We demonstrate that during inflammatory responses the nuclear factor kappa B (NF-kappa B) induces the synthesis of melatonin by macrophages and that macrophage-synthesized melatonin modulates the function of these professional phagocytes in an autocrine manner. Expression of a DsRed2 fluorescent reporter driven by regions of the aa-nat promoter, that encodes the key enzyme involved in melatonin synthesis (arylalkylamine-N-acetyltransferase), containing one or two upstream kappa B binding sites in RAW 264.7 macrophage cell lines was repressed when NF-kappa B activity was inhibited by blocking its nuclear translocation or its DNA binding activity or by silencing the transcription of the RelA or c-Rel NF-kappa B subunits. Therefore, transcription of aa-nat driven by NF-kappa B dimers containing RelA or c-Rel subunits mediates pathogen-associated molecular patterns (PAMPs) or pro-inflammatory cytokine-induced melatonin synthesis in macrophages. Furthermore, melatonin acts in an autocrine manner to potentiate macrophage phagocytic activity, whereas luzindole, a competitive antagonist of melatonin receptors, decreases macrophage phagocytic activity. The opposing functions of NF-kappa B in the modulation of AA-NAT expression in pinealocytes and macrophages may represent the key mechanism for the switch in the source of melatonin from the pineal gland to immune-competent cells during the development of an inflammatory response.

The ethanol electro-oxidation reaction was evaluated using a polycrystalline Au substrate modified with two different amounts of Pt using the galvanic exchange methodology. FTIR results suggest that Pt deposits have a greater ability to break the C-C bond present in the ethanol molecule. However, under potentiostatic conditions both modified Au surfaces undergo faster deactivation in comparison with polycrystalline platinum as indicated by the chronoamperometric results. XPS results indicate the presence of two phases depending on the Pt content. These are: (i) Pt-Au alloy and (ii) segregated Pt. The structural and electronic properties of these phases were related to the differences observed in the catalytic activity.

We investigated the electrochemical oxidation of glycerol on low-index Pt single crystals in acidic media (H2SO4 and HClO4) by cyclic voltammetry and Fourier Transform Infrared (FTIR) spectroscopy and we verified that this is a surface sensitive reaction. Pt(100) and Pt(110) surface structures favor the breaking of the C-C-C bond at low potentials (say 0.05 V), as seen by the formation of CO, one of the adsorbed residues of the glycerol dissociation, which poisons these surfaces even at high potentials. Pt(111) surface structure does not favor the C-C-C bond breaking at potentials as low as 0.05 V. However, Pt(111) is less poisoned by residues of glycerol dissociation and, for this reason, it is more active for glycerol oxidation than Pt(100) and Pt(110) at low potentials. Carbonyl containing compounds and CO2 were detected as reaction products of the glycerol oxidation on all investigated single-crystal Pt surfaces. The ratio between CO2 and carbonyl containing compounds is clearly much higher for Pt(100) and Pt(110) than for Pt(111). (C) 2012 Elsevier Ltd. All rights reserved.

The crystallographically determined structure of biologically active 4,4-dichloro-1,3-diphenyl-4-telluraoct-2-en-1-one, 3, shows the coordination geometry for Te to be distorted psi-pentagonal bipyramidal based on a C2OCl3(lone pair) donor set. Notable is the presence of an intramolecular axial Te center dot center dot center dot O (carbonyl) interaction, a design element included to reduce hydrolysis. Raman and molecular modelling studies indicate the persistence of the Te center dot center dot center dot O(carbonyl) interaction in the solution (CHCl3) and gasphases, respectively. Docking studies of 3' (i.e. original 3 less one chloride) with Cathepsin B reveals a change in the configuration about the vinyl C = C bond. i.e. to E from Z (crystal structure). This isomerism allows the optimisation of interactions in the complex which features a covalent Te-SGCys29 bond. Crucially, the E configuration observed for 3' allows for the formation of a hypervalent Te center dot center dot center dot O interaction as well as an O center dot center dot center dot H-O hydrogen bond with the Gly27 and Glu122 residues, respectively. Additional stabilisation is afforded by a combination of interactions spanning the S1, S2, S1' and S2' sub-sites of Cathepsin B. The greater experimental inhibitory activity of 3 compared with analogues is rationalised by the additional interactions formed between 3' and the His110 and His111 residues in the occluding loop, which serve to hinder the entrance to the active site. Gellan-based polymer electrolytes (PEs), doped with lithium iodide (LiI), were prepared and their electrical properties were characterized. The samples are thermally stable up to 234 degrees C and exhibit ionic conductivity of 3.8 x 10(-4) S/cm at room temperature for the sample doped with 40 wt% of LiI. Addition of 10 wt% of glycerol promotes an increase of the ionic conductivity to 1.5 x 10(-3) S/cm, which remains stable up to 100 degrees C. The activation energies of 2.4 to 12.4 kJ/mol were derived from the Arrhenius model. The repeated ionic conductivity measurements as a function of temperature show that these membranes can be reversibly used between the room temperature and 100 degrees C.

