FISH has been used as a complement to classical cytogenetics in the detection of mosaicism in sex chromosome anomalies. The aim of this study is to describe three cases in which the final diagnosis could only be achieved by FISH. Case 1 was an 8-year-old 46,XY girl with normal female genitalia referred to our service because of short stature. FISH analysis of lymphocytes with probes for the X and Y centromeres identified a 45,X/46,X,idic(Y) constitution, and established the diagnosis of Turner syndrome. Case 2 was a 21-month-old 46,XY boy with genital ambiguity (penile hypospadias, right testis, and left streak gonad). FISH analysis of lymphocytes and buccal smear identified a 45,X/46,XY karyotype, leading to diagnosis of mixed gonadal dysgenesis. Case 3 was a 47,XYY 19-year-old boy with delayed neuromotor development, learning disabilities, psychological problems, tall stature, small testes, elevated gonadotropins, and azoospermia. FISH analysis of lymphocytes and buccal smear identified a 47,XYY/48,XXYY constitution. Cases 1 and 2 illustrate the phenotypic variability of the 45,X/46,XY mosaicism, and the importance of detection of the 45,X cell line for proper management and follow-up. In case 3, abnormal gonadal function could be explained by the 48,XXYY cell line. The use of FISH in clinical practice is particularly relevant when classical cytogenetic analysis yields normal or uncertain results in patients with features of sex chromosome aneuploidy. O síndroma de Turner (TS) tem sido descrito em associação com diversas anomalias dos cromossomas sexuais. Embora a maioria dos individuos com TS não apresentem evidência citogenética de sequências do cromossoma Y, diferentes autores consideram que algumas doentes com TS podem possuir uma linha celular minoritária contendo material do cromossoma Y, que não é detectada pela análise citogenética convencional. A identificação de moisacismos minoritários ou subrepresentados contendo o cromossoma Y é de importância fundamental em termos clínicos devido ao risco aumentado que estas doentes possuem para desenvolvimento de gonadoblastoma. No presente estudo procedeu-se à análise citogenética convencional de linfócitos de sangue periférico obtidos de 22 doentes com TS. Destas doentes, doze possuíam cariotipo 45,X, em sete foram detectados mosaicos com ou sem anomalias estruturais do cromossoma X e nas restantes três foram identificados os seguintes cariotipos: 46,X,i (X)(q10); 46,X,+mar/47,X,idic(Y),+mar e 45,X/46,X,+r. Os estudos moleculares foram realizados em DNA genómico obtido a partir de linfócitos de sangue periférico e de células de mucosa bucal, dois tecidos que derivam de folhetos embrionários diferentes, respectivamente, mesoderme e ectoderme. A pesquisa de moisacismos minoritários envolvendo o cromossoma Y foi efectuada por PCR simples e PCR nested para os seguintes loci específicos do cromossoma Y: SRY (sex determining region Y), TSPY (testis specific protein Y encoded), DYZ3 (locus centromérico) e DAZ1 (deleted in azoospermia). O uso de STSs localizados nos braços curto e longo do cromossoma Y permitiu a caracterização de um idic (Y)e de um cromossoma em anel, detectados em duas das doentes estudadas. A eleveda sensibilidade da PCR nested (1 célila masculina/125 000 células femininas) permitiu excluir a presença de moisacismos minoritários do Y em 20 das 22 doentes com TS. Na doente com um idic(Y) a análise por PCR simples foi posistiva para todos os loci estudados com excepção da região heterocromática. Este resultado permitiu identificar o ponto de quebra no braço longo entre sY158 e sY159, tendo-se confirmado por hibridação in situ de fluorescência (FISH), a duplicação da eurocromatina do braço longo, centrómero e braço curto do cromossoma Y. A caracterização do cromossoma em anel, detectado num das doentes com TS só foi possível por FISH e por PCR. Neste cromossoma, derivado de Y, foi detectada, no braço curto, a delecção da região pseudoautossómica 1 (PARY1)e, no braço longo, a delecção dos intervalos 6 e 7. Contudo, o referido cromossoma foi positivo para os loci SRY, RPS4Y, AMGY, TSPY localizados no braço curto, DYZ3 (centrómero) e, sY85, DFFRY, GY6, sY87, sY113, sY119, sY122, sY126 e RBMY1 localizados no braço longo do cromossoma Y. Este estudo permitiu, assim, excluir a presença de moisacismos minoritários do cromossoma Y em dois tecidos obtidos de 20 doentes com TS, e caracterizar por FISH e análise molecular, um idic(Y) e um cromossoma em anel, em que a natureza deste último não tinha sido identificada por análise citogenética convencional. O risco elevado de desenvolvimento de gonadoblastoma nos indivíduos com TS que possuem sequências do cromossoma Y justifica a aplicação de FISH e PCR para a caracterização de cromossomas marcadores e a utilização de PCR nested para a detecção de moisacismos minoritários do Y sempre que o material deste cromossoma não seja detectado pela análise citogenética convencional em doentes com cariotipo 45,X e/ou virilização.

