Tutkielmassa selvitetään englanti lingua francana (ELF) -näkökulman ilmenemistä kulttuurin ja sen opettamisen periaatteissa. Tarkastelun kohteina ovat viimeaikaiset kielenopetuksen ohjenuoria käsittelevät ohjelmat Euroopassa ja Suomessa: Euroopan neuvoston julkaisema Eurooppalainen viitekehys ja Suomen perus- ja lukio-opetuksen uusimmat vieraiden kielten opetussuunnitelmat. Tutkielmassa sovelletaan kriittisen diskurssianalyysin kolmiportaista analyysikehystä, jonka kuvausosiossa Viitekehyksestä analysoidaan esiin kulttuurisia diskursseja, tulkintaosiossa aineistoa tarkastellaan Euroopan neuvoston kielipolitiikan osana, ja selitysosiossa sitä verrataan opetussuunnitelmista hahmotettuihin diskursseihin. Viitekehyksestä nousi esiin neljä keskeistä diskurssia: 1) kohdekulttuuri-, 2) kulttuurien monimuotoisuus-, 3) monikulttuurisuus- ja 4) oppilaskeskeisyysdiskurssit. Näistä kahdessa viimeisessä oli piirteitä, joiden voi katsoa tukevan ELF-näkökulmaa. Tällaisia olivat mm. usean kulttuurin välillä liikkuminen ja oppilaiden tarpeiden korostaminen. Sen sijaan opetussuunnitelmissa keskitytään ELF:n kannalta liian kapea-alaisesti vain äidinkielisten ja ei-äidinkielisten väliseen viestintään. ELF-lähestymistavan kannalta olisikin tärkeää ymmärtää, että kansainvälisessä viestinnässä englannin kieltä ei voi yhdistää tiettyyn kohdekulttuuriin ja että monikielitaitoisuuden ja lingua francan ei tarvitse olla ristiriidassa keskenään.

The aim of this study was to build a model and analyze how users move in a virtual environment and to explore the experiential dimensions connected with different ways of moving. Due to the lack of previous research on this subject, this was an explorative study. This study also aimed to identify different ways how users move in virtual environments and the background variables connected to them. It was hypothesized that fluent movement in virtual environments is connected to high presence, skill and challenge assessments. Test participants (n = 68) were mostly highly educated young adults. A virtual environment was built using a CAVE -type virtual reality interface. The task was to search for objects that do not belong into a normal house. The participants movement in the virtual house was recorded on a computer. Movement was modelled using a cluster analysis of information entropy based movement measurements, acceleration, amount of stops and time spent being stationary. The experiential dimensions were measured using the EVEQ -questionnaire. We were able to identify four different ways of moving in virtual environments. In respect of background variables, the four groups differed only in the amount of weekly computer usage. However, fluent movement in virtual environments was connected to a high sense of presence. Furthermore, participants who moved fluently in the environment assessed their skills as being high and regarded the use of virtual environment as challenging. The results indicate that different ways of moving affects how people experience virtual environments. Consequently the participants assessment of their skills and level of challenge have an impact on the affective evaluation of the situation at hand. Entropy measures have not been previously applied when studying movement, and in addition the role of movement on the experiential dimensions of virtual environments is an unexplored subject. The movement analysis method introduced here is applicable to other research problems. Finally, this study expands on our knowledge of the special characteristics connected with the experiential dimensions of virtual environments.

It is demanding for children with visual impairment to become aware of the world beyond their immediate experience. They need to learn to control spatial experiences as a whole and understand the relationships between objects, surfaces and themselves. Tactile maps can be an excellent source of information for depicting space and environment. By means of tactile maps children can develop their spatial understanding more efficiently than through direct travel experiences supplemented with verbal explanations. Tactile maps can help children when they are learning to understand environmental, spatial, and directional concepts. The ability to read tactile maps is not self-evident; it is a skill, which must be learned. The main research question was: can children who are visually impaired learn to read tactile maps at the preschool age if they receive structural teaching? The purpose of this study was to develop an educational program for preschool children with visual impairment, the aim of which was to teach them to read tactile maps in order to strengthen their orientation skills and to encourage them to explore the world beyond their immediate experience. The study is a multiple case study describing the development of the map program consisting of eight learning tasks. The program was developed with one preschooler who was blind, and subsequently the program was implemented with three other children. Two of the children were blind from birth, one child had lost her vision at the age of two, and one child had low vision. The program was implemented in a normal preschool. Another objective of the pre-map program was to teach the preschooler with visual impairment to understand the concept of a map. The teaching tools were simple, map-like representations called pre-maps. Before a child with visual impairment can read a comprehensive tactile map, it is important to learn to understand map symbols, and how a three-dimensional model changes to a two-dimensional tactile map. All teaching sessions were videotaped; the results are based on the analysis of the videotapes. Two of the children completed the program successfully, and learned to read a tactile map. The two other children felt happy during the sessions, but it was problematic for them to engage fully in the instruction. One of the two eventually completed the program, while the other developed predominantly emerging skills. The results of the children's performances and the positive feedback from the teachers, assistants and the parents proved that this pre-map program is appropriate teaching material for preschool children who are visually impaired. The program does not demand high-level expertise; also parents, preschool teachers, and school assistants can carry out the program.

