The use of agents targeting EGFR represents a new frontier in colon cancer therapy. Among these, monoclonal antibodies (mAbs) and EGFR tyrosine kinase inhibitors (TKIs) seemed to be the most promising. However they have demonstrated low utility in therapy, the former being effective at toxic doses, the latter resulting inefficient in colon cancer. This thesis work presents studies on a new EGFR inhibitor, FR18, a molecule containing the same naphtoquinone core as shikonin, an agent with great anti-tumor potential. In HT-29, a human colon carcinoma cell line, flow cytometry, immunoprecipitation, and Western blot analysis, confocal spectral microscopy have demonstrated that FR18 is active at concentrations as low as 10 nM, inhibits EGF binding to EGFR while leaving unperturbed the receptor kinase activity. At concentration ranging from 30 nM to 5 μM, it activates apoptosis. FR18 seems therefore to have possible therapeutic applications in colon cancer. In addition, surface plasmon resonance (SPR) investigation of the direct EGF/EGFR complex interaction using different experimental approaches is presented. A commercially available purified EGFR was immobilised by amine coupling chemistry on SPR sensor chip and its interaction to EGF resulted to have a KD = 368 ± 0.65 nM. SPR technology allows the study of biomolecular interactions in real-time and label-free with a high degree of sensitivity and specificity and thus represents an important tool for drug discovery studies. On the other hand EGF/EGFR complex interaction represents a challenging but important system that can lead to significant general knowledge about receptor-ligand interactions, and the design of new drugs intended to interfere with EGFR binding activity.

In der vorliegenden Arbeit wurden unter Verwendung neuer auf der Isophthalsäure basieren-der polymerisierbaren Tensiden carboxylfunktionalisierte Latexpartikel hergestellt, charakte-risiert und funktionalisiert. Im ersten Teil dieser Arbeit wurden 5-(10-Undecenyloxy)isophthalsäure (ISA-Vinyl), 5-(11-(4-Vinylphenoxy)undecyloxy)isophthal-säure (ISA-Sty), 5-(11-(2-Prop-1-enyl)phenoxy)undecyloxy)isophthalsäure (ISA-Pr), 5-(11-(1-Methacryloxy)undecylen)isophthalsäure (ISA-Met) und 5-(10-(3-Methylbut-3-enyl)oxy-1-oxydecylen)isophthalsäure (ISA-Bu) hergestellt. Die Surfmere wurden mit Styrol bzw. mit n-Butylacrylat copolymerisiert. ISA-Bu und ISA-Pr weisen während der Copolymerisation mit Styrol fast ideale Verläufe der Zeit/Umsatz-Kurven auf. Bei der Copolymerisation von ISA-Bu bzw. ISA-Vinyl mit n-Butylmethacrylat wurden ähnliche Ergebnisse erhalten. Die Carboxylgruppen an der Partikeloberfläche wurden mit Halogenderivaten verestert oder mit primären Aminen amidiert. Die funktionalisierten Partikel wurden mit der Polyelektrolyt-titration und konduktometrischen Titration, der IR- und UV-Spektroskopie, der Transmissi-onselektronenmikroskopie, der Fluoreszenzkorrelationsspektroskopie charakterisiert. Es konnte gezeigt werden, dass die Reaktanden kovalent an der Partikeloberfläche gebunden sind. Die Polymerpartikel wurden bei der Herstellung von Immunoassays genutzt. Die Adsorpti-onsisothermen zeigten, dass eine hohe Menge an Rinderserumalbumin an die Partikeloberflä-che physikalisch gebunden werden kann. In dieser Arbeit wurde ein einfaches Immunoassay hergestellt mit Biotin – Avidin als Modellsystem hergestellt. Die Surfmere wurden zur Stabilisierung von Miniemulsionen für die Miniemulsionspolymeri-sation genutzt. Im Laufe dieser Arbeit konnten mit dieser Methode Rylenfarbstoffe in Po-lystyrolpartikel stabilisiert werden.

