Le 1,1'-bi-2-naphtol ou Binol, présentant une chiralité axiale, est un ligand très utilisé en catalyse asymétrique. Au cours des vingt dernières années, le Binol a servi de synthon à l’élaboration de très nombreux ligands permettant la catalyse asymétrique de tous types de réactions, allant de l’hydrogénation, à l’alkylation, en passant par diverses réactions péricycliques. Le grand intérêt pour ce ligand vient de sa versatilité et des nombreuses possibilités de fonctionnalisation qu’il offre, permettant d’altérer ses propriétés catalytiques à volonté, aussi bien en modifiant son caractère électronique, qu’en introduisant des facteurs stériques autour du site catalytique. Parallèlement aux développements de la catalyse par des dérivés de Binol, le domaine des liquides ioniques a connu un intérêt croissant ces dernières années. Les liquides ioniques, sels dont le point de fusion est inférieur à 100°C, cumulent de nombreuses qualités convoitées : faible pression de vapeur, stabilité thermique et chimique et fort pouvoir de solvatation. Dû à ces propriétés, les liquides ioniques ont principalement été étudiés dans l’optique de développer une gamme de solvants recyclables. Alors que les propriétés des liquides ioniques sont facilement modulables en fonction de l’anion et du cation choisi, le concept de liquide ionique à tâche spécifique va plus loin et propose d’introduire directement, sur le cation ou l’anion, un groupement conférant une propriété particulière. En suivant cette approche, plusieurs ligands ioniques ont été rapportés, par simple couplage d’un cation organique à un ligand déjà connu. Étonnamment, le Binol a fait l’objet de très peu de travaux pour l’élaboration de ligands ioniques. Dans cette thèse, nous proposons l’étude d’une famille de composés de type Binol-imidazolium dont les unités Binol et imidazolium sont séparées par un espaceur méthylène. Différents homologues ont été synthétisés en variant le nombre d’unités imidazolium et leur position sur le noyau Binol, la longueur de la chaîne alkyle portée par les unités imidazolium et la nature du contre-anion. Après une étude des propriétés thermiques de ces composés, l’utilisation des Binol-imidazoliums en tant que ligands dans une réaction asymétrique d’éthylation d’aldéhydes aromatique a été étudiée en milieu liquide ionique. La réaction a été conduite en solvant liquide ionique dans le but de recycler aussi bien le ligand Binol-imidazolium que le solvant, en fin de réaction. Cette étude nous a permis de démontrer que la sélectivité de ces ligands ioniques dépend grandement de leur structure. En effet, seuls les Binols fonctionnalisés en positions 6 et 6’ permettent une sélectivité de la réaction d’éthylation. Alors que les dérivés de Binol fonctionnalisés en positions 3 et 3’ ne permettent pas une catalyse énantiosélective, il a déjà été rapporté que ces composés avaient la capacité de complexer des anions. D’autre part, il a déjà été rapporté par notre groupe, que les composés comportant des unités imidazolium pouvaient permettre le transport d’anions à travers des bicouches lipidiques en fonction de leur amphiphilie. Ceci nous a amenés à la deuxième partie de cette thèse qui porte sur les propriétés ionophores des Binols fonctionnalisés en positions 3 et 3’ par des unités imidazoliums. Dans un premier temps, nous nous sommes intéressés à l’étude de la relation structure-activité et au mécanisme de transport de ces composés. Le transport d’anions étant un processus clé dans la biologie cellulaire, l’activité biologique des composés présentant une activité ionophore dans des systèmes modèles (liposomes) a été étudiée par la suite. L’activité antibactérienne des nos composés a été testée sur quatre souches de bactéries. Il s’est avéré que les composés Binol-imidazolium sont actifs uniquement sur les bactéries Gram positives. Finalement, la cytotoxicité des composés présentant une activité antibactérienne a été étudiée sur des cellules humaines.

