While recreational drug use in UK women is prevalent, to date there is little prospective data on patterns of drug use in recreational drug-using women immediately before and during pregnancy. A total of 121 participants from a wide range of backgrounds were recruited to take part in the longitudinal Development and Infancy Study (DAISY) study of prenatal drug use and outcomes. Eighty-six of the women were interviewed prospectively while pregnant and/or soon after their infant was born. Participants reported on use immediately before and during pregnancy and on use over their lifetime. Levels of lifetime drug use of the women recruited were high, with women reporting having used at least four different illegal drugs over their lifetime. Most users of cocaine, 3,4-methylenedioxy-N-methylamphetamine (MDMA) and other stimulants stopped using these by the second trimester and levels of use were low. However, in pregnancy, 64% of the sample continued to use alcohol, 46% tobacco and 48% cannabis. While the level of alcohol use reduced substantially, average tobacco and cannabis levels tended to be sustained at pre-pregnancy levels even into the third trimester (50 cigarettes and/or 11 joints per week). In sum, while the use of ‘party drugs’ and alcohol seems to reduce, levels of tobacco and cannabis use are likely to be sustained throughout pregnancy. The data provide polydrug profiles that can form the basis for the development of more realistic animal models.

Background: The existence of an ecstasy dependence syndrome is controversial. We examined whether the acute after-effects of ecstasy use (i.e., the “come-down”) falsely lead to the identification of ecstasy withdrawal and the subsequent diagnosis of ecstasy dependence. Methods: The Structured Clinical Interview for DSM-IV-TR Disorders: Research Version (SCID-RV) was administered to 214 Australian ecstasy users. Ecstasy withdrawal was operationalized in three contrasting ways: (i) as per DSM-IV criteria; (ii) as the expected after effects of ecstasy (a regular come-down); or (iii) as a substantially greater or longer come-down than on first use (intense come-down). These definitions were validated against frequency of ecstasy use, readiness to change and ability to resist the urge to use ecstasy. Confirmatory factor analyses were used to see how they aligned with the overall dependence syndrome. Results: Come-down symptoms increased the prevalence of withdrawal from 1% (DSM-IV criterion) to 11% (intense come-downs) and 75% (regular come-downs). Past year ecstasy dependence remained at 31% when including the DSM-IV withdrawal criteria and was 32% with intense come-downs, but increased to 45% with regular come-downs. Intense come-downs were associated with lower ability to resist ecstasy use and loaded positively on the dependence syndrome. Regular come-downs did not load positively on the ecstasy dependence syndrome and were not related to other indices of dependence. Conclusion: The acute after-effects of ecstasy should be excluded when assessing ecstasy withdrawal as they can lead to a false diagnosis of ecstasy dependence. Worsening of the ecstasy come-down may be a marker for dependence.

Two new 2-aryl benzofuran derivatives, 3-[7-methoxy-2-(3,4-methylenedioxy phenyl) benzofuran-5-yl]propyl 3-[7-methoxy-2-(3,4-methylenedioxyphenyl)benzo furan-5-yl]propanoate (1) and demethoxy egonol gentiobioside (2), were isolated from the 95% aqueous ethanolic extract of the seeds of Styrax macranthus, together with 7-methoxy-2-(3,4-methylenedioxyphenyl) benzofuran-5-carbaldehyde (3), egonol (4), demethoxy egonol (5), demethyl egonol (6), egonol glucoside (7), egonol gentiobioside (8), egonol gentiotrioside (9), stigmasterol (10), 2,3-dihydroxypropyl tetracosoate (11), and daucosterol (12). In vitro test, compound 2 promote the synthesis of estrogen E2. Nineteen compounds were isolated from the 95% aqueous ethanolic extract of the whole plant of Porandra scandens for the first time. Their structures were identified as (2S,3S,4R)-2-[(2R)-2-hydroxy-heneicosanoylamino]-1,3,4- heneicosanetriol (13), (2S,3S,4R)-2-tetracosanoylamino-1,3,4-octadecanetriol (14), daucosterol (15), β-sitosterol (12), (20S,22E,24R)-5α,8α-epidioxy-ergosta-6,22-diene- 3β-ol (16), 6β-hydroxylstigmast-4-en-3-one (17), 1-glycerol-1-hexadecoate (18), betulinic acid (19), emodin (20), 1-glycerol-1-docosoate (21), p-hydroxybenzaldehyde (22), 1-glycerol-1-heptadecoate (23), aurantiamide acetate (24), 1-glycerol-1- nonadecoate (25), palmatic acid (26), (E)-p-coumaric acid (27), (22E,24S)- 24-metbylcbolesta-7,22-diene-3β,5α,6β-triol (28), 2-deoxycrustecdysone (29), and auranamide (30). The characteristic, natural resource, bioactivity, and the total synthesis of 2-aryl benzofurans were reviewed.

