug/ paw. The ensemble of results permitted us to identify 1 as an antinociceptive compound. The mechanism underlying this antinociceptive effect of 1 remains unknown, but the results suggest that such an effect could be related to pathways associated to vanilloid receptor systems.

A subfamília Caesalpinioideae (Leguminosae) possui cerca de 2.800 espécies, muitas das quais ocorrem no Brasil. Para a região Sul do Brasil são citadas 56 espécies, distribuídas pelos mais diversos ambientes, com importância econômica, social e científica bastante grande, sendo ainda pouco conhecidas do ponto de vista citogenético e taxonômico. Neste estudo foram analisados, quanto ao número de cromossomos, 74 acessos de 27 táxons incluídos em 10 gêneros pertencentes às tribos Cassieae, Caesalpinieae e Cercideae. Os números cromossômicos encontrados foram 2n = 32, 28, 26, 24, 22, 16 e 14. Sete espécies tiveram seus números cromossômicos determinados pela primeira vez: Cassia leptophylla, Senna araucarietorum, S. hilariana, S. neglecta, S. oblongifolia, Chamaecrista repens e Pomaria stipularis. A maioria das espécies apresentaram 2n = 28 cromossomos, sendo observados também 2n = 26, 24 e 22. O gênero Chamaecrista diferenciase dos demais gêneros, pois todos os seus táxons apresentaram 2n = 32, 16 e 14 cromossomos, sendo o primeiro número supostamente originado por poliploidia. O número básico proposto para as espécies estudadas foi x = 14, com os demais números, x = 13, 12 e 11, tendo surgido provavelmente por disploidia e para o gênero Chamaecrista x = 8 e x = 7 para a espécie pertencente à seção Xerocalyx. A poliploidia pareceu importante na diversificação inicial do grupo, com ocorrência de uma série de reduções displóides no decorrer do processo evolutivo. O caráter número de cromossomos mostrou-se relevante na distinção de táxons do gênero Chamaecrista dos demais gêneros, sugerindo, juntamente com outros caracteres analisados e encontrados em literatura, a segregação deste dos demais gêneros pertencentes à tribo Cassieae.

Bioactivity-guided fractionation of several bioactive extracts obtained from Cerrado and Atlantic Forest plant species led to the isolation of potent DNA-damaging piperidine 1-5 and guanidine alkaloids 6-9 from Cassia leptophylla and Pterogyne nitens respectively, two common Leguminosae from Atlantic Forest. By means of biotechnological approach on Maytenus aquifolium, a species from Cerrado, moderate DNA-damaging sesquiterpene pyridine alkaloid 10-11 was isolated. Bioassay-guided fractionation on Casearia sylvestris, a medicinal plant species found in Cerrado and Atlantic Forest, led to the isolation of clerodane diterpenes 12-13 which showed effect on DNA. In addition, we have reported several interesting potent antifungal iridoids: 1β-hydroxy-dihydrocornin (14), 1α-hydroxy-dihydrocornin (15), α-gardiol (16), β-gardiol (17), plumericin (18), isoplumericin (19), 11-O-trans-caffeoylteucrein (20); ester derivative: 2-methyl-4-hydroxy-butyl-caffeoate (21), amide N-[7-(3'.4'-methylenedioxyphenyl)-2Z, 4Z-heptadienoyl] pyrrolidine (22) and triterpene viburgenin (23).

As part of an ongoing research project on Senna and Cassia species, five new pyridine alkaloids, namely, 12'-hydroxy-7'-multijuguinol (1), 12'-hydroxy-8'-multijuguinol (2), methyl multijuguinate (3), 7'-multijuguinol (4), and 8'-multijuguinol (5), were isolated from the leaves of Senna multijuga (syn. Cassia multijuga). Their structures were elucidated on the basis of spectroscopic data analysis. Mass spectrometry was used for confirmation of the positions of the hydroxy groups in the side-chains of 1, 2, 4, and 5. All compounds exhibited weak in vitro acetylcholinesterase inhibitory activity as compared with the standard compound physostigmine.

Neogene climates and vegetation history of western Yunnan are reconstructed on the basis of known fossil plants using the Coexistence Approach (CA) and Leaf Margin Analysis (LMA). Four Neogene leaf floras from Tengchong, Jianchuan and Eryuan in southwestern China are analyzed by the CA, and the paleoclimatic data of one Miocene carpoflora from Longling and three Pliocene palynofloras from Longling, Yangyi and Eryuan are used for comparison. The Miocene vegetation of the whole of West Yunnan is subtropical evergreen broad-leaved forest, and a similar mean annual precipitation is inferred for Tengchong, Longling and Jianchuan. However, by the Late Pliocene a large difference in vegetation occurred between the two slopes of Gaoligong Mountain, western Yunnan. The region of Tengchong retained a subtropical evergreen broad-leaved forest vegetation, whereas in Yangyi and Eryuan a vertical vegetation zonation had developed, which consists, in ascending order, of humid evergreen broad-leaved, needle and broad-leaved mixed evergreen, and coniferous forests. Distinctively, the Late Pliocene vegetational patterns of West Yunnan were already very similar to those of the present, and the Pliocene mean annual precipitation in Tengchong was markedly higher than that of Yangyi and Eryuan. Considering that the overall vegetation of West Yunnan and the precipitation at Yangyi and Eryuan have undergone no distinct change since the Late Pliocene, we conclude that the Hengduan Mountains on the northern boundary of West Yunnan must have arisen after the Miocene and approached their highest elevation before the Late Pliocene. Furthermore, the fact of the eastern portion of the Tibetan Plateau underwent a slight uplift after the Late Pliocene is also supported.