During the last fifteen years organocatalysis emerged as a powerful tool for the enantioselective functionalization of the most different organic molecules. Both C-C and C-heteroatom bonds can be formed in an enantioselective fashion using many types of catalyst and the field is always growing. Many kind of chiral catalysts have emerged as privileged, but among them Proline, cinchona alkaloids, BINOL, and their derivatives showed to be particularly useful chiral scaffolds. This thesis, after a short presentation of many organocatalysts and activation modes, focuses mainly on cinchona alkaloid derived primary amines and BINOL derived chiral Brønsted acids, describing their properties and applications. Then, in the experimental part, these compounds are used for the catalysis of new transformations. The enantioselective Friedel-Crafts alkylation of cyclic enones with naphthols using cinchona alkaloid derived primary amines as catalysts is presented and discussed. The results of this work were very good and this resulted also in a publication. The same catalysts are then used to accomplish the enantioselective addition of indoles to cyclic enones. Many catalysts in combination with many acids as co-catalysts were tried and the reaction was fully studied. Selective N-alkylation was obtained in many cases, in combination with quite good to good enantioselectivities. Also other kind of catalysis were tried for this reaction, with interesting results. Another aza-Michael reaction between OH-free hydroxylamines and nitrostyrene using cinchona alkaloid derived thioureas is briefly discussed. Then our attention focused on Brønsted acid catalyzed transformations. With this regard, the Prins cyclization, a reaction never accomplished in an enantioselective fashion until now, is presented and developed. The results obtained are promising. In the last part of this thesis the work carried out abroad is presented. In Prof. Rueping laboratories, an enantioselective Nazarov cyclization using cooperative catalysis and the enantioselective desymmetrization of meso-hydrobenzoin catalyzed by Brønsted acid were studied.

The implementation of chiral centres within biologically active compounds has been a perplexing yet motivational force in chemistry. This work presents the attempted formation of a concurrent or sequential tandem catalyzed methodology of enantioselective nucleophilic addition and electrophilic cyclization. The 2'- arylalkynyl- aldehyde, ketone, and imine substrates used within were adeptly chosen with a dually activated structure; 1) for nucleophilic addition to the electrophilic substituents; and 2) for carbophilic activation of the alkyne substituent to undergo cyclization. To accomplish the nucleophilic addition, two distinct allylation methodologies were pursued: (/?)-BINOL catalyzed-allylboration and (5)- BINAP-AgF catalyzed-allylsilylation. BINAP catalyzed enantioselective allylation of 2'-arylalkynyl-aldehydes, to form chiral homoallylic alcohols, was successful. Homoallylic alcohols were isolated with high enantio-purity (>80%), which then underwent sequential cyclization to form chiral allylic phthalans, in moderate yields. An application of this methodology towards the construction of biologically active compounds was included with the partial synthesis of the natural product and H. pylori inhibitor, (+)-Spirolaxine methyl ether.

Les ligands de carbènes N-hétérocycliques (NHC) qui possèdent une symétrie C1 attirent beaucoup l’attention dans la littérature. Le présent projet de recherche propose de synthétiser une nouvelle série de ligands NHC C1-symétriques avec deux groupements N-alkyles qui exploitent un relais chiral. Un protocole modulaire et efficace pour la synthèse des sels d’imidazolium chiraux qui servent comme préligands pour les NHC a été développé. Quelques-uns de ces nouveaux ligands ont été installés sur le cuivre et de l’or, créant de nouveaux complexes chiraux. Les nouveaux complexes à base de cuivre ont été évalués comme catalyseurs pour le couplage oxydatif de 2-naphthols. Les ligands C1-symmétriques ont fourni des meilleurs rendements que les ligands C2-symmétriques. Au cours de l’optimisation, des additifs ont été évalués; les additifs à base de pyridine ont fourni des énantiosélectivités modérées tandis que les additifs à base de malonate ont donné des meilleurs rendements de la réaction de couplage oxydatif. Ultérieurement, les additifs à base de malonate ont été appliqués envers l’hétérocouplage de 2-naphthols. Le partenaire de couplage qui est riche en électrons est normalement en grand excès à cause de sa tendance à dégrader. Avec le bénéfice de l’additif, les deux partenaires de couplage peuvent être utilisés dans des quantités équivalentes. La découverte de l’effet des additifs a permis le développement d’un protocole général pour l’hétérocouplage de 2-naphthols.

