The use of capillary electrophoresis (CE) has been restricted to applications having high sample concentrations because of its low sensitivity caused by small injection volumes and, when ultraviolet (UV) detection is used, the short optical path length. Sensitivity in CE can be improved by using more sensitive detection systems, or by preconcentration techniques which are based on chromatographic and/or electrophoretic principles. One of the promising strategies to improve sensitivity is solid phase extraction (SPE). Solid Phase Extraction utilizes high sample volumes and a variety of complex matrixes to facilitate trace detection. To increase the specificity of the SPE a selective solid phase must be chosen. Immunosorbents, which are a combination of an antibody and a solid support, have proven to be an excellent option because of high selectivity of the antibody. This thesis is an exploratory study of the application of immunosorbent-SPE combined with CE for trace concentration of benzodiazepines. This research describes the immobilization and performance evaluation of an immunosorbent prepared by immobilizing a benzodiazepine-specific antibody on aminopropyl silica. The binding capacity of the immunosorbent, measured as µg of benzodiazepine/ gram of immunosorbent, was 39 ± 10. The long term stability of the prepared immunosorbent has been improved by capping the remaining aminopropyl groups by reaction with acetic anhydride. The capped immunosorbent retained its binding capacity after several uses.

Sampling and preconcentration techniques play a critical role in headspace analysis in analytical chemistry. My dissertation presents a novel sampling design, capillary microextraction of volatiles (CMV), that improves the preconcentration of volatiles and semivolatiles in a headspace with high throughput, near quantitative analysis, high recovery and unambiguous identification of compounds when coupled to mass spectrometry. The CMV devices use sol-gel polydimethylsiloxane (PDMS) coated microglass fibers as the sampling/preconcentration sorbent when these fibers are stacked into open-ended capillary tubes. The design allows for dynamic headspace sampling by connecting the device to a hand-held vacuum pump. The inexpensive device can be fitted into a thermal desorption probe for thermal desorption of the extracted volatile compounds into a gas chromatography-mass spectrometer (GC-MS). The performance of the CMV devices was compared with two other existing preconcentration techniques, solid phase microextraction (SPME) and planar solid phase microextraction (PSPME). Compared to SPME fibers, the CMV devices have an improved surface area and phase volume of 5000 times and 80 times, respectively. One (1) minute dynamic CMV air sampling resulted in similar performance as a 30 min static extraction using a SPME fiber. The PSPME devices have been fashioned to easily interface with ion mobility spectrometers (IMS) for explosives or drugs detection. The CMV devices are shown to offer dynamic sampling and can now be coupled to COTS GC-MS instruments. Several compound classes representing explosives have been analyzed with minimum breakthrough even after a 60 min. sampling time. The extracted volatile compounds were retained in the CMV devices when preserved in aluminum foils after sampling. Finally, the CMV sampling device were used for several different headspace profiling applications which involved sampling a shipping facility, six illicit drugs, seven military explosives and eighteen different bacteria strains. Successful detection of the target analytes at ng levels of the target signature volatile compounds in these applications suggests that the CMV devices can provide high throughput qualitative and quantitative analysis with high recovery and unambiguous identification of analytes.

The aim of this study was to compare the performance of the following techniques on the isolation of volatiles of importance for the aroma/flavor of fresh cashew apple juice: dynamic headspace analysis using PorapakQ(®) as trap, solvent extraction with and without further concentration of the isolate, and solid-phase microextraction (fiber DVB/CAR/PDMS). A total of 181 compounds were identified, from which 44 were esters, 20 terpenes, 19 alcohols, 17 hydrocarbons, 15 ketones, 14 aldehydes, among others. Sensory evaluation of the gas chromatography effluents revealed esters (n = 24) and terpenes (n = 10) as the most important aroma compounds. The four techniques were efficient in isolating esters, a chemical class of high impact in the cashew aroma/flavor. However, the dynamic headspace methodology produced an isolate in which the analytes were in greater concentration, which facilitates their identification (gas chromatography-mass spectrometry) and sensory evaluation in the chromatographic effluents. Solvent extraction (dichloromethane) without further concentration of the isolate was the most efficient methodology for the isolation of terpenes. Because these two techniques also isolated in greater concentration the volatiles from other chemical classes important to the cashew aroma, such as aldehydes and alcohols, they were considered the most advantageous for the study of cashew aroma/flavor.

