Dar cumplimiento al proyecto de tesis a través de una experiencia de investigación. Retribuir el costo del posgrado, poniendo en beneficio de la institución, los modestos conocimientos alcanzados. Abordar una investigación estructurada y concreta sobre el colegio Rafael Reyes para dinamizar una mejora integral. Promover cambios pedagógicos en la institución acorde con las nuevas tecnologías. Optimizar los recursos humanos, económicos y tecnológicos con que cuenta el colegio. Generar un clima de reingeniería que conecte el colegio con la tendencia educativa del nuevo milenio. Se ha trabajado de forma teórica y práctica. El primer capítulo hace una breve reseña histórica de la evaluación. El segundo capítulo explica cómo se puede realizar una evaluación de centros educativos. El tercer capítulo ofrece algunas premisas a la hora de evaluar un centro educativo. El cuarto capítulo habla de la calidad en la institución escolar. El quinto capítulo es un breve diagnóstico de la evaluación en el contexto colombiano. El sexto capítulo explica el contexto del colegio general Rafael Reyes. El séptimo capítulo es un modelo de auto-evaluación institucional centrado en el cambio con una propuesta para mejorar algunos aspectos del colegio. Es necesario verificar las políticas de descentralización de los procesos curriculares, debido a la sacralización que se ha generado de la autonomía curricular. De acuerdo con los resultados de la evaluación realizada, están desembocando en la ambigüedad de contenidos. Poniendo en riesgo el nivel académico de la institución, y lo más grave, dejando a la deriva la responsabilidad adquirida de graduar a los bachilleres, que enfrentarán con el tiempo el reto de la universidad, con las herramientas y los conocimientos indispensables para que continúen creciendo integralmente. La institución carece de una plataforma informática que pueda garantizar el aprendizaje actualizado en el área de tecnología, lo que ha producido en los estudiantes una desmotivación casi general, frenando de alguna manera la dinámica que es posible ofrecer mediante este recurso tecnológico. Como quiera que, cada día la educación se encuentra más mediatizada por la tecnología, no se puede mirar a espaldas de ella, sin recibir, como consecuencia lógica, la desmotivación de los jóvenes.

Background and aims Unilateral incompatibility (UI) occurs when pollinations between species are successful in one direction but not in the other. Self-incompatible (SI) species frequently show UI with genetically related, self-compatible (SC) species, as pollen of SI species is compatible on the SC pistil, but not vice versa. Many examples of unilateral incompatibility, and all those which have been studied most intensively, are found in the Solanaceae, particularly Lycopersicon, Solanum, Nicotiana and Petunia. The genus Capsicum is evolutionarily somewhat distant from Lycopersicon and Solanum and even further removed from Nicotiana and Petunia. Unilateral incompatibility has also been reported in Capsicum; however, this is the first comprehensive study of crosses between all readily available species in the genus. Methods All readily available (wild and domesticated) species in the genus are used as plant material, including the three genera from the Capsicum pubescens complex plus eight other species. Pollinations were made on pot-grown plants in a glasshouse. The number of pistils pollinated per cross varied (from five to 40 pistils per plant), depending on the numbers of flowers available. Pistils were collected 24 h after pollination and fixed for 3-24 h. After staining, pistils were mounted in a drop of stain, squashed gently under a cover slip and examined microscopically under ultra-violet light for pollen tube growth. Key results Unilateral incompatibility is confirmed in the C. pubescens complex. Its direction conforms to that predominant in the Solanaceae and other families, i.e. pistils of self-incompatible species, or self-compatible taxa closely related to self-incompatible species, inhibit pollen tubes of self-compatible species. Conclusions Unilateral incompatibility in Capsicum does not seem to have arisen to prevent introgression of self-compatibility into self-incompatible taxa, but as a by-product of divergence of the C. pubescens complex from the remainder of the genus. We examined the effect of the invasive Solanum elaeagnifolium (Solanaceae) on flower visitation patterns and seed set of the co-flowering native Glaucium flavum (Papaveraceae). We observed flowering G. flavum plants in invaded and uninvaded sites and found that G. flavum flowers in uninvaded sites received significantly more total visits. In addition, we hand-pollinated flowers on plants of G. flavum with (i) pure conspecific pollen, (ii) pure S. elaeagnifolium pollen and (iii) three different mixtures of the two types of pollen (containing 25, 50 and 75% invasive pollen). As a control, flowers were left unmanipulated or were permanently bagged. Seed set did not differ significantly between flowers receiving pollen mixtures and pure conspecific pollen. However, in the open pollination treatment, seed set was significantly lower than in the 100% conspecific pollen treatment, which suggests pollen limitation. Bagged flowers had very low seed set. G. flavum was generally resilient against the deposition of S. elaeagnifolium pollen.