The aim of this thesis was to investigate novel techniques to create complex hierarchical chemical patterns on silica surfaces with micro to nanometer sized features. These surfaces were used for a site-selective assembly of colloidal particles and oligonucleotides. To do so, functionalised alkoxysilanes (commercial and synthesised ones) were deposited onto planar silica surfaces. The functional groups can form reversible attractive interactions with the complementary surface layers of the opposing objects that need to be assembled. These interactions determine the final location and density of the objects onto the surface. Photolithographically patterned silica surfaces were modified with commercial silanes, in order to create hydrophilic and hydrophobic regions on the surface. Assembly of hydrophobic silica particles onto these surfaces was investigated and finally, pH and charge effects on the colloidal assembly were analysed. In the second part of this thesis the concept of novel, "smart" alkoxysilanes is introduced that allows parallel surface activation and patterning in a one-step irradiation process. These novel species bear a photoreactive head-group in a protected form. Surface layers made from these molecules can be irradiated through a mask to remove the protecting group from selected regions and thus generate lateral chemical patterns of active and inert regions on the substrate. The synthesis of an azide-reactive alkoxysilane was successfully accomplished. Silanisation conditions were carefully optimised as to guarantee a smooth surface layer, without formation of micellar clusters. NMR and DLS experiments corroborated the absence of clusters when using neither water nor NaOH as catalysts during hydrolysis, but only the organic solvent itself. Upon irradiation of the azide layer, the resulting nitrene may undergo a variety of reactions depending on the irradiation conditions. Contact angle measurements demonstrated that the irradiated surfaces were more hydrophilic than the non-irradiated azide layer and therefore the formation of an amine upon irradiation was postulated. Successful photoactivation could be demonstrated using condensation patterns, which showed a change in wettability on the wafer surface upon irradiation. Colloidal deposition with COOH functionalised particles further underlined the formation of more hydrophilic species. Orthogonal photoreactive silanes are described in the third part of this thesis. The advantage of orthogonal photosensitive silanes is the possibility of having a coexistence of chemical functionalities homogeneously distributed in the same layer, by using appropriate protecting groups. For this purpose, a 3',5'-dimethoxybenzoin protected carboxylic acid silane was successfully synthesised and the kinetics of its hydrolysis and condensation in solution were analysed in order to optimise the silanisation conditions. This compound was used together with a nitroveratryl protected amino silane to obtain bicomponent surface layers. The optimum conditions for an orthogonal deprotection of surfaces modified with this two groups were determined. A 2-step deprotection process through a mask generated a complex pattern on the substrate by activating two different chemistries at different sites. This was demonstrated by colloidal adsorption and fluorescence labelling of the resulting substrates. Moreover, two different single stranded oligodeoxynucleotides were immobilised onto the two different activated areas and then hybrid captured with their respective complementary, fluorescent labelled strand. Selective hybridisation could be shown, although non-selective adsorption issues need to be resolved, making this technique attractive for possible DNA microarrays.

The aim of this thesis was to investigate the synthesis of enantiomerically enriched heterocycles and dehydro-β-amino acid derivatives which can be used as scaffolds or intermediates of biologically active compounds, in particular as novel αvβ3 and α5β1 integrin ligands. The starting materials of all the compounds here synthesized are alkylideneacetoacetates. Alkylidene derivates are very usefull compounds, they are usually used as unsaturated electrophiles and they have the advantage of introducing different kind of functionality that may be further elaborated. In chapter 1, regio- and stereoselective allylic amination of pure carbonates is presented. The reaction proceeds via uncatalyzed or palladium-catalyzed conditions and affords enantiopure dehydro-β-amino esters that are useful precursor of biologically active compounds. Chapter 2 illustrates the synthesis of substituted isoxazolidines and isoxazolines via Michael addition followed by intramolecular hemiketalisation. The investigation on the effect of the Lewis acid catalysis on the regioselectivity of the addition it also reported. Isoxazolidines and isoxazolines are interesting heterocyclic compounds that may be regarded as unusual constrained -amino acids or as furanose mimetics. The synthesis of unusual cyclic amino acids precursors, that may be envisaged as proline analogues, as scaffolds for the design of bioactive peptidomimetics is presented in chapter 3. The synthesis of 2-substituted-3,4-dehydropyrrole derivatives starting from allylic carbonates via a two step allylic amination/ring closing metathesis (RCM) protocol is carried out. The reaction was optimized by testing different Grubbs’ catalysts and carbamate nitrogen protecting groups. Moreover, in view of a future application of these dehydro-β-amino acids as central core of peptidomimetics , the malonate chain was also used to protect nitrogen prior to RCM. Finally, chapter 4 presents the synthesis of two novel different classes of integrin antagonists, one derived from dehydro-β-amino acid prepared as described in chapter 1 and the other one has isoxazolidines synthesized in chapter 2 as rigid constrained core. Since that these compounds are promising RGD mimetics for αvβ3 and α5β1 integrins, they have been submitted to biological assay. and to interpret on a molecular basis their different affinities for the αvβ3 receptor, docking studies were performed using Glide program.

