Polybrominated diphenyl ethers (PBDEs) are a class of brominated flame retardants (BFRs) that have been heavily used in consumer products such as furniture foams, plastics, and textiles since the mid-1970’s. BFRs are added to products in order to meet state flammability standards intended to increase indoor safety in the event of a fire. The three commercial PBDE mixtures, Penta-, Octa-, and DecaBDE, have all been banned in the United States, however, limited use of DecaBDE is still permitted. PBDEs were phased out of production and added to the Stockholm Convention due to concerns over their environmental persistence and toxicity. Human exposure to PBDEs occurs primarily through the inadvertent ingestion of contaminated house dust, as well as though dietary sources. Despite the phase-out and discontinued use of PBDEs, human exposure to this class of chemicals is likely to continue for decades due to the continued use of treated products and existing environmental reservoirs of PBDEs. Extensive research over the years has shown that PBDEs disrupt thyroid hormone (TH) levels and neurodevelopmental endpoints in rodent and fish models. Additionally, there is growing epidemiological evidence linking PBDE exposure in humans to altered TH homeostasis and neurodevelopmental impairments in children. Due to the importance of THs throughout gestation, there is a great need to understand the effects of BFRs on the developing fetus. Specifically, the placenta plays a critical role in the transport, metabolism, and delivery of THs to the fetal compartment during pregnancy and is a likely target for BFR bioaccumulation and endocrine disruption. The central hypothesis of this dissertation research is that BFRs disrupt the activity of TH sulfotransferase (SULT) enzymes, thereby altering TH concentrations in the placenta.

In the first aim of this dissertation research, the concentrations of PBDEs and 2,4,6-TBP were measured in a cohort of 102 placenta tissue samples from an ongoing pregnancy cohort in Durham, NC. Methods were developed for the extraction and analysis of the BFR analytes. It was found that 2,4,6-TBP was significantly correlated with all PBDE analytes, indicating that 2,4,6-TBP may share common product applications with PBDEs or that 2,4,6-TBP is a metabolite of PBDE compounds. Additionally, this was the first study to measure 2,4,6-TBP in human placenta tissues.

In the second aim of this dissertation research, the placenta tissue concentrations of THs, as well as the endogenous activity of deiodinase (DI) and TH SULT enzymes were quantified using the same cohort of 102 placenta tissue samples. Enzyme activity was detected in all samples and this was the first study to measure TH DI and SULT activity in human placenta tissues. Enzyme activities and TH concentrations were compared with BFR concentrations measured in Aim 1. There were few statistically significant associations observed for the combined data, however, upon stratifying the data set based on infant sex, additional significant associations were observed. For example, among males, those with the highest concentrations of BDE-99 in placenta had T3 levels 0.80 times those with the lowest concentration of BDE-99 (95% confidence interval (CI): 0.59, 1.07). Whereas females with the highest concentrations of BDE-99 in placenta had T3 levels 1.50 times those with the lowest concentration of BDE-99 (95% CI: 1.10, 2.04). Additionally, all BFR analyte concentrations were higher in the placenta of males versus females and they were significantly higher for 2,4,6-TBP and BDE-209. 3,3’-T2 SULT activity was significantly higher in female placenta tissues, while type 3 DI activity was significantly higher in male placenta tissues. This research is the first to show sex-specific differences in the bioaccumulation of BFRs in human placenta tissue, as well as differences in TH concentrations and endogenous DI and SULT activity. The underlying mechanisms of these observed sex differences warrant further investigation.

