Two chromatographic methods, gas chromatography with flow ionization detection (GC–FID) and liquid chromatography with ultraviolet detection (LC–UV), were used to determine furfuryl alcohol in several kinds of foundry resins, after application of an optimised extraction procedure. The GC method developed gave feasibility that did not depend on resin kind. Analysis by LC was suitable just for furanic resins. The presence of interference in the phenolic resins did not allow an appropriate quantification by LC. Both methods gave accurate and precise results. Recoveries were >94%; relative standard deviations were ≤7 and ≤0.3%, respectively for GC and LC methods. Good relative deviations between the two methods were found (≤3%).

Phenol is a toxic compound present in a wide variety of foundry resins. Its quantification is important for the characterization of the resins as well as for the evaluation of free contaminants present in foundry wastes. Two chromatographic methods, liquid chromatography with ultraviolet detection (LC-UV) and gas chromatography with flame ionization detection (GC-FID), for the analysis of free phenol in several foundry resins, after a simple extraction procedure (30 min), were developed. Both chromatographic methods were suitable for the determination of phenol in the studied furanic and phenolic resins, showing good selectivity, accuracy (recovery 99–100%; relative deviations <5%), and precision (coefficients of variation <6%). The used ASTM reference method was only found to be useful in the analysis of phenolic resins, while the LC and GC methods were applicable for all the studied resins. The developed methods reduce the time of analysis from 3.5 hours to about 30 min and can readily be used in routine quality control laboratories.

An amperometric biosensor based on cholinesterase (ChE) has been used for the determination of selected carbamate insecticides in vegetable samples. The linear range of the biosensor for the N-methylcarbamates (aldicarb, carbaryl, carbofuran, methomyl and propoxur) varied from 5 x 10(-5) to 50 mg kg(-1). Limits of detection were calculated on the basis that the ChE enzymes were 10% inhibited and varied, depending of the combination ChE (as acetyl- or butyrylcholinesterase) vs. inhibitor (pesticide), from 1 x 10(-4) to 3.5 mg kg(-1). The biosensor-based carbamate determination was compared to liquid chromatography/UV methods. Three vegetable samples were spiked with carbofuran and propoxur at 125 mu g kg(-1) followed by conventional procedures. Good correlations were observed for carbofuran in the vegetable extracts (79, 96 and 91% recoveries for potato, carrot and sweet pepper, respectively), whereas for propoxur unsatisfactory results were obtained. Potato and carrot samples were spiked with 10, 50 and 125 mu g kg(-1) carbofuran, followed by direct determination by the amperometric biosensor. The fortified sampler; resulted in very high inhibition values, and recoveries were: 28, 34 and 99% for potato, and 140, 90 and 101% for carrot, respectively, at these three fortification levels. In recent years the interest in naturally occurring compounds has been increasing worldwide. Indeed, many of the bioactive compounds currently used as medicines have been synthesized based on the structure of natural compounds [1]. In order to obtain bioactive fractions and subsequently isolated compounds derived from natural matrices, several procedures have been carried out. One of these is to separate and assess the concentration of the active compound(s) present in the samples, a step in which the chromatographic techniques stand out [2]. In the present work the mushroom Sui/Ius granulatus (L.) Roussel was chemically characterized by chromatographic techniques coupled to different detectors, in order to evaluate the presence of nutritional and/or bioactive molecules. Some hydrophilic compounds, namely free sugars, were identified by high performance liquid chromatography coupled to a refraction index detector (HPLC-RI), and organic and phenolic acids were assessed by HPLC coupled to a photodiode array detector (HPLC-PDA). Regarding lipophilic compounds, fatty acids weredetermined by gas chromatography with a flame ionization detector (GC-FID) and tocopherols by HPLC-fluorescence detection. Mannitol and trehalose were the main free sugars detected. Different organic acids were also identified (i.e. oxalic, quinic and fumaric acids), as well as phenolic acids (i.e. gallic and p-hydroxybenzoic acids) and the related compound cinnamic acid. Mono- and polyunsaturated fatty acids were the prevailing fatty acids and a-, ~- and ~-tocopherol were the isoforms of vitamin E detected in the samples. Since this species proved to be a source of biologically active compounds, the antioxidant and antimicrobial properties were evaluated. The antioxidant activity was measured through the reducing power, free radical's scavenging activity and lipid peroxidation inhibition of its methanolic extract, and the antimicrobial activity was also tested in Gram positive and Gram negative bacteria and iri different fungi. S. granulatus presented antioxidant properties in all the performed assays, and proved to inhibit the growth of different bacterial and fungal strains. This study is a first step for classifying S. granulatus as a functional food, highlighting the potential of mushrooms as a source of nutraceutical and biologically active compounds.

