Knowledge of the native prokaryotes in hazardous locations favors the application of biotechnology for bioremediation. Independent strategies for cultivation and metagenomics contribute to further microbiological knowledge, enabling studies with non-cultivable about the "native microbiological status and its potential role in bioremediation, for example, of polycyclic aromatic hydrocarbons (HPA's). Considering the biome mangrove interface fragile and critical bordering the ocean, this study characterizes the native microbiota mangrove potential biodegradability of HPA's using a biomarker for molecular detection and assessment of bacterial diversity by PCR in areas under the influence of oil companies in the Basin Petroleum Geology Potiguar (BPP). We chose PcaF, a metabolic enzyme, to be the molecular biomarker in a PCR-DGGE detection of prokaryotes that degrade HPA s. The PCR-DGGE fingerprints obtained from Paracuru-CE, Fortim-CE and Areia Branca-RN samples revealed the occurrence of fluctuations of microbial communities according to the sampling periods and in response to the impact of oil. In the analysis of microbial communities interference of the oil industry, in Areia Branca-RN and Paracuru-CE was observed that oil is a determinant of microbial diversity. Fortim-CE probably has no direct influence with the oil activity. In order to obtain data for better understanding the transport and biodegradation of HPA's, there were conducted in silico studies with modeling and simulation from obtaining 3-D models of proteins involved in the degradation of phenanthrene in the transport of HPA's and also getting the 3-D model of the enzyme PcaF used as molecular marker in this study. Polycyclic aromatic hydrocarbons (PAHs) are a group of compounds that have been the subject of much concern due to their toxic potential. In this study, margarine?s, vegetable cream and mayonnaise available on the Brazilian market were analyzed for pyrene, chrysene, benzo(a)pyrene, benzo(b)fluoranthene and dibenzo(a,h)anthracene. The analytical methodology involved liquid-liquid extraction, clean-up on silica gel column and determination by high performance liquid chromatography using fluorescence detector. Variable levels of contamination were found within differents brands of the same product and within differents batches of the same brand. The total PAH content was in the range of 4.1 to 7.1mug/kg in vegetable cream, 1.7 to 3.9mug/kg in margarine and 1.0 to 21.7mug/kg in mayonnaise. In general the products which according to the label contain corn oil showed the highest levels of contamination. Based on these results and on the importance of fat, oils and derived products for the intake of PAHs, it is recommended that producers of margarine, vegetable creams and mayonnaise start to control the contamination of the vegetable oils used in the elaboration of these products, in order to reduce the exposure of consumers to excessive amounts of potentially carcinogenic compounds.

Aromatic compounds are still poorly evaluated in geochemical studies of Brazilian oil samples. For this reason, the objective of this work was to analyze and evaluate 26 oil samples from four Brazilian sedimentary basins. Aromatic hydrocarbon fractions have been analyzed by gas chromatography coupled to mass spectrometry. The results were also compared with previously evaluated saturated hydrocarbon data. Maturation parameters obtained using the 4- and 1-methyl-dibenzothiophenes and trimethyl-dibenzothiophenes led to the best results because a better grouping was observed. For this correlation, the samples were grouped by maturity degree enabling the routine use of these parameters to evaluate oil maturity levels.

Three bacterial strains were isolated from the activated sludge system of petroleum refinery wastewater, identified by partial sequencing of 16S rDNA, and classified as Acinetobacter genomospecies 3, Bacillus pumilus, and Bacillus flexus. The degradation efficiency of aromatic hydrocarbons was evaluated by gas chromatography with a flame ionization detector. In a mineral medium containing anthracene and phenanthrene and the consortium of microorganisms, the removal efficiency was 96% and 99%, respectively, after 30 days. The good rate of hydrocarbon degradation proves the operational efficiency of the microbial consortium in treating effluents containing these compounds.