Background: There are compelling economic and environmental reasons to reduce our reliance on inorganic phosphate (Pi) fertilisers. Better management of Pi fertiliser applications is one option to improve the efficiency of Pi fertiliser use, whilst maintaining crop yields. Application rates of Pi fertilisers are traditionally determined from analyses of soil or plant tissues. Alternatively, diagnostic genes with altered expression under Pi limiting conditions that suggest a physiological requirement for Pi fertilisation, could be used to manage Pifertiliser applications, and might be more precise than indirect measurements of soil or tissue samples. Results: We grew potato (Solanum tuberosum L.) plants hydroponically, under glasshouse conditions, to control their nutrient status accurately. Samples of total leaf RNA taken periodically after Pi was removed from the nutrient solution were labelled and hybridised to potato oligonucleotide arrays. A total of 1,659 genes were significantly differentially expressed following Pi withdrawal. These included genes that encode proteins involved in lipid, protein, and carbohydrate metabolism, characteristic of Pi deficient leaves and included potential novel roles for genes encoding patatin like proteins in potatoes. The array data were analysed using a support vector machine algorithm to identify groups of genes that could predict the Pi status of the crop. These groups of diagnostic genes were tested using field grown potatoes that had either been fertilised or unfertilised. A group of 200 genes could correctly predict the Pi status of field grown potatoes. Conclusions: This paper provides a proof-of-concept demonstration for using microarrays and class prediction tools to predict the Pi status of a field grown potato crop. There is potential to develop this technology for other biotic and abiotic stresses in field grown crops. Ultimately, a better understanding of crop stresses may improve our management of the crop, improving the sustainability of agriculture.

Eukaryotic genome expansion/retraction caused by LTR-retrotransposon activity is dependent on the expression of full length copies to trigger efficient transposition and recombination-driven events. The Tnt1 family of retrotransposons has served as a model to evaluate the diversity among closely related elements within Solanaceae species and found that members of the family vary mainly in their U3 region of the long terminal repeats (LTRs). Recovery of a full length genomic copy of Retrosol was performed through a PCR-based approach from wild potato, Solanum oplocense. Further characterization focusing on both LTR sequences of the amplified copy allowed estimating an approximate insertion time at 2 million years ago thus supporting the occurrence of transposition cycles after genus divergence. Copy number of Tnt1-like elements in Solanum species were determined through genomic quantitative PCR whereby results sustain that Retrosol in Solanum species is a low copy number retrotransposon (1-4 copies) while Retrolyc1 has an intermediate copy number (38 copies) in S. peruvianum. Comparative analysis of retrotransposon content revealed no correlation between genome size or ploidy level and Retrosol copy number. The tetraploid cultivated potato with a cellular genome size of 1,715 Mbp harbours similar copy number per monoploid genome than other diploid Solanum species (613-884 Mbp). Conversely, S. peruvianum genome (1,125 Mbp) has a higher copy number. These results point towards a lineage specific dynamic flux regarding the history of amplification/activity of Tnt1-like elements in the genome of Solanum species.

O gênero Solanum L. (Solanaceae) compreende mais de 1000 espécies, incluindo táxons de grande interesse econômico por seu valor alimentício e medicinal. Este gênero é dividido em três subgêneros: Bassovia, Solanum e Leptostemonum. O subgênero Leptostemonum é dividido em dez seções, e entre essas destaca-se a seção Torva que possui representantes no sul do Brasil, e cujas espécies têm amplo interesse por apresentarem substâncias ativas de grande utilidade farmacológica. Entretanto, dentro dessa seção existem problemas taxonômicos, inclusive com a presença de indivíduos de morfologia intermediária, que dificultam sua classificação e, conseqüentemente, o seu melhor aproveitamento. Nesse trabalho, foram realizados dois estudos de caráter filogenético a fim de conhecer as relações de parentesco entre as espécies de Solanum seção Torva, presentes no sul do Brasil, e destas com espécies de outras seções do subgênero Leptostemonum. Em ambos os estudos foram utilizados quatro marcadores (genomas nuclear e plastidial): a região ITS (espaçadores internos transcritos do DNA nuclear ribossomal) incluindo ITS1, ITS2 e o gene 5,8S; o íntron trnL e os espaçadores intergênicos trnL-trnF e trnS-trnG do DNA plastidial. O marcador ISSR (Inter Simple Sequence Repeats) foi utilizado para verificar a variabilidade genética entre as espécies de Solanum seção Torva e testar o grau de polimorfismo de quatro “primers” dentro dessa seção. As análises realizadas evidenciaram uma origem monofilética para a seção Torva. Além disso, foi verificada uma relação de parentesco mais acentuado dessa seção com S. melongena, S. jamaicense e S. sisymbriifolium. Dentro da seção Torva foram observados agrupamentos que relacionam a espécie de morfologia intermediária a seus possíveis progenitores S. paniculatum e S. guaraniticum. Os quatro agrupamentos mais freqüentes observados dentro da seção foram: a aproximação de S. guaraniticum, S. bonariense e S. paniculatum X S. guaraniticum; o relacionamento entre S. adspersum e S. tabacifolium; a interação entre S. paniculatum e a espécie de morfologia intermediária; e a aproximação entre S. paniculatum e S. variabile. Este trabalho contribuiu para o conhecimento evolutivo das espécies dessa complexa seção que vem levantando interesse de inúmeros pesquisadores.

