Electrochemical and photophysical analysis of new donoracceptor systems 2 and 3, in which a benzothiadiazole (BTD) unit is covalently linked to a tetrathiafulvalene (TTF) core, have verified that the lowest excited state can be ascribed to an intramolecular-charge-transfer (ICT) (TTF)*(benzothiadiazole) transition. Owing to better overlap of the HOMO and LUMO in the fused scaffold of compound 3, the intensity of the 1ICT band is substantially higher compared to that in compound 2. The corresponding CT fluorescence is also observed in both cases. The radical cation TTF+. is easily observed through chemical and electrochemical oxidation by performing steady-state absorption experiments. Interestingly, compound 2 is photo-oxidized under aerobic conditions.

The synthesis and incorporation into oligonucleotides of C-nucleosides containing the two aromatic, non-hydrogen-bonding nucleobase substitutes biphenyl (I) and bipyridyl (Y) are described. Their homo- and hetero-recognition properties in different sequential arrangements were then investigated via UV-melting curve analysis, gel mobility assays, CD- and NMR spectroscopy. An NMR analysis of a dodecamer duplex containing one biphenyl pair in the center, as well as CD data on duplexes with multiple insertions provide further evidence for the zipper-like interstrand stacking motif that we proposed earlier based on molecular modeling. UV-thermal melting experiments with duplexes containing one to up to seven I- or Y base pairs revealed a constant increase in T(m) in the case of I and a constant decrease for Y. Mixed I/Y base pairs lead to stabilities in between the homoseries. Insertion of alternating I/abasic site- or Y/abasic site pairs strongly decreases the thermal stability of duplexes. Asymmetric distribution of I- or Y residues on either strand of the duplex were also investigated in this context. Duplexes with three natural base pairs at both ends and 50 % of I pairs in the center are still readily formed, while duplexes with blunt ended I pairs tend to aggregate unspecifically. Duplexes with one natural overhang at the end of a I-I base pair tract can both aggregate or form ordered duplexes, depending on the nature of the natural bases in the overhang

This minireview highlights three aspects of our recent work in the area of sugar modified oligonucleotide analogues. It provides an overview over recent results on the conformationally constrained analogue tricyclo-DNA with special emphasis of its antisense properties, it summarizes results on triple-helix forming oligodeoxynucleotides containing pyrrolidino-nucleosides with respect to DNA recognition via the dual recognition mode, and it highlights the advantageous application of the orthogonal oligonucleotidic pairing system homo-DNA in molecular beacons for DNA diagnostics

We prepared and investigated oligonucleotide duplexes of the sequence d(GATGAC(X)(n)GCTAG)d(CTAGC(Y)(n)GTCATC), in which X and Y designate biphenyl- (bph) and pentafluorobiphenyl- ((5F)bph) C-nucleotides, respectively, and n varies from 0-4. These hydrophobic base substitutes are expected to adopt a zipperlike, interstrand stacking motif, in which not only bph/bph or (5F)bph/(5F)bph homo pairs, but also (5F)bph/bph mixed pairs can be formed. By performing UV-melting curve analysis we found that incorporation of a single (5F)bph/(5F)bph pair leads to a duplex that is essentially as stable as the unmodified duplex (n=0), and 2.4 K more stable than the duplex with the nonfluorinated bph/bph pair. The T(m) of the mixed bph/(5F)bph pair was in between the T(m) values of the respective homo pairs. Additional, unnatural aromatic pairs increased the T(m) by +3.0-4.4 K/couple, irrespective of the nature of the aromatic residue. A thermodynamic analysis using isothermal titration calorimetry (ITC) of a series of duplexes with n=3 revealed lower (less negative) duplex formation enthalpies (DeltaH) in the (5F)bph/(5F)bph case than in the bph/bph case, and confirmed the higher thermodynamic stability (DeltaG) of the fluorinated duplex, suggesting it to be of entropic origin. Our data are compatible with a model in which the stacking of (5F)bph versus bph is dominated by dehydration of the aromatic units upon duplex formation. They do not support a model in which van der Waals dispersive forces (induced dipoles) or electrostatic (quadrupole) interactions play a dominant role

When comparing the transporters of three completely sequenced eukaryotic genomes - Saccharomyces cerevisiae, Arabidopsis thaliana and Homo sapiens - transporter types can be distinguished according to phylogeny, substrate spectrum, transport mechanism and cell specificity. The known amino acid transporters belong to five different superfamilies. Two preferentially Na+-coupled transporter superfamilies are not represented in them yeast and Arabidopsis genomes, whereas the other three groups, which often function as H+-coupled systems, have members in all investigated genomes. Additional superfamilies exist for organellar transport, including mitochondrial and plastidic carriers. When used in combination with phylogenetic analyses, functional comparison might aid our prediction of physiological functions for related but uncharacterized open reading frames.

