This study aimed to investigate which genes Cnidaria use for photoreception and test whether Gi alpha subunit protein is involved in the phototransduction cascade, giving additional tools to investigate light-mediated behaviors, as nematocyte firing. Here, I engineered an opsin gene promoter construct useful to test whether nematocyte sensory cells express opsin gene. By determining the expression of one of the unique EST opsin genes of the eyeless hydrozoan Hydra magnipapillata genome in nematocyte sensory cells, we will be able to investigate whether light modulation is an ancestral feature in Cnidaria, and whether regulation of nematocyte discharge by opsin-mediated phototransduction predated this pathway’s function in cnidarian eyes. Nematocytes, the cnidarians stinging cells, discharge nematocysts to capture prey. As nematocysts are energetically expensive, the discharge is tightly regulated and occurs after proper chemical and mechanical stimulation. Cnidarians are also known to display a rich corpus of photobehaviors, which are often associated with activities that involve nematocytes. Previous experiments on nematocyst firing modulation show that light decreases nematocyte firing. This study contributed to confirm that bright light decreases the tendency for nematocytes to discharge in Haliplanella luciae. Similar findings in cubozoan and hydrozoan lead us to believe that light modulation of cnidocytes may be an ancestral feature of Cnidaria. Experimentally, I found no evidence that pertussis toxin, a Gi alpha subunit protein inhibitor, ablates Hydra magnipapillata photobehaviour, preliminary suggesting that Gi alpha subunit protein is not involved in photoresponse. I found no significant association between pertussis toxin and nematocyte firing in Haliplanella luciae both in conditions of dim and bright light, suggesting that Gi alpha subunit protein is not involved in photoresponse. We have preliminary evidence for a prevalence of photoreception over chemoreception, tending toward conditions of bright light. This finding may suggest the involvement of a Gs alpha subunit protein in Haliplanella luciae phototransduction pathway. While nematocyte chemo- and mechano-sensitivity have been extensively studied, further research is necessary to better understand what an ancestral phototransduction cascade looked like, and how opsin-based phototransduction acts to regulate nematocyte discharge.

La necessità di conservazione e recupero di murature, di qualsiasi interesse storico/architettonico, ha evidenziato la necessità di conoscere il più dettagliatamente possibile le caratteristiche strutturali delle opere interessate: un intervento risulterà tanto più efficace e adatto agli scopi prefissi quanto maggiore sarà la conoscenza dell’opera, della sua evoluzione, dei materiali utilizzati per la realizzazione, della tecnica costruttiva e della struttura portante. Spesso è necessario eseguire interventi di adeguamento sismico di comuni edifici su cui poter intervenire più o meno indiscriminatamente, mentre, per opere di interesse storico è necessario ridurre al minimo l’invasività degli interventi: in tutti e due i casi, una buona riuscita dell’intervento dipende dalla conoscenza dell’organismo strutturale sul quale si deve operare. Come spesso accade, anche per opere di recente costruzione, risulta difficile poter recuperare i dati progettuali (disegni e calcoli) e spesso le tecniche e le tipologie utilizzate per le costruzioni si differenziavano da zona a zona, così come diversi erano i materiali da costruzione; risulta quindi evidente che per progettare una serie di interventi di recupero è necessario poter ottenere il maggior numero di informazioni al riguardo. Diverse sono le esperienze maturate in questo campo in tutta Europa e queste hanno mostrato come non è sufficiente intervenire con tecniche innovative per adeguare agli standard attuali di sicurezza opere del passato: infatti, in molti casi, l’applicazione sbagliata di queste tecniche o gli interventi progettati in modo non adeguato non hanno svolto il loro compito e in alcuni casi hanno peggiorato la situazione esistente. Dalle esperienze maturate è stato possibile osservare che anche le migliore tecniche di recupero non possono risultare efficaci senza un’adeguata conoscenza dello stato di degrado degli edifici, del loro assetto strutturale e delle possibili carenze. La diagnostica strutturale si vuole inserire proprio in questo livello potendo fornire ad un progettista tutte le informazioni necessarie per effettuare al meglio ed in maniera efficace gli interventi di recupero e restauro necessari. Oltre questi aspetti, le analisi diagnostiche possono essere utilizzate anche per verificare l’efficacia degli interventi effettuati. Diversi sono gli aspetti che si possono analizzare in un’indagine di diagnostica ed in base alle esigenze e alle necessità del rilievo da effettuare sono varie le tecniche a disposizione, ognuna con le sue peculiarità e potenzialità. Nella realizzazione di questa tesi sono state affrontate diverse problematiche che hanno previsto sia l’analisi di situazioni reali in cantiere, sia lo studio in laboratorio. La prima parte del presente