Ziel dieser Arbeit war es, ausgehend von auxiliargebundenen Piperidinderivaten, unterschiedliche chirale bi- und tricyclische Verbindungen darzustellen. Dazu wurde das 2,3,4,6-Tetra-O-pivaloyl--D-galactosylamin durch Kondensation mit Aldehyden in die entsprechenden Galactosylaldimine überführt, die in einer Lewissäure-katalysierten hochdiastereoselektiven Tandem-Mannich-Michael-Reaktionssequenz mit Danishefsky-Dien zu 2-substituierten Dehydropiperidinonen umgesetzt wurden. Die auf diese Weise zugänglichen chiralen Heterocyclen wurden diastereoselektiv in trans-konfigurierte 5-Bromverbindungen überführt. In einer Thiazolsynthese nach Hantzsch konnten die -Bromketone mit ambidenten Nukleophilen, wie Thiobenzamiden und unsymmetrischen Thioharnstoffderivaten, in niedrigen Ausbeuten zu bicyclischen Tetrahydro-thiazolo[5,4-c]pyridinen umgesetzt werden. Weitere bicyclische Heterocyclen mit einem Tetrahydro-thieno[2,3-c]pyridin-System konnten durch eine Gewald-Cyclisierung an 2-substituierten N-Galactosyl-piperidinonen erhalten werden. Durch Palladium-katalysierte Kreuzkupplungen an heterocyclischen Enoltriflaten, die ausgehend von den N-Galactosyl-dehydropiperidinonen synthetisiert wurden, gelang die Einführung von Aryl-, Alkinyl- und Alkenylsubstituenten in 4-Position des Piperidinringes. Zur Freisetzung der 2,4-disubstituierten Dehydropiperidinen wurde die N-glycosidische Bindung im sauren Milieu gespalten. Verbindungen mit einer exocyclischen Doppelbindung wurden einer Diels-Alder-Reaktion mit N-Phenylmaleinimid zum Aufbau von Isochinolinderivaten eingesetzt. Des Weiteren gelang die Synthese von 2-benzylsubstituierten N-Galactosyl-dehydropiperidinonen, wobei ortho-halogensubstituierte Phenylacetaldehyde eingesetzt wurden. Die in hohen Diastereomerenüberschüssen gebildeten Dehydropiperidinone wurden in die entsprechenden Enoltriflate überführt und einer Domino-Suzuki-Heck-Reaktion unterworfen. In dieser Kaskadenreaktion konnten tricyclische diastereomerenreine Benzomorphanderivate synthetisiert werden.

Die Zielsetzung dieser Arbeit war die Synthese und Markierung, sowie die in vitro- und in vivo-Evaluierung zweier markierter Aminosäure. Es wurden die PET-Tumor-Tracer C1-(2-[18F]Fluoreth- ylamino)-asparagin und S-2-Amino-4-[18F]fluor-butansäure synthetisiert. Die Markierung zum C1-(2-[18F]Fluorethylamino)-asparagin wurde mit 2-[18F]Fluorethylamin als Precursor durchgeführt. Ausgehend von N,N-Dibenzyl-(2-bromethyl)-amin wurde zunächst N,N-Dibenzyl-(2-[18F]fluorethyl)-amin in einer nukleophilen Substiution mit [18F]Fluorid in Acetonitril bei 80 °C mit einer Reaktionsdauer von 5 min dargestellt. Zur Abtrennung überschüssigen [18F]Fluorids wurde das Roh-Produkt auf einer Sep-Pak Plus Kartusche (C18) fixiert und mit Acetonitril eluiert. Die Abspaltung der Benzyl-Schutzgruppen erfolgte durch die Zugabe von Pd/C zu dem eluierten N,N-Dibenzyl-(2-[18F]fluorethyl)-amin und Behandelung der Lösung im leichten Wasserstoffstrom. Im finalen Aufreinigungsschritt wird der Katalysator Pd/C mittels eines Membranfilters abgetrennt. Das 2-[18F]Fluorethylamin wird im Sauren in das Amino-Salz überführt und im Vakuum vom Lösungsmittel befreit. Die Markierung zum C1-(2-[18F]Fluorethylamino)-asparagin wurde in DMF durchgeführt. Die höchsten Ausbeuten werden nach 4 min bei 60 °C erhalten. Die Abspaltung der Schutzgruppe erfolgt durch Trifluoressigsäure bei Raumtemperatur in 8 min. Die Aufreinigung des C1-(2-[18F]Fluorethylamino)-asparagin mittels HPLC und Festphasenextraktion liegt das Produkt in einer isotonischne isotonischen Kochsalzlösung vor. Die in vitro-Versuche wurden mit C1-(2-[18F]Fluorethylamino)-asparagin an 5 verschiedenen Zelllinen durchgeführt, 3 Plattenepitele und 2 Melanome. Hierbei konnte eine erhöhte Akkumulation von C1 -(2-[18F]Fluorethylamino)-asparagin beobachtet werden, die innerhalb von 20 min einen konstanten Wert erreicht. Ein Blockade-Experiment zeigte, dass sich die Aufnahme von C1 -(2-[18F]Fluorethylamino)-asparagin durch gesättinge Asparagin-Lösung bei den Plattenepitelen gar nicht und bei den Melanomen nur leicht vermindern ließ. Da die Aufnahme von C1-(2-[18F]Fluorethylamino)-asparagin in die Zellen höher war als bei FDG bei dem gleichen Versuchsaufbau, wurden in vivo-Versuche angesetzt. Die in vivo-Versuche an Sprague Dawley Ratten mit C1-(2-[18 F]Fluorethylamino)-asparagin zeigten keine messbare Anreicherung von C1-(2-[18F]Fluorethylamino)-asparagin in Tumoren. Fast die komplette Aktivität wurde in der Niere und Blase wiedergefunden. Die Synthese von S-2-Amino-4-[18F]fluor-butansäure wurde über stereodirigierende Auxilliare realisiert. Dazu wurden die geeignetsten Auxillare ausgewählt und auf ihre Eignung verglichen. Der Vergleich der stereodirigierenden Auxilare zeigte, dass das (1R,2R,5R)-2-Hydroxy-2,6,6-trimethyl- bicyclo[3.1.1]hept-3-ylidenamino)-essigsäure tert.-butylester (Laue-Auxillar) am geeignetesten ist. Die 18F-Markierung von S-2-Amino-4-[18F]fluor-butansäure wurde so optimiert, dass eine möglichst hohe stereochemische Reinheit des Produktes erzielt wird. Die optische Reinheit von S-2-Amino-4-[18 F]fluor-butansäure wurde mit > 93 % ee berechnet. Die Synthese des Markierungsvorläufers wurde ausgehend vom Laue-Auxilliar aufgebaut. Als Abgangsgruppe hat sich die Tosylgruppe besonders bewährt. Sie lässt sich unter besonders schonenden Bedingungen in den säurelabilen (1R,2R,5R)-4-Hydroxy-2-(2-hydroxy-2,6,6-trimethyl-bi- cyclo[3.1.1]hept-3-ylidenamino) butansäure tert.