A temática relativa às células estaminais inicia-se na década de 60 com a descoberta da primeira fonte viável destas células: a medula óssea. Diversos estudos permitiram definir a sua função de renovação tecidular e regeneração pós-dano, assim como a sua caraterização num grupo heterogéneo de células indiferenciadas, clonogénicas, definidas pela capacidade de auto-renovação e diferenciação em células maduras. Nos últimos anos, estas células ganharam popularidade face à alternativa terapêutica que representam para muitas doenças, tais como: diabetes, anomalias congénitas, danos do tecido nervoso, Parkinson, Alzheimer e outras alterações degenerativas, exposições pulpares, defeitos periodontais e perda do órgão dentário. Apesar do seu potencial terapêutico, apresentam vários efeitos adversos, especialmente em relação ao seu envolvimento direto (via transformação maligna das MSCs) e indireto (via efeito modulatório das MSCs) no desenvolvimento do cancro. Preconiza-se o seu uso no âmbito da Engenharia Tecidular, introduzindo o processo de regeneração tecidular através da utilização combinada de biomateriais e mediadores biológicos, a fim de proporcionar novas ferramentas para a medicina regenerativa. Mais tarde, tornou-se possível identificar cinco populações de células estaminais de origem dentária (DPSCs, SHEDs, DFPCs, SCAPs e PDLCs) que, para além da sua multipotência e capacidade de diferenciação, constituem fontes acessíveis para recolha. O isolamento destas células constitui ainda uma prática relativamente recente, na qual se torna preponderante isolar células com fenótipo pré-determinado e cultivá-las em meios de cultura adequados. Estudos comprovam que o método de isolamento e as condições de cultura utilizados podem dar origem a diferentes linhas celulares. A conservação é uma prática baseada na convicção de que a medicina regenerativa é o caminho mais promissor para o desenvolvimento da medicina personalizada. Informação adicional relativa à terapia com células estaminais é ainda necessária. Esta utiliza princípios de biomimética altamente desejáveis, pelo que os resultados obtidos têm vindo a despoletar grandes expetativas e a sua implementação na Engenharia Tecidular apresenta-se promissora.

Currently, there are several factors that suggest the need for the school to review their teaching practices: 1) the speed with which information and communication technologies - ICT are spread throughout society and in particular the increasing use of mobile phone by groups in school age;2) the scope and power of integrating ICT convergence of these technological resources, that can be used as teaching resources; 3) the fact that nowadays students grow up in a technological world and make their thought patterns operating in accordance with this reality, etc... In contrast, there are educators unrelated to the appropriation of ICT, presenting, in many cases, resistance to them. There is a incongruity between how individuals use mobile phones, for example, inside and outside the school. There are also educators, schools, public educational departments claiming to be against mobile phones usage; bills and laws regulating their use in educational space, etc... For these controversial aspects we have investigated whether cell phone use as a teaching resource practices mediator in Physics teaching. For this purpose, we developed an applied research with qualitative and exploratory approach about the objectives, adopting participant and the technical procedures. A documental and bibliographical research was taken. First of all, data were collected by e-mail, through a multiple choice questionnaire applied to private school teachers in southern Brazil, to identify the researched reality and develop teaching practices likely to be applied in the studied corpus. Then, such practices were applied in workshops located in Curitiba and Ponta Grossa cities, in Paraná state. During the application, data was collected by the researcher's field notes. Then, we recorded chats about the workshops from an instant message software (msn messenger), and we took photographic records. Data analysis was performed by triangulation. The results showed the need to enhance continuing education courses for teachers and the production of national academic literature, such as articles, dissertations, theses and books that explore the theme of the cell phone and its functionality in educational spaces as an educational resource mediator in the practices of teaching Physics. They are available in the dissertation defended in 2012, and now are also systematized in this book, organized as a final product of research conducted by the Graduate Program in Science Education and Technology UTFPR, Campus Ponta Grossa-PR.

