In the environment humans and biota are generally exposed to chemical mixtures rather than individual chemicals. Therefore, when assessing the environmental risk of chemicals, it is important to consider chemical mixtures and their possible interactions. The main objective of this work focused on the environmental risk assessment of pesticides found in the water of the Alqueva reservoir and their binary combinations. In this aquatic ecosystem several pesticides were above of the environmental quality standards. But in addition, there were several sampling points of the reservoir where ecotoxicity was observed despite the presence of these contaminants at low concentrations. Here, a component-based approach was used to assess the effects of the pesticide mixtures. The effects of the binary combinations of four herbicides, atrazine (ATR), terbuthylazine (TER), simazine (SIM) and metolachlor (MET), on the growth rate of the microalgae Pseudokirchneriella subcapitata and the effects of the binary combinations of the s-triazine herbicides ATR and TER and the insecticide chlorpyrifos (CPF) on the swimming behaviour and acetylcholinesterase (AChE) activity of the zebrafish Danio rerio were assessed using the two reference models of concentration addition (CA) and independent action (IA). Moreover, the combined effects of the herbicides (ATR, TER and MET) and the insecticide CPF were also tested on the swimming behaviour and AChE activity of the aquatic midge Chironomus riparius after the cholinesterases characterization. In this risk characterization, the calculated risk quotients for the herbicides ATR, TER, SIM and MET were higher than 1, meaning that these herbicides present a high risk for the Alqueva ecosystem. As expected, the microalgae P. subcapitata was the most sensitive species to the herbicides. However, despite these herbicides pose no or low risk to other aquatic organisms tested in this study, with EC50 values much higher than the concentrations found in this aquatic ecosystem, they are able to increase the toxic effects of CPF when they are tested in binary mixtures. Moreover, the risk quotients of mixtures of these herbicides present simultaneously in three different locations of the reservoir were also higher than 1, so this confirms the fact that these herbicides when present in mixtures, present a greater risk for this ecosystem than the expected considering each single chemical by its own.

The development of mining activities over thousands of years in the region of Aljustrel is nowadays visible as a vast area of ore tailings, slag and host rocks of sulphides mineralization. The generation of acidic waters by the alteration of pyritic minerals - Acid Mine Drainage (AMD) - causes a significant impact on the river system both in the south of the village (Rib ª. Água Forte) and in the north of it (Rib ª. Água Azeda and Barranco do Farrobo), which is reflected in extremely low pH values (< 3) and high concentrations of As, Cd, Cu, Fe, Mn, Pb, Zn and sulphates. This study aimed to assess the environmental impacts extent, integrating geochemical (surface waters and stream sediments) and biological (diatoms) parameters. Three groups of sites were defined, based on sediments and water analysis, which integration with diatom data showed the same association of groups: Group 1- impacted, with acidic pH (1.9-5.1), high metal contents (0.4-1975 mg L-1) and Fe-Mg-sulphate waters, being metals more bioavailable in waters in cationic form (Me2+); mineralogically the sediments were characterized by phyllosilicates and sulphates/oxy-hydroxysulphate phases, easily solubilized, retaining a high amount of metals when precipitated; dominant taxon was Pinnularia aljustrelica (a new species); Group 2- slightly impacted, weak acid to neutral pH (5.0-6.8), metal contents not so high (0.2-25 mg L-1) and Fe-Mg-sulphate to Mg-chloride waters; dominant taxa were Brachysira neglectissima and Achnanthidium minutissimum; Group 3- unimpacted, alkaline pH (7.0-8.4), low metal contents (0-7 mg L-1) with Mg-chloride waters. In this group, metals were associated to the primary phases (e.g. sulphides), not so easily available; the existence of high chloride contents explained the presence of typical taxa of brackish/marine (e.g. Entomoneis paludosa) waters. Taxonomical aspects of the diatoms were studied (discovery of a new species: Pinnularia aljustrelica Luis, Almeida et Ector sp. nov.), as well as morphometric (size decrease of diatoms valves, as well as the appearance of deformed valves of Eunotia exigua in Group 1 and A. minutissimum in Group 2) and physiological (effective to assess the effects of metals/acidity in the photosynthetic efficiency through PAM Fluorometry) aspects. A study was carried out in an artificial river system (microcosm) that aimed to mimic Aljustrel’s extreme conditions in controlled laboratory conditions. The chronic effects of Fe, SO42- and acidity in field biofilms, inoculated in the artificial rivers, were evaluated as well as their contribution to the communities’ tolerance to metal toxicity, through acute tests with two metals (Cu and Zn). In general, the effects caused by low pH values and high concentrations of Fe and SO42- were reflected at the community level by the decrease in diversity, the predominance of acidophilic species, the decrease in photosynthetic efficiency and the increase of enzymatic (e.g. catalase, superoxide dismutase) and non-enzymatic activities (e.g. total glutathione and total phytochelatins). However, it was possible to verify that acidity performed a protective effect in the communities, upon Cu and Zn addition. A comparative study between Aljustrel mining area and New Brunswick mining area was carried out, both with similar mining and geological conditions, reflected in similar diatom communities in both mines, but in very different geographic and climatic areas.