Gas-phase reactions of model carbosulfonium ions (CH3-S+?=?CH2; CH3CH2-S+?=?CH2 and Ph-S+?=?CH2) and an O-analogue carboxonium ion (CH3-O+?=?CH2) with acyclic (isoprene, 1,3-butadiene, methyl vinyl ketone) and cyclic (1,3-cyclohexadiene, thiophene, furan) conjugated dienes were systematically investigated by pentaquadrupole mass spectrometry. As corroborated by B3LYP/6-311?G(d,p) calculations, the carbosulfonium ions first react at large extents with the dienes forming adducts via simple addition. The nascent adducts, depending on their stability and internal energy, react further via two competitive channels: (1) in reactions with acyclic dienes via cyclization that yields formally [4?+?2+] cycloadducts, or (2) in reactions with the cyclic dienes via dissociation by HSR loss that yields methylenation (net CH+ transfer) products. In great contrast to its S-analogues, CH3-O+?=?CH2 (as well as C2H5-O+?=?CH2 and Ph-O+?=?CH2 in reactions with isoprene) forms little or no adduct and proton transfer is the dominant reaction channel. Isomerization to more acidic protonated aldehydes in the course of reaction seems to be the most plausible cause of the contrasting reactivity of carboxonium ions. The CH2?=?CH-O+?=?CH2 ion forms an abundant [4?+?2+] cycloadduct with isoprene, but similar to the behavior of such alpha,beta-unsaturated carboxonium ions in solution, seems to occur across the C?=?C bond. This work reports aspects of seed germination at different temperatures of Adenanthera pavonina L., a woody Southeast Asian Leguminosae. Germination was studied by measuring the final percentages, the rate, the rate variance and the synchronisation of the individual seeds calculated by the minimal informational entropy of frequencies distribution of seed germination. Overlapping the germinability range with the range for the highest values of germination rates and the minimal informational entropy of frequencies distribution of seed germination, we found that the best temperature for the germination of A. pavonina seeds is 35 ºC. The slope µ of the Arrhenius plot of the germination rates is positive for T < 35 ºC and negative for T > 35 ºC. The activation enthalpies, estimated from closely-spaced points, shows that |ΔH-| < 12 Cal mol-1 occur for temperatures in the range between 25 ºC and 40 ºC. The ecological implication of these results are that this species may germinate very fast in tropical areas during the summer season. This may be an advantage to the establishment of this species under the climatic conditions in those areas.

