The influence of different light regimes on the germination of Australian and English populations of Phalaris paradoxa L. (awned canary-grass) seed was investigated to determine the impact of changing tillage practices on weed infestation. Seeds of all biotypes were highly viable, but differed in levels of innate dormancy (26-99%). In one experiment seed from a single Australian biotype, either enclosed in the spikelet glumes or having the spikelet glumes removed, were exposed to nine light treatments. Germination was stimulated by red and white light, but was inhibited by far-red light. Time to 50%, germination was less for seed enclosed in the spikelet glumes than for naked caryopses, although the final percentage of seed germinating when still enclosed in the spikelet glumes was significantly lower than for naked caryopses. In another experiment, six Australian and English biotypes with varying dormancy characteristics were exposed to eight light treatments. Red light did not stimulate germination in the deeply dormant biotype, however stimulated all other biotypes. Germination in darkness was below 20% in all biotypes except for one where germination was 51%. To overcome dormancy seeds were imbibed and placed in darkness at 16degreesC for either 7 or 14 days prior to exposure to red or white light for a single 15-min period. Dormancy in all biotypes was overcome indicating that a period of burial may decrease the dormancy level and increase seed sensitivity to light. This increased light sensitivity suggests that exposure to light during tillage may stimulate germination in P. paradoxa seed.

Female sexuality has commonly been viewed as the passive counterpart of male sexuality. Building upon Adrienne Rich's theory of compulsive heterosexuality, I would suggest that the fundamental location of this problem lies within the subconscious. Cristina Escofet's stance on this issue is to argue in favor of a deconstruction of Jungian archetypes, revealing their constructed rather than intrinsic character. In this dissertation, I study representative texts by Escofet and Isabel Allende and show not only how they depict patriarchal compulsive heterosexuality, but also try to reconceptualize female sexuality through surrealist and postmodern techniques such as self-reflection, dialogue with our double or Other, and sensorial perception. These techniques are designed to create a new epistemology of jouissance and excess, as defined by contemporary French theory. The significance of my study resides in the interdisciplinary analysis of female sexuality in Hispanic feminist writers. The first chapter proposes that surrealism, postmodernism, and feminism are theoretical frameworks which create new paradigms for social change. In their feminist philosophies, Escofet and Allende emphasize the use of subconscious knowledge as a means of helping them understand the world and create alternative realities. The second chapter shows how Escofet and Allende deconstruct the mysoginist archetype of Eve, which has been largely responsible for identifying women's sexual identity with the disreputable qualities of the femme fatale and whose mirror-image has long plagued women. In accordance with this stereotype, Lillith (Adam's sexually active ex-partner), has typically been portrayed as the negative Other, and for generations the she-devil myth which surrounds her has resurfaced in the media, where she assumes the role of innumerable evil female characters. In the third chapter, I examine how class and race differences have been used to intensify the demonization of different types of sexuality. In the same manner as Lillith and Eve, black and indigenous characters express dissent by retelling their stories in words and performance, and by seeking to form a dialog with their readers. The last chapter deals with the importance of the senses for female characters as they try to create their own sexuality from the fragmented bodies we find in surrealist and postmodern art. In this section we shall see how Luce Irigaray and Hélène Cixous's theories about multiple sexualities are in evidence when Escofet and Allende reconceptualize female sexuality. As no previous scholarship has analyzed the use of the subconscious, the senses, and performance when understanding female sexuality in Latin American literature, this dissertation seeks to provide a tentative exploration of the issues that may help to open up a new field of research in Hispanic feminist cultural studies.