elaborato prevede lo studio delle attività svolte a Palazzo Malvezzi, attuale sede della Provincia di Bologna. L’edificio, di interesse storico, ha subito diverse trasformazioni durate la sua vita ed in alcuni casi, queste, eseguite con tecnologie e materiali inadatti, hanno provocato variazioni nell’assetto statico della struttura; inoltre, il palazzo, è soggetto a movimenti a livello di fondazione in quanto è presente una faglia di subsidenza che attraversa l’edificio. Tutte queste problematiche hanno creato movimenti differenziali alla struttura in elevazione che si sono evidenziati con crepe distribuite in tutto l’edificio. Il primo aspetto analizzato (capitoli 4 e 5) è lo studio della profondità delle fessure presenti nel solaio della sala Rossa, sede dei comunicati stampa e delle conferenze della Provincia. Senza dubbio antiestetiche, le crepe presenti in una struttura, se sottovalutate, possono compromettere notevolmente le funzioni statiche dell’elemento in cui si sviluppano: la funzione di protezione fornita dal solaio o da qualsiasi altro elemento strutturale alle armature in questo immerse, viene meno, offrendo vie preferenziali a possibili situazioni di degrado, specialmente se in condizioni ambientali aggressive. E' facile intuire, quindi, che un aspetto all’apparenza banale come quello delle fessure non può essere sottovalutato. L’analisi è stata condotta utilizzando prove soniche ed impact-echo, tecniche che sfruttano lo stresso principio, la propagazione delle onde elastiche nel mezzo, ma che si differenziano per procedure di prova e frequenze generate nel test. Nel primo caso, la presenza del martello strumentato consente di valutare anche la velocità di propagazione delle onde elastiche, fenomeno utilizzato per ottenere indicazioni sulla compattezza del mezzo, mentre nel secondo non è possibile ricavare queste informazioni in quanto la tecnica si basa solamente sullo studio in frequenza del segnale. L’utilizzo dell’impact-echo è stato necessario in quanto la ristilatura effettuata sulle fessure scelte per l’analisi, non ha permesso di ottenere risultati utili tramite prove soniche ; infatti, le frequenze generate risultano troppo basse per poter apprezzare queste piccole discontinuità materiali. La fase di studio successiva ha previsto l’analisi della conformazione dei solai. Nel capitolo 6, tale studio, viene condotto sul solaio della sala Rossa con lo scopo di individuarne la conformazione e la presenza di eventuali elementi di rinforzo inseriti nelle ristrutturazioni avvenute nel corso della vita del palazzo: precedenti indagini eseguite con endoscopio, infatti, hanno mostrato una camera d’aria ed elementi metallici posizionati al di sotto della volta a padiglione costituente il solaio stesso. Le indagini svolte in questa tesi, hanno previsto l’utilizzo della strumentazione radar GPR: con questa tecnica, basata sulla propagazione delle onde elettromagnetiche all’interno di un mezzo, è possibile variare rapidamente la profondità d’ispezione ed il dettaglio ottenibile nelle analisi cambiando le antenne che trasmettono e ricevono il segnale, caratteristiche fondamentali in questo tipo di studio. I risultati ottenuti hanno confermato quanto emerso nelle precedenti indagini mostrando anche altri dettagli descritti nel capitolo. Altro solaio oggetto d’indagine è quello della sala dell’Ovale (capitoli 7 e 8): costruito per dividere l’antica sala da ballo in due volumi, tale elemento è provvisto al suo centro di un caratteristico foro ovale, da cui ne deriva il nome. La forma del solaio lascia supporre la presenza di una particolare struttura di sostegno che le precedenti analisi condotte tramite endoscopio, non sono riuscite a cogliere pienamente. Anche in questo caso le indagini sono state eseguite tramite tecnica radar GPR, ma a differenza dei dati raccolti nella sala Rossa, in questo caso è stato possibile creare un modello tridimensionale del mezzo investigato; inoltre, lo studio è stato ripetuto utilizzando un’antenna ad elevata risoluzione che ha consentito di individuare dettagli in precedenza non visibili. Un ulteriore studio condotto a palazzo Malvezzi riguarda l’analisi della risalita capillare all’interno degli elementi strutturali presenti nel piano interrato (capitolo 9): questo fenomeno, presente nella maggior parte delle opere civili, e causa di degrado delle stesse nelle zone colpite, viene indagato utilizzando il radar GPR. In questo caso, oltre che individuare i livelli di risalita osservabili nelle sezioni radar, viene eseguita anche un’analisi sulle velocità di propagazione del segnale e sulle caratteristiche dielettriche del mezzo, variabili in base al contenuto d’acqua. Lo scopo è quello di individuare i livelli massimi di risalita per poterli confrontare con successive analisi. Nella fase successiva di questo elaborato vengono presentate le analisi svolte in laboratorio. Nella prima parte, capitolo 10, viene ancora esaminato il fenomeno della risalita capillare: volendo studiare in dettaglio questo problema, sono stati realizzati dei muretti in laterizio (a 2 o 3 teste) e per ognuno di essi sono state simulate diverse condizioni di risalita capillare con varie tipologie di sale disciolto in