Using x-ray absorption spectroscopy (XAS), x-ray emission spectroscopy (XES) and x-ray photoelectron spectroscopy (XPS) in combination with density functional theory (DFT) the changes in electronic and geometric structure of hydrocarbons upon adsorption are determined. The chemical bonding is analyzed and the results provide new insights in the mechanisms responsible for dehydrogenation in heterogeneous catalysis. In the case of alkanes, n-octane and methane are studied. XAS and XES show significant changes in the electronic structure upon adsorption. XES shows new adsorption induced occupied states and XAS shows quenching of CH*/Rydberg states in n-octane. In methane the symmetry forbidden gas phase lowest unoccupied molecular orbital becomes allowed due to broken symmetry. New adsorption induced unoccupied features with mainly metal character appear just above the Fermi level in XA spectra of both adsorbed methane and n-octane. These changes are not observed in DFT total energy geometry optimizations. Comparison between experimental and computed spectra for different adsorbate geometries reveals that the molecular structures are significantly changed in both molecules. The C-C bonds in n-octane are shortened upon adsorption and the C-H bonds are elongated in both n-octane and methane. In addition ethylene and acetylene are studied as model systems for unsaturated hydrocarbons. The validity of both the Dewar-Chatt-Duncanson chemisorption model and the alternative spin-uncoupling picture is confirmed, as well as C-C bond elongation and upward bending of the C-H bonds. The bonding of ethylene to Cu(110) and Ni(110) are compared and the results show that the main difference is the amount of back-donation into the molecular π* orbital, which allows the molecule to desorb molecularly from the Cu(110) surface, whereas it is dehydrogenated upon heating on the Ni(110) surface. Acetylene is found to adsorb in two different adsorption sites on the Cu(110) surface at liquid nitrogen temperature. Upon heating the molecules move into one of these sites due to attractive adsorbate-adsorbate interaction and only one adsorbed species is present at room temperature, at which point the molecules start reacting to form benzene. The bonding of the two species is very similar in both sites and the carbon atoms are rehybridized essentially to sp2.

ZusammenfassungDer humane kationische Aminosäure-Transporter hCAT-1 (CAT für cationic amino acid transporter) gehört zur Familie der Na+- und pH-unabhängigen Transporter für basische Aminosäuren (BAS). Die vorliegende Arbeit befasst sich mit unterschiedlichen Aspekten des hCAT-1-vermittelten Transportes, die in zwei Teilabschnitten behandelt werden. Im ersten Abschnitt wurden die Transporteigenschaften von hCAT-1-exprimierenden X. laevis-Oozyten mit Hilfe von elektrophysiologischen Methoden untersucht und mit denen der Isoformen hCAT-2A und -2B verglichen. Dabei zeigte sich, dass es durch die Expression von hCAT-2A und -2B in Oozyten zur Bildung eines BAS-Potentiales kommt, jedoch nicht durch die Expression von hCAT-1. Hierfür dürfte die hohe Transstimulierbarkeit des hCAT-1-Proteins verantwortlich sein. Obwohl das Membranpotential einer Zelle die Akkumulation von BAS durch die hCAT-Proteine beeinflusst, war bei sehr hohen extrazellulären BAS-Konzentrationen die Akkumulation durch hCAT-1 und -2B im Gegensatz zu hCAT-2A nicht vom Membranpotential abhängig, da unter diesen Bedingungen der Efflux limitierend wirkte. Mit Hilfe der voltage clamp-Methode wurden die L-Arginin-induzierten Maximalströme (Vmax) und die Leitfähigkeiten der hCAT-Proteine bestimmt. Die so ermittelten Vmax-Werte sind nur halb so groß wie die durch Flux-Studien bestimmten. Daher muss von einem Gegentransport an positiver Ladung (Substrat) ausgegangen werden. Weiterhin konnte gezeigt werden, dass die hCAT-Isoformen zwei unterschiedliche Leitfähigkeitszustände für BAS besitzen, die von der intrazellulären BAS-Konzentration abhängig sind. Eine Leitfähigkeitszunahme durch Zugabe von extrazellulärem L-Arginin konnte bei allen hCAT-Isoformen in depletierten Oozyten beobachtet werden. In BAS-beladenen Oozyten führte die Zugabe von L-Arginin dagegen zu keiner (hCAT-1 und hCAT-2B) bzw. zu einer geringen (hCAT-2A) Zunahme der Leitfähigkeit der Transporter. Im Substratgleichgewicht jedoch nahm die Leitfähigkeit der drei untersuchten hCAT-Isoformen in Abhängigkeit von der Substratkonzentration zu. Überraschenderweise wurden für die untersuchten hCAT-Isoformen Leck-Ströme in Abwesenheit von BAS nachgewiesen. An hCAT-2B-exprimierenden Oozyten wurde eine erhöhte Leitfähigkeit für K+-Ionen gezeigt. Die physiologische Bedeutung dieser Kanalfunktion ist jedoch noch völlig ungeklärt. Im zweiten Abschnitt wurde der Mechanismus der Proteinkinase C (PKC)-vermittelten Inhibition der hCAT-1-Transportaktivität untersucht. Hierfür wurden hCAT-1.EGFP-Konstrukte in Oozyten und in U373MG Glioblastom-Zellen exprimiert. Mit Hilfe konfokaler Mikroskopie und Western-Blot-Analysen von biotinylierten Zelloberflächen-Proteinen wurde gezeigt, dass die PKC-vermittelte Reduktion der hCAT-1-Transportaktivität auf einer Reduktion der hCAT-Expression an der Zelloberfläche beruht. Ähnliche Ergebnisse wurden auch mit dem endogen in humanen DLD-1 Kolonkarzinom-Zellen exprimierten hCAT-1 erzielt. Der PKC-Effekt war auch noch nach Entfernung der putativen PKC-Erkennungsstellen am hCAT-1-Protein vorhanden. Daher reguliert die PKC die hCAT-1-Transportaktivität vermutlich über einen indirekten Mechanismus, d. h. nicht über eine direkte Phosphorylierung des hCAT-1-Proteins. Die Veränderung der Zelloberflächenexpression stellt einen neuen Regulationsmechanismus für die CAT-Proteine dar, der erklären kann, warum sich Modifikationen in der CAT-Proteinexpression oft nicht in entsprechenden Veränderungen der Transportaktivität widerspiegeln.

