FADD adaptor and PEA-15/ERK1/2 partners in major depression and schizophrenia postmortem brains: Basal contents and effects of psychotropic treatments.

Enhanced brain apoptosis (neurons and glia) may be involved in major depression (MD) and schizophrenia (SZ), mainly through the activation of the intrinsic (mitochondrial) apoptotic pathway. In the extrinsic death pathway, pro-apoptotic Fas-associated death domain (FADD) adaptor and its non-apoptotic p-Ser194 FADD form have critical roles interacting with other death regulators such as phosphoprotein enriched in astrocytes of 15kDa (PEA-15) and extracellular signal-regulated kinase (ERK). The basal status of FADD (protein and messenger RNA (mRNA)) and the effects of psychotropic drugs (detected in blood/urine samples) were first assessed in postmortem prefrontal cortex of MD and SZ subjects (including a non-MD/SZ suicide group). In MD, p-FADD, but not total FADD (and mRNA), was increased (26%, n=24; all MD subjects) as well as p-FADD/FADD ratio (a pro-survival marker) in antidepressant-free MD subjects (50%, n=10). In contrast, cortical FADD (and mRNA), p-FADD, and p-FADD/FADD were not altered in SZ brains (n=21) regardless of antipsychotic medications (except enhanced mRNA in treated subjects). Similar negative results were quantified in the non-MD/SZ suicide group. In MD, the regulation of multifunctional PEA-15 (i.e., p-Ser116 PEA-15 blocks pro-apoptotic FADD and PEA-15 prevents pro-survival ERK action) and the modulation of p-ERK1/2 were also investigated. Cortical p-PEA-15 was not changed whereas PEA-15 was increased mainly in antidepressant-treated subjects (16-20%). Interestingly, cortical p-ERK1/2/ERK1/2 ratio was reduced (33%) in antidepressant-free when compared to antidepressant-treated MD subjects. The neurochemical adaptations of brain FADD (increased p-FADD and pro-survival p-FADD/FADD ratio), as well as its interaction with PEA-15, could play a major role to counteract the known activation of the mitochondrial apoptotic pathway in MD.

The Cultural Leadership Partners Program of the Iowa Arts Council and Iowa Department of Cultural Affairs (IAC/DCA) engages leaders in Iowa’s arts and cultural community that have demonstrated an exemplary record of programming and managerial excellence, as well as exceptional community service to the citizens of Iowa on a year-round basis. After a rigorous application process, organizations selected to become Cultural Leadership Partners (CLPs) receive annual operating support from IAC/DCA. This document will not only serve to communicate to CLPs what the IAC/DCA has learned from these conversations, but that it will act as a catalyst for further development of the program and inform how IAC/DCA can support, challenge, connect, and partner with Iowa’s leading arts and cultural institutions.

Determinants of the effect of existential behavioral therapy for bereaved partners: a qualitative study.

BACKGROUND: Informal caregivers of palliative patients took part in existential behavioral therapy (EBT), a group intervention comprising mindfulness exercises to reduce psychological distress and improve quality of life. OBJECTIVES: This study examined what the participants perceived as helpful to cope with their loss during the first year of bereavement, particularly with regard to the EBT intervention. DESIGN: Sixteen problem-centered, semi-structured interviews were evaluated with content analysis. RESULTS: Two main categories were found: social support and self-regulation. Social support includes sense of belonging as well as emotional, cognitive, and practical help experienced from others. Mindfulness and acceptance, a clear focus on the positive, and orientation toward the future were helpful strategies of self-regulation; these were also part of the EBT intervention. Mindfulness was understood as permitting emotions and acceptance of one's inner processes, even if they were not pleasant, and was found to be helpful to stop ruminative thinking. CONCLUSIONS: The categories considered as being helpful parallel core elements of EBT and recent grief theories. The intervention was found to be supportive and met the needs of the participants. The interviewees appreciated the continuity of EBT support from palliative care into bereavement.