Rhizoremediation is the use of microbial populations present in the rhizosphere of plants for environmental cleanup. The idea of this work was that bacteria living in the rhizosphere of a nitrogen-fixing leguminous plant, goat's rue (Galega orientalis), could take part in the degradation of harmful monoaromatic hydrocarbons, such as benzene, toluene and xylene (BTEX), from oil-contaminated soils. In addition to chemical (e.g. pollutant concentration) and physical (e.g. soil structure) information, the knowledge of biological aspects (e.g. bacteria and their catabolic genes) is essential when developing the rhizoremediation into controlled and effective bioremediation practice. Therefore, the need for reliable biomonitoring methods is obvious. The main aims of this thesis were to evaluate the symbiotic G. orientalis - Rhizobium galegae system for rhizoremediation of oil-contaminated soils, to develop molecular methods for biomonitoring, and to apply these methods for studying the microbiology of rhizoremediation. In vitro, Galega plants and rhizobia remained viable in m-toluate concentrations up to 3000 mg/l. Plant growth and nodulation were inhibited in 500 mg/l m-toluate, but were restored when plants were transferred to clean medium. In the greenhouse, Galega showed good growth, nodulation and nitrogen fixation, and developed a strong rhizosphere in soils contaminated with oil or spiked with 2000 mg/l m-toluate. The high aromatic tolerance of R. galegae and the viability of Galega plants in oil-polluted soils proved this legume system to be a promising method for the rhizoremediation of oil-contaminated soils. Molecular biomonitoring methods were designed and/or developed further for bacteria and their degradation genes. A combination of genomic fingerprinting ((GTG)5-PCR), taxonomic ribotyping of 16S rRNA genes and partial 16S rRNA gene sequencing were chosen for molecular grouping of culturable, heterogeneous rhizosphere bacteria. PCR primers specific for the xylE gene were designed for TOL plasmid detection. Amplified enzyme-coding DNA restriction analysis (AEDRA) with AluI was used to profile both TOL plasmids (xylE primers) and, in general, aromatics-degrading plasmids (C230 primers). The sensitivity of the direct monitoring of TOL plasmids in soil was enhanced by nested C23O-xylE-PCR. Rhizosphere bacteria were isolated from the greenhouse and field lysimeter experiments. High genetic diversity was observed among the 50 isolated, m-toluate tolerating rhizosphere bacteria in the form of five major lineages of the Bacteria domain. Gram-positive Rhodococcus, Bacillus and Arthrobacter and gram-negative Pseudomonas were the most abundant genera. The inoculum Pseudomonas putida PaW85/pWW0 was not found in the rhizosphere samples. Even if there were no ecological niches available for the bioaugmentation bacterium itself, its conjugative catabolic plasmid might have had some additional value for other bacterial species and thus, for rhizoremediation. Only 10 to 20% of the isolated, m-toluate tolerating bacterial strains were also able to degrade m-toluate. TOL plasmids were a major group of catabolic plasmids among these bacteria. The ability to degrade m-toluate by using enzymes encoded by a TOL plasmid was detected only in species of the genus Pseudomonas, and the best m-toluate degraders were these Pseudomonas species. Strain-specific differences in degradation abilities were found for P.oryzihabitans and P. migulae: some of these strains harbored a TOL plasmid - a new finding observed in this work, indicating putative horizontal plasmid transfer in the rhizosphere. One P. oryzihabitans strain harbored the pWW0 plasmid that had probably conjugated from the bioaugmentation Pseudomonas. Some P. migulae and P. oryzihabitans strains seemed to harbor both the pWW0- and the pDK1-type TOL plasmid. Alternatively, they might have harbored a TOL plasmid with both the pWW0- and the pDK1-type xylE gene. The breakdown of m-toluate by gram-negative bacteria was not restricted to the TOL pathway. Also some gram-positive Rhodococcus erythropolis and Arthrobacter aurescens strains were able to degrade m-toluate in the absence of a TOL plasmid. Three aspects of the rhizosphere effect of G. orientalis were manifested in oil-contaminated soil in the field: 1) G. orientalis and Pseudomonas bioaugmentation increased the amount of rhizosphere bacteria. G. orientalis especially together with Pseudomonas bioaugmentation increased the numbers of m-toluate utilizing and catechol positive bacteria indicating an increase in degradation potential. 2) Also the bacterial diversity, when measured as the amount of ribotypes, was increased in the Galega rhizosphere with or without Pseudomonas bioaugmentation. However, the diversity of m-toluate utilizing bacteria did not significantly increase. At the community level, by using the 16S rRNA gene PCR-DGGE method, the highest diversity of species was also observed in vegetated soils compared with non-vegetated soils. Diversified communities may best guarantee the overall success in rhizoremediation by offering various genetic machineries for catabolic processes. 3) At the end of the experiment, no TOL plasmid could be detected by direct DNA analysis in soil treated with both G. orientalis and Pseudomonas. The detection limit for TOL plasmids was encountered indicating decreased amount of degradation plasmids and thus, the success of rhizoremediation. The use of G. orientalis for rhizoremediation is unique. In this thesis new information was obtained about the rhizosphere effect of Galega orientalis in BTEX contaminated soils. The molecular biomonitoring methods can be applied for several purposes within environmental biotechnology, such as for evaluating the intrinsic biodegradation potential, monitoring the enhanced bioremediation, and estimating the success of bioremediation. Environmental protection by using nature's own resources and thus, acting according to the principle of sustainable development, would be both economically and environmentally beneficial for society. The study examines various uses of computer technology in acquisition of information for visually impaired people. For this study 29 visually impaired persons took part in a survey about their experiences concerning acquisition of infomation and use of computers, especially with a screen magnification program, a speech synthesizer and a braille display. According to the responses, the evolution of computer technology offers an important possibility for visually impaired people to cope with everyday activities and interacting with the environment. Nevertheless, the functionality of assistive technology needs further development to become more usable and versatile. Since the challenges of independent observation of environment were emphasized in the survey, the study led into developing a portable text vision system called Tekstinäkö. Contrary to typical stand-alone applications, Tekstinäkö system was constructed by combining devices and programs that are readily available on consumer market. As the system operates, pictures are taken by a digital camera and instantly transmitted to a text recognition program in a laptop computer that talks out loud the text using a speech synthesizer. Visually impaired test users described that even unsure interpretations of the texts in the environment given by Tekstinäkö system are at least a welcome addition to complete perception of the environment. It became clear that even with a modest development work it is possible to bring new, useful and valuable methods to everyday life of disabled