The acquisition of oligosaccharides from chitosan has been the subject of several studies in the pharmaceutical, biochemical, food and medical due to functional properties of these compounds. This study aimed to boost its production of chitooligosaccharides (COS) through the optimization of production and characterization of chitosanolytic enzymes secreted by microorganisms Paenibacillus chitinolyticus and Paenibacillus ehimensis, and evaluating the antioxidant potential of the products obtained. In the process of optimizing the production of chitosanase were employed strategies Fractional Factorial Experimental Design and Central Composite Rotatable Design. The results identified the chitosan, peptone and yeast extract as the components that influenced the production of chitosanase by these microorganisms. With the optimization of the culture media was possible to obtain an increase of approximately 8.1 times (from 0.043 to 0.35 U.mL U.mL-1) and 7.6 times (from 0.08 U.mL-1 to 0.61 U.mL-1) in the enzymatic activity of chitosanase produced by P. chitinolyticus and P. ehimensis respectively. Enzyme complexes showed high stability in temperature ranges between 30º and 55º C and pH between 5.0 and 9.0. Has seen the share of organic solvents, divalent ions and other chemical agents on the activity of these enzymes, demonstrating high stability of these crude complexes and dependence of Mn2+. The COS generated showed the ability of DPPH radical scavenging activity, reaching a maximum rate of scavenging of 61% and 39% when they were produced with enzymes of P. ehimensis and P. chitinolyticus respectively. The use of these enzymes in raw form might facilitate its use for industrial applications

Neste trabalho foram analisados os parâmetros cinéticos e a atividade colinesterásica de duas espécies de peixes estuarinos: a corvina Micropogonias furnieri (Teleostei, Scianidae) e o bagre Cathorops spixii (Teleostei, Ariidae), buscando avaliar o uso destas espécies como bioindicadores da presença de compostos anticolinesterásicos no meio aquático. Os exemplares de corvina e bagre, respectivamente, foram coletados nos estados do RS (Lagoa dos Patos) e PR (Baía de Paranaguá) no inverno (corvina) e verão (corvina e bagre) em dois pontos de coleta: um local controle e outro poluído. Os peixes foram anestesiados (bezocaína, 200 ppm) e os cérebros dissecados, homogeneizados e centrifugados. Foram estimados os parâmetros cinéticos (Vmax e Kmap), utilizando iodeto de acetiltiocolina como substrato, nas seguintes concentrações: 0,025; 0,05; 0,2; 0,8; 1,6; 3,2 e 9 mM. Nos estudos de inibição enzimática foi utilizado o carbamato eserina em concentrações de 0,3 a 10 mM e de 1x10-4 a 1 µM, a fim de determinar os parâmetros cinéticos de inibição e a concentração de eserina que inibia 50% da atividade colinesterásica (CI50), respectivamente. Os resultados mostraram pouca variação nos valores de Kmap nas duas espécies, porém diferenças significativas nos valores de CI50 foram observadas, indicando que a ChE do cérebro da corvina M. furnieri é resistente à inibição por eserina. Nos estudos de cinética de inibição da ChE do cérebro do bagre C. spixii, foram encontradas diferenças entre alguns parâmetros, quando foram comparados os peixes coletados no local controle com aqueles coletados no local poluído. Houve uma maior atividade colinesterásica de bagres coletados no local poluído (p<0,05), sendo que o mesmo resultado foi observado em exemplares de corvinas M. furnieri coletados durante o inverno no local poluído. Os resultados obtidos in vitro demonstram que a ChE de M. furnieri possui pouca sensibilidade à eserina. 5 Porém, o fato de ter sido registrada inibição colinesterásica nos organismos coletados durante o verão região poluída sugere a possibilidade de alterações significativas na capacidade de bio-oxidação de pesticidas nesta espécie, ou a possibilidade de inibição por outros contaminantes que também afetam a atividade colinesterásica, como por exemplo, metais. No caso de C. spixii, a determinação dos parâmetros cinéticos e de atividade colinesterásica sugere que pode estar ocorrendo alterações bioquímicas nos peixes previamente expostos a contaminantes no ambiente, provavelmente devido a respostas adaptativas ao ambiente impactado. Desta forma, o presente estudo indica a importância de estudos cinéticos prévios, quando se utiliza ou pretende utilizar, a atividade colinesterásica como bioindicador da presença de compostos anticolinesterásicos no ambiente em espécies aquáticas.