After harvest, plants remain living organisms with the capacity to carry out metabolic processes. Thus, from the moment they are detached from the source of nutrients, they become entirely dependent on their own organic reserves [1]. Postharvest changes cannot be stopped, but they can be slowed within certain limits. Therefore, this study was conducted to evaluate the effects induced by storage in the profiles of sugars, organic acids and tocopherols of two leafy vegetables. Wild samples of watercress (Nasturtium officinale R. Br.) and buckler sorrel (Rumex induratus Boiss. & Reut.), from the Northeastern region of Portugal, were analyzed after harvest (control) and after storage in sterilized packages (using the passive modification mode) at 4ºC for 7 or 12 days, respectively. Analyses were performed by high-performance liquid chromatography (HPLC) using different detectors, i.e., a refraction index detector (RID) for free sugars, a photodiode array detector (PDA) for organic acids, and a fluorescence (FP) detector for tocopherols. The storage time decreased the levels of fructose, glucose and total sugars in both leafy vegetables and increased the total organic acids content. The decrease of these sugars can be related to its use by the plant to produce the required energy. Ascorbic acid was detected in buckler sorrel and decreased with storage; while the amount of malic acid increased in both species. Curiously, all the tocopherol isoforms increased in watercress, while buckler sorrel just present higher values of γ- and δ- tocopherols. In fact, the de novo synthesis of these bioactives compounds can be a plant strategy to fight against the reactive species that are produced during storage. The knowledge of the behavior of these compounds during storage that was achieved with this study [2] may contribute to the development of more effective preservation strategies for leafy vegetables.

Smokeless powder additives are usually detected by their extraction from post-blast residues or unburned powder particles followed by analysis using chromatographic techniques. This work presents the first comprehensive study of the detection of the volatile and semi-volatile additives of smokeless powders using solid phase microextraction (SPME) as a sampling and pre-concentration technique. Seventy smokeless powders were studied using laboratory based chromatography techniques and a field deployable ion mobility spectrometer (IMS). The detection of diphenylamine, ethyl and methyl centralite, 2,4-dinitrotoluene, diethyl and dibutyl phthalate by IMS to associate the presence of these compounds to smokeless powders is also reported for the first time. A previously reported SPME-IMS analytical approach facilitates rapid sub-nanogram detection of the vapor phase components of smokeless powders. A mass calibration procedure for the analytical techniques used in this study was developed. Precise and accurate mass delivery of analytes in picoliter volumes was achieved using a drop-on-demand inkjet printing method. Absolute mass detection limits determined using this method for the various analytes of interest ranged between 0.03–0.8 ng for the GC-MS and between 0.03–2 ng for the IMS. Mass response graphs generated for different detection techniques help in the determination of mass extracted from the headspace of each smokeless powder. The analyte mass present in the vapor phase was sufficient for a SPME fiber to extract most analytes at amounts above the detection limits of both chromatographic techniques and the ion mobility spectrometer. Analysis of the large number of smokeless powders revealed that diphenylamine was present in the headspace of 96% of the powders. Ethyl centralite was detected in 47% of the powders and 8% of the powders had methyl centralite available for detection from the headspace sampling of the powders by SPME. Nitroglycerin was the dominant peak present in the headspace of the double-based powders. 2,4-dinitrotoluene which is another important headspace component was detected in 44% of the powders. The powders therefore have more than one headspace component and the detection of a combination of these compounds is achievable by SPME-IMS leading to an association to the presence of smokeless powders.

Frutos in natura de C. xanthocarpa (Myrtaceae), coletados em diferentes estádios de amadurecimento na Floresta Estadual de Assis, município de Assis - SP, foram avaliados por meio de métodos tradicionais de análise, técnicas cromatográficas e de espectrometria de massas e de emissão ótica quanto à composição nutricional, ao perfil químico do óleo volátil e ao teor de elementos inorgânicos. Os resultados mostraram alto teor de água (81,4%); lipídios (1,9%); carboidratos totais (8,9%); fibra alimentar (6,3%); além de quantidades razoáveis de ácido ascórbico (17,8 mg.100 g-1); e traços de riboflavina (0,09 mg.100 g-1). No óleo volátil (0,2%), pôde-se identificar 62 componentes, correspondendo a 100% dos constituintes do óleo, destacando-se dentre eles os monoterpenos α-pineno (15%), o-cimeno (10,8%), β-pineno (10,5%). Entre os minerais (16), os principais elementos foram o K (2084 mg.kg-1), P (149 mg.kg-1), Mg (135 mg.kg-1) e, como microelementos, o Fe (6,4 mg.kg-1), Cu (93,3 mg.100 g-1) e Pb (1,3 mg.kg-1). O valor energético do fruto (57,3 kcal.100 g-1) deve-se quase que exclusivamente aos teores de carboidratos totais (8,9%).
Isolation and analysis of bioactive isoflavonoids and chalcone from a new type of Brazilian propolis

