The aim of this study was to evaluate the effects of 7-epiclusianone (7-epi) on specific virulence attributes of Streptococcus mutans in vitro and on development of dental caries in vivo. 7-Epi was obtained and purified from fruits of Rheedia brasiliensis. We investigated its influence on surface-adsorbed glucosyltransferase (Gtf) B activity, acid production, and viability of S. mutans in biofilms, as well as on caries development using a rodent model. 7-Epi (100 mu g/mL) significantly reduced the activity of surface-adsorbed GtfB (up to 48.0 +/- 1.8 of inhibition at 100 mu g/mL) and glyco-lytic pH-drop by S. mutans in biofilms (125 and 250 mu g/mL) (vs. vehicle control, p < 0.05). In contrast, the test compound did not significantly affect the bacterial viability when compared to vehicle control (15% ethanol, p > 0.05). Wistar rats treated topically with 7-epi (twice daily, 60-s exposure) showed significantly smaller number of and less severe smooth-and sulcal-surface carious lesions (p < 0.05), without reducing the S. mutans viable population from the animals` dental biofilms. In conclusion, the natural compound 7-epiclusianone may be a potentially novel pharmacological agent to prevent and control dental caries disease.

Evaluation of students undertaking fieldwork education placements is a critical process in the health professions. As training programs and practice evolve, systems for assessing students need to be reviewed and updated constantly. In 1995, staff of the occupational therapy training program at the University of Queensland, Australia decided to develop a new tool for assessing student fieldwork performance. Using an action research methodology, a team developed the Student Placement Evaluation Form, a flexible and comprehensive criterion-referenced evaluation tool. The present paper examines action research as an appropriate methodology for considering real-life organisational problems in a systematic and participatory manner. The action research cycles undertaken, including preliminary information gathering, tool development, trial stages and current use of the tool, are detailed in the report. Current and future development of the tool is also described.

Background: Tissue Doppler may be used to quantify regional left ventricular function but is limited by segmental variation of longitudinal velocity from base to apex and free to septal walls. We sought to overcome this by developing a composite of longitudinal and radial velocities. Methods and Results. We examined 82 unselected patients undergoing a standard dobutamine echocardiogram. Longitudinal velocity was obtained in the basal and mid segments of each wall using tissue Doppler in the apical views. Radial velocities were derived in the same segments using an automated border detection system and centerline method with regional chords grouped according to segment location and temporally averaged. In 25 patients at low probability of coronary disease, the pattern of regional variation in longitudinal velocity (higher in the septum) was the opposite of radial velocity (higher in the free wall) and the combination was homogenous. In 57 patients undergoing angiography, velocity in abnormal segments was less than normal segments using longitudinal (6.0 +/- 3.6 vs 9.0 +/- 2.2 cm/s, P = .01) and radial velocity (6.0 +/- 4.0 vs 8.0 +/- 3.9 cm/s, P = .02). However, the composite velocity permitted better separation of abnormal and normal segments (13.3 +/- 5.6 vs 17.5 +/- 4.2 cm/s, P = .001). There was no significant difference between the accuracy of this quantitative approach and expert visual wall motion analysis (81% vs 84%, P = .56). Conclusion: Regional variation of uni-dimensional myocardial velocities necessitates site-specific normal ranges, probably because of different fiber directions. Combined analysis of longitudinal and radial velocities allows the derivation of a composite velocity, which is homogenous in all segments and may allow better separation of normal and abnormal myocardium.

Mental health reform in the western developed world has resulted in new models of care and changed work practices for all mental health professionals. Occupational therapists, as with other mental health professionals, have been required to assume new roles and responsibilities. Literature from the United Kingdom has reflected concern about this new way of working. The aim of this exploratory study was to examine the current work practices of and issues faced by Australian mental health occupational therapists. One hundred and forty-eight respondents (74%) answered an occupational therapy practice in mental health questionnaire. The results from this survey suggest that there are two quite distinct groups of occupational therapists working in mental health settings in Australia. One group works as rehabilitation therapists in traditional activity-focused work roles. The other group works as case manager therapists and employs a much broader spectrum of clinical and support roles. The issues facing therapists include the development and maintenance of a clearly defined role, generic case management versus discipline-specific roles, recruitment and retention, the need for research and evidence-based practice, professional standing, and education and professional development. The concerns over the role of occupational therapy in mental health were similar to those in previous British studies. The implications of these findings include a need for education and training at the undergraduate and postgraduate levels to equip mental health occupational therapists with both discipline-specific and generic skills.

