Vitis vinifera L., the most widely cultivated fruit crop in the world, was the starting point for the development of this PhD thesis. This subject was exploited following on two actual trends: i) the development of rapid, simple, and high sensitive methodologies with minimal sample handling; and ii) the valuation of natural products as a source of compounds with potential health benefits. The target group of compounds under study were the volatile terpenoids (mono and sesquiterpenoids) and C13 norisoprenoids, since they may present biological impact, either from the sensorial point of view, as regards to the wine aroma, or by the beneficial properties for the human health. Two novel methodologies for quantification of C13 norisoprenoids in wines were developed. The first methodology, a rapid method, was based on the headspace solid-phase microextraction combined with gas chromatography-quadrupole mass spectrometry operating at selected ion monitoring mode (HS-SPME/GC-qMS-SIM), using GC conditions that allowed obtaining a C13 norisoprenoid volatile signature. It does not require any pre-treatment of the sample, and the C13 norisoprenoid composition of the wine was evaluated based on the chromatographic profile and specific m/z fragments, without complete chromatographic separation of its components. The second methodology, used as reference method, was based on the HS-SPME/GC-qMS-SIM, allowing the GC conditions for an adequate chromatographic resolution of wine components. For quantification purposes, external calibration curves were constructed with β-ionone, with regression coefficient (r2) of 0.9968 (RSD 12.51 %) and 0.9940 (RSD of 1.08 %) for the rapid method and for the reference method, respectively. Low detection limits (1.57 and 1.10 μg L-1) were observed. These methodologies were applied to seventeen white and red table wines. Two vitispirane isomers (158-1529 L-1) and 1,1,6-trimethyl-1,2-dihydronaphthalene (TDN) (6.42-39.45 μg L-1) were quantified. The data obtained for vitispirane isomers and TDN using the two methods were highly correlated (r2 of 0.9756 and 0.9630, respectively). A rapid methodology for the establishment of the varietal volatile profile of Vitis vinifera L. cv. 'Fernão-Pires' (FP) white wines by headspace solid-phase microextraction combined with comprehensive two-dimensional gas chromatography with time-of-flight mass spectrometry (HS-SPME/GCxGC-TOFMS) was developed. Monovarietal wines from different harvests, Appellations, and producers were analysed. The study was focused on the volatiles that seem to be significant to the varietal character, such as mono and sesquiterpenic compounds, and C13 norisoprenoids. Two-dimensional chromatographic spaces containing the varietal compounds using the m/z fragments 93, 121, 161, 175 and 204 were established as follows: 1tR = 255-575 s, 2tR = 0,424-1,840 s, for monoterpenoids, 1tR = 555-685 s, 2tR = 0,528-0,856 s, for C13 norisoprenoids, and 1tR = 695-950 s, 2tR = 0,520-0,960 s, for sesquiterpenic compounds. For the three chemical groups under study, from a total of 170 compounds, 45 were determined in all wines, allowing defining the "varietal volatile profile" of FP wine. Among these compounds, 15 were detected for the first time in FP wines. This study proposes a HS-SPME/GCxGC-TOFMS based methodology combined with classification-reference sample to be used for rapid assessment of varietal volatile profile of wines. This approach is very useful to eliminate the majority of the non-terpenic and non-C13 norisoprenic compounds, allowing the definition of a two-dimensional chromatographic space containing these compounds, simplifying the data compared to the original data, and reducing the time of analysis. The presence of sesquiterpenic compounds in Vitis vinifera L. related products, to which are assigned several biological properties, prompted us to investigate the antioxidant, antiproliferative and hepatoprotective activities of some sesquiterpenic compounds. Firstly, the antiradical capacity of trans,trans-farnesol, cis-nerolidol, α-humulene and guaiazulene was evaluated using chemical (DPPH• and hydroxyl radicals) and biological (Caco-2 cells) models. Guaiazulene (IC50= 0.73 mM) was the sesquiterpene with higher scavenger capacity against DPPH•, while trans,trans-farnesol (IC50= 1.81 mM) and cis-nerolidol (IC50= 1.48 mM) were more active towards hydroxyl radicals. All compounds, with the exception of α-humulene, at non-cytotoxic levels (≤ 1 mM), were able to protect Caco-2 cells from oxidative stress induced by tert-butyl