To increase transient expression of recombinant proteins in Chinese hamster ovary cells, we have engineered their protein synthetic capacity by directed manipulation of mRNA translation initiation. To control this process we constructed a nonphosphorylatable Ser51Ala site-directed mutant of eIF2, a subunit of the trimeric eIF2 complex that is implicated in regulation of the global rate of mRNA translation initiation in eukaryotic cells. Phosphorylation of eIF2 by protein kinases inhibits eIF2 activity and is known to increase as cells perceive a range of stress conditions. Using single-and dual-gene plasmids introduced into CHO cells by electroporation, we found that transient expression of the eIF2 Ser51Ala mutant with firefly luciferase resulted in a 3-fold increase in reporter activity, relative to cells transfected with reporter only. This effect was maintained in transfected cells for at least 48 h after transfection. Expression of the wild-type eIF2 protein had no such effect. Elevated luciferase activity was associated with a reduction in the level of eIF2 phosphorylation in cells transfected with the mutant eIF2 construct. Transfection of CHO cells with the luciferase-only construct resulted in a marked decrease in the global rate of protein synthesis in the whole cell population 6 h post-transfection. However, expression of the mutant Ser51Ala or wild-type eIF2 proteins restored the rate of protein synthesis in transfected cells to a level equivalent to or exceeding that of control cells. Associated with this, entry of plasmid DNA into cells during electroporation was visualized by confocal microscopy using a rhodamine-labeled plasmid construct expressing green fluorescent protein. Six hours after transfection, plasmid DNA was present in all cells, albeit to a variable extent. These data suggest that entry of naked DNA into the cell itself functions to inhibit protein synthesis by signaling mechanisms affecting control of mRNA translation by eIF2. This work therefore forms the basis of a rational strategy to generically up-regulate transient expression of recombinant proteins by simultaneous host cell engineering.

International non-governmental organisations (NGOs) are powerful political players who aim to influence global society. In order to be effective on a global scale, they must communicate their goals and achievements in different languages. Translation and translation policy play an essential role here. Despite NGOs’ important position in politics and society, not much is known about how these organisations, who often have limited funds available, organise their translation work. This study aims to contribute to Translation Studies, and more specifically to investigating institutional translation, by exploring translation policies at Amnesty International, one of the most successful and powerful human rights NGOs around the world. Translation policy is understood as comprising three components: translation management, translation practices, and translation beliefs, based on Spolsky’s study of language policy (2004). The thesis investigates how translation is organised and what kind of policies different Amnesty offices have in place, and how this is reflected in their translation products. The thesis thus also pursues how translation and translation policy impact on the organisation’s message and voice as it is spread around the world. An ethnographic approach is used for the analysis of various data sets that were collected during fieldwork. These include policy documents, guidelines on writing and translation, recorded interviews, e-mail correspondence, and fieldnotes. The thesis at first explores Amnesty’s global translation policy, and then presents the results of a comparative analysis of local translation policies at two concrete institutions: Amnesty International Language Resource Centre in Paris (AILRC-FR) and Amnesty International Vlaanderen (AIVL). A corpus of English source texts and Dutch (AIVL) and French (AILRC-FR) target texts are analysed. The findings of the analysis of translation policies and of the translation products are then combined to illustrate how translation impacts on Amnesty’s message and voice. The research results show that there are large differences in how translation is organised depending on the local office and the language(s), and that this also influences the way in which Amnesty’s message and voice are represented. For Dutch and French specifically, translation policies and translation products differ considerably. The thesis describes how these differences are often the result of different beliefs and assumptions relating to translation, and that staff members within Amnesty are not aware of the different conceptions of translation that exist within Amnesty International as a formal institution. Organising opportunities where translation can be discussed (meetings, workshops, online platforms) can help in reducing such differences. The thesis concludes by suggesting that an increased awareness of these issues will enable Amnesty to make more effective use of translation in its fight against human rights violations.

By examining the pictorial content of the veintena section of the Primeros Memoriales, the manusctipt compiled by fray Bernardino de Sahagún, I identify new pieces of evidence on the origin of these illustrations and their authors. A carefull analisis of this material suggests that it is strongly embedded in the pre-Hispanic tradition and that it is doubtful that their iconographic sources originated in Tepeopolco, as it is widely believed.