people. Unconventional production process of the system appeared to be efficient as well. Achieved results and the proposed working model offer one suggestion for giving enough attention to easily overlooked needs of the people with special abilities. Ongoing habitat loss and fragmentation threaten much of the biodiversity that we know today. As such, conservation efforts are required if we want to protect biodiversity. Conservation budgets are typically tight, making the cost-effective selection of protected areas difficult. Therefore, reserve design methods have been developed to identify sets of sites, that together represent the species of conservation interest in a cost-effective manner. To be able to select reserve networks, data on species distributions is needed. Such data is often incomplete, but species habitat distribution models (SHDMs) can be used to link the occurrence of the species at the surveyed sites to the environmental conditions at these locations (e.g. climatic, vegetation and soil conditions). The probability of the species occurring at unvisited location is next predicted by the model, based on the environmental conditions of those sites. The spatial configuration of reserve networks is important, because habitat loss around reserves can influence the persistence of species inside the network. Since species differ in their requirements for network configuration, the spatial cohesion of networks needs to be species-specific. A way to account for species-specific requirements is to use spatial variables in SHDMs. Spatial SHDMs allow the evaluation of the effect of reserve network configuration on the probability of occurrence of the species inside the network. Even though reserves are important for conservation, they are not the only option available to conservation planners. To enhance or maintain habitat quality, restoration or maintenance measures are sometimes required. As a result, the number of conservation options per site increases. Currently available reserve selection tools do however not offer the ability to handle multiple, alternative options per site. This thesis extends the existing methodology for reserve design, by offering methods to identify cost-effective conservation planning solutions when multiple, alternative conservation options are available per site. Although restoration and maintenance measures are beneficial to certain species, they can be harmful to other species with different requirements. This introduces trade-offs between species when identifying which conservation action is best applied to which site. The thesis describes how the strength of such trade-offs can be identified, which is useful for assessing consequences of conservation decisions regarding species priorities and budget. Furthermore, the results of the thesis indicate that spatial SHDMs can be successfully used to account for species-specific requirements for spatial cohesion - in the reserve selection (single-option) context as well as in the multi-option context. Accounting for the spatial requirements of multiple species and allowing for several conservation options is however complicated, due to trade-offs in species requirements. It is also shown that spatial SHDMs can be successfully used for gaining information on factors that drive a species spatial distribution. Such information is valuable to conservation planning, as better knowledge on species requirements facilitates the design of networks for species persistence. This methods and results described in this thesis aim to improve species probabilities of persistence, by taking better account of species habitat and spatial requirements. Many real-world conservation planning problems are characterised by a variety of conservation options related to protection, restoration and maintenance of habitat. Planning tools therefore need to be able to incorporate multiple conservation options per site, in order to continue the search for cost-effective conservation planning solutions. Simultaneously, the spatial requirements of species need to be considered. The methods described in this thesis offer a starting point for combining these two relevant aspects of conservation planning.

The actin cytoskeleton is essential for many cellular processes, including motility, morphogenesis, endocytosis and signal transduction. Actin can exist in monomeric (G-actin) or filamentous (F-actin) form. Actin filaments are considered to be the functional form of actin, generating the protrusive forces characteristic for the actin cytoskeleton. The structure and dynamics of the actin filament and monomer pools are regulated by a large number of actin-binding proteins in eukaryotic cells. Twinfilin is an evolutionarily conserved small actin monomer binding protein. Twinfilin is composed of two ADF/cofilin-like domains, separated by a short linker and followed by a C-terminal tail. Twinfilin forms a stable, high affinity complex with ADP-G-actin, inhibits the nucleotide exchange on actin monomers, and prevents their assembly into filament ends. Twinfilin was originally identified from yeast and has since then been found from all organisms studied except plants. Not much was known about the role of twinfilin in the actin dynamics in mammalian cells before this study. We set out to unravel the mysteries still covering twinfilins functions using biochemistry, cell biology, and genetics. We identified and characterized two mouse isoforms for the previously identified mouse twinfilin-1. The new isoforms, twinfilin-2a and -2b, are generated from the same gene through alternative promoter usage. The three isoforms have distinctive expression patterns, but are similar biochemically. Twinfilin-1 is the major isoform during development and is expressed in high levels in almost all tissues examined. Twinfilin-2a is also expressed almost ubiquitously, but at lower levels. Twinfilin-2b turned out to be a muscle-specific isoform, with very high expression in heart and skeletal muscle. It seems all mouse tissues express at least two twinfilin isoforms, indicating that twinfilins are important regulators of actin dynamics in all cell and tissue types. A knockout mouse line was generated for twinfilin-2a. The mice homozygous for this knockout were viable and developed normally, indicating that twinfilin-2a is dispensable for mouse development. However, it is important to note that twinfilin-2a shows similar expression pattern to twinfilin-1, suggesting that these proteins play redundant roles in mice. All mouse isoforms were shown to be able to sequester actin filaments and have higher affinity for ADP-G-actin than ATP-G-actin. They are also able to directly interact with heterodimeric capping protein and PI(4,5)P2 similar to yeast twinfilin. In this study we also uncovered a novel function for mouse twinfilins; capping actin filament barbed ends. All mouse twinfilin isoforms were shown to possess this function, while yeast and Drosophila twinfilin were not able to cap filament barbed ends. Twinfilins localize to the cytoplasm but also to actin-rich regions in mammalian cells. The subcellular localizations of the isoforms are regulated differently, indicating that even though twinfilins biochemical functions in vitro are very similar, in vivo they can play different roles through different regulatory pathways. Together, this study show that twinfilins regulate actin filament assembly both by sequestering actin monomers and by capping filament barbed ends, and that mammals have three biochemically similar twinfilin isoforms with partially overlapping expression patterns.