The proposal in my thesis has been the study of Stereoselective α-alkylation through SN1 type reaction. SN1 type reaction involves a stabilized and reactive carbocation intermediate By taking advantages of stability of particular carbocations, the use of carbocations in selective reactions has been important. In this work has been necessary to know the stability and reactivity of carbocations. And the work of Mayr group has helped to rationalize the behaviour and reactivity between the carbocations and nucleophiles by the use of Mayr’s scale of reactivity. The use of alcohols to performed the stable and reactive carbocations have been the key in my thesis. The direct nucleophilic substitution of alcohols has been a crucial scope in the field of organic synthesis, because offer a wide range of intermediates for the synthesis of natural products and pharmaceutics synthesis. In particular the catalytic nucleophilic direct substitution of alcohols represents a novel methodology for the preparation of a variety of derivatives, and water only as the sub-product in the reaction. The stereochemical control of the transformation C-H bond into stereogenic C-C bond adjacent to carbonyl functionalized has been studied for asymmetric catalysis. And the field of organocatalysis has introduced the use of small organic molecule as catalyst for stereoselective transformations. Merging these two concepts Organocatalysis and Mayr’s scale, my thesis has developed a new approach for the α-alkylation of aldehydes and ketones through SN1 type reaction.

Zusammenfassung Nanokomposite aus Polymeren und Schichtsilikaten werden zumeist auf der Basis natürlicher Tone wie Montmorillonit hergestellt. Für NMR- und EPR-Untersuchungen der Tensidschicht, die das Silikat mit dem Polymer kompatibilisiert, ist der Eisengehalt natürlicher Tone jedoch abträglich, weil er zu einer Verkürzung der Relaxationszeiten und zu einer Linienverbreiterung in den Spektren führt. Dieses Problem konnte überwunden werden, indem als Silikatkomponente eisenfreies, strukturell wohldefiniertes Magadiit hydrothermal synthetisiert und für die Kompositbildung eingesetzt wurde. Die Morphologie des Magadiits wurde durch Rasterelektronenmikroskopie charakterisiert und der Interkalationsgrad von schmelzinterkalierten Polymer-Nanokompositen wurde durch Weitwinkelröntgenstreuung bestimmt. Polymere mit Carbonylgruppen scheinen leichter zu interkalieren als solche ohne Carbonylgruppen. Polycaprolacton interkalierte sowohl in Oragnomagadiite auf der Basis von Ammoniumtensiden als auch in solche auf der Basis von Phosphoniumtensiden. Die Dynamik auf einer Nanosekundenzeitskala und die Struktur der Tensidschicht wurden mittels ortsspezifisch spinmarkierter Tensidsonden unter Nutzung von Dauerstrich- (CW) und Puls-Methoden der elektronenparamagnetischen Resonanzspektroskopie (EPR) untersucht. Zusätzlich wurde die statische 2H-Kernmagnetresonanz (NMR) an spezifisch deuterierten Tensiden angewendet, um die Tensiddynamik auf einer komplementären Zeitskala zwischen Mikrosekunden und Millisekunden zu erfassen. Sowohl die CW-EPR- als auch die 2H-NMR-Ergebnisse zeigen eine Beschleunigung der Tensiddynamik durch Interkalation von Polycaprolacton auf, während sich in den nichtinterkalierten