The aim of the present study was to examine the association between milk protein polymorphism and fatty acids profiles of bovine milk. Milk samples were collected from each of 55 Reggiana cows during early, mid and late lactation, respectively, in two farms within the production area of Parmigiano Reggiano cheese. Identification and quantification of fatty acids were performed by gas chromatography. Milk fatty acid composition using cows of differing κ-casein (κ-Cn) and β-lactoglobulin (β-Lg) phenotypes was investigated. Statistically significant results regarding the associations between milk fatty acid composition and κ-Cn phenotype were found, in particular, κ-Cn BB seems to influence de novo fatty acid synthesis in the mammary gland. Also κ-Cn AB seems to have the same effect. Proportions of C10:0 (2,29a AA; 2,53b AB; 2,59b BB), C12:0 (2,77a AA; 3,17b AB; 3,20b BB) and C14:0 (9,22a AA; 10,25b AB; 10,27b BB) were higher in the milk from cows with κ-Cn phenotype AB and BB vs κ-Cn phenotype AA (p<0,05). Conversely C18:0 (7,84b AA; 7,20a,b AB; 6,94a BB) and C18:1 (19,19b AA; 16,81a AB; 16,79a BB) were lower in the milk from cows with κ-Cn phenotype AB and BB vs κ-Cn phenotype AA. The association between milk fatty acid composition and β-Lg phenotype was not statistically significant, except for some fatty acids. In particular, C12:0 (3,05a AA; 3,04a AB; 3,33b BB) was higher in the milk from cows with β-Lg phenotype BB vs β-Lg phenotype AA and AB (p<0,05). Concentrations of C18:0 (6,93a AA; 7,86b AB; 6,59a BB) and C18:1 (16,74a,b AA; 18,24b AB; 16,07a BB) were lower in the milk from cows with β-Lg phenotype AA and BB vs β-Lg phenotype AB (p<0,05). Moreover this research, carried out in farms within the Parmigiano Reggiano cheese district, analysed also the size distribution of fat globules in bulk milk of Reggiana and Frisona breed cows. In particular, the size distribution of individual milk fat globules of Reggiana cows with differing κ-Cn phenotypes was considered. From first observations, no statistically significant differences were observed.

I materiali zirconiferi, in particolare i silicati di zirconio e l’ossido di zirconio, vengono ampiamente utilizzati in diverse applicazioni industriali: fra queste, l’industria delle piastrelle ceramiche e di produzione dei materiali refrattari ne consuma quantitativi consistenti. Misure di spettrometria gamma condotte su diversi campioni di materiale zirconifero (farine di zirconio e sabbie zirconifere) utilizzati nell’industria ceramica per la produzione del gres porcellanato e delle piastrelle smaltate, hanno messo in evidenza valori di concentrazione di attività superiori a quelli presenti mediamente sulla crosta terrestre (35, 30 Bqkg-1 per il 238U e il 232Th, rispettivamente [Unscear, 2000]). L’aggiunta del materiale zirconifero nella preparazione delle piastrelle ceramiche in particolare di quelle smaltate (in una percentuale in peso pari al 10-20%) del gres porcellanato (in una percentuale in peso di 1-10%) e delle lamine di gres porcellanato “sottile” (in una percentuale in peso dell’ordine del 30%), conferisce al prodotto finale un alto grado di bianco, buone caratteristiche meccaniche ed un elevato effetto opacizzante, ma comporta anche un arricchimento in radionuclidi naturali. L’obiettivo principale di questo lavoro di tesi è stato quello di mettere a punto una metodologia (teorica e sperimentale) per valutare l’incremento di esposizione che possono ricevere un lavoratore standard di una industria ceramica ed una persona del pubblico che soggiorna in un ambiente rivestito con piastrelle. Da un lato, la presenza di radioattività nelle sabbie zirconifere, utilizzate come materie prime per la produzione di piastrelle, porta a considerare il problema dell’incremento di esposizione che può subire un lavoratore impiegato in ambiente di lavoro dove sono stoccati e manipolati consistenti quantitativi di materiale, dall’altro il contenuto di radioattività nel prodotto finito porta a considerare l’esposizione per una persona della popolazione che soggiorna in una stanza rivestita con materiale ceramico. Le numerose misure effettuate, unitamente allo sviluppo dei modelli necessari per valutare le dosi di esposizione per le persone del pubblico e dei lavoratori impiegati nel processo di produzione di piastrelle ceramiche, hanno permesso di mettere a punto una procedura che fornisce le garanzie necessarie per dichiarare accettabili le condizioni di lavoro nelle industrie ceramiche e più in generale il rispetto delle norme radioprotezionistiche per gli occupanti di ambienti rivestiti con piastrelle italiane.

