Two forms of the activated beta(1)-adrenoceptor exist, one that is stabilized by (-)-noradrenaline and is sensitive to blockade by (-)-propranolol and another which is stabilized by partial agonists such as (-)-pindolol and (-)-CGP 12177 but is relatively insensitive to (-)-propranolol. We investigated the effects of stimulation of the propranolol-resistant PI-adrenoceptor in the human heart. Myocardium from non-failing and failing human hearts were set up to contract at 1 Hz. In right atrium from non-ailing hearts in the presence of 200 nM (-)-propranolol, (-)-CGP 12177 caused concentration-dependent increases in contractile force (-logEC(50)[M] 7.3+/-0.1, E-max 23+/-1% relative to maximal (-)-isoprenaline stimulation of beta(1)- and beta(2)-adrenoceptors, n=86 patients), shortening of the time to reach peak force (-logEC(50)[M] 7.4+/-0.1, E-max 37+/-5%, n=61 patients) and shortening of the time to reach 50% relaxation (t(50%), -logEC(50)[M] 7.3+/-0.1, E-max 33+/-2%, n=61 patients). The potency and maxima of the positive inotropic effects were independent of Ser49Gly- and Gly389Arg-beta(1)-adrenoceptor polymorphisms but were potentiated by the phosphodiesterase inhibitor 3-isobutyl-1-methylxanthine (-logEC(50)[M] 7.7+/-0.1, E-max 68+/-6%, n=6 patients, P

Two methods were compared for determining the concentration of penetrative biomass during growth of Rhizopus oligosporus on an artificial solid substrate consisting of an inert gel and starch as the sole source of carbon and energy. The first method was based on the use of a hand microtome to make sections of approximately 0.2- to 0.4-mm thickness parallel to the substrate surface and the determination of the glucosamine content in each slice. Use of glucosamine measurements to estimate biomass concentrations was shown to be problematic due to the large variations in glucosamine content with mycelial age. The second method was a novel method based on the use of confocal scanning laser microscopy to estimate the fractional volume occupied by the biomass. Although it is not simple to translate fractional volumes into dry weights of hyphae due to the lack of experimentally determined conversion factors, measurement of the fractional volumes in themselves is useful for characterizing fungal penetration into the substrate. Growth of penetrative biomass in the artificial model substrate showed two forms of growth with an indistinct mass in the region close to the substrate surface and a few hyphae penetrating perpendicularly to the surface in regions further away from the substrate surface. The biomass profiles against depth obtained from the confocal microscopy showed two linear regions on log-linear plots, which are possibly related to different oxygen availability at different depths within the substrate. Confocal microscopy has the potential to be a powerful tool in the investigation of fungal growth mechanisms in solid-state fermentation. (C) 2003 Wiley Periodicals, Inc.

A new questionnaire, the Maternal Mental State Input Inventory (MMSII) was created to measure mothers' preferences for introducing and elaborating on mental states in conversation with their young children. In two studies, the questionnaire was given to mothers of young children, and the children's theory of mind (ToM) development was assessed with standard tasks. In both studies, the questionnaire exhibited good internal reliability, and significant correlations emerged between mothers' self-reported preferences for elaborated, explanatory talk about the mental states and children's theory of mind performance. Further, mothers' conversational preferences, as measured by the MMSII, were the best predictors of children's theory of mind development when relevant control variables were included in the analyses. These results converge with naturalistic observational research that has demonstrated links between mothers' conversational styles and their children's theory of mind. They go further in suggesting that mothers' tendencies toward elaborated, explanatory talk about a range of mental states is particularly beneficial to children's theory of mind development. Low-temperature (15 K) single-crystal neutron-diffraction structures and Raman spectra of the salts (NX4)(2)[CU(OX2)(6)](SO4)(2), where X = H or D, are reported. This study is concerned with the origin of the structural phase change that is known to occur upon deuteration. Data for the deuterated salt were measured in the metastable state, achieved by application of 500 bar of hydrostatic pressure at similar to303 K followed by cooling to 281 K and the subsequent release of pressure. This allows for the direct comparison between the hydrogenous and deuterated salts, in the same modification, at ambient pressure and low temperature. The Raman spectra provide no intimation of any significant change in the intermolecular bonding. Furthermore, structural differences are few, the largest being for the long Cu-O bond, which is 2.2834(5) and 2.2802(4) Angstrom for the hydrogenous and the deuterated salts, respectively. Calorimetric data for the deuterated salt are also presented, providing an estimate of 0.17(2) kJ/mol for the enthalpy difference between the two structural forms at 295.8(5) K. The structural data suggest that substitution of hydrogen for deuterium gives rise to changes in the hydrogen-bonding interactions that result in a slightly reduced force field about the copper(II) center. The small structural differences suggest different relative stabilities for the hydrogenous and deuterated salts, which may be sufficient to stabilize the hydrogenous salt in the anomalous structural form.

In their 1994 study Taxation and Representation, Deacon and Golding point to the extensive use of press and public relations professionals by governments to promote policy, and to outmanouvre their opponents. With the UK specifically in mind, they warn: 'we cannot ignore the massive expansion of the public relations state.' (p.6). What distinguishes their approach from the more usual preoccupation with the use of 'spin' to 'package' political leaders is a focus on the institutionalisation of public relations within government. In this paper, I explore the utililty of the concept, and consider what the broad features of an Australian 'PR State' might be.