hydroperoxide. The activity of the compounds under study was also evaluated as antiproliferative agents. Guaiazulene and cis-nerolidol were able to more effectively arrest the cell cycle in the S-phase than trans,trans-farnesol and α-humulene, being the last almost inactive. The relative hepatoprotection effect of fifteen sesquiterpenic compounds, presenting different chemical structures and commonly found in plants and plant-derived foods and beverages, was assessed. Endogenous lipid peroxidation and induced lipid peroxidation with tert-butyl hydroperoxide were evaluated in liver homogenates from Wistar rats. With the exception of α-humulene, all the sesquiterpenic compounds under study (1 mM) were effective in reducing the malonaldehyde levels in both endogenous and induced lipid peroxidation up to 35% and 70%, respectively. The developed 3D-QSAR models, relating the hepatoprotection activity with molecular properties, showed good fit (R2LOO > 0.819) with good prediction power (Q2 > 0.950 and SDEP < 2%) for both models. A network of effects associated with structural and chemical features of sesquiterpenic compounds such as shape, branching, symmetry, and presence of electronegative fragments, can modulate the hepatoprotective activity observed for these compounds. In conclusion, this study allowed the development of rapid and in-depth methods for the assessment of varietal volatile compounds that might have a positive impact on sensorial and health attributes related to Vitis vinifera L. These approaches can be extended to the analysis of other related food matrices, including grapes and musts, among others. In addition, the results of in vitro assays open a perspective for the promising use of the sesquiterpenic compounds, with similar chemical structures such as those studied in the present work, as antioxidants, hepatoprotective and antiproliferative agents, which meets the current challenges related to diseases of modern civilization.

Nesta tese, realizada no âmbito do Programa Doutoral em Química da Universidade de Aveiro, foram desenvolvidas duas famílias de receptores sintéticos: macrocíclicos baseados na plataforma tetraazacalix[2]areno[2]triazina; e acíclicos construídos a partir de diaminas simples. A plataforma macrocíclica foi decorada nos átomos de azoto em ponte com unidades de reconhecimento molecular contendo fragmentos com grupos amida para o reconhecimento de aniões ou com grupos ácidos carboxílicos para a coordenação de metais de transição. Os receptores acíclicos foram obtidos por acoplamento de diaminas (etilenodiamina, orto-fenilenodiamina ou 2-aminobenzilamina) com uma unidade lipofílica incorporando um anel heterocíclico (derivados de oxadiazole ou furano) e com um derivado isocianato. Estas moléculas assimétricas com um grupo amida e um grupo ureia como unidades de reconhecimento molecular foram avaliadas como receptores e transportadores transmembranares de aniões biologicamente relevantes (Cl- e HCO3-). Os resultados experimentais obtidos serão descritos ao longo de três capítulos, após um primeiro capítulo bibliográfico. No Capítulo 1 começa-se por fazer uma revisão bibliográfica sucinta sobre o desenvolvimento recente de receptores funcionais baseados em azacalixarenos bem como das suas aplicações, designadamente no reconhecimento molecular. Numa segunda parte apresenta-se uma revisão sucinta de receptores derivados de (tio)ureias, relacionados com os receptores sintetizados no âmbito desta tese e com propriedades de reconhecimento e transporte transmembranar de aniões. No Capítulo 2 reporta-se uma série de macrociclos novos com os átomos de azoto em ponte de tetraazacalix[2]areno[2]triazina funcionalizados com bromoacetato de metilo. Foram preparados três novos macrociclos com quatro grupos éster, como braços pendentes, a partir de percursores tetraazacalix[2]areno[2]triazina com os anéis de triazina substituídos com cloro, metilamina ou hexilamina. Os grupos acetato foram hidrolisados em condições básicas, tendo cada um dos derivados dialquilamina originado um composto com quatro grupo carboxílicos, enquanto o análogo diclorado originou uma mistura de compostos com dois grupos carboxílico e com os átomos de cloro substituídos por grupos hidroxilo. Subsequentemente, as propriedades de coordenação dos derivados alquilamina para cobre(II) foram avaliadas por espectroscopia de UV-Vis, tendo-se obtido constantes de estabilidades semelhantes (logk ≈ 6,7). No Capítulo 3 descrevem-se três macrociclos obtidos através da funcionalização dos átomos de azoto em ponte