A generalized Gierer-Meinhardt model has been used to account for the transplantation experiments in Hydra. In this model, a cross inhibition between the two organizing centres (namely, head and foot) are assumed to be the only mode of interaction in setting up a stable morphogen distribution for the pattern formation in Hydra.

Mycotoxins are secondary metabolites of filamentous fungi. They pose a health risk to humans and animals due to their harmful biological properties and common occurrence in food and feed. Liquid chromatography/mass spectrometry (LC/MS) has gained popularity in the trace analysis of food contaminants. In this study, the applicability of the technique was evaluated in multi-residue methods of mycotoxins aiming at simultaneous detection of chemically diverse compounds. Methods were developed for rapid determination of toxins produced by fungal genera of Aspergillus, Fusarium, Penicillium and Claviceps from cheese, cereal based agar matrices and grains. Analytes were extracted from these matrices with organic solvents. Minimal sample clean-up was carried out before the analysis of the mycotoxins with reversed phase LC coupled to tandem MS (MS/MS). The methods were validated and applied for investigating mycotoxins in cheese and ergot alkaloid occurrence in Finnish grains. Additionally, the toxin production of two Fusarium species predominant in northern Europe was studied. Nine mycotoxins could be determined from cheese with the method developed. The limits of quantification (LOQ) allowed the quantification at concentrations varying from 0.6 to 5.0 µg/kg. The recoveries ranged between 96 and 143 %, and the within-day repeatability (as relative standard deviation, RSDr) between 2.3 and 12.1 %. Roquefortine C and mycophenolic acid could be detected at levels of 300 up to 12000 µg/kg in the mould cheese samples analysed. A total of 29 or 31 toxins could be analysed with the method developed for agar matrices and grains, with the LOQs ranging overall from 0.1 to 1250 µg/kg. The recoveries ranged generally between 44 and 139 %, and the RSDr between 2.0 and 38 %. Type-A trichothecenes and beauvericin were determined from the cereal based agar and grain cultures of F. sporotrichioides and F. langsethiae. T-2 toxin was the main metabolite, the average levels reaching 22000 µg/kg in the grain cultures after 28 days of incubation. The method developed for ten ergot alkaloids from grains allowed their quantification at levels varying from 0.01 to 10 µg/kg. The recoveries ranged from 51 to 139 %, and the RSDr from 0.6 to 13.9 %. Ergot alkaloids were measured in barley and rye at average levels of 59 and 720 µg/kg, respectively. The two most prevalent alkaloids were ergocornine and ergocristine. The LC/MS methods developed enabled rapid detection of mycotoxins in such applications where several toxins co-occurred. Generally, the performance of the methods was good, allowing reliable analysis of the mycotoxins of interest with sufficiently low quantification limits. However, the variation in validation results highlighted the challenges related to optimising this type of multi-residue methods. New data was obtained about the occurrence of mycotoxins in mould cheeses and of ergot alkaloids in Finnish grains. In addition, the study revealed the high mycotoxin-producing potential of two common fungi in Finnish crops. The information can be useful when risks related to fungal and mycotoxin contamination will be assessed.

El siguiente trabajo está realizado sobre un proyecto de inversión referido a la incorporación de un hotel boutique de lujo como alternativa innovadora y de gran calidad en el rubro hotelero. En este sentido, la idea del proyecto es lograr que el viajero tenga una experiencia única alojándose en este hotel, apelando a despertar todos los sentidos. El desafío es lograr un hotel con identidad y carácter en donde se descubra la verdadera esencia del lugar.