This paper examines the use of on-line discussion as a medium for learning in a pre-service teacher education program. As part of an Education Studies course student teachers engaged in a discussion of issues related to technology and equity in schools. The design of the task and the subsequent analysis of the on-line text were part of a research project investigating whether and how communications technology can be used to integrate and extend the learning of teacher education students. The main argument developed in the paper is that through the on-line activity distinctive sets of writing practices were created. These practices enabled students to make connections between the often disparate parts of teacher education programs-theory and practice, campus and school, research and experience. Investiga as relações entre a formação continuada de professores e as práticas de partilha das experiências construídas na atuação docente nos anos iniciais do ensino fundamental, numa escola da Rede Municipal de Ensino de Marilândia–ES. Questiona, com base nas narrativas de cinco professoras, como a experiência compartilhada nos processos de formação continuada de professores dos anos iniciais do ensino fundamental na Rede Municipal de Ensino de Marilândia–ES é vivida e percebida por elas e quais sentidos são produzidos sobre a formação continuada. Pressupõe a perspectiva de uma formação continuada em processo, ao longo da vida, desenvolvida em redes coletivas de trabalho e partilha de experiências no cotidiano escolar e na vida das professoras. Em face dessa complexidade, a pesquisa de caráter qualitativo e abordagem metodológica ancorada na pesquisa narrativa (auto)biográfica tem, como procedimentos de investigação, as entrevistas biográficas direcionadas para a construção de narrativas orais e escritas, a observação participante dos sujeitos da pesquisa em momentos formativos instituídos, a análise de documentos da Secretaria Municipal de Educação de Marilândia e da escola e a aplicação de questionário para composição de dados acerca do perfil dos sujeitos da pesquisa. A definição do caminho metodológico deste estudo teve inspiração em Benjamin e Larrosa e, no campo da formação de professores, sustentação nos referenciais de Nóvoa, Souza, Freire e Giroux. Os resultados apontam uma variedade de sentidos sobre a formação continuada de professores, desde formas mais institucionalizadas até iniciativas menos formais, vividas por meio da partilha de experiências e de processos (auto)formativos, que contribuem para o desenvolvimento pessoal e profissional do professor e organizacional da escola, de modo mais aproximado do trabalho pedagógico na unidade escolar e na sala de aula. Aponta a necessidade da valorização das experiências dos professores e do investimento no estudo das práticas. Considera a existência, entre os professores, de uma responsividade compartilhada sobre a formação do outro, compondo um coletivo instituinte na escola. Assinala a potencialidade da pesquisa narrativa (auto)biográfica como opção metodológica que possibilita o entrecruzamento dos movimentos investigativos e formativos.

The determination of aminotranferases levels is very useful in the diagnosis of hepatopathies. In recent years, an elevated serum ALT level in blood donors has been associated with an increased risk of post-transfusion hepatitis (PTH). The purpose of the study was to research the factors associated with elevated ALT levels in a cohort of voluntary blood donors and to evaluate the relationship between increased ALT levels and the development of hepatitis C (HCV) infection. 166 volunteer blood donors with elevated ALT at the time of their first donation were studied. All of the donors were questioned about previous hepatopathies, exposure to hepatitis, exposure to chemicals, use of medication or drugs, sexual behaviour, contact with blood or secretions and their intake of alcohol. Every three months, the serum levels of AST, ALT, alkaline phosphatase, gamma glutamyl transpeptidase, cholesterol, triglyceride and glycemia are assessed over a two year follow-up. The serum thyroid hormone levels as well as the presence of auto-antibodies were also measured. Abdominal ultrasound was performed in all patients with persistently elevated ALT or AST levels. A needle biopsy of liver was performed in 9 donors without definite diagnostic after medical investigation. The presence of anti-HCV antibodies in 116 donors were assayed again the first clinical evaluation. At the end of follow-up period (2 years later) 71 donors were tested again for the presence of anti-HCV antibodies. None of donors resulted positive for hepatitis B or hepatitis C markers during the follow-up. Of the 116 donors, 101 (87%) had persistently elevated ALT serum levels during the follow-up. Obesity and alcoholism were the principal conditions related to elevated ALT serum levels in 91/101 (90.1%) donors. Hypertriglyceridemia, hypercholesterolemia, hypothyroidism and diabetes mellitus also were associated with increased ALT levels. Only 1/101 (0.9%) had mild chronic active non A-G viral hepatitis and 3/101 (2.9%) had liver biopsy with non-specific reactive hepatitis. The determination of ALT levels was not useful to detect donors infected with HCV at donation in Brazil, including the initial seronegative anti-HCV phase.