In order to harness the unique properties of nanoparticles for novel clinical applications and to modulate their uptake into specific immune cells we designed a new library of homo- and hetero-functional fluorescence-encoded gold nanoparticles (Au-NPs) using different poly(vinyl alcohol) and poly(ethylene glycol)-based polymers for particle coating and stabilization. The encoded particles were fully characterized by UV-Vis and fluorescence spectroscopy, zeta potential and dynamic light scattering. The uptake by human monocyte derived dendritic cells in vitro was studied by confocal laser scanning microscopy and quantified by fluorescence-activated cell sorting and inductively coupled plasma atomic emission spectroscopy. We show how the chemical modification of particle surfaces, for instance by attaching fluorescent dyes, can conceal fundamental particle properties and modulate cellular uptake. In order to mask the influence of fluorescent dyes on cellular uptake while still exploiting its fluorescence for detection, we have created hetero-functionalized Au-NPs, which again show typical particle dependent cellular interactions. Our study clearly prove that the thorough characterization of nanoparticles at each modification step in the engineering process is absolutely essential and that it can be necessary to make substantial adjustments of the particles in order to obtain reliable cellular uptake data, which truly reflects particle properties.

In der anthropologischen Sammlung des Musée de l'Homme in Paris befinden sich etwa 23000 Objekte, darunter Skelette des modernen Menschen und des Neandertalers, Mumien, Haarproben, ethnographische Büsten und Moulagen. Aufgrund der kolonialen Vergangenheit Frankreichs bilden rezente und historische Schädelserien aus den ehemaligen Kolonien einen Schwerpunkt der Sammlung, die vor allem im 19. Jahrhundert zusammengetragen wurde. Unter den Überresten befinden sich auch die Skelette namentlich bekannter Individuen, die daher besonders von möglichen Rückgabeforderungen betroffen sind und einen sensiblen Umgang mit dieser Thematik erfordern. Am Beispiel dreier Serien von algerischen Schädeln (Berber, Kabylen, Chaouia) sollen die Chancen einer präventiven Dokumentation mittels 3D-Oberflächenscannings erläutert sowie die Möglichkeiten und Grenzen der anthropologischen Untersuchung dieses stark selektierten Materials vorgestellt werden. Die Schädel wurden Ende des 19. Jhs. von französischen Militärärzten in der damaligen Kolonie Algerien gesammelt und später der Société d’anthropologie de Paris übergeben. In dieser Studie wurden die Schädel zunächst einer anthropologischen Basisuntersuchung unterzogen (Individualalter, Geschlecht, Metrik, Zahnstatus). Basierend auf den metrischen Daten von 105 Schädeln wurde die Homo- und Heterogenität der drei ethnisch und kulturell verschiedenen Populationen untersucht (Berber: n=69, Kabylen: n=24, Chaouia: n=12). Einige Individuen heben sich morphologisch und metrisch von dem übrigen Material ab und könnten einen subsaharischen Ursprung vermuten lassen. Vor dem Hintergrund dieser Untersuchung soll über den wissenschaftlichen und politischen Umgang mit musealen Altsammlungen und den Sinn oder Unsinn deren anthropologischer Bearbeitung diskutiert werden. Die Rekonstruktion des Kontextes, aus dem die menschlichen Überreste stammen, soll dabei im Vordergrund stehen.

Since the origin of early Homo species during the Late Pliocene, interactions of humans with scavenging birds and mammals have changed in form through shifting ecological scenarios. How humans procured meat during the Quaternary Period changed from confrontational scavenging to hunting; shepherding of wild animals; and, eventually, intensive husbandry of domesticated animals. As humans evolved from carcass consumers to carcass providers, the overall relationship between humans and scavengers shifted from competition to facilitation. These changing interactions have translated into shifting provisioning (by signaling carcass location), regulating (e.g., by removing animal debris and controlling infectious diseases), and cultural ecosystem services (e.g., by favoring human language and social cooperation skills or, more recently, by enhancing ecotourism) provided by scavenging vertebrates. The continued survival of vultures and large mammalian scavengers alongside humans is now severely in jeopardy, threatening the loss of the numerous ecosystem services from which contemporary and future humans could benefit.

