Pseudomonas syringae is a model bacterial pathogen that penetrates the leaf to reach the plant apoplast, where it replicates causing disease. In order to do that, the pathogen must interfere and suppress a two-tiered plant defense response: PTI (PAMP-Triggered Immunity, or basal resistance) and ETI (Effector-Triggered Immunity). P. syringae uses a type III secretion system to directly deliver effector proteins inside the plant cell cytosol, many of which are known to suppress PTI, some of which are known to trigger ETI, and a handful of which are known to suppress ETI. Bacterial infection can also trigger a systemic plant defense response that protects the plant against additional pathogen attacks known as SAR (Systemic Acquired Resistance). We are particularly interested in the molecular and cellular mechanisms involved in effector-mediated defense evasion by P. syringae, in particular those involved in the suppression of ETI and SAR, and/or mediation of hormone signaling. Here we present data describing effector-mediated interference with plant immunity, by means of acetylation of a key positive regulator of local and systemic responses. Our work identifies a novel plant target for effector function, and characterizes its function. This work illustrates how analyzing the means by which a given effector interferes with its target can provide novel information regarding eukaryotic molecular mechanisms.

Both the end of the twentieth century and the beginning of the twenty-first century have been characterized as a period of major political, economic, social and cultural transformations. Two of the major consequences of the political-economical crisis of the end of last century are the restructuring of capitalist production, and the consolidation of neoliberalism as a worldwide phenomenon. This new world political-economical scenario has influenced, in a dialectic way, the contemporary urban development. In that sense, "new" spatial processes and new paradigms in both urban management and urban planning have gained shape. In this context of urban transformations, the central areas of western cities, also known as historic centers, are being increasingly (re)valued. Since the Second World War, the historic centers urban areas which have great infrastructure and symbolic relevance had been undergoing a process of evasion of population and activities, undeniably linked to the neglect of government authorities. However, in recent decades, the question of historic centers rehabilitation has acquired a growing interest, academically and in political agendas. The object of this dissertation is to focus on how the government of each Brazil and Portugal has dealt with the issue of historic center rehabilitation through programs of urban rehabilitation

Several teams of researchers at multiple universities are currently measuring annual and seasonal fluxes of carbon dioxide and other greenhouses gases (nitrous oxide and methane) in riparian wetlands and upland forests in the Tenderfoot Creek Experimental Forest (TCEF), a subalpine watershed in the Little Belt Mountains, Montana. In the current thesis, the author characterized the geochemistry and stable carbon isotope composition of shallow groundwater, soil water, and stream water in upper Stringer Creek, near sites that are being investigated for gas chemistry and microbial studies. It was hypothesized that if methanogenesis were a dominant process in the riparian wetlands of upper Stringer Creek, then this should impart a characteristic signal in the measured stable isotopic composition of dissolved inorganic carbon in shallow groundwater. For the most part, the major solute composition of shallow groundwater in upper Stringer Creek was similar to that of the stream. However, several wells completed in wetland soil had highly elevated concentrations of Fe2+ and Mn2+ which were absent in the well-oxygenated surface water. Use of sediment pore-water samplers (peepers) demonstrated a rapid increase in Fe2+ and Mn2+ with depth, most feasibly explained by microbial reduction of Fe- and Mn-oxide minerals. In general, the pH of shallow groundwater was lower than that of the stream. Since concentrations of CO2 in the groundwater samples were consistently greater than atmospheric pCO2, exchange of CO2 gas across the stream/air interface occurred in one direction, from stream to air. Evasion of CO2 partly explains the higher pH values in the stream. Microbial processes involving breakdown of organic carbon, including aerobic respiration, anaerobic respiration, and methanogenesis, explain the occurrence of excess CO2 in the groundwater. In general, the isotopic composition of total dissolved inorganic carbon (DIC) decreased with increasing DIC concentration, consistent with aerobic and/or anaerobic respiration being the dominant metabolic process in shallow groundwater. However, a minority of wells contained high DIC concentrations that were anomalously heavy in u13C, and these same wells had elevated concentrations of dissolved methane. It is concluded that the wells with isotopically-heavier DIC have likely been influenced by acetoclastic methanogenesis. Results from shallow groundwater wells and one of the peeper samplers suggest a possible link between methanogenesis and bacterial iron reduction.