In this work, we discuss the use of multi-way principal component analysis combined with comprehensive two-dimensional gas chromatography to study the volatile metabolites of the saprophytic fungus Memnoniella sp. isolated in vivo by headspace solid-phase microextraction. This fungus has been identified as having the ability to induce plant resistance against pathogens, possibly through its volatile metabolites. Adequate culture media was inoculated, and its headspace was then sampled with a solid-phase microextraction fiber and chromatographed every 24 h over seven days. The raw chromatogram processing using multi-way principal component analysis allowed the determination of the inoculation period, during which the concentration of volatile metabolites was maximized, as well as the discrimination of the appropriate peaks from the complex culture media background. Several volatile metabolites not previously described in the literature on biocontrol fungi were observed, as well as sesquiterpenes and aliphatic alcohols. These results stress that, due to the complexity of multidimensional chromatographic data, multivariate tools might be mandatory even for apparently trivial tasks, such as the determination of the temporal profile of metabolite production and extinction. However, when compared with conventional gas chromatography, the complex data processing yields a considerable improvement in the information obtained from the samples. In this work, the volatile chromatographic profiles of roasted Arabica coffees, previously analyzed for their sensorial attributes, were explored by principal component analysis. The volatile extraction technique used was the solid phase microextraction. The correlation optimized warping algorithm was used to align the gas chromatographic profiles. Fifty four compounds were found to be related to the sensorial attributes investigated. The volatiles pyrrole, 1-methyl-pyrrole, cyclopentanone, dihydro-2-methyl-3-furanone, furfural, 2-ethyl-5-methyl-pyrazine, 2-etenyl-n-methyl-pyrazine, 5-methyl-2-propionyl-furan compounds were important for the differentiation of coffee beverage according to the flavour, cleanliness and overall quality. Two figures of merit, sensitivity and specificity (or selectivity), were used to interpret the sensory attributes studied.

In order to protect food from pathogenic microorganisms as well as increase its shelf-life, while keeping sensorial properties (e.g., odor and taste), which are important properties required by spice buyers, it is necessary to analyze volatile formation from irradiation of medicinal and food herbs. Possible changes in the odor of these herbs are evaluated by characterizing different radiation doses and effects on sensorial properties, in order to allow better application of the irradiation technology. The aim of the present study was to analyze volatile formation on cinnamon (Laurus cinnamomum) samples after gamma irradiation. These samples were irradiated into plastic packages using a (60)Co facility. Radiation doses applied were 0, 5, 10, 15, 20 and 25 kGy. For the analysis of the samples, solid-phase microextraction (SPME) was applied, while for the analysis of volatile compounds, CG/MS. Spice irradiation showed the highest decrease in volatile compounds. For L. cinnamomum, the irradiation decreased volatile compounds by nearly 56% and 89.5%, respectively, comparing to volatile from a sample which had not been previously irradiated. A rapid and simple method was optimized for determination of Delta(9)-tetrahydrocannabinol (Delta(9)-THC), cannabidiol (CBD), and cannabinol (CBN) contents in cannabis products by gas chromatography with flame-ionization detection (GC-FID), using diazepam as internal standard. All parameters of validation of the method such as linearity, intraassay precision, and limits of detection and quantification of the analytes were satisfactory. Using the described method, cannabinoid contents of 55 cannabis product samples seized in Sao Paulo City, Brazil, in 2006 and 2007 were measured. Delta(9)-THC content in marijuana and hashish samples varied between 0.08% and 5.5%, with an average of 2.5%. The phenotypic ratio showed that the products were able to be designated as ""drug type.""

A sensitive and reproducible stir bar-sorptive extraction and high-performance liquid chromatography-UV detection (SBSE/HPLC-UV) method for therapeutic drug monitoring of carbamazepine, carbamazepine-10,11-epoxide, phenytoin and phenobarbital in plasma samples is described and compared with a liquid:liquid extraction (LLE/HPLC-UV) method. Important factors in the optimization of SBSE efficiency such as pH, extraction time and desorption conditions (solvents, mode magnetic stir, mode ultrasonic stir, time and number of steps) assured recoveries ranging from 72 to 86%, except for phenytoin (62%). Separation was obtained using a reverse phase C-18 column with UV detection (210 nm). The mobile phase consisted of water: acetonitrile (78:22, v/v). The SBSE/HPLC-UV method was linear over a working range of 0.08-40.0 mu g mL(-1) for carbamazepine, carbamazepine-10,11-epoxide and phenobarbital and 0.125-40.0 mu g mL(-1) for phenytoin, The intra-assay and inter-assay precision and accuracy were studied at three concentrations (1.0, 4.0 and 20.0 mu g mL(-1)). The intra-assay coefficients of variation (CVs) for all compounds were less than 8.8% and all inter-CVs were less than 10%. Limits of quantification were 0.08 mu g mL(-1) for carbamazepine, carbamazepine-10,11-epoxide and phenobarbital and 0.125 mu g mL(-1) for phenytoin. No interference of the drugs normally associated with antiepileptic drugs was observed. Based on figures of merit results, the SBSE/HPLC-UV proved adequate for antiepileptic drugs analyses from therapeutic levels. A sensitive and precise stir bar sorptive extraction (SBSE) combined with LC (SBSE/LC) analysis is described for simultaneous determination of methyl, ethyl, propyl, and butyl parabens in commercial cosmetic products in agreement with the European Union Cosmetics Directive 76/768/EEC. Important factors in the optimization of SB SE efficiency are discussed, such as time and temperature of extraction, pH, and ionic strength of the sample, matrix effects, and liquid desorption conditions by different modes (magnetic stirring, ultrasonic). The LOQs of the SBSE/LC method ranged from 30 to 200 ng/mg, with linear response over a dynamic range, from the LOQ to 2.5 mu g/mg, with a coefficient of determination higher than 0.993. The interday precision of the SBSE/LC method presented a coefficient of variation lower than 5%. The effectiveness of the proposed method was proven for analysis of commercial cosmetic products such as body creams, antiperspirant creams, and sunscreens.