Os biofilmes podem ser definidos como uma comunidade complexa, constituída principalmente por água, microrganismos e substâncias poliméricas extracelulares (EPS), estando aderidos a uma superfície sólida. Um biofilme pode ser constituído por uma única espécie ou, na maioria dos casos, por inúmeras espécies, sendo as bactérias, fungos, algas e protozoários os principais microrganismos, embora as bactérias representem o grupo dominante. Os biofilmes desempenham um papel bastante relevante nos sistemas de biomassa fixa no tratamento de águas residuais, oferecendo diversas vantagens. Assim, o seu estudo e a compreensão das suas características torna-se essencial para a garantia de eficiências elevadas nestes sistemas. O presente trabalho pretendeu contribuir para o estudo de biofilmes de diferentes idades, nomeadamente na identificação e caracterização dos microrganismos dominantes. A idade do biofilme é um parâmetro fundamental na exploração de sistemas de biomassa fixa, uma vez que já se demonstrou ser vantajoso no controlo da espessura do biofilme. O crescimento dos biofilmes foi possível devido a um dispositivo experimental concebido para se obter biofilmes de determinada espessura (ou idade). O método adoptado neste estudo para a identificação dos microrganismos foi o FISH (Hibridação in situ de fluorescência), uma vez que este permite a identificação, quantificação e localização espacial de microrganismos directamente nos seus habitats naturais. Através dos resultados obtidos, pôde constatar-se que a idade de biofilme de 0,5 – 1 dia apresentou maiores diferenças no perfil microbiano comparativamente com as restantes idades (4 – 5 dias e 5 – 6 dias), parecendo demonstrar uma menor abundância de tipos de bactérias. Também se concluiu que, para a idade de biofilme menor (0,5 – 1 dia), as bactérias presentes em maior abundância foram as Gammaproteobacteria, para a idade de 4 a 5 dias foram as Betaproteobacteria, Cytophaga-Flavobacteria e Actinobacteria, e para a idade de 5 a 6 dias foram as Betaproteobacteria e Cytophaga-Flavobacteria. Os resultados obtidos possibilitaram um conhecimento mais aprofundado dos microrganismos presentes em biofilmes de idades distintas, podendo contribuir para a optimização de sistemas de biomassa fixa. Assim, se se adoptar este tipo de conhecimento em ETAR por sistemas de biomassa fixa no tratamento de águas residuais, poder-se-á contribuir para a melhoria do funcionamento da instalação, principalmente no que diz respeito à sua eficiência de tratamento.

Human multipotent mesenchymal stromal cells (MSCs), also known as mesenchymal stem cells, have become an important and attractive therapeutic tool since they are easily isolated and cultured, have in vitro expansion potential, substantial plasticity and secrete bioactive molecules that exert trophic effects. The human umbilical cord as a cell source for cell therapy will help to avoid several ethical, political, religious and technical issues. One of the main issues with SC lines from different sources, mainly those of embryonic origin, is the possibility of chromosomal alterations and genomic instability during in vitro expansion. Cells isolated from one umbilical cord exhibited a rare balanced paracentric inversion, likely a cytogenetic constitutional alteration, karyotype: 46,XY,inv(3)(p13p25~26). Important genes related to cancer predisposition and others involved in DNA repair are located in 3p25~26. Titanium is an excellent biomaterial for bone-implant integration; however, the use can result in the generation of particulate debris that can accumulate in the tissues adjacent to the prosthesis, in the local bone marrow, in the lymph nodes, liver and spleen. Subsequently may elicit important biological responses that aren´t well studied. In this work, we have studied the genetic stability of MSC isolated from the umbilical cord vein during in vitro expansion, after the cryopreservation, and under different concentrations and time of exposition to titanium microparticles. Cells were isolated, in vitro expanded, demonstrated capacity for osteogenic, adipogenic and chondrogenic differentiation and were evaluated using flow cytometry, so they met the minimum requirements for characterization as MSCs. The cells were expanded under different concentrations and time of exposition to titanium microparticles. The genetic stability of MSCs was assessed by cytogenetic analysis, fluorescence in situ hybridization (FISH) and analysis of micronucleus and other nuclear alterations (CBMN). The cells were able to internalize the titanium microparticles, but MSCs preserve their morphology, differentiation capacity and surface marker expression profiles. Furthermore, there was an increase in the genomic instability after long time of in vitro expansion, and this instability was greater when cells were exposed to high doses of titanium microparticles that induced oxidative stress. It is necessary always assess the risks/ benefits of using titanium in tissue therapy involving MSCs, considering the biosafety of the use of bone regeneration using titanium and MSCs. Even without using titanium, it is important that the therapeutic use of such cells is based on analyzes that ensure quality, security and cellular stability, with the standardization of quality control programs appropriate. In conclusion, it is suggested that cytogenetic analysis, FISH analysis and the micronucleus and other nuclear alterations are carried out in CTMH before implanting in a patient