The adequacy of anesthesia has been studied since the introduction of balanced general anesthesia. Commercial monitors based on electroencephalographic (EEG) signal analysis have been available for monitoring the hypnotic component of anesthesia from the beginning of the 1990s. Monitors measuring the depth of anesthesia assess the cortical function of the brain, and have gained acceptance during surgical anesthesia with most of the anesthetic agents used. However, due to frequent artifacts, they are considered unsuitable for monitoring consciousness in intensive care patients. The assessment of analgesia is one of the cornerstones of general anesthesia. Prolonged surgical stress may lead to increased morbidity and delayed postoperative recovery. However, no validated monitoring method is currently available for evaluating analgesia during general anesthesia. Awareness during anesthesia is caused by an inadequate level of hypnosis. This rare but severe complication of general anesthesia may lead to marked emotional stress and possibly posttraumatic stress disorder. In the present series of studies, the incidence of awareness and recall during outpatient anesthesia was evaluated and compared with that of in inpatient anesthesia. A total of 1500 outpatients and 2343 inpatients underwent a structured interview. Clear intraoperative recollections were rare the incidence being 0.07% in outpatients and 0.13% in inpatients. No significant differences emerged between outpatients and inpatients. However, significantly smaller doses of sevoflurane were administered to outpatients with awareness than those without recollections (p<0.05). EEG artifacts in 16 brain-dead organ donors were evaluated during organ harvest surgery in a prospective, open, nonselective study. The source of the frontotemporal biosignals in brain-dead subjects was studied, and the resistance of bispectral index (BIS) and Entropy to the signal artifacts was compared. The hypothesis was that in brain-dead subjects, most of the biosignals recorded from the forehead would consist of artifacts. The original EEG was recorded and State Entropy (SE), Response Entropy (RE), and BIS were calculated and monitored during solid organ harvest. SE differed from zero (inactive EEG) in 28%, RE in 29%, and BIS in 68% of the total recording time (p<0.0001 for all). The median values during the operation were SE 0.0, RE 0.0, and BIS 3.0. In four of the 16 organ donors, EEG was not inactive, and unphysiologically distributed, nonreactive rhythmic theta activity was present in the original EEG signal. After the results from subjects with persistent residual EEG activity were excluded, SE, RE, and BIS differed from zero in 17%, 18%, and 62% of the recorded time, respectively (p<0.0001 for all). Due to various artifacts, the highest readings in all indices were recorded without neuromuscular blockade. The main sources of artifacts were electrocauterization, electromyography (EMG), 50-Hz artifact, handling of the donor, ballistocardiography, and electrocardiography. In a prospective, randomized study of 26 patients, the ability of Surgical Stress Index (SSI) to differentiate patients with two clinically different analgesic levels during shoulder surgery was evaluated. SSI values were lower in patients with an interscalene brachial plexus block than in patients without an additional plexus block. In all patients, anesthesia was maintained with desflurane, the concentration of which was targeted to maintain SE at 50. Increased blood pressure or heart rate (HR), movement, and coughing were considered signs of intraoperative nociception and treated with alfentanil. Photoplethysmographic waveforms were collected from the contralateral arm to the operated side, and SSI was calculated offline. Two minutes after skin incision, SSI was not increased in the brachial plexus block group and was lower (38 ± 13) than in the control group (58 ± 13, p<0.005). Among the controls, one minute prior to alfentanil administration, SSI value was higher than during periods of adequate antinociception, 59 ± 11 vs. 39 ± 12 (p<0.01). The total cumulative need for alfentanil was higher in controls (2.7 ± 1.2 mg) than in the brachial plexus block group (1.6 ± 0.5 mg, p=0.008). Tetanic stimulation to the ulnar region of the hand increased SSI significantly only among patients with a brachial plexus block not covering the site of stimulation. Prognostic value of EEG-derived indices was evaluated and compared with Transcranial Doppler Ultrasonography (TCD), serum neuron-specific enolase (NSE) and S-100B after cardiac arrest. Thirty patients resuscitated from out-of-hospital arrest and treated with induced mild hypothermia for 24 h were included. Original EEG signal was recorded, and burst suppression ratio (BSR), RE, SE, and wavelet subband entropy (WSE) were calculated. Neurological outcome during the six-month period after arrest was assessed with the Glasgow-Pittsburgh Cerebral Performance Categories (CPC). Twenty patients had a CPC of 1-2, one patient had a CPC of 3, and nine patients died (CPC 5). BSR, RE, and SE differed between good (CPC 1-2) and poor (CPC 3-5) outcome groups (p=0.011, p=0.011, p=0.008, respectively) during the first 