In 1691, eighteen years after its original publication, Samuel Pufendorf’s De officio hominis et civis appeared in English translation in London, bearing the title The Whole Duty of Man, According to the Law of Nature. This translation, by Andrew Tooke (1673–1732), professor of geometry at Gresham College, passed largely unaltered through two subsequent editions, in 1698 and 1705, before significant revision and augmentation in the fourth edition of 1716. Unchanged, this text was then reissued as the fifth and final edition of 1735, which is here republished for the first time since.1 Five editions, spanning almost half a century, bear testimony to the English appetite for Pufendorf’s ideas.

Cobalamins are stored in high concentrations in the human liver and thus are available to participate in the regulation of hepatotropic virus functions. We show that cyanocobalamin (vitamin B12) inhibited the H(IV internal ribosome entry site (IRES)-dependent translation of a reporter gene in vitro in a dose-dependent manner without significantly affecting the cap-dependent mechanism. Vitamin B12 failed to inhibit translation by IRES elements from encephalomyocarditis virus (EMCV) or classical swine fever virus (CSFV), We also demonstrate a relationship between the total cobalamin concentration in human sera and HCV viral load (a measure of viral replication in the host), The mean viral load was two orders of magnitude greater when the serum cobalamin concentration was above 200 pM (P < 0.003), suggesting that the total cobalamin concentration in an HCV-infected liver is biologically significant in HCV replication.

The effect of cyanocobalamin (CNCbl, vitamin 1312) on hepatitis C virus internal ribosome entry site (HCV IRES)-dependent initiation of translation was studied by ribosomal toeprinting and sucrose gradient centrifugation analysis. These results suggested that CNCbl did not inhibit HCV IRES-dependent translation by a competitive binding mechanism. CNCbl allowed 80 S elongation complex formation on the mRNA, but stalled the initiation at that point, effectively trapping the 80 S ribosomal complexes on the HCV TRES. CNCbl had no effect on cap-dependent mRNA, consistent with the known mRNA specificity of this translational inhibitor. To help elucidate the mechanism, comparative data were collected for the well-characterised translation inhibitors cycloheximide and 5'-guanylyl-imidophosphate, Although CNCbl stalled HCV IRES-dependent translation at approximately the same step in initiation as cycloheximide, the mechanisms of these two inhibitors are distinct. AIMS/HYPOTHESIS: Chronic exposure of pancreatic beta cells to proinflammatory cytokines leads to impaired insulin secretion and apoptosis. ARE/poly(U)-binding factor 1 (AUF1) belongs to a protein family that controls mRNA stability and translation by associating with adenosine- and uridine-rich regions of target messengers. We investigated the involvement of AUF1 in cytokine-induced beta cell dysfunction. METHODS: Production and subcellular distribution of AUF1 isoforms were analysed by western blotting. To test for their role in the control of beta cell functions, each isoform was overproduced individually in insulin-secreting cells. The contribution to cytokine-mediated beta cell dysfunction was evaluated by preventing the production of AUF1 isoforms by RNA interference. The effect of AUF1 on the production of potential targets was assessed by western blotting. RESULTS: MIN6 cells and human pancreatic islets were found to produce four AUF1 isoforms (p42>p45>p37>p40). AUF1 isoforms were mainly localised in the nucleus but were partially translocated to the cytoplasm upon exposure of beta cells to cytokines and activation of the ERK pathway. Overproduction of AUF1 did not affect glucose-induced insulin secretion but promoted apoptosis. This effect was associated with a decrease in the production of the anti-apoptotic proteins, B cell leukaemia/lymphoma 2 (BCL2) and myeloid cell leukaemia sequence 1 (MCL1). Silencing of AUF1 isoforms restored the levels of the anti-apoptotic proteins, attenuated the activation of the nuclear factor-κB (NFκB) pathway, and protected the beta cells from cytokine-induced apoptosis. CONCLUSIONS/INTERPRETATION: Our findings point to a contribution of AUF1 to the deleterious effects of cytokines on beta cell functions and suggest a role for this RNA-binding protein in the early phases of type 1 diabetes.