acqua (cloruro di sodio e solfato di sodio). Lo scopo è di valutare i livelli di risalita osservabili per diverse tipologie di sale e per diverse concentrazioni dello stesso. Ancora una volta è stata utilizzata la tecnica radar GPR che permette non solo di valutare tali livelli, ma anche la distribuzione dell’umidità all’interno dei provini, riuscendo a distinguere tra zone completamente sature e zone parzialmente umide. Nello studio è stata valutata anche l’influenza delle concentrazioni saline sulla propagazione del segnale elettromagnetico. Un difetto delle tecniche di diagnostica è quello di essere strumenti qualitativi, ma non quantitativi: nel capitolo 11 viene affrontato questo problema cercando di valutare la precisione dei risultati ottenuti da indagini condotte con strumentazione radar GPR. Studiando gli stessi provini analizzati nel capitolo precedente, la tecnica radar è stata utilizzata per individuare e posizionare i difetti (muretti a 3 teste) e le pietre (muretti a 2 teste) inserite in questi elementi: i risultati ottenuti nella prova sono stati confrontati, infine, con la reale geometria. Nell’ultima parte (capitolo 12), viene esaminato il problema dell’individuazione dei vuoti all’interno di laterizi: per questo scopo sono state create artificialmente delle cavità di diversa forma e a diverse profondità all’interno di laterizi pieni che, dopo un trattamento in cella climatica per aumentarne la temperatura, sono stati sottoposti a riprese termografiche nella fase di raffreddamento. Lo scopo di queste prove è quello di valutare quale sia la massima differenza di temperatura superficiale causata dai diversi difetti e per quale intervallo di temperature questa si verifica.

My research PhD work is focused on the Electrochemically Generated Luminescence (ECL) investigation of several different homogeneous and heterogeneous systems. ECL is a redox induced emission, a process whereby species, generated at electrodes, undergo a high-energy electron transfer reaction to form excited states that emit light. Since its first application, the ECL technique has become a very powerful analytical tool and has widely been used in biosensor transduction. ECL presents an intrinsically low noise and high sensitivity; moreover, the electrochemical generation of the excited state prevents scattering of the light source: for all these characteristics, it is an elective technique for ultrasensitive immunoassay detection. The majority of ECL systems involve species in solution where the emission occurs in the diffusion layer near to the electrode surface. However, over the past few years, an intense research has been focused on the ECL generated from species constrained on the electrode surface. The aim of my work is to study the behavior of ECL-generating molecular systems upon the progressive increase of their spatial constraints, that is, passing from isolated species in solution, to fluorophores embedded within a polymeric film and, finally, to patterned surfaces bearing “one-dimensional” emitting spots. In order to describe these trends, I use different “dimensions” to indicate the different classes of compounds. My thesis was mostly developed in the electrochemistry group of Bologna with the supervision of Prof Francesco Paolucci and Dr Massimo Marcaccio. With their help and also thanks to their long experience in the molecular and supramolecular ECL fields and in the surface investigations using scanning probe microscopy techniques, I was able to obtain the results herein described. Moreover, during my research work, I have established a new collaboration with the group of Nanobiotechnology of Prof. Robert Forster (Dublin City University) where I spent a research period. Prof. Forster has a broad experience in the biomedical field, especially he focuses his research on film surfaces biosensor based on the ECL transduction. This thesis can be divided into three sections described as follows: (i) in the fist section, homogeneous molecular and supramolecular ECL-active systems, either organic or inorganic species (i.e., corannulene, dendrimers and iridium metal complex), are described. Driving force for this kind of studies includes the search for new luminophores that display on one hand higher ECL efficiencies and on the other simple mechanisms for modulating intensity and energy of their emission in view of their effective use in bioconjugation applications. (ii) in the second section, the investigation of some heterogeneous ECL systems is reported. Redox polymers comprising inorganic luminophores were described. In such a context, a new conducting platform, based on carbon nanotubes, was developed aimed to accomplish both the binding of a biological molecule and its electronic wiring to the electrode. This is an essential step for the ECL application in the field of biosensors. (iii) in the third section, different patterns were produced on the electrode surface using a Scanning Electrochemical Microscopy. I developed a new methods for locally functionalizing an inert surface and reacting this surface with a luminescent probe. In this way, I successfully obtained a locally ECL active platform for multi-array application.