-butylester einführen. Bei der 18F-Markierung von S-2-Amino-4-[18F]fluor-butansäure mittels n.c.a. [18F]Fluorid hat sich die Wahl des Basensystems als besonders wichtig erwiesen. Die maximale Ausbeuten von 55% wurde mit Oxalat als Basensystem in Acetonitril bei 80 °C und einer Reaktionszeit von 15 min erzielen. Die Abspaltung der Schutzgruppen und die Abtrennung des Produktes wird in mehreren Schritten durchgeführt einschließlich einer HPLC-Abtrennung. Es wird nach 150 min Synthesedauer das gereinigte S-2-Amino-4-[18F]fluor-butansäure in isotonischer Kochsalzlösung mit einer Ausbeute von > 10 % RCA erhalten. Bei in vivo-Versuchen an Sprague Dawley Ratten reicherte sich der Hauptteil der Aktivität in der Niere an und nur weniger als ein halbes Prozent der applizierten Aktivität fand sich in den Tumoren wieder. Nach 10 min wurde ein maximaler Wert erreicht, der sich bis zum Ende der Messung nicht verändert. Das Verhältnis von Tumoraktivität zu unspezifisch-gebundener Aktivität betrug 2,2. Damit liegt das Verhältnis im Bereich der meisten klinisch eingesetzten PET-Tumor-Tracer wie dem des O-(2-[18F]Fluorethyl)-L-tyrosin mit 1,5.

The aim of this Ph.D. project has been the photophysical and photochemical characterization of new photo- and redox-active supramolecular systems. In particular we studied two different classes of compounds: metal complexes and dendrimers. Two different families of bis-cyclometalated neutral Ir(III) complexes are presented and their photophysical properties are discussed. The first family of complexes contains two 2-phenylpyridyl (ppy) or 2-(4,6-difluorophenyl)pyridyl (F2ppy) cyclometalated ligands and an ancillary ligand constituted by a phenol-oxazoline (phox), which can be substituted in the third position with a fluorine group (Fphox). In the second part of this study, we present another family of bis-cyclometalated Ir(III) complexes in which the ancillary ligand could be a chiral or an achiral bis-oxazoline (box). We report on their structural, electrochemical, photophysical, and photochemical properties. Complexes containing phox and Fphox ancillary ligands show blue luminescence with very high quantum yield, while complexes with box ligands do not show particularly interesting photophysical properties. Surprisingly these complexes give an unexpected photoreaction when irradiated with UV light in presence of dioxygen. This photoreaction originates a stable, strong blue emitting and particularly interesting photoproduct. Three successive generations of a family of polyethyleneglycol (PEG)-coated Pd(II) tetrabenzoporphyrin (PdTBP)-based dendritic nanoprobes are presented, and their ability to sensitize singlet oxygen and inflict cellular photodamage are discussed. It was found that the size of the dendrimer has practically no effect on the singlet oxygen sensitization efficiency, that approximate the unity, in spite of the strong attenuation of the triplet quenching rate with an increase in the dendrimer generation. Nevertheless, when compared against a commonly used singlet oxygen sensitizer, as Photofrin, the phosphorescent probes were found to be non-phototoxic. The lack of phototoxicity is presumably due to the inability of PEGylated probes to associate with cell surfaces and/or penetrate cellular membranes. The results suggest that protected phosphorescent probes can be safely used for oxygen measurements in biological systems in vivo. A new family of two photoswitchable (G0(Azo) and G1(Azo)) dendrimers with an azobenzene core, two cyclam units as coordination sites for metal ions, and luminescent naphthalene units at the periphery have been characterized and their coordination abilities have been studied. Because of their proximity, the various functional groups of the dendrimer may interact, so that the properties of the dendrimers are different from those exhibited by the separated functional units. Both the naphthalene fluorescence and the azobenzene photoisomerization can be observed in the dendrimer, but it has been shown that (i) the fluorescent excited state of the naphthalene units is substantially quenched by excimer and exciplex formation and by energy transfer to the azobenzene units, and (ii) in the latter case the fluorescence quenching is accompanied by the photosensitized isomerization of the trans → cis, and, with higher efficiency, the cis → trans reaction. Complexation of these dendrimers, both trans and cis isomers, with Zn(II) ions shows that complexes of 1:1 and 2:1 metal per dendrimer stoichiometry are formed showing different photophysical and photochemical properties compared to the corresponding free ligands. Practically unitary efficiency of the sensitized isomerization of trans → cis and cis → trans reaction is observed, as well as a slight increase in the naphthalene monomer emission. These results are consistent with the coordination of the cyclam amine units with Zn(II), which prevents exciplex formation. No indication of a concomitant coordination of both cyclam to a single metal ion has been obtained both for trans and cis isomer.

