Cyclic voltammetry and electrochemical impedance spectroscopy were used to study the surface acid-base property of carboxylic acid-terminated self-assembled monolayers (SAMs). A carboxylic acid-terminated thiol, such as thioctic acid (1,2-dithiolane-3-pentanoic add), was self-assembled on gold electrodes. Electron transfer between the bulk solution and the SAM modified electrode was studied at different pH using Fe(CN)(6)(3-) as a probe. The surface pK(a) of thioctic acid was determined by cyclic voltammetry and electrochemical impedance spectroscopy to be 5.6 +/- 0.1 and 5.8 +/- 0.1, respectively. The method is compared with other methods of monolayer pK(a) measurement.

Multilayer films composed of heteropolyanions (HPAS, SiMo11 VO405-) and cationic polymer poly(diallyldimethylammonium chloride) on 4-aminothiophenol self-assembled-monolayer were fabricated by electrochemical growth. Growth processes of the composite films were characterized by cyclic voltammetry. The results prove the third redox peak of Mo increases more rapidly, otherwise the other Mo redox peaks increase very slowly when the number of layers of heteropolyanions is greater. The peak potentials of composite films shift linearly to negative position with higher pH, which implies that protons are involved in the redox processes of HPA. The investigation of electrocatalytic behaviors of composite films shows a good catalytic activity for the reductions of HNO2 and BrO3-. Catalytic currents increase with increasing number of layers of heteropolyanions, moreover, the catalytic currents have a good linear relationship with the concentrations of BrO3-.

A surface plasmon resonance biosensor has been used to determine antibody activity in serum. As a model system, the interaction of mouse IgG and sheep anti-mouse IgG polyclonal antibody was investigated in real time. The factors, including pH value, ionic strength, protein concentration, influencing electrostatic adsorption of mouse IgG protein onto carboxylated dextran-coated sensor chip surface, were studied. The procedures of mouse IgG protein immobilization and immune reaction were monitored in real time. The regeneration effect using the different elution reagents was also investigated. The same mouse IgG immobilized surface can be used for 100 cycles of binding and elution with only 0.38% loss per regeneration in reactivity. The results show that the surface plasmon resonance biosensor is a rapid, simple, sensitive, accurate and reliable detection technique for real-time immunoassay of antibody activity. Three beta -blockers (propranolol, timolol, acebutolol) were separated by capillary electrophoresis (CE) and detected by end-column electrochemical detection (EC). In the present work, a carbon fiber (33 mum) electrode was used as the working electrode. The effect of the buffer concentration, buffer pH, detection potential and separation voltage on the separation of analytes and behavior of electrochemical detection was systematically investigated. The optimum conditions determined were as following: 40 cm length, 25 mum i.d. capillary; 17.5 kV separation voltage; 2 s injection at 15 kV; 70mM phosphate buffer, pH 3.5; detection potential + 1.2V (vs. Ag/AgCl). Under these conditions, the linear ranges of beta -blockers were over three orders of magnititude and the low detection limit of 10(-8)M was obtained. This method was also applied to detect the simulated urine sample.

A new type of inorganic-organic hybrid material incorporating carbon powder and alpha -type 2:18-molybdodiphosphate (P2Mo18) in a methyltrimethoxysilane (MTMOS) based gel has been produced by a sol-gel process and used to fabricate a chemically modified electrode. The P2Mo18-doped carbon ceramic composite electrode was characterized using SEM and cyclic voltammetry. Square-wave voltammetry with an excellent sensitivity was exploited to conveniently investigate the dependence of current and half-wave potential (E-1/2) on pH. The chemically modified electrode has some advantages over the modified film electrodes constructed by the conventional methods, such as long-term stability, reproducibility, and especially repeatability of surface-renewal by simple polishing in the event of surface fouling or dopant leaching. In addition, the modified electrode shows a good catalytic activity for the electrochemical reduction of bromate in an acidic aqueous solution. (C) 2000 Elsevier Science B.V. All rights reserved.

An amperometric tyrosinase enzyme electrode for the determination of phenols was developed by a simple and effective immobilization method using sol-gel techniques. A grafting copolymer was introduced into sol-gel solution and the composition of the resultant organic-inorganic composite material was optimized, the tyrosinase retained its activity in the sol-gel thin film and its response to several phenol compounds was determined at 0 mV vs. Ag/AgCl (sat. KCI). The dependences of the current response on pH, oxygen level and temperature were studied, and the stability of the biosensor was also evaluated. The sensitivity of the biosensor for catechol, phenol and p-cresol was 59.6, 23.1 and 39.4 muA/mM, respectively. The enzyme electrode maintained 73% of its original activity after intermittent use for three weeks when storing in a dry state at 4 degreesC. (C) 2000 Elsevier Science S.A. All rights reserved.

With the cyclic voltammetry and quartz crystal microbalance (QCM), the oxidation process and the electrodeposition behavior were studyied during the electrochemical oxidation of 2-mercaptobenzimidazol in aqueous solution. The E-pH diagram was also gained. These results showed the oxidation reaction was one electron reaction. The results from X-ray photoelectron spectrometry verified that the 2-mercaptobenzimidazol was oxidized to bisbenzimidazoyl disulfide.