Opuntia fícus - indica (L.) Mill is a cactacea presents in the Caatinga ecosystem and shows in its chemical c omposition flavonoids, galacturonic acid and sugars. Different hydroglicolic (EHG001 and EHG002) and hydroethanolic subsequently lyophilized (EHE001 and EHE002) extracts were developed. The EHE002 had his preliminary phytochemical composition investigated by thin layer chromatography (TLC) and we observed the predominance of flavonoids. Different formulations were prepared as emulsions with Sodium Polyacrylate (and) Hydrogenated Polydecene (and) Trideceth - 6 (Rapithix® A60), and Polyacrylamide (and) C13 - 14 I soparaffin (and) Laureth - 7 (Sepigel® 305), and gel with Sodium Polyacrylate (Rapithix® A100). The sensorial evaluation was conducted by check - all - that - apply method. There were no significant differences between the scores assigned to the formulations, howe ver, we noted a preference for those formulated with 1,5% of Rapithix® A100 and 3,0% of Sepigel® 305. These and the formulation with 3% Rapithix® A60 were tested for preliminary and accelerated stability. In accelerated stability study, samples were stored at different temperatures for 90 days. Organoleptic characteristics, the pH values and rheological behavior were assessed. T he emulsions formulated with 3,0% of Sepigel® 305 and 1,5% of Rapithix® A60 w ere stable with pseudoplastic and thixotropic behavior . The moisturizing clinical efficacy of the emulsions containing 3,0% of Sepigel® 305 containing 1 and 3% of EHG001 was performed using the capacitance method (Corneometer®) and transepidermal water lost – TEWL evaluation ( Tewameter®). The results showed t hat the formulation with 3% of EHG001 increased the skin moisturizing against the vehicle and the extractor solvent formulation after five hours. The formulations containing 1 and 3% of EHG001 increased skin barrier effect by reducing transepidermal water loss up to four hours after application.

This thesis will discuss the topic architectural atmospheres, based on Peter Zumthor's works and narrative. The subject "architectural atmospheres" is defined in the study as the "complex and general condition, realized by the user during the experience of a place". Atmospheres are composed of the overlap of all physical qualities that compound a space and its perception, conditioned by individual issues of the user. The atmosphere is realized through all human body receivers, in a multi-sensorial process, and affects qualitatively the spacial experience of the places created by architecture. The study has three chapters. The first chapter is a reflection on the issues that architectural atmospheres present. It passes through the concept and definition of the subject and includes a compilation of design principles that act on the architectural atmosphere composition in a variety of projects by a range of professionals. In the second chapter, the procedures adopted by Peter Zumthor in his design process that focus on atmosphere composition are studied. In the third and last chapter, Brother Claus Chapel (Wachendorf, German) and Kolumba Museum (Colon, German) projects, both by Peter Zumthor, are analyzed, in order to comprehend the complex role of elements that constitute the architectural atmosphere of each of these places. The methodology used throughout the thesis consists of bibliography and documental analysis, based on books, plans and material searched on Internet. Furthermore, the project analysis counted with the personal experience of the author of this thesis upon visiting the studied buildings. Finally, in conclusion, it is realized that when architecture is made in order to touch the user, expressed by its built objects, it can be provide a vast conceptual and theoretical basis that valorizes and enriches the experience of its use. It (the architectural atmosphere) is related to an approach that uses the same elements of any other construction, but distinguishes itself by the connections that it implies with humans and its environment. In the face of new patterns unveiled for contemporary architecture, this study contribution is based around the opportunity to understand this particular relationship between user and architectural object, identified as a phenomenological approach of architecture, as well as to update the subject bibliography, still scarce among Brazilian universities.