Marine algae are one of the major sources of biologic compounds. In extracellular matrix of these organisms there are sulfated polysaccharides that functions as structural components and provides protection against dehydration. The fraction 1.0 (F1.0) rich in sulfated galactans obtained from red seaweed Hypnea musciformis was physicochemical characterized and evaluated for pharmacologic activity through antioxidant activity, cytotoxic action on erythrocytes, anticoagulant, stimulatory action under antithrombotic heparan sulfate synthesis and their effects on cell proliferation and cycle cell progression. The main components of F1.0 were carbohydrates (49.70 ± 0.10%) and sulfate (44.59 ± 0.015%), presenting phenolic compounds (4.79 ± 0.016%) and low protein contamination (0.92 ± 0.001%). Fraction 1.0 showed polidisperse profile and signs in infrared analysis in 1262, 1074 and 930, 900 and 850 attributed to sulfate esters S=O bond, presence of a 3,6- anidrogalactose C-O bond, non-sulfated β-D-galactose and a C-O-SO4 bond in galactose C4, respectively. The fraction rich in sulfated galactans exhibited strong antioxidant action under lipid peroxidation assay with IC50 of 0.003 mg/mL. Besides the inhibition of hemolysis induced by H2O2 in erythrocytes treated with F1.0, this fraction did not promote significant cytotoxity under erythrocytes membranes. F1.0 exhibited low anticoagulant activity causing moderate direct inhibition of enzimatic activity of thrombin. This fraction promoted stimulation around of 4.6 times on this synthesis of heparan sulfate (HS) by rabbit aortic endothelial cells (RAEC) in culture when was compared with non treated cells. The fraction of this algae displayed antiproliferative action under RAEC cells causing incresing on cell number on S fase, blocking the cycle cell progression. Thus F1.0 presented cytostatic and no cytotoxic action under this cell lineage. These results suggest that F1.0 from H. musciformis have antioxidant potential which is a great effect for a compound used as food and in food industry which could be an alternative to food industry to prevent quality decay of lipid containing food due to lipid peroxidation. These polysaccharides prevent the lipid peroxidation once the fraction in study exhibited strong inhibitory action of this process. Furthermore that F1.0 present strong antithrombotic action promoting the stimulation of antithrombotic HS synthesis by endothelial cells, being important for thrombosis preventing, by its inhibitory action under reactive oxygen species (ROS) in some in vitro methods, being involved in promotion of hypercoagulability state.

Heparin is a pharmaceutical animal widely used in medicine due to its potent anticoagulant effect. Furthermore, it has the ability to inhibit the proliferation, invasion and adhesion of cancer cells to vascular endothelium. However, its clinical applicability can be compromised by side effects such as bleeding. Thus, the search for natural compounds with low bleeding risk and possible therapeutic applicability has been targeted by several research groups. From this perspective, this study aims to evaluate the hemorrhagic and anticoagulant activities and citotoxic effect for different tumor cell lines (HeLa, B16-F10, HepG2, HS-5,) and fibroblast cells (3T3) of the Heparin-like from the crab Chaceon fenneri (HEP-like). The HEP-like was purified after proteolysis, ion-exchange chromatography, fractionation with acetone and characterized by electrophoresis (agarose gel) and enzymatic degradation. Hep-like showed eletroforetic behavior similar to mammalian heparin, and high trisulfated /Nacetylated disaccharides ratio. In addition, HEP-like presented low in vitro anticoagulant activity using aPTT and a minor hemorrhagic effect when compared to mammalian heparin. Furthermore, the HEP-like showed significant cytotoxic effect (p<0.001) on HeLa, HepG2 and B16-F10 tumor cells with IC50 values of 1000 ug/mL, after incubation for 72 hours. To assess the influence of heparin-like on the cell cycle in HeLa cells, analysis was performed by flow cytometry. The results of this analysis showed that HEP-like influence on the cell cycle increasing S phase and decreasing phase G2. Thus, these properties of HEP-like make these compounds potential therapeutic agents

This report shows 2232 times purification of a βNAcetylhexosaminidase from hepatic extracts from the sea mammal Sotalia fluviatilis homogenate with final recovery of 8,4%. Sequenced steps were utilized for enzyme purification: ammonium sulfate fractionation, Biogel A 1.5 m, chitin, DEAESepharose and hydroxyapatite chromatographies. The protein molecular mass was estimated in 10 kDa using SDSPAGE and confirmed by MALDITOF. It was found to have an optimal pH of 5.0 and a temperature of 60°C. Using pnitrophenylNAcetylβDglycosaminide apparent Km and Vmax values were of 2.72 mM and 0.572 nmol/mg/min, respectively. The enzyme was inhibited by mercury chloride (HgCl2) and sodium dodecil sulfate (SDS)