The rational construction of the house. The writings and projects of Giuseppe Pagano Description, themes and research objectives The research aims at analysing the architecture of Giuseppe Pagano, which focuses on the theme of dwelling, through the reading of 3 of his house projects. On the one hand, these projects represent “minor” works not thoroughly known by Pagano’s contemporary critics; on the other they emphasise a particular methodological approach, which serves the author to explore a theme closely linked to his theoretical thought. The house project is a key to Pagano’s research, given its ties to the socio-cultural and political conditions in which the architect was working, so that it becomes a mirror of one of his specific and theoretical path, always in a state of becoming. Pagano understands architecture as a “servant of the human being”, subject to a “utilitarian slavery” since it is a clear, essential and “modest” answer to specific human needs, free from aprioristic aesthetic and formal choices. It is a rational architecture in sensu stricto; it constitutes a perfect synthesis between cause and effect and between function and form. The house needs to accommodate these principles because it is closely intertwined with human needs and intimately linked to a specific place, climatic conditions and technical and economical possibilities. Besides, differently from his public and common masterpieces such as the Palazzo Gualino, the Istituto di Fisica and the Università Commerciale Bocconi, the house projects are representative of a precise project will, which is expressed in a more authentic way, partially freed from political influences and dogmatic preoccupations and, therefore, far from the attempt to research a specific expressive language. I believe that the house project better represents that “ingenuity”, freshness and “sincerity” that Pagano identifies with the minor architecture, thereby revealing a more authentic expression of his understanding of a project. Therefore, the thesis, by tracing the theoretical research of Pagano through the analysis of some of his designed and built works, attempts to identify a specific methodological approach to Pagano’s project, which, developed through time, achieves a certain clarity in the 1930s. In fact, this methodological approach becomes more evident in his last projects, mainly regarding the house and the urban space. These reflect the attempt to respond to the new social needs and, at the same time, they also are an expression of a freer idea of built architecture, closely linked with the place and with the human being who dwells it. The three chosen projects (Villa Colli, La Casa a struttura d’acciaio and Villa Caraccio) make Pagano facing different places, different customers and different economic and technical conditions, which, given the author’s biography, correspond to important historical and political conditions. This is the reason why the projects become apparently distant works, both linguistically and conceptually, to the point that one can define them as ”eclectic”. However, I argue that this eclecticism is actually an added value to the architectural work of Pagano, steaming from the use of a method which, having as a basis the postulate of a rational architecture as essence and logic of building, finds specific variations depending on the multiple variables to be addressed by the project. This is the methodological heritage that Pagano learns from the tradition, especially that of the rural residential architecture, defined by Pagano as a “dictionary of the building logic of man”, as an “a-stylistic background”. For Pagano this traditional architecture is a clear expression of the relationships between a theme and its development, an architectural “fact” that is resolved with purely technical and utilitarian aims and with a spontaneous development far from any aprioristic theoretical principle. Architecture, therefore, cannot be an invention for Pagano and the personal contribution of each architect has to consider his/her close relationship with the specific historical context, place and new building methods. These are basic principles in the methodological approach that drives a great deal of his research and that also permits his thought to be modern. I argue that both ongoing and new collaborations with younger protagonists of the culture and architecture of the period are significant for the development of his methodology. These encounters represent the will to spread his own understanding of the “new architecture” as well as a way of self-renewal by confronting the self with new themes and realities and by learning from his collaborators. Thesis’ outline The thesis is divided in two principal parts, each articulated in four chapters attempting to offer a new reading of the theory and work of Pagano by emphasising the central themes of the research. The first chapter is an introduction to the thesis and to the theme of the rational house, as understood and developed in its typological and technical aspects by Pagano and by other protagonists of the Italian rationalism of the 1930s. Here the attention is on two different aspects defining, according to Pagano, the house project: on the one hand, the typological renewal, aimed at defining a “standard form” as a clear and essential answer to certain needs and variables of the project leading to different formal expressions. On the other, it focuses on the building, understood as a technique to “produce” architecture, where new technologies and new materials are not merely tools but also essential elements of the architectural work. In this way the villa becomes different from the theme of the common house or from that of the minimalist house, by using rules in the choice of material and in the techniques that are every time different depending on the theme under exploration and on the contingency of place. It is also visible the rigorous rationalism that distinguishes the author's appropriation of certain themes of rural architecture. The pages of “Casabella” and the events of the contemporary Triennali form the preliminary material for the writing of this chapter given that they are primary sources to individuate projects and writings produced by Pagano and contemporary architects on this theme. These writings and projects, when compared, reconstruct the evolution of the idea of the rational house and, specifically, of the personal research of Pagano. The second part regards the reading of three of Pagano’s projects of houses as a built verification of his theories. This section constitutes the central part of the thesis since it is aimed at detecting a specific methodological approach showing a theoretical and ideological evolution expressed in the vast edited literature. The three projects that have been chosen explore the theme of the house, looking at various research themes that the author proposes and that find continuity in the affirmation of a specific rationalism, focussed on concepts such as essentiality, utility, functionality and building honesty. These concepts guide the thought and the activities of Pagano, also reflecting a social and cultural period. The projects span from the theme of the villa moderna, Villa Colli, which, inspired by the architecture of North Europe, anticipates a specific rationalism of Pagano based on rigour, simplicity and essentiality, to the theme of the common house, Casa a struttura d’acciaio, la casa del domani, which ponders on the definition of new living spaces and, moreover, on new concepts of standardisation, economical efficiency and new materials responding to the changing needs of the modern society. Finally, the third project returns to the theme of the, Villa Caraccio, revisiting it with new perspectives. These perspectives find in the solution of the open plant, in the openness to nature and landscape and in the revisiting of materials and local building systems that idea of the freed house, which express clearly a new theoretical thought. Methodology It needs to be noted that due to the lack of an official Archive of Pagano’s work, the analysis of his work has been difficult and this explains the necessity to read the articles and the drawings published in the pages of «Casabella» and «Domus». As for the projects of Villa Colli and Casa a struttura d’acciaio, parts of the original drawings have been consulted. These drawings are not published and are kept in private archives of the collaborators of Pagano. The consultation of these documents has permitted the analysis of the cited works, which have been subject to a more complete reading following the different proposed solutions, which have permitted to understand the project path. The projects are analysed thought the method of comparison and critical reading which, specifically, means graphical elaborations and analytical schemes, mostly reconstructed on the basis of original projects but, where possible, also on a photographic investigation. The focus is on the project theme which, beginning with a specific living (dwelling) typology, finds variations because of the historico-political context in which Pagano is embedded and which partially shapes his research and theoretical thought, then translated in the built work. The analysis of the work follows, beginning, where possible, from a reconstruction of the evolution of the project as elaborated on the basis of the original documents and ending on an analysis of the constructive principles and composition. This second phase employs a methodology proposed by Pagano in his article Piante di ville, which, as expected, focuses on the plant as essential tool to identify the “true practical and poetic qualities of the construction”(Pagano, «Costruzioni-Casabella», 1940, p. 2). The reading of the project is integrated with the constructive analyses related to the technical aspects of the house which, in the case of Casa a struttura d’acciaio, play an important role in the project, while in Villa Colli and in Villa Caraccio are principally linked to the choice of materials for the construction of the different architectural elements. These are nonetheless key factors in the composition of the work. Future work could extend this reading to other house projects to deepen the research that could be completed with the consultation of Archival materials, which are missing at present. Finally, in the appendix I present a critical selection of the Pagano’s writings, which recall the themes discussed and embodied by the three projects. The texts have been selected among the articles published in Casabella and in other journals, completing the reading of the project work which cannot be detached from his theoretical thought. Moving from theory to project, we follow a path that brings us to define and deepen the central theme of the thesis: rational building as the principal feature of the architectural research of Pagano, which is paraphrased in multiple ways in his designed and built works.