Der Austausch von Spurengasen und Aerosolpartikeln zwischenAtmosphäre und Biosphäre spielt eine wichtige Rolle in derAtmosphärenphysik und -chemie. Wälder repräsentieren sowohleine signifikante Senke als auch Quelle für Spurengase undPartikel und tragen somit maßgeblich zu derenatmosphärischem Budget bei. Strahlungsnebel beeinflußt durchAufnahme, Entfernen und Prozessieren von Aerosolpartikelnund löslichen Spurengasen deren Konzentrationen in derGasphase. In dieser Arbeit wird erstmalig ein Modell präsentiert,welches die Simulation des Austausches zwischen Atmosphäreund Biosphäre unter Berücksichtigung der dynamischenWechselwirkung zwischen Strahlungsnebel, Blattflächenwasserund Mehrphasenchemie ermöglicht. Numerische Fallstudien mitfolgenden Schwerpunkten werden präsentiert: - Einfluß von Vegetation und Blattflächenwasser auf diezeitlichen und räumlichen Schwankungen derGrößenabhängigkeit der Flüssigphasenkonzentrationen inNebeltropfen, - Einfluß von Blattflächenwasser auf dieTrockendepositionsflüsse von Ammoniak im Wald - Simulationenwurden mit einem neuen dynamischen Depositionsmodelldurchgeführt und mit dem Widerstandsansatz verglichen -, - Einfluß von physikalischen und chemischen Prozessen aufdie Reduktion von NO- und Isoprenemissionen aus demWaldbestand verglichen mit den primären Emissionen.

ZusammenfassungDie Analyse von Isotopenverhältnissen ist von wachsender Bedeutung bei der Untersuchung von Quellen, Senken und chemischen Reaktionswegen atmosphärischer Spurengase. Distickstoffoxid (N2O) hat vier isotopisch einfach substituierte Spezies: 14N15N16O, 15N14N16O, 14N217O und 14N218O. In der vorliegenden Arbeit wurden massenspektrometrische Methoden entwickelt, die eine komplette Charakterisierung der Variationen im Vorkommen dieser Spezies ermöglichen. Es wird die bisher umfassendste Darstellung dieser Variationen in Troposphäre und Stratosphäre gegeben und mit Bezug auf eine Reihe von Laborexperimenten detailliert interpretiert.Die Laborexperimente machen einen großen Anteil dieser Doktorarbeit aus und konzentrieren sich auf die Isotopenfraktionierung in den stratosphärischen N2O-Senken, d. h. Photolyse und Reaktion mit elektronisch angeregten Sauerstoffatomen, O(1D). Diese Prozesse sind von dominantem Einfluß auf die Isotopenzusammensetzung von atmosphärischem N2O. Potentiell wichtige Parameter wie Temperatur- und Druckvariationen, aber auch Veränderungen der Wellenlänge im Fall der Photolyse wurden berücksichtigt. Photolyse bei stratosphärisch relevanten Wellenlängen > 190 nm zeigte immer Anreicherungen von 15N in beiden Stickstoffatomen des verbleibenden N2O wie auch in 17O und 18O. Die Anreicherungen waren am mittelständigen N-Atom signifikant höher als am endständigen N (mit mittleren Werten für 18O) und stiegen zu größeren Wellenlängen und niedrigeren Temperaturen hin an. Erstmalig wurden für 18O und 15N am endständigen N-Atom Isotopenabreicherungen bei 185 nm-Photolyse festgestellt. Im Gegensatz zur Photolyse waren die Isotopenanreicherungen bei der zweiten wichtigen N2O-Senke, Reaktion mit O(1D) vergleichsweise gering. Jedoch war das positionsabhängige Fraktionierungsmuster dem der Photolyse direkt entgegengesetzt und zeigte größere Anreicherungen am endständigen N-Atom. Demgemäß führen beiden Senkenprozesse zu charakteristischen Isotopensignaturen in stratosphärischem N2O. Weitere N2O-Photolyseexperimente zeigten, daß 15N216O in der Atmosphäre höchstwahrscheinlich mit der statistisch zu erwartenden Häufigkeit vorkommt.Kleine stratosphärische Proben erforderten die Anpassung der massenspektrometrischen Methoden an Permanentflußtechniken, die auch für Messungen an Firnluftproben von zwei antarktischen Stationen verwendet wurden. Das 'Firnluftarchiv' erlaubte es, den gegenwärtigen Trend und die präindustriellen Werte der troposphärischen N2O-Isotopensignatur zu bestimmen. Ein daraus konstruiertes globales N2O-Isotopenbudget ist im Einklang mit den besten Schätzungen der Gesamt-N2O-Emissionen aus Böden und Ozeanen.17O-Messungen bestätigten die Sauerstoffisotopenanomalie in atmosphärischem N2O, zeigten aber auch, daß N2O-Photolyse die Sauerstoffisotope gemäß einem massenabhängigen Fraktionierungsgesetz anreichert. Eine troposphärische Ursache für einen Teil des Exzeß-17O wurde vorgeschlagen, basierend auf der Reaktion von NH2 mit NO2, wodurch die Sauerstoffisotopenanomalie von O3 über NO2 an N2O übertragen wird.