Plodia interpunctella (Indian meal moth) is a cosmopolitan pest that attacks not only a wide range of stored grain as well other food products. Due to its economic importance several researches have focused in a method with ability to control this pest with few or no damage to the environment. The study of digestive enzymes inhibitors, lectins and chitin-binding proteins, has often been proposed as an alternative to reduce insect damage. In this study we report the major classes of digestive enzymes during larval growth in P. Interpunctella, being those proteinases actives at pH 9.5 and optimum temperature of 50 oC to both larvae of the 3rd instar and pre-pupal stage of development. In vitro and zymogram assays presented the effects of several inhibitors, such as SBTI, TLCK and PMSF to intestinal homogenate of 3rd instar larvae of 62%, 92% and 87% of inhibition and In pre-pupal stage of 87%, 62 % and 55% of inhibition, respectively. Zymograms showed inhibition of two low molecular masses protein bands by TLCK and that in presence of SBTI were retarded. These results are indicative of predominance of digestive serine proteinases in gut homogenate from Plodia interpunctella larvae. This serine proteinase was then used as a target to evaluate the effect of SBTI on larvae in in vivo assay. Effect of SBTI on mortality and larval mass was not observed at until 4% of concentration (w/w) in diets. Chitin, another target to insecticidal proteins, was observed by chemical method. Moreover, optic microscopy confirmed the presence of a peritrophic membrane. Established this target, in vivo effect of EvV, a chitin binding vicilin, evaluated during the larval development of P. interpunctella and was obtained a LD50 of 0,23% and WD50 of 0,27% to this protein. Mechanism of action was proposed through of the in vivo digestibility of EvV methodology. During the passage through the larval digestive tract was observed that EvV was susceptible to digestive enzymes and a reactive fragment, visualized by Western blotting, produced by digestion was recovered after dissociation of the peritrophic membrane. The bound of EvV to peritrophic membrane was confirmed by immunohystochemical assays that showed strong immunofluorescent signal of EvV-FITC binding and peritrophic membrane. These results are a indicative that vicilins could be utilized as potential insecticide to Plodia interpunctella and a control methods using EvV as bioinsecticide should be studied to reduce lost caused by storage insect pests

Propósito y método del estudio: Cuando se ingieren grandes cantidades de fruto de Karwinskia humboldtiana se produce una intoxicación aguda, que ocasiona daño en múltiples órganos, falla respiratoria y muerte en pocos días. En la intoxicación accidental y experimental con este fruto, se ha reportado daño histológico en pulmones, hígado y riñones. Se sabe que el daño histológico a estos órganos, además de la falla multiorgánica, son situaciones comunes cuando existe daño pancreático, sin embargo, hasta la fecha, el páncreas no ha sido estudiado en esta intoxicación. En este trabajo examinamos el efecto que ocasiona la intoxicación aguda con el fruto de Karwinskia humboldtiana en el páncreas en la rata Wistar. Contribuciones y conclusiones: En este trabajo se encontró daño progresivo confinado a la porción exocrina del páncreas, iniciando con reducción en el tamaño de los acinos pancreáticos, así como del número de gránulos de zimógeno, presencia de vesículas de apariencia autofágica y apoptosis, seguidos de edema, infiltrado inflamatorio, necrosis y pérdida completa de la arquitectura acinar. Cabe señalar que la morfología de los islotes de Langerhans se mantuvo conservada en todos los tiempos evaluados en este trabajo. Mediante las técnicas Western Blot e inmunofluorescencia, analizamos la expresión y localización de proteínas implicadas en la autofagia (LC3-I y LC3-II) y en la apoptosis (caspasa-3). Observamos que las proteínas LC3-I y LC3-II se encuentran expresadas en todos los tiempos experimentales, mientras que la proteína caspasa-3 se expresó únicamente a las 48 h de la intoxicación con Karwinskia humboldtiana. Mediante ensayos de histoquímica enzimática para la citocromo oxidasa c, comprobamos que desde las 24 h de intoxicación, existen cambios en la forma, tamaño y localización de las mitocondrias, organelos implicados en la muerte celular. Asimismo, realizamos la evaluación de la función pancreática mediante la determinación de la actividad de la amilasa sérica, sin encontrar diferencia significativa entre los grupos estudiados. Todos estos datos indican que el daño inducido por una dosis alta de fruto de Karwinskia humboldtiana en la rata Wistar, es consistente con una pancreatitis aguda necrotizante que afecta exclusivamente el páncreas exocrino. Este modelo de pancreatitis puede ser útil para el estudio de los mecanismos celulares y moleculares implicados en este padecimiento, así como para el ensayo de posibles tratamientos para esta y otras enfermedades pancreáticas.