"Bioactive compounds" are extranutritional constituents that typically occur in small quantities in food. They are being intensively studied to evaluate their effects on health. Bioactive compounds include both water soluble compounds, such as phenolics, and lipidic substances such as n-3 fatty acids, tocopherols and sterols. Phenolic compounds, tocopherols and sterols are present in all plants and have been studied extensively in cereals, nuts and oil. n-3 fatty acids are present in fish and all around the vegetable kingdom. The aim of the present work was the determination of bioactive and potentially toxic compounds in cereal based foods and nuts. The first section of this study was focused on the determination of bioactive compounds in cereals. Because of that the different forms of phytosterols were investigated in hexaploid and tetraploid wheats. Hexaploid cultivars were the best source of esterified sterols (40.7% and 37.3% of total sterols for Triticum aestivum and Triticum spelta, respectively). Significant amounts of free sterols (65.5% and 60.7% of total sterols for Triticum durum and Triticum dicoccon, respectively) were found in the tetraploid cultivars. Then, free and bound phenolic compounds were identified in barley flours. HPLCESI/ MSD analysis in negative and positive ion mode established that barley free flavan-3- ols and proanthocyanidins were four dimers and four trimers having (epi)catechin and/or (epi)gallocatechin (C and/or GC) subunits. Hydroxycinnamic acids and their derivatives were the main bound phenols in barley flours. The results obtained demonstrated that barley flours were rich in phenolic compounds that showed high antioxidant activity. The study also examined the relationships between phenolic compounds and lipid oxidation of bakery. To this purpose, the investigated barley flours were used in the bakery production. The formulated oven products presented an interesting content of phenolic compounds, but they were not able to contain the lipid oxidation. Furthermore, the influence of conventional packaging on lipid oxidation of pasta was evaluated in n-3 enriched spaghetti and egg spaghetti. The results proved that conventional packaging was not appropriated to preserve pasta from lipid oxidation; in fact, pasta that was exposed to light showed a high content of potentially toxic compounds derived from lipid oxidation (such as peroxide, oxidized fatty acids and COPs). In the second section, the content of sterols, phenolic compounds, n-3 fatty acids and tocopherols in walnuts were reported. Rapid analytical techniques were used to analyze the lipid fraction and to characterize phenolic compounds in walnuts. Total lipid chromatogram was used for the simultaneous determination of the profile of sterols and tocopherols. Linoleic and linolenic acids were the most representative n-6 and n-3 essential dietary fatty acids present in these nuts. Walnuts contained substantial amounts of γ- and δ-tocopherol, which explained their antioxidant properties. Sitosterol, Δ5-avenasterol and campesterol were the major free sterols found. Capillary electrophoresis coupled to DAD and microTOF was utilized to determine phenolic content of walnut. A new compound in walnut ((2E,4E)- 8-hydroxy-2,7-dimethyl-2,4-decadiene-1,10-dioic acid 6-O-β-D-glucopiranosyl ester, [M−H]− 403.161m/z) with a structure similar to glansreginins was also identified. Phenolic compounds corresponded to 14–28% of total polar compounds quantified. Aglycone and glycosylated ellagic acid represented the principal components and account for 64–75% of total phenols in walnuts. However, the sum of glansreginins A, B and ((2E,4E)-8-hydroxy- 2,7-dimethyl-2,4-decadiene-1,10-dioic acid 6-O-β-D-glucopiranosyl ester was in the range of 72–86% of total quantified compounds.

This PhD thesis describes the application of some instrumental analytical techniques suitable to the study of fundamental food products for the human diet, such as: extra virgin olive oil and dairy products. These products, widely spread in the market and with high nutritional values, are increasingly recognized healthy properties although their lipid fraction might contain some unfavorable components to the human health. The main aim of this PhD research project was the evaluation of the biological effects of bioactive compounds derived from edible plants, with particular attention on their possibility to counteract oxidative damage and inflammation. After a preliminary study of in vitro antioxidant activity, regarding the modification eventually occurring after home freezing and cooking of edible vegetables, cultured mammalian cells were used as experimental model systems. Soluble extract and essential oils derived from different cultivars of Brassicaceae and Lamiaceae were tested as possible tools for the counteraction of the oxidative damage due to reactive oxygen species (ROS), underlining differences related to cultivar and agronomic techniques. Since accumulating evidence indicates that phytochemicals exhibit several additional properties in complex biological systems, a nutrigenomic approach was used to further explain the biological activity of a green tea extract, and to evidence the anti-inflammatory role of bioactive compounds derived from different foods. Overall, results obtained could contribute to a better understanding of the potential health benefit of plant foods.