Zusammenfassung Um zu einem besseren Verständnis des Prozesses der Biomineralisation zu gelangen, muss das Zusammenwirken der verschiedenen Typen biologischer Makromoleküle, die am Keimbildungs- und Wachstumsprozess der Minerale beteiligt sind, berücksichtigt werden. In dieser Arbeit wird ein neues Modellsystem eingeführt, das aus einem SAM (self-assembled monolayer) mit verschiedenen Funktionalitäten und unterschiedlichen, gelösten Makromolekülen besteht. Es konnte gezeigt werden, dass die Kristallisation von Vaterit (CaCO3) sowie Strontianit (SrCO3) Nanodrähten der Präsenz von Polyacrylat in Kooperation mit einer COOH-funktionalisierten SAM-Oberfläche zugeschrieben werden kann. Die Kombination bestehend aus einer polaren SAM-Oberfläche und Polyacrylat fungiert als Grenzfläche für die Struktur dirigierende Kristallisation von Nanodraht-Kristallen. Weiter konnte gezeigt werden, dass die Phasenselektion von CaCO3 durch die kooperative Wechselwirkung zwischen einer SAM-Oberfläche und einem daran adsorbierten hb-Polyglycerol kontrolliert wird. Auch die Funktionalität einer SAM-Oberfläche in Gegenwart von Carboxymethyl-cellulose übt einen entscheidenden Einfluss auf die Phasenselektion des entstehenden Produktes aus. In der vorliegenden Arbeit wurden Untersuchungen an CaCO3 zur homogenen Keimbildung, zur Nukleation in Gegenwart eines Proteins sowie auf Kolloiden, die als Template fungieren, mittels Kleinwinkel-Neutronenstreuung durchgeführt. Die homogene Kristallisation in wässriger Lösung stellte sich als ein mehrstufiger Prozess heraus. In Gegenwart des Eiweißproteins Ovalbumin konnten drei Phasen identifiziert werden, darunter eine anfänglich vorhandene amorphe sowie zwei kristalline Phasen.

A study of maar-diatreme volcanoes has been perfomed by inversion of gravity and magnetic data. The geophysical inverse problem has been solved by means of the damped nonlinear least-squares method. To ensure stability and convergence of the solution of the inverse problem, a mathematical tool, consisting in data weighting and model scaling, has been worked out. Theoretical gravity and magnetic modeling of maar-diatreme volcanoes has been conducted in order to get information, which is used for a simple rough qualitative and/or quantitative interpretation. The information also serves as a priori information to design models for the inversion and/or to assist the interpretation of inversion results. The results of theoretical modeling have been used to roughly estimate the heights and the dip angles of the walls of eight Eifel maar-diatremes — each taken as a whole. Inversemodeling has been conducted for the Schönfeld Maar (magnetics) and the Hausten-Morswiesen Maar (gravity and magnetics). The geometrical parameters of these maars, as well as the density and magnetic properties of the rocks filling them, have been estimated. For a reliable interpretation of the inversion results, beside the knowledge from theoretical modeling, it was resorted to other tools such like field transformations and spectral analysis for complementary information. Geologic models, based on thesynthesis of the respective interpretation results, are presented for the two maars mentioned above. The results gave more insight into the genesis, physics and posteruptive development of the maar-diatreme volcanoes. A classification of the maar-diatreme volcanoes into three main types has been elaborated. Relatively high magnetic anomalies are indicative of scoria cones embeded within maar-diatremes if they are not caused by a strong remanent component of the magnetization. Smaller (weaker) secondary gravity and magnetic anomalies on the background of the main anomaly of a maar-diatreme — especially in the boundary areas — are indicative for subsidence processes, which probably occurred in the late sedimentation phase of the posteruptive development. Contrary to postulates referring to kimberlite pipes, there exists no generalized systematics between diameter and height nor between geophysical anomaly and the dimensions of the maar-diatreme volcanoes. Although both maar-diatreme volcanoes and kimberlite pipes are products of phreatomagmatism, they probably formed in different thermodynamic and hydrogeological environments. In the case of kimberlite pipes, large amounts of magma and groundwater, certainly supplied by deep and large reservoirs, interacted under high pressure and temperature conditions. This led to a long period phreatomagmatic process and hence to the formation of large structures. Concerning the maar-diatreme and tuff-ring-diatreme volcanoes, the phreatomagmatic process takes place due to an interaction between magma from small and shallow magma chambers (probably segregated magmas) and small amounts of near-surface groundwater under low pressure and temperature conditions. This leads to shorter time eruptions and consequently to structures of smaller size in comparison with kimberlite pipes. Nevertheless, the results show that the diameter to height ratio for 50% of the studied maar-diatremes is around 1, whereby the dip angle of the diatreme walls is similar to that of the kimberlite pipes and lies between 70 and 85°. Note that these numerical characteristics, especially the dip angle, hold for the maars the diatremes of which — estimated by modeling — have the shape of a truncated cone. This indicates that the diatreme can not be completely resolved by inversion.