El presente Proyecto de Fin de Máster consiste en crear una herramienta software capaz de monitorizar y gestionar la actividad de Hydra, una herramienta de gestión de entornos distribuidos, para que su estrategia de balanceo de carga se adecúe al modelo creado por GloBeM, una metodología de análisis de entornos distribuidos. GloBeM, que es una metodología externa, puede analizar y crear un modelo de máquina de estados finitos a partir de un sistema distribuido concreto. Hydra, una herramienta también externa, es un sistema de gestión de entornos cloud recientemente desarrollado y de código abierto, con un sistema de balanceo de carga efectivo pero algo limitado. El software construido recoge el modelo creado por GloBeM y lo analiza. A partir de ahí, monitoriza en tiempo real y a una frecuencia determinada la actividad de Hydra y el sistema cloud que ésta gestiona, y reconfigura sus parámetros para que su desempeño se ciña a lo estipulado por el modelo de GloBeM, extendiendo así el sistema de balanceo de carga original de Hydra.---ABSTRACT---This Master's Thesis Project involves creating a software able to monitor and manage the activity of Hydra, a tool for managing distributed environments, in order to adjust its load balancing strategy to the model created by GloBeM, an analysis methodology for distributed environments. GloBeM, which is an external methodology, can analyse and create a finite-state machine model from a particular cloud system. Hydra, also an external tool, is an open source management system for cloud environments recently developed, with a relatively limited system of load balancing. The created software gets the model created by GloBeM as an input and analyses it. From there, it monitors in real time and at a certain frequency Hydra’s activity and the cloud system that it manages, and reconfigures its parameters to adjust its performance to the stipulations by the GloBeM’s model, extending Hydra's original load balancing system.

Pax proteins are a family of transcription factors with a highly conserved paired domain; many members also contain a paired-type homeodomain and/or an octapeptide. Nine mammalian Pax genes are known and classified into four subgroups: Pax-1/9, Pax-2/5/8, Pax-3/7, and Pax-4/6. Most of these genes are involved in nervous system development. In particular, Pax-6 is a key regulator that controls eye development in vertebrates and Drosophila. Although the Pax-4/6 subgroup seems to be more closely related to Pax-2/5/8 than to Pax-3/7 or Pax-1/9, its evolutionary origin is unknown. We therefore searched for a Pax-6 homolog and related genes in Cnidaria, which is the lowest phylum of animals that possess a nervous system and eyes. A sea nettle (a jellyfish) genomic library was constructed and two pax genes (Pax-A and -B) were isolated and partially sequenced. Surprisingly, unlike most known Pax genes, the paired box in these two genes contains no intron. In addition, the complete cDNA sequences of hydra Pax-A and -B were obtained. Hydra Pax-B contains both the homeodomain and the octapeptide, whereas hydra Pax-A contains neither. DNA binding assays showed that sea nettle Pax-A and -B and hydra Pax-A paired domains bound to a Pax-5/6 site and a Pax-5 site, although hydra Pax-B paired domain bound neither. An alignment of all available paired domain sequences revealed two highly conserved regions, which cover the DNA binding contact positions. Phylogenetic analysis showed that Pax-A and especially Pax-B were more closely related to Pax-2/5/8 and Pax-4/6 than to Pax-1/9 or Pax-3/7 and that the Pax genes can be classified into two supergroups: Pax-A/Pax-B/Pax-2/5/8/4/6 and Pax-1/9/3/7. From this analysis and the gene structure, we propose that modern Pax-4/6 and Pax-2/5/8 genes evolved from an ancestral gene similar to cnidarian Pax-B, having both the homeodomain and the octapeptide.

In response to 9/11, the U.S. launched airstrikes in Afghanistan against al Qaeda to diminish its ability to conduct future attacks. Next, the U.S. turned its attention to Iraq. A series of problems and missteps there subsequently created an opportunity for al Qaeda to evolve, aligning itself with active insurgencies and other terrorist groups around the world. This paper identifies the largely preemptive strategies employed by the U.S. and analyzes how those efforts positively and negatively impacted the War on Terror. While some significant gains were made, the results show that al Qaeda remains a threat to the international community and demands more effective U.S. and international counter-radicalization strategies, including diplomacy and aid, to curb its global appeal.