Il confronto in corso tra gli esperti di management sanitario sui dipartimenti ospedalieri, la crescente attenzione sui modelli di organizzazione a rete e le indagini sui servizi sanitari condotte con strumenti di analisi dei network hanno rappresentato la base su cui sviluppare il disegno dello studio. La prospettiva relazionale e le tecniche di social network analysis (Sna) sono state impiegate in un indagine empirica effettuata presso tre Dipartimenti Ospedalieri dell’Azienda USL di Bologna per osservare la struttura delle relazioni che intercorrono nell’ambito dei dipartimenti, tra le unità operative e tra i clinici, al fine di assicurare il quotidiano svolgersi dei processi clinico assistenziali dei pazienti. L’indagine si è posta tre obiettivi. Il primo è quello di confrontare la rete delle relazioni “reali” che intercorrono tra unità operative e tra clinici con le relazioni “progettate” attraverso l’afferenza delle unità operative nei dipartimenti e dei singoli clinici nelle unità operative. In sostanza si tratta di confrontare, con intenti esclusivamente conoscitivi, la struttura organizzativa formale – istituzionale con quella “informale”, che emerge dalle relazioni giornaliere tra i professionisti. In secondo luogo si intende comprendere se e come i fattori di natura attributiva che caratterizzano i singoli rispondenti, (es. età, sesso, laurea, anni di permanenza in azienda, ecc.) incidano sulla natura e sull’intensità delle relazioni e delle collaborazioni intrattenute con i colleghi all’interno dell’azienda. L’analisi ha un intento “esplicativo”, in quanto si cerca di indagare come le similitudini nelle caratteristiche individuali possano o meno incidere sull’intensità degli scambi e quindi delle collaborazioni tra professionisti. Il terzo obiettivo è volto a comprendere se e come i fattori attributivi e/o relazionali siamo in grado di spiegare l’attitudine mostrata dai singoli professionisti rispetto l’adozione di un approccio alla pratica clinica ispirato all’Evidence based medicine. Lo scopo è quello di verificare se la disponibilità / orientamento ad operare in una prospettiva evidence based sia più legata ad elementi e caratteristiche personali piuttosto che all’influenza esercitata da coloro con i quali si entra in contatto per motivi lavorativi. La relativa semplicità della fase di indagine ha indotto ad arricchire i contenuti e gli obiettivi originari del lavoro allo scopo di correlare indicatori relazionali e attributivi con indicatori di “performance”, in particolare di efficienza e appropriatezza. Le relazioni sono state rilevate attraverso un questionario sociometrico inserito in uno spazio web accessibile dalla rete ospedaliera e compilato online da parte dei medici. Il questionario è stato organizzato in tre sezioni: la prima per la raccolta di informazioni anagrafiche e dati attributivi dei clinici; la seconda volta a raccogliere i dati relazionali, funzionali e di consulenza, verso le equipe di professionisti (unità operative) e verso i singoli colleghi clinici; la terza sezione è dedicata alla raccolta di informazioni sull’utilizzo delle evidenze scientifiche a supporto della propria pratica clinica (consultazione di riviste, banche dati, rapporti di HTA, etc,) e sulla effettiva possibilità di accesso a tali strumenti. L’azienda ha fornito i dati di struttura e la base dati degli indicatori di attività delle UO arruolate nello studio. La compliance complessiva per i tre dipartimenti è stata pari a circa il 92% (302 rispondenti su un campione di 329 medici.). Non si sono rilevate differenze significative sulla compliance per i tre dipartimenti considerati. L’elaborazione dei dati è stata effettuata mediante specifici software per l’analisi delle reti sociali, UCINET 6 per il calcolo degli indicatori relazionali (centralità, densità, structural holes etc.), e Pajek per l’analisi grafica dei network. L’ultima fase è stata realizzata con l’ausilio del software statistico STATA vers. 10. L’analisi dei risultati è distinta in due 2 fasi principali. In primis è stato descritto il network di relazioni professionali rilevate, sono stai calcolati i relativi indicatori di centralità relazionale e verificato il grado di sovrapposizione tra struttura formale dei dipartimenti in studio con le relazioni informali che si stabiliscono tra di essi nell’ambito clinico. Successivamente è stato analizzato l’impatto che le relazioni esercitano sulla propensione da parte dei singoli medici a utilizzare nuove evidenze scientifiche I primi risultati emersi dallo studio forniscono interessanti evidenze, con