La aplicación de las neurociencias en el conocimiento del ser humano ha demostrado cómo el empleo de estímulos sensoriales y experienciales influyen en la atención, el agrado, las emociones, el compromiso –engagement- y la memoria -recuerdo y reconocimiento- de las marcas en entornos publicitarios relacionados con la decisión de compra. El objeto a través del empleo del Neuromarketing es determinar las áreas y funciones de las activaciones cerebrales que modulan la conducta del consumidor relacionadas con la mercadotecnia y el proceso de compra en entornos comerciales. Asimismo identifica el efecto que la publicidad genera en hombres y mujeres al encontrar diferencias de género de cómo les impacta la publicidad como la utilización de personajes célebres a cada uno de ellos, con los que adecuar los mensajes a los segmentos de mercado que tiendan a la excelencia de la eficiencia publicitaria con los que ayudar a los protagonistas clave del sector de la marketing y la publicidad. A través de un estudio propio de Neuromarketing se analiza el comportamiento de hombres y mujeres durante la visualización de los anuncios publicitarios como recurso estratégico para mejorar la eficiencia publicitaria en el diseño y la comunicación de un producto, servicio o campaña publicitaria antes, durante y después del lanzamiento.La medición publicitaria realizada con algunas de las técnicas de Neuroimagen más precisas del mercado, el Electroencefalograma (EEG) y biométricas; el Ritmo Cardíaco (HR) y Respuesta Galvánica de la Piel (GSR), han registrado el aumento del nivel de atención, agrado, emociones o motivación cuando se genera una necesidad o deseo en la demanda de los consumidores mejorando las actuales técnicas de Investigación de Mercado del marketing tradicional. Estas investigaciones permiten presentar la creación del modelo de eficiencia en el diseño y comunicación publicitaria SIADEM (-SENSORY, INTENTION, ATTENTION, DESIRE, EMOTION, MEMORY-) que tiene en cuenta los elementos básicos en los que se debe basar los mensajes publicitarios de cualquier marca para atraer el interés del cerebro consumidor.

No atual contexto de mudança e incerteza no qual vivem as empresas, torna-se fundamental para a continuidade e sucesso do seu negócio, a adoção de estratégias diferenciadoras que permitam aos clientes a perceção de um valor superior dos seus produtos e serviços. O presente trabalho relata as atividades desenvolvidas no âmbito do estágio curricular realizado numa pequena e média empresa de cariz familiar, a Vipex-Comércio e Industria de Plásticos S.A. O estágio foi desenvolvido na área de marketing, tendo sido exercidas atividades de comunicação, sobretudo externa, captação de novos clientes (prospeção) e a aplicação de elementos de marketing sensorial. Numa primeira fase deste relatório é feita a apresentação da empresa Vipex-Comércio e Industria de Plásticos S.A., entidade onde se realizou o estágio curricular, e descritas as principais funções e tarefas desempenhadas ao longo de 8 meses. Numa segunda fase, é realizada uma abordagem teórica sobre a comunicação, prospeção de clientes e marketing sensorial. A literatura enumera os múltiplos benefícios agregados à prospeção e fidelização de clientes. Adicionalmente, é enfatizado que o papel das experiências sensoriais é um novo fator a ter em conta na captação e fidelização dos clientes.

No setor alimentar, o controlo da qualidade dos produtos e dos processos é uma etapa essencial, uma vez que é por este meio que se avaliam os padrões exigidos, quer a nível de legislação, quer a nível de mercado. O presente trabalho teve, como objetivos principais, o desenvolvimento de um novo produto alimentar com adição de algas, bem como o controlo de qualidade a nível de uma unidade fabril de produção de sumos e polpas naturais, à base de hortofrutícolas. De forma a garantir a qualidade na obtenção de produtos, foi realizado, diariamente, o controlo de entradas dos produtos hortofrutícolas, o acompanhamento de processos nas linhas de produção da fábrica e a análise das amostras de referência. Considerando a tendência e potencial de crescimento apresentados pelo setor das bebidas, nomeadamente dos sumos de fruta naturais, juntamente com a incessante procura por ingredientes naturais ricos em vários nutrientes e com propriedades bioativas, tem-se vindo a registar o lançamento de um número considerável de bebidas inovadoras, onde as macroalgas têm merecido um papel de destaque. Desta forma, procedeu-se à elaboração de dois protótipos, nomeadamente, sumo de framboesa e sumo de espinafres, com a adição do extrato da alga Gelidium corneum, estabilizados através do processo de Hiperpressão a frio, com o intuito de se avaliar o impacto desse extrato nos diferentes parâmetros de qualidade dos sumos. No que respeita aos parâmetros físico-químicos, o pH e teor de sólidos solúveis (TSS) foram avaliados após produção (t0), no tempo intermédio (t15) e no tempo final (t30). A cor foi determinada no t0 e no t30 e os restantes parâmetros, como acidez titulável, quantificação total de polifenóis, capacidade de redução do radical DPPH, teor de proteína bruta, teor de cinzas, teor de minerais e oligoelementos e teor de vitaminas (A, B1, B2 e C) foram avaliados no tempo final (t30). Foram, também, avaliados os parâmetros microbiológicos mais significativos, nomeadamente, microrganismos totais a 30 ˚C, Escherichia coli, Salmonella spp, bolores e leveduras (ao t0, t15 e t30) e realizou-se uma avaliação sensorial ao 29.º dia após produção. Estes sumos apresentaram-se microbiologicamente estáveis, não se tendo verificado crescimento microbiológico durante o período de armazenamento de 30 dias. A análise dos resultados físico-químicos permitiram constatar que a adição do extrato da alga Gelidium corneum aos sumos influenciou as suas características, tendo havido um aumento significativo do pH com a consequente diminuição da acidez titulável e um aumento do teor de sólidos solúveis, em comparação com os sumos controlo. Em relação à avaliação da cor, no caso das amostras de sumos de framboesa, verificou-se que a adição do extrato de algas influenciou o parâmetro a*. No entanto, ambas as amostras permitiram a manutenção deste parâmetro, ao longo do tempo. Quanto ao parâmetro b*, as duas amostras de sumo com algas demonstraram uma evolução mais estável, em relação aos respetivos controlos. No que respeita ao teor em minerais, a adição do extrato da alga demonstrou aumentar os teores de magnésio, sódio, potássio e iodo. Em termos sensoriais, o sumo de framboesa com algas obteve maior aceitação por parte do painel de provadores, podendo-se constatar que o atributo da cor foi o que mostrou maiores variações, entre as duas amostras.