The Brazilian Cerrado houses a hugely diverse biota and is considered a conservation hotspot. One of the greatest threats to the integrity of this ecosystem is introduced African grasses, which can competitively exclude native grasses and cause changes in the microclimate and other disturbances. The Cerrado is a mosaic vegetation that provides different combinations, both spatially and temporally, of conditions that can become natural stressors to the herbaceous vegetation (water, nutrient and light availability). These mosaics are reflected in differences in relationships among native and invasive species, affecting competition and creating situations (place/season) that are more, or less, susceptible to invasion. The present study aimed to identify the different biological responses of native (Aristida recurvata, Aristida setifolia, Axonopus barbigerus, Echinolaena inflexa, Gymnopogon spicatus, Paspalum gardnerianum, Paspalum stellatum, Schizachyrium microstachyum, Schizachyrium sanguineum) and invasive (Melinis minutiflora and Andropogon gayanus) grasses to variations in natural stressors and to disturbance (fire and clipping), in order to understand changes in ecosystem functioning and competition processes between the grasses, and to understand invasion dynamics in this ecosystem. The presence of invasive species proved to affect the ecosystem functioning by increasing soil feeding activity. These differences were no longer observed in the dry season or when fires were frequent, showing that water availability and fire are more detrimental to soil feeding activity than is the vegetation. Laboratory experiments showed that both drought and flood simulated scenarios damaged both species, although the invasive species performed better under all watering conditions and responded better to fertilization. Underlying mechanisms such as the efficiency of photosynthesis and antioxidant mechanisms helped to explain this behavior. The invasive species grew faster and showed less cellular damage and a healthier photosystem, reflected in higher assimilation rates under stress. These differences between the native and invasive species were reduced with clipping, especially in dry soil with no fertilization, where the native species recovered better in relation to the pre-clipping levels. Flooding was as stressful as drought, but the invasive species can bypass this issue by growing an extensive root system, especially in the better-drained soils. Fire is more detrimental than clipping, with a slower recovery, while post-fire temperatures affect the germination of both invasive and native seeds and may be an important factor influencing the persistence of a diverse biota. This approach will finally contribute to the choice of the appropriate management techniques to preserve the Cerrado’s biodiversity.

