Premise of the study: A set of eight microsatellite (simple sequence repeat [SSR]) markers for Lippia alba, an important medicinal and cosmetic plant, was developed to aid studies of genetic diversity and to define efficient strategies for breeding programs. Methods and Results: Using a (CT)(8)- and (GT)(8)-enriched library, a total of 11 SSR loci were developed and optimized in L. alba. Of the 11 loci, eight were found to be polymorphic after screening 61 accessions from two populations. The parameters used to characterize loci were expected heterozygosity (H-e) and number of alleles. A total of 44 alleles were identified, with an average of 5.5 alleles per loci, which were moderately to highly informative according to H-e. Conclusions: These new SSR markers have potential for informing genetic diversity, allele mining, and mapping studies and will be used to generate information for breeding programs of L. alba

The microencapsulation of Lippia sidoides extracts in blends of carbohydrates was investigated. The extraction conditions were determined through a 2(2) factorial design. The effects of the plant:solvent ratio (A - 7.5:100 and 15:100 m/m) and the extraction time (B - 30 and 90 min) on thymol content of extractive solutions were evaluated, using a 2:1 (v/v) of ethanol:water at a temperature of 50 degrees C, as a solvent system. The selected extract was subjected to spray drying. Blends of maltodextrin and gum arabic at different proportions (4:1; 3:2; 2:3; 0:1) (m/m) were used as encapsulating material. The protective effects of the maltodextrin and gum arabic blends were evaluated by determination of the thymol retention in the dried product, which ranged from 70.2 to 84.2% (related to the content in the extractive solution). An increase in the gum arabic to maltodextrin (DE10) ratio has positive effect on thymol retention. L. sidoides extracts and spray-dried products showed antifungal activity against tested fungal strains (Candida albicans - ATCC 64548, Candida glabrata - ATCC 90030, Candida krusei - ATCC 6258, and Candida parapsilosis - ATCC 22019), evidencing their potential as a natural antifungal agent for medicinal, food, and cosmeceutical purposes. A commercial casein hydrolysate was microencapsulated in liposomes produced with non-purified soy lecithin, cryoprotected with two different disaccharides and lyophilized. The encapsulation efficiency of casein hydrolysate ranged from 30 to 40%. The powders were analyzed by differential scanning calorimetry (DSC), scanning electron micrography (SEM), infrared spectroscopy (FTIR) and wide angle X-ray diffraction (WAXD). DSC data revealed the presence of an exothermal transition in empty lyophilized liposomes, which was ascribed to the presence of a quasicrystalline lamellar phase (intermediary characteristics between the L-beta and L-c phases). The addition of peptides to the liposomal system caused the disappearance of this exothermic phenomenon, as they were located in the polar headgroup portion of the bilayer, causing disorder and preventing the formation of the quasicrystalline phase. Infrared data indicated the presence of the peptides in the lyophilized formulations and showed that the cryoprotectants interacted effectively with the polar heads of phospholipids in the bilayer.

The schistosomicidal effects of pimaradienoic acid (PA) and two derivatives, obtained by fungal transformation in the presence of Aspergillus ochraceus, were investigated. PA was the only compound with antischistosomal activity among the three diterpenes studied, with the ability to significantly reduce the viability of the parasites at concentrations ranging from 25 to 100 mu M. PA also promoted morphological alterations of the tegument of Schistosoma mansoni, separated all the worm couples, and affected the production and development of eggs. Moreover, this compound was devoid of toxicity toward human fibroblasts. In a preliminary in vivo experiment, PA at a dose of 100 mg/kg significantly diminished the number of parasites in infected Balb/c mice. Taken together, these results show that PA may be potentially employed in the discovery of novel schistosomicidal agents, and that diterpenes are an important class of natural compounds for the investigation of agents capable of fighting the parasite responsible for human schistosomiasis.

