Geologic and Engineering Properties of Pleistocene Materials in Iowa, Iowa Highway Research Board Bulletin No. 20, 1960

This bulletin is a compilation of the reports on completed research done for the Iowa State Highway Research Board Project HR-1, "The Loess and Glacial Till Materials of Iowa; an Investigation of Their Physical and Chemical Properties and Techniques for Processing Them to Increase Their All-Weather Stability for Road Construction.” The research, started in 1950, was done by the Iowa Engineering Experiment Station under its project 283-S. The project was supported by funds from the Iowa State Highway Commission.

Cytoskeletal and DNA structure abnormalities result from bypass of requirement for the cdc10 start gene in the fission yeast Schizosaccharomyces pombe.

The cdc10 gene of the fission yeast S. pombe is required for traverse of the start control in late G1 and commitment to the mitotic cell cycle. To increase our understanding of the events which occur at start, a pseudoreversion analysis was undertaken to identify genes whose products may interact with cdc10 or bypass the requirement for it. A single gene, sct1+ (suppressor of cdc ten), has been identified, mutation of which suppresses all conditional alleles and a null allele of cdc10. Bypass of the requirement for cdc10+ function by sct1-1 mutations leads to pleiotropic defects, including microtubule, microfilament and nuclear structural abnormalities. Our data suggest that sct1 encodes a protein that is dependent upon cdc10+ either for its normal function or expression, or is a component of a checkpoint that monitors execution of p85cdc10 function.

Maintenance requirement and deposition efficiency of lysine in pigs

The objective of this work was to determine the maintenance requirement and the deposition efficiency of lysine in growing pigs. It was used the incomplete changeover experimental design, with replicates over time. Twelve castrated pigs with average body weight (BW) of 52±2 kg were kept in metabolism crates with a controlled temperature of 22ºC. The diets were formulated to supply 30, 50, 60, and 70% of the expected requirements of standardized lysine, and provided at 2.6 times the energy requirements for maintenance. The trial lasted 24 days and was divided into two periods of 12 days: seven days for animal adaptation to the diet and five days for sample collection. The increasing content of lysine in the diet did not affect dry matter intake of the pigs. The amount of nitrogen excreted was 47% of the nitrogen intake, of which 35% was excreted through feces and 65% through urine. The estimated endogenous losses of lysine were 36.4 mg kg-1 BW0.75. The maintenance requirement of lysine for pigs weighing around 50 kg is 40.4 mg kg-1 BW0.75, and the deposition efficiency of lysine is 90%.

Modification of the monomer composition of polyhydroxyalkanoate synthesized in Saccharomyces cerevisiae expressing variants of the beta-oxidation-associated multifunctional enzyme.

Expression by Saccharomyces cerevisiae of a polyhydroxyalkanoate (PHA) synthase modified at the carboxy end by the addition of a peroxisome targeting signal derived from the last 34 amino acids of the Brassica napus isocitrate lyase (ICL) and containing the terminal tripeptide Ser-Arg-Met resulted in the synthesis of PHA. The ability of the terminal peptide Ser-Arg-Met and of the 34-amino-acid peptide from the B. napus ICL to target foreign proteins to the peroxisome of S. cerevisiae was demonstrated with green fluorescent protein fusions. PHA synthesis was found to be dependent on the presence of both the enzymes generating the beta-oxidation intermediate 3-hydroxyacyl-coenzyme A (3-hydroxyacyl-[CoA]) and the peroxin-encoding PEX5 gene, demonstrating the requirement for a functional peroxisome and a beta-oxidation cycle for PHA synthesis. Using a variant of the S. cerevisiae beta-oxidation multifunctional enzyme with a mutation inactivating the B domain of the R-3-hydroxyacyl-CoA dehydrogenase, it was possible to modify the PHA monomer composition through an increase in the proportion of the short-chain monomers of five and six carbons.

Benchmarking project of the Department of Electrical Engineering of Lappeenranta University of Technology

Lappeenrannan teknillisen korkeakoulun sähkötekniikan osasto muutti 1.8.2005 sähkötekniikan tutkinnon kaksiportaiseksi ja vastaamaan näin Bologna-prosessia ja Suomen yliopistolainsäädäntöä. Tutkinnonuudistuksen myötä osasto haluaa varmistaa sähkötekniikan tutkintojen laadun ja vertailtavuuden sekä parantaa opiskelijoidensekä henkilökunnan liikkuvuutta. Tutkintojen laatu ja vertailtavuus osoitetaan sähkötekniikan osaston benchmark-projektilla, jossa kerätään tietoja maisteri- ja tohtorintutkintoa tarjoavista eurooppalaisista yliopistoista. Diplomityö käsittää BM-projektin kolmannen vaiheen suunnittelun ja toteutuksen sisältäen teoriaa benchmark-projekteille tyypillisistä toimintatavoista. Hyväksi havaitut menetelmiä, kuten kyselyt ja matriisit, on tässä työssä otettu soveltuvin osin sähkötekniikan osaston BM-projektin työkaluiksi. Diplomityössä analysoidaan työkalujen avulla BM-kumppaneilta kerättyjä tietoja sekä esitetään ratkaisuja, miten sähkötekniikan osastolla voidaan jatkaa parhaiden toimintatapojen löytämistä.

Tarjoustiedon hyödyntäminen asiakasräätälöivän yrityksen toimitusketjussa

Diplomityössä tutkittiin asiakasräätälöityjä tuotteita valmistavan yrityksen tarjous-toimitus-prosessin tehostamismahdollisuuksia. Tutkimuksen tavoitteena on tuoda tarjouksista saatava informaatio ennen kaikkea yrityksen hankintatoimen hyödynnettäväksi. Tarkoitusta varten kehitettiin toimintamalli, jonka avulla myynnistä saatava ennusteinformaatio saadaan koko tuotantoprosessin hyödynnettäväksi. Toimintamallissa pyritään huomioimaan myös asiakaskohtaisen suunnittelunja valmistuksen tarpeita. Toimintamallia kehitettäessä havaittiin tiedonkulun yrityksessä perustuvan tarjousvaiheessa voimakkaasti henkilösuhteisiin. Tieto kulkee sääntelemättömästi henkilöltä toiselle, eikä kokonaisuutta hallita. Tämän tilanteen parantamiseksi toimintamallissa otetaan käyttöön toiminnanohjaus järjestelmän ominaisuuksia, joita ei aiemmin ole käytetty. Tarjoukset kootaan jatkossa toiminnanohjausjärjestelmään myyntitarjoustietokannaksi, josta niitä voidaan hyödyntää materiaalitarvelaskennan lähtötietoina. Kohdeyrityksessä materiaalitarvelaskennan täysmääräinen käyttöönotto on ensimmäinen olennainen vaihe kohti tarjoustiedon järjestelmällistä hyödyntämistä.