Salt marshes are highly productive intertidal habitats that serve as nursery grounds for many commercially and economically important species. Because of their location and physical and biological characteristics, salt marshes are considered to be particularly vulnerable to anthropogenic inputs of oil hydrocarbons. Sediment contamination with oil is especially dangerous for salt marsh vegetation, since low molecular weight aromatic hydrocarbons can affect plants at all stages of development. However, the use of vegetation for bioremediation (phytoremediation), by removal or sequestration of contaminants, has been intensively studied. Phytoremediation is an efficient, inexpensive and environmental friendly approach for the removal of aromatic hydrocarbons, through direct incorporation by the plant and by the intervention of degrading microbial populations in the rhizosphere (microbe-assisted phytoremediation). Rhizosphere microbial communities are enriched in important catabolic genotypes for degradation of oil hydrocarbons (OH) which may have a potential for detoxification of the sediment surrounding the roots. In addition, since rhizosphere bacterial populations may also internalize into plant tissues (endophytes), rhizocompetent AH degrading populations may be important for in planta AH degradation and detoxification. The present study involved field work and microcosms experiments aiming the characterization of relevant plant-microbe interactions in oilimpacted salt marshes and the understanding of the effect of rhizosphere and endosphere bacteria in the role of salt marsh plants as potential phytoremediation agents. In the field approach, molecular tools were used to assess how plant species- and OH pollution affect sediment bacterial composition [bulk sediment and sediment surrounding the roots (rhizosphere) of Halimione portulacoides and Sarcocornia perennis subsp. perennis] in a temperate estuary (Ria de Aveiro, Portugal) chronically exposed to OH pollution. In addition, the 16S rRNA gene sequences retrieved in this study were used to generate in silico metagenomes and to evaluate the distribution of potential bacterial traits in different microhabitats. Moreover, a combination of culture-dependent and -independent approaches was used to investigate the effect of oil hydrocarbons contamination on the structure and function of endophytic bacterial communities of salt marsh plants.Root systems of H. portulacoides and S. perennis subsp. perennis appear to be able to exert a strong influence on bacterial composition and in silico metagenome analysis showed enrichment of genes involved in the process of polycyclic aromatic hydrocarbon (PAH) degradation in the rhizosphere of halophyte plants. The culturable fraction of endophytic degraders was essentially closely related to known OH-degrading Pseudomonas species and endophytic communities revealed sitespecific effects related to the level of OH contamination in the sediment. In order to determine the effects of oil contamination on plant condition and on the responses in terms of structure and function of the bacterial community associated with plant roots (rhizosphere, endosphere), a microcosms approach was set up. The salt marsh plant Halimione portulacoides was inoculated with a previous isolated Pseudomonas sp. endophytic degrader and the 2-methylnaphthalene was used as model PAH contaminant. The results showed that H. portulacoides health and growth were not affected by the contamination with the tested concentration. Moreover, the decrease of 2-methylnaphthalene at the end of experiment, can suggest that H. portulacoides can be considered as a potential plant for future uses in phytoremedition approaches of contaminated salt marsh. The acceleration of hydrocarbon degradation by inoculation of the plants with the hydrocarbon-degrading Pseudomonas sp. could not, however, be demonstrated, although the effects of inoculation on the structure of the endophytic community observed at the end of the experiment indicate that the strain may be an efficient colonizer of H. portulacoides roots. The results obtained in this work suggest that H. portulacoides tolerates moderate concentrations of 2-methylnaphthalene and can be regarded as a promising agent for phytoremedition approaches in salt marshes contaminated with oil hydrocarbons. Plant/microbe interactions may have an important role in the degradation process, as plants support a diverse endophytic bacterial community, enriched in genetic factors (genes and plasmids) for hydrocarbon degradation.