O objetivo deste artigo ?? compreender a percep????o dos gestores, que atuam com o planejamento estrat??gico de uma organiza????o p??blica, sobre o construto de comprometimento no trabalho, bem como a influ??ncia dessa percep????o em suas pr??ticas cotidianas de gest??o. Para tanto, realizou-se, inicialmente, pesquisa bibliogr??fica focando quest??es relacionadas ao sentido e natureza do construto, bem como sobre os estudos realizados sobre o tema do comprometimento no contexto da administra????o p??blica. Em seguida, foram coletados dados com gestores mediante entrevista com roteiro semiestruturado, os quais foram analisados por meio da t??cnica de an??lise de conte??do. As evid??ncias indicam que os gestores associam o v??nculo ao trabalho a quest??es atitudinais e de inten????es comportamentais e que suas pr??ticas cotidianas s??o influenciadas pela percep????o de comprometimento dos indiv??duos, admitindo que as decis??es tomadas, motivadas por essa percep????o, podem trazer consequ??ncias negativas tanto para a organiza????o quanto para o indiv??duo.

Refletindo sobre modelo de carreira mais adequado para a Administra????o P??blica nesta era de economia globalizada e de maiores exig??ncias de efici??ncia e flexibilidade na gest??o dos aparatos governamentais, questiona-se aqui a id??ia de que o modelo burocr??tico deva ser substitu??do pelo modelo gerencial. Considerando essa id??ia apenas parcialmente verdadeira, nossa argumenta????o vai na seguinte dire????o: como a burocracia ?? a forma espec??fica de organiza????o do aparato administrativo do Estado de Direito, este modelo n??o pode ser rejeitado, sob pena de comprometimento desse Estado. Mas ele precisa ser flexibilizado, superando e reduzindo o formalismo excessivo e a rigidez que sua realiza????o hist??rica possa ter gerado nos diferentes pa??ses. Igualmente, deve ser aperfei??oado em seus mecanismos de controle, at?? como condi????o para sua manuten????o eficaz nos governos democr??ticos contempor??neos.

O objetivo deste paper ?? demonstrar a todos aqueles que trabalham com a disciplina Administra????o P??blica quais s??o os principais autores e obras da literatura nacional/internacional recente, articulando-os sob os aspectos anal??tico, metodol??gico e te??rico-conceitual, no sentido de contribuir para a cumulatividade da gera????o do conhecimento e forma????o de panorama nessa ??rea, no per??odo selecionado (1994/2002). Para tanto, foi adotada metodologia espec??fica para esse objetivo, apresentada por KEINERT (2000) em fun????o das caracter??sticas estruturais da disciplina. A literatura foi agrupada em cinco loci contemplando a vertente p??s-burocr??tica da gest??o p??blica, os impactos da globaliza????o no aparelho do Estado, as condicionantes do ordenamento pol??tico e econ??mico nacional e internacional, as novas tend??ncias no ciclo de pol??ticas p??blicas e a gest??o de pol??ticas sociais.As principais conclus??es da pesquisa apontam para a adequa????o do instrumental anal??tico adotado para compreens??o da literatura em Administra????o P??blica e tamb??m para a baixa verticalidade e cumulatividade da pesquisa nesse campo, com pouca sistematiza????o e densidade anal??tica no que se refere a determinados temas.

O modelo brasileiro das organiza????es sociais representa uma das respostas poss??veis ?? crise do aparelho do Estado no ??mbito da presta????o dos servi??os sociais. Essas entidades s??o percebidas como uma forma de parceria do Estado com as institui????es privadas de fins p??blicos (perspectiva ex parte principe) ou, sob outro ??ngulo, uma forma de participa????o popular na gest??o administrativa (perspectiva ex parte populi). No texto s??o tematizadas as diferen??as e semelhan??as entre o marco legal das organiza????es sociais e das entidades de utilidade p??blica no Brasil, as notas distintivas entre a disciplina dos servi??os privados de interesse p??blico e dos servi??os p??blicos, bem como o que distingue juridicamente o modelo das organiza????es sociais de processos de privatiza????o e terceiriza????o. Em todos esses temas os juristas aparecem como protagonistas na determina????o dos limites do modelo das organiza????es sociais, evidenciando que processos de reforma normativa exigem, para serem eficazes, uma concomitante reforma na mentalidade dos agentes p??blicos.

O artigo contrap??e modelos alternativos de organiza????o das Institui????es Federais de Ensino Superior (IFES) propostos pelo Minist??rio da Administra????o e Reforma do Estado (MARE), no ??mbito do Programa de Reforma do Estado, e pelo Minist??rio da Educa????o e do Desporto (MEC). Procura tamb??m avaliar de que forma as solu????es propostas enfrentam alguns problemas caracter??sticos dessas institui????es, levantando quest??es sobre a viabilidade desses modelos. Apresenta, ainda, algumas sugest??es para a abordagem dos problemas apontados.