Background: We report the validation of a method for the determination of acetaldehyde, acetone, methanol, and ethanol in biological fluids using manual headspace sample introduction and an acetonitrile internal standard. Method: This method uses a capillary column (I = 30 m, I.D. = 0.25 mm, dF = 0.25 mu m) installed in a gas chromatography-flame ionization detector (GC-FID) apparatus with a run time of 7.5 minutes. Results: Analysis of the retention times and the resolution of the analyte peaks demonstrated excellent separation without widening of the peaks. Precision and accuracy were good (interassay precision < 15% and recovery between 85% and 115%) in both blood and urine. Conclusion: The method was linear (r > 0.09) over the analytical measurement range (AMR) of each analyte.

Da crescente necessidade de alimentos e da necessidade de travar a destruição de culturas por animais e insectos foram sintetizados os pesticidas. Por entre uma vasta gama de pesticidas alguns são desreguladores endócrinos o que se traduz num perigo para a saúde humana pois pode despoletar alterações nos seres vivos mesmo em concentrações muito baixas. Devido a diversos factores nomeadamente a lixiviação, ventos e outros agentes ambientais assim como a presença de terrenos agrícolas junto a diversos rios, estes encontram-se contaminados com pesticidas desreguladores endócrinos. O objectivo deste trabalho foi avaliar quais os pesticidas desreguladores endócrinos presentes nas águas de rios da região Norte. Neste trabalho utilizou-se a técnica de microextracção em fase sólida recorrendo a uma fibra de PDMS (100 μm). Para tal as condições de optimização foram testadas nomeadamente a quantidade de NaCl, quantidade de metanol, temperatura do injector e tempo de exposição. Os parâmetros obtidos óptimos foram 0 % de NaCl, tempo de exposição de 45 min, temperatura do injector de 260 ºC e 2,5 % de metanol. Conseguiu-se a separação de todos os pesticidas desreguladores endócrinos com a seguinte programação temperaturas: inicio a 60ºC por um minuto seguido de um aumento de 20 °C/min até aos 200 °C onde permanece por um minuto e de seguida um aumento de 5 °C/min até aos 245 °C onde permanece por 40 min. Fizeram-se curvas de calibração entre 0,01 μg/L e 10 μg/L. Constatou-se no entanto uma falta de reprodutibilidade entre as injecções utilizando esta técnica. Os rios analisados foram o Rio Douro, Rio Tâmega, Ria de Aveiro, Rio Lima, Rio Minho, Rio Sousa, Rio Águeda, Rio Cávado e Rio Leça. No Rio Tâmega foram encontrados os seguintes pesticidas: diazinão, α-HCH, β-HCH, δ-HCH, lindano, HCB, simazina/ atrazina, vinclozolina, alacloro, 2,4-D, malatião, aldrina, bifentrina, metoxicloro e fenvalerato. No rio Douro estão presentes HCB, simazina/ atrazina, vinclozolina, 2,4-D, malatião, aldrina, fenvalerato e deltametrina. No rio Lima encontra-se diazinão, α-HCH, δ-HCH, 2,4-D, HCB, vinclozolina, lindano, simazina/atrazina, alacloro, malatião, aldrina, fenvalerato e deltametrina. No rio Sousa os pesticidas encontrados foram: diazinão, HCB, aldrina, α-HCH, β-HCH, δ-HCH, lindano, simazina/ atrazina, 2,4-D, cipermetrina, alacloro, fenvalerato e malatião. No rio Cávado estão presentes o diazinão, α-HCH, β-HCH, δ-HCH, lindano, HCB, 2,4-D, malatião, metoxicloro, cipermetrina e o fenvalerato. Na ria de Aveiro encontrou-se o diazinão, α-HCH, β-HCH, δ-HCH, lindano, HCB, simazina/atrazina, 2,4-D, Malatião e aldrina. No rio Águeda estão presentes o diazinão, HCB, 2,4-D, aldrina e malatião. E por último no rio Leça esta presente o diazinão, 2,4-D, alacloro, malatião, aldrina, cipermetrina e fenvalerato. A importância deste trabalho reside na demonstração da presença destes pesticidas, desreguladores endócrinos nas águas superficiais da região Norte.