K0.5Na0.5NbO3 (KNN), is the most promising lead free material for substituting lead zirconate titanate (PZT) which is still the market leader used for sensors and actuators. To make KNN a real competitor, it is necessary to understand and to improve its properties. This goal is pursued in the present work via different approaches aiming to study KNN intrinsic properties and then to identify appropriate strategies like doping and texturing for designing better KNN materials for an intended application. Hence, polycrystalline KNN ceramics (undoped, non-stoichiometric; NST and doped), high-quality KNN single crystals and textured KNN based ceramics were successfully synthesized and characterized in this work. Polycrystalline undoped, non-stoichiometric (NST) and Mn doped KNN ceramics were prepared by conventional ceramic processing. Structure, microstructure and electrical properties were measured. It was observed that the window for mono-phasic compositions was very narrow for both NST ceramics and Mn doped ceramics. For NST ceramics the variation of A/B ratio influenced the polarization (P-E) hysteresis loop and better piezoelectric and dielectric responses could be found for small stoichiometry deviations (A/B = 0.97). Regarding Mn doping, as compared to undoped KNN which showed leaky polarization (P-E) hysteresis loops, B-site Mn doped ceramics showed a well saturated, less-leaky hysteresis loop and a significant properties improvement. Impedance spectroscopy was used to assess the role of Mn and a relation between charge transport – defects and ferroelectric response in K0.5Na0.5NbO3 (KNN) and Mn doped KNN ceramics could be established. At room temperature the conduction in KNN which is associated with holes transport is suppressed by Mn doping. Hence Mn addition increases the resistivity of the ceramic, which proved to be very helpful for improving the saturation of the P-E loop. At high temperatures the conduction is dominated by the motion of ionized oxygen vacancies whose concentration increases with Mn doping. Single crystals of potassium sodium niobate (KNN) were grown by a modified high temperature flux method. A boron-modified flux was used to obtain the crystals at a relatively low temperature. XRD, EDS and ICP analysis proved the chemical and crystallographic quality of the crystals. The grown KNN crystals exhibit higher dielectric permittivity (29,100) at the tetragonal-to-cubic phase transition temperature, higher remnant polarization (19.4 μC/cm2) and piezoelectric coefficient (160 pC/N) when compared with the standard KNN ceramics. KNN single crystals domain structure was characterized for the first time by piezoforce response microscopy. It could be observed that <001> - oriented potassium sodium niobate (KNN) single crystals reveal a long range ordered domain pattern of parallel 180° domains with zig-zag 90° domains. From the comparison of KNN Single crystals to ceramics, It is argued that the presence in KNN single crystal (and absence in KNN ceramics) of such a long range order specific domain pattern that is its fingerprint accounts for the improved properties of single crystals. These results have broad implications for the expanded use of KNN materials, by establishing a relation between the domain patterns and the dielectric and ferroelectric response of single crystals and ceramics and by indicating ways of achieving maximised properties in KNN materials. Polarized Raman analysis of ferroelectric potassium sodium niobate (K0.5Na0.5)NbO3 (KNN) single crystals was performed. For the first time, an evidence is provided that supports the assignment of KNN single crystals structure to the monoclinic symmetry at room temperature. Intensities of A′, A″ and mixed A′+A″ phonons have been theoretically calculated and compared with the experimental data in dependence of crystal rotation, which allowed the precise determination of the Raman tensor coefficients for (non-leaking) modes in monoclinic KNN. In relation to the previous literature, this study clarifies that assigning monoclinic phase is more suitable than the orthorhombic one. In addition, this study is the basis for non-destructive assessments of domain distribution by Raman spectroscopy in KNN-based lead-free ferroelectrics with complex structures. Searching a deeper understanding of the electrical behaviour of both KNN single crystal and polycrystalline materials for the sake of designing optimized KNN materials, a comparative study at the level of charge transport and point defects was carried out by impedance spectroscopy. KNN single crystals showed lower conductivity than polycrystals from room temperature up to 200 ºC, but above this temperature polycrystalline KNN displays lower conductivity. The low temperature (T < 200 ºC) behaviour reflects the different processing