de tetraazacalix[2]areno[2]triazina com grupos amida derivados de N-Boc-etilenodiamina, benzilamina e (S)-metilbenzilamina. A afinidade destes receptores para a série de aniões carboxilato (oxalato, malonato, succinato, glutarato, diglicolato, pimelato, suberato, fumarato, maleato, ftalato e isoftalato) e inorgânicos (Cl-, H2PO4- e SO42-) por titulação de RMN de 1H, foi avaliada. Estes macrociclos conjuntamente com os descritos no Capítulo 2 são os primeiros exemplos reportados na literatura de receptores sintéticos baseados na plataforma de tetraazacalix[2]areno[2]triazina com grupos funcionais nos azotos em ponte. O receptor derivado de N-Boc-etilenodiamina, com oito grupos N-H, entre os três receptores, é o que apresenta maior afinidade para os aniões estudados. No Capítulo 4 é descrita a síntese 59 compostos acíclicos (vide supra) obtidos em três passos de síntese com bons rendimentos. No design desta biblioteca de moléculas a afinidade para aniões dos grupos ureia foi modelada pela inserção de diferentes substituintes arilo ou alquilo, com propriedades electrónicas distintas. A introdução destes grupos em conjugação com um anel de oxadiazole ou furano permitiu também modelar a lipofília destes compostos. A afinidade destes receptores para aniões cloreto e bicarbonato, e em alguns casos para fumarato e maleato, foi investigada por titulação de RMN de 1H. Estes compostos apresentaram constantes de associações compatíveis com o transporte transmembranar de cloreto. Por outro lado estes receptores apresentaram afinidades elevadas para fumarato e maleato, com seletividade para este último. São também discutidos os resultados dos ensaios de transporte de cloreto por estes receptores através de vesículas de em POPC. No Capítulo 5 encontram-se as conclusões gerais desta tese de Doutoramento. No Capitulo 6 encontram-se os dados espectroscópicos e os restantes detalhes experimentais para todos os compostos sintetizados.

The present work has the merit of exploring an insight into the activation of defence genes of Quercus suber during response to infection by Phytophthora cinnamomi. Thus, cDNA-AFLP methodology was used to identify gene fragments differentially present in the mRNA profiles of host cells of micropropagated Q. suber plantlets roots infected with zoospores of P. cinnamomi at different post challenge time points. Six candidate genes were selected based on their interesting cDNA-AFLP expression patterns and homology to genes known to play a role in defence. These six genes encode a cinnamyl alcohol dehydrogenase 2 (QsCAD2), a protein disulphide isomerase (QsPDI), a CC-NBS-LRR resistance protein (QsRPc), thaumatin-like protein (QsTLP), chitinase (QsCHI) and a 1,3-beta glucanase (QsGLU). The current work has been successful in evaluation of the expression of these genes by qRT-PCR. Data analysis revealed that transcript levels of QsRPc, QsCHI, QsCAD2 and QsPDI increased during the early hours of inoculation, while transcript profiles of thaumatin-like protein showed decreasing. No expression was detected for 1,3-beta-glucanase (QsGLU). Furthermore, the choice of suitable reference genes in any new experimental system is absolutely crucial in qRT-PCR; for this reason in this study and for the first time a set of potential reference genes were analyzed and validated for qRT-PCR normalization in the patho-system Phytophthora-Q. suber. Four candidate reference genes polimerase II (QsRPII), eukaryotic translation initiation factor 5A(QsEIF-5A), b-tubulin (QsTUB) and a medium subunit family protein of Clathrin adaptor complexes (QsCACs) were evaluated to determine the most stable internal references in Q. suber. Analysis of stability of genes was carried out using Genex software. Results indicated all these four potential reference genes assumed stable expression. Data analysis revealed that QsRPII and QsCACs were the two most stable genes, while genes QsTUB and QsEIF-5A were the third and the fourth most stable gene, respectively. In this study, a plasmid-based quantitative PCR method was developed to measure P. cinnamomi colonization during infection process of Q. suber. Plasmid-based detection of P. cinnamomi showed a gradual accumulation of the pathogen DNA in cork oak root tips up to 24 h post infection. The higher increase in P. cinnamomi/plasmid DNA ratio occurred between 18 and 24 h. One of the primary objectives of this research was to study the effect of cinnamomins (elicitins secreted by P. cinnamomin) on inducing defence mechanism against the pathogen, as recent histological and