This study evaluated the effect of indole-3-acetic acid (IAA) in the activation of goats preantral follicles (FOPA). Were used four pairs of ovaries of adult mixed breed goats. Each ovarian pair was divided into 23 fragments. One fragment was fixed for histology and other fragment was using to follicular isolation procedure. The remaining fragments were cultured in 1.0 ml of Minimum Essential Medium (MEM) or MEM supplemented with IAA at concentrations of 10, 40, 100, 500 or 1000 ng/mL. The in vitro culture was performed at 39°C in incubator with 5% CO2 for 1, 3 and 5 days. After in vitro culture was evaluated histological integrity and viability of FOPA. The addition of 100 ng/ml of IAA to MEM showed a significant increase in follicles transition in the third day of in vitro culture, characterizing follicular activation. Moreover, this concentration was obtained maintaining the histological integrity of PAF by the fifth day of in vitro culture. The viability test confirmed the results of histology. Thus, we conclude that IAA can promote the activation of goats FOPA

This in situ study investigated, using scanning electron microscopy, the effect of stimulated saliva on the enamel surface of bovine and human substrates submitted to erosion followed by brushing abrasion immediately or after one hour. During 2 experimental 7-day crossover phases, 9 previously selected volunteers wore intraoral palatal devices, with 12 enamel specimens (6 human and 6 bovine). In the first phase, the volunteers immersed the device for 5 minutes in 150 ml of a cola drink, 4 times a day (8h00, 12h00, 16h00 and 20h00). Immediately after the immersions, no treatment was performed in 4 specimens (ERO), 4 other specimens were immediately brushed (0 min) using a fluoride dentifrice and the device was replaced into the mouth. After 60 min, the other 4 specimens were brushed. In the second phase, the procedures were repeated but, after the immersions, the volunteers stimulated the salivary flow rate by chewing a sugar-free gum for 30 min. Enamel superficial alterations of all specimens were then evaluated using a scanning electron microscope. Enamel prism core dissolution was seen on the surfaces submitted to erosion, while on those submitted to erosion and to abrasion (both at 0 and 60 min) a more homogeneous enamel surface was observed, probably due to the removal of the altered superficial prism layer. For all the other variables - enamel substrate and salivary stimulation -, the microscopic pattern of the enamel specimens was similar.

The objective of this study was to assess the salivary residual effect of fluoride dentifrice on human enamel subjected to an erosive challenge. This crossover in situ study was performed in two phases (A and B), involving ten volunteers. In each phase, they wore acrylic palatal appliances, each containing 3 human enamel blocks, during 7 days. The blocks were subjected to erosion by immersion of the appliances in a cola drink for 5 minutes, 4 times a day. Dentifrice was used to brush the volunteers’ teeth, 4 times a day, during 1 minute, before the appliance was replaced into the mouth. In phases A and B the dentifrices used had the same formulation, except for the absence (PD) or presence (FD) of fluoride, respectively. Enamel alterations were determined using profilometry, microhardness (%SMHC), acid- and alkali-soluble F analysis. The data were tested using ANOVA (p < 0.05). The concentrations (mean ± SD) of alkali- and acid-soluble F (µgF/cm²) were, respectively, PD: 1.27ª ± 0.70/2.24A ± 0.36 and FD: 1.49ª ± 0.44/2.24A ± 0.67 (p > 0.05). The mean wear values (± SD, µm) were PD: 3.63ª ± 1.54 and FD: 3.54ª ± 0.90 (p > 0.05). The mean %SMHC values (± SD) were PD: 89.63ª ± 4.73 and FD: 87.28ª ± 4.01 (p > 0.05). Thus, we concluded that the residual fluoride from the fluoride-containing dentifrice did not protect enamel against erosion.

The aim of this study was to evaluate in situ changes in the alveolar crest bone height around immediate implant-supported crowns in comparison to tooth-supported crowns (control) with the cervical margins located at the bone crest level, without occlusal load. In Group I, after extraction of 12 mandibular premolars from 4 adult dogs, implants from Branemark System (MK III TiU RP 4.0 x 11.5 mm) were placed to retain complete acrylic crowns. In Group II, premolars were prepared to receive complete metal crowns. Sixteen weeks after placement of the crowns (38 weeks after tooth extraction), the height of the alveolar bone crest was measured with a digital caliper. Data were analyzed statistically by the Mann-Whitney test at 5% significance level. The in situ analysis showed no statistically significant difference (p=0.880) between the implant-supported and the tooth-supported groups (1.528 + 0.459 mm and 1.570 + 0.263 mm, respectively). Based on the findings of the present study, it may be concluded that initial peri-implant bone loss may result from the remodeling process necessary to establish the biological space, similar to which occurs with tooth-supported crowns.