Mikrokompositen mit Polystyrol die Tensiddynamik verlangsamt. Die Rotationskorrelationszeiten und Aktivierungsenergien offenbaren verschiedene Regime der Tensiddynamik. In Polystyrol-Mikrokompositen entspricht die Übergangstemperatur zwischen den Regimen der Glasübergangstemperatur von Polystyrol, während sie in Polycaprolacton-Nanokompositen bei der Schmelztemperatur von Polycaprolacton liegt. Durch die erhebliche Verlängerung der Elektronenspin-Relaxationszeiten bei Verwendung von eisenfreiem Magadiit können Messdaten hoher Qualität mit Puls-EPR-Experimenten erhalten werden. Insebsondere wurden die Vier-Puls-Elektron-Elektron-Doppelresonanz (DEER), die Elektronenspinechoenveloppenmodulation (ESEEM) und die Elektronen-Kern-Doppelresonanz (ENDOR) an spinmarkierten sowie spezifisch deuterierten Tensiden angewandt. Die ENDOR-Ergebnisse legen ein Model der Tensidschicht nahe, in dem zusätzlich zu den Oberflächenlagen auf dem Silikat eine wohldefinierte mittlere Lage existiert. Dieses Modell erklärt auch Verdünnungseffekte durch das Polymer in Kompositen mit Polycaprolacton und Polystyrol. Die umfangreiche Information aus den Magnetresonanztechniken ergänzt die Information aus konventionellen Charakterisierungstechniken wie Röntgendiffraktion und Transmissionselektronenmikroskopie und führt so zu einem detaillierteren Bild der Struktur und Dynamik der Tensidschicht in Nanokompositen aus Polymeren und Schichtsilikaten.

Das Stannylen SnHyp2 (Hyp = Si(SiMe3)3) reagiert mit den Übergangsmetallhydrid-Komplexen Cp2MH2 (Cp = C5H5, M = Mo, W) in einer alpha-Additionsreaktion zu Cp2MSn(H)Hyp2. Ferner bilden sich unter Abspaltung von HSi(SiMe3)3 auch vierkernige Verbindungen der Form [Cp2MSn(H)Hyp]2, welche für M = Mo röntgendiffraktometrisch nachgewiesen wurden. Erhöht man den sterischen Anspruch der Silylreste des Stannylens, so nimmt die Tendenz der Adduktbildung ab, so dass nur die vierkernigen Komplexe nachweisbar sind. Im Fall für SnSit2 (Sit = Si(SiMe3)2SiMe2tBu) konnten sogar Stereoisomere der vierkernigen Verbindungen [Cp2MoSn(H)Sit]2 nachgewiesen werden. Im Gegensatz dazu reagiert das Plumbylen PbHyp2 mit Cp2MoH2 in einer Substitutionsreaktion zu dem nachgewiesenen Cp2Mo(H)Hyp. Bei größer werdendem Silylrest werden andere Reaktionsabläufe bevorzugt. Auf dem Forschungsgebiet der Ein-Elektronen-Reduktion von Halogenstannanen und -plumbanen XER2R´ (X = Halogen; E = Sn, Pb; R = Silylrest; R = Aryl- / Alkylrest) mit dem 19-Elektronen-Komplex Decamethylcobaltocen CoCp*2 (Cp* = C5Me5) konnten Unterschiede zwischen den Verbindungen des Zinns und des Bleis festgestellt werden: Bei der Reduktion von Halogenstannanen fallen die erwarteten Decamethylcobaltocenium-Halogenide [CoCp*2]X aus und ESR-Messungen bestätigen die Anwesenheit von Stannylradikalen, während bei der Reduktion von hypersilylierten Halogenplumbanen unterschiedliche Folgeprodukte entstehen und Plumbylradikale via ESR-Spektroskopie nicht feststellbar sind. Bei alkylhaltigen Halogenplumbanen XPbHyp2R (R = Alkyl) findet eine Spaltung der Pb-C-Bindung statt und es bilden sich Plumbate der Form [CoCp*2][PbHyp2X], während die Reduktion von einem arylhaltigen Halogenplumban IPbHyp2Ph (Ph = Phenyl) das erwartete Decamethylcobaltocenium-Salze ergibt.