Cannabinoids (CBs) can be classified as: phytocannabinoids, the constituents of the Cannabis sativa plant; synthetic cannabinoids lab-synthesized and the endocannabinoids that are endogenous lipid mediators. Cannabinoid compounds activate cannabinoid receptors – CB1 and CB2. The most prevalent psychoactive phytocannabinoid is Δ9tetrahydrocannabinol (THC), but more than 60 different CBs were already identified in the plant. The best characterized endocannabinoids (eCBs) are anandamide (AEA) and 2arachidonoylglycerol (2-AG), that are involved in several physiological processes including synaptic plasticity, pain modulation, energy homeostasis and reproduction. On the other hand, some synthetic cannabinoids that were initially designed for medical research, are now used as drugs of abuse. During the period of placental development, highly dynamic processes of remodeling occur, involving proliferation, apoptosis, differentiation and invasion of trophoblasts. It is known that a tight control of eCBs levels is required for normal pregnancy progression and that eCBs are involved in trophoblast cells turnover. Therefore, by sharing activation of the same receptors, exposure to exocannabinoids either by recreational or medicinal use may lead to alterations in the eCBs levels and in the endocannabinoid system homeostasis In this work, it was studied the impact of CBs in BeWo trophoblastic cells and in primary cultures of human cytotrophoblasts. Cells were treated for 24 hours with different concentrations of THC, the synthetic cannabinoid WIN‐55,212 (WIN) and 2-AG. Treatment with THC did not affect BeWo cells viability while WIN and 2-AG caused a dose-dependent viability loss. Morphological studies together with biochemical markers indicate that 2-AG is able to induce apoptosis in cytotrophoblasts. On the other hand, morphological studies after acridine orange staining suggest that autophagy may take part in WIN-induced loss of cell viability. All cannabinoids caused a decrease in mitochondrial membrane potential (Δψm) but only 2-AG led to ROS/RNS generation, though no changes in glutathione levels were observed. In addition, ER-stress may be involved in the 2-AG induced-oxidative stress, as preliminary results point to an increase in CCAAT-enhancer-binding protein homologous protein (CHOP) expression. Besides the decrease in cell viability, alterations in cell cycle progression were observed. WIN treatment induced a cell cycle arrest in G0/G1 phase, whereas 2-AG induced a cell cycle arrest in G2/M phase. Here it is reinforced the relevance of cannabinoid signaling in fundamental processes of cell proliferation and cell death in trophoblast cells. Since cannabis-based drugs are the most consumed illicit drugs worldwide and some of the most consumed recreational drugs by pregnant women, this study may contribute to the understanding of the impact of such substances in human reproduction.

A encontrabilidade da informação é um elemento que se situa entre as funcionalidades de um ambiente informacional analógico, digital ou híbrido e as características dos sujeitos informacionais. Deriva da mediação infocomunicacional, visto que está relacionada aos processos que compõem o fluxo infocomunicacional, desde a produção até a apropriação da informação. Considerando que os profissionais da informação, de informática e os próprios usuários de ambientes informacionais são mediadores, percebe-se a influência de suas ações mediadoras na encontrabilidade da informação. Com o objetivo de compreender como a mediação infocomunicacional praticada por esses mediadores pode influenciar a encontrabilidade da informação em ambientes informacionais, foram realizadas pesquisas bibliográfica, descritiva e documental, com abordagem qualitativa, o que viabilizou uma discussão entre os conceitos estudados, tornando possível realizar a análise do processo de autoarquivamento no Repositório Institucional da Universidade Federal do Rio Grande do Norte. A partir da técnica de observação foram mapeadas as ações praticadas pelos diferentes mediadores no referido ambiente, tendo como base as dimensões top-down e bottom-up do Modelo de Encontrabilidade da Informação (MEI). A partir do mapeamento, foram identificadas as ações infocomunicacionais e tecnológicas realizadas pelos diferentes mediadores no referido Repositório, constatando a hipótese de que suas ações interferem significativamente na encontrabilidade da informação.

Alzheimer’s Disease (AD) is a neurodegenerative disorder neuropathologically characterized by the presence of extracellular senile plaques, intracellular neurofibrillary tangles and synaptic loss. Neuroinflammation has been associated with some neurodegenerative diseases, such as AD. In AD, increased Aβ production and aggregation, have a fundamental role in the activation of the inflammatory process. In turn, this could be fundamental in the early stages of this pathology, regarding the Aβ clearance and brain protection. However, chronic inflammation leads to an increase of the inflammatory mediators, such as cytokines, released by activated microglia, astrocytes, and neurons. The excessive production of these inflammatory components promotes alterations in both amyloid precursor protein (APP) expression and processing, stimulating the increase of Aβ accumulation and abnormal tau phosphorylation. This results in neurotoxic effects, irreversible damage and neuronal loss. Chronic inflammation is a feature of AD however, little is known about the effects of some chemokines on its pathogenesis. Thus, the main aim of this thesis was to study the impact of the interleukin-8 (IL-8) and monocyte chemoattractant protein-1 (MCP-1) on apoptosis, APP and tau. The both studied chemokines resulted in small alterations regarding the cytotoxicity on SH-SY5Y differentiated cells, being a significant increase in apoptosis observed only for the MCP-1 at the highest concentration. For the APP processing no significant differences were obtained, although a tendency to increase at different concentrations and periods was registered for both IL-8 and MCP-1. With respect to tau and other cytoskeleton-associated proteins, it was possible to observe a tendency to increase in the phosphorylated residue (Ser396) at the higher concentrations, as well as alterations on actin and tubulin with an increase on acetylated-α tubulin. This effect can be translated by neuronal architectural and survival alterations. Therefore additional studies could contribute to a better understanding of the way that these chemokines act on AD pathogenesis.