conditions of both ceramics and single crystals, which account for less defects prone to charge transport in the case of single crystals. As temperature increases (T > 200 ºC) single crystals become more conductive than polycrystalline samples, in which grain boundaries act as barriers to charge transport. For even higher temperatures the conductivity difference between both is increased due to the contribution of ionic conduction in single crystals. Indeed the values of activation energy calculated to the high temperature range (T > 300 ºC) were 1.60 and 0.97 eV, confirming the charge transport due to ionic conduction and ionized oxygen vacancies in single crystals and polycrystalline KNN, respectively. It is suggested that single crystals with low defects content and improved electromechanical properties could be a better choice for room temperature applications, though at high temperatures less conductive ceramics may be the choice, depending on the targeted use. Aiming at engineering the properties of KNN polycrystals towards the performance of single crystals, the preparation and properties study of (001) – oriented (K0.5Na0.5)0.98Li0.02NbO3 (KNNL) ceramics obtained by templated grain growth (TGG) using KNN single crystals as templates was undertaken. The choice of KNN single crystals templates is related with their better properties and to their unique domain structure which were envisaged as a tool for templating better properties in KNN ceramics too. X-ray diffraction analysis revealed for the templated ceramics a monoclinic structure at room temperature and a Lotgering factor (f) of 40% which confirmed texture development. These textured ceramics exhibit a long range ordered domain pattern consisting of 90º and 180º domains, similar to the one observed in the single crystals. Enhanced dielectric (13017 at TC), ferroelectric (2Pr = 42.8 μC/cm2) and piezoelectric (d33 = 280 pC/N) properties are observed for textured KNNL ceramics as compared to the randomly oriented ones. This behaviour is suggested to be due to the long range ordered domain patterns observed in the textured ceramics. The obtained results as compared with the data previously reported on texture KNN based ceramics confirm that superior properties were found due to ordered repeated domain pattern. This study provides an useful approach towards properties improvement of KNN-based piezoelectric ceramics. Overall, the present results bring a significant contribution to the pool of knowledge on the properties of sodium potassium niobate materials: a relation between the domain patterns and di-, ferro-, and piezo-electric response of single crystals and ceramics was demonstrated and ways of engineering maximised properties in KNN materials, for example by texturing were established. This contribution is envisaged to have broad implications for the expanded use of KNN over the alternative lead-based materials.

In recent years, the search for a environmentally friendly products has increased. One of the major challenges has been the demand for biodegradable materials that can replace plastic. If a few decades ago, plastic replaced, for example, the ivory in billiard balls, and in other products, saving the lives of thousands elephants, nowadays a replacement for that plastic is being searched, to prevent the change of the environmental conditions, essential to life in harmonly with the fauna and flora that the human specie has, in recent years, destroyed. Plastic is a petroleum derivate, whose price has been growing exponentially, mainly due to the fact of beind a cheap material and also to enable the production of products that are essential to modern life. Therefore, the petrochemical era is going to come to an end and a new environmentally sustainable era, based on biodegradable materials from renewable sources, will follow. The change to green routes only will be possible with the support of the major companies, and the implementation of drastic governmental law. Poly(lactic acid), PLA, is produced from the lactose present in the corn or sugarcane and has been intensively studied in recent years because if some limitants properties required its extrusion are overcome, it has the potential to replace the traditional polymers. PLA have high brittleness, low toughness and low tensile elongation. In this work, natural antioxidant (alpha-tocopherol) and synthetics antioxidants (BHT ant TBHQ) were added to the PLA with the aim not only to improve their flexibility, but also to create an active packaging to extend the shelf life of the foods and improve the organoleptic properties by preventing food losses. The impact of the addition of antioxidants into the PLA films, in its mechanical, thermal and barrier properties were studied by FTIR, DSC, SEM, AFM, DMA, TGA, QCM and time-lag techniques.

