La régulation de l’expression des gènes est ce qui permet à nos cellules de s’adapter à leur environnement, de combattre les infections ou, plus généralement, de produire la quantité exacte de protéine nécessaire pour répondre à un besoin spécifique. Parmi les joueurs les plus importants dans cette régulation de l’expression des gènes on retrouve les microARN (miARN). Ces petits ARN de 22 nucléotides sont présents chez la majorité des espèces multicellulaires et sont responsables du contrôle direct de plus de 30% des gènes exprimant des protéines chez les vertébrés. La famille de miARN lethal-7 (let-7) est composée de miARN parmi les plus connus et ayant des fonctions cruciales pour la cellule. La régulation du niveau des miARN let-7 est essentielle au bon développement cellulaire. La biogenèse de ces miARN, du transcrit primaire jusqu’à leur forme mature, est régulée principalement par Lin28, une protéine pluripotente très conservée. Cette protéine est composée d’un domaine cold shock (CSD) et de deux domaines de liaison au zinc. C’est grâce à ces domaines de liaison à l’ARN que Lin28 peut lier et inhiber la maturation des miARN let-7. L’objectif de cette thèse est de caractériser l’interaction entre Lin28 et le microARN précurseur let-7g afin de mieux comprendre le rôle de cette protéine dans l’inhibition de la biogenèse du miARN. À l’aide de techniques biochimiques et biophysiques, nous avons d’abord défini les principaux déterminants de l’interaction entre Lin28 et la boucle terminale du miARN précurseur let-7g (TL-let-7g). Nous avons conclu que le domaine C-terminal de Lin28, composé d’un motif riche en lysines et arginines ainsi que de deux motifs de liaison au zinc, permet à la protéine de lier spécifiquement et avec haute affinité un renflement riche en guanine conservé chez les précurseurs de la famille let-7. Aussi, parce que la séquence et la spécificité de liaison à l’ARN de ce domaine C-terminal sont semblables à celles de la protéine NCp7 du VIH, nous avons défini ce dernier comme le domaine NCp7-like de Lin28. Par la suite, nous avons caractérisé la multimérisation de trois protéines Lin28 sur la boucle terminale de pre-let-7g. Ceci a permis de réconcilier d’apparentes contradictions retrouvées dans la littérature actuelle concernant les sites de liaison de Lin28 lors de sa liaison aux miARN précurseurs. Nous avons identifié trois sites de liaison à haute affinité sur TL-let-7g qui sont liés dans un ordre précis par trois protéines Lin28. Lors de la formation du complexe multimérique, le CSD permet une déstabilisation de l’ARN, ce qui rend accessible plusieurs sites de liaison. Le domaine NCp7-like permet plutôt un assemblage ordonné de la protéine et facilite la liaison initiale de cette dernière. Ces nouveaux résultats rendent possible la mise au point d’un nouveau modèle de l’interaction entre Lin28 et le miARN précurseur let-7g. En conclusion, les études réalisées dans cette thèse apportent une meilleure compréhension des mécanismes moléculaires impliqués dans la régulation post-transcriptionnelle d’une importante famille de miARN et permettront de guider les futures études dans le domaine de recherche en pleine effervescence qu’est celui de la biogenèse des miARN.

H-1- and C-13-NMR spectroscopy and FT-Raman spectroscopy are used to investigate the properties of a polymer gel dosimeter post-irradiation. The polymer gel (PACT) is composed of acrylamide, N,N'-methylene-bisacrylamide, gelatin, and water. The formation of a polyacrylamide network within the gelatin matrix follows a dose dependence nonlinearly correlated to the disappearance of the double bonds from the dissolved monomers within the absorbed dose range of 0-50 Gy. The signal from the gelatin remains constant with irradiation. We show that the NMR spin-spin relaxation times (T-2) of PAGs irradiated to up to 50 Gy measured in a NMR spectrometer and a clinical magnetic resonance imaging scanner can be modeled using the spectroscopic intensity of the growing polymer network. More specifically, we show that the nonlinear T-2 dependence against dose can be understood in terms of the fraction of protons in three different proton pools. (C) 2000 John Wiley & Sons, Inc.

Matrix metalloproteinase (MMP) inhibition has been shown to reduce dentin caries progression, but its role in dental erosion has not yet been assessed. This study tested the hypothesis that gels containing MMP inhibitors (epigallocatechin gallate-EGCG and chlorhexidine) can prevent dental erosion. Volunteers (n = 10) wore palatal devices containing bovine dentin blocks (n = 10/group) treated for 1 min with EGCG at 10 (EGCG10) or 400 mu M (EGCG400), chlorhexidine at 0.012%, F at 1.23% (NaF), and no vehicle (placebo). Erosion was performed with Coca-Cola (R) (5 min) 4X/day during 5 days. The wear, assessed by profilometry (mean +/- SD, mu m), was significantly reduced by the gels containing MMP inhibitors (0.05 +/- 0.02(a), 0.04 +/- 0.02(a), and 0.05 +/- 0.02(a) for EGCG10, EGCG400, and chlorhexidine, respectively) when compared with NaF (0.79 +/- 0.35(b)) and placebo gels (1.77 +/- 0.35(b)) (Friedman and Dunn`s tests, p < 0.01). The use of gels delivering MMP inhibitors was shown to prevent erosion and opens a new perspective for protection against dental erosion.