The mushrooms have been object of intense research in view of its potential raising of application in different sectors of the pharmacology and alimentary industry. Among diverse bioactive composites of polyssacharides nature that exist in the fungus the glucans are much searched. These are polymers of glucose and classified as the type of glicosidic linking [α, β]. Peroxisome proliferator-activated receptors (PPARs), ranscription factors belonging to the family of nuclear receptors that bind themselves o specific agonists, have shown their importance in controlling the inflammatory process. The aim of this study was to perform a chemical characterization of extract rom the mushroom Caripia montagnei, assess its antiinflammatory and antibacterial effect and determine if this effect occurs via PPAR. This mushroom is composed of carbohydrates (63.3±4.1%), lipids (21.4l±0.9%) and proteins (2.2± 0.3%). The aqueous solution resulting from the fractionation contained carbohydrates (98.7±3.3%) and protein (1.3±0.25%). Analyses of infrared spectrophotometry and of nuclear magnetic esonance demonstrated that the extract of mushroom C. montagnei is rich in β-glucans. In hioglycolate-induced peritonitis, the C. montagnei glucans (50 mg/kg) educed the inflammatory process in 65.5±5.2% and agonists, pharmacological igands, for PPAR: Wy-14643 (49.3±6.1%), PFOA (48.9±3.8%) and clofibrate in 45.2±3.2%. Sodium diclofenac showed a reduction of 81.65±0.6%. In the plantar edema, the glucans from C. montagnei (50 mg/kg) and L-NAME reduced the edema to a similar degree 91.4±0.3% and 92.8±0,5 %, respectively. In all the groups tested, nitric oxide (NO), an inflammation mediator, showed a significant reduction in the nitrate/nitrite levels when compared to the positive control (P<0.001). The C. montagnei glucans did not show cytotoxicity in the concentrations tested (2.5, 5.0, 10.0, 20.0 and 40.0 µg/100 µL). Antibacterial activity demonstrated that, unlike total extract, there was no inhibition of bacterial growth. The C. montagnei glucans show great potential for antiinflammatory applications. This effect suggests that it is mediated by PPAR activation and by COX and iNOS inhibition

The central interest of this thesis is to comprehend how the public action impels the formation and transformation of the tourist destinies. The research was based on the premise that the public actions are the result of the mediation process of state and non-state actors considered important in a section, which interact aiming for prevailing their interests and world visions above the others. The case of Porto de Galinhas beach, in Pernambuco, locus of the investigation of this thesis, allowed the analysis of a multiplicity of actors on the formation and implementation of local actions toward the development of the tourism between the years 1970 and 2010, as well as permitted the comprehension of the construction of the referential on the interventions made. This thesis, of a qualitative nature, has as theoretical support the cognitive approach of analysis of the public policies developed in France, and it has as main exponents the authors Bruno Jobert and Pierre Muller. This choice was made by the emphasis on the cognitive and normative factors of the politics, which aspects are not very explored in the studies of public policies in Brazil. As the source of the data collection, documental, bibliographic and field researches were utilized to the (re)constitution of the formation and transformation in the site concerned. The analysis techniques applied were the content and the documental analysis. To trace the public action referential, it started by the characterization of the touristic section frontiers and the creation of images by the main international body: the World Tourism Organization, of which analysis of the minutes of the meetings underscored guidelines to the member countries, including Brazil, which compounds the global-sectorial reference of the section. As from the analysis of the evolution of the tourism in the country, was identified that public policies in Brazil passed by transformations in their organization over the years, indicating changes in the referential that guided the interventions. These guidelines and transformations were identified in the construction of the tourist destination of Porto de Galinhas, of which data was systematized and presented in four historical periods, in which were discussed the values, the standard, the algorithms, the images and the important mediators. It has been revealed that the State worked in different roles in the decades analyzed in local tourism. From the 1990s, however, new actors were inserted in the formulation and implementation of policies developed, especially for local hotelkeepers. These, through their association, establishes a leadership relation in the local touristic section, thereby, they could set their hegemony and spread their own interest. The leadership acquired by a group of actors, in the case of Porto de Galinhas, does not mean that trade within the industry were neutralized, but that there is a cognitive framework that confronts the actors involved. In spite of the advances achieved by the work of the mediators in the last decades, that resulted in an amplification and diversification of the activity in the area, as well as the consolidation at the beach, as a tourist destiny of national standout, the position of the place is instable, concerned to the competitiveness, once that there is an situation of social and environmental unsustainability

