Adoptive Cell Transfer (ACT) Therapy is a cancer treatment that enhances and utilizes the body’s own immune system. However, this treatment has had limited success in clinical trials. We hypothesized that this is due to the immunosuppressive, acidic microenvironment of cancer tumors. We tested the effects of acidic, neutral, and basic environments in vitro on cytotoxic T lymphocyte (CTL) survival, activation, migration and killing ability and on cancer cell survival. We found that CTLs have most optimum survival, activation, and migration in a neutral environment, while the optimal extracellular conditions for EG-7 lymphoma are slightly acidic and B16-OVA melanoma survives best in physiological conditions. Future research should further study the killing ability of T cells in the three different environments and look to move to in vivo experiments.

This article discusses the potential of audio games based on the evaluation of three projects: a story-driven audio role-playing game (RPG), an interactive audiobook with RPG elements, and a set of casual sound-based games. The potential is understood, both in popularity and playability terms. The first factor is connected to the degree of players’ interest, while the second one to the degree of their engagement in sound-based game worlds. Although presented projects are embedded within the landscape of past and contemporary audio games and gaming platforms, the authors reach into the near future, concluding with possible development directions for this non-visual interactive entertainment.

Tendo em conta o cenário atual de aumento de população idosa e da esperança média de vida, de proliferação dos meios tecnológicos e da necessidade de permanecer ativo para a melhoria da qualidade de vida, a criação de produtos, que aliem a tecnologia ao exercício físico, mostra-se uma mais-valia. A presente dissertação possui quatro objetivos:I) Identificar as tipologias de exercícios adequados ao público-alvo II) planeamento de três vídeos tutoriais de exercício físico para apoio ao cidadão sénior; III) realização dos três vídeos tutoriais de exercício físico para apoio ao cidadão sénior; IV) avaliação dos três vídeos tutoriais junto a cidadãos seniores para compreender se as características presentes nos tutoriais são adequadas para o público. Para a concretização dos objetivos, anteriormente mencionados, foram utilizadas três diferentes fases metodológicas durante este projeto: Na primeira fase foi utilizada uma metodologia exploratória, isto é, foram estudados trabalhos anteriormente realizados relativos a esta temática, para auxiliar a construção dos tutoriais; Na segunda fase recorrendo à metodologia de investigação de desenvolvimento, foi concretizado um protótipo dos vídeos tutoriais, apresentados a um grupo de especialistas em ambiente de focus group, onde foram sugeridas alterações para melhor se adaptarem ao público-alvo, e posteriormente foi realizada uma versão final dos tutoriais. Por último, os tutoriais foram apresentados a cidadãos seniores para serem avaliados a nível de: vídeo, áudio, exercícios e texto, com o intuito de compreender se estes se encontravam adequados ao público sénior. Em suma, no final da presente investigação consegue-se responder à questão: quais as caraterísticas que devem possuir os tutoriais audiovisuais, de apoio à atividade física, destinados a um público sénior.

PURPOSE: To quantitatively analyze and compare the fundoscopic features between fellow eyes of retinal angiomatous proliferation and typical exudative age-related macular degeneration and to identify possible predictors of neovascularization. METHODS: Retrospective case-control study. Seventy-nine fellow eyes of unilateral retinal angiomatous proliferation (n = 40) and typical exudative age-related macular degeneration (n = 39) were included. Fundoscopic features of the fellow eyes were assessed using digital color fundus photographs taken at the time of diagnosis of neovascularization in the first affected eye. Grading was performed by two independent graders using RetmarkerAMD, a computer-assisted grading software based on the International Classification and Grading System for age-related macular degeneration. RESULTS: Baseline total number and area (square micrometers) of drusen in the central 1,000, 3,000, and 6,000 μm were considerably inferior in the fellow eyes of retinal angiomatous proliferation, with statistically significant differences (P < 0.05) observed in virtually every location (1,000, 3,000, and 6,000 μm). A soft drusen (≥125 μm) area >510,196 μm2 in the central 6,000 μm was associated with an increased risk of neovascularization (hazard ratio, 4.35; 95% confidence interval [1.56-12.15]; P = 0.005). CONCLUSION: Baseline fundoscopic features of the fellow eye differ significantly between retinal angiomatous proliferation and typical exudative age-related macular degeneration. A large area (>510,196 μm2) of soft drusen in the central 6,000 μm confers a significantly higher risk of neovascularization and should be considered as a phenotypic risk factor.