Exchange of Product-related Data between a Pulp and Paper Machinery Supplier and Its Partners

Kiihtyvä kilpailu yritysten välillä on tuonut yritykset vaikeidenhaasteiden eteen. Tuotteet pitäisi saada markkinoille nopeammin, uusien tuotteiden pitäisi olla parempia kuin vanhojen ja etenkin parempia kuin kilpailijoiden vastaavat tuotteet. Lisäksi tuotteiden suunnittelu-, valmistus- ja muut kustannukset eivät saisi olla suuria. Näiden haasteiden toteuttamisessa yritetään usein käyttää apuna tuotetietoja, niiden hallintaa ja vaihtamista. Andritzin, kuten muidenkin yritysten, on otettava nämä asiat huomioon pärjätäkseen kilpailussa. Tämä työ on tehty Andritzille, joka on maailman johtavia paperin ja sellun valmistukseen tarkoitettujen laitteiden valmistajia ja huoltopalveluiden tarjoajia. Andritz on ottamassa käyttöön ERP-järjestelmän kaikissa toimipisteissään. Sitä halutaan hyödyntää mahdollisimman tehokkaasti, joten myös tuotetiedot halutaan järjestelmään koko elinkaaren ajalta. Osan tuotetiedoista luo Andritzin kumppanit ja alihankkijat, joten myös tietojen vaihto partnereiden välillä halutaan hoitaasiten, että tiedot saadaan suoraan ERP-järjestelmään. Tämän työn tavoitteena onkin löytää ratkaisu, jonka avulla Andritzin ja sen kumppaneiden välinen tietojenvaihto voidaan hoitaa. Tämä diplomityö esittelee tuotetietojen, niiden hallinnan ja vaihtamisen tarkoituksen ja tärkeyden. Työssä esitellään erilaisia ratkaisuvaihtoehtoja tiedonvaihtojärjestelmän toteuttamiseksi. Osa niistä perustuu yleisiin ja toimialakohtaisiin standardeihin. Myös kaksi kaupallista tuotetta esitellään. Tarkasteltavana onseuraavat standardit: PaperIXI, papiNet, X-OSCO, PSK-standardit sekä RosettaNet. Lisäksi työssä tarkastellaan ERP-järjestelmän toimittajan, SAP:in ratkaisuja tietojenvaihtoon. Näistä vaihtoehdoista parhaimpia tarkastellaan vielä yksityiskohtaisemmin ja lopuksi eri ratkaisuja vertaillaan keskenään, jotta löydettäisiin Andritzin tarpeisiin paras vaihtoehto.

Independent of its transcriptional function, a pool of interferon regulatory factor (IRF)-3 activated downstream of MAVS can bind to and activate cytosolic Bax, resulting in Bax translocation to the mitochondria and initiation of the intrinsic apoptotic pathway.

Software SMEs asStrategic Partners in the Partner Programs of Large International Companies

Tutkielman tavoitteena oli tutkia pienten- ja keskisuurten (PK) ohjelmistoyritysten kansainvälisiä kumppanuuksia. Päätavoitteena oli löytää keinoja kuinka PK-ohjelmistoyritykset voisivat tulla strategisiksi kumppaneksi suurten kansainvälisten yritysten kumppanuusohjelmissa. Lisäksi tutkielmassa oli tavoitteena selvittää kuinka kumppaneiden välistä sitoutumista voitaisiin vahvistaa, jotta PK-ohjelmistoyritykset voisivat saavuttaa todellista lisäarvoa ja kansainvälistä kasvua kumppanuusohjelmien kautta. Tutkielma jakaantuu teoreettiseen ja empiiriseen osaan. Teoreettinen osa keskittyy tarkastelemaan korkean teknologian markkinointia ohjelmistoalalla sekä kansainvälisiä kumppanuuksia. Suurten yritystenkumppanuusohjelmia ei ole tutkittu suomalaisten PK-ohjelmistoyritysten näkökulmasta, minkä vuoksi empiirinen tutkimus on perusteltua. Empiirinen tutkimus toteutettiin laadullisena case-tutkimuksena ja tutkimusmenetelmänä käytettiin puolistrukturoitua haastattelua. Tutkimustulokset osoittavat, että strategisen kumppanin aseman saavuttaminen on pitkä ja haastava matka PK-yrityksille. Suurten kansainvälisten yritysten kumppanuusohjelmat ovat useimmiten monimutkaisia ja todellisen lisäarvon saavuttaminen kumppanuusohjelman kautta vaatii paljon resursseja PK-yrityksiltä. Jotta PK-yritykset voisivat saavuttaa ja säilyttää strategisen kumppanin aseman kumppanuusohjelmassa, vaatii se aktiivista ja päivittäistä vuorovaikutusta kumppaneiden kesken. Erityisesti tiiviit henkilösuhteet oikeiden avainhenkilöiden kanssa ovat välttämättömyys. Läheiset kontaktit mahdollistavat sen, että PK-yritykset voivat ainakin osittain ohittaa kumppanuusohjelman byrokratian, mikä lisää luottamusta ja sitoutumista kumppanuussuhteessa sekä edistää kansainvälistä kasvua ja menestystä liiketoiminnassa.