Objectives The extract and essential oil of clove (Syzygium aromaticum) are widely used because of their medicinal properties. Eugenol is the most important component of clove, showing several biological properties. Herein we have analysed the immunomodulatory/anti-inflammatory effect of clove and eugenol on cytokine production (interleukin (IL)-1 beta, IL-6 and IL-10) in vitro. Methods Macrophages were incubated with clove or eugenol (5, 10, 25, 50 or 100 mg/well) for 24 h. Concentrations that inhibited the production of cytokines were used before or after incubation with lipopolysaccharide (LPS), to verify a preventive or therapeutic effect. Culture supernatants were harvested for measurement of cytokines by enzyme-linked immunosorbent assay. Key findings Clove (100 mg/well) inhibited IL-1 beta, IL-6 and IL-10 production and exerted an efficient action either before or after LPS challenge for all cytokines. Eugenol did not affect IL-1 beta production but inhibited IL-6 and IL-10 production. The action of eugenol (50 or 100 mg/well) on IL-6 production prevented efficiently effects of LPS either before or after its addition, whereas on IL-10 production it counteracted significantly LPS action when added after LPS incubation. Conclusions Clove exerted immunomodulatory/anti-inflammatory effects by inhibiting LPS action. A possible mechanism of action probably involved the suppression of the nuclear factor-kB pathway by eugenol, since it was the major compound found in clove

The oregano is a plant, rich in essential oil and very used as spice in the preparation of foods. The objective of this paper was to analyze the viability of irrigation for oregano in Presidente Prudente, São Paulo state, Brazil, including economic risk factors, their effect on irrigation total cost, as well as the different pumping kinds. The Monte Carlo simulation was utilized to study the economic factors: fixed cost, labor, maintenance, pumping and water. The use of irrigation for the oregano in the region of Presidente Prudente is indicated because of its economic feasibility and the reduced risks. The average values of the benefit/cost for all water depths tested were higher than 1, indicating viability. The use of irrigation promoted lower risks compared to the non irrigated crop. The micro irrigation system presented greater sensitivity to changes of prices of the equipment associated to the variation of the useful life of the system. The oregano selling price was the most important factor involved in annual net profit. The water cost was the factor of lesser influence on the total cost. Due to the characteristic of high drip irrigation frequency there was no difference between the tariffs based in use hour of electric energy classified as green and blue, which are characterized by applying different rates on the energy consumption and demand according to the hours of day and times of the year. For the studied region it was recommended drip irrigation water management of oregano with the daily application of 100% of pan evaporation Class A using electric motor with tariffs blue or green.

Objective: The objective of this study was to compare the effects of a commercial CPC (cetylpyridinium chloride) mouthrinse containing 0.07% CPC (Crest® ProHealth Rinse) versus those provided by a commercial essential flavor oil mouthrinse (Listerine® Antiseptic) on dental plaque accumulation and prevention of gingivitis in an unsupervised 6 month clinical study. Methods: This was a double blind, 6-month, parallel group, positive controlled study involving 128 subjects who were balanced and randomly assigned to either positive control (essential oil) or experimental (CPC) mouthrinse treatment groups. The CPC mouthrinse passed proposed performance assays by the FDA for an OTC CPC mouthrinse. At baseline, subjects received a dental prophylaxis and began unsupervised rinsing twice daily with 20 ml. of their assigned mouthrinse for 30 seconds after brushing their teeth for 1 min. Subjects were assessed for gingivitis and gingival bleeding by the Gingival Index (GI) of Loe and Silness and plaque by the Silness and Loe Plaque Index (PI) at baseline and after 3 and 6 months of product use. Oral soft tissue health was also assessed. Microbiological samples were also taken for community profiling by the DNA-DNA checkerboard method. Results: Results show that after 3 and 6 months use there was no significant difference (p = 0.05) between the CPC and essential oil mouthrinse treatment groups for overall gingivitis status, gingival bleeding, and plaque. At 6 months the covariant (baseline) –adjusted mean GI and bleeding sites numbers for the CPC and essential oil mouthrinses were 0.52 and 0.53 and 5.5 and 6.3, respectively. Both mouth rinses were well tolerated by the subjects. Microbiological community profiles were similar for the 2 treatment group. Conclusion: This study shows that the 0.07% CPC mouthrinse can provide similar plaque and gingivitis benefits to those provided by an essential oil mouthrinse over a 6 month period.