Studio, a livello teorico basandosi sulla teoria DFT, del precursore dell’acido fosforico di Brønsted derivato dall’R-BINOL, delineazione delle conformazioni possibili e della loro stabilità come supporto base alla via sperimentale. Ricerca di una via di sintesi ottimizzata, basata sulla reazione di Suzuki-coupling, che permetta di ottenere i precursori ipotizzati in precedenza (si veda la figura 9) con buone rese e successiva separazione degli atropoisomeri mediante HPLC semipreparartiva. Caratterizzazione completa degli isomeri conformazionali utilizzando tecniche spettroscopiche (NMR ed ECD), studio cinetico per ottenere il valore della barriera rotazionale sperimentale (ΔG#) dei gruppi metilnaftalenici attraverso equilibrazione termica e confronto con i dati teorici. Ricerca di una via di sintesi che consenta la funzionalizzazione dei precursori ad acidi fosforici senza alterazione della conformazione atropoisomerica e con assenza di racemizzazione ed equilibrazione. Studio dell’attività catalitica dei vari atropoisomeri in una sintesi asimmetrica nota per osservare sia come varia la reattività in funzione della conformazione molecolare, sia per ottenere confronti con altri organocatalizzatori conosciuti.

Extensive experimental and computational studies have been carried out on the enantioselective titanium(IV)-catalyzed cyanobenzoylation of aldehydes using 1:n Binolam:Ti(OiPr)4 mixtures as precatalysts, with the purpose of identifying the key mechanistic aspects governing enantioselectivity. HCN and isopropyl benzoate were detected in the reacting mixtures. This, as well as the reaction’s response to the presence of an exogenous base, and the failure to react in the presence of Binol:Ti(OiPr)4 mixtures, led us to propose not a direct cyanobenzoylation but an indirect process involving enantioselective hydrocyanation followed by O-benzoylation. Computational work provided positive evidence for the intervention of both indirect and direct cyanobenzoylation routes, the former being faster. However, the standard Curtin–Hammett-based optimization search ended with unsatisfactory results. Experimental and computational DFT studies (B3LYP/6-31G*) led us to conclude that: (1) the overall cyanobenzoylation of aldehydes catalyzed by 1:n Binolam:Ti(OiPr)4 mixtures involves an enantioselective hydrocyanation followed by an stereochemically inert O-benzoylation; (2) the initial complexes prevailing in a 1:1 Binolam:Ti(OiPr)4 mixture are the solvated mononuclear monomer 5·2(iPrOH) and solvated dinuclear dimer 9·2(iPrOH), whereas 9·2(iPrOH) is the major component in a 1:2 or higher 1:n mixture; (3) since the slowest step is that of benzoylation of ligated iPrOH which yields the actual catalysts 5–9, the catalytic system fits into a non-Curtin–Hammett framework, the final products deriving from a kinetic quench of the competing routes; and (4) accordingly, catalysis by 1:1 Binolam:Ti(OiPr)4 mixtures should involve cyanobenzoylations promoted by mononuclear 5, contaminated with those promoted by some dinuclear open dimer 9, whereas cyanobenzoylations catalyzed by a 1:2 and higher 1:n mixtures should be the result of catalysis promoted by the large amounts of dinuclear open dimer 9.

The lithiation, of the secondary chloride 2, catalyzed by binaphthyl derivatives, i.e. BINAM 4, BINOL 5, BINAP 6, H8-BINAP 7, Tol-BINAP 8, 2,2’-bis(pyrrolidin-1-yl)-1,1’-binaphthalene 9, and 2,2’-dimethyl-1,1’-binaphthalene 11, in the presence of different ketones has been studied, yielding the corresponding alcohol derivatives 3 and 12-16 in moderate to good yields. Binaphthyl derivative 11 has revealed to be very active as catalyst in the lithiation process at room temperature, and has allowed the preparation of the alcohol derivatives with enantioselectivities up to 50%.

