Exogenous androgenic steroids applied to pregnant sheep programmes a PCOS-like phenotype in female offspring. Via ultrasound guidance we applied steroids directly to ovine fetuses at d62 and d82 of gestation, and examined fetal (day 90 gestation) and postnatal (11 months old) pancreatic structure and function. Of three classes of steroid agonists applied (androgen - Testosterone propionate (TP), estrogen - Diethystilbesterol (DES) and glucocorticoid - Dexamethasone (DEX)), only androgens (TP) caused altered pancreatic development. Beta cell numbers were significantly elevated in prenatally androgenised female fetuses (P=0.03) (to approximately the higher numbers found in male fetuses), whereas alpha cell counts were unaffected, precipitating decreased alpha:beta cell ratios in the developing fetal pancreas (P=0.001), sustained into adolescence (P=0.0004). In adolescence basal insulin secretion was significantly higher in female offspring from androgen-excess pregnancies (P=0.045), and an exaggerated, hyperinsulinaemic response to glucose challenge (P=0.0007) observed, whereas prenatal DES or DEX treatment had no effects upon insulin secretion. Postnatal insulin secretion correlated with beta cell numbers (P=0.03). We conclude that the pancreas is a primary locus of androgenic stimulation during development, giving rise to postnatal offspring whose pancreas secreted excess insulin due to excess beta cells in the presence of a normal number of alpha cells.

Orthobunyaviruses are the largest genus within the Bunyaviridae family, with over 170 named viruses classified into 18 serogroups (Elliott and Blakqori, 2001; Plyusnin et al., 2012). Orthobunyaviruses are transmitted by arthropods and have a tripartite negative sense RNA genome, which encodes 4 structural proteins and 2 non-structural proteins. The non-structural protein NSs is the primary virulence factor of orthobunyaviruses and potent antagonist of the type I interferon (IFN) response. However, sequencing studies have identified pathogenic viruses that lack the NSs protein (Mohamed et al., 2009; Gauci et al., 2010). The work presented in this thesis describes the molecular and biological characterisation of divergent orthobunyaviruses. Data on plaque morphology, growth kinetics, protein profiles, sensitivity to IFN and activation of the type I IFN system are presented for viruses in the Anopheles A, Anopheles B, Capim, Gamboa, Guama, Minatitlan, Nyando, Tete and Turlock serogroups. These are complemented with complete genome sequencing and phylogenetic analysis. Low activation of IFN by Tete serogroup viruses, which naturally lack an NSs protein, was also further investigated by the development of a reverse genetics system for Batama virus (BMAV). Recombinant viruses with mutations in the virus nucleocapsid protein amino terminus showed higher activation of type I IFN in vitro and data suggests that low levels of IFN are due to lower activation rather than active antagonism. The anti-orthobunyavirus activity of IFN-stimulated genes IFI44, IFITMs and human and ovine BST2 were also studied, revealing that activity varies not only within the orthobunyavirus genus and virus serogroups but also within virus species. Furthermore, there was evidence of active antagonism of the type I IFN response and ISGs by non-NSs viruses. In summary, the results show that pathogenicity in man and antagonism of the type I IFN response in vitro cannot be predicted by the presence, or absence, of an NSs ORF. They also highlight problems in orthobunyavirus classification with discordance between classical antigen based data and phylogenetic analysis.

Background: The use of artificial endoprostheses has become a routine procedure for knee and hip joints while ankle arthritis has traditionally been treated by means of arthrodesis. Due to its advantages, the implantation of endoprostheses is constantly increasing. While finite element analyses (FEA) of strain-adaptive bone remodelling have been carried out for the hip joint in previous studies, to our knowledge there are no investigations that have considered remodelling processes of the ankle joint. In order to evaluate and optimise new generation implants of the ankle joint, as well as to gain additional knowledge regarding the biomechanics, strain-adaptive bone remodelling has been calculated separately for the tibia and the talus after providing them with an implant. Methods: FE models of the bone-implant assembly for both the tibia and the talus have been developed. Bone characteristics such as the density distribution have been applied corresponding to CT scans. A force of 5,200 N, which corresponds to the compression force during normal walking of a person with a weight of 100 kg according to Stauffer et al., has been used in the simulation. The bone adaptation law, previously developed by our research team, has been used for the calculation of the remodelling processes. Results: A total bone mass loss of 2% in the tibia and 13% in the talus was calculated. The greater decline of density in the talus is due to its smaller size compared to the relatively large implant dimensions causing remodelling processes in the whole bone tissue. In the tibia, bone remodelling processes are only calculated in areas adjacent to the implant. Thus, a smaller bone mass loss than in the talus can be expected. There is a high agreement between the simulation results in the distal tibia and the literature regarding. Conclusions: In this study, strain-adaptive bone remodelling processes are simulated using the FE method. The results contribute to a better understanding of the biomechanical behaviour of the ankle joint and hence are useful for the optimisation of the implant geometry in the future.