Einleitung Folgt man den aktuellen Sportentwicklungsberichten, dann sehen sich zunehmend mehr Sportvereine mit Mitgliederfluktuationen sowie stagnierenden bzw. zurückgehenden Mitglie- derzahlen konfrontiert (Lamprecht et al. 2012). Jedoch werden nicht alle Vereine in gleichem Maße mit instabilen Mitgliedschaftsverhältnissen konfrontiert. So gibt es mit Blick auf die Mitgliederstruktur Vereine, die – aufgrund ihrer spezifischen situativen und strukturellen Bedingungen – kaum Probleme mit Mitgliederfluktuation und Vereinsaustritten haben, wohin- gegen andere Vereine mitunter erhebliche Mitgliederrückgänge verzeichnen. Demnach ist zu vermuten, dass sich das soziale Handeln der Vereinsmitglieder je nach Organisationsprofil der Vereine unterscheidet. Zwar werden Verknüpfungen von Individual- und korrespondierenden Strukturdaten innerhalb der Sportvereinsforschung bereits seit geraumer Zeit gefordert (z.B. Nagel, 2007), aber bis heute nicht konsequent umgesetzt. Es stellt sich deshalb die Frage, welche organisations- und individuumsbezogenen Faktoren für die Mitgliederbindung in Sportvereinen eine Rolle spielen? Theoretisch-methodisches Vorgehen Im Zusammenhang mit der Frage der Mitgliederbindung wird davon ausgegangen, dass kontextuelle Bedingungen individuelle Entscheidungen strukturieren und somit persönliche Handlungsketten beeinflussen können (Coleman, 1990). Auf dieser Grundlage wird ein Mehr- ebenenmodell entwickelt, das neben individuellen Merkmalen auch die Strukturbedingungen von Sportvereinen berücksichtigt, die im Zusammenhang mit der individuellen Wahlhandlung zwischen stabiler Mitgliedschaft oder Austritt stehen. Der organisationale Kontext Sportverein wird dabei als Interessenorganisation konzeptualisiert, der mit seinen Kontexteigenschaften als Gelegenheits- und Opportunitätsstruktur, als kultureller sowie sozialer Bezugsrahmen gewisse Anreize schafft („Logik der Situation“), die gemäss individueller Präferenzen („Logik der Selektion“) zu Parametern des Mitgliederhandelns werden können. Die aus dem Modell abgeleiteten Annahmen werden auf der Grundlage einer Mitglieder- befragung (n = 1.434) in 36 Schweizer Sportvereinen empirisch geprüft. Die adäquate Methode, welche die hierarchische Datenstruktur (jede Messung auf der Individualebene kann eindeutig einer Messung auf der Vereinsebene zugeordnet werden) adäquat berücksichtigt und folglich das entwickelte theoretische Modell statistisch umsetzt, ist die Mehrebenenanalyse (z.B. Hox, 2002). Entsprechend wird der Einfluss der Individual- und Kontextebene auf die Mitglieder- bindung in Sportvereinen anhand unterschiedlicher Mehrebenenmodelle (Random Intercept, Random Slope sowie Cross-Level Interaktionen) geschätzt. Ergebnisse Die Analysen machen deutlich, dass sich die dauerhafte Mitgliedschaft in Sportvereinen nicht allein auf individuelle Merkmale der Mitgliedschaft, wie eine ausgeprägte Verbundenheit, ein positiv wahrgenommenes soziales Miteinander, die Zufriedenheit mit der Vereinsarbeit sowie die ehrenamtliche Mitarbeit zurückführen lässt. Darüber hinaus nehmen auch vereinsspezi-fische Strukturbedingungen Einfluss auf die Mitgliederbindung, wobei in ländlich geprägten Sportvereinen und in Vereinen, die Geselligkeit explizit fordern und in denen das Vereinsziel sportlicher Erfolg eher eine untergeordnete Rolle spielt, die Austrittswahrscheinlichkeit geringer ist. Diskussion Die Befunde machen deutlich, dass für eine dauerhafte Mitgliedschaft sowohl zweckorientierte Nutzenüberlegungen als auch solidargemeinschaftliche Handlungsorientierungen eine zentrale Rolle spielen, so dass eine ausschließliche Dienstleistungs- bzw. Kundenorientierung als Strategie der Mitgliederbindung in Sportvereinen, wie sie vielfach (auch von Verbänden) nahegelegt wird, zu kurz greifen dürfte. Weiterhin zeigt sich, dass der Sportverein als Ort der Geselligkeit nicht nur Werte des sozialen Miteinanders und solidarischen Verhaltens vermittelt, sondern auch als Katalysator der Stabilität der Mitgliedschaft wirkt, sofern entsprechende Gelegenheiten zur Verfügung stehen. Im Zusammenhang mit der Mitgliederbindung scheint damit gerade jene vereinskulturelle Orientierung von Vereinen bedeutsam, die im Zuge der Modernisierung von Vereinsangeboten gern als überholt erachtet wird. During the elongation cycle of protein biosynthesis, tRNAs traverse through the ribosome by consecutive binding to the 3 ribosomal binding sites (A-, P-, and E- sites). While the ribosomal A- and P-sites have been functionally well characterized in the past, the contribution of the E-site to protein biosynthesis is still poorly understood in molecular terms. Previous studies suggested an important functional interaction of the terminal residue A76 of E-tRNA with the nucleobase of the universally conserved 23S rRNA residue C2394. Using an atomic mutagenesis approach to introduce non-natural nucleoside analogs into the 23S rRNA, we could show that removal of the nucleobase or the ribose 2'-OH at C2394 had no effect on protein synthesis. On the other hand, our data disclose the importance of the highly conserved E-site base pair G2421-C2395 for effective translation. Ribosomes with a disrupted G2421-C2395 base pair are defective in tRNA binding to the E-site. This results in an impaired translation of genuine mRNAs, while homo-polymeric templates are not affected. Cumulatively our data emphasize the importance of E-site tRNA occupancy and in particular the intactness of the 23S rRNA base pair G2421-C2395 for productive protein biosynthesis.