Les inventaires archéologiques représentent une part importante des travaux archéologiques qui sont effectués en sol québécois. Il s’agit d’interventions qui sont réalisées en milieux urbains ou ruraux, mais également dans des milieux forestiers parfois éloignés. Lors de ces interventions, plusieurs informations de nature spatiale sont prises, tant pour évaluer la présence ou l’absence de vestiges archéologiques que pour décrire l’environnement et les perturbations observées. Les données issues de ces travaux sont de manière générale enregistrées sur le terrain sur des supports papiers, pour être éventuellement transférées en format numérique après l’intervention, et la localisation sur le terrain est effectuée à l’aide de cartes, de la boussole et d’un appareil GPS. Ce projet cherche à évaluer la contribution potentielle d’une solution géomatique mobile qui permettrait d’assister les archéologues sur le terrain, tant au niveau de la saisie de données que de la géolocalisation sur le terrain. Afin d’identifier l’intérêt, les attentes et les besoins prioritaires d’une telle solution, une enquête a été réalisée auprès de la communauté archéologique. Subséquemment, afin de démontrer qu’une solution géomatique mobile pourrait être développée afin de soutenir les travaux archéologiques, un appareil mobile adapté aux contraintes du terrain archéologique a été identifié et un prototype d’application a été développé, sur la base des résultats de l’enquête obtenus. Il nous a par la suite été possible de faire prendre en main la solution mobile développée par une archéologue et un essai en milieu contrôlé a pu être réalisé afin d’avoir une évaluation critique de l’application et de proposer des pistes d’amélioration. Les résultats obtenus indiquent que la technologie géomatique mobile est aujourd’hui suffisamment accessible pour pouvoir bien s’adapter aux réalités de terrain et ainsi venir répondre à certains besoins des professionnels de terrain tout en demeurant abordable. Malgré quelques pistes d’amélioration qui ont été identifiées, l’enthousiasme suscité par l’application développée démontre l’intérêt des utilisateurs à intégrer ces nouvelles technologies. La démarche de développement employée est également novatrice puisqu’elle permet de développer, en l’espace de quelques mois, une application adaptée à un contexte de terrain professionnel particulier, tout en demeurant grandement modulable et pouvant s’adapter à d’autres domaines d’application.

The use of electrocardiogram nowadays, is very important in diagnosis of heart disease. The emergent increase of portable technology provides medica] monitoring of vital signs allowing freedom ofmovement and watching during normal activity of the patient. In this shidy, it is described the development of a prototype of an ambulatory cardiac monitoring system using 3 leads. The systems consists on conversion of an analog signal, having been previously processed and conditioned, into digital ECG signal and after processed with a microcontroller (MCU). The heartbeat rate can be observed in an LCD display. The LCD display is also used as the interface during the setup process. Ali digital data stream can be stored on a SD memory card llowing the ECG signa] to be accessed later on a PC.

Roots normally grow in darkness, but they may be exposed to light. After perceiving light, roots bend to escape from light (root light avoidance) and reduce their growth. How root light avoidance responses are regulated is not well understood. Here, we show that illumination induces the accumulation of flavonols in Arabidopsis thaliana roots. During root illumination, flavonols rapidly accumulate at the side closer to light in the transition zone. This accumulation promotes asymmetrical cell elongation and causes differential growth between the two sides, leading to root bending. Furthermore, roots illuminated for a long period of time accumulate high levels of flavonols. This high flavonol content decreases both auxin signaling and PLETHORA gradient as well as superoxide radical content, resulting in reduction of cell proliferation. In addition, cytokinin and hydrogen peroxide, which promote root differentiation, induce flavonol accumulation in the root transition zone. As an outcome of prolonged light exposure and flavonol accumulation, root growth is reduced and a different root developmental zonation is established. Finally, we observed that these differentiation-related pathways are required for root light avoidance. We propose that flavonols function as positional signals, integrating hormonal and ROS pathways to regulate root growth direction and rate in response to light.