Industrial activities are the major sources of pollution in all environments. Depending on the type of industry, various levels of organic and inorganic pollutants are being continuously discharged into the environment. Although, several kinds of physical, chemical, biological or the combination of methods have been proposed and applied to minimize the impact of industrial effluents, few have proved to be totally effective in terms of removal rates of several contaminants, toxicity reduction or amelioration of physical and chemical properties. Hence, it is imperative to develop new and innovative methodologies for industrial wastewater treatment. In this context nanotechnology arises announcing the offer of new possibilities for the treatment of wastewaters mainly based on the enhanced physical and chemical proprieties of nanomaterials (NMs), which can remarkably increase their adsorption and oxidation potential. Although applications of NMs may bring benefits, their widespread use will also contribute for their introduction into the environment and concerns have been raised about the intentional use of these materials. Further, the same properties that make NMs so appealing can also be responsible for producing ecotoxicological effects. In a first stage, with the objective of selecting NMs for the treatment of organic and inorganic effluents we first assessed the potential toxicity of nanoparticles of nickel oxide (NiO) with two different sizes (100 and 10-20 nm), titanium dioxide (TiO2, < 25 nm) and iron oxide (Fe2O3, ≈ 85x425 nm). The ecotoxicological assessment was performed with a battery of assays using aquatic organisms from different trophic levels. Since TiO2 and Fe2O3 were the NMs that presented lower risks to the aquatic systems, they were selected for the second stage of this work. Thus, the two NMs pre-selected were tested for the treatment of olive mill wastewater (OMW). They were used as catalyst in photodegradation systems (TiO2/UV, Fe2O3/UV, TiO2/H2O2/UV and Fe2O3/H2O2/UV). The treatments with TiO2 or Fe2O3 combined with H2O2 were the most efficient in ameliorating some chemical properties of the effluent. Regarding the toxicity to V. fischeri the highest reduction was recorded for the H2O2/UV system, without NMs. Afterwards a sequential treatment using photocatalytic oxidation with NMs and degradation with white-rot fungi was applied to OMW. This new approach increased the reduction of chemical oxygen demand, phenolic content and ecotoxicity to V. fischeri. However, no reduction in color and aromatic compounds was achieved after 21 days of biological treatment. The photodegradation systems were also applied to treat the kraft pulp mill and mining effluents. For the organic effluent the combination NMs and H2O2 had the best performances in reduction the chemical parameters as well in terms of toxicity reduction. However, for the mine effluent the best (TiO2/UV and Fe2O3/UV) were only able to significantly remove three metals (Zn, Al and Cd). Nonetheless the treatments were able of reducing the toxicity of the effluent. As a final stage, the toxicity of solid wastes formed during wastewater treatment with NMs was assessed with Chironomus riparius larvae, a representative species of the sediment compartment. Certain solid wastes showed the potential to negatively affect C. riparius survival and growth, depending on the type of effluent treated. This work also brings new insights to the use of NMs for the treatment of industrial wastewaters. Although some potential applications have been announced, many evaluations have to be performed before the upscaling of the chemical treatments with NMs.

Potenciais agentes de biocontrolo foram isolados a partir da microbiota epifítica de folhas e frutos, de citrinos e pomóideas, de diferentes pomares, durante diferentes campanhas e de distintas condições de armazenamento. A atividade antagonista de 1465 microrganismos isolados foi testada em ensaios in vivo em pomóideas face a Penicillium expansum (104 esporos/ml) e em citrinos face a P. digitatum (105 esporos/ml) em frutos feridos e inoculados artificialmente. Aproximadamente 7,6% dos isolados reduziu a severidade (diâmetro da podridão) e a incidência (% podres) em mais de 25%, menos de 3% reduziu ambos os parâmetros em mais de 50%, mas apenas 4 microrganismos preencheram os critérios de seleção, redução da incidência e severidade em mais de 75%. Dos 4 e