In the third aim of this dissertation research, the effects of BFRs were examined in a human choriocarcinoma placenta cell line, BeWo. Michaelis-Menten parameters and inhibition curves were calculated for 2,4,6-TBP, 3-OH BDE-47, and 6-OH BDE-47. 2,4,6-TBP was shown to be the most potent inhibitor of 3,3’-T2 SULT activity with a calculated IC50 value of 11.6 nM. It was also shown that 2,4,6-TBP and 3-OH BDE-47 exhibit mixed inhibition of 3,3’-T2 sulfation in BeWo cell homogenates. Next, a series of cell culture exposure experiments were performed using 1, 6, 12, and 24 hour exposure durations. Once again, 2,4,6-TBP was shown to be the most potent inhibitor of basal 3,3’-T2 SULT activity by significantly decreasing activity at the high and medium dose (1 M and 0.5 M, respectively) at all measured time points. Interestingly, BDE-99 was also shown to inhibit basal 3,3’-T2 SULT activity in BeWo cells following the 24 hour exposure, despite exhibiting no inhibitory effects in the BeWo cell homogenate experiments. This indicates that BDE-99 must act through a pathway other than direct enzyme inhibition. Following exposures, the TH concentrations in the cell culture growth media and mRNA expression of TH-related genes were also examined. There was no observed effect of BFR treatment on these endpoints. Future work should focus on determining the downstream biological effects of TH SULT disruption in placental cells, as well as the underlying mechanisms of action responsible for reductions in basal TH SULT activity following BFR exposure.

This was one of the first studies to measure BFRs in a cohort of placenta tissue samples from the United States and the first study to measure THs, DI activity, and SULT activity in human placenta tissues. This research provides a novel contribution to our growing understanding of the effects of BFRs on TH homeostasis within the human placenta, and provides further evidence for sex-specific differences within this important organ. Future research should continue to investigate the effects of environmental contaminants on TH homeostasis within the placenta, as this represents the most critical and vulnerable stage of human development.

The karyophilic properties of the human immunodeficiency virus, type I (HIV-1) pre-integration complex (PIC) allow the virus to infect non-dividing cells. To better understand the mechanisms responsible for nuclear translocation of the PIC, we investigated nuclear import of HIV-1 integrase (IN), a PIC-associated viral enzyme involved in the integration of the viral genome in the host cell DNA. Accumulation of HIV-1 IN into nuclei of digitonin-permeabilized cells does not result from passive diffusion but rather from an active transport that occurs through the nuclear pore complexes. HIV-1 IN is imported by a saturable mechanism, implying that a limiting cellular factor is responsible for this process. Although IN has been previously proposed to contain classical basic nuclear localization signals, we found that nuclear accumulation of IN does not involve karyopherins alpha, beta1, and beta2-mediated pathways. Neither the non-hydrolyzable GTP analog, guanosine 5'-O-(thiotriphosphate), nor the GTP hydrolysis-deficient Ran mutant, RanQ69L, significantly affects nuclear import of IN, which depends instead on ATP hydrolysis. Therefore these results support the idea that IN import is not mediated by members of the karyopherin beta family. More generally, in vitro nuclear import of IN does not require addition of cytosolic factors, suggesting that cellular factor(s) involved in this active but atypical pathway process probably remain associated with the nuclear compartment or the nuclear pore complexes from permeabilized cells.

As collections of archived digital documents continue to grow the maintenance of an archive, and the quality of reproduction from the archived format, become important long-term considerations. In particular, Adobe s PDF is now an important final form standard for archiving and distributing electronic versions of technical documents. It is important that all embedded images in the PDF, and any fonts used for text rendering, should at the very minimum be easily readable on screen. Unfortunately, because PDF is based on PostScript technology, it allows the embedding of bitmap fonts in Adobe Type 3 format as well as higher-quality outline fonts in TrueType or Adobe Type 1 formats. Bitmap fonts do not generally perform well when they are scaled and rendered on low-resolution devices such as workstation screens. The work described here investigates how a plug-in to Adobe Acrobat enables bitmap fonts to be substituted by corresponding outline fonts using a checksum matching technique against a canonical set of bitmap fonts, as originally distributed. The target documents for our initial investigations are those PDF files produced by (La)TEXsystems when set up in a default (bitmap font) configuration. For all bitmap fonts where recognition exceeds a certain confidence threshold replacement fonts in Adobe Type 1 (outline) format can be substituted with consequent improvements in file size, screen display quality and rendering speed. The accuracy of font recognition is discussed together with the prospects of extending these methods to bitmap-font PDF files from sources other than (La)TEX.