ultra-structural studies showed that P. cinnamomi was restricted to the outer cortex root fragments pre-treated with capsicien and cryptogein, suggesting that elicitins can stimulate plant defence reactions against P. cinnamomi. To complement these studies and to have a clear view of the nature of the interaction, the role of cinnamomins in the production of the oxidative burst [ROS and ROS scavenging enzymes such as superoxide dismutase (SOD), catalase (CAT) and peroxidase (POD)] and in the defence responses was evaluated. Cork oak seedlings were pretreated with alpha-cinnamomin and then inoculated with P. cinnamomi mycelia. Results showed a significant higher production of reactive oxygen species (ROS) (H2O2 and O2•-) in elicitin and non-elicitin treated roots in interaction with P. cinnamomi in comparison to the corresponding control. The plant group inoculated with the pathogen after cinnamomin treatment showed an earlier increase in H2O2 production but this was lower as compared with that group inoculated with P. cinnamomi alone. Also, in elicitin pre-treated group generally, a lower level of O2•− production during infection was observed as compared with inoculated roots with P. cinnamomi alone without elicitin treatment. Furthermore, in this study, we evaluated activities of antioxidant enzymes upon challenge with P. cinnamomi, with and without pretreatment with alpha cinnamomin. Results indicated that the activities of defense enzymes POD, SOD and CAT increased after P. cinnamomi inoculation when compared with those in the control group. Also, in the group treated with alpha-cinnamomin followed by P. cinnamomi inoculation, a higher level of enzymatic activities was detected as compared with elicitin non-treated group, which suggest the protective effect of alpha-cinnamomin against the pathogen due to higher elevated levels of defense enzymes POD, SOD and CAT during the infection period. Furthermore, a sensitive qPCR method was applied to measure the pathogen biomass in elicited and non-elicited Q. suber roots challenged with P. cinnamomi to elucidate the effect of cinnamomins on the colonization of P. cinnamomi. Plasmid-based quantification of P. cinnamomi showed a significant decrease in accumulation of the pathogen DNA in cork oak roots after treatment with alpha and beta-cinnamomins which attest the role of cinnamomins in promoting defense responses in cork oak against P. cinnamomi invasion.

O desenvolvimento de tecnologias para a deteção de vírus é um tema de grande interesse em medicina de diagnóstico. Neste contexto, os biossensores têm-se revelado importantes ferramentas bioanalíticas, na medida em que permitem efetuar deteções moleculares de forma rápida, específica, reprodutível e com baixo custo associado. No âmbito desta dissertação, estabeleceram-se metodologias para a deteção fluorescente do fator de infetividade viral (Vif) do VIH através de um nanossensor ótico funcionalizado com anticorpos recombinantes anti-Vif, fragmentos variáveis de cadeia simples 4BL. Quantum dots carboxílicos conjugados a proteínas Vif são excitados a um comprimento de onda de 405 nm, transmitindo luz fluorescente a 605 nm a um fotodetetor de silício amorfo hidrogenado com um filtro de fluorescência integrado. Para implementar um sistema de deteção sensível e eficiente, estudaram-se estratégias de ativação de superfície em chips de vidro. A variabilidade das condições experimentais de um protocolo de silanização com (3-mercaptopropil)-trimetoxisilano (MPTS) foi avaliada através da análise de ângulos de contacto e densidades moleculares de fluoresceína funcionalizada com maleimida. Estabeleceu-se que, uma incubação dos substratos por 4 h com 5% (w/v) MPTS, seguida de cura a 110 ºC por 2 h, e redução com 10 mM ditiotreitol por 30 min, promove a formação de camadas de organosilanos de qualidade com grupos tiol reativos bem orientados. Estratégias de imobilização do elemento recetor foram testadas em substratos de vidro ativados e em sistemas de microfluídica vidro/PDMS, tomando por base o tag de afinidade, glutationa S-transferase (GST), do anticorpo recombinante. Vidros funcionalizados com glutationa e anti-GST mostraram-se igualmente reativos ao anticorpo GST-4BL, resultando uma capacidade de ligação do antigénio de aproximadamente 4.76 x 1010 moléculas/cm2 de QD-Vif, o que corresponde a 15% da cobertura máxima de superfície. Dos ensaios em sistemas de microfluídica resultaram sinais fracos e poucos reprodutíveis, destacando a necessidade de desenvolver metodologias de funcionalização mais apropriadas.
