Polyamine polymers have attracted attention due to their ability to demonstrate pH dependent cationic nature and presence of highly reactive pendant amino groups. These amino groups make them suitable for a host of applications through cross-linking and derivatization. As a result the end use application of a polyamine is largely driven by the number of amino groups and the way they are attached to the polymer backbone. Thus, this piece of work describes the synthesis and investigation of properties of a novel aliphatic polyamine, poly(methylene amine); that carries maximum number of amino group on its backbone. The target polymer, poly(methylene amine); was synthesized via two major steps viz.1.synthesis of precursor polymers of poly(methylene amine) and 2. Hydrolysis of the precursor polymers to obtain poly(methylene amine). The precursor polymers poly (1,3-diacetylimidazole-2-one)(6) and poly(1,3-diformyldihydroimidazol-2-one)(7) were synthesized via radical polymerization of their respective monomers. The monomers were polymerized in bulk as well as in solution at different reaction conditions. The maximum molecular weights were achieved by polymerizing the monomers in bulk (Mn = 6.5 x 104 g/mol and Mw = 2.13 x 105 g/mol) of 6. The precursor polymers were hydrolyzed under strong reaction conditions in ethanol in presence of NaOH, LiCl at 170°C to yield poly(methylene amine). The process of hydrolysis was monitored by IR spectroscopy. The solution properties of poly(methylene amine) and its hydrochloride were investigated by viscosimetry and light scattering. The reduced viscosity of poly (methylene amine) hydrochloride as a function of polymer concentration demonstrated a behavior typical of cationic polyelectrolyte. With decrease in polymer concentration the reduced viscosity of poly(methylene amine) hydrochloride increased gradually. The dynamic light scattering studies also revealed behaviors of a polyelectrolyte. Poly(methylene amine) was reacted with electrophiles to yield novel materials. While the attachment of alkyl group onto the nitrogen would increase nucleophilicity, it would also impose steric hindrance. As a result the degree of substitution on poly(methylene amine) would be governed by both the factors. Therefore, few model reactions with electrophiles were performed on polvinylamine under similar reaction conditions in order to make a comparative evaluation. It was found that under similar reaction conditions the degree of substitution was higher in case of polyvinylamine in comparison with poly (methylene amine).This shows that the steric hindrance outweighs nucleophilicity while deciding degree of substitution of electrophiles on poly(methylene amine). The modification was further extended to its use as an initiator for ring opening polymerization of benzyloxy protected N-carboxyanhydride of z-Lysine. The resulting polymer had an interesting brush like architecture. The solid state morphology of this polymer was investigated by SAXS. The 2D-WAXS diffractograms revealed hexagonal morphology of peptide segments without formation of alpha helices.

Die vorliegende Dissertation beschreibt die Verschmelzung der Konzepte von konjugierten Polyelektrolyten und amphiphilen Kammpolymeren in Form von konjugierten, Poly(2,7-carbazol)-basierenden Polyelektrolytkammpolymeren mit Poly(L-lysin)seitenketten sowie Alkyl- oder Polyethylenglykolsubstituenten. Die Synthese wurde durch die Suzuki-Polykondensation von monodispersen Makromonomeren erreicht. Hierbei fand die Precursor-Synthesestrategie Anwendung. In diesem Ansatz war die ε-Aminofunktion des Lysins mit einer Benzoyloxycarbonylschutzgruppe geschützt. Der Aufbau der benötigten monodispersen Makromonomere erfolgte durch die Kupplung von Poly(L-lysin)ketten an den Carbazolbaustein mittels eines aktivierten Esters. Eine Besonderheit der hergestellten Kammpolymere lag in den konformativen Eigenschaften seiner einzelnen Komponenten. Dabei konnte die Konformation der Poly(L-lysin)seitenketten infolge ihres Polyelektrolyt- und Peptidcharakters gezielt mit Hilfe des pH-Wertes und der Ionenstärke variiert werden, wohingegen das konjugierte Rückgrat seine steife Konformation beibehielt. Infolge des Polyelektrolytcharakters zeigte sich zudem, dass die Polymere in sauren und neutralen, wässrigen Lösungen zu großen Teilen in Form von Domänenstrukturen auftraten, während im Basischen eine sofortige Aggregation eintrat. Ein weiteres Merkmal der vorgestellten Polyelektrolytkammpolymere war ihr amphiphiler Charakter. Diese Amphiphilie der in dieser Arbeit vorgestellten Polyelektrolytkammpolymere beeinflusste dabei maßgeblich ihre unstrukturierte Anordnung in Lösung, in der festen Phase sowie an Oberflächen.