Esta tesis doctoral explora las percepciones que tienen las jóvenes estudiantes de origen marroquí sobre el sistema educativo, sobre sí mismas y sobre su particular manera de construir el vínculo social. Para afrontar estos objetivos se analiza, por una parte, cómo es el sistema educativo marroquí en el que han sido escolarizadas en una primera etapa la mayoría de las estudiantes, así como los cambios sociales que se están produciendo en Marruecos en los que las mujeres están teniendo un papel decisivo. Por otra parte, se explora cómo se ha ido produciendo la incorporación de estas jóvenes al sistema educativo español y cómo su presencia junto con la de todo el alumnado de origen inmigrante ha derivado en determinadas toma de decisiones respecto al tratamiento educativo de la diversidad cultural por parte del Estado y de las Comunidades Autónomas. La opción metodológica adoptada para el análisis empírico ha sido la Teoría Fundamentada (Grounded Theory) combinada principalmente con el concepto de entramado propuesto por Elias (1982). El trabajo de campo ha estado orientado fundamentalmente a la realización de entrevistas en profundidad a las jóvenes estudiantes, la mitad de ellas estudiantes universitarias, y al profesorado, educadores/as y mediadores/as de los institutos de educación secundaria con mayor presencia de alumnado femenino de origen marroquí. Del análisis de las experiencias educativas de las jóvenes, de sus inquietudes personales y de sus propuestas se pone de manifiesto la importancia de la condición de estudiantes en sus trayectorias vitales principalmente porque les permite pensarse a sí mismas y afirmar como mujeres con derecho a decidir sobre sus propias vidas. De ahí que haya en sus discursos una crítica a la deriva multiculturalista de la escuela al incidir en el respeto y reconocimiento de la identidad cultural y no tanto en la reivindicación de la igualdad y de la autonomía y libertad individuales. Las jóvenes reclaman que la institución educativa les permite un devenir similar al de la población autóctona sin ser encasilladas desde criterios comunitaristas. Reclaman, en definitiva, a la escuela la recuperación de los valores de la modernidad y una defensa sin fisuras de la igualdad entre hombres y mujeres.

Studies indicate that several components were isolated from medicinal plants, which have antibacterial, antifungal, antitumor and anti-inflammatory properties. Sepsis is characterized by a systemic inflammation which leads to the production of inflammatory mediators exacerbated by excessive activation of inflammatory cells and disseminated intravascular coagulation (DIC), in which the human neutrophil elastase plays an important role in its pathogenesis. Several epidemiological studies suggest that components of plants, especially legumes, can play a beneficial role in reducing the incidence of different cancers. A chymotrypsin inhibitor of Kunitz (Varela, 2010) was purified from seeds of Erythrina velutina (Mulungu) by fractionation with ammonium sulfate, affinity chromatography on Trypsin-Sepharose, Chymotrypsin-Sepharose and ion exchange chromatography on Resource Q 1 ml (GE Healthcare) in system FPLC / AKTA. The inhibitor, called EvCI, had a molecular mass of 17 kDa determined by SDS-PAGE. The purified protein was able to inhibit human neutrophil elastase (HNE), with an IC50 of 3.12 nM. The EvCI was able to inhibit both pathways of HNE release stimulated by PAF and fMLP (75.6% and 65% respectively). The inhibitor also inhibited leukocyte migration in septic mice about 87% and prolonged the time of coagulation and inhibition factor Xa. EvCI showed neither hemolytic activity nor cytotoxicity. EvCI showed a selective antiproliferative effect to HepG2 cell lines with IC50 of 0.5 micrograms per milliliter. These results suggest EvCI as a molecule antagonist of PAF / fMLP and a potential use in fighting inflammation related disorders, disseminated intravascular coagulation (DIC) and cancer