Sähköisen hankintatyökalun kehittäminen ja käyttöönotto. Case: Inex Partners Oy

Internetin yleistyminen on luonut uudenlaiset puitteet yritysten väliselle sähköiselle kaupankäynnille. Vanhat EDI-pohjaiset järjestelmät koetaan kalliiksi ja sitoviksi ja niiden sijasta rakennetaan www-teknologiaan pohjautuvia järjestelmiä. Tämän pro gradu-tutkielman tarkoituksena oli kuvata mahdollisimman tarkkaan erään web-pohjaisen hankintatyökalun kehittämis- ja käyttöönottoprojekti sekä arvioida sen onnistumista sekä käyttöönoton vaikutuksia. Case yrityksenä tutkielmassa toimii valikoima- ja logistiikkayhtiö Inex Partners Oy. Tutkielma on luonteeltaan toimintatutkimus, sillä tutkija on osa tutkimuskohdetta. Työn teoriaosassa käsitellään hankintatoimen strategiaa toimittajien valinnan ja toimittajasuhteiden hallinnan näkökulmasta. Tämän jälkeen esitellään sähköistä kaupankäyntiä ja sen hankintatoimelle tarjoamia sovelluksia. Kehitysprojektin tuotoksena syntyi sähköinen SRM-toimittajaportaali, joka mahdollistaa tarjouspyyntöjen lähettämisen ja vastaanottamisen Inexin ja tavarantoimittajien välillä. Portaali on integroitu Inexin toiminnanohjausjärjestelmään (ERP) siten, että tarjouspyyntöjen perusteella muodostuneet ostotilaukset siirtyvät järjestelmästä toiseen. Tutkielman johtopäätöksinä esitetään, että läpikäyty kehitysprojekti oli lähtökohtaisesti oikeintoteutettu ja yrityksen strategian mukainen. Tuotoksena saadun toimittajaportaalin istuvuutta tuoteryhmän hankintastrategiaan ei kuitenkaan oltu otettu tarpeeksi huomioon. Teknisen toteutuksen monimutkaisuus asettaa myös haasteensa työkalun käytölle. Jatkokehitystarpeiksi puolestaan esitetään portaalin räätälöimistä Inexin muiden tuoteryhmien hankinnan tueksi sekä portaalin huutokauppa- toiminnallisuuden kehittämistä ja käyttöönotttoa.

Implementation of Customer Relationship Management system to support channel partners

Työn tavoitteena oli selvittää, kokevatko matkapuhelinalalla toimivat kanavapartnerit heitä varten suunnitellun asiakassuhdehallintajärjestelmän lisäarvopalveluna vaiko eivät. Työ toteutettiin asiakastyytyväisyystutkimuksen avulla, jossa mitattiin erilaisten partnereiden asennetta palvelua kohtaan. Työn tuloksena ilmeni, että palvelussa olevan informaation määrällä ei ollut suurta vaikutusta asiakkaiden tyytyväisyyteen. Syyt ovat pikemminkin yleisissä asenteissa vastaavia palveluja kohtaan.

Pienyrityksen investointiprosessi. Case: STL-Partners Oy

Tutkielman tavoitteena oli tutkia pienyrityksen investointiprosessia ja seurata sen edistymistä kohdeyrityksessä. Tavoite pyrittiin saavuttamaan kirjallisuustutkimuksen avulla ja STL-Partners Oy:n toimitilainvestoinnin suunnitteluprosessia seuraamalla. Tutkimus toteutettiin pääasiassa tapaustutkimuksena, mutta se sisältää myös päätöksentekometodologisia osia. Tutkielman alussa esitettiin kirjallisuuten pohjautuva investointiprosessin malli. Kohdeyrityksen investointiprosessia seurattiin esitetyn mallin vaiheiden mukaisesti. Loppuosassa selvitettiin investoinnin eri rahoitusvaihtoehdot ja tehtiin kohdeyritykseltä saatujen tietojen pohjalta laskelma investoinnin kannattavuudesta. Tutkimuksessa todettiin, että myös pienessä yrityksessä investointien kannattavuuden arvioinnissa voidaan hyödyntää investointiteorian malleja. Tutkimuksessa selvitettiin myös perusteet investoinnin kannattavuuden arvioinnille. Tutkimuksessa selvitettyjen tietojen avullalla kohdeyritys voi tehdä lopullisen investointipäätöksen.