Nesse trabalho é descrita a síntese de três hidrazidas graxas monosubstituídas [R1C(O)NHNH2, onde R1 =C15H31, C17H35, C17H33] derivadas dos ácidos graxos palmítico, esteárico e oléico, respectivamente, utilizando duas novas metodologias, primeiramente a partir de monocloridrato de hidrazina em presença de hidróxido de sódio e, em um segundo momento, usando dicloridrato de hidrazina e metóxido de sódio. Ambas as metodologias foram realizadas em metanol como solvente, sendo investigadas as proporções estequiométricas específicas para cada método utilizado. A síntese das hidrazidas graxas com as 4-alcóxi-1,1,1-trialometil-3-alquen-2-onas substituídas [R3C(O)C= C(R2 )(OR), onde R3 = CF3, CCl3; R2 = Me, 4-Me-C6H4, 4- OMe-C6H4, 4-Br-C6H4, 4-Cl-C6H4, 4-F-C6H4], através da reação de ciclocondensação do tipo [3+2] catalisada por BF3.MeOH, favoreceu a síntese regioespecífica de duas novas séries de N-acil trialometil pirazolinas graxas, com rendimentos de bons a excelentes (80 a 90%) e alto grau de pureza, tendo suas estruturas caracterizadas através de dados de Espectroscopia de Infravermelho, Cromatografia Gasosa Acoplada a Espectrometria de Massas e Ressonância Magnética Nuclear de 1H e 13C.

Les troubles respiratoires du sommeil ont une prévalence élevée dans la population générale, l’apnée obstructive du sommeil étant le plus important de ces troubles. Malgré tout, une grande proportion des patients avec apnée ne sont pas diagnostiqués. La méthode la plus complète pour diagnostiquer l’apnée est l’enregistrement d’une nuit de sommeil par polysomnographie, aussi appelée enregistrement de type 1, qui est un processus long et coûteux. Pour surmonter ces difficultés, des appareils d’enregistrements portables (ou de type 3) ont été développés. Toutefois, ces enregistrements de type 3 ne capturent pas l’activité cérébrale, mesurée avec l’électroencéphalographie (EEG). Le présent mémoire décrit une étude comparative entre les enregistrements de type 1 et de type 3. Tous les événements respiratoires d’apnée, d’hypopnée et d’éveils liés à un effort respiratoire (RERA, en anglais) seront analysés ainsi que les éveils cérébraux (ou éveils EEG) et les éveils autonomiques. Ces éveils autonomiques se définissent par une hausse de la fréquence cardiaque suite à un événement respiratoire. Pour enrichir les analyses, les variables respiratoires suivantes ont été étudiées : une chute de la saturation en oxygène de 4 % (ODI), l’index d’apnée-hypopnée (IAH), l’indice de perturbations respiratoires avec apnées + hypopnées + RERAs et les éveils EEG (RDIe, en anglais) et le RDI incluant les éveils autonomiques définis par une augmentation de la fréquence cardiaque de 5 bpm (RDIa5). L’objectif de la présente étude est d’évaluer la proportion d’événements respiratoires avec éveils autonomiques ainsi que leur impact sur le RDI des enregistrements de type 1 et 3. L’hypothèse suggère que les événements avec éveils autonomiques auraient un plus grand impact sur le RDI des enregistrements de type 3 contrairement au type 1. Cette étude inclut 72 sujets ayant suivi un enregistrement de polysomnographie complète de type 1 ainsi que 79 sujets différents ayant suivi un enregistrement ambulatoire de type 3. Les analyses suivantes ont été effectuées : 1) les pourcentages d’événements associés avec seulement des éveils autonomiques dans les enregistrements de type 1 et de type 3 ; 2) les fréquences de migration entre les catégories basses et élevées de sévérité de l’AHI en prenant en compte les événements associés avec seulement des éveils autonomiques ; 3) les Bland-Altman (B-A) pour mesurer l’accord entre l’AHI, le RDIe et le RDIa5 (type 1), et entre l’AHI et le RDIa5 (type 3) et ; 4) les corrélations de Pearson et les coefficients de corrélation intraclasse (ICC) pour mesurer l’accord entre l’AHI, le RDIe et le RDIa5 (type 1), et entre l’AHI et le RDIa5 (type 3). L’utilisation du critère de RDIa5 permet d’ajouter 49 % d’événements comptés avec l’AHI pour les enregistrements de type 1, et 51 % d’événements pour ceux de type 3. La présente étude montre que les éveils autonomiques ont un impact similaire autant pour le RDI des enregistrements de type 3 que de type 1. En conclusion, on peut affirmer que le RDIa5 est acceptable et fiable pour les enregistrements de type 3.