Hidrocarbonetos aromáticos policíclicos (HAPs) são um grupo de compostos que têm sido objeto de muitas pesquisas devido ao seu potencial tóxico. Neste estudo, margarinas, cremes vegetais e maioneses disponíveis no mercado brasileiro foram analisadas quanto ao teor de pireno, fluoranteno, benzo(a)antraceno, criseno, benzo(b)fluoranteno, benzo(k)fluoranteno, benzo(a)pireno e dibenzo(a,h)antraceno. A metodologia analítica envolveu extração líquido-líquido, limpeza em coluna de sílica gel e determinação por cromatografia líquida de alta eficiência com detector de fluorescência. Níveis variáveis de contaminação foram encontrados em marcas diferentes do mesmo produto e em lotes diferentes da mesma marca. O teor médio de HAPs totais encontrados esteve na faixa de 4,1 a 7,1mig/kg em cremes vegetais, de 1,7 a 3,9mig/kg em margarinas e de 1,0 a 21,7mig/kg em maioneses. Em geral, os produtos que declaravam conter óleo de milho em sua formulação mostraram os maiores níveis de contaminação. Com base nestes resultados e na importância de gorduras, óleos e produtos derivados como fonte de ingestão de HAPs, recomenda-se aos produtores de margarinas, cremes vegetais e maioneses que iniciem o controle da contaminação dos óleos vegetais utilizados na elaboração destes produtos, de forma a reduzir a exposição do consumidor à quantidades excessivas de compostos potencialmente carcinogênicos.

Industrial activities, oil spills and its derivatives, as well as the incomplete combustion of fossil fuels have caused a great accumulation of hydrocarbons in the environment. The number of microorganisms on the planet is estimated at 1030 and prokaryotes the most abundant. They colonized diverse environments for thousands of years, including those considered extreme and represent an untapped source of metabolic and genetic diversity with a large biotechnological potential. It is also known that certain microorganisms have the enzymatic capacity to degrade petroleum hydrocarbons and, in many ecosystems, there is an indigenous community capable of performing this function. The metagenomic has revolutionized the microbiology allowing access uncultured microbial communities, being a powerful tool for elucidation of their ecological functions and metabolic profiles, as well as for identification of new biomolecules. Thus, this study applied metagenomic approaches not only for functional selection of genes involved in biodegradation and emulsification processes of the petroleum-derived hydrocarbons, but also to describe the taxonomic and metabolic composition of two metagenomes from aquatic microbiome. We analyzed 123.116 (365 ± 118 bp) and 127.563 sequences (352 ± 120 bp) of marine and estuarine metagenomes, respectively. Eight clones were found, four involved in the petroleum biodegradation and four were able to emulsify kerosene indicating their abilities in biosurfactants synthesis. Therefore, the metagenomic analyses performed were efficient not only in the search of bioproducts of biotechnological interest and in the analysis of the functional and taxonomic profile of the metagenomes studied as well

Volatile Organic Compounds are pollutants coming mainly from activities that use fossil fuels. Within this class are the BTEX (benzene, toluene, ethylbenzene and xylenes) compounds that are considered hazardous. Among the various existing techniques for degradation of pollutants, there is advanced oxidation using H2O2 generating hidoxil radical ( OH). In this work, the mesoporous material of MCM-41 was synthesized by hydrothermal method and then was used as support, the impregnation of titanium by the method of synthesis with excess solvent to obtain the catalyst Ti-MCM-41. The catalyst was used in the reaction catalyzed removal of BTEX in water using H2O2 as oxidant. The materials were characterized by: XRD, TG/DTG, FTIR, nitrogen adsorption-desorption and FRX-EDX, in order to verify the method of impregnation of the mesoporous titanium support was effective. Catalytic tests were carried out in reactors of 20 mL containing BTEX (100.0 μg/L), H2O2 (2.0 M) and Ti-MCM-41 (2.0 g/L) in acid medium. The reaction occurred for 5 h at 60 °C and analysis were performed by gas chromatography with photoionization detector and static headspace sampler. The characterizations have proven the effectiveness of the synthesis method used and the incorporation of titanium lt in the support. The catalytic tests showed satisfactory results with conversion of more than 95 % for the studied compounds, where the catalyst 48% Ti-MCM-41 showed a higher removal efficiency of the compounds under study

The oil and biodiesel are oily substances which, if spilled or thrown out in the environment, can cause serious damages. In this context, bioremediation techniques can be used to the recovering of sites degradated by mineral or vegetable oil, besides them, the biodegradation. However, it is necessary that there are microorganism strains used to degradate the contaminant, previously selected for the fast, efficient and minimum impact remediation. The main objective of this study was to isolate microorganism strains coming from different ecological recesses and verify their potentiality on biodegradating oil and biodiesel. Strains of bacteria, fungi and yeasts were isolated from soils contaminated by hydrocarbons (diesel, gasoline, grease) before, by doing striations on culture plates. On the biodegradation tests, it was used the redox indicator, 2,6-dichlorophenol indophenol (DCPIP), then, after the indicador’s discoloration, the absorbance of the suspension obtained was measured. 46 different microorganism strains were isolated, and the oil, when it was used as a carbon resource, it was degradate easier than the biodiesel. The experiments showed that, in a period, the site impacted by oil spills recovers itself by the biodegradation, in aerobic conditions, by the redox activity.

