Durante los últimos 30 años se han creado una gran cantidad de índices e indicadores para evaluar la práctica totalidad de los países bajo distintas premisas. La tesis parte de un análisis detallado de más de un centenar de estos índices diferenciados entre los enfocados al desarrollo y los enfocados a la competitividad económica (véase Módulo I anexo) y tras esto, dentro del estudio teórico nos hemos centrado en 35 indicadores relacionados con la tecnología (capítulo 3, apartado 3.3.). La justificación, el objetivo de la investigación y la estructura de la tesis se presenta en el capítulo 2. Respecto a la metodología, tal y como se plantea en la hipótesis (apartado 2.2.), se presentan los criterios de selección de seis grupos de países (EP, EPC, EC, ECI, EI y EM)1, que se van a evaluar. Posteriormente se plantea el Protocolo de Cálculo para el total de grupos seleccionados dentro de los periodos 2005-2006 y 2007-2008 y se realiza una profunda evaluación estadística como se plantea dentro de la coherencia estadística explicada en el apartado 3.4.4.5. (también se dispone de los cálculos dentro de los Módulos II, III, IV y V anexos). Tras la metodología establecemos la construcción de un índice sintético NRI(A) y, tras esto, estudiamos las relaciones así como la interpretación de los resultados (capítulo 4, apartado 4.1.). Una vez obtenidos los resultados realizamos la validación de los mismos para el periodo 2007-2015 (capítulo 4 - apartado 4.2. – y los Módulos VI y VII anexos). En el capítulo 5, evaluamos el nivel de preparación tecnológico y su relación con la competitividad para los seis grupos de países (véase desde los apartados 5.1.y 5.2.). Dentro de cada uno de los seis grupos de países, sabemos las variables que cualitativamente tienen que priorizarse para mejorar el nivel de preparación tecnológica de los mismos. Estas variables inicialmente son sesenta y ocho – año 2007-08 –, y al final de la implementación del método se reducen notablemente. Estas variables finales, llamadas Indicadores Clave de Actuación (ICA), se agrupan – vía análisis factorial – en Factores Clave de Actuación que nos simplifican lo planteado. Para cada grupo de países se realizan los conglomerados de acuerdo a sus valores dentro de las ICAs en busca de singularidades y se ha llevado a cabo un análisis minucioso en función de los Indicadores Clave de Actuación. La Tesis, plantea científicamente como podemos evaluar el nivel de preparación tecnológica y su relación con la competitividad, desde un índice sintético creado NRI(A), que contempla únicamente Indicadores Clave de Actuación (variables seleccionadas) a partir de las variables originales del Network Readiness Index (NRI(R)) . Por último se plantea dentro de las conclusiones, capítulo 6, diferentes líneas de investigación, desarrollando dos de ellas que se pueden encontrar en el Modulo VIII anexo. Por un lado presentamos una línea de investigación centrada en 29 economías africanas (EA) de las que disponemos información fidedigna y por otro lado una segunda línea en la que nos centramos en la evaluación de España respecto a sus naciones coetáneas. La principal voluntad de la presente tesis doctoral, es simplificar la evaluación del nivel de preparación tecnológica y la relación de esta con la competitividad a partir de la creación de un índice sintético propio NRI(A). ABSTRACT - During the last 30 years, many institutions have been evaluating and endless range of variables in practically all of the world´s economies. This Thesis is the product of a detail analysis of more than one hundred indicators / index, which we have divided into two parts: those focused on development and those focused on economic competitiveness (see module I annex). Secondly, in our theoretical research we have concentrated on those indicators, which are related to technology (chapters 3, section 3.3). The selection criteria of the six economic groups to be evaluated are included in our methodology, as mentioned in the hypothesis (see section 2.2.). Subsequently the calculation procedure is also presented for all of the groups selected between the periods 2005-2006 and 2007-2008. Next, we perform a statistical study, which is presented accordingly in the segment dealing with statistics, section 3.4.4.5. The calculations are provided in modules I, II, III, IV and V annex. After the methods segment of the Thesis, we develop our argument, in which we presented the explanation of the relations as well as the interpretation of the results. Also at the chapter 4 you can find the result validation from 2007 till 2015. Finally in chapters 6, we evaluate the conclusions for the six economic groups (see section 6.2.). The Thesis scientifically explains the way in which we evaluate economic competitiveness in 135 countries from a standpoint of strictly technological variables. Six groups of countries are evaluated, being divided by criteria, which homogenize the economies under review. We recognize that the variables of each economic group should be prioritized in order to better their competitiveness. Initially the group consisted of 68 variables, a number which was considerably reduced after the implementation of our methodology. Likewise, these final variables, dubbed “key performance indicators”, were grouped into factors (key performance factors), which greatly simplify the prioritization process. At the same time, conglomerates have been created for each economic group according to their value concerning the selected variables. A detailed country – by – country analysis of their positioning in each of the six groups was conducted for each of the mathematically selected key performance indicators. Finally, at the Conclusion we introduce new research lines and between them we focus on two research lines in which ones we are working with (see chapter 6). We basically try to apply the multivariable analysis method, the factorial analysis and the conglomerates to designed and implemented our method first in a geographically group of countries (Africa) and secondly to evaluate and develop the public policies for Spain for the development of its competitively, comparing Spain to his coetaneous countries in Europe (see Module VIII). The main objective of this Doctoral Thesis is to noticeably simplify the comparison of the Network Readiness Index and its relation with the economic competitiveness of the countries using a new synthetic index design by us.