Activity-directed fractionation and purification processes were employed to identify isoflavonoids with antioxidant and antimicrobial activities from Brazilian red propolis. Crude propolis was extracted with ethanol (80%. v/v) and fractioned by liquid-liquid extraction technique using hexane and chloroform. Since chloroform fraction showed strong antioxidant and antimicrobial activities it was purified and isolated using various chromatographic techniques. Comparing our spectral data (UV, NMR, and mass spectrometry) with values found in the literature, we identified two bioactive isoflavonoids (vestitol and neovestitol), together with one chalcone (isoliquiritigenin). Vestitol presented higher antioxidant activity against beta-carotene consumption than neovestitol. The antimicrobial activity of these three compounds against Staphylococcus aureus, Streptococcus mutans, and Actinomyces naeslundii was evaluated and we concluded that isoliquiritigenin was the most active one with lower MIC, ranging from 15.6 to 62.5 mu g/mL. Our results showed that Brazilian red propolis has biologically active isoflavonoids that may be used as a mild antioxidant and antimicrobial for food preservation. (C) 2010 Elsevier B.V. A presente dissertação tem como principal propósito avaliar o desempenho energético e a qualidade do ar interior do edifício principal do Parque Biológico de Vila Nova de Gaia (PBG). Para esse efeito, este estudo relaciona os termos definidos na legislação nacional em vigor até à presente data, e referentes a esta área de atuação, em particular, os presentes no SCE, RSECE, RCCTE e RSECE-QAI. Para avaliar o desempenho energético, procedeu-se numa primeira fase ao processo de auditoria no local e posteriormente à realização de uma simulação dinâmica detalhada, cuja modelação do edifício foi feita com recurso ao software DesignBuilder. Após a validação do modelo simulado, por verificação do desvio entre os consumos energéticos registados nas faturas e os calculados na simulação, igual a 5,97%, foi possível efetuar a desagregação dos consumos em percentagem pelos diferentes tipos de utilizações. Foi também possível determinar os IEE real e nominal, correspondendo a 29,9 e 41.3 kgep/m2.ano, respetivamente, constatando-se através dos mesmos que o edifício ficaria dispensado de implementar um plano de racionalização energética (PRE) e que a classe energética a atribuir é a C. Contudo, foram apresentadas algumas medidas de poupança de energia, de modo a melhorar a eficiência energética do edifício e reduzir a fatura associada. Destas destacam-se duas propostas, a primeira propõe a alteração do sistema de iluminação interior e exterior do edifício, conduzindo a uma redução no consumo de eletricidade de 47,5 MWh/ano, com um período de retorno de investimento de 3,5 anos. A segunda está relacionada com a alteração do sistema de produção de água quente para o aquecimento central, através do incremento de uma caldeira a lenha ao sistema atual, que prevê uma redução de 50 MWh no consumo de gás natural e um período de retorno de investimento de cerca de 4 anos. Na análise realizada à qualidade do ar interior (QAI), os parâmetros quantificados foram os exigidos legalmente, excetuando os microbiológicos. Deste modo, para os parâmetros físicos, temperatura e humidade relativa, obtiveram-se os resultados médios de 19,7ºC e 66,9%, respetivamente, ligeiramente abaixo do previsto na legislação (20,0ºC no período em que foi feita a medição, inverno). No que diz respeito aos parâmetros químicos, os valores médios registados para as concentrações de dióxido de carbono (CO2), monóxido de carbono (CO), ozono (O3), formaldeído (HCHO), partículas em suspensão (PM10) e radão, foram iguais a 580 ppm, 0,2 ppm, 0,06 ppm, 0,01 ppm, 0,07 mg/m3 e 196 Bq/m3, respetivamente, verificando-se que estão abaixo dos valores máximos de referência presentes no regulamento (984 ppm, 10,7 ppm, 0,10 ppm, 0,08 ppm, 0,15 mg/m3 e 400 Bq/m3). No entanto, o parâmetro relativo aos compostos orgânicos voláteis (COV) teve um valor médio igual a 0,84 ppm, bastante acima do valor máximo de referência (0,26 ppm). Neste caso, terá que ser realizada uma nova série de medições utilizando meios cromatográficos, para avaliar qual(ais) são o(s) agente(s) poluidor(es), de modo a eliminar ou atenuar as fontes de emissão.

Background Over the years, food industry wastes have been the focus of a growing interest due to their content in high added-value compounds. A good example are the olive oil by-products (OOBP), which retain a great amount of phenolic compounds during olive oil production. Their structure and biological properties justify their potential use as antioxidants in other food products. The efficient recovery of phenolic compounds has been extensively studied and optimized in order to maximize their reintroduction in the food chain and contribute to a higher valorization and better management of wastes from olive oil industry. Scope and approach This paper reviews the most representative phenolic compounds described in OOBP and their biological properties. New extraction procedures to efficiently recover these compounds and the most advanced chromatographic techniques that have been used for a better understanding of the phenolic profile of these complex matrices are also referred. Finally, this paper reports the main applications of OOBP, with emphasis on their phenolic content as natural antioxidants for food applications. Key findings and conclusions Besides their antioxidant activity, phenolic compounds from OOBP have also shown antimicrobial and antitumoral properties. Their application as food antioxidants requires new extraction techniques, including the use of non-toxic solvents and, in a pilot scale, the use of filters and adsorbent resins. The inclusion of phenolic compounds from OOBP in some food matrices have improved not only their antioxidant capacity but also their sensory attributes.