PI3K and AKT: Unfaithful Partners in Cancer.

The phosphatidylinositol 3-kinase (PI3K)/AKT signaling pathway regulates multiple cellular processes. An overactivation of the pathway is frequently present in human malignancies and plays a key role in cancer progression. Hence, its inhibition has become a promising approach in cancer therapy. However, the development of resistances, such as the abrogation of negative feedback mechanisms or the activation of other proliferative signaling pathways, has considerably limited the anticancer efficacy of PI3K/AKT inhibitors. In addition, emerging evidence points out that although AKT is acknowledged as the major downstream effector of PI3K, both PI3K and AKT can operate independently of each other in cancer, revealing another level of complexity in this pathway. Here, we highlight the complex relationship between PI3K and AKT in cancer and further discuss the consequences of this relationship for cancer therapy.

Infection with human papillomaviruses of sexual partners of women having cervical intraepithelial neoplasia

Epidemiological studies show that human papillomaviruses (HPV) are strongly related to cervical cancer and cervical intraepithelial neoplasias (CIN). Unlike the case for women, there are no consistent data on the natural history of HPV in the male population even though these viruses are prevalent in males. We carried out a prospective study to assess the prevalence of HPV in males as well as the factors that determine such infections in 99 male sexual partners of women with CIN. The genitalia of the males were physically examined and subjected to peniscopy for the collection of scrapings which were subjected to the polymerase chain reaction and restriction fragment length polymorphism to detect HPV. Of the 99 males sampled, 54 (54.5%) were positive for HPV DNA, 24% of whom presented normal peniscopy, 28% presented evident clinical lesions and 48% isolated lesions consistent with subclinical infection. In the HPV-negative group, 53% showed normal peniscopy, 4% presented evident clinical lesions and 42% isolated lesions consistent with subclinical infection. The study detected a statistically significant association (P < 0.02, Pearson chi-square test) between HPV infection and both the mean number of sexual partners which a male had during his life and the mean number of sexual partners in the year prior to testing. Viral types 6 and 11 were most frequently encountered. The study shows that infection with HPV was frequent in male sexual partners of women with CIN.

Human papillomavirus infection among sexual partners attending a Sexually Transmitted Disease Clinic in Rio de Janeiro, Brazil

Cervical cancer is a major source of illness and death among women worldwide and genital infection with oncogenic human papillomavirus (HPV) its principal cause. There is evidence of the influence of the male factor in the development of cervical neoplasia. Nevertheless, the pathogenic processes of HPV in men are still poorly understood. It has been observed that different HPV types can be found among couples. The objective of the present study was to investigate HPV infections in female patients (n = 60 females/group) as well as in their sexual partners and to identify the concordance of HPV genotypes among them. By using the polymerase chain reaction, we detected a 95% prevalence of HPV DNA in women with cervical intraepithelial neoplasia (CIN) compared to 18.3% in women with normal cervical epithelium, with a statistically significant difference (P < 0.001). The HPV DNA prevalence was 50% in male partners of women with CIN and 16.6% in partners of healthy women. In the control group (healthy women), only 9 couples were simultaneously infected with HPV, and only 22.2% of them had the same virus type, showing a weak agreement rate (kappa index = 0.2). Finally, we observed that HPV DNA was present in both partners in 30 couples if the women had CIN, and among them, 53.3% shared the same HPV type, showing moderate agreement, with a kappa index of 0.5. This finding supports the idea of circulation and recirculation of HPV among couples, perpetuating HPV in the sexually active population, rather than true recurrences of latent infections.

Identification and description of the development of the relationships between mentor and mentee teachers in the Halton Board of Education "Partners in the Classroom" Program

Fifteen mentoring pairs of teachers were randomly selected from each group of teachers that had participated in the Halton Board of Education "Partners in the Classroom" program during 1988/89, 1989/90, and 1990/91. Each teacher was personally interviewed. Interviews were recorded, transcriptions were prepared and examined and analyzed. During the first part of the interview questions were asked regarding personal and professional demographics. The purpose of the second part of the interview was to gain information relating to the development of the relationships, over a three-year period, between mentor and mentee teacher participants in the "Partners in the Classroom" program. The analysis of the data suggest that there are identifiable changes in the development of the relationship between the mentor teacher and the mentee teacher over time. Implications from the study results that could enhance the induction program for new teachers are discussed.