Two efficient, regio- and stereo controlled synthetic approaches to the synthesis of racemic analogs of pancratistatin have been accomplished and they serve as the model systems for the total synthesis of optically active 7-deoxy-pancratistatin. In the Diels-Alder approach, an efficient [4+2] cycloaddition of 3,4-methylenedioxyco- nitrostyrene with Danishefsky's diene to selectively form an exo-nitro adduct has been developed as the key step in the construction of the C-ring of the target molecule. In the Michael addition approach, the key step was a conjugate addition of an organic zinc-cuprate to the 3,4-methylenedioxy-(B-nitrostyrene, followed by a diastereocontroUed closure to form the cyclohexane C-ring of the target molecule via an intramolecular nitro-aldol cyclization on a neutral alumina surface. A chair-like transition state for such a cyclization has been established and such a chelation controlled transition state can be useful in the prediction of diastereoselectivity in other related 6-exo-trig nitroaldol reactions. Cyclization of the above products fi^om both approaches by using a Bischler-Napieralski type reaction afforded two lycoricidine derivatives 38 and 50 in good yields. The initial results from the above modeling studies as well as the analysis of the synthetic strategy were directed to a chiral pool approach to the total synthesis of optically active 7-deoxy-pancratistatin. Selective monsilylation and iodination of Ltartaric acid provided a chiral precursor for the proposed key Michael transformation. The outlook for the total synthesis of 7-deoxy-pancratistatin by this approach is very promising.A concise synthesis of novel designed, optically pure, Cz-symmetrical disulfonylamide chiral ligands starting from L-tartaric acid has also been achieved. This sequence employs the metallation of indole followed by Sfj2 replacement of a dimesylate as the key step. The activity for this Cz-symmetric chiral disulfonamide ligand in the catalytic enantioselective reaction has been confirmed by nucleophilic addition to benzaldehyde in the disulfonamide-Ti (0-i-Pr)4-diethylzinc system with a 48% yield and a 33% e.e. value. Such a ligand tethered with a suitable metal complex should be also applicable towards the total synthesis of 7-deoxy-pancratistatin.

Extracts from Holostylis reniformis were tested in vivo against Plasmodium berghei and in vitro against a chloroquine-resistant strain of Plasmodium falciparum. The hexane extract of the roots was the most active, causing 67% reduction of parasitemia in vivo. From this extract, six lignans, including a new (7 ' R,8S,8 ' S)-3 ',4 '-methylenedioxy-4,5-dimethoxy-2,7 '-cyclolignan-7-one, were isolated and tested in vitro against P. falciparum. The three most active lignans showed 50% inhibitor concentrations of <= 0.32 mu M. An evaluation of minimum lethal dose (30%) values showed low toxicity for these lignans in a hepatic cell line (Hep G2A16). Therefore, these compounds are potential candidates for the development of antimalarial drugs.

Bioactivity-guided fractionation of a CH2Cl2 extract from leaves of Piper hispidum (Piperaceae) yielded a new pyrrolidine amide, N-[7-(3',4'-methylenedioxyphenyl)-2(Z),4(Z)-heptadienoyl] pyrrolidine 1, in addition to two known amides N-[5-(3',4'-methylenedioxyphenyl)-2(E)-pentadienoyl]pyrrolidine and N-[2-(3',4'-methylenedioxy-6'-methoxyphenyl)-2(Z)-propenoyl]pyrrolidine. The structure of compound 1 was elucidated by interpretation of spectral data, including ES-MS. Compound 1 showed antifungal activity against Cladosporium sphaerospermum.

From the seeds of Virola surinamensis, which were collected near Altamira and near Maraba, Para State, Brazil, the following substances were isolated by chromatographic techniques: two dibenzylbutanediol lignans, dihydrocubebin and the new dihydrocubebin monolaurate, two furofuran lignans, sesamin and asarinin, three dibenzylbutyrolactol lignans, cubebin, β-O- methylcubebin and α-O-methylcubebin, one dibenzylbutyrolactone lignan, hinokinin, one aryltetralin neolignan, galbulin, two tetrahydrofuran neolignans, galgravin and the new 4'-hydroxy-3'-methoxy-3,4-methylenedioxy- 8.8',7.O.7'-neolignan, one flavone, tithonine, one isoflavone, irisolidone, and two new polyketides, 3-hydroxy-1-(15-phenylpentadecanoyl)-2,6- cyclohexanedione and 1-(5-phenylpentanoyl)-2,6-cyclohexanedione. Different chemical constitutions of the fruits from the two localities were observed.