A procura de piscinas para a prática de atividades desportivas, recreativas e/ou terapêuticas tem sofrido um aumento gradual ao longo do tempo. No entanto, nas piscinas existem vários perigos associados à sua utilização. Relativamente aos perigos químicos, a utilização de desinfetantes à base de cloro, bromo ou compostos derivados vai, por um lado, inativar microrganismos patogénicos mas, por outro, dar origem a subprodutos ao reagir com compostos orgânicos presentes na água. Os trihalometanos são um exemplo de subprodutos que se podem formar e, entre os compostos principais, estão o clorofórmio (TCM), bromodiclorometano (BDCM), clorodibromometano (CDBM) e bromofórmio (TBM). Este trabalho teve como objetivo o desenvolvimento de uma metodologia analítica para a determinação de trihalometanos em água e ar de piscinas e a sua aplicação a um conjunto de amostras. Para a análise dos compostos, foi utilizada a microextração em fase sólida no espaço de cabeça (HS-SPME) com posterior quantificação dos compostos por cromatografia gasosa com detetor de captura eletrónica (GC-ECD). Foi realizada uma otimização das condições de extração dos compostos em estudo em amostras de água, através da realização de dois planeamentos experimentais. As condições ótimas são assim obtidas para uma temperatura de extração de 45ºC, um tempo de extração de 25 min e um tempo de dessorção de 5 min. Foram analisadas amostras de águas de piscina cedidas pelo Centro de Estudos de Águas, sendo avaliada a aplicação da técnica HS-SPME e o efeito de matriz. O modo como se manuseiam as soluções que contêm os compostos em estudo influencia os resultados devido ao facto destes serem bastante voláteis. Concluiu-se também que existe efeito de matriz, logo a concentração das amostras deverá ser determinada através do método de adição de padrão. A caraterização da água de piscinas interiores permitiu conhecer a concentração de trihalometanos (THMs). Foram obtidas concentrações de TCM entre 4,5 e 406,5 μg/L sendo que apenas 4 das 27 amostras analisadas ultrapassam o valor limite imposto pelo Decreto-Lei nº306/2007 (100 μg/L) no que diz respeito a águas de consumo humano e que é normalmente utilizado como valor indicativo para a qualidade das águas de piscina. Relativamente à concentração obtida no ar de uma piscina interior, foi detetada uma concentração média de 224 μg/m3 de TCM, valor muito abaixo dos 10000 μg/m3 impostos pelo Decreto-lei nº24/2012, como valor limite para exposição profissional a agentes químicos.

The present paper describes a procedure to isolate volatiles from rock-rose (Cistus ladanifer L.) using simultaneous distillation–extraction (SDE). High-value volatile compounds (HVVC) were selected and the influence of the extraction conditions investigated. The effect of the solvent nature and extraction time on SDE efficiency was studied. The best performance was achieved with pentane in 1 h operation. The extraction efficiencies ranged from 65% to 85% and the repeatability varied between 4% and 6% (as a CV%). The C. ladanifer SDE extracts were analysed by headspace solid phase microextraction (HS-SPME) followed by gas chromatography with flame ionization detection (GC-FID). The HS-SPME sampling conditions such as fiber coating, temperature, ionic strength and exposure time were optimized. The best results were achieved with an 85 µm polyacrylate fiber for a 60 min headspace extraction at 40ºC with 20% (w/v) of NaCl. For optimized conditions the recovery was in average higher than 90% for all compounds and the intermediate precision ranged from 4 to 9% (as CV %). The volatiles α-pinene (22.2 mg g−1 of extract), 2,2,6-trimethylcyclohexanone (6.1 mg g−1 of extract), borneol (3.0 mg g−1 of extract) and bornyl acetate (3.9 mg g−1 of extract) were identified in the SDE extracts obtained from the fresh plant material.