Proper ion channels’ functioning is a prerequisite for a normal cell and disorders involving ion channels, or channelopathies, underlie many human diseases. Long QT syndromes (LQTS) for example may arise from the malfunctioning of hERG channel, caused either by the binding of drugs or mutations in HERG gene. In the first part of this thesis I present a framework to investigate the mechanism of ion conduction through hERG channel. The free energy profile governing the elementary steps of ion translocation in the pore was computed by means of umbrella sampling simulations. Compared to previous studies, we detected a different dynamic behavior: according to our data hERG is more likely to mediate a conduction mechanism which has been referred to as “single-vacancy-like” by Roux and coworkers (2001), rather then a “knock-on” mechanism. The same protocol was applied to a model of hERG presenting the Gly628Ser mutation, found to be cause of congenital LQTS. The results provided interesting insights about the reason of the malfunctioning of the mutant channel. Since they have critical functions in viruses’ life cycle, viral ion channels, such as M2 proton channel, are considered attractive targets for antiviral therapy. A deep knowledge of the mechanisms that the virus employs to survive in the host cell is of primary importance in the identification of new antiviral strategies. In the second part of this thesis I shed light on the role that M2 plays in the control of electrical potential inside the virus, being the charge equilibration a condition required to allow proton influx. The ion conduction through M2 was simulated using metadynamics technique. Based on our results we suggest that a potential anion-mediated cation-proton exchange, as well as a direct anion-proton exchange could both contribute to explain the activity of the M2 channel.

In dieser Arbeit wurde die Elektronenemission von Nanopartikeln auf Oberflächen mittels spektroskopischen Photoelektronenmikroskopie untersucht. Speziell wurden metallische Nanocluster untersucht, als selbstorganisierte Ensembles auf Silizium oder Glassubstraten, sowie ferner ein Metall-Chalcogenid (MoS2) Nanoröhren-Prototyp auf Silizium. Der Hauptteil der Untersuchungen war auf die Wechselwirkung von fs-Laserstrahlung mit den Nanopartikeln konzentriert. Die Energie der Lichtquanten war kleiner als die Austrittsarbeit der untersuchten Proben, so dass Ein-Photonen-Photoemission ausgeschlossen werden konnte. Unsere Untersuchungen zeigten, dass ausgehend von einem kontinuierlichen Metallfilm bis hin zu Clusterfilmen ein anderer Emissionsmechanismus konkurrierend zur Multiphotonen-Photoemission auftritt und für kleine Cluster zu dominieren beginnt. Die Natur dieses neuen Mechanismus` wurde durch verschiedenartige Experimente untersucht. Der Übergang von einem kontinuierlichen zu einem Nanopartikelfilm ist begleitet von einer Zunahme des Emissionsstroms von mehr als eine Größenordnung. Die Photoemissions-Intensität wächst mit abnehmender zeitlicher Breite des Laserpulses, aber diese Abhängigkeit wird weniger steil mit sinkender Partikelgröße. Die experimentellen Resultate wurden durch verschiedene Elektronenemissions-Mechanismen erklärt, z.B. Multiphotonen-Photoemission (nPPE), thermionische Emission und thermisch unterstützte nPPE sowie optische Feldemission. Der erste Mechanismus überwiegt für kontinuierliche Filme und Partikel mit Größen oberhalb von mehreren zehn Nanometern, der zweite und dritte für Filme von Nanopartikeln von einer Größe von wenigen Nanometern. Die mikrospektroskopischen Messungen bestätigten den 2PPE-Emissionsmechanismus von dünnen Silberfilmen bei „blauer“ Laseranregung (hν=375-425nm). Das Einsetzen des Ferminiveaus ist relativ scharf und verschiebt sich um 2hν, wenn die Quantenenergie erhöht wird, wogegen es bei „roter“ Laseranregung (hν=750-850nm) deutlich verbreitert ist. Es zeigte sich, dass mit zunehmender Laserleistung die Ausbeute von niederenergetischen Elektronen schwächer zunimmt als die Ausbeute von höherenergetischen Elektronen nahe der Fermikante in einem Spektrum. Das ist ein klarer