A utilização de vermicompostos de diferentes resíduos agroindustriais na produção de mudas de acácianegra pode ser uma alternativa de reutilização de resíduos e aumentar a produção de mudas. Assim, os objetivos deste trabalho foram avaliar o crescimento e a concentração de nutrientes em mudas de acácianegra, cultivadas em substratos com diferentes vermicompostos de resíduos orgânicos agroindustriais. Instalou-se em casa de vegetação 11 diferentes tratamentos: T1) vermicomposto de esterco bovino (EB); T2) vermicomposto de esterco ovino (EO); T3) vermicomposto de lodo de parbolização de arroz (LP); T4) tratamento controle (sem adubação); T5) tratamento controle com adução mineral (NPK); T6) mistura de EB e LP; T7) mistura de EO e LP; T8) mistura de EB e vermicomposto de resíduos de alimentos (RA); T9) mistura de EO e RA; T10) mistura de EB e vermicomposto de resíduos de frutas (RF); T11) mistura de EO e RF. Após 180 dias de cultivo em recipiente com capacidade de cinco litros, foram analisadas a massa seca e a concentração de nutrientes na parte aérea da acácia-negra, e a concentração de nutrientes no solo, após o cultivo. A adição do esterco bovino, bem como a mistura de esterco bovino e resíduos alimentícios favoreceram o incremento de matéria seca das plantas de acácia-negra. Os resultados mostraram que as concentrações de nutrientes nas plantas, com exceção de Fe e Mn, variaram com adição de vermicompostos no solo. Os tratamentos T3 e T6 elevaram as concentrações em P, N, Zn de Cu nas folhas de acácia-negra. Além disso, a adição dos vermicompostos ao solo aumentou a disponibilidade de nutrientes para as plantas, mesmo após o cultivo, especialmente com relação ao fósforo, potássio e magnésio, sendo uma alternativa viável e eficaz na produção de mudas, podendo substituir a utilização de adubação mineral.

This work presents an application of optical fiber sensors based on Bragg gratings integrated to a transtibial prosthesis tube manufactured with a polymeric composite systrem of epoxy resin reinforced with glass fiber. The main objective of this study is to characterize the sensors applied to the gait cycle and changes in the gravity center of a transtibial amputee, trough the analysis of deformation and strengh of the transtibial prosthesis tube. For this investigation it is produced a tube of the composite material described above using the molding method of resin transfer (RTM) with four optical sensors. The prosthesis in which the original tube is replaced is classified as endoskeletal, has vacuum fitting, aluminium conector tube and carbon fiber foot cushioning. The volunteer for the tests was a man of 41 years old, 1.65 meters tall, 72 kilograms and left-handed. His amputation occurred due to trauma (surgical section is in the medial level, and was made below the left lower limb knee). He has been a transtibial prosthesis user for two years and eight months. The characterization of the optical sensors and analysis of mechanical deformation and tube resistance occurred through the gait cycle and the variation of the center of gravity of the body by the following tests: stand up, support leg without the prosthesis, support in the leg with the prosthesis, walk forward and walk backward. Besides the characterization of optical sensors during the gait cycle and the variation of the gravity center in a transtibial amputated, the results also showed a high degree of integration of the sensors in the composite and a high mechanical strength of the material.

Background: Material wear testing is an important technique in the development and evaluation of materials for use in implant for total knee arthroplasty. Since a knee joint induces a complex rolling-gliding movement, standardised material wear testing devices such as Pin-on-Disc or Ring-on-Disc testers are suitable to only a limited extent because they generate pure gliding motion only.Methods: A rolling-gliding wear simulator was thus designed, constructed and implemented, which simulates and reproduces the rolling-gliding movement and loading of the knee joint on specimens of simplified geometry. The technical concept was to run a base-plate, representing the tibia plateau, against a pivoted cylindrical counter-body, representing one femur condyle under an axial load. A rolling movement occurs as a result of the friction and pure gliding is induced by limiting the rotation of the cylindrical counter-body. The set up also enables simplified specimens handling and removal for gravimetrical wear measurements. Long-term wear tests and gravimetrical wear measurements were carried out on the well known material pairings: cobalt chrome-polyethylene, ceramic-polyethylene and ceramic-ceramic, over three million motion cycles to allow material comparisons to be made.Results: The observed differences in wear rates between cobalt-chrome on polyethylene and ceramic on polyethylene pairings were similar to the differences of published data for existing material-pairings. Test results on ceramic-ceramic pairings of different frontal-plane geometry and surface roughness displayed low wear rates and no fracture failures.Conclusions: The presented set up is able to simulate the rolling-gliding movement of the knee joint, is easy to use, and requires a minimum of user intervention or monitoring. It is suitable for long-term testing, and therefore a useful tool for the investigation of new and promising materials which are of interest for application in knee joint replacement implants. © 2010 Richter et al; licensee BioMed Central Ltd.