particolare riguardo al dato di un discreto grado di “sovrapposizione” tra struttura formale e informale delle unità organizzative in studio e a correlazioni significative tra fattori relazionali e attitudine dei medici verso l’utilizzo dell’approccio EBM. Altre evidenze, in specie la correlazione tra “centralità” degli attori organizzativi e alcuni indicatori di performance /appropriatezza, meritano ulteriori approfondimenti e una definitiva validazione. In conclusione lo studio dimostra che la prospettiva relazionale e la Sna consentono di porre in evidenza caratteristiche dei dipartimenti, dei suoi attori e delle loro reti di reciproche relazioni, in grado di favorire la comprensione di alcune dinamiche ricercate proprio attraverso l’organizzazione dipartimentale e quindi di specifico interesse per il management, i clinici e quanti altri impegnati nella gestione e nello sviluppo di questo modello di organizzazione dell’ospedale.

Nuclear Magnetic Resonance (NMR) is a branch of spectroscopy that is based on the fact that many atomic nuclei may be oriented by a strong magnetic field and will absorb radiofrequency radiation at characteristic frequencies. The parameters that can be measured on the resulting spectral lines (line positions, intensities, line widths, multiplicities and transients in time-dependent experi-ments) can be interpreted in terms of molecular structure, conformation, molecular motion and other rate processes. In this way, high resolution (HR) NMR allows performing qualitative and quantitative analysis of samples in solution, in order to determine the structure of molecules in solution and not only. In the past, high-field NMR spectroscopy has mainly concerned with the elucidation of chemical structure in solution, but today is emerging as a powerful exploratory tool for probing biochemical and physical processes. It represents a versatile tool for the analysis of foods. In literature many NMR studies have been reported on different type of food such as wine, olive oil, coffee, fruit juices, milk, meat, egg, starch granules, flour, etc using different NMR techniques. Traditionally, univariate analytical methods have been used to ex-plore spectroscopic data. This method is useful to measure or to se-lect a single descriptive variable from the whole spectrum and , at the end, only this variable is analyzed. This univariate methods ap-proach, applied to HR-NMR data, lead to different problems due especially to the complexity of an NMR spectrum. In fact, the lat-ter is composed of different signals belonging to different mole-cules, but it is also true that the same molecules can be represented by different signals, generally strongly correlated. The univariate methods, in this case, takes in account only one or a few variables, causing a loss of information. Thus, when dealing with complex samples like foodstuff, univariate analysis of spectra data results not enough powerful. Spectra need to be considered in their wholeness and, for analysing them, it must be taken in consideration the whole data matrix: chemometric methods are designed to treat such multivariate data. Multivariate data analysis is used for a number of distinct, differ-ent purposes and the aims can be divided into three main groups: • data description (explorative data structure modelling of any ge-neric n-dimensional data matrix, PCA for example); • regression and prediction (PLS); • classification and prediction of class belongings for new samples (LDA and PLS-DA and ECVA). The aim of this PhD thesis was to verify the possibility of identify-ing and classifying plants or foodstuffs, in different classes, based on the concerted variation in metabolite levels, detected by NMR spectra and using the multivariate data analysis as a tool to inter-pret NMR information. It is important to underline that the results obtained are useful to point out the metabolic consequences of a specific modification on foodstuffs, avoiding the use of a targeted analysis for the different metabolites. The data analysis is performed by applying chemomet-ric multivariate techniques to the NMR dataset of spectra acquired. The research work presented in this thesis is the result of a three years PhD study. This thesis reports the main results obtained from these two main activities: A1) Evaluation of a data pre-processing system in order to mini-mize unwanted sources of variations, due to different instrumental set up, manual spectra processing and to sample preparations arte-facts; A2) Application of multivariate chemiometric models in data analy-sis.

