Diplodia corticola is regarded as the most virulent fungus involved in cork oak decline, being able to infect not only Quercus species (mainly Q. suber and Q. ilex), but also grapevines (Vitis vinifera) and eucalypts (Eucalyptus sp.). This endophytic fungus is also a pathogen whose virulence usually manifests with the onset of plant stress. Considering that the infection normally culminates in host death, there is a growing ecologic and socio-economic concern about D. corticola propagation. The molecular mechanisms of infection are hitherto largely unknown. Accordingly, the aim of this study was to unveil potential virulence effectors implicated in D. corticola infection. This knowledge is fundamental to outline the molecular framework that permits the fungal invasion and proliferation in plant hosts, causing disease. Since the effectors deployed are mostly proteins, we adopted a proteomic approach. We performed in planta pathogenicity tests to select two D. corticola strains with distinct virulence degrees for our studies. Like other filamentous fungi D. corticola secretes protein at low concentrations in vitro in the presence of high levels of polysaccharides, two characteristics that hamper the fungal secretome analysis. Therefore, we first compared several methods of extracellular protein extraction to assess their performance and compatibility with 1D and 2D electrophoretic separation. TCA-Acetone and TCA-phenol protein precipitation were the most efficient methods and the former was adopted for further studies. The proteins were extracted and separated by 2D-PAGE, proteins were digested with trypsin and the resulting peptides were further analysed by MS/MS. Their identification was performed by de novo sequencing and/or MASCOT search. We were able to identify 80 extracellular and 162 intracellular proteins, a milestone for the Botryosphaeriaceae family that contains only one member with the proteome characterized. We also performed an extensive comparative 2D gel analysis to highlight the differentially expressed proteins during the host mimicry. Moreover, we compared the protein profiles of the two strains with different degrees of virulence. In short, we characterized for the first time the secretome and proteome of D. corticola. The obtained results contribute to the elucidation of some aspects of the biology of the fungus. The avirulent strain contains an assortment of proteins that facilitate the adaptation to diverse substrates and the identified proteins suggest that the fungus degrades the host tissues through Fenton reactions. On the other hand, the virulent strain seems to have adapted its secretome to the host characteristics. Furthermore, the results indicate that this strain metabolizes aminobutyric acid, a molecule that might be the triggering factor of the transition from a latent to a pathogenic state. Lastly, the secretome includes potential pathogenicity effectors, such as deuterolysin (peptidase M35) and cerato-platanin, proteins that might play an active role in the phytopathogenic lifestyle of the fungus. Overall, our results suggest that D. corticola has a hemibiotrophic lifestyle, switching from a biotrophic to a necrotrophic interaction after plant physiologic disturbances.This understanding is essential for further development of effective plant protection measures.

Temas que, por otra parte, marcan derroteros de búsqueda, de formulación, interrogantes que se vertieron en investigaciones que comprometen la escritura misma de las cosas. Todos los aquí presentes son estudiantes de posgrado de la carrera de filosofía, alumnos de dos seminarios que extendieron, como prácticas de formación y discusión sus trabajos en el “Coloquio Reflexiones sobre el saber, el poder, la verdad y las prácticas de sí”. Hoy, parte de estos trabajos, en los que los estudiantes arriesgaron sus propias tesis, sus pesquisas, la entrañable cualidad de quedar comprometidos con el discurso y haciendo “uso” de lo que el propio Foucault deseaba, cada uno hace de sí una parte de esa obra que es la estética de sí. Cardiogenesis is a delicate and complex process that requires the coordination of an intricate network of pathways and the different cell types. Therefore, understanding heart development at the morphogenetic level is an essential requirement to uncover the causes of congenital heart disease and to provide insight for disease therapies. Mouse Cerberus like 2 (Cerl2) has been defined as a Nodal antagonist in the node with an important role in the Left-Right (L/R) axis establishment, at the early embryonic development. As expected, Cerl2 knockout mice (Cerl2-/-) showed multiple laterality defects with associated cardiac failure. In order to identify the endogenous role of Cerl2 during heart formation independent of its described functions in the node, we accurately analyzed animals where laterality defects were not present. We thereby unravel the consequences of Cerl2 lossof- function in the heart, namely increased left ventricular thickness due to hyperplasia of cardiomyocytes and de-regulated expression of cardiac genes. Furthermore, the Cerl2 mutant neonates present impaired cardiac function. Once that the cardiac expression of Cerl2 is mostly observed in the left ventricle until around midgestration, this result suggest a specific regulatory role of Cerl2 during the formation of the left ventricular myoarchitecture. Here, we present two possible molecular mechanisms underlying the cardiac Cerl2 function, the regulation of Cerl2 antagonist in activation of the TGFßs/Nodal/Activin/Smad2 signaling identified by increased Smad2 phosphorilation in Cerl2-/- hearts and the negative feedback between Cerl2 and Wnt/ß-catenin signaling in heart formation. In this work and since embryonic stem cells derived from 129 mice strain is extensively used to produce targeted mutants, we also present echocardiographic reference values to progressive use of juveniles and young adult 129/Sv strain in cardiac studies. In addition, we investigate the cardiac physiology of the surviving Cerl2 mutants in 129/Sv background over time through a follow-up study using echocardiographic analysis. Our results revealed that Cerl2-/- mice are able to improve and maintain the diastolic and most of systolic cardiac physiologic parameters as analyzed until young adult age. Since Cerl2 is no longer expressed in the postnatal heart, we suggest that an intrinsic and compensatory mechanism of adaptation may be active for recovering the decreased cardiac function found in Cerl2 mutant neonates. Altogether, these data highlight the role of Cerl2 during embryonic heart development in mice. Furthermore, we also suggest that Cerl2-/- may be an interesting model to uncover the molecular, cellular and physiological mechanisms behind the improvement of the cardiac function, contributing to the development of therapeutic approaches to treat heart failures.

