The aim of this study was to evaluate whether yerba maté alcoholic extracts at very low concentrations (0.01 and 0.1%), prevent/retard lipid peroxidation in beef hamburgers without impairing sensory acceptability. For this TBARs and hexanal levels, fatty acid profile and cholesterol oxides were evaluated as oxidation parameters in beef hamburgers during 90 days' storage. The addition of 0.01% yerba maté ethanolic extracts proved inefficient in restraining the lipid peroxidation while the addition of 0.1% resulted in efficient antioxidant activity. Sensory evaluation of hamburger containing 0.1% yerba maté ethanolic extracts showed good acceptability. Yerba maté ethanolic extracts could entirely or partially replace the phenolic synthetic antioxidants in beef hamburgers only when used at above legally allowed concentrations for antioxidant additives (0.01%).

Yerba mate´ (Ilex paraguariensis) is rich in polyphenols, especially chlorogenic acids. Evidence suggests that dietary polyphenols could play a role in glucose absorption and metabolism. The aim of this study was to evaluate the antidiabetic properties of yerba mate´ extract in alloxan-induced diabetic Wistar rats. Animals (n ) 41) were divided in four groups: nondiabetic control (NDC, n=11), and diabetic yerba maté (DY, n = 10). The intervention consisted in the administration of yerba mate´ extract in a 1 g extract/ kg body weight dose for 28 days; controls received saline solution only. There were no significant differences in serum glucose, insulin, and hepatic glucose-6-phosphatase activity between the groups that ingested yerba maté extract (NDY and DY) and the controls (NDC and DC). However, the intestinal SGLT1 gene expression was significantly lower in animals that received yerba maté both in upper (p = 0.007) and middle (p < 0.001) small intestine. These results indicate that bioactive compounds present in yerba maté might be capable of interfering in glucose absorption, by decreasing SGLT1 expression

Yerba maté extracts have in vitro antioxidant capacity attributed to the presence of polyphenolic compounds, mainly chlorogenic acids and dicaffeoylquinic acid derivatives. DPPH is one of the most used assays to measure the antioxidant capacity of pure compounds and plant extracts. It is difficult to compare the results between studies because this assay is applied in too many different conditions by the different research groups. Thus, in order to assess the antioxidant capacity of yerba maté extracts, the following procedure is proposed: 100 µL of an aqueous dilution of the extracts is mixed in duplicate with 3.0 mL of a DPPH 'work solution in absolute methanol (100 µM.L-1), with an incubation time of 120 minutes in darkness at 37 ± 1 °C, and then absorbance is read at 517 nm against absolute methanol. The results should be expressed as ascorbic acid equivalents or Trolox equivalents in mass percentage (g% dm, dry matter) in order to facilitate comparisons. The AOC of the ethanolic extracts ranged between 12.8 and 23.1 g TE % dm and from 9.1 to 16.4 g AAE % dm. The AOC determined by the DPPH assay proposed in the present study can be related to the total polyphenolic content determined by the Folin-Ciocalteu assay.

Yerba mate extract (Ilex paraguariensis) is a Source of phenolic compounds that possesses in vitro antioxidant activities and may contribute to a reduction in the risk of cardiovascular disease. In this Study we examined the acute effects of the consumption of mate infusion on ex vivo plasma and low-density lipoprotein (LDL) oxidation, plasma antioxidant capacity, and platelet aggregation. Twelve healthy fasted subjects ingested 500 mL. of mate infusion and blood samples were collected before and I h after mate intake. Lipid peroxidation of plasma and LDL was monitored by the measurement of cholesteryl-ester hydroperoxides (CE-OOH) and cholesterol oxides. The plasma antioxidant capacity was measured as ferric-reducing antioxidant potential (FRAP). Platelet aggregation was evaluated in platelet-rich plasma Stimulated with adenosine diphosphate and coagulation was tested in platelet-poor plasma. Ingestion of mate infusion diminished the ex vivo oxidizability of both plasma and LDL particles. After mate intake, the CE-OOH levels were around 50% lower in plasma oxidized with copper or 2,2`-azobis[-2-amidine-propane-hydrochloride] (AAPH) and the lag time to plasma oxidation increased 2-fold (P < 0.05). Copper- and AAPH-induced LDL peroxidation were also inhibited by around 50% and 20%, respectively, after mate Consumption (P < 0.05). The levels of various oxysterols were significantly reduced in oxidized-plasma and LDL (P < 0.05) and FRAP increased by 7.7% after mate intake (P < 0.01). However. mate consumption did not inhibit platelet aggregation or blood coagulation. In summary, intake of yerba mate infusion improved the antioxidant capacity and the resistance of plasma and LDL particles to ex vivo lipid peroxidation. (C) 2008 Elsevier Ltd. All rights reserved.