Methicillin-resistant Staphylococcus aureus (MRSA) Sequence Type (ST)1, Clonal Complex(CC)1, SCCmec V is one of the major Livestock-Associated (LA-) lineages in pig farming industry in Italy and is associated with pigs in other European countries. Recently, it has been increasingly detected in Italian dairy cattle herds. The aim of this study was to analyse the differences between ST1 MRSA and methicillin-susceptible S. aureus (MSSA) from cattle and pig herds in Italy and Europe and human isolates. Sixty-tree animal isolates from different holdings and 20 human isolates were characterized by pulsed-field gel electrophoresis (PFGE), spa-typing, SCCmec typing, and by micro-array analysis for several virulence, antimicrobial resistance, and strain/host-specific marker genes. Three major PFGE clusters were detected. The bovine isolates shared a high (≥90% to 100%) similarity with human isolates and carried the same SCCmec type IVa. They often showed genetic features typical of human adaptation or present in human-associated CC1: Immune evasion cluster (IEC) genes sak and scn, or sea; sat and aphA3-mediated aminoglycoside resistance. Contrary, typical markers of porcine origin in Italy and Spain, like erm(A) mediated macrolide-lincosamide-streptograminB, and of vga(A)-mediated pleuromutilin resistance were always absent in human and bovine isolates. Most of ST(CC)1 MRSA from dairy cattle were multidrug-resistant and contained virulence and immunomodulatory genes associated with full capability of colonizing humans. As such, these strains may represent a greater human hazard than the porcine strains. The zoonotic capacity of CC1 LA-MRSA from livestock must be taken seriously and measures should be implemented at farm-level to prevent spill-over.

Many vertebrates are known to show behavioural lateralization, whereby they differentially use one side of their body or either of their bilateral organs or limbs. Behavioural lateralization often manifests in a turning bias in fishes, with some individuals showing a left bias and others a right bias. Such biases could be the source of considerable conflict in fish schools given that there may be considerable social pressure to conform to the group to maintain effective group evasion. Here, we show that predation pressure is a major determinant of the degree of lateralization, both in a relative and absolute sense, in yellow-and-blueback fusiliers (Caesio teres), a schooling fish common on coral reefs. Wild-caught fish showed a bias for right turning. When predation pressure was experimentally elevated or relaxed, the strength of lateralization changed. Higher predation pressure resulted in an increase in the strength of lateralization. Individuals that exhibited the same turning bias as the majority of individuals in their group had improved escape performance compared with individuals that were at odds with the group. Moreover, individuals that were right-biased had improved escape performance, compared with left-biased ones. Plasticity in lateralization might be an important evolutionary consequence of the way gregarious species respond to predators owing to the probable costs associated with this behaviour.

The expression of a gene from transcription of the DNA into pre-messenger RNA (pre-mRNA) over translation of messenger RNA (mRNA) into protein is constantly monitored for errors. This quality control is necessary to guarantee successful gene expression. One quality control mechanism important to this thesis is called nonsense-mediated mRNA decay (NMD). NMD is a cellular process that eliminates mRNA transcripts harboring premature translation termination codons (PTCs). Furthermore, NMD is known to regulate certain transcripts with long 3′ UTRs. However, some mRNA transcripts are known to evade NMD. The mechanism of NMD activation has been subjected to many studies whereas NMD evasion or suppression still remains rather elusive. It has previously been shown that the cytoplasmic poly(A)-binding protein (PABPC1) is able to suppress NMD of certain transcripts. In this study I show that PABPC1 is able to suppress NMD of a long 3′ UTR-carrying reporter when tethered immediately downstream of the termination codon. I further am able to show the importance of the interaction between PABPC1 and eIF4G for NMD suppression, whereas the interaction between PABPC1 and eRF3a seems dispensable. These results indicate an involvement of efficient translation termination and potentially ribosome recycling in NMD suppression. I am able to show that if PABPC1 is too far removed from the terminating ribosome NMD is activated. After showing the importance of PABPC1 recruitment directly downstream of a terminating ribosome in NMD suppression, I am further able to demonstrate several different methods by which PABPC1 can be recruited. Fold-back of the poly(A)-tail mediated by two interacting proteins on opposite ends of a 3′ UTR manages to bring PABPC1 bound to the poly(A)-tail into close proximity of the terminating ribosome and therefore suppress NMD. Furthermore, small PAM2 peptides that are known to interact with the MLLE domain of PABPC1 are able to strongly suppress NMD initiated by either a long 3′ UTR or an EJC. I am also able to show the NMD antagonizing power of recruited PABPC1 for the known endogenous NMD target β-globin PTC39, which is responsible for the disease β-thalassemia. This shows the potential medical implications and application of suppressing NMD by recruiting PABPC1 into close proximity of a terminating ribosome.