The biosensor based on surface plasmon resonance(SPR) technology is a very useful tool to study the interaction between biomolecles. The main advantages of this technique is to "visualize" macromolecular interactions directly in real time, and in a label-free mode rather than indirect methods like enzyme-linked immunosorbent assays (ELISAs). We immobilize human serum albumin (HSA) to the carboxymethyldextran-modified sensor chip surface covalently to detect the activity of anti-HSA in serum, and regenerate the surface with .1 mol/L phosphoric acid. The results show that SPR biosensor can detect the activity of anti-HSA in real-time quickly and the sensor chip can be used over 100 cycles.

A novel glucose biosensor based on cast lipid film was developed. This model of biological membrane was used to supply a biological environment on the surface of the electrode, moreover it could greatly reduce the interference and effectively exclude hydrophilic electroactive material from reaching the detecting surface. TTF was selected as a mediator because of its high electron-transfer efficiency, and it was incorporated in the lipid film firmly. Glucose oxidase was immobilized in hydrogel covered on the lipid film. The effects of pH, operating potential were explored for the optimum analytical performance by using amperometric method. The response time of the biosensor was less than 20 s, and the linear range is up to 10 mmol l(-1) (corr. coeff. 0.9932) with the detection limit of 2 x 10(-5) mol l(-1). Two modification methods for multilayer formation, i.e. immersion growth and electrochemical growth, were studied comparatively for their influence on the electrochemical behavior and the electrocatalytic properties of the thus-fabricated SiMo11V-containing multilayer films. Electrochemical growth was proven to be a more suitable method than immersion growth in preparing uniform ultrathin multilayer self-assemblies with good functions. We investigated the effects of scan rate and pH on the electrochemical behavior of the monolayer and multilayer films. We also compared the electrocatalytic effects on the reduction of BrO3- and HNO2 by the multilayer films prepared by the two methods. Moreover, the influence of multilayer thickness and the identity of the outermost layer on the electrocatalytic properties were studied. Much higher catalytic currents appeared on the thicker multilayer films than those on the thinner ones. On the other hand, the catalytic currents became smaller when the multilayer films with SiMo11V as the outermost layer were covered with an additional quarternized poly(4-vinylpyridine) layer partially complexed with osmium bis(2,2'-bypyridine) chloride (QPVP-Os layer). These influences were believed to be due to the different quantities of electrocatalyst loaded in the multilayer films and the blocking effect of the QPVP-Os outermost layer. The characteristic electrochemical mechanics of azobenzene derivative self-assembled monolayers is discussed in present paper. It is presented that the structure inhibition is one of the most important factors in the increase of electrochemical reactive energy. A corresponding mathematical model was established based on Levich and Marcus's theory. Moreover, computational program was written to simulate the decrease of apparent rate constant (k(app)) of electron transfer with increasing surface concentration.

Capillary electrophoresis (CE) with end-column electrochemical detection (EC) of barbituric acid (BA) and 2-thiobarbituric acid (TA) has been described. Under optimum condition, BA and TA were separated satisfactorily, and a response of high sensitivity and stability was obtained at a detection potential of 1.25 V versus Ag/AgCl. Optimized end-column detection provides detection limit as low as 0.5 and 0.1 mu M for BA and TA, respectively. The calibration graph was linear over three orders of magnitude. The relative standard deviations (n = 10) of peak currents and migration times obtained for both BA and TA were 3.4, 3.7, and 1.7, 1.2%, respectively. The self-assembled monolayer(SAM) of 11-mercaptoundecanoic acid [HS(CH2)(10)COOH] was formed on a gold electrode and the effect of the charge of end group on the electrochemical response of Fe(CN)(6)(3-) at the SAM modified electrode was studied by cyclic voltammetry. At high pH, when the -COOH groups are dissociated, the current of Fe(CN)(6)(3-) is suppressed; as the solution pH is lowered, the current of Fe(CN)(6)(3-) increases. The electrochemical titration curve was obtained by correlating the currents of Fe(CN)(6)(3-) to the different pH values of electrolyte, from which the surface pK(a) was obtained to be 3. 0+/-0. 2. Furthermore, the reason of small pK(a) value was explained using SAMs of different surface coverage.

A hydrogen peroxide biosensor was fabricated by coating a sol-gel-peroxidase layer onto a Nafion-methylene green modified electrode. Immobilization of methylene green (MG) was attributed to the electrostatic force between MG(+) and the negatively charged sulfonic acid groups in Nafion polymer, whereas immobilization of horseradish peroxidase was attributed to the encapsulation function of the silica sol-gel network. Cyclic voltammetry and chronoamperometry were employed to demonstrate the feasibility of electron transfer between sol-gel-immobilized peroxidase and a glassy carbon electrode. Performance of the sensor was evaluated with respect to response time, sensitivity as well as operational stability. The enzyme electrode has a sensitivity of 13.5 mu A mM(-1) with a detection limit of 1.0 x 10(-7) M H2O2, and the sensor achieved 95% of the steady-state current within 20 s.