Die Dissertation beschreibt die Darstellung und Charakterisierung von neuartigen, amphiphilen Carbazol- und Fluoren-(co)-polymeren, die infolge ihres strukturellen Aufbaus sowohl selbstorganisierende als auch optoelektronische Eigenschaften kombinierten. Zum einen wurden Kammpolymere mit konjugierten, steifen Rückgraten und flexiblen sowie konformativ schaltbaren Polyelektrolytseitenketten dargestellt und auf eine pH-abhängige Selbstorganisation in Lösung und an Oberflächen untersucht. Des Weiteren wurden neutrale, methanollösliche Polyfluorene synthetisiert, die in Kombination mit einem unpolaren Polyindenofluorenderivat zum Aufbau mehrschichtiger PLEDs mittels nasschemischer Verfahren eingesetzt wurden. Zum anderen fand die Synthese von amphiphilen Polyfluorenen statt, die als Emulgatoren zur Stabilisierung von inversen Emulsionen eingesetzt wurden. Dabei konnten mit Hilfe eines bereits für die Darstellung von anorganisch-organischen Kern-Schale-Partikeln etablierten in situ-Verfahrens formanisotrope, kristalline Zinkoxidkerne mit konjugierter Polymerhülle erhalten werden.

In dieser Arbeit werden formstabile, amphiphile, oberflächenstrukturierte Polyphenylendendrimere (PPDs) mit verschiedenen Oberflächenpolaritäten beschrieben. Die physikalisch-chemischen Eigenschaften dieser Makromoleküle wurden studiert, welche ein gutes Verständnis der Nanoumgebung amphiphiler PPDs lieferten. Auch lichtinduzierte Polaritätsänderung wurde untersucht. Mit dem Konzept einer gleichmäßigen Verteilung polarer Bereiche auf der Peripherie hydrophober PPPs gelang es, Transportsysteme für Fettsäuren und Zytostatika zu erzeugen, welche charakteristische Merkmale natürlicher Transportproteine wie Albumin in sich vereinen. Hierzu zählen eine stabile dreidimensionale Form, die Ausbildung von Bindungstaschen sowie eine definierte strukturierte Oberfläche aus hydrophilen und hydrophoben Bereichen. Die Verfügbarkeit von lipophilen Bindungstaschen übertrifft sogar die des Albumins. Im Gegensatz zu Polymeren kann die Wirkstoffaufnahme bei PPDs exakt bestimmt werden. Die Anpassung der peripheren Gruppen beeinflusst den zellulären Aufnahmemechanismus. Es konnten effiziente Zellaufnahmen in A549-Zellen sowie der Transport und die intrazelluläre Freisetzung von Doxorubicin erreicht werden. Manche PPDs bieten eine Größe und Architektur, die es ermöglicht, Endothelzellen des Gehirns zu durchdringen. Es wurde auch der andere Extremfall untersucht, indem alle polaren Gruppen auf einer Hemisphäre akkumuliert wurden. Zur Darstellung solcher Janus-Dendrimere wurde ein neues Synthesekonzept herausgearbeitet und die erhaltenen Janus-Dendrimere mittels Lichtstreuung untersucht, wobei definierte perlenschnurartige Aggregate gefunden wurden. Weiterhin wurden semifluorierte Amphiphile vorgestellt, welche die Möglichkeit zur Selbstorganisation durch Nanophasenseparation bieten.

Die vorliegende Arbeit behandelt die Darstellung von neuen fluorierten Amphiphilen in Hinblick auf deren spätere Anwendung zur Modifikation von flüssig-gasförmigen, flüssig-flüssigen und flüssig-festen Grenzflächen. Jedes der drei Themengebiete richtete sich an eine andere Grenzfläche. Die erste Aufgabe wurde mit der Darstellung und Charakterisierung von Fluortensiden kurzen Perfluoralkyl-Ketten erfüllt, welche als möglichen umweltfreundlichen Ersatz für die Derivate der langkettigen Perfluoroktancarbonsäure und Perfluoroktansulfonsäure Anwendung finden. Es wurden Fluortenside entwickelt, welche die Oberflächenspannung von Wasser auf bis zu 18 mN/m absenken können. Der zweite Fokus lag auf der Stabilisierung einer fluorierten Emulsion, die sich für die carbo-kationische Polymerisationen eignete. Im Rahmen der Arbeit wurde das neue Blockcopolymer Poly(isobutylen-b-pentafluorstyrol) dargestellt. Neben der Homopolymerisation von Isobutylen zur PIB-Partikelsynthese wurde die Eignung des Stabilisator-Emulsionssystems durch die Darstellung von hart-weichen PS-PIB-Hybridpartikeln erfolgreich verfolgt. Dazu wurde die Synthese von quervernetzten PS-Kernpartikeln und der anschließenden Umhüllung durch PIB in einem weiteren Schritt. Im dritten Teil wurde die Adsorption von amphiphilen Fluorpolymeren zur Solvophobierung von rauen anorganischen Oberflächen untersucht. Es konnten sowohl superhydrophobe als auch superoleophobe Eigenschaften erzielt werden.