Within the supply chain, the customer does not simply buy parts or services from suppliers, but also buys supplier capabilities, which results in quality products and services. Having a tool that handles the Inspection as well as the Nonconformance, Complaint, Corrective Action and Concession processes is key to successfully track the supplier performance. Taking as a case study a Supplier Quality Management (SQM) currently in operation in an Original Equipment Manufacturer (OEM) for automotive industry, this paper presents a platform to support a Supplier Quality Management System (SQMS), that fits the technical specification ISO/TS 16949 requirements. This prototype is composed by a web platform and a mobile App, having flexibility and mobility as key main characteristics.

During the lifetime of a research project, different partners develop several research prototype tools that share many common aspects. This is equally true for researchers as individuals and as groups: during a period of time they often develop several related tools to pursue a specific research line. Making research prototype tools easily accessible to the community is of utmost importance to promote the corresponding research, get feedback, and increase the tools’ lifetime beyond the duration of a specific project. One way to achieve this is to build graphical user interfaces (GUIs) that facilitate trying tools; in particular, with web-interfaces one avoids the overhead of downloading and installing the tools. Building GUIs from scratch is a tedious task, in particular for web-interfaces, and thus it typically gets low priority when developing a research prototype. Often we opt for copying the GUI of one tool and modifying it to fit the needs of a new related tool. Apart from code duplication, these tools will “live” separately, even though we might benefit from having them all in a common environment since they are related. This work aims at simplifying the process of building GUIs for research prototypes tools. In particular, we present EasyInterface, a toolkit that is based on novel methodology that provides an easy way to make research prototype tools available via common different environments such as a web-interface, within Eclipse, etc. It includes a novel text-based output language that allows to present results graphically without requiring any knowledge in GUI/Web programming. For example, an output of a tool could be (a structured version of) “highlight line number 10 of file ex.c” and “when the user clicks on line 10, open a dialog box with the text ...”. The environment will interpret this output and converts it to corresponding visual e_ects. The advantage of using this approach is that it will be interpreted equally by all environments of EasyInterface, e.g., the web-interface, the Eclipse plugin, etc. EasyInterface has been developed in the context of the Envisage [5] project, and has been evaluated on tools developed in this project, which include static analyzers, test-case generators, compilers, simulators, etc. EasyInterface is open source and available at GitHub2.

Angiotensin II (Ang II) and platelet-derived growth factor-BB (PDGF-BB) are associated with excessive cell migration, proliferation and many growth-related diseases. However, whether these agents utilise similar mechanisms to trigger vascular pathologies remains to be explored. The effects of Ang II and PDGF-BB on coronary artery smooth muscle cell (CASMC) migration and proliferation were investigated via Dunn chemotaxis assay and the measurement of [3H]thymidine incorporation rates, respectively. Both atherogens produced similar degrees of cell migration which were dramatically inhibited by mevastatin (10 nM). However, the inhibitory effects of losartan (10 nM) and MnTBAP (a free radical scavenger; 50 μM) were found to be unique to Ang II-mediated chemotaxis. In contrast, MnTBAP, apocynin (an antioxidant and phagocytic NADPH oxidase inhibitor; 500 μM), mevastatin and pravastatin (100 nM) equally suppressed both Ang II and PDGF-BB-induced cellular growth. Although atherogens produced similar changes in NADPH oxidase, NOS and superoxide dismutase activities, they differentially regulated antioxidant glutathione peroxidase activity which was diminished by Ang II and unaffected by PDGF-BB. Studies with signal transduction pathway inhibitors revealed the involvement of multiple pathways i.e. protein kinase C, tyrosine kinase and MAPK in Ang II- and/or PDGF-BB-induced aforementioned enzyme activity changes. In conclusion, Ang II and PDGF-BB may induce coronary atherosclerotic disease formation by stimulating CASMC migration and proliferation through agent-specific regulation of oxidative status and utilisation of different signal transduction pathways.