Korean mondshood root polysaccharides (KMPS) isolated from the root of Aconitum coreanum (Lévl.) Rapaics have shown anti-inflammatory activity, which is strongly influenced by their chemical structures and chain conformations. However, the mechanisms of the anti-inflammatory effect by these polysaccharides have yet to be elucidated. A RG-II polysaccharide (KMPS-2E, Mw 84.8 kDa) was isolated from KMPS and its chemical structure was characterized by FT-IR and NMR spectroscopy, gas chromatography–mass spectrometry and high-performance liquid chromatography. The backbone of KMPS-2E consisted of units of [→6) -β-D-Galp (1→3)-β-L-Rhap-(1→4)-β-D-GalpA-(1→3)-β-D-Galp-(1→] with the side chain →5)-β-D-Arap (1→3, 5)-β-D-Arap (1→ attached to the backbone through O-4 of (1→3,4)-L-Rhap. T-β-D-Galp is attached to the backbone through O-6 of (1→3,6)-β-D-Galp residues and T-β-D-Ara is connected to the end group of each chain. The anti-inflammatory effects of KMPS-2E and the underlying mechanisms using lipopolysaccharide (LPS) - stimulated RAW 264.7 macrophages and carrageenan-induced hind paw edema were investigated. KMPS-2E (50, 100 and 200 µg/mL) inhibits iNOS, TLR4, phospho-NF-κB–p65 expression, phosphor-IKK, phosphor-IκB-α expression as well as the degradation of IκB-α and the gene expression of inflammatory cytokines (TNF-α, IL-1β, iNOS and IL-6) mediated by the NF-κB signal pathways in macrophages. KMPS-2E also inhibited LPS-induced activation of NF-κB as assayed by electrophorectic mobility shift assay (EMSA) in a dose-dependent manner and it reduced NF-κB DNA binding affinity by 62.1% at 200µg/mL. In rats, KMPS-2E (200 mg/kg) can significantly inhibit carrageenan-induced paw edema as ibuprofen (200 mg/kg) within 3 h after a single oral dose. The results indicate that KMPS-2E is a promising herb-derived drug against acute inflammation.

Los procesos de acogimiento familiar que se producen en cada país dependen de factores históricos y culturales que dan lugar a grandes diferencias internacionales. A pesar de que las comparativas internacionales ofrecen un medio de intercambio de experiencias, lo que permite el aprendizaje mutuo y la transferencia de buenas prácticas, a menudo encontramos enormes barreras para su realización debido, entre otros factores, al limitado acceso a los datos sobre la práctica de la protección infantil. Frente a estas limitaciones, este artículo ofrece una investigación comparativa realizada en España y Portugal, cuyo objetivo ha sido la evaluación del acogimiento familiar en dos países donde la investigación en el ámbito de la protección infantil há sido tradicionalmente desatendida. Sobre una muestra de 357 casos en España y 289 en Portugal, se realizó un estudio del perfil de características de los niños acogidos, las familias de origen y los acogedores, así como de los procesos de acogida en ambos países. La comparativa reveló importantes diferencias relacionadas con la mayor edad al inicio del acogimiento de los niños en España; el perfil de los acogedores, con edades más avanzadas, un bajo nivel educativo y llevando a cabo acogidas de más de un niño simultaneamente en Portugal. El estudio comparado nos ha permitido identificar áreas que requieren mayor atención en ambos países, como la renovación generacional del banco de acogedores en Portugal o la necesidad de acelerar los procesos de toma de decisiones para la entrada en acogimiento familiar en España. Se pretende que este artículo sirva de aliento para la recogida de datos y comparación con otros países iberoamericanos.

Estadística corriente de la pesca de anchoveta, la cual fue iniciada con el Informe 1 cubriendo el periodo comprendido entre Octubre y diciembre de 1961. Esta vez han sido introducidas algunas pequeñas reformas: 1) Se presentan estadísticas de desembarque de anchoveta de acurdo a las dos fuentes disponibles de información: a) desembarque por viajes de embarcaciones y b) cantidades de pescado adquiridas diariamente por las fabricas harinera, 2) los cuadros en los que se dan los datos de desembarque por categorías de tamaños de embarcación incluyen las cantidades máximas desembarcadas y por cualquier embarcación en su grupo y presentan, además, los valores promedios 3) la distribución mensual de las embarcaciones por grupos de tamaño que en el primer informe fue dado por puestos, es presentado ahora en conjunto para todo el país.

El estudio trata de la situación de 1968, desde el punto de vista técnico, de la industria del procesamiento de pescado para consumo humano en el país, es decir, del pescado fresco, refrigerado, congelado, conservas, productos salados y secos, ahumados, marinados y salsas. Para cada uno de estos reglones de la industria se especifica las fluctuaciones en su producción, consumo y exportación durante los últimos 5 años. Además se indica la ubicación de los centros de producción a lo largo del litoral; las facilidades que disponen, los métodos de fabricación; maquinaria empleada, capacidades, materia prima utilizada, tipo de productos elaborados. Así mismo, se exponen los problemas que confrontan y se dan algunas recomendaciones que podrían contribuir a su mayor eficiencia y desarrollo.