Relatório EPE - Relatório de estágio em Educação Pré-Escolar: O presente relatório de estágio foi realizado no âmbito da Unidade Curricular (UC) de Prática Pedagógica Supervisionada na Educação Pré-Escolar, inserida no Mestrado em Educação Pré-Escolar e Ensino do 1.º Ciclo do Ensino Básico. Sendo que este perspetiva o percurso de formação da mestranda, importa conceptualizar os referenciais teóricos e legais que sustentaram a sua ação e a metodologia utlizada durante todo o processo – investigação-ação – que inclui os seguintes processos educativos: a observação, a planificação, a ação, a avaliação e a reflexão. A Prática Pedagógica Supervisionada realizou-se na valência de creche, no Colégio Novo da Maia e teve como principais objetivos o desenvolvimento das competências profissionais e disposições subjacentes ao Perfil Geral e Específico de Desempenho do Educador de Infância. Apoiando a sua prática através da observação, recolha de dados, análise constante da ação e reflexão crítica permanente, a formanda, regulou-se pela metodologia de investigação-ação enquanto “processo em que os participantes analisam as suas próprias práticas educativas de uma forma sistemática e aprofundada” (Coutinho et al, 2009, p.360), com vista ao desenvolvimento de competências relativas a um educador enquanto investigador da própria ação. Importa acrescentar que o período de estágio foi de 210 horas e decorreu em díade de formação. A colaboração entre todos os atores intervenientes no contexto da prática pedagógica supervisionada “visa a construção de um perfil profissional que promova o desenvolvimento das competências socioprofissionais e pessoais, de forma fundamentada, reflexiva, integrada e autonomizante, à luz do princípio da aprendizagem ao longo da vida” (Ribeiro, 2013, p.2).

O presente relatório tem como intuito refletir sobre o desenvolvimento pessoal e profissional da mestranda, a sua experiência e os conhecimentos adquiridos em resultado da prática pedagógica supervisionada desenvolvida ao nível da Educação Pré-Escolar e do 1.º Ciclo do Ensino Básico ao longo deste mestrado. Ao longo da prática, a ação da mestranda foi orientada pela metodologia de investigação-ação, constituída por várias etapas cíclicas e articuladas (observação, planificação, ação, reflexão e avaliação) numa perspetiva socio construtivista. Esta estratégia possibilitou a planificação de atividades adequadas às caraterísticas específicas de cada grupo. Para isso foi indispensável adotar uma atitude profissional indagadora, de caráter investigativo e crítico-reflexivo, que permitiu agir intencionalmente em cada situação, procurando atender às necessidades e interesses de cada criança. A mestranda desenvolveu a sua forma pessoal de pensar e agir no decorrer da sua prática baseando-se em quadros teóricos e concetuais, visando uma diferenciação educativa e uma colaboração profissional e reflexiva. Em suma, os estágios desenvolvidos nos dois níveis educativos promoveram a edificação de uma postura profissional reflexiva e investigativa que favoreceu a tomada de decisões em contexto de prática. Neste contexto, foram reafirmadas competências profissionais e pessoais e abriu-se caminho para uma formação ao longo da vida, essencial para um docente generalista.

The development of human cell models that recapitulate hepatic functionality allows the study of metabolic pathways involved in toxicity and disease. The increased biological relevance, cost-effectiveness and high-throughput of cell models can contribute to increase the efficiency of drug development in the pharmaceutical industry. Recapitulation of liver functionality in vitro requires the development of advanced culture strategies to mimic in vivo complexity, such as 3D culture, co-cultures or biomaterials. However, complex 3D models are typically associated with poor robustness, limited scalability and compatibility with screening methods. In this work, several strategies were used to develop highly functional and reproducible spheroid-based in vitro models of human hepatocytes and HepaRG cells using stirred culture systems. In chapter 2, the isolation of human hepatocytes from resected liver tissue was implemented and a liver tissue perfusion method was optimized towards the improvement of hepatocyte isolation and aggregation efficiency, resulting in an isolation protocol compatible with 3D culture. In chapter 3, human hepatocytes were co-cultivated with mesenchymal stem cells (MSC) and the phenotype of both cell types was characterized, showing that MSC acquire a supportive stromal function and hepatocytes retain differentiated hepatic functions, stability of drug metabolism enzymes and higher viability in co-cultures. In chapter 4, a 3D alginate microencapsulation strategy for the differentiation of HepaRG cells was evaluated and compared with the standard 2D DMSO-dependent differentiation, yielding higher differentiation efficiency, comparable levels of drug metabolism activity and significantly improved biosynthetic activity. The work developed in this thesis provides novel strategies for 3D culture of human hepatic cell models, which are reproducible, scalable and compatible with screening platforms. The phenotypic and functional characterization of the in vitro systems performed contributes to the state of the art of human hepatic cell models and can be applied to the improvement of pre-clinical drug development efficiency of the process, model disease and ultimately, development of cell-based therapeutic strategies for liver failure.

Ultrastructural observations of spermatogenesis and sperm development of Saccocoelioides godoyi, an intestinal parasite of Leporinus friderici (Bloch, 1794) are described. The irregular-shaped spermatogonia form a peripheral layer, and show a prominent nucleus. Spermatocytes are larger than spermatogonia, and in the early stage present synaptonemal complex. Spermatids show nuclei smaller than the spermatocytes. Spermiogenesis is characterized by outgrowth of the zone of differentiation, presenting basal bodies, separated by an intercentriolar body. At the end of this process, the spermatozoa are released into the residual cytoplasmic mass. The spermatozoa of S. godoyi are elongate, similar to the pattern described for other Digenea, showing nuclei, mitochondria and two axonemes with the 9+1 configuration. The peripheral cortical microtubules on the dorsal and ventral faces are laterally interrupted.