OBJETIVOS: avaliar a expressão de erbB-2 e dos receptores hormonais para estrógeno e progesterona (RE/RP) nas regiões de transição entre as frações in situ e invasoras de neoplasias ductais da mama (CDIS e CDI, respectivamente). MÉTODOS: oitenta e cinco casos de neoplasias mamárias, contendo regiões contíguas de CDIS e CDI, foram selecionados. Espécimes histológicos das áreas de CDIS e de CDI foram obtidos através da técnica de tissue microarray (TMA). As expressões da erbB-2 e dos RE/RP foram avaliadas por meio de imunoistoquímica convencional. A comparação da expressão da erbB-2 e dos RE/RP nas frações in situ e invasoras da mama foi realizada com emprego do teste de McNemar. Os intervalos de confiança foram determinados em 5% (p=0,05). Foram calculados coeficientes de correlação intraclasse (ICC) para avaliar a concordância na tabulação cruzada da expressão de erbB-2 e RE/RP nas frações de CDIS e CDI. RESULTADOS: a expressão da erbB-2 não diferiu entre as áreas de CDIS e CDI (p=0,38). Comparando caso a caso suas áreas de CDIS e CDI, houve boa concordância na expressão da erbB-2 (coeficiente de correlação intraclasse, ICC=0,64), dos RP (ICC = 0,71) e dos RE (ICC = 0,64). Considerando apenas tumores cujo componente in situ apresentasse áreas de necrose (comedo), o ICC para erbB-2 foi de 0,4, comparado a 0,6 no conjunto completo de casos. Os ICC não diferiram substancialmente daqueles obtidos com o conjunto completo de espécimes em relação aos RE/RP: para RE, ICC=0,7 (versus 0,7 no conjunto completo), e para RP, ICC=0,7 (versus 0,6 no conjunto completo). CONCLUSÕES: nossos achados sugerem que as expressões de erbB-2 e RE/RP não diferem nos componentes contíguos in situ e invasivo em tumores ductais da mama.

Since 1964, the Center for Geochronological Research - CPGeo, one of the interdepartmental centers of the Instituto de Geociências (IG) of São Paulo University, has developed studies related to several geological processes associated with different rock types. Thermal Ionization Mass Spectrometry Isotopic Dilution (ID-TIMS) has been the technique widely used in the CPGeo U-Pb Laboratory. It provides reliable and accurate results in age determination of superposed events. However, the open-system behavior such as Pb-loss, the inheritance problem and metamictization processes allow and impel us to a much richer understanding of the power and limitations of U-Pb geochronology and thermochronology. In this article, we present the current methodology used at the CPGeo-IGc-USP U-Pb laboratory, the improvements on ID-TIMS method, and report high-precision U-Pb data from zircon, monazite, epidote, titanite, baddeleyite and rutile from different rock types of several domains of the Brazilian south-southeast area, Argentina and Uruguay.

Interference by autofluorescence is one of the major concerns of immunofluorescence analysis of in situ hybridization-based diagnostic assays. We present a useful technique that reduces autofluorescent background without affecting the tissue integrity or direct immunofluorescence signals in brain sections. Using six different protocols, such as ammonia/ethanol, Sudan Black B (SBB) in 70% ethanol, photobleaching with UV light and different combinations of them in both formalin-fixed paraffin-embedded and frozen human brain tissue sections, we have found that tissue treatment of SBB in a concentration of 0.1% in 70% ethanol is the best approach to reduce/eliminate tissue autofluorescence and background, while preserving the specific fluorescence hybridization signals. This strategy is a feasible, non-time consuming method that provides a reasonable compromise between total reduction of the tissue autofluorescence and maintenance of specific fluorescent labels.

The objective of this study was to evaluate the effect of a polyclonal antibody preparation (PAP) against specific ruminal bacteria on the in situ degradability of dry-grounded maize grain (DMG), high moisture maize silage (HMMS) starch and citrus pulp (CiPu) pectin. Nine ruminally cannulated cows were used in a 3 x 3 Latin square design, replicated three times in a factorial arrangement of treatments of two rumen modifiers represented by monensin and PAP plus a control group, and the three energy sources (DMG, HMMS and CiPu). Each period had 21 days, where 16 were used for adaptation to treatment and five for data collection. The group treated with PAP showed an effect on the soluble fraction (""a"") of DMG starch, decreasing it by respectively 45.3% and 45.4% compared to the CON and MON groups. No effect of PAP was observed for any in situ degradability parameters of starch from HMMS or pectin of CiPu. It was concluded that the polyclonal antibody preparation had limited effect on the in situ degradability of the tested energy sources.