A group of transposons, named maT, with characteristics intermediate between mariner and Tc1 transposons, is described. Two defective genomic copies of MdmaT from the housefly Musca domestica, with 85% identity, were found flanking and imbedded in the MdalphaE7 esterase gene involved in organophosphate insecticide resistance. Two cDNA clones, with 99% identity to each other and 72%-89% identity to the genomic copies were also obtained, but both represented truncated versions of the putative open reading frame. A third incomplete genomic copy of MdmaT was also identified upstream of the putative M. domestica period gene. The MdmaT sequences showed high identity to the transposable element Bmmar1 from the silk-worm moth, Bombyx mori, and to previously unidentified sequences in the genome of Caenorhabditis elegans. A total of 16 copies of full-length maT sequences were identified in the C elegans genome, representing three variants of the transposon, with 34%-100% identity amongst them. Twelve of the copies, named CemaT1, were virtually identical, with eight of them encoding a putative full length, intact transposase. Secondary structure predictions and phylogenetic analyses confirm that maT elements belong to the mariner-Tc1 superfamily of transposons, but their intermediate sequence and predicted structural characteristics suggest that they belong to a unique clade, distinct from either mariner-like or Tc1-like elements.

The study evaluated the efficiency of diagnostic laboratory methods to detect anti-Toxoplasma gondii antibodies in paired serum and aqueous humour samples from experimentally infected pigs. 18-mixed breed pigs were used during the experiment; these were divided into two groups, G I (infected group, n = 10) and G2 (uninfected group, n = 8). Infection was performed with 4 x 10(4) VEG strain oocysts at day 0 by the oral route in G1 animals. All pigs were euthanized at day 60, when retina, aqueous humour, and blood samples were collected. Anti-T gondii antibody levels were assessed in serum (s) and aqueous humour (ah) by indirect immunofluorescence assay (IFA), modified agglutination test (MAT), m-ELISA (using crude membranes from T gondii tachyzoites as antigen) and r-ELISA (using rhoptries from T gondii tachyzoites as antigen). Polymerase chain reactions (PCR) of samples from the retina were performed by using Tox4 and Tox5 primers. Antibody titers of G1 animals ranged from 128 to 1024 and from 16 to 256 in serum and aqueous humour, respectively. There were differences in the correlation coefficients between IFA(s) x IFA (ah) (r = 0.62, P = 0.05), MAT(s) x MAT (ah) (r = 0.97, P < 0.0001); however, there was no significant difference between r-ELISA(s) x r-ELISA (ah) (r = 0. 14, P = 0.7). Antibodies present in serum and aqueous humour recognized similar antigens. Samples of retina were positive by PCR in 30% (3/10) of infected pigs. G2 animals remained without antibody levels and were PCR negative throughout the experiment. These results suggest that the use of a combination of tests and immunoblotting for paired aqueous humour and serum samples could improve the sensitivity and specificity for the diagnosis of ocular toxoplasmosis. A homocistinúria devida à deficiência da enzima cistationina -sintetase ou “homocistinúria clássica” é uma doença metabólica rara (1/344 000 RN), de transmissão autossómica recessiva e caracterizada por elevada heterogeneidade clínica, que frequentemente contribui para o diagnóstico tardio. Existe tratamento efetivo, se instituído antes de se instalarem sintomas irreversíveis, pelo que tem sido incluída num número considerável de programas de rastreio neonatal. O rastreio baseia-se na determinação dos níveis plasmáticos de metionina, por espectrometria de massa em tandem (ms/ms), mas conduz à identificação de muitos casos falsos-positivos, portadores de uma condição com significado clínico não completamente esclarecido, a deficiência em metionina adenosiltransferase (MAT I/III). Ambas as condições são rastreadas na Galiza e em Portugal desde 2000 e 2004, respetivamente. Desde então, foram identificados três doentes com homocistinúria clássica e 44 doentes com deficiência em MAT I/III. Uma forma dominante, e aparentemente benigna, desta última condição, associada à mutação p.R264H, parece ser muito frequente na Península Ibérica. A implementação de um teste de segunda linha, consistindo na determinação da homocisteína total, permitiria reduzir consideravelmente o número de RN identificados com deficiência em MAT I/III e melhorar a especificidade e valor positivo preditivo do rastreio da homocistinúria clássica.