24 h after arrest. WSE was borderline higher in the good outcome group between 24 and 48 h after arrest (p=0.050). All patients with status epilepticus died, and their WSE values were lower (p=0.022). S-100B was lower in the good outcome group upon arrival at the intensive care unit (p=0.010). After hypothermia treatment, NSE and S-100B values were lower (p=0.002 for both) in the good outcome group. The pulsatile index was also lower in the good outcome group (p=0.004). In conclusion, the incidence of awareness in outpatient anesthesia did not differ from that in inpatient anesthesia. Outpatients are not at increased risk for intraoperative awareness relative to inpatients undergoing general anesthesia. SE, RE, and BIS showed non-zero values that normally indicate cortical neuronal function, but were in these subjects mostly due to artifacts after clinical brain death diagnosis. Entropy was more resistant to artifacts than BIS. During general anesthesia and surgery, SSI values were lower in patients with interscalene brachial plexus block covering the sites of nociceptive stimuli. In detecting nociceptive stimuli, SSI performed better than HR, blood pressure, or RE. BSR, RE, and SE differed between the good and poor neurological outcome groups during the first 24 h after cardiac arrest, and they may be an aid in differentiating patients with good neurological outcomes from those with poor outcomes after out-of-hospital cardiac arrest.

To a large extent, lakes can be described with a one-dimensional approach, as their main features can be characterized by the vertical temperature profile of the water. The development of the profiles during the year follows the seasonal climate variations. Depending on conditions, lakes become stratified during the warm summer. After cooling, overturn occurs, water cools and an ice cover forms. Typically, water is inversely stratified under the ice, and another overturn occurs in spring after the ice has melted. Features of this circulation have been used in studies to distinguish between lakes in different areas, as basis for observation systems and even as climate indicators. Numerical models can be used to calculate temperature in the lake, on the basis of the meteorological input at the surface. The simple form is to solve the surface temperature. The depth of the lake affects heat transfer, together with other morphological features, the shape and size of the lake. Also the surrounding landscape affects the formation of the meteorological fields over the lake and the energy input. For small lakes the shading by the shores affects both over the lake and inside the water body bringing limitations for the one-dimensional approach. A two-layer model gives an approximation for the basic stratification in the lake. A turbulence model can simulate vertical temperature profile in a more detailed way. If the shape of the temperature profile is very abrupt, vertical transfer is hindered, having many important consequences for lake biology. One-dimensional modelling approach was successfully studied comparing a one-layer model, a two-layer model and a turbulence model. The turbulence model was applied to lakes with different sizes, shapes and locations. Lake models need data from the lakes for model adjustment. The use of the meteorological input data on different scales was analysed, ranging from momentary turbulent changes over the lake to the use of the synoptical data with three hour intervals. Data over about 100 past years were used on the mesoscale at the range of about 100 km and climate change scenarios for future changes. Increasing air temperature typically increases water temperature in epilimnion and decreases ice cover. Lake ice data were used for modelling different kinds of lakes. They were also analyzed statistically in global context. The results were also compared with results of a hydrological watershed model and data from very small lakes for seasonal development.

Peruna kestää A-virusta estämällä sen leviämistä Peruna on maissin ohella maailman kolmanneksi tärkein ravintokasvi vehnän ja riisin jälkeen. Perunaa lisätään kasvullisesti mukuloita istuttamalla, jolloin virukset siirtyvät sairaiden siemenmukuloiden välityksellä kasvukaudesta toiseen. Virustauteja voi torjua ainoastaan terveen siemenperunan ja kestävien lajikkeiden avulla. Kestävyys perustuu usein siihen, että kasvi estää viruksen leviämisen tartuntakohdasta välttyäkseen virustaudilta. Tässä työssä tutkittiin kolmea perunan A-viruksen (PVA) liikkumista estävää kestävyysmekanismia perunassa. Lisäksi työn kokeelliseen osaan oleellisesti kuuluvaa virustartutusta varten kehitettiin uusi paranneltu versio geenipyssystä. Tämä itse rakennettu laite optimoitiin PVA:n tartuttamiseen mahdollisimman helposti ja pienin käyttökustannuksin. Tutkimuksen kohteena olleessa perunan risteytysjälkeläistössä oli PVA:ta kestäviä kasveja (ryhmä nnr), jotka estivät viruksen liikkumisen aiheuttamatta oireita tartutuskohdassa, sekä kasveja, joissa PVA aiheutti kuolioläikkinä näkyvän yliherkkyysvasteen (ryhmä HR). Molemmissa kestävyystyypeissä virus pystyi monistumaan ja leviämään solusta soluun paikallisesti, mutta liikkuminen muihin kasvinosiin nilan kautta