The abuse of antibiotics and the emergence of multi-drug resistant bacterial strains have created the need to explore alternative methods of controlling microbial pathogens. The bacteriocin family of antimicrobial peptides has been proposed as one such alternative to classic antibiotics. Nisin A belongs to the subgroup of bacteriocins called the lantibiotics, which contain several unusual amino acids as a consequence of enzyme-mediated post-translational modifications. As nisin is produced by generally regarded as safe (GRAS) microorganisms, it could potentially be applied in a clinical setting. However, as lantibiotics are naturally produced in such small quantities, this can hinder their industrial potential. In order to overcome this, several approaches can be utilised. For example, given the gene encoded nature of lantibiotics, genetic engineering approaches can be implemented in order to yield variants with enhanced properties. Here, the use of mutagenesis-based strategies was employed to obtain a derivative of nisin with enhanced bioactivity in vitro. Investigations with purified peptide highlighted the enhanced specific activity of this variant, nisin M21V, against food-borne Listeria monocytogenes strains. Furthermore, this specific enhanced bioactivity was evident in a mouse model of listeriosis. Reductions in bioluminescence and microbial counts in organs from infected mice were observed following treatment with nisin M21V compared to that of wild-type nisin A. Peptide bioengineering approaches were also implemented to obtain additional novel derivatives of nisin. The generation of “S5X” and “S33X” banks (representing a change of natural serines at positions 5 and 33 to all possible alternative residues) by a combination of site-saturation and site-directed mutagenesis led to the identification of several derivatives exhibiting improved stability. This allowed the rational design of variants with enhanced stability compared to that of wild type nisin. Another means of tackling issues associated with lantibiotic yield is to combine lantibiotics with other antimicrobials. This could circumvent the need for enhanced production while also reducing concentrations of the peptide antimicrobials. We observed that combinations of nisin variants and low levels of plant essential oils (thymol, carvacrol, trans-cinnamaldehyde) significantly controlled Gram negative foodborne pathogens in in vitro assays compared to nisin A-essential oil combinations. This enhanced control was also evident in model food systems. Nisin variants used in conjunction with carvacrol significantly reduced numbers of E. coli O157:H7 in apple juice while a commercial nisin preparation used in combination with citric acid significantly controlled C. sakazakii in infant milk formula. It is noteworthy that while nisin is generally associated with Gram positive targets, upon combination with plant essential oils the spectrum of inhibition was broadened to Gram negative targets.