A cultura da bananeira é de grande importância económica para a Ilha da Madeira. A procura de formas de agricultura mais sustentáveis, como é o caso da agricultura biológica, tem levado ao desenvolvimento de estratégias alternativas aos produtos fitofarmacêuticos que possam ser utilizadas pelos agricultores. O objectivo geral deste trabalho foi o contribuir para a agricultura da Madeira desenvolvendo uma estratégia de luta contra o nemátode-das-lesões-radiculares, Pratylenchus goodeyi, utilizando Solanum sisymbriifolium e S. nigrum como cultura armadilha ou como biofumigante. Os isolados de P. goodeyi foram obtidos a partir da cultura in vitro em discos de cenoura e de raízes de bananeira infectadas pelo nemátode. Apesar de P. goodeyi se ter reproduzido nas plantas de S. sisymbriifolium e de S. nigrum, os factores de reprodução foram muito baixos (0,001), podendo estas plantas serem consideradas como resistentes ou, pelo menos, hospedeiros fracos. Os efeitos da incorporação, de diferentes partes de S. sisymbriifolium e S. nigrum (raízes, raízes+parte aérea e parte aérea), no solo com plantas de bananeira, infectadas com P. goodeyi, foram avaliados em relação ao crescimento das plantas de bananeira e à reprodução do nemátode. Verificou-se que qualquer uma das espécies de Solanum influenciou o crescimento das plantas de bananeira, principalmente daquelas em que as raízes foram incorporadas no solo. A reprodução de P. goodeyi nas plantas de bananeira, apenas infectadas com o nemátode, foi superior à sua reprodução nas plantas em que ocorreu incorporação no solo. Relativamente aos efeitos dos extractos aquosos das plantas de Solanum na mortalidade de P. goodeyi concluiu-se que os extractos de S. sisymbriifolium foram os mais eficazes, principalmente na concentração de 250 mg/ml. De acordo com os resultados obtidos, conclui-se que as plantas S. sisymbriifolium e S. nigrum poderão ser utilizadas como biofumigante e como adubo verde, contribuindo para melhorar a qualidade e a quantidade das produções, eliminando os efeitos negativos, de produtos de origem química, sobre a saúde e o ambiente.

The control of Pratylenchus goodeyi a common nematode parasite of banana crop in Madeira Island can benefit from searching for natural nematicides through plants extracts. With this aim we submitted Solanum nigrum and S. sisymbriifolium dried plants to a sequential extraction in the solvent sequence of dichloromethane, acetone, ethanol and water, and to na aqueous extraction of the fresh and dried plants. Analyses with the extracts at several concentrations were used to assess mobility and mortality on P. goodeyi. Results showed that the water extract and aqueous extracts from both plants at a concentration of 10 mg/mL affected nematode mobility and caused mortality but the acetone extract from S. nigrum was the most efficient, causing 100% mortality whereas dichloromethane had no effect on P. goodeyi. Determination of the lipophilic and phenolic compounds present in the two most effective Solanum extracts (acetone and water) and in dichloromethane extract revealed that some of these compounds had nematicidal activity. S. nigrum acetone extract (10 mg/mL) was used to find out the nematicidal potential following the effect at gene expression level and nematode behaviour. Genes coding for calreticulin and beta-1,4- endoglucanase related to parasitism and translocon-associated protein putatively connected to stress were obtained and its relative expression assessed in nematodes exposed to the extract. Results revealed that expression of Pg-CRT decreased showing to influence the infection, Pg-ENG remained steady and Pg-TRAPδ was induced over time exposure. Biological assays showed that P. goodeyi mobility and ability to infect the banana roots were affected as a decrease in the number of nematodes that reached the roots was obtained with the increased exposure time to the extract being implicated in the infection success. The information obtained from this thesis showed that S. nigrum has potential to be used for the development of a new control strategy against plant-parasitic nematodes.