Objectives were to evaluate the role of canonical WNT signaling in development of the preimplantation embryo. Signaling was activated with 2-Amino-4-(3,4-(methylenedioxy)benzylamino)-6-(3-methoxyphenyl)pyrimidine (AMBMP) and inhibited with Dickkopf-related protein 1 (DKK1). Treatment of bovine embryos with AMBMP at day 5 after insemination decreased development to the blastocyst stage at day 7 and reduced numbers of trophectoderm and inner cell mass cells. At high concentrations, AMBMP caused disorganization of the inner cell mass. DKK1 blocked actions of AMBMP but did not affect development in the absence of AMBMP. Examination of gene expression in day 6 morulae by microarray revealed expression of 16 WNT genes and other genes involved in WNT signaling; differences in relative expression were confirmed by PCR for 7 genes. In conclusion, the preimplantation embryo possesses a functional WNT signaling system and activation of the canonical pathway can inhibit embryonic development.

Do extrato hexânico das partes aéreas de Aristolochia peltato-deltoidea Hoehne (Aristolochiaceae) foram isolados duas novas lignanas epiméricas do tipo dibenzilbutirolactol, rel-(8R, 8 S, 9S)-3,4-dimetoxi-3 ,4 -metilenodioxi-9b-etoxi- e rel-(8R, 8 S, 9R)-3,4-dimetoxi-3 ,4 -metilenodioxi-9a-etoxi-lignanas-8.8 ,9.O.9 , além da neolignana benzofurânica eupomatenóide-7, a-tocoferilquinona, b-sitosterol e estigmasterol. Do extrato clorofórmico foram isoladas duas lignanas dibenzilbutirolactonas diasteroisoméricas: rel-(8R, 8 R)- e rel-(8R, 8 S)-3,4-dimetoxi-3 ,4 -metilenodioxi-9-oxo-lignanas-8.8 ,9.O.9 . A composição química das frações apolares do extrato hexânico também foi analisada por CG/EM. Dentre os componentes detectados, dez foram identificados. As estruturas dos compostos isolados foram elucidadas utilizando-se métodos espectrométricos.

Das folhas de Vatairea guianensis Aublé foram isoladas quatro isoflavonas identificadas como, 5,3',-diidroxi-4'-metoxi-2",2"-dimetilpirano-(5",6":8,7)-isoflavona ( 1: ), 5,7-diidroxi-3',4'-metilenodioxi-8-prenil-isoflavona ( 2: ) e 5,3'-diidroxi-4'-metoxi-7-O-β-glicopiranosídeo-8-prenil-isoflavona ( 3: ) e derrona ( 4: ), juntamente com cinco triterpenos identificados em mistura de lupeol, α-amirina, β-amirina, germanicol e ácido betulínico. As substâncias 1: 3: são novos produtos naturais, porém 1: e 2: já foram citados como produtos de síntese. No entanto, todas essas substâncias são relatadas pela primeira vez para essa espécie. Suas estruturas químicas foram elucidadas com base nos seus dados de ressonância magnética nuclear (RMN) 1D e 2D e por espectrometria de massas de alta resolução. O extrato etanólico das folhas e os compostos 1-3: foram avaliados quanto ao seu potencial sequestrador do radical DPPH• (2,2-difenil-1-picril-hidrazila) e os resultados mostram que o extrato apresentou alta atividade (CI50 = 6,2 ± 0,4 µg mL-1), enquanto as substâncias testadas apresentaram baixo poder antioxidante (CI50 ≥ 29,5 ± 2,5 µg mL-1) quando comparadas com TROLOX (CI50 = 4,5 ± 0,4 µg mL-1).

This study assessed the effects of the serotonin (5-HT) and norepinephrine (NE) transporter inhibitor duloxetine on the effects of 3,4-methylenedioxy-methamphetamine (MDMA, ecstasy) in vitro and in 16 healthy subjects. The clinical study used a double-blind, randomized, placebo-controlled, four-session, crossover design. In vitro, duloxetine blocked the release of both 5-HT and NE by MDMA or by its metabolite 3,4-methylenedioxyamphetamine from transmitter-loaded human cells expressing the 5-HT or NE transporter. In humans, duloxetine inhibited the effects of MDMA including elevations in circulating NE, increases in blood pressure and heart rate, and the subjective drug effects. Duloxetine inhibited the pharmacodynamic response to MDMA despite an increase in duloxetine-associated elevations in plasma MDMA levels. The findings confirm the important role of MDMA-induced 5-HT and NE release in the psychotropic effects of MDMA. Duloxetine may be useful in the treatment of psychostimulant dependence.