The aims of this study were: (1) to correlate surface (SH) and cross-sectional hardness (CSH) with microradiographic parameters of artificial enamel lesions; (2) to compare lesions prepared by different protocols. Fifty bovine enamel specimens were allocated by stratified randomisation according to their initial SH values to five groups and lesions produced by different methods: MC gel (methylcellulose gel/lactic acid, pH 4.6, 14 days); PA gel (polyacrylic acid/lactic acid/hydroxyapatite, pH 4.8, 16 h); MHDP (undersaturated lactate buffer/methyl diphosphonate, pH 5.0, 6 days); buffer (undersaturated acetate buffer/fluoride, pH 5.0, 16 h), and pH cycling (7 days). SH of the lesions (SH(1)) was measured. The specimens were longitudinally sectioned and transverse microradiography (TMR) and CSH measured at 10- to 220-mu m depth from the surface. Overall, there was a medium correlation but non-linear and variable relationship between mineral content and root CSH. root SH(1) was weakly to moderately correlated with surface layer properties, weakly correlated with lesion depth but uncorrelated with integrated mineral loss. MHDP lesions showed the highest subsurface mineral loss, followed by pH cycling, buffer, PA gel and MC gel lesions. The conclusions were: (1) CSH, as an alternative to TMR, does not estimate mineral content very accurately, but gives information about mechanical properties of lesions; (2) SH should not be used to analyse lesions; (3) artificial caries lesions produced by the protocols differ, especially considering the method of analysis. Aluminium (At) tolerance in plants may be conferred by reduced binding of Al in the cell wall through low root cation exchange capacity (CEC) or by organic acid exudation. Root CEC is related to the degree of esterification (DE) of pectin in the cell wall, and pectin hydrolysis plays a role in cell expansion. Therefore, it was hypothesised that Al-tolerant plants with a low root CEC maintain pectin hydrolysis in the presence of Al, allowing cell expansion to continue. Irrespective of the DE, binding of Al to pectin reduced the enzymatic hydrolysis of Al-pectin gels by polygalacturonase (E.C. 3.2.1.15). Pectin gels with calcium (Ca) were slightly hydrolysed by polygalacturonase. It was concluded, therefore, that Al tolerance conferred by low root CEC is not mediated by the ability to maintain pectin hydrolysis. Citrate and malate, but not acetate, effectively dissolved Al-pectate gel and led to hydrolysis of the dissolved pectin by polygalacturonase. The organic acids did not dissolve Ca-pectate, nor did they increase pectin hydrolysis by polygalacturonase. It was concluded that exudation of some organic acids can remove Al bound to pectin and this could alleviate toxicity, constituting a tolerance mechanism. We investigate the thermodynamics and percolation regimes of model binary mixtures of patchy colloidal particles. The particles of each species have three sites of two types, one of which promotes bonding of particles of the same species while the other promotes bonding of different species. We find up to four percolated structures at low temperatures and densities: two gels where only one species percolates, a mixed gel where particles of both species percolate but neither species percolates separately, and a bicontinuous gel where particles of both species percolate separately forming two interconnected networks. The competition between the entropy and the energy of bonding drives the stability of the different percolating structures. Appropriate mixtures exhibit one or more connectivity transitions between the mixed and bicontinuous gels, as the temperature and/or the composition changes.

A simple protocol is described for the silver staining of polyacrylamide gradient gels used for the separation of restriction fragments of kinetoplast DNA [schizodeme analysis of trypanosomatids (Morel et al., 1980)]. The method overcomes the problems of non-uniform staining and strong background color which are frequently encountered when conventional protocols for silver staining of linear gels. The method described has proven to be of general applicability for DNA, RNA and protein separations in gradient gels.