Para determinar el poder antioxidante y conservante del aceite esencial de tomillo mendocino (Acantholippia seriphioides) en hamburguesas funcionales, conservadas a 4 ± 0.5 °C, se elaboraron medallones utilizando: 83 % carne vacuna 5 % grasa vacuna 5 % salvado de avena 5 % texturizado de soja 2 % sustituto graso 0.08 g NaCl/kg aceite esencial de tomillo (AET) 106 ufc/g Se estudió: • tipo de envasado: bolsas de poliamidapolietileno; atmósfera modificada: 70 % N 2 y 30 % CO2, y vacío; • tiempo de almacenamiento: 0, 1, 2, 3 y 4 semanas; • dosis de tomillo: 0 y 150 mg AET/kg Los datos se analizaron de acuerdo con un arreglo factorial 2 (tipo de envasado) x 5 (tiempos de almacenamiento) x 2 (dosis de AET) en un diseño de parcelas subdivididas, con 4 repeticiones. Se evaluaron TBA, pH, NBV, color, olor y carga microbiana. Las tres primeras variables se estudiaron mediante el ANOVA. Para color y olor se recurrió a un análisis sensorial descriptivo. La carga microbiana se representó gráficamente. El AET disminuyó el TBA, independientemente del tipo de envasado utilizado. El pH disminuyó en el tiempo siguiendo un modelo polinomial de 2° grado. El NBV resultó significativo para: envasamiento al vacío y 0 mg/kg AET. El AET mantiene los atributos sensoriales durante las dos primeras semanas pero no disminuye la carga microbiana. Conclusión: el AET tiene efecto antioxidante pero no conservante en hamburguesas funcionales de carne vacuna bajo las condiciones del ensayo.

El objetivo del presente trabajo fue estudiar la actividad tóxica del aceite esencial de laurel (Laurus nobilis L.) y del cineol, monoterpeno cíclico considerado un aleloquímico puro, sobre Brevicoryne brassicae L. en repollo (Brassica oleracea var. capitata L.). Las concentraciones de aceite esencial utilizadas fueron: 1; 1,5; 2 y 3%, y para cineol: 0,5; 1,5 y 2,5%, ambas formuladas en solución acuosa con 2% de oleato de polietilenglicol como emulsionante. Se utilizaron dos técnicas de aplicación: papeles impregnados y pulverización directa. A las 24 horas del tratamiento se evaluó el porcentaje de mortalidad. Los resultados se analizaron por ANOVA de dos vías y Test de Tukey. Se encontraron diferencias significativas para los productos evaluados en todas las concentraciones ensayadas. Las técnicas de aplicación no dieron diferencias significativas. La mayor mortalidad correspondió a 52% para el aceite esencial de laurel y 27,5% para cineol a las mayores concentraciones en ambos casos. Se concluye que los productos ensayados podrían ser una herramienta para el Manejo Integrado de Plagas para control de áfidos en cultivos hortícolas.

Se trabajó con aceite esencial obtenido por arrastre por vapor de Origanum x applii (criollo) y Origanum x majoricum (mendocino), cultivados en La Consulta, Mendoza, Argentina. Para evaluar su poder antioxidante y conservante en alimentos se determinó: rendimiento, polifenoles totales, cuantificación de timol y carvacrol mediante cromatografía en capa fina de alta resolución (HPTLC) y capacidad de secuestro de radicales libres (DPPH). La actividad conservante se evaluó in vitro, por el método de difusión en agar, frente a cultivos puros de cinco bacterias patógenas y seis cepas de bacterias lácticas que alteran alimentos. El rendimiento en aceite esencial de estos oréganos oscila entre los valores informados por distintos autores para variedades cultivadas en otras regiones. Se destaca el contenido de polifenoles de ambos en relación con los valores mencionados en la bibliografía para otros Origanum, prevaleciendo el timol sobre el carvacrol. Esto se condice con un superior nivel de secuestro de radicales libres de ambos respecto de los citados en la literatura. Los oréganos de La Consulta inhibieron todas las cepas ensayadas y fueron más efectivos contra Staphylococcus aureus. Lo expuesto corrobora el poder antioxidante y conservante del Origanum x applii y del Origanum x majoricum.