A sexualidade de Lea e Raquel, o útero, as mandrágoras e o corpo de Jacó são fatores que definem o alicerce do nosso texto como espaços de diálogo, mediação e estrutura do cenário. O destaque principal está sob o capítulo 30.14-16 que retrata a memória das mandrágoras. Como plantas místicas elas dominam o campo religioso e como plantas medicinais elas são utilizadas para solucionar problemas biológicos. As instituições e sociedades detentoras de uma ideologia e de leis que regulamentam uma existência apresentam na narrativa, duas irmãs, mas também esposas de um mesmo homem que, manipuladas por essa instituição que minimiza e oprime a mulher, principalmente a estéril, confina-as como simples objeto de sexualidade e mantenedoras da descendência por meio da maternidade. A memória das mandrágoras é sinal de que a prática existente circundava uma religião não monoteísta. Ela existia sociologicamente por meio de sincretismos, força e poderes sócio-culturais e religiosos. Era constituída das memórias de mulheres que manipulavam e dominavam o poder sagrado para controle de suas necessidades. O discurso dessas mulheres, em nossa unidade, prova que o discurso dessa narrativa não se encontra somente no plano individual, mas também se estende a nível comunitário, espaço que as define e lhes concede importância por meio do casamento e dádivas da maternidade como continuidade da descendência. São mulheres que dominaram um espaço na história com suas lutas e vitórias, com atos de amor e de sofrimento, de crenças e poderes numa experiência religiosa dominada pelo masculino que vai além do nosso conhecimento atual. As lutas firmadas na fé e na ideologia dessas mulheres definiram e acentuaram seu papel de protagonistas nas narrativas 9 bíblicas que estudamos no Gênesis. A conservação dessas narrativas, e do espaço teológico da época, definiu espaços, vidas, gerações e tribos que determinaram as gerações prometidas e fecharam um ciclo: o da promessa de Iahweh quanto à descendência desde Abraão. Os mitos e as crenças foram extintos para dar espaço a uma fé monoteísta, mas a experiência religiosa

Neurons undergoing targeted photolytic cell death degenerate by apoptosis. Clonal, multipotent neural precursor cells were transplanted into regions of adult mouse neocortex undergoing selective degeneration of layer II/III pyramidal neurons via targeted photolysis. These precursors integrated into the regions of selective neuronal death; 15 ± 7% differentiated into neurons with many characteristics of the degenerated pyramidal neurons. They extended axons and dendrites and established afferent synaptic contacts. In intact and kainic acid-lesioned control adult neocortex, transplanted precursors differentiated exclusively into glia. These results suggest that the microenvironmental alterations produced by this synchronous apoptotic neuronal degeneration in adult neocortex induced multipotent neural precursors to undergo neuronal differentiation which ordinarily occurs only during embryonic corticogenesis. Studying the effects of this defined microenvironmental perturbation on the differentiation of clonal neural precursors may facilitate identification of factors involved in commitment and differentiation during normal development. Because photolytic degeneration simulates some mechanisms underlying apoptotic neurodegenerative diseases, these results also suggest the possibility of neural precursor transplantation as a potential cell replacement or molecular support therapy for some diseases of neocortex, even in the adult.

The positions of ≈4,800 individual miniature inverted-repeat transposable element (MITE)-like repeats from four families were mapped on the Caenorhabditis elegans chromosomes. These families represent 1–2% of the total sequence of the organism. The four MITE families (Cele1, Cele2, Cele14, and Cele42) displayed distinct chromosomal distribution profiles. For example, the Cele14 MITEs were observed clustering near the ends of the autosomes. In contrast, the Cele2 MITEs displayed an even distribution through the central autosome domains, with no evidence for clustering at the ends. Both the number of elements and the distribution patterns of each family were conserved on all five C. elegans autosomes. The distribution profiles indicate chromosomal polarity and suggest that the current genetic and physical maps of chromosomes II, III, and X are inverted with respect to the other chromosomes. The degree of conservation of both the number and distribution of these elements on the five autosomes suggests a role in defining specific chromosomal domains.