estyi. Ryhmän nnr kasveissa PVA-tartunta ei aiheuttanut tilastollisesti merkitsevää muutosta useimpien geenien ilmenemiseen tartuntakohdassa. Ainoastaan geeniperhe, joka ilmentää tiettyä proteinaasi-inhibiittoria (PI), reagoi PVA:han 24 tuntia tartutuksesta. Kun tämän PVA:han reagoivan geeniperheen jäsenet hiljennettiin nnr- perunalinjoissa, ne muuttuivat alttiiksi PVA:lle ja virus levisi tartuntakohdasta muihin kasvinosiin. Tulos osoittaa, että PI on viruskestävyystekijä. Lisäksi muut tutkimuksessa saadut tulokset tukevat mahdollisuutta, että PI estää PVA:n P1-proteinaasin toimintaa. HR-linjoissa todettiin erilaisiin puolustusvasteisiin liittyvien PR-geenien aktivoitumista PVA-tartunnan seurauksena, mutta myös ilman sitä kasvien kasvettua mullassa noin neljä viikkoa. Sen sijaan solukkoviljelyssä tai vasta kaksi viikkoa mullassa kasvaneissa kasveissa vastaavaa ei vielä todettu. Tulos viittaa siihen, että HR-perunat reagoivat herkemmin ympäristöön ja/tai kasvin kehitysasteeseen laukaisten puolustusvasteita, jotka saattavat parantaa kestävyyttä taudinaiheuttajia vastaan. Kolmas tutkittu kestävyystyyppi havaittiin Pito-perunalajikkeessa. Se muistutti nnr-kestävyyttä siten, että myös siinä viruksen liikkuminen nilassa muihin kasvinosiin estyi. PVA:n todettiin pysähtyvän vasta lehtiruodin tyvelle muodostuvaan irtoamisvyöhykkeeseen, mitä havainnollistettiin käyttämällä muunnettua PVA-rotua, joka tuotti UV-valossa fluoresoivaa vihreää valoa. Tulos viittaa siihen, että virus ei pääse kulkemaan vyöhykkeeseen kuuluvan suojaavan kerroksen läpi, jollei sillä ole pääsyä nilaan. Tällainen kestävyys on tarpeen, jotta virus ei voi korvata nilakuljetusta solusta soluun leviämisellä. Tulokset tuovat uusia näkökulmia kasvien viruskestävyyteen ja auttavat selittämään viruksen nilakuljetuksen estymistä sekä solusta soluun leviämisen pysähtymistä kestävissä kasveissa.

Hyvinkääläiset työläisnaiset ottivat vuoden 1918 sisällissotaan osaa punakaartin riveissä ruoanlaittajina, sairaanhoitajina sekä asetta kantaneina sotilaina. Valkokaartia tukemaan saapuneet saksalaissotilaat saavuttivat Hyvinkään huhtikuun 20. päivä, ja pitäjä valloitettiin. Osa hyvinkääläisnaisista oli tässä vaiheessa siirtynyt jo pohjoisemmille rintamille. Valkoiset etenivät kuitenkin voittoisasti kohti pohjoista, ja suurin osa hyvinkääläisnaisista oli antautunut 1. toukokuuta mennessä. Tämä tutkimus tarkastelee Hyvinkään punakaartilaisnaisiin kohdistuneita kuulustelututkimuksia ja toimenpiteitä sisällissodan jälkiselvittelyissä, jotka alkoivat huhtikuussa ja jatkuivat vuoden loppuun saakka. Samalla tuon esiin runsaasti uutta tietoa Hyvinkään punakaartilaisnaisten toimista sisällissodassa sekä heidän taustoistaan. Tärkeimpiä lähteitä tutkimuksessani ovat erilaiset Kansallisarkistossa säilytettävät oikeusasiakirjat: Hyvinkään pitäjän kenttäoikeuksien pöytäkirjat, viralliset esitutkintapöytäkirjat sekä valtiorikosoikeuksien ja valtiorikosylioikeuden aktit. Lisäksi olen etsinyt tietoa sodassa ja sen jälkiselvittelyissä surmansa saaneista punaisista hyvinkääläisnaisista. Tässä tärkeimpinä lähteinäni ovat olleet Suomen Sotasurmat 1914–1922 -projektin nimitiedosto sekä valkokaartin kirjanpidosta löytyneet vangitsemistiedot. Olen hyödyntänyt suuren tietomäärän käsittelyssä tilastollisia menetelmiä. Tarkastelen naisia kuitenkin paljon myös yksilötasolla, sillä juuri yksilötarinat rikastuttavat paikallishistoriallista tutkimusta. Tutkimukseni perusteella punaisiksi luokiteltuja hyvinkääläisnaisia menehtyi sisällissodan jälkiselvittelyissä 27 – heistä suurin osa Lahden ja Hämeenlinnan seudulla, missä viimeiset taistelut käytiin. Menehtyneistä naisista moni oli vasta 16–17-vuotias. Myös hyvinkääläisnaisia joutui suurille vankileireille – tässä yhteydessä tarkastelen kahden naisen vankileirimuistelmia. Kotipaikkakunnalla tutkittiin samanaikaisesti kenttäoikeuksissa 64 hyvinkääläisnaista. Kenttäoikeuksien tehtävänä oli kartoittaa punakaartilaisten toimintaa sekä heidän tietojansa paikkakunnalla ja lähiseuduilla tapahtuneista murhista sekä ryöstöistä. Kenttäoikeuksissa kuulustelluista naisista valtaosa vapautettiin lyhyen vankeuden jälkeen. Jälkiselvittelyjen virallisessa vaiheessa 116 hyvinkääläistä punakaartilaisnaista joutui kuulusteluihin. Heistä vajaa puolet oli toiminut sodan aikana erilaisissa huoltotehtävissä mm. punakaartin ruokalassa. 25 naista oli epäiltynä Hyvinkäällä huhtikuussa perustetussa aseellisessa naiskaartissa toimimisesta. Loput olivat toimineet punaisten hallinnossa tai sairaanhoitotehtävissä kotipaikkakunnalla ja rintamilla. Kuulustelujen perusteella 81 naista sai vuoden 1918 jälkipuoliskolla syytteen valtiorikosoikeudessa; heistä suurin osa avunannosta valtiopetokseen. Tutkimukseni kattaa näin kaikkiaan noin 150 hyvinkääläistä punakaartilaisnaista. Tutkimukseni valottaa heidän toimintaansa sodan aikana sekä arvioi rankaisutoimenpiteiden motiiveja. Tutkimukseni mukaan asetta kantaneisiin naisiin suhtauduttiin jälkiselvittelyissä keskimäärin hyvin ankarasti. Muiden punakaartilaisnaisten osalta rankaisutoimenpiteissä ei ollut yhtenäistä linjaa. Perunankuorinta punakaartin ruokalassa saattoi olla tuomittava rikos siinä missä kaartilaisten matkoilla liikkuminen ja ryöstetyn tavaran kätkeminen.