A ideia de que as Matemáticas de Portugal (e de Espanha) atravessaram, depois de um período áureo nos Descobrimentos, um longo deserto onde não foi possível florescerem Mestres, nem escolas, nem cultura científica, nem investigação de relevo foi, durante muito tempo, reiteradamente veiculada, inclusivé através de alguns dos nossos mais referenciados historiadores da Matemática, como é o caso de Gomes Teixeira ou de Rey Pastor. Mas a verdade é que o estudo da História das Matemáticas em Portugal tem, na última década, vivido um interesse crescente onde sobressaem, em particular, uma leitura menos enviesada sobre, por exemplo, o papel educativo dos Jesuítas ou a publicação das obras completas de Pedro Nunes. Está-se assim a contribuir para uma compreensão mais completa da História geral de Portugal, de que a História da Ciência e da Cultura faz parte. José Anastácio da Cunha (1744-1787) foi figura de proa no século XVIII português. Sabíamo-lo matemático que, sem nunca ter saído de Portugal, havia sido capaz de antecipar, em mais de 50 anos, os esforços de matemáticos franceses e alemães para fundar a Matemática com rigor. Sabíamo-lo também autor de uma vasta e diversificada obra de inegável importância matemática mas, igualmente, autor de textos poéticos. Agora, com o projecto que denominámos de MAT2, centramo-nos em José Anastácio da Cunha e pretendemos, se possível, ir ainda mais além. Partimos de uma descoberta, árdua mas com final feliz, em um Arquivo de família: o da Casa de Mateus. Sentimo-nos, com esta “sorte”, privilegiados e gratos por nos ter sido gentilmente concedido o acesso a um vasto conjunto de documentos únicos (diários de viagens, notas de aulas e correspondência) que incluem memórias autógrafas e inéditas de Anastácio da Cunha. Organizámo-nos, cientes do trabalho árduo que temos pela frente, multi e interdisciplinarmente englobando a Matemática (nas suas múltiplas especializações) e a História (incluindo a da Matemática) mas também contando com a Física, a Informática, os estudos militares ou a Arquivística e as Humanidades; reunimos académicos, mais e menos veteranos, com investigadores jovens e juntámos valências nacionais e estrangeiras. No presente artigo daremos conta do percurso trilhado, até agora, pelo projecto MAT2.

Depuis plus de 30 ans, Maïté se bat sur le terrain avec les femmes, spécialement avec des femmes victimes de violences conjugales. Elle est cofondatrice de l'APIAF (Association pour la Promotion d'Initiatives Autonomes des Femmes), centre d'accueil pour des femmes en difficulté. « Lieu d'écoute, d'information, d'orientation et d'insertion pour les femmes en difficulté, avec un accueil spécifique pour les femmes victimes de violences conjugales », l'APIAF fut créée à Toulouse en 1981 et adhère à la Fédération Nationale Solidarité Femmes (FNSF). Chaque année, l'APIAF accueille environ 1000 femmes victimes de violences conjugales et 1200 enfants. Maïté participe aussi au comité d'éthique et au service de formation de la FNSF. Quartiers Nord de Toulouse. Durant cette période, en tant que militante féministe, elle s'est intéressée à la pratique culturelle de la vidéo et à son utilisation comme outil d'expression engagée. Plusieurs de ses documentaires et reportages ont été tournés avec Carol Prestat. Maïté utilise cet outil dans ses interventions, tant auprès des femmes - parmi les documentaires qu'elle a réalisés : "Cinq femmes et des mariages", "Des dames comme tout le monde" (avec des femmes chômeuses) et "Prends garde à toi!" (sur l'intervention professionnelle auprès de femmes victimes de violences conjugales) - que des jeunes - une de ces vidéos, « Salah, Malik, Beurs », a donné naissance au groupe de musique Zebda. Ce travail avec les jeunes l'a amenée à mettre sur pied l'Association Vitécri, visant à réaliser des films et organiser des activités culturelles avec des jeunes des quartiers populaires issu·e·s de l'immigration postcoloniale.