Estrogen receptor β (ERβ) is expressed at high levels in both neurons and glial cells of the central nervous system. The development of ERβ knockout (BERKO) mice has provided a model to study the function of this nuclear receptor in the brain. We have found that the brains of BERKO mice show several morphological abnormalities. There is a regional neuronal hypocellularity in the brain, with a severe neuronal deficit in the somatosensory cortex, especially layers II, III, IV, and V, and a remarkable proliferation of astroglial cells in the limbic system but not in the cortex. These abnormalities are evident as early as 2 mo of age in BERKO mice. As BERKO mice age, the neuronal deficit becomes more pronounced, and, by 2 yr of age, there is degeneration of neuronal cell bodies throughout the brain. This is particularly evident in the substantia nigra. We conclude that ERβ is necessary for neuronal survival and speculate that this gene could have an important influence on the development of degenerative diseases of the central nervous system, such as Alzheimer's disease and Parkinson's disease, as well as those resulting from trauma and stroke in the brain.

Cleavage and polyadenylation of mRNA 3′ ends in Saccharomyces cerevisiae requires several factors, one of which is cleavage factor I (CF I). Purification of CF I activity from yeast extract has implicated numerous proteins as functioning in both cleavage and/or polyadenylation. Through reconstitution of active CF I from separately expressed and purified proteins, we show that CF I contains five subunits, Rna14, Rna15, Pcf11, Clp1, and Hrp1. These five are necessary and sufficient for reconstitution of cleavage activity in vitro when mixed with CF II, and for specific polyadenylation when mixed with polyadenylation factor I, purified poly(A) polymerase, and poly(A) binding protein. Analysis of the individual protein–protein interactions supports an architectural model for CF I in which Pcf11 simultaneously interacts with Rna14, Rna15, and Clp1, whereas Rna14 bridges Rna15 and Hrp1.

We had earlier identified the pcnB locus as the gene for the major Escherichia coli poly(A) polymerase (PAP I). In this report, we describe the disruption and identification of a candidate gene for a second poly(A) polymerase (PAP II) by an experimental strategy which was based on the assumption that the viability of E. coli depends on the presence of either PAP I or PAP II. The coding region thus identified is the open reading frame f310, located at about 87 min on the E. coli chromosome. The following lines of evidence support f310 as the gene for PAP II: (i) the deduced peptide encoded by f310 has a molecular weight of 36,300, similar to the molecular weight of 35,000 estimated by gel filtration of PAP II; (ii) the deduced f310 product is a relatively hydrophobic polypeptide with a pI of 9.4, consistent with the properties of partially purified PAP II; (iii) overexpression of f310 leads to the formation of inclusion bodies whose solubilization and renaturation yields poly(A) polymerase activity that corresponds to a 35-kDa protein as shown by enzyme blotting; and (iv) expression of a f310 fusion construct with hexahistidine at the N-terminus of the coding region allowed purification of a poly(A) polymerase fraction whose major component is a 36-kDa protein. E. coli PAP II has no significant sequence homology either to PAP I or to the viral and eukaryotic poly(A) polymerases, suggesting that the bacterial poly(A) polymerases have evolved independently. An interesting feature of the PAP II sequence is the presence of sets of two paired cysteine and histidine residues that resemble the RNA binding motifs seen in some other proteins.

The primate temporal cortex has been demonstrated to play an important role in visual memory and pattern recognition. It is of particular interest to investigate whether activity-dependent modification of synaptic efficacy, a presumptive mechanism for learning and memory, is present in this cortical region. Here we address this issue by examining the induction of synaptic plasticity in surgically resected human inferior and middle temporal cortex. The results show that synaptic strength in the human temporal cortex could undergo bidirectional modifications, depending on the pattern of conditioning stimulation. High frequency stimulation (100 or 40 Hz) in layer IV induced long-term potentiation (LTP) of both intracellular excitatory postsynaptic potentials and evoked field potentials in layers II/III. The LTP induced by 100 Hz tetanus was blocked by 50-100 microM DL-2-amino-5-phosphonovaleric acid, suggesting that N-methyl-D-aspartate receptors were responsible for its induction. Long-term depression (LTD) was elicited by prolonged low frequency stimulation (1 Hz, 15 min). It was reduced, but not completely blocked, by DL-2-amino-5-phosphonovaleric acid, implying that some other mechanisms in addition to N-methyl-DL-aspartate receptors were involved in LTD induction. LTD was input-specific, i.e., low frequency stimulation of one pathway produced LTD of synaptic transmission in that pathway only. Finally, the LTP and LTD could reverse each other, suggesting that they can act cooperatively to modify the functional state of cortical network. These results suggest that LTP and LTD are possible mechanisms for the visual memory and pattern recognition functions performed in the human temporal cortex.