Nell’ambito dell’ingegneria dei tessuti, la possibilità di rigenerazione del miocardio post-infartuale è un argomento “caldo”, che suscita grandi speranze ma solleva altrettanto grandi interrogativi - sostenuti dal sussistere di dubbi di base sulle scelte operative praticabili. Esiste tuttavia concordanza nel considerare fondamentale l’utilizzo di un “supporto” che possa mantenere nella sede peri-infartuale le cellule competenti. Infatti, la semplice iniezione di cellule staminali per via endovenosa o direttamente nell’area infartuata non si è dimostrata particolarmente efficace, soprattutto a causa della cospicua perdita cellulare che si verifica rapidamente dopo il trapianto. Ci si orienta quindi verso la strategia di seminare cellule in grado di transdifferenziare in senso muscolare cardiaco su un materiale biocompatibile in vitro e di impiantare successivamente il costrutto ottenuto in vivo dove ci si attende il riassorbimento del biomateriale e l’integrazione delle cellule. Tuttavia, mentre in altri settori della medicina - quali ortopedia e dermatologia - l’impiego di pseudotessuti ingegnerizzati ha già permesso di conseguire ottimi risultati nella rigenerazione di tessuti danneggiati, allo stato attuale, i progressi ottenuti nell’ambito della rigenerazione del miocardio infartuato appaiono ancora aneddotici e distanti dall’ottenere protocolli condivisi per l’impiego in clinica. Il lavoro presentato in questa ricerca, condotto grazie alla sinergia di competenze interdisciplinari negli ambiti chimico, biologico e dell’ingegneria biomedica meccanica ed elettronica, è uno studio di fattibilità di una metodica standardizzata in grado di indirizzare cellule staminali mesenchimali (MSCs) indifferenziate verso l’acquisizione in vitro di caratteri fenotipici confrontabili con quelli delle cellule muscolari cardiache attraverso il paradigma della coltura dinamica in bioreattore. Il prototipo di bioreattore impiegato, in quanto sviluppato originalmente nel corso di questa attività di ricerca, presenta rispetto ad altri strumenti descritti l’innovazione e il vantaggio di non richiedere l’utilizzo di un incubatore, in quanto esso stesso permette di coltivare cellule al suo interno in condizioni controllate di temperatura, pH e concentrazione di CO2. La sua flessibilità operativa consente di impostare e controllare da personal computer leggi di moto di qualsiasi forma anche con cicliche molto veloci. Infine, la presenza di estensimetri in grado di misurare finemente la variazione di tensione esercitata sulla matrice polimerica utilizzata, posta in trazione tra due afferraggi, permette di applicare nel tempo una forza di stiramento costante, ottenendo deformazioni controllate e risultati riproducibili in termini di modificazioni cellulari. Il superamento delle problematiche sorte durante la fase di messa a punto del sistema, che deve essere ritenuto parte integrante del lavoro di sviluppo condotto, ha permesso di studiare l’adattamento di MSCs allo stiramento ciclico, mostrando che questo effettivamente determina alcune differenze fenotipiche rispetto al controllo statico. Inoltre le cellule hanno acquistato una disposizione orientata lungo l’asse longitudinale delle fibre, dato questo particolarmente importante se si considera la disposizione ordinata delle cellule del miocardio, le quali costituiscono un vero e proprio sincizio, indispensabile per una diffusione sincrona dell’impulso elettrico di contrazione. La creazione di uno pseudotessuto cardiaco ottimale richiederà ovviamente ulteriore lavoro, ma la metodica qui presentata si propone al tempo stesso come uno strumento di studio e come una strategia operativa per un approccio innovativo e standardizzabile alla medicina rigenerativa del miocardio.