FXYD3 (Mat-8) proteins are regulators of Na,K-ATPase. In normal tissue, FXYD3 is mainly expressed in stomach and colon, but it is also overexpressed in cancer cells, suggesting a role in tumorogenesis. We show that FXYD3 silencing has no effect on cell proliferation but promotes cell apoptosis and prevents cell differentiation of human colon adenocarcinoma cells (Caco-2), which is reflected by a reduction in alkaline phosphatase and villin expression, a change in several other differentiation markers, and a decrease in transepithelial resistance. Inhibition of cell differentiation in FXYD3-deficient cells is accompanied by an increase in the apparent Na+ and K+ affinities of Na,K-ATPase, reflecting the absence of Na,K-pump regulation by FXYD3. In addition, we observe a decrease in the maximal Na,K-ATPase activity due to a decrease in its turnover number, which correlates with a change in Na,K-ATPase isozyme expression that is characteristic of cancer cells. Overall, our results suggest an important role of FXYD3 in cell differentiation of Caco-2 cells. One possibility is that FXYD3 silencing prevents proper regulation of Na,K-ATPase, which leads to perturbation of cellular Na+ and K+ homeostasis and changes in the expression of Na,K-ATPase isozymes, whose functional properties are incompatible with Caco-2 cell differentiation.

Yerba mate (Ilex paraguariensis) is a tree species native to the subtropical regions of South America, and is found in Brazil predominantly in the southern region. Despite the historical importance in this region, so far, studies on crop nutrition to improve yields are scarce. Thus, this study evaluated the effect of potassium rates on K soil availability, and the yield and nutritional status of yerba mate. The experiment was conducted in São Mateus do Sul, State of Paraná, on a Humox soil, where K2O rates of 0, 20, 40, 80, 160, and 320 kg ha-1 were tested on 7-year-old plantations. The experiment was harvested 24 months after installation by removing approximately 95 % of the canopy that had sprouted from the previous harvest. The soil was evaluated for K availability in the layers 0-10, 0-20, 10-20, and 20-40 cm. The plant parts leaf fresh matter (LM), twigs (TW), thick branches (BR) and commercial yerba mate (COYM), i.e., LM+TW, were analyzed. In addition, the relationship between fresh matter/dry matter (FM/DM) and K concentration in LM, AG and BR were evaluated. The fertilization increased K availability in all evaluated soil layers, indicating good mobility of the nutrient even at low rates. Yerba mate responded positively to increasing K2O rates with higher yields of all harvested components. The crop proved K-demanding, with a maximum COYM yield of 28.5 t ha-1, when 72 mg dm-3 K was available in the 0-20 cm layer. Yerba mate in the plant production stage requires soil K availability at medium to high level; in clayey soil with low K availability, a rate of 300 kg ha-1 K2O should be applied at 24 month intervals to obtain high yields. A leaf K concentration of 16.0 g ha-1 is suitable for yerba mate in the growth stage.

Purpose: The HR-NBL1 trial of the European SIOP Neuroblastoma Group randomised 2 MAT regimens to demonstrate superiority based on event free survival (EFS).Method: Response eligibility criteria prior to randomisation after Rapid COJEC Induction (J Clin Oncol, 2010) 3 4 2 courses of TVD (Cancer, 2003) included complete bone marrow remission andA ^ 3, but improved, mIBG positive spots. The MAT regimens were BuMel (oral busulfan till 2006, 4_150 mg/m2 in 4 equal doses, or after 2006 intravenous use according to body weight and melphalan 140 mg/m__/day) and CEM (carboplatin ctn. infusion (4xAUC 4.1 mg/ml.min/day), etoposide ctn. infusion (4_338 mg/m__day or 4_200 mg/m__/ day*), melphalan (3_70 mg/m__/day or 3_60 mg/m__/day*. *reduced if GFR<100 ml/ min/1.73m__)). A minimum of 3_10E6 CD34/kgBW PBSC were requested. VOD prophylaxis included ursadiol, but not prophylactic defibrotide. Local control included surgery and radiotherapy of 21 Gy. A total of 598 high risk neuroblastoma patients were randomised (296 BuMel, 302 CEM). The median age at randomisation was 3 years (1-17.2).Results: A significant difference in EFS in favour of BuMel (3-years EFS 49% vs. 33%) was observed as well as for overall survival (3-years OS 60% vs. 48%, p¼0.004) with a median follow up of 3 years. This difference was mainly related to the relapse and progression incidence, which was significantly (p<0.001) lower with BuMel (48% vs. 60%). The severe toxicity rate up to day 100 (ICU and toxic deaths) was below 10%, but was significantly higher for CEM (p¼0.014). The acute toxic death rate was 3% for BuMel and 5% for CEM (NS). The acute MAT toxicity profile favours the BuMel regimen in spite of a total VOD incidence of 18% (grade 3:5%). Based on these results and following advice from the DMC, the randomisation was closed early.