The studies conducted during my Phd thesis were focused on two different directions: 1. In one case we tried to face some long standing problems of the asymmetric aminocatalysis as the activation of encumbered carbonyl compounds and the control of the diastereoisomeric ratio in the diastero- and enantioselective construction of all carbon substituted quaternary stereocenters adjacent a tertiary one. In this section (Challenges) was described the asymmetric aziridination of ,-unsaturated ketones, the activation of ,-unsaturated -branched aldehydes and the Michael addition of oxindoles to enals and enones. For the activation via iminium ion formation of sterically demanding substrates, as ,-unsaturated ketones and ,-unsaturated -branched aldehydes, we exploited a chiral primary amine in order to overcome the problem of the iminium ion formation between the catalyst and encumbered carbonylic componds. For the control of diastereoisomeric ratio in the diastero- and enantioselective construction of all carbon substituted quaternary stereocenters adjacent a tertiary one we envisaged that a suitable strategy was the Michael addition to 3 substituted oxindoles to enals activated via LUMO-lowering catalysis. In this synthetic protocol we designed a new bifunctional catalyst with an amine moiety for activate the aldehyde and a tioureidic fragment for direct the approach of the oxindole. This part of the thesis (Challenges) could be considered pure basic research, where the solution of the synthetic problem was the goal itself of the research. 2. In the other hand (Molecules) we applied our knowledge about the carbonylic compounds activation and about cascade reaction to the synthesis of three new classes of spirooxindole in enantiopure form. The construction of libraries of these bioactive compounds represented a scientific bridge between medicinal chemistry or biology and the asymmetric catalysis.

Many physiological and pathological processes are mediated by the activity of proteins assembled in homo and/or hetero-oligomers. The correct recognition and association of these proteins into a functional complex is a key step determining the fate of the whole pathway. This has led to an increasing interest in selecting molecules able to modulate/inhibit these protein-protein interactions. In particular, our research was focused on Heat Shock Protein 90 (Hsp90), responsible for the activation and maturation and disposition of many client proteins [1], [2] [3]. Circular Dichroism (CD) spectroscopy, Surface Plasmon Resonance (SPR) and Affinity Capillary Electrophoresis (ACE) were used to characterize the Hsp90 target and, furthermore, its inhibition process via C-terminal domain driven by the small molecule Coumermycin A1. Circular Dichroism was used as powerful technique to characterize Hsp90 and its co-chaperone Hop in solution for secondary structure content, stability to different pHs, temperatures and solvents. Furthermore, CD was used to characterize ATP but, unfortunately, we were not able to monitor an interaction between ATP and Hsp90. The utility of SPR technology, on the other hand, arises from the possibility of immobilizing the protein on a chip through its N-terminal domain to later study the interaction with small molecules able to disrupt the Hsp90 dimerization on the C-terminal domain. The protein was attached on SPR chip using the “amine coupling” chemistry so that the C-terminal domain was free to interact with Coumermycin A1. The goal of the experiment was achieved by testing a range of concentrations of the small molecule Coumermycin A1. Despite to the large difference in the molecular weight of the protein (90KDa) and the drug (1110.08 Da), we were able to calculate the affinity constant of the interaction that was found to be 11.2 µm. In order to confirm the binding constant calculated for the Hsp90 on the chip, we decided to use Capillary Electrophoresis to test the Coumermycin binding to Hsp90. First, this technique was conveniently used to characterize the Hsp90 sample in terms of composition and purity. The experimental conditions were settled on two different systems, the bared fused silica and the PVA-coated capillary. We were able to characterize the Hsp90 sample in both systems. Furthermore, we employed an application of capillary electrophoresis, the Affinity Capillary Electrophoresis (ACE), to measure and confirm the binding constant calculated for Coumermycin on Optical Biosensor. We found a KD = 19.45 µM. This result compares favorably with the KD previously obtained on biosensor. This is a promising result for the use of our novel approach to screen new potential inhibitors of Hsp90 C-terminal domain.