Chiral complexes formed by privileged phosphoramidites derived from chiral binol and optically pure Davies’ amines, and copper(II) triflate, silver(I) triflate or silver(I) benzoate are excellent catalysts for the general 1,3-dipolar cycloaddition between nitroalkenes and azomethine ylides generated from α-amino acid derived imino esters. These three methods can be conducted at room temperature to afford the exo-cycloadducts (4,5-trans-2,5-cis-4-nitroprolinates) with high diastereoselectivity and high enantioselectivity. In general, the three procedures are complementary but silver catalysts are more versatile and less sensitive to sterically congested starting materials.

The primary focus of this thesis was the development of a novel chiral tether that could be used to control axial chirality around a newly formed aryl-aryl bond, and the extension of this methodology to the model synthesis of gomisin M1. In chapter 1, a review detailing the use of chiral tethers in the synthesis of atropisomers is discussed. The use of a variety of chiral molecules including 1,2-diols, 1,3-diols and other diol-based tethers, as well as amine-based and miscellaneous tethers are detailed. In chapter 2, the rationale behind the design of our novel molecular tethers, along with the subsequent synthesis of three chiral 1,3-diol-based tethers, is outlined. The method by which the enantiopurity of these diols was determined is also reviewed. This chapter also includes the attempted Mitsunobu and intramolecular couplings in the model synthesis of BINOL. Chapter 3 discusses the synthesis of suitable aryl halide substrates, and their employment in the attempted tether-controlled asymmetric model synthesis of gomisin M1. A comprehensive investigation into the attempted intramolecular biaryl coupling of these tethered substrates is also included. The non-stereoselective model synthesis of gomisin M1 is outlined in chapter 4. The installation of the desired biaryl linkage and the subsequent attempted intramolecular McMurry couplings are discussed. The impact of different protecting groups in the molecule on the intramolecular McMurry reaction is also outlined. Chapter 5 details the full experimental procedures, including spectroscopic and analytical data for the compounds prepared during this research.

In this review article recent developments in the asymmetric transfer hydrogenation of imines from 2008 up to today are presented. The main methodology involves either metal-catalyzed procedures in the presence of a chiral ligand or organocatalyzed technologies using a Hantzsch ester and a chiral BINOL-derived phosphoric acid. The most important procedures are collected, paying special attention to the application of this methodology in synthetic organic chemistry.

The gold(I)-catalyzed chemoselective dearomatization of β-naphthols is reported through a straightforward approach via [3,3]-sigmatropic rearrangement /allene-cyclyzation cascade processes. Easily accessed naphthyl-propargyl ethers and derivatives in this work are employed as starting materials. Delightfully, an array of deoramatized dyhydrofuryl -naphthalen-2(1H)-ones featured densely functional groups are obtained in high yields (up to 98%) in 10 min reaction time under extremely mild reaction conditions like reagent grade solvent and exposure to air. The potential of accessing to high enantioselectivety on the dearomatized dyhydrofuryl- naphthalen-2(1H)-ones is also approved by the good ee (65%) relying on (R)-xylyl- BINAP(AuCl)2. In addition, complete theoretical elucidation of the reaction pathway is also proposed which addresses a rationale for essential motivation such as regio- and chemoselectivity. Moreover, an efficient gold catalyzed intermolecular dearomatization of substituted β-naphthols with allenamides is presented here. PPh3AuTFA (5 mol %) approves the efficient dearomatively allylation protocol under mild conditions and exhibits high tolerance on substrates scope (24 examples) in good to excellent yield accompanied with high regioselectivity and stereoselectivity. Moreover, the synergistic catalytic system also highlight the synergistic function between the [PPh3Au]+ (π-acid) and TFA− (Lewis base). At last, a new chiral BINOL phosphoric acid silver salt is successfully synthesized and used as the chiral counter anion, which strongly promotes the enantioselectivity (up to 92%). At last but not least, crucially, SmI2 induced enantioselective formal synthesis of strychnine, a complex alkaloid and a classical target used to benchmark new synthetic methods is developed. Enantioselective dearomatising radical cyclisation on to the indole unit and further ET will then give organosamarium that is quenched diastereoselectively by the ester to deliver Strychnine in 7 steps.