Treball de recerca realitzat per un alumne d'ensenyament secundari i guardonat amb un Premi CIRIT per fomentar l'esperit científic del Jovent l'any 2009. Aquest treball de recerca es basa en l'experimentació i, posteriorment, l'obtenció i anàlisi de resultats de l'experiment creador d'anells de Liesegang. Aquest experiment, consistent en la precipitació d'un compost en una base gelificada formant anells distanciats logarítmicament els uns dels altres, ha estat durant més d'un segle objecte d'investigació de moltíssims científics, els quals no han sabut mai treure'n una explicació lògica i raonable d'aquest rar comportament. L'autor ha pretès recrear els curiosos anells intentant formar-los amb diferents inhibidors i compostos als trobats en la bibliografia. Després de realitzar més d'una trentena d'experiments, s'ha realitzat una anàlisi exhaustiva dels resultats. Aquest apartat ha estat un dels més enriquidors, ja que s'han dut a terme en ell comparacions sorprenents i troballes molt curioses, com per exemple la similitud entre els anells de Liesegang i les estructures de Turing, la qual intenta explicar les formes presents en els ocels dels éssers vius; i l'aparició d'anells de Liesegang segons l’òptica visual, efecte inexistent en l’àmplia bibliografia consultada. A més a més, també s'han efectuat una sèrie d'estudis: un en què es confirmen les distàncies logarítmiques entre els anells i on es realitza una comparació entre les dades empíriques i el patró matemàtic; i un altre en què s'estudia el comportament dels anells al variar els factors que regulen la velocitat de reacció.

Several different sample preparation methods for two-dimensional electrophoresis (2-DE) analysis of Leishmania parasites were compared. From this work, we were able to identify a solubilization method using Nonidet P-40 as detergent, which was simple to follow, and which produced 2-DE gels of high resolution and reproducibility.

OBJECTIVE To analyze the effectiveness of 2% and 4% papain gels in tissue repair of venous ulcers. METHOD Quasi-experimental study with consecutive sample of 16 patients with 30 venous ulcers treated at the outpatient clinic of a teaching hospital, from April to November in 2011, using a form for clinical assessment of the patient and its lesion. Variables were analyzed by Wilcoxon and McNemar test (p < 0.05). RESULTS Most participants were female; aged between 51 and 59 years; obese; with hypertension. Regarding ulcers, there was an average decrease of 7.9 cm2 (50% of its original size) in 90 days; 20% of the ulcers completely healed within 56.67 days. There was an increase in epithelialization, significant reduction in the slough and edema, improved depth, in the type and amount of exudate (p < 0.0001). CONCLUSION 2% and 4% papain gels were effective in healing venous ulcers.

Pressure-driven and temperature-driven transitions of two thermoresponsive polymers, poly(N-isopropylacrylamide) (pNIPAM) and poly(N-vinylisobutyramide) (pNVIBA)), in both a soluble linear polymer form and a cross-linked hydro-gel form, were examined by a dynamic light-scattering method and direct microscopic observation, respectively. Their behavior was compared with that of protein systems. Changes in some characteristic parameters in the time-intensity correlation functions of dynamic light-scattering measurement of aqueous solutions of pNIPAM at various pressures and temperatures showed no essential differences during temperature and pressure scanning and, as a whole, the motions of polymers in aqueous solutions were similar in two types of transitions until chain shrinkage occurred. The gels (cross-linked polymer gels) prepared from the thermoresponsive polymers also showed similar volume transitions responding to the pressure and temperature increase. In temperature transitions, however, gels showed drastic volume shrinkage with loss of transparency, while pressure-induced transition showed a slow recovery of transparency while keeping the size, after first transient drastic volume shrinkage with loss of transparency. At a temperature slightly higher than the transition under atmospheric temperature, so-called reentry of the volume change and recovery of the transparency were observed during the pressure-increasing process, which implies much smaller aggregation or non-aggregated collapsed polymer chains in the gel at higher pressures, indicating a certain mechanistic difference of the dehydration processes induced by temperature and pressure.

In this Master’s Thesis work the rheological properties of different polysaccharide gels have been studied. The results of this study are used as a starting point for further investigations of potential applications. In order to understand rheological behavior of studied materials, the commercial hydrocolloids such as sodium carboxymethyl cellulose, xanthan gum and guar gum were used as reference and comparison material for rheological studies. As a part the rheological research the development and implementation of proper measurement methods for studied materials were carried out. In the literature review, short introductions of studied materials and application areas of rheological modifiers are summarized. In addition, basic rheological concepts and key fundamentals are explained. In the experimental part the focus was on the rheological characterization of aqueous suspensions of studied materials. Especially, gel strength and solution stability were investigated. The rheological measurements included both rotational and oscillatory measurements in different conditions, where several chemical and physical properties were measured with Anton Paar MCR302 dynamic rotational rheometer. Studied polysaccharide gels can be clearly defined to be shear thinning and thixotropic materials. They have strong gel forming properties even at low concentrations, which explains the superior thickening behavior for some of the samples. Along with rheological characterization of selected materials the factors behind different phenomena were investigated. To reveal value and potential use of polysaccharide gels the influence of various factors such as concentration, temperature and ionic strength were determined. The measurements showed a clear difference between studied materials under investigated external parameters.