La productividad del orégano (Origanum vulgare L.) está determinada por la conjunción entre cantidad de biomasa acumulada y contenido de aceite esencial hasta el momento de su cosecha. Numerosos autores han constatado que dicho contenido es máximo al momento de floración pero los procesos que determinan la ocurrencia de la misma son poco claros en esta especie. A través de la prolongación artificial del fotoperíodo, se evaluó la sensibilidad fotoperiódica de dos subespecies tradicionales de orégano (Compacto: Origanum vulgare ssp. vulgare y Criollo: Origanum vulgare ssp. hirtum Ietsw.) y su incidencia en el desarrollo, duración de fases fenológicas y en la dinámica de crecimiento. Se encontró que ambas responden al aumento del fotoperíodo reduciendo la longitud de su ciclo. Ante estas condiciones, las mismas difirieron en la magnitud de su respuesta, siendo la subespecie Criollo más sensible que Compacto. Esto sugiere que el umbral fotoperiódico de inducción a floración es menor en el orégano Criollo que en el orégano Compacto. El acortamiento de la fase de desarrollo vegetativo en ambas subespecies generó menor número de nudos y longitud de ramas finales (disminución más notoria en la subespecie Criollo). El fotoperíodo extendido generó un cambio en el modelo de crecimiento de la longitud de ramas de lineal a lineal con meseta o cuadrático.

(E)-β-Farnesene is a sesquiterpene semiochemical that is used extensively by both plants and insects for communication. This acyclic olefin is found in the essential oil of peppermint (Mentha x piperita) and can be synthesized from farnesyl diphosphate by a cell-free extract of peppermint secretory gland cells. A cDNA from peppermint encoding (E)-β-farnesene synthase was cloned by random sequencing of an oil gland library and was expressed in Escherichia coli. The corresponding synthase has a deduced size of 63.8 kDa and requires a divalent cation for catalysis (Km for Mg2+ ≈ 150 μM; Km for Mn2+ ≈ 7 μM). The sesquiterpenoids produced by the recombinant enzyme, as determined by radio-GC and GC-MS, are (E)-β-farnesene (85%), (Z)-β-farnesene (8%), and δ-cadinene (5%) with the native C15 substrate farnesyl diphosphate (Km ≈ 0.6 μM; Vrel = 100) and Mg2+ as cofactor, and (E)-β-farnesene (98%) and (Z)-β-farnesene (2%) with Mn2+ as cofactor (Vrel = 80). With the C10 analog, GDP, as substrate (Km = 1.5 μM; Vrel = 3 with Mg2+ as cofactor), the monoterpenes limonene (48%), terpinolene (15%), and myrcene (15%) are produced.

The biosynthesis of the monoterpenes limonene and carvone in the fruit of caraway (Carum carvi L.) proceeds from geranyl diphosphate via a three-step pathway. First, geranyl diphosphate is cyclized to (+)-limonene by a monoterpene synthase. Second, this intermediate is stored in the essential oil ducts without further metabolism or is converted by limonene-6-hydroxylase to (+)-trans-carveol. Third, (+)-trans-carveol is oxidized by a dehydrogenase to (+)-carvone. To investigate the regulation of monoterpene formation in caraway, we measured the time course of limonene and carvone accumulation during fruit development and compared it with monoterpene biosynthesis from [U-14C]Suc and the changes in the activities of the three enzymes. The activities of the enzymes explain the profiles of monoterpene accumulation quite well, with limonene-6-hydroxylase playing a pivotal role in controlling the nature of the end product. In the youngest stages, when limonene-6-hydroxylase is undetectable, only limonene was accumulating in appreciable levels. The appearance of limonene-6-hydroxylase correlates closely with the onset of carvone accumulation. At later stages of fruit development, the activities of all three enzymes declined to low levels. Although this correlates closely with a decrease in monoterpene accumulation, the latter may also be the result of competition with other pathways for substrate.