Um reator em batelada, aerado, com biomassa imobilizada de Aspergillus niger AN400 foi operado durante 10 ciclos de 7 dias para remover benzeno (200 mg.L-1), tolueno (200 mg.L-1) e xileno (50 mg.L-1) - BTX - e de nutrientes de meio basal. O reator era alimentado semanalmente com 4 L do meio e glicose - 1 g.L-1, na Fase I, e 0,5 g.L-1, na Fase II. Os BTX foram detectados até o quarto dia de operação, em todos os ciclos. As melhores eficiências médias de remoção foram na Fase I: 75%de matéria orgânica solúvel, 80% de ortofosfato e 77% de amônia. O reator pode ser uma alternativa viável para tratamento de águas poluídas com BTX, porém há a necessidade de estudar o comportamento do reator durante período de operação mais longo e com ciclos reacionais mais curtos, bem como da identificação dos metabólitos produzidos.

Hidrocarbonetos Aromáticos Policíclicos (HAPs) e seus derivados Halogenados (HHAPs) são conhecidos como um conjunto de contaminantes ambientais, classificados como potencialmente tóxicos, mutagénicos e carcinogénicos, sendo um assunto de extrema importância e preocupação para a saúde pública. Diversos estudos têm reportado o potencial efeito tóxico destes compostos após longos períodos de exposição, pelo uso de métodos colorimétricos como o ensaio de brometo de 3-[4,5- dimetil-tiazol-2-il]-2,5-difeniltetrazólio (MTT) e vermelho neutro. Contudo, apesar de estes métodos clássicos de avaliação da citotoxicidade in vitro permitirem, de forma rápida, identificar as propriedades nocivas dos compostos, não permitem compreender os mecanismos moleculares subjacentes responsáveis pela sua toxicidade. Deste modo, no presente estudo, para avaliar os efeitos à exposição aos compostos Pireno (Pir) e seu derivado resultante da desinfeção de águas por bromação, 1-BromoPireno (1-BrPir), foram utilizados outros métodos complementares como avaliação do stress oxidativo e vários fenómenos associados à morte celular programada. Os resultados obtidos comprovam não só a existência de efeitos citotóxicos dos poluentes quando acumulados, como também possíveis efeitos genotóxicos. Assim, pelos métodos complementares, foi possível a identificação de alvos moleculares passiveis de ação farmacológica, o que pode constituir o primeiro passo para o desenho de estratégias terapêuticas que visem prevenir ou tratar os danos provocados por estes poluentes no Homem.

Os Hidrocarbonetos Aromáticos Policíclicos (HAPs) são contaminantes persistentes em meio aquoso. Estes compostos são conhecidos pelas suas propriedades carcinogénicas, mutagénicas e genotóxicas. O principal objetivo deste trabalho consistiu na avaliação das potencialidades de subprodutos da indústria corticeira, como adsorventes alternativos para a remoção de cinco HAPs em meio aquoso: benzo(a)pireno, benzo(ghi)perileno, benzo(b)fluoranteno, benzo(k)fluoranteno e indeno(1,2,3-cd)pireno. A metodologia analítica para quantificar os HAPs envolveu a preparação das amostras, através da técnica de extração em fase sólida (SPE), e a quantificação dos compostos analisados por cromatografia líquida com detetor de fluorescência (LC-FLD). O método foi otimizado e validado, obtendo-se limites de quantificação de 0,004 μg/L para todos os HAPs. Os estudos incidiram na utilização de uma amostra de cortiça, pó de aglomerado de cortiça expandida (PACE), obtida por aglutinação de cortiça em condições hidrotérmicas, a qual nos estudos preliminares revelou desempenho semelhante aos carvões ativados. Com exceção do benzo(ghi)perileno, os resultados mostram que o processo de adsorção dos HAPs na amostra PACE segue uma cinética de pseudo-segunda ordem e as isotérmicas ajustam-se ao modelo de Langmuir.