The current work aimed to study the antitumour activity of a phenolic extract of the edible mushroom Leccinum vulpinum Watling, rich essentially in hydroxybenzoic acids. In a first approach, the mushroom extract was tested against cancer cell growth by using four human tumour cell lines. Given the positive results obtained in these initial screening experiments and the evidence of some studies for an inverse relationship between mushroom consumption and breast cancer risk, a detailed study of the bioactivity of the extract was carried out on MCF-7 cells. Once the selected cell line to precede the work was the breast adenocarcinoma cell line, the human breast non-malignant cell line MCF-10A was used as control. Overall, the extract decreased cellular proliferation and induced apoptosis. Furthermore, the results also suggest that the extract causes cellular DNA damage. Data obtained highlight the potential of mushrooms as a source of biologically active compounds, particularly with antitumour activity.

Tesi hau Solarus AB enpresaren konzentratzailedun eguzki kolektore fotovoltaiko termikoei (C-PVT) buruz doa eta bi helburu nagusi ditu. Lehena Solarus-eko oraingo diseinuaren alderaketak diseinatzea da, MaReCo (Maximum Reflector Collector) diseinuaren eta parabola puruaren alderaketa batzuekin batera. Diseinu hauetan eguzki zelulen ebaketa berriak daude barruan eta 4 busbar-eko eguzki zeluletan oinarritua dago. Honi esker analisi sakon bat egin ahalko da hargailu eta estruktura diseinuak konparatuz. Bigarren helburua Solarus AB-k Gävleko unibertsitatean (HiG) kokaturik dituen kolektoreen errendimendu elektriko eta termikoa aztertzean datza. Datuak simulazio eta software espezifikoen bidez lortu dira eta ondoren Microsoft Excel®-en aztertu. Bi proiektu txikiagoak egin dira ere enpresan, bata eguzki kolektore fotovoltaiko termikoen merkatuaren ikerketan datza eta bestea eguzki kolektoreen produkzio prozesuaren gida batean. Hargailuen eta estrukturaren diseinu berriak preparatuta utzi dira prototipoen hurreneko eraikuntzarako eta proiektuarekin jarraitzeko etorkizuneko lan bat planeatu da. Unibertsitateko instalakuntzaren analisiari dagokionez, errendimendu elektriko eta termikoa estimatuena baino nabarmenki txikiagoak izan dira.

O avanço das tecnologias de informação continua a mudar os paradigmas de ensino e aprendizagem. Os meios disponíveis são cada vez mais diversificados e, com a necessidade de procurar novos estudantes e diversificar o público-alvo, as instituições de ensino superior estão a repensar os seus modelos de negócio e estratégias pedagógicas. A proliferação de dispositivos móveis catalisa uma aposta crescente no ensino a distância (EaD) no sentido de proporcionar aprendizagens em mobilidade (m-learning). No entanto, as soluções existentes para m-learning são ainda pouco adaptadas às recentes metodologias de EaD, na maioria das vezes funcionando como extensão de um ambiente virtual de aprendizagem ou com muito foco nos conteúdos. Sendo a Universidade Aberta (UAb) a única instituição de ensino superior público em Portugal de ensino a distância, com um modelo pedagógico próprio, constitui um natural caso de aplicação de tecnologia móvel em novos contextos de aprendizagem, importando por isso estudar e desenhar os mecanismos de interação mais adequados com professores e estudantes em mobilidade. Adotou-se neste trabalho a metodologia Design Science Research, tendo sido identificadas as características e comportamentos de potenciais utilizadores, e definidas as funcionalidades que devem ser disponibilizadas na primeira versão de uma aplicação para dispositivos móveis (app) no contexto do ensino a distância. É proposto o design da interface dessa app, usando o modelo da UAb como caso de aplicação, e disponibilizada uma lista de orientações para o desenvolvimento do protótipo funcional. Da investigação realizada, concluiu-se que a interface proposta constitui um modelo válido para o desenho de uma app para aprendizagens em mobilidade, no regime de ensino de uma universidade virtual. A partir deste modelo, as instituições de ensino superior podem desenvolver apps adaptando-se ao avanço das Tecnologias de Informação e Comunicação e ficarem alinhadas com as necessidades dos seus alunos e docentes, particularmente se dispuserem de oferta formativa a distância.