The synthesis and incorporation into oligodeoxynucleotides of two novel derivatives of bicyclothymidine carrying a cationic diaminopropyl or lysine unit in the C(6′)-β position is described. Compared to unmodified DNA these oligonucleotides show Tm-neutral behavior when paired against complementary DNA and are destabilizing when paired against RNA. Unaided uptake experiments of a decamer containing five lys-bcT units into HeLa and HEK293T cells showed substantial internalization with mostly cytosolic distribution which was not observed in the case of an unmodified control oligonucleotide.

The impact of di-cationic pentamidine-analogues against Toxoplama gondii (Rh- and Me49-background) was investigated. The 72 h-growth assays showed that the arylimidamide DB750 inhibited the proliferation of tachyzoites of T. gondii Rh and T. gondii Me49 with an IC(50) of 0.11 and 0.13 muM, respectively. Pre-incubation of fibroblast monolayers with 1 muM DB750 for 12 h and subsequent culture in the absence of the drug also resulted in a pronounced inhibiton of parasite proliferation. However, upon 5-6 days of drug exposure, T. gondii tachyzoites adapted to the compound and resumed proliferation up to a concentration of 1.2 muM. Out of a set of 32 di-cationic compounds screened for in vitro activity against T. gondii, the arylimidamide DB745, exhibiting an IC(50) of 0.03 muM and favourable selective toxicity was chosen for further studies. DB745 also inhibited the proliferation of DB750-adapted T. gondii (IC(50)=0.07 muM). In contrast to DB750, DB745 also had a profound negative impact on extracellular non-adapted T. gondii tachyzoites, but not on DB750-adapted T. gondii. Adaptation of T. gondii to DB745 (up to a concentration of 0.46 muM) was much more difficult to achieve and feasible only over a period of 110 days. In cultures infected with DB750-adapted T. gondii seemingly intact parasites could occasionally be detected by TEM. This illustrates the astonishing capacity of T. gondii tachyzoites to adapt to environmental changes, at least under in vitro conditions, and suggests that DB745 could be an interesting drug candidate for further assessments in appropriate in vivo models.

Macrophages become activated by bacterial endotoxin (lipopolysaccharide) and other stimuli to release proinflammatory cytokines and NO. To prevent release of toxic or potentially lethal quantities of these factors, the state of macrophage activation is counter-regulated by anti-inflammatory mediators (e.g., glucocorticoid hormones, interleukin 10, and transforming growth factor type β). Fetuin, a negative acute-phase protein, recently was implicated as an anti-inflammatory mediator, because it is required for macrophage deactivation by spermine. In the present studies, we found that fetuin is necessary for macrophages to respond to CNI-1493, a tetravalent guanylhydrazone inhibitor of p38 mitogen-activated protein kinase phosphorylation. Fetuin dose-dependently increases macrophage uptake of CNI-1493, which can be specifically inhibited by anti-human fetuin antibodies. Anti-human fetuin antibodies render primary human peripheral blood mononuclear cells insensitive to deactivation by CNI-1493. Thus, macrophages use fetuin as an opsonin for cationic-deactivating molecules, both endogenous (e.g., spermine) and pharmacologic (e.g., CNI-1493). This role of fetuin as an opsonic participant in macrophage-deactivating mechanisms has implications for understanding and manipulating the innate immune response.