Tämä tutkimus tarkastelee pastori Väinö Kanteleen työtä lähetystyöntekijänä Kiinassa hänen kolmannella työkaudellaan 1948 - 1951. Tutkimuksen keskeisimmät lähteet ovat Kanteleen perheelleen osoittamat päiväkirjamaiset kirjeet sekä Suomen Lähetysseuran (SLS) johtokunnan kokousten ja lähettien kokousten pöytäkirjat. Tutkimus on mikrohistoriallinen sivuten henkilö- ja lähetyshistoriaa. Kantele saapui Yungšunin asemalle maaliskuussa 1948. Asemalle oli kaivattu kokenutta mieslähettiä. Työkauden alku sujui hyvin Kiinan poliittisen tilanteen levottomuudesta huolimatta. Tilanne kuitenkin kiristyi kommunistien saadessa lisää kannatusta. SLS kutsui lähetit pois työalueelta keväällä 1949. Kantele päätti jäädä Kiinaan. Yungšunin seurakunta oli toivonut hänen jäämistään. Lisäksi mahdollisuudet lähetystyön tekemiseen olivat yhä olemassa huolimatta Kiinan kansantasavallan perustamisesta lokakuussa 1949. Olosuhteet alkoivat kuitenkin asteittain kiristyä. Hajanaisten sotilasjoukkojen sekä kommunistiarmeijan liikkuminen ympäri Kiinaa häiritsivät lähetystyötä. Sotilaat muun muassa valtasivat lähetysasemia käyttöönsä. Ongelmana olivat myös inflaatio ja postin kulun epävarmuus. Henkilökohtaisesti Kanteletta vaivasi myös tunne oman uskonsa heikkoudesta. Vuonna 1950 Kiinan poliittiset uudistukset rajoittivat lähetystyötä entisestään. Heinäkuussa julkaistu Kirkkojen manifesti vaati kirkkojen pikaista irrottautumista ulkomaisista yhteyksistään, käytännössä lähetysjärjestöistä. Samana vuonna aloitettiin maareformi. Seurakuntiin asetettiin kokoontumiskielto reformin toteuttamisen ajaksi. Yungšunissa kieltoa ei kumottu enää Kanteleen työkauden aikana. Uudistusten myötä Kanteleen liikkumisvapautta rajoitettiin. Viranomaiset hakivat hänet välillä moneksi päiväksi kuulusteluihin. Lopulta hän eli käytännössä vankina asemallaan. Tammikuussa 1951 hän koki, ettei lähetystyön tekemiseen ole mahdollisuuksia, ja anoi maastapoistumislupaa viranomaisilta. Alkoi pitkä ja epävarma odotus. Kantele alkoi laulaa kiinankielisiä virsiä kovaan ääneen asemalla koska koki, että se oli ainoa jäljellä oleva keino levittää kristinuskon sanomaa. Tapansa vuoksi hänet tunnettiin laulavana evankelistana. Tie kotiin avautui lopulta heinäkuussa 1951. Kantele koki työnsä lähetyssaarnaajana tärkeäksi ja rakkaaksi. Hän oli valmis työskentelemään hankalissa olosuhteissa. Hän oli hyvin ihmisläheinen ja kuvaili kirjeissään paljon tapaamiaan ihmisiä. Kantele oli käytännön ihminen eikä hänen kirjoituksissaan ole havaittavissa taipumusta opillisiin pohdintoihin.