Aims: Ripening evaluation of two different Pecorino cheese varieties ripened according either to a traditional method in plant and in cave. Different ripening features have been analyzed in order to evaluate the cave as possible ripening environment with the aim of obtaining a peculiar product which could also establish an added value to the cultural heritage of the local place in which it has been originally manufactured. Methods and Results: Chemical-physical features of Pecorino cheese have been initially analyzed into two different ripening environments and experimentations, among which: pH, weight reduction and subsequent water activity. Furthermore, the microbial composition has been characterized in relationship with the two different ripening environments, undertaking a variety of microbial groups, such as: lactic bacteria, staphylococci, yeasts, lactococci, enterobacteria, enterococci. Besides, an additional analysis for the in-cave adaptability evaluation has been the identification of biogenic amines inside the Pecorino cheese (2-phenilethylamine, putrescine, cadaverine, hystidine, tyramine, spermine and spermidine). Further analysis were undertaken in order to track the lipid profile evolution, reporting the concentration of the cheese free fatty acids in object, in relation with ripening time, environment and production. In order to analyse the flavour compounds present in Pecorino cheese, the SPME-GC-MS technique has been widely employed. As a result, it is confirmed the trend showed by the short-chain free fatty acids, that is to say the fatty acids which are mostly involved in conveying a stronger flavor to the cheese. With the purpose of assessing the protheolytic patterns of the above-mentioned Pecorino cheese in the two different ripening environments and testing methods, the technique SDS-PAGE has been employed into the cheese insoluble fraction, whereas the SDS-PAGE technique has been carried out into the cheese soluble portion. Furthermore, different isolated belonging to various microbial groups have been genotypically characterized though the ITS-PCR technique with the aim to identify the membership species. With reference to lactic bacillus the characterized species are: Lactobacillus brevis, Lactobacillus curvatus and Lactobacillus paraplantarum. With reference to lactococci the predominant species is Lactococcus lactis, coming from the employed starter used in the cheese manufacturing. With reference to enterococcus, the predominant species are Enterococcus faecium and Enterococcus faecalis. Moreover, Streptococcus termophilus and Streptococcus macedonicus have been identified too. For staphylococci the identified species are Staphyilococcus equorum, Staphylococcus saprophyfiticus and Staphylococcus xylosus. Finally, a sensorial analysis has been undertaken through on one side a consumer test made by inexperienced consumers, and on the other side through a panel test achieved by expert consumers. From such test Pecorino cheese ripened in cave were found to be more pleasant in comparison with Pecorino cheese ripened in plant. Conclusions: The proposed approach and the undertaken analysis showed the cave as preferential ripening environment for Pecorino cheese and for the development of a more palatable product and safer for consumers’ health.

L'attività di ricerca descritta in questa tesi fornisce linee guida per la progettazione di arti protesici inferiori, con particolare riferimento alla progettazione di protesi a basso costo. La necessità di efficienti protesi a basso costo risulta infatti sentita nei Paesi in via di sviluppo ma anche dalle fasce meno abbienti dei paesi occidentali. Al fine di comprendere le strategie adottate dall'apparato locomotorio per muoversi con le protesi sono analizzati il cammino fisiologico, le protesi presenti sul mercato ed infine le modalità con cui le loro prestazioni sono valutate. Con il presente lavoro, dopo aver osservato la presenza di una scarsa strutturazione della metodologia di progettazione che riguarda specialmente il settore del basso costo, si propone una metodologia il più possibile oggettiva e ripetibile tesa ad individuare quali sono gli aspetti essenziali di una protesi per garantire al paziente una buona qualità di vita. Solo questi aspetti dovranno essere selezionati al fine di ottenere la massima semplificazione della protesi e ridurre il più possibile i costi. Per la simulazione delle attività di locomozione, in particolare del cammino, è stato elaborato un apposito modello spaziale del cammino. Il modello proposto ha 7 membri rigidi (corrispondenti a piedi, tibie, femori e bacino) e 24 gradi di libertà. Le articolazioni e l'appoggio dei piedi al suolo sono modellati con giunti sferici. La pianta del piede consente tre possibili punti di appoggio. I criteri di realizzazione delle simulazioni possono comprendere aspetti energetici, cinematici e dinamici considerati come obiettivo dall'apparato locomotorio. In questa tesi vengono trattati in particolare gli aspetti cinematici ed è mostrata un'applicazione della procedura nella quale vengono dapprima identificati i riferimenti fisiologici del cammino e quindi simulato il cammino in presenza di una menomazione al ginocchio (eliminazione della flessione in fase di appoggio). Viene quindi lasciato a sviluppi futuri il completamento della procedura e la sua implementazione in un codice di calcolo.