Using a group of structurally related cytofectins, the effects of different vehicle constituents and mixing techniques on the physical properties and biological activity of lipoplexes were systematically examined. Physical properties were examined using a combination of dye accessibility assays, centrifugation, gel electrophoresis and dynamic light scattering. Biological activity was examined using in vitro transfection. Lipoplexes were formulated using two injection vehicles commonly used for in vivo delivery (PBS pH 7.2 and 0.9% saline), and a sodium phosphate vehicle previously shown to enhance the biological activity of naked pDNA and lipoplex formulations. Phosphate was found to be unique in its effect on lipoplexes. Specifically, the accessible pDNA in lipoplexes formulated with cytofectins containing a γ-amine substitution in the headgroup was dependent on alkyl side chain length and sodium phosphate concentration, but the same effects were not observed when using cytofectins containing a β-OH headgroup substitution. The physicochemical features of the phosphate anion, which give rise to this effect in γ-amine cytofectins, were deduced using a series of phosphate analogs. The effects of the formulation vehicle on transfection were found to be cell type-dependent; however, of the formulation variables examined, the liposome/pDNA mixing method had the greatest effect on transgene expression in vitro. Thus, though predictive physical structure relationships involving the vehicle and cytofectin components of the lipoplex were uncovered, they did not extrapolate to trends in biological activity.

Effective gene therapy for lung tissue requires the use of efficient vehicles to deliver the gene of interest into lung cells. When plasmid DNA encoding chloramphenicol acetyltransferase (CAT) was administered intranasally to BALB/c mice without carrier lipids, CAT activity was detected in mouse lung extracts. Plasmid DNA delivered with optimally formulated commercially available transfection reagents expressed up to 10-fold more CAT activity in lung than observed with naked DNA alone. Liposome formulations consisting of (+/-)-N-(3-aminopropyl)-N,N-dimethyl-2,3-bis (dodecyloxy)-1-propanaminium bromide (GAP-DLRIE) plus the neutral colipid dioleoylphosphatidylethanolamine (DOPE) enhanced CAT expression by more than 100-fold relative to plasmid DNA alone. A single administration of GAP-DLRIE liposome-CAT DNA complexes to mouse lung elicited peak expression at days 1-4 posttransfection, followed by a gradual return to baseline by day 21 postadministration. Readministration of GAP-DLRIE liposome CAT complexes at day 21 led to another transient peak of reporter gene expression. Histological examination of lungs treated with GAP-DLRIE complexed beta-galactosidase DNA revealed that alveolar epithelial cells were the primary locus of expression and that up to 1% of all alveoli contained epithelial cells expressing the transgene.

We propose a mechanism for oligonucleotide (ODN) release from cationic lipid complexes in cells that accounts for various observations on cationic lipid-nucleic acid-cell interactions. Fluorescent confocal microscopy of cells treated with rhodamine-labeled cationic liposome/ fluorescein-labeled ODN (F-ODN) complexes show the F-ODN separates from the lipid after internalization and enters the nucleus leaving the fluorescent lipid in cytoplasmic structures. ODN displacement from the complex was studied by fluorescent resonance energy transfer. Anionic liposome compositions (e.g., phosphatidylserine) that mimic the cytoplasmic facing monolayer of the cell membrane released ODN from the complex at about a 1:1 (-/+) charge ratio. Release was independent of ionic strength and pH. Physical separation of the F-ODN from monovalent and multivalent cationic lipids was confirmed by gel electrophoresis. Fluid but not solid phase anionic liposomes are required, whereas the physical state of the cationic lipids does not effect the release. Water soluble molecules with a high negative linear charge density, dextran sulfate, or heparin also release ODN. However, ATP, spermidine, spermine, tRNA, DNA, polyglutamic acid, polylysine, bovine serum albumin, or histone did not release ODN, even at 100-fold charge excess (-/+). Based upon these results, we propose that the complex, after internalization by endocytosis, induces flip-flop of anionic lipids from the cytoplasmic facing monolayer. Anionic lipids laterally diffuse into the complex and form a charged neutralized ion-pair with the cationic lipids. This leads to displacement of the ODN from the cationic lipid and its release into the cytoplasm.

Two cationic lipids, bis-guanidinium-spermidine-cholesterol (BGSC) and bis-guanidinium-trencholesterol (BGTC)-cholesterol derivatives bearing two guanidinium groups-have been synthesized and tested as artificial vectors for gene transfer. They combine the membrane compatible features of the cholesterol subunit and the favorable structural and high pKa features of the guanidinium functions for binding DNA via its phosphate groups. Reagent BGTC is very efficient for transfection into a variety of mammalian cell lines when used as a micellar solution. In addition, both BGTC and BGSC present also a high transfection activity when formulated as liposomes with the neutral phospholipid dioleoylphosphatidyl ethanolamine. These results reveal the usefulness of cholesterol derivatives bearing guanidinium groups for gene transfer.