Current procedures for flood risk estimation assume flood distributions are stationary over time, meaning annual maximum flood (AMF) series are not affected by climatic variation, land use/land cover (LULC) change, or management practices. Thus, changes in LULC and climate are generally not accounted for in policy and design related to flood risk/control, and historical flood events are deemed representative of future flood risk. These assumptions need to be re-evaluated, however, as climate change and anthropogenic activities have been observed to have large impacts on flood risk in many areas. In particular, understanding the effects of LULC change is essential to the study and understanding of global environmental change and the consequent hydrologic responses. The research presented herein provides possible causation for observed nonstationarity in AMF series with respect to changes in LULC, as well as a means to assess the degree to which future LULC change will impact flood risk. Four watersheds in the Midwest, Northeastern, and Central United States were studied to determine flood risk associated with historical and future projected LULC change. Historical single framed aerial images dating back to the mid-1950s were used along with Geographic Information Systems (GIS) and remote sensing models (SPRING and ERDAS) to create historical land use maps. The Forecasting Scenarios of Future Land Use Change (FORE-SCE) model was applied to generate future LULC maps annually from 2006 to 2100 for the conterminous U.S. based on the four IPCC-SRES future emission scenario conditions. These land use maps were input into previously calibrated Soil and Water Assessment Tool (SWAT) models for two case study watersheds. In order to isolate effects of LULC change, the only variable parameter was the Runoff Curve Number associated with the land use layer. All simulations were run with daily climate data from 1978-1999, consistent with the 'base' model which employed the 1992 NLCD to represent 'current' conditions. Output daily maximum flows were converted to instantaneous AMF series and were subsequently modeled using a Log-Pearson Type 3 (LP3) distribution to evaluate flood risk. Analysis of the progression of LULC change over the historic period and associated SWAT outputs revealed that AMF magnitudes tend to increase over time in response to increasing degrees of urbanization. This is consistent with positive trends in the AMF series identified in previous studies, although there are difficulties identifying correlations between LULC change and identified change points due to large time gaps in the generated historical LULC maps, mainly caused by unavailability of sufficient quality historic aerial imagery. Similarly, increases in the mean and median AMF magnitude were observed in response to future LULC change projections, with the tails of the distributions remaining reasonably constant. FORE-SCE scenario A2 was found to have the most dramatic impact on AMF series, consistent with more extreme projections of population growth, demands for growing energy sources, agricultural land, and urban expansion, while AMF outputs based on scenario B2 showed little changes for the future as the focus is on environmental conservation and regional solutions to environmental issues.

