Hypercapitalism: Language, new media and social perceptions of value

Using examples from contempoary policy and business discourses, and exemplary historical texts dealing with the notion of value, I put forward an argument as to why a critical scholarship that draws on media history, language analysis, philosophy and political economy is necessary to understand the dynamics of what is being called 'the global knowledge economy'. I argue that the social changes associated with new modes of value determination are closely associated with new media form.

Spontaneous generation and survival of blood dendritic cells in mononuclear cell culture without exogenous cytokines

Studies on purified blood dendritic cells (DCs) are hampered by poor viability in tissue culture. We, therefore, attempted to study some of the interactions/relationships between DCs and other blood cells by culturing unseparated peripheral blood mononuclear cell (PBMC) preparations in vitro. Flow cytometric techniques were used to undertake a phenotypic and functional analysis of DCs within the cultured PBMC population. We discovered that both the CD11c(+) and CD11c(-) CD123(hi) DC subsets maintained their viability throughout the 3-day culture period, without the addition of exogenous cytokines. This viability was accompanied by progressive up-regulation of the surface costimulatory (CD40, CD80, CD86) and activation (CMRF-44, CMRF-56, CD83) molecules. The survival and apparent production of DCs in PBMC culture (without exogenous cytokines) and that of sorted DCs (with cytokines) were evaluated and compared by using TruCOUNT analysis. Absolute DC counts increased (for CD123hi and CD11c+ subsets) after overnight culture of PBMCs. Single-cell lineage depletion experiments demonstrated the rapid and spontaneous emergence of new in vitro generated DCs from CD14(+)/CD16(+) PBMC radioresistant precursors, additional to the preexisting ex vivo DC population. Unlike monocyte-derived DCs, blood DCs increased dextran uptake with culture and activation. Finally, DCs obtained after culture of PBMCs for 3 days were as effective as freshly isolated DCs in stimulating an allogeneic mixed leukocyte reaction. Mitigated directness in Honglou meng: directive speech acts and politeness in eighteenth century Chinese

This paper is situated within a theoretical discussion that asserts a positive correlation between indirectness and politeness, and uses these claims as a springboard for an examination of polite ways of issuing directives in eighteenth century Chinese. On the basis of an analysis of directive speech acts from dialogue in the novel Honglou meng, it argues that the concept of indirectness, as it applies to the illocutionary transparency of individual speech acts, has no particular value in the culture and language of eighteenth century Chinese men. It is found that other linguistic devices such as particles, the reduplication of verbs, terms of address, and the presence and sequencing of supportive moves are far more significant to the communication of politeness. The findings suggest a need to rethink current theoretical positions on this subject and move beyond the analysis of individual speech acts to examine the role discourse structure and management play in the enactment of politeness. YouTubers as satirists: humour and remix in online video

This article aims to discuss the role humour plays in politics, particularly in a media environment overflowing with user-generated video. We start with a genealogy of political satire, from classical to Internet times, followed by a general description of “the Downfall meme,” a series of videos on YouTube featuring footage from the film Der Untergang and nonsensical subtitles. Amid video-games, celebrities, and the Internet itself, politicians and politics are the target of such twenty-first century caricatures. By analysing these videos we hope to elucidate how the manipulation of images is embedded in everyday practices and may be of political consequence, namely by deflating politicians' constructed media image. The realm of image, at the centre of the Internet's technological culture, is connected with decisive aspects of today's social structure of knowledge and play. It is timely to understand which part of “playing” is in fact an expressive practice with political significance.

As manifestações públicas de cultura popular têm sido abordadas por diferentes áreas do conhecimento (antropologia, sociologia, história, etnografia e outras), mas as ciências da comunicação têm-lhes dado relativamente menos atenção. Do estudo de festas populares, sobretudo na investigação literária da sua matriz religiosa, passamos à investigação e à análise dos processos comunicacionais destas manifestações. O encontro com uma nova área das ciências da comunicação, a folkcomunicação, foi decisivo para a opção por paradigmas e instrumentos de análise que nos têm apoiado na investigação em curso sobre comunicação popular. Este texto tem como principal objectivo apresentar a origem e a evolução desta teoria comunicacional, Folkcomunicação, teoria inspirada na Escola de Chicago. Trata-se de um artigo teórico/conceptual, resultado de pesquisa bibliográfica e análise do Estado da Arte em relação à cultura popular e à folkcomunicação.