Several studies have shown the presence of liver mitochondrial dysfunction during sepsis. TLR3 recognizes viral double-stranded RNA and host endogenous cellular mRNA released from damaged cells. TLR3 ligand amplifies the systemic hyperinflammatory response observed during sepsis and in sepsis RNA escaping from damaged tissues/cells may serve as an endogenous ligand for TLR3 thereby modulating immune responses. This study addressed the hypothesis that TLR3 might regulate mitochondrial function in cultured human hepatocytes. HepG2 cells were exposed to TLR-3 ligand (dsRNA--polyinosine-polycytidylic acid; Poly I:C) and mitochondrial respiration was measured. Poly I:C induced a reduction in maximal mitochondrial respiration of human hepatocytes which was prevented partially by preincubation with cyclosporine A (a mitochondrial permeability transition pore-opening inhibitor). Poly-I:C induced activation of NF-κB, and the mitochondrial dysfunction was accompanied by caspase-8 but not caspase-3 activation and by no major alterations in cellular or mitochondrial ultrastructure.

The conversion of alkylboranes to the corresponding alkanes is classically performed via protonolysis of alkylboranes. This simple reaction requires the use of severe reaction conditions, that is, treatment with a carboxylic acid at high temperature (>150 degrees C). We report here a mild radical procedure for the transformation of organoboranes to alkalies. 4-tert-Butylcatechol, a well-established radical inhibitor and antioxidant, is acting as a source of hydrogen atoms. An efficient chain reaction is observed due to the exceptional reactivity of phenoxyl radicals toward alkylboranes. The reaction has been applied to a wide range of organoboron derivatives such as B-alkylcatecholboranes, trialkylboranes, pinacolboronates, and alkylboronic acids. Furthermore, the so far elusive rate constants for the hydrogen transfer between secondary alkyl radical and catechol derivatives have been experimentally determined. Interestingly, they are less than 1 order of magnitude slower than that of tin hydride at 80 degrees C, making catechols particularly attractive for a wide range of transformations involving C-C bond formation.

The IkappaB kinase (IKK) complex controls processes such as inflammation, immune responses, cell survival and the proliferation of both normal and tumor cells. By activating NFkappaB, the IKK complex contributes to G1/S transition and first evidence has been presented that IKKalpha also regulates entry into mitosis. At what stage IKK is required and whether IKK also contributes to progression through mitosis and cytokinesis, however, has not yet been determined. In this study, we use BMS-345541, a potent allosteric small molecule inhibitor of IKK, to inhibit IKK specifically during G2 and during mitosis. We show that BMS-345541 affects several mitotic cell cycle transitions, including mitotic entry, prometaphase to anaphase progression and cytokinesis. Adding BMS-345541 to the cells released from arrest in S-phase blocked the activation of Aurora A, B and C, Cdk1 activation and histone H3 phosphorylation. Additionally, treatment of the mitotic cells with BMS-345541 resulted in precocious cyclin B1 and securin degradation, defective chromosome separation and improper cytokinesis. BMS-345541 was also found to override the spindle checkpoint in nocodazole-arrested cells. In vitro kinase assays using BMS-345541 indicate that these effects are not primarily due to a direct inhibitory effect of BMS-345541 on mitotic kinases such as Cdk1, Aurora A or B, Plk1 or NEK2. This study points towards a new potential role of IKK in cell cycle progression. Since deregulation of the cell cycle is one of the hallmarks of tumor formation and progression, the newly discovered level of BMS-345541 function could be useful for cell cycle control studies and may provide valuable clues for the design of future therapeutics.