The Mn K-edge x-ray absorption spectra for the pure S states of the tetranuclear Mn cluster of the oxygen-evolving complex of photosystem II during flash-induced S-state cycling have been determined. The relative S-state populations in samples given 0, 1, 2, 3, 4, or 5 flashes were determined from fitting the flash-induced electron paramagnetic resonance (EPR) multiline signal oscillation pattern to the Kok model. The edge spectra of samples given 0, 1, 2, or 3 flashes were combined with EPR information to calculate the pure S-state edge spectra. The edge positions (defined as the zero-crossing of the second derivatives) are 6550.1, 6551.7, 6553.5, and 6553.8 eV for S0, S1, S2, and S3, respectively. In addition to the shift in edge position, the S0--> S1 and S1--> S2 transitions are accompanied by characteristic changes in the shape of the edge, both indicative of Mn oxidation. The edge position shifts very little (0.3 eV) for the S2--> S3 transition, and the edge shape shows only subtle changes. We conclude that probably no direct Mn oxidation is involved in this transition. The proposed Mn oxidation state assignments are as follows: S0 (II, III, IV, IV) or (III, III, III, IV), S1 (III, III, IV, IV), S2 (III, IV, IV, IV), S3 (III, IV, IV, IV).

Activity-dependent plasticity is thought to underlie both formation of appropriate synaptic connections during development and reorganization of adult cortical topography. We have recently cloned many candidate plasticity-related genes (CPGs) induced by glutamate-receptor activation in the hippocampus. Screening the CPG pool for genes that may contribute to neocortical plasticity resulted in the identification of six genes that are induced in adult visual cortical areas in response to light. These genes are also naturally induced during postnatal cortical development. CPG induction by visual stimulation occurs primarily in neurons located in cortical layers II-III and VI and persists for at least 48 hr. Four of the visually responsive CPGs (cpg2, cpg15, cpg22, cpg29) are previously unreported genes, one of which (cpg2) predicts a "mini-dystrophin-like" structural protein. These results lend molecular genetic support to physiological and anatomical studies showing activity-dependent structural reorganization in adult cortex. In addition, these results provide candidate genes the function of which may underlie mechanisms of adult cortical reorganization.

The symbiotic pattern of expression of Rhizobium meliloti N2-fixation genes is tightly coupled with the histological organization of the alfalfa root nodule and thus is under developmental control. N2-fixation gene expression is induced very sharply at a particular zone of the nodule called interzone II-III that precedes the zone where N2 fixation takes place. We show here that this coupling can be disrupted, hereby resulting in ectopic expression of N2-fixation genes in the prefixing zone II of the nodule. Uncoupling was obtained either by using a R. meliloti strain in which a mutation rendered N2-fixation gene expression constitutive with respect to oxygen in free-living bacterial cultures or by placing nodules induced by a wild-type R. meliloti strain in a microoxic environment. These results implicate oxygen as a key determinant of the symbiotic pattern of N2-fixation gene expression.

The phenotype and antigenic specificity of cells secreting interleukin (IL) 4, IL-6, and interferon gamma was studied in mice during primary and secondary immune responses. T lymphocytes were the major source of interferon gamma, whereas non-B/non-T cells were the dominant source of IL-4 and IL-6 in the spleens of immunized animals. Cytokine-secreting non-B/non-T cells expressed surface receptors for IgE and/or IgG types II/III. Exposing these cells to antigen-specific IgE or IgG in vivo (or in vitro) "armed" them to release IL-4 and IL-6 upon subsequent antigenic challenge. These findings suggest that non-B/non-T cells may represent the "natural immunity" analogue of CD4+ T helper type 2 cells and participate in a positive feedback loop involved in the perpetuation of T helper type 2 cell responses.

O Opúsculo em favor do insensato de Gaunilo constitui uma cerrada discussão filosófica da expressão “esse in intellectu” (existir no intelecto), ponto determinante do argumento construído por Anselmo no Proslogion II-III para demonstrar a verdade da existência de Deus a partir da existência no intelecto da sua definição enquanto “algo maior que o qual nada pode ser pensado”. Analisam-se aqui as razões de Gaunilo e as suas concepções de intelecção e de cogitação para desmontar o ponto de apoio principal do argumento de Anselmo, que, por sua vez, respondeu ao seu crítico com um Livro apologético. Indaga-se, em consequência, a clarificação por Anselmo sobre a intellectio e a cogitatio e se nela os dois conceitos são reposicionados para evitar as críticas de Gaunilo e assim continuar a manter que a razão, por si mesma, sem auxílio da experiência ou da fé, pode obter a certeza da existência que refuta o dito do insensato “Deus não existe”.