The transition metal-catalyzed allylic alkylation (Tsuji-Trost type reaction) is a powerful tool for C-C, C-N, and C-O bond formation, which has been widely applied to organic chemistry over the last decades. Typical substrates for this transformation are activated allylic compounds such as halides, esters, carbonates, carbamates, phosphates, and so on. However, use of these substrates is associated with the disadvantage of generating a stoichiometric amount of chemical waste. Furthermore, these starting materials have to be prepared in an extra step from the corresponding allylic alcohol. Thus, ideal substrates would be the allylic alcohols themselves, with water being the only byproduct in this case. However, the scarse propensity of the hydroxyl moiety to act as good leaving group has significantly limited their use so far. During the last decade significant efforts have been made in order to develop more atom-economical and environmentally-friendly allylic alkylation protocols by employing allylic alcohols directly. In this PhD dissertation two main projects addressing this topic are presented. “Project 1” deals with the development of new metal-catalyzed intramolecular Friedel-Crafts (FC) allylic alkylations of electron-rich (PAPER A), as well as challenging electron-poor arenes (PAPER B) with alcohols. In “Project 2”, gold(I)-catalyzed intramolecular and stereoselective allylic alkylation reactions are reported. In particular, a FC alkylation of indole-containing allylic alcohols is presented in PAPER C. While, an O-alkylation of aminol-containing allylic alcohols is reported in PAPER D. To the best of knowledge, these reports represent the first example of gold(I)-catalyzed stereoselective alkylations with alcohols.

Dendrimere spielen als strukturtreue Nanopartikel eine herausragende Rolle. Ziel dieser Arbeit war, Dendrimere mit einer hohen Dichte an photoaktiven Chromophoren herzu-stellen und zu untersuchen. Dazu wurden die terminalen Aminogruppen von Poly(propylenimin)dendrimeren 1. und 2. Generation, Astramol DAB-Am-4R und DAB-Am-8R, mit Stilbenen und Styrylstilbenen als Chromophor verknüpft. Mittels Wittig-Horner- und Heck-Reaktion wurden (E)-Stilbene aufgebaut, die auf der einen Seite drei Propoxygruppen zur Verbesserung der Löslichkeit und auf der anderen Seite eine passende Funktionalität zur Verknüpfung mit dem dendritischen Core tragen. Als Verknüpfungsmethoden wurden die Verknüpfung als Amid (PSDA), Schiffsche Base (PSDS) und Harnstoff (PSDH) getestet. Die Schiffschen Basen wurden außerdem zur Erhöhung der Hydrolysestabilität zum sekundären Amin reduziert (PSDR und PQDR). Durch die Verknüpfung mit dem Core werden die stilbenoiden Chromophore sehr stark photoaktiviert. Das beruht auf einem Singulett-Energietransfer (Förster-Mechanismus) von Chromophor zu Chromophor. Dieser Prozeß konkurriert zu den Deaktivierungsprozessen, verlängert die mittlere S1-Lebensdauer und erhöht somit die Chancen der Photochemie. Der Styrylstilben-Chromophor hat darüber hinaus einen erheblichen Teil seiner UV-Absorbtion bereits im Tageslicht und photopolymerisiert daher bereits im Tageslicht. Vor allem bei den Dendrimeren 2. Generation stellte sich die Frage nach der vollständigen, d.h. achtfachen Umsetzung; das Core sollte als Knäuel vorliegen, die Arme zum Teil nach innen gefaltet und somit dem Reaktand nur bedingt zugänglich. Auch dort konnten unter optimierten Reaktionsbedingungen alle Aminogruppen umgesetzt werden. Die vollständige Umsetzung der Dendrimere wurde mittels NMR und massenspektroskopischen Methoden untersucht. Bei den Absorptionsspektren der Dendrimere 1. Generation ändert sich die Lage der Maxima je nach Art der Verknüpfung der Chromophore mit dem Core. Die Verlängerung des Chromophors um eine Styryleinheit bedingt eine beträchtliche Rotverschiebung. Die Lage der Emissionsmaxima differiert stärker als die Lage der Absorptionsmaxima. Den geringsten Stokes-Shift weist der Harnstoff auf, dann folgt das sekundäre Amin, dann die Schiffsche Base. Dies weist auf unterschiedlich relaxierte S1-Geometrien hin. Die Verbindungen PSDS1, PSDR1 und PSDH1 aus 3,4,5-Tripropoxystilbeneinheit und Astramol-Core 1. Generation DAB-Am-4 wurden in einer Konzentration von 10-5 mol/L belichtet. Der vollständige Photoabbau durch Belichtung in Chloroform mit einer Xenon-Lampe erfolgte ohne jeglichen Filter innerhalb von zehn Minuten (PSDH1), 20 Minuten (PSDR1) und einer Stunde (PSDS1). Allen drei Verbindungen gemeinsam ist das Entstehen eines intermediären neuen Maximums geringer Intensität, das um etwa 100 nm bathochrom verschoben ist. Das Harnstoffsystem weist außerdem ein weiteres intermediäres Maximum bei 614 nm auf. Diese Maxima können (laut früherer Untersuchungen) durch Oxidation entstandenen chinoiden Strukturen zugeordnet werden, deren Lebensdauer (im Sekundenbereich) zu kurz für eine NMR-Charakterisierung ist. PSDR1 wurde außerdem bei höheren Konzentrationen (10-4 und 10-3 mol/L) mit einer Quecksilberlampe mit Pyrex-Filter (lambda > 300 nm) belichtet. Dabei wird, wie erwartet, eine Verbreiterung der NMR-Signale beobachtet. Es bildet sich zunächst cis-Stilben. Außerdem läßt sich bei 4.3 ppm ein Signal beobachten, das von inter- oder intramolekular gebildeten Methinprotonen herrührt. Auch wenn laut MOPAC- und Kraftfeldrechnung die Doppelbindungen ungünstig für eine [pi2s + pi2s]-Cyclodimerisierung zueinander stehen, kann im photochemisch angeregten Zustand eine Geometrie vorherrschen, die die intramolekulare Kopf-Kopf-Cyclobutanbildung ermöglicht. Die massenspektrometrischen Untersuchungen der Belichtungsprodukte (FD, ESI, MALDI-TOF) zeigen als höchste Masse lediglich das Monomer. Allerdings kann dadurch nicht auf eine rein intramolekulare Reaktion geschlossen werden. Die fortschreitende statistische CC-Verknüpfung kann schnell zu vernetzten Nanopartikeln führen, die im Massenspektrometer nicht fliegen. Die NMR-Spektren der mit zunehmender Vernetzung immer schlechter löslich werdenden Teilchen belegen die Oligomerisierung.

In this thesis, the development of an enantioselective oxidation of α-branched aldehydes using covalent organocatalysis is described. At state of the art, the asymmetric organocatalysis approach, gave often serous difficulties for these kind of substrate respect “classic” aldehydes. We have used a primary cinchona alkaloid derived amine (specially the 9-epi-NH2-CDA) to develop the reaction in combinations with additives. With benzoyl peroxide as oxidant and 2-phenylpropionaldehyde as reference substrate, we have tried to optimize this system but we not found great results about enantiomeric excess.

During this internship, the α-alkylation of branched aldehydes was taken into consideration. An enantiopure Betti’s base derivative was used as catalyst, applying a new concept in catalysis: organocatalysis. The Betti’s base may be of particular interest for organic chemists working in the field of “reactions catalysed by enantiopure small organic molecules”, in particular for the ones interested in enantiopure primary amines. The potential of secondary amines as catalysts has certainly been known for years. It is indeed more innovative to conduct reactions using primary amine derivatives as catalyst. In this work, the efficacy of the primary amine was checked first. Then, the focus was set on finding optimal reaction conditions. Finally, to have a more complete picture of the structure of the compounds used in the project, experimental and computational IR spectra were compared, after the method was validated. Durante il periodo di tirocinio è stata presa in esame la reazione di α-alchilazione di aldeidi branched, utilizzando un derivato dell’ammina di Betti come catalizzatore enantiopuro ed applicando un nuovo tipo di catalisi: l’organocatalisi. Questi composti possono essere di particolare interesse per lavori in chimica organica, nel campo delle reazioni catalizzate da “piccole” molecole organiche, in particolare da ammine primarie a chiralità definita; la potenzialità delle ammine secondarie chirali come catalizzatori è certamente nota da anni, ma innovativo è condurre il tutto con l’impiego di un derivato amminico primario. Altri aspetti significativi sono gli apparenti e innumerevoli vantaggi, dal punto di vista economico ed ambientale, oltre che operativo e sintetico, derivanti dal nuovo tipo di catalisi. In un primo momento è stata verificata l’efficacia dell’ammina primaria sintetizzata nella reazione in progetto, quindi sono state individuate le condizioni di reazione ottimali. Infine, per un’analisi più completa di alcune molecole organiche e dopo un’opportuna validazione del metodo utilizzato, sono stati ottenuti a livello computazionale gli spettri IR delle molecole di sintesi prodotto e catalizzatore.