Resumo: a fraude fiscal é um dos crimes que pode dar origem a um outro crime, o crime de branqueamento de capitais e/ou lavagem de dinheiro, como é mais conhecido por exemplo no Brasil. O branqueamento de capitais é apenas um dos exemplos do branqueamento de vantagens. Toda esta actividade pode ser incluída, em sentido amplo, na chamada corrupção internacional. Quanto mais internacionais forem estas actividades – fraude fiscal e branqueamento de capitais -, mais eficazes serão os seus proveitos. A fraude fiscal é um crime que pode atingir montantes mais elevados no caso de poder ser praticado em cenários internacionais. O branqueamento de capitais é também um crime que pode atingir montantes mais elevados no caso de poder ser praticado em cenários internacionais. Assim, a corrupção internacional em sentido amplo pode ser apelidada como mercado ideal para a criminalidade. Estamos também a falar de multinacionais do crime. § Abstract: tax fraud is a crime that can lead to another crime, the crime of money laundering. Money laundering is just one example of the whitening benefits. All this activity can be included in a broad sense, the international corruption. The more international are these activities - tax evasion and money laundering - the more effective will be the income. Tax fraud is a crime that can achieve higher amounts if it can be practiced in international scenarios. Money laundering is also a crime that can achieve higher amounts if it can be practiced in international scenarios. Thus, the international corruption in the broad sense can be dubbed as ideal market for the crime. We are talking also about transnational crime.

El contrabando técnico representa un problema para los hacedores de política económica pues tiene efectos perversos en temas fundamentales como la hacienda pública, la competencia de mercado y la informalidad. Sin embargo, a pesar de ser un problema tan importante son pocos los esfuerzos empíricos que se han hecho para estudiar los incentivos que están detrás de esta práctica ilegal, tanto a nivel global como para el caso colombiano. En este trabajo se desarrolla un modelo teórico a partir del cual se estudian los incentivos para la existencia del contrabando, y sus conclusiones se contrastan con una aplicación empírica en la que se utilizan datos de importaciones (reporte de origen y destino) de 24 sectores económicos (583 productos) provenientes de 84 países entre 1998 y 2013. Con estos datos se estima un modelo de panel de datos en el que se encuentra que hay una relación positiva entre la corrupción y el contrabando y también entre los aranceles y el contrabando técnico, indicando que se presenta una mayor subfacturación en productos que tienen aranceles altos y provienen de países más corruptos.

Portugal is characterized by a significant asymmetry in the population distribution/density and economic activity as well as in social and cultural dynamics. This means very diverse landscapes, differences in regional development, sustainability and quality of life, mainly between urban and rural areas. A consequence coherent with the contemporary dynamics: urbanization of many rural areas that loose their productive-agricultural identity and, simultaneously, the reintegration in urban areas of spaces and activities with more rural characteristics. In this process of increasing complexity of organization of the landscape is essential to restore the continuum naturale (between urban and rural areas) allowing closer links to both ways of life. A strategy supported in the landscape, which plays important functions for public interest, in the cultural, social, ecological and environmental fields. At the same time, constitutes an important resource for economic activity, as underlined in the European Landscape Convention. Based on this assumption, and using a multi-method approach, the study aims to analyse a) the links between urban and rural areas in Portugal and b) the reasons why these territories are chosen by individuals as places of work and mobility, residence or evasion, culture and leisure, tranquillity or excitement – meaning overall well-being. Primary information was obtained by a questionnaire survey applied to a convenience sample of the Portuguese population. Secondary data and information will be collected on the official Portuguese Statistics (INE and PORDATA). Understanding the urban-rural links is essential to support policy measures, take advantage from the global changes and challenge many of the existing myths.