Kalkkunoiden teuraskuljetuksissa lintuja pidetään kuljetuspäällyksissä, joiden korkeuden on esitetty olevan riittämätön suurimpien kalkkunakukkojen kuljetukseen, mutta tieteellisiä tutkimuksia aiheesta on vähän. Jalostuksella saavutetut nopeakasvuiset ja suuret lihakset voivat lisäksi altistaa kalkkunat mahdollisesti kivuliaille lihassairauksille. Tutkielman tavoitteena oli selvittää lihasentsyymiarvojen vaikutusta kalkkunoiden käyttäytymiseen erikorkuisissa kuljetuspäällyksissä. Tutkimuksessa analysoitiin kreatiinikinaasin (CK) ja aspartaattiaminotransferaasin (ASAT) aktiivisuutta kalkkunoiden seerumissa, sillä näiden solunsisäisten entsyymien yhtäaikainen esiintyminen seerumissa on todettu olevan merkki lihasvauriosta ja täten kuvaavan eläimen heikentynyttä hyvinvointia. Tutkimuksessa käytettiin 36 lihantuotantoon jalostettua kalkkunakukkoa. Kutakin lintua testattiin kahtena eri päivänä noin viikon välein. Testattavien lintujen paino oli keskimäärin 16,5 ± 0,2 kiloa. Linnut olivat eri testikerroilla satunnaistetusti erikorkuisissa häkeissä. Häkkikorkeudet olivat 40, 55 ja 90 cm. Linnut olivat paikallaan olevissa häkeissä kuusi tuntia, jonka ajan niiden käyttäytymistä videoitiin. Kustakin linnusta otettiin verinäyte yhdellä testauskerralla kuvaamisen päätyttyä ja seerumista analysoitiin CK ja ASAT. Kalkkunoiden CK-aktiivisuus oli 25450,5 ±10402,6 IU/l ja ASAT-aktiivisuus 625,0 ±143,7 IU/l. Häkkikorkeudella ei ollut tilastollisesti merkitsevää korrelaatiota CK- eikä ASAT-aktiivisuuden kanssa (p = 0,86 ja p = 0,68), mutta CK- ja ASAT-arvot korreloivat positiivisesti keskenään (p < 0,001). Sekä CK- että ASAT-aktiivisuuksien ollessa korkeat linnut makasivat vähemmän 55 cm ja 90 cm korkeissa häkeissä testijakson ensimmäisinä tunteina. Paino ja CK-aktiivisuus olivat positiivisesti korreloituneet (p = 0,001). Kalkkunoiden lihasentsyymiarvoista on saatavilla heikosti tietoa, mutta verrattuna moniin muihin eläinlajeihin kalkkunoiden seerumin CK- ja ASAT-arvot ovat huomattavan korkeat. 40 cm korkeissa häkeissä linnut eivät voineet tilan ahtauden vuoksi seistä normaalissa asennossa jalat ojennettuina kuten muissa häkkikorkeuksissa. Linnuilla kivun liittymistä lihassairauksiin ei ole kuvattu, mutta kipu on varsin todennäköistä akuutissa vaiheessa. Kalkkunat siis saattavat mahdollisuuksien mukaan välttää makaamista kivun vuoksi. Jalostuksella saatu rintalihaksen nopea kasvu suhteettoman suureksi on todettu voivan aiheuttaa lihasvaurioita. On mahdollista, että jo suuri koko itsessään saattaa aiheuttaa kalkkunoille kipua. Tekijät, jotka kohottavat seerumin lihasentsyymiarvoja, aiheuttavat joka tapauksessa myös hyvinvointiongelmia. Nykyisin käytössä olevat matalat, 40 cm korkeat, kuljetuspäällykset voivat heikentää etenkin suurikokoisten kalkkunakukkojen kuljetuksen aikaista hyvinvointia, koska ne estävät lintuja seisomasta luonnollisessa asennossa ja niissä liikkuminen on muutenkin hyvin rajoitettua. Jotta jalostettujen kalkkunoiden lihasentsyymiarvoista voitaisiin tehdä tarkempia johtopäätöksiä, tarvitaan lisää tutkimuksia lihasentsyymiarvojen perustason määrittämiseksi. Lisätutkimuksia tarvitaan myös kalkkunoiden mahdollisesti kokeman kivun ja lihasentsyymiarvojen välisestä yhteydestä.

Biological membranes are tightly linked to the evolution of life, because they provide a way to concentrate molecules into partially closed compartments. The dynamic shaping of cellular membranes is essential for many physiological processes, including cell morphogenesis, motility, cytokinesis, endocytosis, and secretion. It is therefore essential to understand the structure of the membrane and recognize the players that directly sculpt the membrane and enable it to adopt different shapes. The actin cytoskeleton provides the force to push eukaryotic plasma membrane in order to form different protrusions or/and invaginations. It has now became evident that actin directly co-operates with many membrane sculptors, including BAR domain proteins, in these important events. However, the molecular mechanisms behind BAR domain function and the differences between the members of this large protein family remain largely unresolved. In this thesis, the structure and functions of the I-BAR domain family members IRSp53 and MIM were thoroughly analyzed. By using several methods such as electron microscopy and systematic mutagenesis, we showed that these I-BAR domain proteins bind to PI(4,5)P2-rich membranes, generate negative membrane curvature and are involved in the formation of plasma membrane protrusions in cells e.g. filopodia. Importantly, we characterized a novel member of the BAR-domain superfamily which we named Pinkbar. We revealed that Pinkbar is specifically expressed in kidney and epithelial cells, and it localizes to Rab13-positive vesicles in intestinal epithelial cells. Remarkably, we learned that the I-BAR domain of Pinkbar does not generate membrane curvature but instead stabilizes planar membranes. Based on structural, mutagenesis and biochemical work we present a model for the mechanism of the novel membrane deforming activity of Pinkbar. Collectively, this work describes the mechanism by which I-BAR domain proteins deform membranes and provides new information about the biological roles of these proteins. Intriguingly, this work also gives evidence that significant functional plasticity exists within the I-BAR domain family. I-BAR proteins can either generate negative membrane curvature or stabilize planar membrane sheets, depending on the specific structural properties of their I-BAR domains. The results presented in this thesis expand our knowledge on membrane sculpting mechanisms and shows for the first time how flat membranes can be generated in cells.