Hinweis auf eine Koexistenz verschiedener Emissionsmechanismen in einem Spektrum. Um die Größenabhängigkeit des Emissionsverhaltens theoretisch zu verstehen, wurde ein statistischer Zugang zur Lichtabsorption kleiner Metallpartikel abgeleitet und diskutiert. Die Elektronenemissionseigenschaften bei Laseranregung wurden in zusätzlichen Untersuchungen mit einer anderen Anregungsart verglichen, der Passage eines Tunnelstroms durch einen Metall-Clusterfilm nahe der Perkolationsschwelle. Die elektrischen und Emissionseigenschaften von stromtragenden Silberclusterfilmen, welche in einer schmalen Lücke (5-25 µm Breite) zwischen Silberkontakten auf einem Isolator hergestellt wurden, wurden zum ersten Mal mit einem Emissions-Elektronenmikroskop (EEM) untersucht. Die Elektronenemission beginnt im nicht-Ohmschen Bereich der Leitungsstrom-Spannungskurve des Clusterfilms. Wir untersuchten das Verhalten eines einzigen Emissionszentrums im EEM. Es zeigte sich, dass die Emissionszentren in einem stromleitenden Silberclusterfilm Punktquellen für Elektronen sind, welche hohe Emissions-Stromdichten (mehr als 100 A/cm2) tragen können. Die Breite der Energieverteilung der Elektronen von einem einzelnen Emissionszentrum wurde auf etwa 0.5-0.6 eV abgeschätzt. Als Emissionsmechanismus wird die thermionische Emission von dem „steady-state“ heißen Elektronengas in stromdurchflossenen metallischen Partikeln vorgeschlagen. Größenselektierte, einzelne auf Si-Substraten deponierte MoS2-Nanoröhren wurden mit einer Flugzeit-basierten Zweiphotonen-Photoemissions-Spektromikroskopie untersucht. Die Nanoröhren-Spektren wiesen bei fs-Laser Anregung eine erstaunlich hohe Emissionsintensität auf, deutlich höher als die SiOx Substratoberfläche. Dagegen waren die Röhren unsichtbar bei VUV-Anregung bei hν=21.2 eV. Eine ab-initio-Rechnung für einen MoS2-Slab erklärt die hohe Intensität durch eine hohe Dichte freier intermediärer Zustände beim Zweiphotonen-Übergang bei hν=3.1 eV.

In dieser Arbeit werden die mikroskopischen, chemischen und spektroskopischen Charakteristika von 260 natürlichen Smaragden und 66 synthetischen „Smaragden“ untersucht. Die Konzentrationen der chemischen Elemente von Smaragden wurden mit Hilfe der LA-ICP-MS und EMS bestimmt. Ergänzende Raman- und IR spektroskopische Methoden ermöglichen es, die Herkunft der verschiedenen Smaragde und ihrer synthetischen Analoga zu bestimmen. Auf Grund der verschiedenen Gehalte von Si, Al und Be können synthetische „Smaragde“ von natürlichen getrennt werden. Die Smaragde von Malipo, Chivor und auch synthetische „Smaragde“ können von allen anderen natürlichen Smaragden wegen der unterschiedlichen Cr-, V-, und Fe-Gehalte von einander getrennt werden. Wegen der unterschiedlichen Mg-, Na-, K-Gehalte lassen sich eher „schiefer-gebundene“ Smaragde identifizieren. Dabei wird festgestellt, dass die Unterscheidung in „schiefer-„ und „nichtschiefer-gebundene“ Smaragd-Vorkommen im Wesentlichen nur die Endglieder einer offensichtlich kristallchemisch sehr variablen Mineralchemie der Berylle, bzw. Smaragde beschreibt, dass damit aber keinesfalls eine petrologisch vertretbare Trennung belegbar ist, sondern dass Smaragde nur das jeweils regierende chemische Regime unter geeigneten Druck-Temperatur-Bedingungen widerspiegeln. Einschlussmerkmale spielen eine große Rolle bei der Unterscheidung verschiedener Lagerstätten und Herstellungsmethoden. Zum Beispiel können die Smaragde der drei Lagerstätten Santa Terezinha, Chivor, und Kafubu mit Hilfe ihrer charakteristischen Pyriteinschlüsse identifiziert werden. Die Band-Positionen und FWHM -Werte der Raman-Bande bei 1068 cm-1 und der IR-Bande bei 1200 cm-1 ermöglichen eine Differenzierung zwischen synthetischen und natürlichen Smaragden, und können darüber hinaus auch Auskunft geben über die Lagerstätte. Zusammen mit chemischen Messwerten kann bewiesen werden, dass diese Banden von Si-O Schwingungen verursacht werden. Die Raman- und IR-Banden im Bereich der Wasserschwingungen und insbesondere das IR-Band um 1140 cm-1 führen zur Trennung von Flux-Synthesen, Hydrothermal-Synthesen und natürlichen Smaragden.