We recently reported on the synthesis and pairing properties of the DNA analogue bicyclo[3.2.1]amide DNA (bca-DNA). In this analogue the nucleobases are attached via a linear, 4-bond amide-linker to a structurally preorganized sugar-phosphate backbone unit. To define the importance of the degree of structural rigidity of the bca-backbone unit on the pairing properties, we designed the structurally simpler cyclopentane amide DNA (cpa-DNA), in which the bicyclo[3.2.1]-scaffold was reduced to a cyclopentane unit while the base-linker was left unchanged. Here we present a synthetic route to the enantiomerically pure cpa-DNA monomers and the corresponding phosphoramidites containing the bases A and T, starting from a known, achiral precursor in 9 and 12 steps, respectively. Fully modified oligodeoxynucleotides were synthesized by standard solid-phase oligonucleotide chemistry, and their base-pairing properties with complementary oligonucleotides of the DNA-, RNA-, bca-DNA-, and cpa-DNA-backbones were assessed by UV melting curves and CD-spectroscopic methods. We found that cpa-oligoadenylates form duplexes with complementary DNA that are less stable by -2.7 degrees C/mod. compared to DNA. The corresponding cpa-oligothymidylates do not participate in complementary base-pairing with any of the investigated backbone systems except with its own (homo-duplex). As its congener bca-DNA, cpa-DNA seems to prefer left-handed helical duplex structures with DNA or with itself as indicated by the CD spectra

cpa-DNA monomers containing the bases adenine and thymine have been synthesized starting from the known compound 1 in 12 steps. Partially and fully modified cpa-thymidine and cpa-adenosine containing oligodeoxynucleotides were synthesized by standard oligonucleotide chemistry. Fully modified homo-cpa-A sequences lead to duplex destabilization by -1.4 degrees C/mod. relative to DNA. As its congener bca-DNA, cpa-DNA prefers left-handed duplex formation where possible

Cleverly designed molecular building blocks provide chemists with the tools of a powerful molecular-scale construction set. They enable them to engineer materials having a predictable order and useful solid-state properties. Hence, it is in the realm of supramolecular chemistry to follow a strategy for synthesizing materials which combine a selected set of properties, for instance from the areas of magnetism, photophysics and electronics. As a successful approach, host/guest solids which are based on extended anionic, homo- and bimetallic oxalato-bridged transition-metal compounds with two-and three-dimensional connectivities have been investigated. In this report, a brief review is given on the structural aspects of this class of compounds followed by a presentation of a thermal and magnetic study for two distinct, heterometallic oxalato-bridged layer compounds.

Despite increased scientific interest in the phenomenon of large-scale land acquisitions (LSLA), accurate data on implementation processes remain sparse. This paper aims at filling this gap by providing empirical in-depth knowledge on the case of the Swiss-based Addax Bioenergy Ltd. in Sierra Leone. Extensive fieldwork allowed the interdisciplinary research team 1) the identification of different actors that are necessary for the implementation on a vertical level and 2) the documentation of the heterogeneous group of project affected people’s perceptions and strategies on a horizontal level. Findings reveal that even a project labeled as best-practice example by UN agencies triggers a number of problematic processes for affected communities. The loss of natural resources that comes along with the land lease and the lack of employment possibilities mostly affects already vulnerable groups. On the other hand, strategies and resistance of local people also affect the project implementation. This shows that the horizontal and vertical levels are not separate entities. They are linked by social networks, social interactions, and means of communication and both levels take part in shaping the project’s impacts.