Iowa’s traffic safety culture is influenced by laws and policies, enforcement methods, driver education, roadway engineering, and drivers’ behaviors. The Center for Social and Behavioral Research at the University of Northern Iowa was contracted by the Iowa Department of Transportation to conduct a general population survey of adult Iowans. Telephone interviews were conducted with 1,088 adult Iowans from October to December 2011. A dual-frame (cell phone and landline) sampling design was used. The interview covered a wide range of traffic safety topics (e.g., traffic safety policies, enforcement techniques, and distracted driving). Most Iowans said driving in Iowa is about as safe now as it was 5 years ago; however, one-fourth said driving in Iowa is less safe now. There are a number of driving-related behaviors many adult Iowans consider serious threats to traffic safety and never acceptable to do while driving. Yet, many Iowans report often seeing other drivers engaging in these behaviors and admit engaging in some themselves. For example, nearly 1 in 5 adult Iowa drivers said they have sent or read a text message or email while driving in the past 30 days despite this being prohibited since July of 2011. A slight majority said they support using cameras on highways, interstates, and city streets to automatically ticket drivers for speeding, with even stronger support for red light cameras. A comprehensive approach to traffic safety in Iowa is required to encourage protective factors that enhance traffic safety and reduce the impact of detrimental factors.

A admiración que destilou no seu tempo e durante o século XIX o bo sentido común das obras de Benjamin Franklin (1706-1790) plasmouse na tradución dos seus libros a un considerable número de linguas. The Way to Wealth (El camino de la fortuna o Cómo hacerse rico), publicado en 1758, contén máis de cen máximas que instan ao traballo honrado, á orde e ao aforro, condenando calquera sinal de preguiza ou desidia. O noso corpus nútrese dunha selección de trece aforismos que teñen relación precisamente con este vicio que condena á miseria, a preguiza. En cambio, o home de ben que aplique as virtudes contidas en The Way to Wealth, aconselladas a modo de refráns, verá, segundo o autor, prosperar esplendidamente a súa economía. Pretendemos analizar, dende os puntos de vista tradutolóxico e contrastivo, o corpus de dúas traducións españolas deste libriño de Franklin: unha anónima, publicada en Barcelona, en 1891; e a de Alberto Lena de 1999.

Diplomityön tavoitteena oli selvittää jakeluverkkoyhtiön verkon jälleenhankinta-arvon määrittämiseen tarvittavat lähtötiedot sähkönjakeluverkon rakentamiskustannuksia selvittäneellä kyselyllä. Työssä on käsitelty sähkönjakeluverkon jälleenhankinta-arvon, nykyarvon ja nykykäyttöarvon määritysmenetelmiä. Lisäksi on esitelty laeissa, asetuksissa ja suosituksissa jakeluverkkotoiminnalle asetettuja vaatimuksia. Tulokset on esitelty komponenteittain ja tutkimalla kuinka paljon kahden kuvitteellisen sähkönjakeluverkon jälleenhankinta-arvo muuttuu. Palkkakustannusten nousu on lisännyt kustannuksia, mutta vastaavasti verkonrakentamisen tehostuminen sekä tässä laskentamallissa esitetyt kustannuksien määrittelyt (esimerkkinä muuntajat) ovat alentaneet yleiskustannuksia, joten kokonaisuutena verkon rakentamisen kustannukset ovat joidenkin komponenttien osalta alentuneet tässä työssä käytettyihin KA 2:95, KA 2:96 ja KA 2:97 kustannustietoihin verrattuna.

Osakkeenomistajien keskinaisia suhteita sekä osakkeenomistajien suhdetta yhtiöön ja kolmanteen sääntelevät muun muassa osakeyhtiölain (29.9.1978/734) saannokset ja yhtiöjärjestyksen määräykset. Osakeyhtiölaissa on vain rajoitetusti otettu huomioon se, että osakeyhtiöiden tarkoitukset vaihtelevat ja että ne ovat erikokoisia. Saannokset soveltuvat parhaiten suuren tai keskisuuren osakeyhtiön osakaspiirin tarpeisiin. Osakassopimuksella pyritään tavallisesti siihen, että asiat olisivat ennen niiden käsittelemistä yhtiön elimissä jo edeltä kasin sovittu. Paatosten tekemistä ei kuitenkaan voida osakkeenomistajien keskinaisella sopimuksella siirtää osakeyhtiölaissa sääntelemättömille elimille. Osakassopimuksilla pyritään varmistamaan osakkaiden oikeudet yhtiön elinkaaren aikana muuttuvissa tilanteissa. Yhdistävänä tekijänä niissä on valta ja sen käyttö. Osakassopimukset ovat jaettavissa vallan keskittämistä, hajauttamista ja tasapainon ylläpitämistä koskeviin sopimuksiin. Osakassopimuksella saatetaan pyrkiä säilyttämään osakkeenomistajien osakassopimuksen tekohetken aikaiset vaikutusmahdollisuudet muuttumattomina heidän keskinaisissa suhteissaan ja suhteissa ulkopuolisiin intressiryhmiin.