Membrane proteins play a major role in every living cell. They are the key factors in the cell’s metabolism and in other functions, for example in cell-cell interaction, signal transduction, and transport of ions and nutrients. Cytochrome c oxidase (CcO), as one of the membrane proteins of the respiratory chain, plays a significant role in the energy transformation of higher organisms. CcO is a multi centered heme protein, utilizing redox energy to actively transport protons across the mitochondrial membrane. One aim of this dissertation is to investigate single steps in the mechanism of the ion transfer process coupled to electron transfer, which are not fully understood. The protein-tethered bilayer lipid membrane is a general approach to immobilize membrane proteins in an oriented fashion on a planar electrode embedded in a biomimetic membrane. This system enables the combination of electrochemical techniques with surface enhanced resonance Raman (SERRS), surface enhanced reflection absorption infrared (SEIRAS), and surface plasmon spectroscopy to study protein mediated electron and ion transport processes. The orientation of the enzymes within the surface confined architecture can be controlled by specific site-mutations, i.e. the insertion of a poly-histidine tag to different subunits of the enzyme. CcO can, thus, be oriented uniformly with its natural electron pathway entry pointing either towards or away from the electrode surface. The first orientation allows an ultra-fast direct electron transfer(ET) into the protein, not provided by conventional systems, which can be leveraged to study intrinsic charge transfer processes. The second orientation permits to study the interaction with its natural electron donor cytochrome c. Electrochemical and SERR measurements show conclusively that the redox site structure and the activity of the surface confined enzyme are preserved. Therefore, this biomimetic system offers a unique platform to study the kinetics of the ET processes in order to clarify mechanistic properties of the enzyme. Highly sensitive and ultra fast electrochemical techniques allow the separation of ET steps between all four redox centres including the determination of ET rates. Furthermore, proton transfer coupled to ET could be directly measured and discriminated from other ion transfer processes, revealing novel mechanistic information of the proton transfer mechanism of cytochrome c oxidase. In order to study the kinetics of the ET inside the protein, including the catalytic center, time resolved SEIRAS and SERRS measurements were performed to gain more insight into the structural and coordination changes of the heme environment. The electrical behaviour of tethered membrane systems and membrane intrinsic proteins as well as related charge transfer processes were simulated by solving the respective sets of differential equations, utilizing a software package called SPICE. This helps to understand charge transfer processes across membranes and to develop models that can help to elucidate mechanisms of complex enzymatic processes.

In the last few years the resolution of numerical weather prediction (nwp) became higher and higher with the progresses of technology and knowledge. As a consequence, a great number of initial data became fundamental for a correct initialization of the models. The potential of radar observations has long been recognized for improving the initial conditions of high-resolution nwp models, while operational application becomes more frequent. The fact that many nwp centres have recently taken into operations convection-permitting forecast models, many of which assimilate radar data, emphasizes the need for an approach to providing quality information which is needed in order to avoid that radar errors degrade the model's initial conditions and, therefore, its forecasts. Environmental risks can can be related with various causes: meteorological, seismical, hydrological/hydraulic. Flash floods have horizontal dimension of 1-20 Km and can be inserted in mesoscale gamma subscale, this scale can be modeled only with nwp model with the highest resolution as the COSMO-2 model. One of the problems of modeling extreme convective events is related with the atmospheric initial conditions, in fact the scale dimension for the assimilation of atmospheric condition in an high resolution model is about 10 Km, a value too high for a correct representation of convection initial conditions. Assimilation of radar data with his resolution of about of Km every 5 or 10 minutes can be a solution for this problem. In this contribution a pragmatic and empirical approach to deriving a radar data quality description is proposed to be used in radar data assimilation and more specifically for the latent heat nudging (lhn) scheme. Later the the nvective capabilities of the cosmo-2 model are investigated through some case studies. Finally, this work shows some preliminary experiments of coupling of a high resolution meteorological model with an Hydrological one.

The Alzheimer’s disease (AD), the most prevalent form of age-related dementia, is a multifactorial and heterogeneous neurodegenerative disease. The molecular mechanisms underlying the pathogenesis of AD are yet largely unknown. However, the etiopathogenesis of AD likely resides in the interaction between genetic and environmental risk factors. Among the different factors that contribute to the pathogenesis of AD, amyloid-beta peptides and the genetic risk factor apoE4 are prominent on the basis of genetic evidence and experimental data. ApoE4 transgenic mice have deficits in spatial learning and memory associated with inflammation and brain atrophy. Evidences suggest that apoE4 is implicated in amyloid-beta accumulation, imbalance of cellular antioxidant system and in apoptotic phenomena. The mechanisms by which apoE4 interacts with other AD risk factors leading to an increased susceptibility to the dementia are still unknown. The aim of this research was to provide new insights into molecular mechanisms of AD neurodegeneration, investigating the effect of amyloid-beta peptides and apoE4 genotype on the modulation of genes and proteins differently involved in cellular processes related to aging and oxidative balance such as PIN1, SIRT1, PSEN1, BDNF, TRX1 and GRX1. In particular, we used human neuroblastoma cells exposed to amyloid-beta or apoE3 and apoE4 proteins at different time-points, and selected brain regions of human apoE3 and apoE4 targeted replacement mice, as in vitro and in vivo models, respectively. All genes and proteins studied in the present investigation are modulated by amyloid-beta and apoE4 in different ways, suggesting their involvement in the neurodegenerative mechanisms underlying the AD. Finally, these proteins might represent novel potential diagnostic and therapeutic targets in AD.