Introducción: La periodontitis crónica y la artritis reumatoide (AR) son desórdenes inflamatorios crónicos caracterizados por la destrucción de tejidos, la reabsorción ósea y la producción de citoquinas proinflamatorias. Objetivos: Analizar la relación entre ambas enfermedades y la posible influencia del tratamiento de una sobre la otra. Material y métodos: Se realiza una revisión bibliográfica acerca de la patogénesis de la AR y la EP y los aspectos relacionados. Resultados: La patogénesis de ambas enfermedades muestra semejanzas notables. Bacterias periodontopatógenas, como Porphyromonas gingivalis, y mediadores inflamatorios, como el factor de necrosis tumoral y la proteína C reactiva, juegan un papel importante en la AR. La EP es un factor importante en la respuesta a la terapia en pacientes con AR. Conclusiones: Ambas enfermedades tienen una patogenia común. La prevalencia de enfermedad periodontal (EP) es mayor en pacientes con AR y viceversa. Aunque el control de la EP mediante tratamiento periodontal no quirúrgico parece mejorar los signos y síntomas en ambas enfermedades, hacen falta estudios más rigurosos con mayor número de casos y de mayor tiempo de evolución.

La Universidad EAFIT ha venido consolidando un programa sistemático de investigación alrededor de la pregunta por la economía política de la periferia colombiana. En particular, desde el Centro de Análisis Político —adscrito al Departamento de Gobierno y Ciencias Políticas— se han abordado asuntos de narcotráfico, madera, oro y poder político. En esta misma línea, el proyecto de investigación que da origen a este libro tiene el propósito de descifrar los arreglos microinstitucionales que enmarcan las acciones de corrupción y cooptación en el escenario de la contratación estatal con recursos de regalías de minería aurífera. Este texto ofrece una lectura alternativa a la dinámica social de la minería de oro. En lugar de la consabida maldición de los recursos naturales, se exponen conceptos y evidencia empírica para pensar y hacer posible una minería con mejores resultados sociales. Concebir el oro como fortuna es una tarea factible que no solo compete a los gobiernos locales sino también a los actores extralocales. Esta propuesta está dividida en dos partes: resultados de la investigación y diálogo entre aproximaciones conceptuales. La primera parte comienza con una mirada panorámica que sintetiza el marco analítico, los conceptos generales, aspectos metodológicos y hallazgos principales; luego, en el segundo capítulo, se detallan las categorías analíticas de la investigación, y se destaca la relevancia de la mercancía política como rasgo distintivo de la corrupción y la cooptación del Estado. El tercer capítulo exhibe los resultados del trabajo de campo en clave del marco de análisis institucional y desarrollo (IAD) de Elinor Ostrom; allí se evidencian las condiciones de adversidad del contexto y la presencia de reglas formales e informales que dan forma a los acuerdos que soportan la arena de acción minera. Para finalizar la primera parte, se presenta el ejercicio econométrico a partir del cual se concluye que existen atributos de comunidad que son factores explicativos de la disposición a la confianza y a la acción colectiva, esto es, que en condiciones de adversidad también afloran las vivencias comunitarias como detonantes que permiten establecer acuerdos para resolver dilemas colectivos. La segunda parte del libro consta de cuatro capítulos encaminados a soportar el proceso de construcción de pactos de gobernanza minera. En primer lugar, el capítulo cinco aborda las nociones de accountability y gobernanza como herramientas para el análisis de experiencias de transparencia, control ciudadano y fortalecimiento institucional. El capítulo siguiente hace un recorrido conceptual y empírico para proponer la noción de accountability societal como escenario para redirigir el sentido de la representación política y apropiar la noción de “cosa pública”. En el séptimo capítulo se estudia el derecho a la participación de la población afectada por proyectos de desarrollo en Colombia y se concluye con la necesidad de identificar y respetar las vías legítimas, pero no legisladas, de movilización social que pueden ensanchar el marco jurídico actual. A manera de conclusión, el último capítulo recoge ideas desarrolladas a lo largo del libro —desde una perspectiva más divulgativa y propositiva— con la intención de materializar un primer eslabón que permita avanzar hacia políticas públicas mineras en sintonía con las realidades y potencialidades locales. Por último, es necesario agradecer el apoyo económico de Colciencias para llevar a cabo esta investigación. También, en nombre del equipo de investigadores y de los colegas que se sumaron a esta propuesta, agradecemos la actitud abierta y constructiva de los pobladores de los territorios mineros que, con sus testimonios y hospitalidad, brindan enseñanzas vitales y hacen llamados de atención para reconocer que los académicos, al igual que los burócratas y los políticos, somos meros mediadores entre la vivencia cotidiana y las decisiones públicas. El desafío intelectual y práctico consiste en reconocer los activos comunitarios como insumo valioso para la construcción de nuevas políticas públicas.