pelos resultados obtidos em ensaios de seleção secundária e de determinação da concentração mínima eficaz, destacaram-se e selecionaram-se 2 microrganismos, uma bactéria isolada de laranjas „Valencia late‟ e identificada como pertencente ao grupo das Enterobactérias, Pantoea agglomerans PCB-1 e uma levedura isolada a partir da superfície de maças 'Bravo de Esmolfe' identificada como Metschnikowia andauensis PBC-2, com o objetivo de usar dois modelos distintos de agentes de biocontrol, uma bactéria e uma levedura. P. agglomerans é um agente de biocontrolo já conhecido. A estirpe PBC-1 isolada neste trabalho mostrou ter elevada eficácia face aos principais agentes patogénicos na póscolheita de citrinos e pomóideas. M. andauensis é uma levedura recentemente descoberta e a estirpe PBC-2 diz respeito à primeira referência desta espécie como agente de biocontrolo, a qual foi recentemente objeto de concessão de Patente de Invenção Nacional nº 105210, como uma nova estirpe desta espécie para uso como agente de biocontrolo das doenças de póscolheita de frutos. A concentração mínima eficaz dos antagonistas revelou estar dentro dos limites para o seu desenvolvimento comercial. M. andauensis PBC-2 quando aplicada à concentração de 5×106 ufc/ml, permitiu uma redução da incidência e da severidade de 62 e 70%, respetivamente, e quando aplicada a 1×107 ufc/ml, uma redução de 90% de incidência e de 95% de severidade. P. agglomerans PBC-1 aplicada a 1 × 108 ufc/ml em maçãs e em citrinos no controlo de P. expansum e P. digitatum, respetivamente, propiciaram uma redução significativa de cerca de 86% de cada um dos agentes patogénicos. O espectro de ação de M. andauensis PBC-2 foi avaliado, verificando-se o controlo efetivo face a Rhizopus stolonifer, P. expansum e Botritys cinerea, em pera 'Rocha' e em diferentes cultivares de maçã e contra P. digitatum e Penicillium italicum em clementinas e laranjas de diferentes cultivares. Durante 4 épocas, a eficácia de M. andauensis PBC-2, foi avaliada e comparada com o fungicida sintético mais usado comercialmente, Imazalil, em ensaios semicomerciais. Os resultados assemelharam-se aos obtidos com o fungicida, a redução da incidência do bolor azul foi de 90% em maças armazenadas durante 3 meses a 1±0.5 ºC, seguido de 7 dias à temperatura ambiente, para simular o tempo de prateleira. Em ensaios do estudo da dinâmica populacional, verificou-se que o agente de biocontrolo M. andauensis PBC-2 tem uma excelente capacidade colonizadora e que consegue crescer e sobreviver nas feridas, mas também na superfície dos frutos, armazenados à temperatura ambiente e em condições de frio. Pelo contrário, M. andauensis PBC- não apresentou capacidade de sobrevivência no suco gástrico simulado, começando a população a diminuir imediatamente após exposição e passadas 48 h não restava população viável. Diferentes meios de cultura usualmente descritos na produção de leveduras foram testados na produção de M. andauensis PBC-2. Aquele que apresentou a maior população viável ao fim de 40 h de incubação foi o meio YPD, entretanto escolhido para estudos posteriores. O pH mais favorável ao crescimento foi de 6,5, não se observando diferenças significativas entre os crescimentos a 25 e 30 ºC. Estudou-se ainda o efeito da concentração das duas fontes de azoto do meio YPD e elegeu-se a combinação de 10 g/l de extrato de levedura e 20 g/l de peptona. Nos estudos de produção de biomassa dos dois potenciais agentes de biocontrolo, analisou-se o efeito da sacarose, frutose e glucose, como fontes de carbono e otimizou-se a concentração destes açúcares no crescimento em Erlenmeyer. Após 20 h de incubação a população viável de P. agglomerans PBC-1 atingiu 3.9×109, 1.5×109, 3.9×109 ufc/ml, respetivamente. No caso de M. andauensis PBC-2, foi obtida a população de 1.2×108, 5.3×108 e 1,3×108 ufc/ml, com glucose, sacarose e frutose, após 40 h. Os resultados permitem concluir que os dois agentes têm capacidade de metabolizar os açúcares testados, contudo e atendendo à produtividade de biomassa, rendimento, disponibilidade e custo, optou-se por usar a sacarose como fonte de carbono nos restantes ensaios, nomeadamente na transição de Erlenmeyer para reator biológico. Na produção de P. agglomerans PBC-1 escolheu-se o meio SAC (5 g/l sacarose e 5 g/l extrato de levedura). O meio YPS (12.5g/l sacarose, 10 g/l extrato de levedura, 20 g/l peptona) foi usado no aumento de escala de M. andauensis