Objetivo: Analisar as associações entre a potência muscular dos membros inferiores e a saúde óssea avaliada através de ultrassonografia quantitativa. Métodos: A amostra incluiu 63 crianças de 9 anos de idade. A velocidade de som (VS) do rádio e da tíbia foi avaliada através de ultrassonografia quantitativa, a potência de salto foi estimada a partir de um salto vertical com contramovimento e a maturidade somática foi determinada a partir da estimação do pico de velocidade em altura. O estado geral de saúde e o historial de fraturas foram avaliados através de questionário. As associações entre medidas dos parâmetros ósseos e a potência de salto foram analisadas através de correlações bivariadas, com as variáveis expressas em valores absolutos e relativos (estandardizados). Resultados: Foram observadas associações positivas entre a maturidade somática e a potência de salto expressa tanto em valores absolutos como relativos (p <0,05). A maturidade somática correlacionou-se ainda positivamente com a VS da tíbia nas raparigas (r=0,358, p=0,045), enquanto nos rapazes se verificou uma associação negativa, embora não significativa (r=-0,290, p=0,126). Observou-se uma associação ou propensão para associação negativa entre a VS da tíbia e a potência de salto nos rapazes (r=-0,490, p=0,007) e nas raparigas (r=-0,344, p=0,054). Conclusão: A potência de salto não parece constituir um bom marcador da saúde óssea de rapazes e raparigas de 9 anos de idade quando avaliada através de ultrassom.

Propósito y Método del Estudio: Introducción. La generación de implantes para el tratamiento y regeneración de afecciones del tejido óseo representan una enorme oportunidad de desarrollo e innovación para muchos campos de la salud humana. Metodología: nosotros generamos un implante de tres componentes (I-3C) constituido por células madre mesenquimales (CMMs), transducidas ex vivo con un vector adenoviral que expresa una combinación de las proteínas morfogenéticas de hueso 2 y 7 (AdBMP2/7), embebidas en una matriz ósea desmineralizada (MOD). Este implante fue probado en un modelo ovino (Ovis aries) en una lesión sometida a carga. Resultados: los ensayos in vitro demostraron que el tratamiento seleccionado con mayor potencial osteogénico es el que contiene la combinación AdBMP2/7 comprobado por PCR en tiempo real, Western Blot y tinciones histológicas e inmunohistoquímicas para las proteínas marcadoras de inducción a hueso, osteocalcina y colágeno tipo I. El implante fue colocado en la zona de la lesión, creada por una distracción en la diáfisis media de la tibia. Se probaron tres grupos de experimentación: control 1 sin implante (S-I); control 2 implante con CMMs (I-CMMs); implante de 3 componentes CMMs modificadas genéticamente con adenovirus que expresan proteínas morfogenéticas de hueso 2 y 7 que se encuentran embebidas en una matriz de hueso desmineralizado (I-3C). Resultados: el seguimiento radiográfico por 10 semanas después de la distracción ósea demostró una reducción en el tiempo de consolidación del grupo I-3C. La tomografía computarizada demostró en ese mismo grupo, una forma y estructura del hueso postmortem muy parecida a la de una tibia sin lesión. Estos hallazgos fueron corroborados por los ensayos de compresión e histológicos, demostrando que la calidad del hueso nuevo formado fue mayor que la de los grupos sin implante y con CMMs sin modificación genética. Contribuciones y Conclusiones: se logró desarrollar un implante de tres componentes constituido por células madre mesenquimales (CMMs), transducidas ex vivo con un vector adenoviral que expresa una combinación de las proteínas morfogenéticas de hueso 2 y 7 (AdBMP2/7), que nos permitirá continuar con los estudios clínicos necesarios para evaluar principalmente defectos en huesos y enfermedades relacionadas con el sistema óseo.