Aunque hace más de 50 años que se describió que la glutamato descarboxilasa (GAD) lleva a cabo la descarboxilación del glutamato para producir GABA, y en animales ha sido muy estudiada debido al papel del GABA como neurotransmisor, la información disponible sobre las GADs de plantas es aún limitada, conociéndose sólo algunos aspectos de la regulación por calcio de su actividad enzimática o de expresión de algunos de los genes de su familia génica. El GABA es un metabolito que tradicionalmente se ha asociado a estrés, pero su papel en plantas todavía no está claro. En las últimas dos décadas los resultados experimentales obtenidos sobre la GAD y el GABA, destacando las alteraciones fenotípicas mostradas por plantas tratadas con GABA y por plantas transgénicas para GAD, han generado preguntas interesantes sobre el posible papel de este metabolito y la enzima en señalización en plantas. En plantas, son varios los papeles que se han propuesto para el metabolismo del GABA tales como su participación como componente del metabolismo del carbono y del nitrógeno (Fait y col., 2008), protección frente especies reactivas de oxigeno (Liu y col., 2011), regulación de la expresión génica incluyendo la regulación de genes implicados en la síntesis de hormonas (Khatiresan y col., 1997; Shi y col., 2010; Lancien y Roberts, 2006) y señalización a larga distancia (Beuve y col., 2004) y en gradiente guiando el crecimiento del tubo polínico (Palanivelu y col., 2013). Nuestro grupo de investigación ha sugerido un papel novedoso para la producción de GABA durante la xilogénesis en pino (Molina-Rueda y col., 2010, 2015). En base a estos antecedentes, los objetivos planteados para este trabajo han sido: la asignación de posibles funciones a las GADs de Populus en condiciones normales de crecimiento y en estrés abióticos, estudiar la adquisición del dominio de unión a calmodulina (CaMBD) de las GADs de plantas vasculares y analizar el efecto del GABA y del glutamato en las raíces de Populus. Las conclusiones que se derivan de los resultados de este trabajo se detallan a continuación. El dominio de unión a calmodulina de la GAD de plantas esta conservado en GADs de plantas consideradas ancestros de plantas vasculares y ausente en plantas no vasculares, lo que sitúa juntos en la evolución los eventos de adquisición del dominio de unión a CaM y el desarrollo del tejido vascular de plantas. Los resultados similares de la localización de GABA en xilema y una expresión GAD asociada a la formación de madera de reacción tanto en pino como en chopo apuntan a un papel relevante de la producción de GABA durante la xilogénesis en leñosas. La familia génica GAD posee seis genes codificando todos ellos para proteínas aparentemente funcionales y susceptibles de ser reguladas por calcio. Esta familia génica ha sufrido duplicaciones y eventos de especialización durante la evolución de Populus. Este trabajo ha posibilitado la asociación entre papeles específicos y los diferentes genes de esta familia. Introducción: A mediados de los años 70’s del siglo pasado el descubrimiento de la tecnología del ADN recombinante marca el inicio de la era de la biotecnología moderna. La implementación de estas tecnologías permitió la utilización de organismos como sistemas de expresión que a lo largo de los años ha generado la producción de una gran variedad de productos biológicos. Dentro de estos sistemas Pichia pastoris es un sistema de expresión ampliamente utilizado debido a sus características tales como la producción de proteínas en grandes cantidades, la liberación de los productos al medio de cultivo, la obtención de productos complejos que requieren modificaciones postraduccionales típicas de los eucariotas o que contienen puentes disulfuro, entre otras. Nocardia brasiliensis es una bacteria parcialmente ácido-alcohol resistente la cual forma colonias granulares, con hifas aéreas escasas, sus colonias exhiben un color anaranjado pardo con bordes en blanco. N. brasiliensis es patógena para el ser humano y es el agente causal del actinomicetoma. El actinomicetoma es una enfermedad crónica generalmente localizada en las extremidades. Se caracteriza por ser un proceso lento de tumefacción con nódulos, abscesos y fístulas.La Superóxido Dismutasa (SOD) es una enzima reductora polimérica que cataliza la conversión del ión superóxido a peróxido de hidrógeno y oxígeno molecular. La SOD ha sido propuesta como un factor de virulencia de microorganismos patógenos, cuya acción consiste en bloquear los efectores oxidativos del estallido respiratorio iniciado por los fagocítos en el fagolisosoma. Este mecanismo ha sido descrito para bacterias de los géneros Mycobacterium, Rhodococcus y Nocardia. Objetivo: producir y caracterizar la Superóxido Dismutasa A (SODA) de Nocardia brasiliensis en Pichia pastoris. Metodología: se realizó el diseño de primers adicionando secuencias de sitios de corte para las enzimas XhoI y AvrII, así como una cola de histidinas en el extremo 5’ para la amplificación del gen sodA de N. brasiliensis a partir del ADN genómico de Nocardia brasiliensis. El amplicón se clonó en el vector de expresión pPIC9. Se llevó a cabo la transformación por electroporación de levaduras Pichia pastoris GS115. La producción de SOD se llevó a cabo en inducciones de 96 h con metanol como agente inductor. Los sobrenadantes se dializaron con membranas de celulosa. Los dializados se observaron por SDS-PAGE y western blot. Se analizó la actividad funcional de la enzima con el SOD Assay kit de Sigma Aldrich. Resultados: Por reacción en cadena de la polimerasa se obtuvo una secuencia de 625 pb correspondiente al gen sodA. El fragmento se ligó al vector de expresión pPIC9 y fue caracterizado con las enzimas de restricción XhoI y AvrII. Las cepas trasformadas de P. pastoris GS115 se caracterizaron con el gen aox1 obteniendo cepas Mut+ y Muts. Los análisis por SDSPAGE mostraron bandas no observadas en el control negativo de expresión mientras en los western blot solo una de las clonas mostró señal. Los análisis de actividad funcional sugieren inhibición de la reacción enzimática infiriendo presencia de la proteína SOD en el medio dializado. Conclusiones: Se logró la construcción del sistema de expresión Pichia pastoris con el casete de expresión de la SOD de N. brasiliensis. Así como la generación de cepas Mut+y Muts. En los ensayos de actividad funcional se observó inhibición de la reacción enzimática.