A espécie endêmica G. elliptica R. E. Fries não apresentava estudos fitoquímicos e biológicos detalhados na literatura. Assim, o objetivo desse trabalho foi avaliar a composição química e as propriedades biológicas dos óleos essenciais, extratos brutos, alcaloides totais, tortas, frações das tortas, amostras isoladas dessa espécie. O material vegetal foi coletado em Paranapiacaba (Santo André, SP, Brasil). O óleo essencial extraído das folhas por destilação à vapor apresentou um rendimento de 0,2%. A análise histológica das folhas encontrou óleo em células oleíferas localizadas no parênquima esponjoso. A composição do óleo (CG-EM) indicou espatulenol e óxido de cariofileno como compostos majoritários. Os alcaloides totais foram obtidos dos extratos brutos das folhas e dos galhos e analisados por CG-EM, identificando quatro aporfinas (nornuciferina, estefarina, corituberina e asimilobina) e duas protoberberinas (discretamina e caseadina). Os alcaloides totais foram fracionados em coluna cromatográfica ou por Extração em Fase Sólida e purificados por cromatografia em camada preparativa, originando duas amostras (Amostra 9 e 10). Na Amostra 9, foram identificados dois alcaloides aporfinicos nornuciferina e asimilobina (CG-EM e RMN-1H). Na Amostra 10, foram identificados (LC-EM/EM) cinco alcaloides aporfínicos (desidronantenina, glaunidina, liriodenina, N-óxido de oliverina e telikovina) e um alcaloide protoberberínico (caseadina). Caseadina, glaunidina, N-óxido de oliverina e telikovina não foram previamente identificados em Guatteria. Os resíduos dos extratos brutos livres de alcaloides foram fracionados pelo método de partição com solventes de polaridade crescente. Os extratos brutos e as frações acetato de etila e butanólicas de folhas e galhos apresentaram flavonoides (NP-PEG). Nos ensaios biológicos, a melhor atividade antioxidante (sequestro do radical DPPH) foi encontrada para a fração clorofórmica dos galhos (EC50=24,25±1,14 µg/mL) e a torta dos galhos (EC50=26,23±4,20 µg/mL). No ensaio antimicrobiano pelo método turbidimétrico a atividade mais importante foi obtida contra Staphylococcus aureus (ATCC 6538) para os alcaloides totais dos galhos (CIM/CBM=0,12±0,01/0,26 mg/mL) e das folhas (CIM/CBM=0,21±0,01/0,28 mg/mL), e fração hexânica das folhas (CIM/CBM=0,24±0,02/>1 mg/mL). Uma alta atividade antitumoral foi observada frente a células humanas de mama (MCF-7) para Amostra 10 (IC50=2,28±0,18 µg/mL), fração de acetato de etila das folhas (IC50=4,47±0,40 µg/mL), óleo essencial (IC50=7,01±0,23 µg/mL) e os alcaloides totais das folhas (IC50=9,32±0,36 µg/mL). Para as células de próstata (PC-3), foi encontrada atividade para a Amostra 10 (IC50=1,37±0,36 µg/mL) e o óleo essencial (IC50=5,32±0,35 µg/mL). A futura aplicação dos extratos e frações de G. elliptica como um agente antitumoral parece ser segura, pois mantiveram uma viabilidade celular maior do que 90% no ensaio de citotoxicidade com culturas de fibroblasto murino (BALB/c 3T3, ATCC CCL-163) nas concentrações onde a atividade antitumoral foi promissora (<30 µg/mL) contra MCF-7 e/ou PC-3.