Nature leads, we follow. But nanotechnologists are in hot pursuit, in designing controllable structures that can mimic naturally occurring and artificially synthesized materials on a common platform. The supramolecular chemistry concerns the investigation of nature principles to produce fascinating complexed and functional molecular assemblies, as well as the utilization of these principles to generate novel devices and materials, potentially useful for sensing, catalysis, transport and other applications in medical or engineering science. The work presented in this thesis is a compilation of different synthetic methods to achieve inorganic-organic hybrid nanomaterials. Silicatein, a protein enzyme, which acts both as a catalyst and template for the formation of silica needles in marine sponges, has been used for the biosynthesis of semiconductor metal oxides on surfaces. Silicatein was immobilized on gold (111) surfaces using alkane thiol, as well as on a novel self-assembly of NTA on top of a “cushion” of reactive ester polymer has been successfully employed to make functionalised surfaces. The immobilization of silicatein on surfaces was monitored by surface plasmon spectroscopy, atomic force microscopy and confocal laser scanning microscopy. Surface bound silicatein retains its biocatalytic activity, which was demonstrated by monitoring its hydrocatalytic activity to catalyse the synthesis of biosilica, biotitania, and biozirconia. The synthesis of semiconductor metal oxides was characterized using scanning electron microscopy. This hydrolytic biocatalyst is used to synthesize the gold nanoparticles. The gold nanoparticles are formed by reduction of tetrachloroaurate, AuCl4-, by the action of sulfhydryl groups hidden below the surface groups of the protein. The resulting gold nanoparticles which are stabilized by surface bound silicatein further aggregate to form Au nanocrystals. The shape of the nanocrystals obtained by using recombinant silicatein is controlled through chiral induction by the protein during the nucleation of the nanocrystals. As an extension of this work, TiO2 nanowires were functionalized using polymeric ligand which incorporates the nitrilotriacetic acid (NTA) linker in the back bone to immobilize His-tagged silicatein onto the TiO2 nanowires. The surface bound protein not only retains its original hydrolytic properties, but also acts as a reductant for AuCl4- in the synthesis of hybrid TiO2/silicatein/Au nanocomposites. Functionalized, monocrystalline rutile TiO2 nanorods were prepared from TiCl4 in aqueous solution in the presence of dopamine. The surface bound organic ligand controls the morphology as well as the crystallinity and the phase selection of TiO2. The surface amine groups can be tailored further with functional molecules such as dyes. As an example, this surface functionality is used for the covalent binding of a fluorescent dye,4-chloro-7- nitrobenzylurazene (NBD) to the TiO2 nanorods. The polymeric ligands have been used successfully for the in-situ and post-functionalization of TiO2 nanoparticles. Besides to chelating dopamine anchor group the multifunctional ligand system presented here incorporates a modifier molecule which allows the binding of functional molecules (here the dyes pyrene, NBD, and Texas Red) as well as additional entities which allow tailoring the solubility of inorganic nanocrystals in different solvents. A novel method for the surface functionalization of fullerene-type MoS2 nanoparticles and subsequently binding these nanoparticles onto TiO2 nanowires has been reported using polymeric ligands. The procedure involves the complexation of IF-MoS2 with a combination of Ni2+ via an umbrella-type nitrilotriacetic acid (NTA) and anchoring them to the sidewalls of TiO2 nanowires utilizing the hydroxyl groups of dopamine present in the main contents of polymeric ligand. A convenient method for the synthesis of Au/CdS nanocomposites has been presented, which were achieved through the novel method of thiol functionalization of gold colloids. The thermodynamically most stable phase of ZrO2 (cubic) has been obtained at much lower temperature (180°C). These nanoparticles are highly blue fluorescent, with a high surface area.

Since conjugated polymers, i.e. polymers with spatially extended pi-bonding system have offered unique physical properties, unobtainable for conventional polymers, significant research efforts directed to better understanding of their chemistry, physics and engineering have been undertaken in the past two and half decades. In this thesis we discuss the synthesis, characterisation and investigation of conjugated semiconducting organic materials for electronic applications. Owing to the versatile properties of metal-organic hybrid materials, there is significant promise that these materials can find use in optical or electronic devices in the future. In addressing this issue, the synthesis of bisthiazol-2-yl-amine (BTA) based polymers is attempted and their metallation is investigated. The focus of this work has been to examine whether the introduction of coordinating metal ions onto the polymer backbone can enhance the conductivity of the material. These studies can provide a basis for understanding the photophysical properties of metal-organic polymers based on BTA. In their neutral (undoped) form conjugated polymers are semiconductors and can be used as active components of plastics electronics such as polymer light-emitting diodes, polymer lasers, photovoltaic cells, field-effect transistors, etc. Toward this goal, it is an objective of the study to synthesize and characterize new classes of luminescent polymeric materials based on anthracene and phenanthrene moieties. A series of materials based on polyphenylenes and poly(phenyleneethynylene)s with 9,10-anthrylene subunits are not only presented but the synthesis and characterization of step-ladder and ladder poly(p-phenylene-alt-anthrylene)s containing 9,10-anthrylene building groups within the main chain are also explored. In a separate work, a series of soluble poly-2,7- and 3,6-phenanthrylenes are synthesized. This can enable us to do a systematic investigation into the optical and electronic properties of PPP-like versus PPV-like. Besides, the self-organization of 3,6-linked macrocyclic triphenanthrylene has been investigated by 2D wide-angle X-ray scattering experiments performed on extruded filaments in solution and in the bulk. Additionally, from the concept that donor-acceptor materials can induce efficient electron transfer, the covalent incorporation of perylene tetracarboxydiimide (PDI) into one block of a poly(2,7-carbazole) (PCz)-based diblock copolymer and 2,5-pyrrole based on push-pull type material are achieved respectively.