PBC-2. A otimização da produção em reator biológico de P. agglomerans PCB-1 foi realizada submetendo o microrganismo a diferentes condições hidrodinâmicas, testando-se arejamentos, dois tipos de turbina (hélice; rusthon) e dispersores (poroso, em L). Foram igualmente estudados, o efeito da concentração inicial do inoculo e a adição programada da fonte de carbono. Embora tenham sido testadas diferentes variações, o perfil dos diferentes parâmetros analisados foi idêntico, a população máxima viável foi de 3-5×109 ufc/ml. A diferença mais notória foi observada na fermentação com concentração inicial de 107 cfu/ml, que permitiu encurtar em cinco horas a fase lag, o que pode significar uma redução de tempo de fermentação, e consequentemente uma redução de custos. Visando a produção de biomassa a baixo custo, fator importante na implementação de um sistema de controlo biológico, estudou-se a possibilidade de utilizar subprodutos e resíduos da indústria alimentar no crescimento dos agentes de biocontrolo. Subprodutos da indústria de alfarroba e subprodutos e resíduos da indústria de sumos de citrinos foram utilizados na produção de P. agglomerans PBC-1 e de M. andauensis PBC-2, respetivamente. Desta forma, para além de uma redução dos custos de produção, pretendeu-se a valorização de um subproduto (no caso de alfarroba e do bagaço de citrinos) e mitigar os efeitos nefastos de um resíduo (licor), com elevada carga poluente, que gera graves problemas ambientais e que pode ditar o encerramento desta unidade industrial, com efeitos devastadores para a economia local. Realizaram-se extrações de subprodutos da indústria de alfarroba, a diferentes razões sólido/líquido, tempos e temperaturas, de forma a maximizar a extração de açúcares. A potencialidade de utilizar o extrato de açúcares obtido, na produção de P. agglomerans PBC-1 foi avaliada, em ensaios de crescimento em Erlenmeyer, com consequente transição para reator mecanicamente agitado. Os perfis de crescimento da cultura crescida com subprodutos da indústria de alfarroba assemelharam-se aos observados com sacarose como fonte de carbono. A biomassa viável produzida com subprodutos da indústria de alfarroba, foi de 4- 7×109 ufc/ml, o que permite concluir que é uma alternativa viável à produção deste microrganismo. A produção de M. andauensis PBC-2 com subprodutos da indústria de sumos de citrinos, foi estudada em Erlenmeyer tendo como objetivo conhecer os perfis de crescimento do microrganismo, bem como, estudar a melhor combinação desta fonte de carbono e sua concentração. O bagaço de citrinos e um resíduo líquido, que se denominou de licor, foram testados com resultados comparáveis à produção obtida com meio usado como standard, (YPS), sem comprometer a atividade antagonista do agente de biocontrolo. Posteriormente, foi realizada a produção em reator mecanicamente agitado, escolhendo-se para tal o meio YL (10 g/l extrato de levedura e licor à concentração de açúcares de 12.5 g/l). Os parâmetros de crescimento da cultura foram semelhantes aos obtidos com a fonte de carbono comercial. Após aproximadamente 40 h de incubação, a população viável de M. andauensis PCB-2 atingiu 3.1×108 ufc/ml. A produtividade de biomassa e rendimento foi de 0.435 g/l.h e 1.502 g/g, respetivamente, comparável a produtividade de biomassa (0.432 g/l.h) e rendimento (1.4416 g/g) observado no meio YPS. Os resultados obtidos, são uma base sólida para o aumento de escala a um nível laboratorial e semi-industrial, permitiram concluir que é exequível produzir M. andauensis a baixo custo e representam uma possível alternativa para um resíduo. Em estudos dos possíveis modos de ação, de M. andauensis PBC-2, conclui-se que, este agente de biocontrolo, não tem como modos de ação a produção de antibióticos ou de voláteis, uma vez que, não se verificou inibição do crescimento dos agentes patogénicos. A competição por ferro e a produção de enzimas líticas por M. andauensis PBC-2 foi estudada em meios com diferentes concentrações de ferro e em um meio de cultura, com paredes celulares de fungos, como única fonte de carbono. Os resultados obtidos neste estudo sugerem que a produção e secreção de enzimas líticas não é o principal ou o mais importante modo de ação do agente de controlo biológico PBC-2, uma vez que a produção de quitinase observada ao 5 e 7º dia de incubação foi muito baixa, e não foi observada a produção de β-1,3- glucanases e proteases.