O objetivo da simulação é confrontar os instruendos com situações que permitam recriar as condições mais próximas da realidade, para que o processo de aprendizagem possa atingir os objetivos da formação. As polícias estrangeiras já utilizam simuladores nas áreas do ensino da condução, tiro, inativação de engenhos explosivos e fiscalização rodoviária. Cada equipamento ensina e desenvolve as capacidades dos agentes policiais, que por sua vez, contribui para a promoção da eficácia policial. Este estudo estabelece a ligação entre os conceitos de simulação e de formação no sentido de tentar desenvolver as linhas orientadoras da formação policial da GNR com recurso à simulação. Abstract: The intent of simulations is to immerse trainees in situations wich provide to recreate conditions similar to reality, for the purpose that the training process would reach training objectives. Law enforcement agencies abroad already use simulators for driver training, shooting, explosive ordinance disposal and road and transport enforcement. Each equipment teaches and enhance skills of individual officers which, in turn, will contribute to promote police effectiveness. This study aims to link the concepts of simulation and training in order to develop the guidelines of the GNR police training using simulators.

Vast montado areas are threatened by degradation, as the result of a long history of land use changes. Since improved pastures have been installed aiming soil quality improvement and system sustainability, it is crucial to evaluate the effects of these management changes on soil organic matter status and soil biological activity, as soil quality indicators. Therefore, a 35-yr old improved pasture and a natural pasture were studied, considering areas beneath tree canopy and in the open. Total organic C, total N, hot water soluble (HWS) and particulate (POM) C, microbial biomass C (MBC) and N (MBN), C mineralization rate (CMR) and net N mineralization rate (NMR) were determined. In addition, for a 1-yr period, soil β-glucosidase, urease, proteases and acid phosphomonoesterase were periodically determined. Improved pasture promoted the increase of soil C and N through POM-C increment, particularly beneath the trees canopies. The two study pastures did not show differences regarding soil microbial biomass, but variations in CMR, HWS-C and N availability (proteases and urease activities) suggest divergent soil microbial communities.