English Language Learning Inside and Outside the Classroom in Portugal

This study investigates the way of learning the English language in Portugal. First-year students of the faculty of Social Sciences and Humanities of New University of Lisbon were selected as participants in the case study. As data collection tools a questionnaire and focus-groups were used. 115 students completed the designed questionnaire and after that 12 students were selected for the more detailed focus-group discussions. Results of the research show that most part of the students´ English knowledge is received from outside the classroom by means of movies, songs, computer games, the Internet, communication with friends and other sources. Also, the results show that motivation is very important in language learning process and motivated students acquire the language faster and easier.

Lexicultura na Língua Chinesa e na Lexicografia Bilingue de Chinês-Português

No contexto de internacionalização de hoje, acompanhando os contactos cada vez mais intensos entre o mundo chinês e o mundo lusófono, é exigida a maior atenção à questão da lexicultura na comunicação interlingual e intercultural, pois o que preocupa as partes envolvidas não é apenas descodificar os signos meramente linguísticos, mas também perceber a cultura que a língua, nomeadamente as unidades lexicais transportam, visando um comportamento culturalmente correto e adequado nessa comunicação. Partindo desta preocupação, este trabalho tenta, através da abordagem e análise da língua, da escrita e do léxico chineses, comprovar que o chinês, devido às suas peculiaridades inexistentes em outras línguas do mundo, constitui o vivo exemplo que exemplifica e alarga o conceito de lexicultura, pois a própria língua, sobretudo a sua escrita e as suas unidades lexicais, além de refletirem a cultura popular e partilhada, também contam verdadeiras histórias da Humanidade e da China. Baseando-se nesta abordagem e análise, a tese apresenta um novo modelo de dicionário cultural chinês-português, com propostas concretas de seleção de vedetas para a sua macroestrutura e de definição de vedetas na sua microestrutura, ao serviço da comunicação entre os dois mundos em questão.

In vitro chondrogenic commitment of human Wharton's jelly stem cells by co-culture with human articular chondrocytes.

Wharton's jelly stem cells (WJSCs) are a potential source of transplantable stem cells in cartilage-regenerative strategies, due to their highly proliferative and multilineage differentiation capacity. We hypothesized that a non-direct co-culture system with human articular chondrocytes (hACs) could enhance the potential chondrogenic phenotype of hWJSCs during the expansion phase compared to those expanded in monoculture conditions. Primary hWJSCs were cultured in the bottom of a multiwell plate separated by a porous transwell membrane insert seeded with hACs. No statistically significant differences in hWJSCs duplication number were observed under either of the culture conditions during the expansion phase. hWJSCs under co-culture conditions show upregulations of collagen type I and II, COMP, TGFβ1 and aggrecan, as well as of the main cartilage transcription factor, SOX9, when compared to those cultured in the absence of chondrocytes. Chondrogenic differentiation of hWJSCs, previously expanded in co-culture and monoculture conditions, was evaluated for each cellular passage using the micromass culture model. Cells expanded in co-culture showed higher accumulation of glycosaminoglycans (GAGs) compared to cells in monoculture, and immunohistochemistry for localization of collagen type I revealed a strong detection signal when hWJSCs were expanded under monoculture conditions. In contrast, type II collagen was detected when cells were expanded under co-culture conditions, where numerous round-shaped cell clusters were observed. Using a micromass differentiation model, hWJSCs, previously exposed to soluble factors secreted by hACs, were able to express higher levels of chondrogenic genes with deposition of cartilage extracellular matrix components, suggesting their use as an alternative cell source for treating degenerated cartilage.
