The central role of translation regulation in the control of critical cellular processes has long been recognized. Yet the systematic exploration of quantitative changes in translation at a genome-wide scale in response to specific stimuli has only recently become technically feasible. Using a genetic approach, we have identified new Arabidopsis weak-ethylene insensitive mutants that also display defects in translation, which suggested the existence of a previously unknown molecular module involved in ethylene-mediated translation regulation of components of this signaling pathway. To explore this link in detail, we implemented for Arabidopsis the ribosome-footprinting technology, which enables the study of translation at a whole-genome level at single codon resolution[1]. Using ribosome-footprinting we examined the effects of short exposure to ethylene on the Arabidopsis translatome looking for ethylene-triggered changes in translation rates that could not be explained by changes in transcript levels. The results of this research, in combination with the characterization of a subset of the aforementioned weak-ethylene insensitive mutants that are defective in the UPF genes (core-components of the nonsense-mediated mRNA decay machinery), uncovered a translation-based branch of the ethylene signaling pathway[2]. In the presence of ethylene, translation of a negative regulator of ethylene signaling EBF2 is repressed, despite induced transcription of this gene. These translational effects of ethylene require the long 3´UTR of EBF2 (3´EBF2), which is recognized by the C-terminal end of the key ethylene-signaling protein EIN2 (EIN2C) in the cytoplasm once EIN2C is released from the ER-membrane by proteolytic cleavage. EIN2C binds the 3´EBF2, recruits the UPF proteins and moves to P-bodies, where the translation of EBF2 in inhibited despite its mRNA accumulation. Once the ethylene signal is withdrawn, the translation of the stored EBF2 mRNAs is resumed, thus rapidly dampening the ethylene response. These findings represent a mechanistic paradigm of gene-specific regulation of translation in response to a key growth regulator. Translation regulatory elements can be located in both 3′ and 5′ UTRs. We are now focusing on the ead1 and ead2 mutants, another set of ethylene-signaling mutants defective in translational regulation. Ribosome-footprinting on the ead1 mutant revealed an accumulation of translating ribosomes in the 5´UTRs of uORF-containing genes and reduction in the levels of ribosomes in the main ORF. The mutant is also impaired in the translation of GFP when this reporter is fused to WT 5´UTR of potential EAD1 targets but not when GFP is fused to the uORF-less versions of the same 5´UTRs. Our hypothesis is that EAD1/2 work as a complex that is required for the efficient translation of mRNAs that have common structural (complex 5´UTR with uORFs) and functional (regulation of key cellular processes) features. A macieira é uma árvore de clima temperado pertencente à família Rosaceae. Árvores de macieira entram em um período de dormência durante o inverno, o que garante a sobrevivência dessas plantas frente a temperaturas abaixo de zero e permite à planta retomar o crescimento vegetativo e reprodutivo na primavera. A indução, progressão e liberação da dormência é dependente de temperaturas abaixo de 7,2°C e é regulada por um mecanismo molecular endógeno de gemas. Em estudos anteriores do Laboratório de Genética Molecular Vegetal, foi identificado um importante QTL no cromossomo 9 de macieira que explica mais de 50% da variação fenotípica observada para o tempo de floração. Dois genes candidatos, MdFLC-like e MdPRE1, foram localizados dentro do intervalo de confiança deste QTL. Na planta modelo Arabidopsis thaliana, FLC e PRE1 estão envolvidos no processo de floração e crescimento, respectivamente, o que reforça um possível papel de MdFLC-like e MdPRE1 na regulação do tempo de floração em macieira. No presente estudo, está sendo investigado o papel dos genes MdFLC-like e MdPRE1 na progressão e liberação da dormência em gemas de macieira.

The Lemnaceae family lacks morphologic and anatomical characteristics with comparative aims and for that reason the molecular studies has been necessary a differentiation. In Costa Rica there are two species of Lemna: Lemna aequinoctialis and Lemna valdiviana. With the objective of a molecular characterization and differentiation of both species were extracted isoenzymes esterases. The study was carried out in the Molecular Genetics Laboratory of the School of Biological Sciences at Universidad Nacional, Heredia, Costa Rica. The technical electrophoresis in gel of polyacrylamida SDS-PAGE was used. It was difference in the pattern of isoenzymes between both species. According to previous molecular studies based on morphology and anatomy, flavonoides, isoenzymes, and DNA sequences of certain genes both species are very different; in addition they belong to different monophyletics groups. On the other hand, the importance of this molecular characterization is its possible use in water treatment, because the esterases have had important in the bioremediation. Also, species of the Lemnaceae family are used in the water treatment like bioremediation.

A missense G209A mutation of the alpha-synuclein gene was recently described in a large Contursi kindred with Parkinson's disease (PD). The objective of this study is to determine if the mutation G209A of the alpha-synuclein gene was present in 10 Brazilian families with PD. PD patients were recruited from movement disorders clinics of Brazil. A family history with two or more affected in relatives was the inclusion criterion for this study. The alpha-synuclein G209A mutation assay was made using polymerase chain reaction and the restriction enzyme Tsp45I. Ten patients from 10 unrelated families were studied. The mean age of PD onset was 42.7 years old. We did not find the G209A mutation in our 10 families with PD. Our results suggest that alpha-synuclein mutation G209A is uncommon in Brazilian PD families.

Yerba mate (Ilex paraguariensis) is rich in several bioactive compounds that can act as free radical scavengers. Since oxidative DNA damage is involved in various pathological states such as cancer, the aim of this study was to evaluate the antioxidant activity of mate tea as well as the ability to influence DNA repair in male Swiss mice. Forty animals were randomly assigned to four groups. The animals received three different doses of mate tea aqueous extract, 0.5, 1.0 or 2.0 g/kg, for 60 days. After intervention, the liver, kidney and bladder cells were isolated and the DNA damage induced by H2O2 was investigated by the comet assay. The DNA repair process was also investigated for its potential to protect the cells from damage by the same methodology. The data presented here show that mate tea is not genotoxic in liver, kidney and bladder cells. The regular ingestion of mate tea increased the resistance of DNA to H2O2-induced DNA strand breaks and improved the DNA repair after H2O2 challenge in liver cells, irrespective of the dose ingested. These results suggest that mate tea could protect against DNA damage and enhance the DNA repair activity. Protection may be afforded by the antioxidant activity of the mate tea's bioactive compounds