Studies in Iowa have long documented the vulnerability of wells with less than 50 feet (15 meters) of confining materials above the source aquifer to contamination from nitrate and various pesticides. Recent studies in Wisconsin have documented the occurrence of viruses in untreated groundwater, even in wells considered to have little vulnerability to contamination from near-surface activities. In addition, sensitive methods have become available for analyses of pharmaceuticals and pesticides. This study represents the first comprehensive examination of contaminants of emerging concern in Iowa’s groundwater conducted to date, and one of the first conducted in the United States. Raw groundwater samples were collected from 66 public supply wells during the spring of 2013, when the state was recovering from drought conditions. Samples were analyzed for 206 chemical and biological parameters; including 20 general water-quality parameters and major ions, 19 metals, 5 nutrients, 10 virus groups, 3 species of pathogenic bacteria, 5 microbial indicators, 108 pharmaceuticals, 35 pesticides and pesticide degradates, and tritium. The wells chosen for this study represent a diverse range of ages, depths, confining material thicknesses, pumping rates, and land use settings. The most commonly detected contaminant group was pesticide compounds, which were present in 41% of the samples. As many as 6 pesticide compounds were found together in a sample, most of which were chloroacetanilide degradates. While none of the measured concentrations of pesticide compounds exceeded current benchmark levels, several of these compounds are listed on the U.S. Environmental Protection Agency’s Contaminant Candidate List and could be subject to drinking water standards in the future. Despite heavy use in the past decade, glyphosate was not detected, and its metabolite, aminomethylphosphonic acid, was only detected in two of 60 wells tested (3%) at the detection limit of 0.02 μg/L. Pharmaceutical compounds were detected in 35% of 63 samples. Of the 14 pharmaceuticals detected, six had reported concentrations above the method reporting limit, with the maximum reported concentration of 826 ng/L for acetaminophen. Diphenhydramine was the only pharmaceutical to have two detections above the reporting limit, at 24.5 and 145 ng/L. Eight pharmaceuticals had confirmed detections at concentrations below the method reporting limit. Caffeine was the most frequently detected pharmaceutical compound (25%), followed by the caffeine metabolite, 1,7-dimethylxanthine (16%). Microorganisms were detected in 21% of the wells using quantitative polymerase chain reaction methodologies. The most frequently detected microorganism was the pepper mild mottle virus (PMMV), a plant pathogen found in human waste. PMMV was detected in 17% of samples at concentrations ranging from 0.4 to 6.38 gene copies per liter. GII norovirus, human polyomavirus, bovine polyomavirus, and Campylobacter were also detected, while adenovirus, enterovirus, GI norovirus, swine hepatitis E, Salmonella, and enterohemmorhagic E. coli were not detected. No correlations were found between viruses or pathogenic bacteria and microbial indicators. Wells with less than 50 feet (15 meters) of confining material were shown to have greater incidence of surface-related contaminants; however, significant relationships (p<0.05) between confining layer thickness and contaminants were only found for nitrate and herbicides.

Nariño y Cauca son dos de los departamentos de Colombia más afectados por la violencia. La reciente firma de un acuerdo de un cese bilateral de hostilidades con las Fuerzas Armadas Revolucionarias de Colombia (FARC) y los diálogos de La Habana son apenas el comienzo de la Construcción de Paz (CP) que implica el esfuerzo continuo de diferentes actores (gubernamentales, sector privado, sociedad civil y organismos multilaterales) para lograr no solo una paz negativa sino una paz positiva. El apoyo al emprendimiento es una estrategia implementada por el Gobierno y por los stakeholders que participan en el proceso del posconflicto, que tiene como finalidad respaldar el proceso de integración económica de las víctimas y desmovilizados. El presente documento es un estudio exploratorio elaborado por medio de una investigación cualitativa en la temática de emprendimiento, instituciones y CP en los departamentos de Nariño y Cauca. Se utilizó una estrategia metodológica denominada Matrices de Stakeholders para representar gráficamente la influencia institucional sobre la toma decisiones e implementación de los stakeholders sobre las reformas o políticas de emprendimiento y CP en estos dos departamentos. En esta investigación se encontró que i) en general, las instituciones del gobierno de los de Nariño y Cauca son extractivas y limitan la participación de la sociedad; ii) los stakeholders de la sociedad civil a pesar de tener cierta organización y voz no están en capacidad de generar influencia más que a nivel local o comunitario; iii) los vacíos dejados por las instituciones extractivas del gobierno tienden a ser llenados por instituciones inclusivas de stakeholders del sector privado y de organismos multilaterales.

Se describe la variante homocigota c.320-2A>G de TGM1 en dos hermanas con ictiosis congénita autosómica recesiva. El clonaje de los transcritos generados por esta variante permitió identificar tres mecanismos moleculares de splicing alternativos.
